For a better understanding of living tissues and materials, it is essential to study the intricate spatial relationship between cells and their surrounding tissue on the nanoscale, with a need for 3D, high‐resolution imaging techniques. In the case of bone, focused ion beam‐scanning electron microscopy (FIB‐SEM) operated in the backscattered electron (BSE) mode proves to be a suitable method to image mineralized areas with a nominal resolution of 5 nm. However, as clinically relevant samples are often resin‐embedded, the lack of atomic number (Z) contrast makes it difficult to distinguish the embedding material from unmineralized parts of the tissue, such as osteoid, in BSE images. Staining embedded samples with iodine vapor has been shown to be effective in revealing osteoid microstructure by 2D BSE imaging. Based on this idea, an iodine (Z = 53) staining protocol is developed for 3D imaging with FIB‐SEM, investigating how the amount of iodine and exposure time influences the imaging outcome. Bone samples stained with this protocol also remain compatible with confocal laser scanning microscopy to visualize the lacunocanalicular network. The proposed protocol can be applied for 3D imaging of tissues exhibiting mineralized and nonmineralized regions to study physiological and pathological biomineralization.

Osteocyte lacuno-canalicular network (LCN) is comprised of micrometre-sized pores and submicrometric wide channels in bone. Accumulating evidence suggests multiple functions of this network in material transportation, mechanobiological signalling, mineral homeostasis and bone remodelling. Combining rhodamine staining and confocal laser scanning microscopy, the longitudinal cross-sections of six mouse tibiae were imaged, and the connectome of the network was quantified with a focus on the spatial heterogeneities of network density, connectivity and length of canaliculi. In-vivo loading and double calcein labelling on these tibiae allowed differentiating the newly formed bone from the pre-existing regions. The canalicular density of the murine cortical bone varied between 0.174 and 0.243 μm/μm3, and therefore is three times larger than the corresponding value for human femoral midshaft osteons. The spatial heterogeneity of the network was found distinctly more pronounced across the cortex than along the cortex. We found that in regions with a dense network, the LCN conserves its largely tree-like character, but increases the density by including shorter canaliculi. The current study on healthy mice should serve as a motivating starting point to study the connectome of genetically modified mice, including models of bone diseases and of reduced mechanoresponse.

The direct electrochemical conversion of ethanol, a sustainable fuel, is an alternative sustainable technology of the future. In this study, membrane electrode assemblies with different electrode configurations for an alkaline direct ethanol fuel cell were fabricated and tested in a fuel cell device. The configurations include a catalyst-coated substrate (CCS), a catalyst-coated membrane (CCM), and a mixture of these two fabrication options. Two different anion exchange membranes were used to perform a comprehensive analysis. The fabricated CCSs and CCMs were characterized with single cell measurements, electro­chemical impedance spectroscopy and scanning electron microscopy. In addition, the swelling behavior of the membranes in alkaline solution was investigated in order to obtain information for CCM production. The results of the experimental electrochemical tests show that the CCS approach provides higher power densities (42.4 mW cm-2) than the others, regardless of the membrane type.

The sluggish kinetics of the ethanol oxidation reaction (EOR) and the related development of low-cost, highly active and stable anode catalysts still remains the major challenge in alkaline direct ethanol fuel cells (ADEFCs). In this respect, we synthesized a PdNiBi nanocatalyst on reduced graphene oxide (rGO) via a facile synthesis method. The prepared composite catalyst was physicochemically characterized by SEM, STEM, EDX, ICP-OES and XRD to analyze the morphology, particle distribution and size, elemental composition and structure. The electrochemical activity and stability towards EOR in alkaline media were examined using the thin-film rotating disk electrode technique. The results reveal well-dispersed and strongly anchored nanoparticles on the rGO support, providing abundant active sites. The PdNiBi/rGO presents a higher EOR activity and stability compared to a commercial Pd/C ascribed to a high ECSA and synergistic effects between Pd, Ni and Bi and the rGO material. These findings suggest PdNiBi/rGO as a promising anode catalyst in ADEFC applications.

SP7 is a transcription factor required for osteoblast differentiation and bone formation. A neomorphic mutation in SP7 was found to alter DNA binding specificity, causing a complex skeletal disorder in both mice and humans.

The shelf-life of catalyst ink for fabricating polymer electrolyte fuel cells (PEFCs) is relevant for large-scale manufacturing with unforeseen production stops. In this study, the storage effects on the physicochemical characteristics of catalyst ink (Pt/C, Nafion, 2-propanol, water) and subsequently manufactured catalyst layers are investigated. Sedimentation analysis showed that catalyst particles are not fully stabilized by charge interaction induced by Nafion. Acetone was found to be an oxidation product, even in freshly prepared ink with platinum catalyzing the reaction. Rotating disk electrode analysis revealed that the electrochemically active surface area is, overall, minimally increased by storage, and the selectivity towards water formation (4-electron pathway) is unharmed within the first 48 h of storage. MEAs prepared from stored ink reach almost the same current density level after conditioning via potential cycling. The open-circuit voltage (OCV) increases due to increased catalyst availability. Scanning electron microscopy and mercury intrusion porosimetry showed that with increasing acetone content, the pore structure becomes finer, with a higher specific surface area. Electrochemical impedance spectroscopy revealed that this results in a more hindered mass transfer but lowered charge transfer resistance. The MEA with the highest OCV and power output and the lowest overall cell resistance was fabricated from catalyst ink stored for a duration of four weeks.

Crosslinked anion exchange membranes (AEMs) made from poly(vinyl alcohol) (PVA) as a backbone polymer and different approaches to functional group introduction were prepared by means of solution casting with thermal and chemical crosslinking. Membrane characterization was performed by SEM, FTIR, and thermogravimetric analyses. The performance of AEMs was evaluated by water uptake, swelling degree, ion exchange capacity, OH- conductivity, and single cell tests. A combination of quaternized ammonium poly(vinyl alcohol) (QPVA) and poly(diallyldimethylammonium chloride) (PDDMAC) showed the highest conductivity, water uptake, and swelling among other functional group sources. The AEM with a combined mass ratio of QPVA and PDDMAC of 1:0.5 (QPV/PDD0.5) has the highest hydroxide conductivity of 54.46 mS cm-1. The single fuel cell tests with QPV/PDD0.5 membrane yield the maximum power density and current density of 8.6 mW cm-2 and 47.6 mA cm-2 at 57 °C. This study demonstrates that PVA-based AEMs have the potential for alkaline direct ethanol fuel cells (ADEFCs) application.

Objective: The objective of the study was to specify the thickness of Zn and Pb accumulation within the tidemark (TM), a narrow structure between the non-calcified and the calcified articular cartilage. It is considered an active or resting calcification front. This banded structure of the cartilage-bone interface is known to undergo changes in osteoarthritis. Therefore, gaining knowledge about this structure is of interest. Methods: Femoral head samples were collected from patients suffering from various bone diseases, 6 samples have been investigated. Thin bone slices (3 ​μm thick) were measured with high resolution synchrotron micro-X-ray fluorescence (SR micro-XRF) analysis using a beam with dimensions of 500 ​× ​800 ​nm2. The tidemark region was found in all analyzed samples. The Savitzky-Golay filter was used to smooth the measured imaging data and Kaplan-Meier estimation to gain reliable tidemarks medians for Pb and Zn. To our knowledge this was the first time that these methods have been applied to gain information on histological structures obtained by elemental imaging. Results: The thickness of the Zn and Pb layer ranged from about 3 to 11 ​μm for Zn and 4–14.5 ​μm for Pb. Our Zn ratios (TM/matrix) were found to be 1.5-3-fold ratio between Zn tidemark values and in mineralized matrix and are similar in all samples. Conclusions: The determined thickness of the layer is much smaller than found in previous measurements with the beam having 20 ​× ​14 ​μm2 size. The Zn ratios agree with our previous findings.

In this work, fully polysaccharide based membranes were presented as self-standing, solid polyelectrolytes for application in anion exchange membrane fuel cells (AEMFCs). For this purpose, cellulose nanofibrils (CNFs) were modified successfully with an organosilane reagent, resulting in quaternized CNFs (CNF (D)), as shown by Fourier Transform Infrared Spectroscopy (FTIR), Carbon-13 (C13) nuclear magnetic resonance (13C NMR), Thermogravimetric Analysis (TGA)/Differential Scanning Calorimetry (DSC), and ζ-potential measurements. Both the neat (CNF) and CNF(D) particles were incorporated in situ into the chitosan (CS) membrane during the solvent casting process, resulting in composite membranes that were studied extensively for morphology, potassium hydroxide (KOH) uptake and swelling ratio, ethanol (EtOH) permeability, mechanical properties, ionic conductivity, and cell performance. The results showed higher Young’s modulus (119%), tensile strength (91%), ion exchange capacity (177%), and ionic conductivity (33%) of the CS-based membranes compared to the commercial Fumatech membrane. The addition of CNF filler improved the thermal stability of the CS membranes and reduced the overall mass loss. The CNF (D) filler provided the lowest (4.23 × 10−5 cm2 s−1) EtOH permeability of the respective membrane, which is in the same range as that of the commercial membrane (3.47 × 10−5 cm2s−1). The most significant improvement (~78%) in power density at 80 °C was observed for the CS membrane with neat CNF compared to the commercial Fumatech membrane (62.4 mW cm−2 vs. 35.1 mW cm−2). Fuel cell tests showed that all CS-based anion exchange membranes (AEMs) exhibited higher maximum power densities than the commercial AEMs at 25 °C and 60 °C with humidified or non-humidified oxygen, demonstrating their potential for low-temperature direct ethanol fuel cell (DEFC) applications.

In recent years, there has been considerable interest in anion exchange membrane fuel cells (AEMFCs) as part of fuel cell technology. Anion exchange membranes (AEMs) provide a significant contribution to the development of fuel cells, particularly in terms of performance and efficiency. Polymer composite membranes composed of quaternary ammonium poly(vinyl alcohol) (QPVA) as electrospun nanofiber mats and a combination of QPVA and poly(diallyldimethylammonium chloride) (PDDA) as interfiber voids matrix filler were prepared and characterized. The influence of various QPVA/PDDA mass ratios as matrix fillers on anion exchange membranes and alkaline fuel cells was evaluated. The structural, morphological, mechanical, and thermal properties of AEMs were characterized. To evaluate the AEMs’ performances, several measurements comprise swelling properties, ion exchange capacity (IEC), hydroxide conductivity (σ), alkaline stability, and single-cell test in fuel cells. The eQP-PDD0.5 acquired the highest hydroxide conductivity of 43.67 ms cm−1 at 80 °C. The tensile strength of the membranes rose with the incorporation of the filler matrix, with TS ranging from 23.18 to 24.95 Mpa. The peak power density and current density of 24 mW cm−2 and 131 mA cm−2 were achieved with single cells comprising eQP-PDD0.5 membrane at 57 °C.

Alkaline direct ethanol fuel cells (DEFCs) represent an efficient energy conversion device for sustainable ethanol fuel. In this study, a design with new structural parameters for the anodic flow field of the alkaline DEFC was modeled with the aid of computational fluid dynamics and was then actually constructed. Single-cell tests were performed to evaluate the impact of the developed design on fuel cell performance. The results show that fuel cell performance significantly increased when using the improved design in the low-temperature range. The higher the temperature in the cell, the lower the influence of the flow field structure on performance. In addition, the influence of external factors, such as the orientation of the cell, the preheating of the fuel, and the direction of the two fuel flows relative to each other (co-current and counter-current), are shown.

Bone formation and resorption are typically coupled, such that the efficacy of anabolic osteoporosis treatments may be limited by bone destruction. The multi-kinase inhibitor YKL-05–099 potently inhibits salt inducible kinases (SIKs) and may represent a promising new class of bone anabolic agents. Here, we report that YKL-05–099 increases bone formation in hypogonadal female mice without increasing bone resorption. Postnatal mice with inducible, global deletion of SIK2 and SIK3 show increased bone mass, increased bone formation, and, distinct from the effects of YKL-05–099, increased bone resorption. No cell-intrinsic role of SIKs in osteoclasts was noted. In addition to blocking SIKs, YKL-05–099 also binds and inhibits CSF1R, the receptor for the osteoclastogenic cytokine M-CSF. Modeling reveals that YKL-05–099 binds to SIK2 and CSF1R in a similar manner. Dual targeting of SIK2/3 and CSF1R induces bone formation without concomitantly increasing bone resorption and thereby may overcome limitations of most current anabolic osteoporosis therapies.

In this study, Ag-MnxOy/C composite catalysts deposited on reduced graphene oxide (rGO) and, for the first time on N-doped graphene oxide (NGO), were prepared via a facile synthesis method. The influence of the carbon support material on the activity and stability of the oxygen reduction reaction (ORR) and on the tolerance to ethanol in alkaline medium was focused and investigated. The physicochemical properties of the Ag-MnxOy/C catalysts were analyzed by X-ray diffraction (XRD), scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDS), Brunauer–Emmett–Teller (BET) method, atomic absorption spectroscopy (AAS), inductively coupled plasma-mass spectrometry (ICP-MS), and thermogravimetric gas analysis (TGA). Electrochemical characterization was performed by rotating disk electrode (RDE) experiments. The results show that the active manganese species MnO2 was assembled as nanorods and nanospheres on rGO and NGO, respectively. Ag was assumed to be present as very small or amorphous particles. Similar redox processes for Ag-MnxOy/rGO and Ag-MnxOy/NGO were examined via cyclic voltammetry. The Ag-MnxOy/rGO resulted in a more negative diffusion limiting current density of −3.01 mA cm−2 compared to Ag-MnxOy/NGO. The onset potential of approximately 0.9 V vs. RHE and the favored 4-electron transfer pathway were independent of the support material. Ag-MnxOy/NGO exhibited a higher ORR stability, whereas Ag-MnxOy/rGO showed a better ethanol tolerance.

Bone mineral density distributions (BMDDs) are a measurable property of bone tissues that depends strongly on bone remodelling and mineralisation processes. These processes can vary significantly in health and disease and across skeletal sites, so there is high interest in analysing these processes from experimental BMDDs. Here, we propose a rigorous hypothesis-testing approach based on a mathematical model of mineral heterogeneity in bone due to remodelling and mineralisation, to help explain differences observed between the BMDD of human femoral cortical bone and the BMDD of human trabecular bone. Recent BMDD measurements show that femoral cortical bone possesses a higher bone mineral density, but a similar mineral heterogeneity around the mean compared to trabecular bone. By combining this data with the mathematical model, we are able to test whether this difference in BMDD can be explained by (i) differences in turnover rate; (ii) differences in osteoclast resorption behaviour; and (iii) differences in mineralisation kinetics between the two bone types. We find that accounting only for differences in turnover rate is inconsistent with the fact that both BMDDs have a similar spread around the mean, and that accounting for differences in osteoclast resorption behaviour leads to biologically inconsistent bone remodelling patterns. We conclude that the kinetics of mineral accumulation in bone matrix must therefore be different in femoral cortical bone and trabecular bone. Although both cortical and trabecular bone are made up of lamellar bone, the different mineralisation kinetics in the two types of bone point towards more profound structural differences than usually assumed.

Osteocytes interconnect with each other forming an intricate cell network within the mineralized bone matrix. One important function of the osteocyte network is the mechano-regulation of bone remodeling, where a possible mechanism includes the fluid flow through the porosity housing the cell network - the osteocyte lacuno-canalicular network (OLCN). In our study the OLCN in human osteons was three-dimensionally imaged with the aim to obtain a quantitative description of the canalicular density and spatial variations of this quantity within osteons. The topology of the OLCN was determined by first staining the bone samples with rhodamine, then imaging the OLCN with confocal laser scanning microscopy and finally using image analysis to obtain a skeletonized version of the network for further analysis. In total 49 osteons were studied from the femoral cortical bone of four different middle-aged healthy women. The mean canalicular density given as length of the canaliculi in a unit volume was 0.074 ± 0.015 μm/μm3 (corresponding to 74 km/cm3). No correlation was found between the canalicular density and neither the size of the osteon nor the volume fraction occupied by osteocyte lacunae. Within osteons the canalicular density varied substantially with larger regions without any network. On average the canalicular density decreases when moving from the Haversian canal outwards towards the cement line. We hypothesize that a decrease in accessible canaliculi with tissue age as a result of micropetrosis can reduce the local mechanosensitivity of the bone. Systematic future studies on age- and disease-related changes on the topology of the OLCN have to demonstrate the diagnostic potential of the presented characterization method.

The influence of treatment with alendronate (ALN), teriparatide (TPTD) or concurrent treatment with both on the human bone matrix mineralization has not yet been fully elucidated. For this purpose we analyzed quadruple fluorochrome labelled transiliac bone biopsy samples (n = 66) from postmenopausal osteoporotic women with prior and ongoing ALN (ALN-Rx arm) or without ALN (Rx-Naïve arm) after 7 months treatment with cyclic or daily TPTD or without TPTD using quantitative backscattered electron imaging and confocal scanning laser microscopy. Additionally to the bone mineralization density distribution (BMDD) of entire cancellous and cortical compartments, we measured the mineralization kinetics, i.e. the calcium concentration between the younger (Ca_DL2) and older double labels (Ca_DL1), and in interstitial bone (Ca_int) in a subset of the biopsy cohort.We found the BMDD from the patients with prior and ongoing ALN generally shifted to higher calcium concentrations compared to those without ALN (average degree of mineralization in cancellous bone Cn.CaMean + 3.1%, p

The design and study of efficient polymer-based drug delivery systems for the controlled release of anticancer drugs is one of the pillars of nanomedicine. The fight against metastatic and invasive cancers demands therapeutic candidates with increased and selective toxicity towards malignant cells, long-term activity and reduced side effects. In this sense, polyphosphazene nanocarriers were synthesized for the sustained release of the anticancer drugs camptothecin (CPT) and epirubicin (EPI). Linear poly(dichloro)phosphazene was modified with lipophilic tocopherol or testosterone glycinate, with antioxidant and antitumor activity, and with hydrophilic Jeffamine M1000 to obtain different polyphosphazene nanocarriers. It allowed us to encapsulate the lipophilic CPT and the more hydrophilic EPI. The encapsulation process was carried out via solvent exchange/precipitation, attaining a 9.2–13.6 wt% of CPT and 0.3–2.4 wt% of EPI. CPT-loaded polyphosphazenes formed 140–200 nm aggregates in simulated body physiological conditions (PBS, pH 7.4), resulting in an 80–100-fold increase of CPT solubility. EPI-loaded polyphosphazenes formed 250 nm aggregates in an aqueous medium. CPT and EPI release (PBS, pH 7.4, 37 °C) was monitored for 202 h, being almost linear during the first 8 h. The slow release of testosterone and tocopherol was also sustained for 150 h in PBS (pH 7.4 and 6.0) at 37 °C. The co-delivery of testosterone or tocopherol and the anticancer drugs from the nanocarriers was expected. Cells of the human breast cancer cell line MCF-7 demonstrated good uptake of anticancer-drug-loaded nanocarriers after 6 h. Similarly, MCF-7 spheroids showed good uptake of the anticancer-drug-loaded aggregates after 72 h. Almost all anticancer-drug-loaded polyphosphazenes exhibited similar or superior toxicity against MCF-7 cells and spheroids when compared to raw anticancer drugs. Additionally, cell-cycle arrest in the G2/M phase was increased in response to the drug-loaded nanocarriers. Almost no toxicity of anticancer-drug-loaded aggregates against primary human lung fibroblasts was observed. Furthermore, the aggregates displayed no hemolytic activity, which is in contrast to the parent anticancer drugs. Consequently, synthesized polyphosphazene-based nanocarriers might be potential nanomedicines for chemotherapy.

ABSTRACT Schnurri‐3 (Shn3) is an essential regulator of postnatal skeletal remodeling. Shn3‐deficient mice (Shn3–/–) have high bone mass; however, their bone mechanical and material properties have not been investigated to date. We performed three‐point bending of femora, compression tests of L3 vertebrae. We also measured intrinsic material properties, including bone mineralization density distribution (BMDD) and osteocyte lacunae section (OLS) characteristics by quantitative backscatter electron imaging, as well as collagen cross‐linking by Fourier transform infrared microspectroscopy of femora from Shn3–/– and WT mice at different ages (6 weeks, 4 months, and 18 months). Abstract Background Whilst hypocalcemic complications from vitamin D deficiency are considered rare in high-income countries, they are highly prevalent among Black, Asian and Minority Ethnic (BAME) group with darker skin. To date, the extent of osteomalacia in such infants and their family members is unknown. Our aim was to investigate clinical, cardiac and bone histomorphometric characteristics, bone matrix mineralization in affected infants and to test family members for biochemical evidence of osteomalacia. Case presentation Three infants of BAME origin (aged 5–6 months) presented acutely in early-spring with cardiac arrest, respiratory arrest following seizure or severe respiratory distress, with profound hypocalcemia (serum calcium 1.22–1.96 mmol/L). All infants had dark skin and vitamin D supplementation had not been addressed during child surveillance visits. All three had severely dilated left ventricles (z-scores + 4.6 to + 6.5) with reduced ejection fraction (25–30%; normal 55–70), fractional shortening (7 to 15%; normal 29–40) and global hypokinesia, confirming hypocalcemic dilated cardiomyopathy.
