10. Short Serious Games Creation under the Paradigm of Software Process and Competencies as Software Requirements. Case Study: Elementary Math Competencies

Abstract

Development of digital resources is difficult due to their particular complexity relying on pedagogical aspects. Another aspect is the lack of well-defined development processes, experiences documented, and standard methodologies to guide and organize game development. Added to this, there is no documented technique to ensure correct implementation of a competency in a videogame. This research proposes a Short Serious Game Development Process founded in Software Engineering paradigms and complemented by previous efforts on large scale development of digital learning resources. This paper focuses in a technique called Competency-Based Decomposition that achieves implementing a formal competency into a short serious game, with which the formal learning process will be complemented to improve the way students learn. Through a case study will be demonstrated its utility by implementing the process and the technique in the whole of mathematics competencies for sixth grade of elementary school in Mexico. The result of applying the proposed process for study case is a collection of video games that satisfactorily implements the competencies and its contents, its expected learning and its knowledge areas specified for sixth grade of elementary school in Mexico by the Ministry of Public Education.

Abstract

Legionella longbeachae and Legionella pneumophila are the most common causative agents of Legionnaires' disease. While the clinical manifestations caused by both species are similar, species-specific differences exist in environmental niches, disease epidemiology, and genomic content. One such difference is the presence of a genomic locus predicted to encode a capsule. Here, we show that L. longbeachae indeed expresses a capsule in post-exponential growth phase as evidenced by electron microscopy analyses, and that capsule expression is abrogated when deleting a capsule transporter gene. Capsule purification and its analysis via HLPC revealed the presence of a highly anionic polysaccharide that is absent in the capsule mutant. The capsule is important for replication and virulence in vivo in a mouse model of infection and in the natural host Acanthamoeba castellanii. It has anti-phagocytic function when encountering innate immune cells such as human macrophages and it is involved in the low cytokine responses in mice and in human monocyte derived macrophages, thus dampening the innate immune response. Thus, the here characterized L. longbeachae capsule is a novel virulence factor, unique among the known Legionella species, which may aid L. longbeachae to survive in its specific niches and which partly confers L. longbeachae its unique infection characteristics. Author summary: Legionella longbeachae can cause a severe pneumonia, known as Legionnaires' disease. In Australia and New Zealand, L. longbeachae is the predominant species causing up to 50% of all infections due to Legionella. However, L. longbeachae virulence factors are nearly unknown. Here, we show that L. longbeachae expresses a capsule that is a major virulence factor of this pathogen as it is important for virulence in vivo in mice and in the environment in its natural host Acanthamoeba castellanii. It dampens the innate immune response in mice and human cells. Our study sheds light on an understudied environmental pathogen and identifies a new virulence feature of Legionellae. Abstract

Complications from diabetic retinopathy such as diabetic macular edema (DME) and proliferative diabetic retinopathy (PDR) constitute leading causes of preventable vision loss in working-age patients. Since vascular endothelial growth factor (VEGF) plays a major role in the pathogenesis of these complications, VEGF inhibitors have been the cornerstone of their treatment. Anti-VEGF monotherapy is an effective but burdensome treatment for DME. However, due to the intensive and burdensome treatment, most patients in routine clinical practice are undertreated, and therefore, their outcomes are compromised. Even in adequately treated patients, persistent DME is reported anywhere from 30% to 60% depending on the drug used. PDR is currently treated by anti-VEGF, panretinal photocoagulation (PRP) or a combination of both. Similarly, a number of eyes, despite these treatments, continue to progress to tractional retinal detachment and vitreous hemorrhage. Clearly there are other molecular pathways other than VEGF involved in the pathogenesis of DME and PDR. One of these pathways is the angiopoietin–Tie signaling pathway. Angiopoietin 1 (Ang1) plays a major role in maintaining vascular quiescence and stability. It acts as a molecular brake against vascular destabilization and inflammation that is usually promoted by angiopoietin 2 (Ang2). Several pathological conditions including chronic hyperglycemia lead to Ang2 upregulation. Recent regulatory approval of the bi-specific antibody, faricimab, may improve long term outcomes in DME. It targets both the Ang/Tie and VEGF pathways. The YOSEMITE and RHINE were multicenter, double-masked, randomized non-inferiority phase 3 clinical trials that compared faricimab to aflibercept in eyes with center-involved DME. At 12 months of follow-up, faricimab demonstrated non-inferior vision gains, improved anatomic outcomes and a potential for extended dosing when compared to aflibercept. The 2-year results of the YOSEMITE and RHINE trials demonstrated that the anatomic and functional results obtained at the 1 year follow-up were maintained. Short term outcomes of previously treated and treatment-naive eyes with DME that were treated with faricimab during routine clinical practice suggest a beneficial effect of faricimab over other agents. Abstract

The thermal stability and degradation mechanisms of three semicrystalline polymers (polyethylene terephthalate [PET]. high-density polyethylene [HDPE]. and polyamide 6 [nylon 6]) were studied. Thermogravimetric traces were acquired first at heating rates of 1[degrees]C/min and 10[degrees]C/min, and it was determined that the heating rate significantly affected the thermal decay curves of the three polymers. The results allowed the selection of specific temperatures at which to carry out heating and cooling cycles from room temperature to the molten state. The thermal behaviors of HDPE, nylon 6, and PET each had particular characteristics. HDPE showed the highest thermal resistance, whereas nylon 6 displayed the lowest. PET had the lowest activation energy for degradation, 93.5 kJ/mol, and retained 14wt% after thermal recycling with no influence of molecular weight. Thermal cycling also revealed gradual morphological changes in HDPE, nylon 6, and PET, and their crystals changed from regular to branched spherulites with variations in the nucleation patterns. Abstract

Featured Application: The methods described here can be applied in interferometric measurements of dynamic events that are challenging to analyze using conventional approaches incapable of evaluating a single interferogram with closed fringes. Frequency-guided sequential demodulation (FSD) is a technique used to recover the phase of closed fringe patterns when only a single interferogram is available. FSD is a sequential method that follows simple and intuitive processing steps for interferogram demodulation, guided by the frequency of the fringes. The objective of this work is to apply an optimizer block that feeds from the first stages of the FSD and make feedback in such a manner as to optimize the search for guided frequencies. This approach significantly reduces demodulation errors, especially when dealing with high-frequency fringe patterns. The results of using the FSD with and without optimizers are compared. The optimizers used are the Harmonic Search Optimization (HSO) as well as the Particle Swarm Optimization (PSO). Results show that FSD with PSO delivers better results compared with other methods here analyzed. [ABSTRACT FROM AUTHOR]
