Abstract A network of marine reserves can enhance yield in depleted fisheries by protecting populations, particularly large, old spawners that supply larvae for interspersed fishing grounds. The ability of marine reserves to enhance sustainable fisheries is much less evident. We report empirical evidence of a marine reserve network improving yield regionally for a sustainable spiny lobster fishery, apparently through the spillover of adult lobsters and behavioral adaptation by the fishing fleet. Results of a Before-After, Control-Impact analysis found catch, effort, and Catch-Per-Unit Effort increased after the establishment of marine reserves in the northern region of the fishery where fishers responded by fishing intensively at reserve borders, but declined in the southern region where they vacated once productive fishing grounds. The adaptation of the northern region of the fishery may have been aided by a history of collaboration between fishers, scientists, and managers, highlighting the value of collaborative research and education programs for preparing fisheries to operate productively within a seascape that includes a large marine reserve network.

Abstract Macrocystis pyrifera (giant kelp), is a brown macroalga of great ecological importance as a primary producer and structure-forming foundational species that provides habitat for hundreds of species. It has many commercial uses (e.g. source of alginate, fertilizer, cosmetics, feedstock). One of the limitations to exploiting giant kelp’s economic potential and assisting in giant kelp conservation efforts is a lack of genomic tools like a high quality, contiguous reference genome with accurate gene annotations. Reference genomes attempt to capture the complete genomic sequence of an individual or species, and importantly provide a universal structure for comparison across a multitude of genetic experiments, both within and between species. We assembled the giant kelp genome of a haploid female gametophyte de novo using PacBio reads, then ordered contigs into chromosome level scaffolds using Hi-C. We found the giant kelp genome to be 537 MB, with a total of 35 scaffolds and 188 contigs. The assembly N50 is 13,669,674 with GC content of 50.37%. We assessed the genome completeness using BUSCO, and found giant kelp contained 94% of the BUSCO genes from the stramenopile clade. Annotation of the giant kelp genome revealed 25,919 genes. Additionally, we present genetic variation data based on 48 diploid giant kelp sporophytes from three different Southern California populations that confirms the population structure found in other studies of these populations. This work resulted in a high-quality giant kelp genome that greatly increases the genetic knowledge of this ecologically and economically vital species.

Foundation species are essential to ecosystem function, but their role as habitat providers is predicated on their spatial dominance. Worldwide, kelps, seagrasses, corals, and other marine foundation species have declined. This is true also for rockweeds, the canopy-forming analog of subtidal kelp forests in temperate rocky intertidal ecosystems. On the west coast of North America, dense beds of the rockweed Silvetia compressa occur across large biogeographic regions, benefitting numerous species by ameliorating physical stress caused by sun exposure, desiccation, heat, and wave disturbance. Like many rockweed species, Silvetia is long-lived, slow-growing, and short-dispersing – characteristics that reduce its resilience to disturbance. Using a generalized additive mixed-effects model with explicit spatial effects, we analyzed canopy cover data from 30 sites spanning 18 years, and we tested the hypothesis that Silvetia population trends are tightly linked to atmospheric climate conditions, particularly Santa Ana wind events (SAWs): strong, hot, and dry downslope winds that originate inland and move offshore. We found that the rockweed had declined markedly, particularly at sites south of the major biogeographic break, Point Conception (PC), including the California Channel Islands and southern California mainland, and a highly significant negative effect of dewpoint depression, a measure of moisture content in the atmosphere, on Silvetia cover across all three regions in this study. Our results suggest that any increases in the frequency or intensity of SAWs are likely to lead to large declines and possible extirpation of Silvetia, as well as the important ecological services the species provides.

Sessile invertebrates in Californian kelp forests exhibit prolonged changes to community structure following a marine heatwave event.

Abstract The aim of the present study was to evaluate in vitro and in vivo the acaricidal activity of two forms of geranium (Pelargonium graveolens) (PG). These two forms were the P. graveolens essential oil nanoemulsion (PGN), and the PG in combination with the sesame oil (SO), PGSO). These forms were first evaluated in vitro for their adulticidal, ovicidal, and larvicidal activities against the different stages of acaricide-resistant Rhipicephalus annulatus (Say). Geranium nanoemulsion was prepared and then characterized by UV–Vis spectrophotometer, and zeta droplet size measurement. The results revealed that LC50 of the PG against the adult ticks was attained at concentration of 7.53% while it was decreased to 1.91% and 5.60% for PGSO and PGN, respectively. Also, the LC50 of PGN and PGSO were reached at concentrations of 1.688 and 0.944%, respectively against the larvae while the LC50 of the PG was reached at concentration of 3.435% for. The combination of PGN with PG exhibited non-significant ovicidal effect meanwhile PGSO showed significant ovicidal effect even at the low concentration (2.5%). The PGSO and PGN formulations were applied in a field trial to control the ticks of the naturally infested cattle. PGSO and PGN significantly reduced the tick burden to 74.83% and 87.97%, respectively at 3 weeks post-application with performance better than the deltamethrin (29.88%). In conclusion, the two PG forms can be used as suitable alternatives to control R. annulatus tick and they need further modifications for effective field application.

Abstract Foundation species, such as trees, corals, grasses, oysters, and rockweeds, must be common and abundant to effectively modify the physical environment and increase biodiversity by buffering environmental stress. Yet many of these important species have been declining due to disease, climate change, and other factors. A prime example is the precipitous population decline of marine rockweeds, which is attributed to increased urbanization and its accompanying impacts. Rockweeds provide three‐dimensional habitat in harsh rocky intertidal environments and regulate ecosystem functioning, essential roles that no substitute species are capable of filling. Recovery of impacted rockweed populations is typically slow and unpredictable due to their limited dispersal capacity. These issues have motivated efforts to conserve remaining populations of rockweeds and reestablish or enhance depleted ones. Successfully doing so requires a robust understanding of factors that affect survival of the species and the processes that influence ecosystem structure, along with rigorous scientific testing of restoration methods and the factors that affect restoration success. In this comprehensive review, we summarize the current knowledge of rockweed ecology, highlight studies that could inform restoration practices, and recommend ways to improve our ability to implement scalable restoration of rockweeds and accompanying ecosystem‐wide benefits.

Abstract Background The pathogenesis of hematogenous orthopaedic implant-associated infections (HOIAI) remains largely unknown, with little understanding of the influence of the physis on bacterial seeding. Since the growth velocity in the physis of long bones decreases during aging, we sought to evaluate the role of the physis on influencing the development of Staphylococcus aureus HOIAI in a mouse model comparing younger versus older mice. Methods In a mouse model of HOIAI, a sterile Kirschner wire was inserted retrograde into the distal femur of younger (5–8-week-old) and older (14–21-week-old) mice. After a 3-week convalescent period, a bioluminescent Staphylococcus aureus strain was inoculated intravenously. Bacterial dissemination to operative and non-operative legs was monitored longitudinally in vivo for 4 weeks, followed by ex vivo bacterial enumeration and X-ray analysis. Results In vivo bioluminescence imaging and ex vivo CFU enumeration of the bone/joint tissue demonstrated that older mice had a strong predilection for developing a hematogenous infection in the operative legs but not the non-operative legs. In contrast, this predilection was less apparent in younger mice as the infection occurred at a similar rate in both the operative and non-operative legs. X-ray imaging revealed that the operative legs of younger mice had decreased femoral length, likely due to the surgical and/or infectious insult to the more active physis, which was not observed in older mice. Both age groups demonstrated substantial reactive bone changes in the operative leg due to infection. Conclusions The presence of an implant was an important determinant for developing a hematogenous orthopaedic infection in older but not younger mice, whereas younger mice had a similar predilection for developing periarticular infection whether or not an implant was present. On a clinical scale, diagnosing HOIAI may be difficult particularly in at-risk patients with limited examination or other data points. Understanding the influence of age on developing HOIAI may guide clinical surveillance and decision-making in at-risk patients.

Abstract Biodiversity is changing at an accelerating rate at both local and regional scales. Beta diversity, which quantifies species turnover between these two scales, is emerging as a key driver of ecosystem function that can inform spatial conservation. Yet measuring biodiversity remains a major challenge, especially in aquatic ecosystems. Decoding environmental DNA (eDNA) left behind by organisms offers the possibility of detecting species sans direct observation, a Rosetta Stone for biodiversity. While eDNA has proven useful to illuminate diversity in aquatic ecosystems, its utility for measuring beta diversity over spatial scales small enough to be relevant to conservation purposes is poorly known. Here we tested how eDNA performs relative to underwater visual census (UVC) to evaluate beta diversity of marine communities. We paired UVC with 12S eDNA metabarcoding and used a spatially structured hierarchical sampling design to assess key spatial metrics of fish communities on temperate rocky reefs in southern California. eDNA provided a more-detailed picture of the main sources of spatial variation in both taxonomic richness and community turnover, which primarily arose due to strong species filtering within and among rocky reefs. As expected, eDNA detected more taxa at the regional scale (69 vs. 38) which accumulated quickly with space and plateaued at only ~ 11 samples. Conversely, the discovery rate of new taxa was slower with no sign of saturation for UVC. Based on historical records in the region (2000–2018) we found that 6.9 times more UVC samples would be required to detect 50 taxa compared to eDNA. Our results show that eDNA metabarcoding can outperform diver counts to capture the spatial patterns in biodiversity at fine scales with less field effort and more power than traditional methods, supporting the notion that eDNA is a critical scientific tool for detecting biodiversity changes in aquatic ecosystems.

Abstract Sea urchins are key grazers in coastal seas, where they can survive a variety of conditions and diets, enhancing their ecological impact on kelp forests and other ecosystems. Using 16S rRNA gene sequencing, we characterized bacterial communities associated with guts of the two dominant sea urchin species in southern California, the red urchin Mesocentrotus franciscanus, and the purple urchin Strongylocentrotus purpuratus. Our results show that the two urchin species have distinct gut microbiomes that vary with habitat. The taxonomic composition of their microbiomes suggests that they may facilitate digestion of food and be a source of nutrition themselves. These results highlight the role of microbiomes within macroorganisms as an extended ecological trait, and suggest that microbes may be crucial to resource use and partitioning in co‐occurring species.

Monitoring is a crucial tool for measuring the progress and success of environmental policies and management programs. While many studies have evaluated the effectiveness of biodiversity sampling methods, few have compared their efficiency, which is crucial given the funding constraints present in all conservation efforts. In this study we demonstrate how existing analytical tools can be applied to (i) assess the relationship between sampling effort and resulting confidence in biodiversity metrics, and (ii) compare the efficiency of different methods for monitoring biodiversity. We tested this methodology on data from marine fish surveys, including: roving surveys within permanent areas, randomly placed belt transects, and randomly placed transects conducted by citizen scientists using a reduced species list. We constructed efficiency curves describing how increasing effort spent on each method reduced uncertainty in biodiversity estimates and the associated ability to detect change in diversity. All programs produced comparable measurements of species diversity for all metrics despite substantial differences in the species being surveyed by each method. The uncertainty of diversity estimations fell faster and reached a lower level for the roving diver method. Strikingly, the transect method conducted by citizen scientists performed almost identically to the more taxonomically resolved transect method conducted by professional scientists, suggesting that sampling strategies that recorded only a subset of species could still be effective, as long as the excluded species were chosen strategically. The methodology described here can guide decisions about how to measure biodiversity and optimize the resources available for monitoring, ultimately improving management outcomes.

Abstract Coexistence of similar species can be influenced by the intensity of interspecific interactions, which often depends on the availability of limiting resources. Habitat availability varies strongly with tidal phase in many intertidal ecosystems, potentially affecting interspecific interaction strength, particularly for mobile species. Four closely related species of highly mobile intertidal detritivores (talitrid amphipods Megalorchestia californiana, M. corniculata, M. benedicti, M. minor) inhabit sandy beaches in southern California, where they consume wave‐cast macroalgal wrack originating on coastal reefs. Their coexistence suggests that mechanisms, such as niche separation, are operating to weaken competition among these species. To evaluate this possibility, we explored how tidal phase may mediate temporal and spatial patterns of habitat use among these closely related congeners. We hypothesized that neap tides that reduce intertidal habitat would strengthen temporal separation between species, whereas spatial separation would be greater during spring tides when more habitat is available. We investigated these questions during spring and neap tide phases using (1) comparisons of intertidal distributions of burrowed amphipods and (2) observations of surface activity of amphipods from pitfall traps and mesocosms. We found significant effects of tide phase and species identity on mean intertidal positions and separation of burrowed amphipods. Intertidal distributions of the four species overlapped during neap tide and were significantly separated during spring tide when more intertidal habitat was available. Surface activity patterns differed among species and were more widely separated in time during neap tide than during spring tide. Consequently, the cumulative activity time of all species on neap tides was twice that observed during spring tides. Our findings suggest that mobile intertidal species, like these sympatric talitrid amphipods, can avoid interspecific competition by shifting their activity patterns with tide phase and beach condition. As rising sea levels reduce beach habitat, interspecific competition among these important intertidal consumers may increasingly influence their behavior and coexistence.

Tools for noninvasive detection of bacterial pathogens are needed but are not currently available for clinical use. We have previously shown that para-aminobenzoic acid (PABA) rapidly accumulates in a wide range of pathogenic bacteria, motivating the development of related PET radiotracers. In this study, 11C-PABA PET imaging was used to accurately detect and monitor infections due to pyogenic bacteria in multiple clinically relevant animal models. 11C-PABA PET imaging selectively detected infections in muscle, intervertebral discs, and methicillin-resistant Staphylococcus aureus–infected orthopedic implants. In what we believe to be first-in-human studies in healthy participants, 11C-PABA was safe, well-tolerated, and had a favorable biodistribution, with low background activity in the lungs, muscles, and brain. 11C-PABA has the potential for clinical translation to detect and localize a broad range of bacteria.

The southern cattle fever tick, Rhipicephalus (Boophilus) microplus, is the most economically important ectoparasite of cattle worldwide. A limitation for sustainable control and eradication is the emergence of acaricide resistance among tick populations. Molecular diagnostic tools offer the opportunity to detect resistance rapidly, which can be complemented with confirmatory bioassays with larvae and adult ticks that are more resource and time consuming to generate. Synthetic pyrethroid resistance is one of the most prevalent and well-studied forms of resistance in arthropods, being linked with target site alterations in the sodium ion channel gene. Here, we report research on a novel molecular method to detect mutations in the para-sodium channel gene of R. microplus associated with acaricide resistance that is based on quantitative PCR high-resolution melt (HRM) analysis. Genomic DNA fragments of domains II and III of the para-sodium channel gene were amplified by real-time PCR in the presence of EVA®Green dye to test resistant and susceptible reference ticks from the U.S., Brazil, and Mexico. Larval packet tests with discriminating doses and a modified lethal time analysis were performed to confirm resistance to permethrin, cypermethrin, deltamethrin, and flumethrin in laboratory strains. Tick specimens collected from cattle that were inspected at the United States Port-of-Entry at the Texas-Mexico border were also genotyped. Previously described mutations associated with pyrethroid resistance (T170C, C190A, G184C, and T2134A) were successfully detected by qPCR-HRM in different genotypes and confirmed by sequencing. A novel non-synonymous SNP located at domain III (C2136A) and the G215T mutation in domain II, previously described only in Asian R. microplus and R. australis, were also detected with the HRM and confirmed by sequencing. This technique could be adapted for high-throughput screening, detection, and discovery of allele-specific mutations in cattle tick outbreak populations to inform eradication strategies in the USA. This knowledge could also be applied to integrated control programs in other parts of the world where R. microplus is endemic and where similar SNPs have been identified associated with pyrethroid resistance. This study highlights the existence of several mutations in the para-sodium channel gene in different combinations in field populations of R. microplus from Mexico. Abstract Background The southern cattle fever tick (SCFT), Rhipicephalus (Boophilus) microplus, remains endemic in Puerto Rico. Systematic treatment programmes greatly reduced and even eradicated temporarily this tick from the island. However, a systemic treatment programme that includes integrated management practices for livestock against SCFT remains to be established in the island. We describe a spatially-explicit, individual-based model that simulates climate–livestock–SCFT–landscape interactions. This model was developed as an investigative tool to aid in a research project on integrated management of the SCFT that took place in Puerto Rico between 2014 and 2017. We used the model to assess the efficacy of tick suppression and probability of tick elimination when applying safer acaricides at 3-week intervals to different proportions of a herd of non-lactating dairy cattle. Results Probabilities of eliminating host-seeking larvae from the simulated system decreased from ≈ 1 to ≈ 0 as the percentage of cattle treated decreased from 65 to 45, with elimination probabilities ≈ 1 at higher treatment percentages and ≈ 0 at lower treatment percentages. For treatment percentages between 65% and 45%, a more rapid decline in elimination probabilities was predicted by the version of the model that produced higher densities of host-seeking larvae. Number of weeks after the first acaricide application to elimination of host-seeking larvae was variable among replicate simulations within treatment percentages, with within-treatment variation increasing markedly at treatment percentages ≤ 65. Number of weeks after first application to elimination generally varied between 30 and 40 weeks for those treatment percentages with elimination probabilities ≈ 1. Conclusions Explicit simulation of the spatial and temporal dynamics of off-host (host-seeking) larvae in response to control methods should be an essential element of research that involves the evaluation of integrated SCFT management programmes. This approach could provide the basis to evaluate novel control technologies and to develop protocols for their cost-effective use with other treatment methods.

Abstract Ecosystems across the United States are changing in complex and surprising ways. Ongoing demand for critical ecosystem services requires an understanding of the populations and communities in these ecosystems in the future. This paper represents a synthesis effort of the U.S. National Science Foundation‐funded Long‐Term Ecological Research (LTER) network addressing the core research area of “populations and communities.” The objective of this effort was to show the importance of long‐term data collection and experiments for addressing the hardest questions in scientific ecology that have significant implications for environmental policy and management. Each LTER site developed at least one compelling case study about what their site could look like in 50–100 yr as human and environmental drivers influencing specific ecosystems change. As the case studies were prepared, five themes emerged, and the studies were grouped into papers in this LTER Futures Special Feature addressing state change, connectivity, resilience, time lags, and cascading effects. This paper addresses the “connectivity” theme and has examples from the Phoenix (urban), Niwot Ridge (alpine tundra), McMurdo Dry Valleys (polar desert), Plum Island (coastal), Santa Barbara Coastal (coastal), and Jornada (arid grassland and shrubland) sites. Connectivity has multiple dimensions, ranging from multi‐scalar interactions in space to complex interactions over time that govern the transport of materials and the distribution and movement of organisms. The case studies presented here range widely, showing how land‐use legacies interact with climate to alter the structure and function of arid ecosystems and flows of resources and organisms in Antarctic polar desert, alpine, urban, and coastal marine ecosystems. Long‐term ecological research demonstrates that connectivity can, in some circumstances, sustain valuable ecosystem functions, such as the persistence of foundation species and their associated biodiversity or, it can be an agent of state change, as when it increases wind and water erosion. Increased connectivity due to warming can also lead to species range expansions or contractions and the introduction of undesirable species. Continued long‐term studies are essential for addressing the complexities of connectivity. The diversity of ecosystems within the LTER network is a strong platform for these studies.

Ticks from the genus Rhipicephalus have enormous global economic impact as ectoparasites of cattle. Rhipicephalus microplus and Rhipicephalus annulatus are known to harbor infectious pathogens such as Babesia bovis, Babesia bigemina, and Anaplasma marginale. Having reference quality genomes of these ticks would advance research to identify druggable targets for chemical entities with acaricidal activity and refine anti-tick vaccine approaches. We sequenced and assembled the genomes of R. microplus and R. annulatus, using Pacific Biosciences and HiSeq 4000 technologies on very high molecular weight genomic DNA. We used 22 and 29 SMRT cells on the Pacific Biosciences Sequel for R. microplus and R. annulatus, respectively, and 3 lanes of the Illumina HiSeq 4000 platform for each tick. The PacBio sequence yields for R. microplus and R. annulatus were 21.0 and 27.9 million subreads, respectively, which were assembled with Canu v. 1.7. The final Canu assemblies consisted of 92,167 and 57,796 contigs with an average contig length of 39,249 and 69,055 bp for R. microplus and R. annulatus, respectively. Annotated genome quality was assessed by BUSCO analysis to provide quantitative measures for each assembled genome. Over 82% and 92% of the 1066 member BUSCO gene set was found in the assembled genomes of R. microplus and R. annulatus, respectively. For R. microplus, only 189 of the 1066 BUSCO genes were missing and only 140 were present in a fragmented condition. For R. annulatus, only 75 of the BUSCO genes were missing and only 109 were present in a fragmented condition. The raw sequencing reads and the assembled contigs/scaffolds are archived at the National Center for Biotechnology Information.

The coastal zone provides foraging opportunities for insular populations of terrestrial mammals, allowing for expanded habitat use, increased dietary breadth, and locally higher population densities. We examined the use of sandy beach resources by the threatened island fox (Urocyon littoralis) on the California Channel Islands using scat analysis, surveys of potential prey, beach habitat attributes, and stable isotope analysis. Consumption of beach invertebrates, primarily intertidal talitrid amphipods (Megalorchestia spp.) by island fox varied with abundance of these prey across sites. Distance-based linear modeling revealed that abundance of giant kelp (Macrocystis pyrifera) wrack, rather than beach physical attributes, explained the largest amount of variation in talitrid amphipod abundance and biomass across beaches. δ13C and δ15N values of fox whisker (vibrissae) segments suggested individualism in diet, with generally low δ13C and δ15N values of some foxes consistent with specializing on primarily terrestrial foods, contrasting with the higher isotope values of other individuals that suggested a sustained use of sandy beach resources, the importance of which varied over time. Abundant allochthonous marine resources on beaches, including inputs of giant kelp, may expand habitat use and diet breadth of the island fox, increasing population resilience during declines in terrestrial resources associated with climate variability and long-term climate change.

The emerging sector of offshore kelp aquaculture represents an opportunity to produce biofuel feedstock to help meet growing energy demand. Giant kelp represents an attractive aquaculture crop due to its rapid growth and production, however precision farming over large scales is required to make this crop economically viable. These demands necessitate high frequency monitoring to ensure outplant success, maximum production, and optimum quality of harvested biomass, while the long distance from shore and large necessary scales of production makes in person monitoring impractical. Remote sensing offers a practical monitoring solution and nascent imaging technologies could be leveraged to provide daily products of the kelp canopy and subsurface structures over unprecedented spatial scales. Here, we evaluate the efficacy of remote sensing from satellites and aerial and underwater autonomous vehicles as potential monitoring platforms for offshore kelp aquaculture farms. Decadal-scale analyses of the Southern California Bight showed that high offshore summertime cloud cover restricts the ability of satellite sensors to provide high frequency direct monitoring of these farms. By contrast, daily monitoring of offshore farms using sensors mounted to aerial and underwater drones seems promising. Small Unoccupied Aircraft Systems (sUAS) carrying lightweight optical sensors can provide estimates of canopy area, density, and tissue nitrogen content on the time and space scales necessary for observing changes in this highly dynamic species. Underwater color imagery can be rapidly classified using deep learning models to identify kelp outplants on a longline farm and high acoustic returns of kelp pneumatocysts from side scan sonar imagery signal an ability to monitor the subsurface development of kelp fronds. Current sensing technologies can be used to develop additional machine learning and spectral algorithms to monitor outplant health and canopy macromolecular content, however future developments in vehicle and infrastructure technologies are necessary to reduce costs and transcend operational limitations for continuous deployment in an offshore setting.

Mottled skin pigmentation and solar lentigines from chronic photodamage with aging involve complex interactions between keratinocytes and melanocytes. However, the precise signaling mechanisms that could serve as therapeutic targets are unclear. Herein, we report that expression of nuclear factor erythroid 2–related factor 2 (NRF2), which regulates reduction-oxidation reactions, is altered in solar lentigines and photodamaged skin. Moreover, mottled skin pigmentation in humans could be treated with topical application of the NRF2 inducer sulforaphane (SF). Similarly, UV light–induced pigmentation of WT mouse ear skin could be treated or prevented with SF treatment. Conversely, SF treatment was unable to reduce UV-induced ear skin pigmentation in mice deficient in NRF2 or in mice with keratinocyte-specific conditional deletion of IL-6Rα. Taken together, NRF2 and IL-6Rα signaling are involved in the pathogenesis of UV-induced skin pigmentation, and specific enhancement of NRF2 signaling could represent a potential therapeutic target.

Post-surgical implant-associated spinal infection is a devastating complication commonly caused by Staphylococcus aureus. Biofilm formation is thought to reduce penetration of antibiotics and immune cells, contributing to chronic and difficult-to-treat infections. A rabbit model of a posterior-approach spinal surgery was created, in which bilateral titanium pedicle screws were interconnected by a plate at the level of lumbar vertebra L6 and inoculated with a methicillin-resistant S. aureus (MRSA) bioluminescent strain. In vivo whole-animal bioluminescence imaging (BLI) and ex vivo bacterial cultures demonstrated a peak in bacterial burden by day 14, when wound dehiscence occurred. Structures suggestive of biofilm, visualized by scanning electron microscopy, were evident up to 56 days following infection. Infection-induced inflammation and bone remodeling were also monitored using 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography (PET) and computed tomography (CT). PET imaging signals were noted in the soft tissue and bone surrounding the implanted materials. CT imaging demonstrated marked bone remodeling and a decrease in dense bone at the infection sites. This rabbit model of implant-associated spinal infection provides a valuable preclinical in vivo approach to investigate the pathogenesis of implant-associated spinal infections and to evaluate novel therapeutics.

Abstract Nitrate concentrations routinely fall below levels required to sustain growth of giant kelp (Macrocystis pyrifera) during summer and autumn in the Santa Barbara Channel, yet growth continues. We found urea to be consistently present at concentrations of 0.48–1.82 μM, accounting for greater than 20% of the dissolved fixed nitrogen pool during summer (14% overall). Field experiments indicate direct uptake of urea by giant kelp at a rate of 0.19 μmol N g dw−1 h−1, comparable to rates for ammonium (0.18 μmol N g dw−1 h−1) but lower than for nitrate (0.39 μmol N g dw−1 h−1). Co‐occurring phytoplankton took up nitrate, urea, and ammonium, 2‐, 15‐, and 39‐fold faster than giant kelp; however, the nitrogen uptake advantage of phytoplankton varies by substrate and season. Together, our results suggest that urea is readily used by giant kelp and may help to sustain growth throughout the year.

The growth of marine aquaculture over the 21st century is a promising venture for food security because of its potential to fulfill the seafood deficit in the future. However, to maximize the use of marine space and its resources, the spatial planning of marine aquaculture needs to consider the regimes of climate variability in the oceanic environment, which are characterized by large-amplitude interannual to decadal fluctuations. It is common to see aquaculture spatial planning schemes that do not take variability into consideration. This assumption may be critical for management and for the expansion of marine aquaculture, because projects require investments of capital and need to be profitable to establish and thrive. We analyze the effect of climate variability on the profitability of hypothetical mussel aquaculture systems in the Southern California Bight. Using historical environmental data from 1981 to 2008, we combine mussel production and economics models at different sites along the coast to estimate the Net Present Value as an economic indicator of profitability. We find that productivity of the farms exhibits a strong coherent behavior with marketed decadal fluctuations that are connected to climate of the North Pacific Basin, in particular linked to the phases of the North Pacific Gyre Oscillation (NPGO). This decadal variability has a strong impact on profitability both temporally and spatially, and emerges because of the mussels’ dependence on multiple oceanic environmental variables. Depending on the trend of the decadal regimes in mussel productivity and the location of the farms, these climate fluctuations will affect cost recovery horizon and profitability for a given farm. These results suggest that climate variability should be taken into consideration by managers and investors on decision making to maximize profitability.

Measurements of the status and trends of key indicators for the ocean and marine life are required to inform policy and management in the context of growing human uses of marine resources, coastal development, and climate change. Two synergistic efforts identify specific priority variables for monitoring: Essential Ocean Variables (EOVs) through the Global Ocean Observing System (GOOS), and Essential Biodiversity Variables (EBVs) from the Group on Earth Observations Biodiversity Observation Network (GEO BON) (see Data Sheet 1 in Supplementary Materials for a glossary of acronyms). Both systems support reporting against internationally agreed conventions and treaties. GOOS, established under the auspices of the Intergovernmental Oceanographic Commission (IOC), plays a leading role in coordinating global monitoring of the ocean and in the definition of EOVs. GEO BON is a global biodiversity observation network that coordinates observations to enhance management of the world's biodiversity and promote both the awareness and accounting of ecosystem services. Convergence and agreement between these two efforts are required to streamline existing and new marine observation programs to advance scientific knowledge effectively and to support the sustainable use and management of ocean spaces and resources. In this context, the Marine Biodiversity Observation Network (MBON), a thematic component of GEO BON, is collaborating with GOOS, the Ocean Biogeographic Information System (OBIS), and the Integrated Marine Biosphere Research (IMBeR) project to ensure that EBVs and EOVs are complementary, representing alternative uses of a common set of scientific measurements. This work is informed by the Joint Technical Commission for Oceanography and Marine Meteorology (JCOMM), an intergovernmental body of technical experts that helps international coordination on best practices for observing, data management and services, combined with capacity development expertise. Characterizing biodiversity and understanding its drivers will require incorporation of observations from traditional and molecular taxonomy, animal tagging and tracking efforts, ocean biogeochemistry, and ocean observatory initiatives including the deep ocean and seafloor. The partnership between large-scale ocean observing and product distribution initiatives (MBON, OBIS, JCOMM, and GOOS) is an expedited, effective way to support international policy-level assessments (e.g., the Intergovernmental Science-Policy Platform on Biodiversity and Ecosystem Services or IPBES), along with the implementation of international development goals (e.g., the United Nations Sustainable Development Goals).

Submarine canyons have been shown to positively influence pelagic and benthic biodiversity and ecosystem function. In the eastern Bering Sea, several immense canyons lie under the highly productive “green belt” along the continental slope. Two of these, Pribilof and Zhemchug canyons, are the focus of current conservation interest. We used a maximum entropy modeling approach to evaluate the importance of these two canyons, as well as canyons in general, as habitat for gorgonian (alcyonacean) corals, pennatulacean corals, and sponges, in an area comprising most of the eastern Bering Sea slope and outer shelf. These invertebrates create physical structure that is a preferred habitat for many mobile species, including commercially important fish and invertebrates. We show that Pribilof canyon is a hotspot of structure-forming invertebrate habitat, containing over 50% of estimated high-quality gorgonian habitat and 45% of sponge habitat, despite making up only 1.7% of the total study area. The amount of quality habitat for gorgonians and sponges varied in other canyons, but canyons overall contained more high-quality habitat for structure-forming invertebrates compared to other slope areas. Bottom trawling effort was not well correlated with habitat quality for structure-forming invertebrates, and bottom-contact fishing effort in general, including longlining and trawling, was not particularly concentrated in the canyons examined. These results suggest that if conserving gorgonian coral habitat is a management goal, canyons, particularly Pribilof Canyon, may be a prime location to do this without excessive impact on fisheries.

Cu is an essential trace element but can be highly toxic to aquatic organisms at elevated concentrations. Greater use of CuO engineered nanoparticles (ENPs) may lead to increased concentrations of CuO ENPs in aquatic environments causing potential ecological injury. We examined the toxicity of CuO ENPs to marine mussels and the influence of mussels on the fate and transport of CuO ENPs. We exposed marine mussels to 1, 2, or 3 mg L−1 CuO ENPs for four weeks, and measured clearance rate, rejection, excretion and accumulation of Cu, and mussel shell growth. Mussel clearance rate was 48% less, and growth was 68% less, in mussels exposed to 3 mg L−1 than in control animals. Previous studies show 100% mortality at 1 mg Cu L−1, suggesting that CuO ENPs are much less toxic than ionic Cu, probably due to the slow dissolution rate of the ENPs. Mussels rejected and excreted CuO ENPs in biodeposits containing as much as 110 mg Cu g−1, suggesting the potential for magnification in sediments. Mussels exposed to 3 mg L−1 CuO ENPs accumulated 79.14 ± 12.46 µg Cu g−1 dry weight, which was 60 times more Cu than in control animals. Our results suggest that mussels have the potential to influence the fate and transport of CuO ENPs and potentially cause magnification of CuO ENPs in mussel bed communities, creating a significant source of Cu to marine benthos.

We explored the use of stable nitrogen (N) isotope analysis to assess trophic position of consumers in two marine ecosystems: the kelp forests of southern California and a coral atoll in the tropical Pacific. The δ15N values of consumers in both ecosystems increased from known herbivores (invertebrates and fish) to higher-level consumers (predatory invertebrates and fish). In the absence of data on trophic enrichment in 15N for our study species, we used the oft-cited value of +3.4‰ increase in δ15N value per trophic level and estimates of the δ15N producer baseline value to estimate trophic position. The trophic position of consumers computed using N isotopes compared favorably to published observations of diet. Nitrogen isotope analysis revealed that some of our higher-level fish consumers from rocky reefs (i.e., some rockfish) were feeding largely on invertebrates rather than on fish, as is often assumed. Our analysis also suggests that higher-level consumers on coral reefs may consume more herbivorous prey (i.e., both fishes and invertebrates) than previously reported. Our data support the use of nitrogen isotope values to assess trophic position and, thus, their utility as one metric with which to explore the effects of short- and longer-term natural and human-induced changes on kelp forest and coral reef food webs.

Two distant Antioquian cattle farms where systemic and topical acaricides had previously failed to control infestations by Rhipicephalus ( Boophilus) microplus were studied. An initial in vivo study was conducted using single subcutaneous injections with a long-acting formulation of ivermectin (630 μg/kg). Injections were made at 3-month intervals on animals at each farm to evaluate the therapeutic and persistent efficacy of ivermectin against R. microplus. Body tick counts and reproductive parameters of semi- or fully engorged females ≥5 mm) were assessed at 10-day intervals, and since no negative control group could be included, values were compared against those for day 0. Although there was an overall reduction of 50%-75% in tick numbers that persisted for 30-40 days, it was not significantly different at one of the farms and not enough to afford protection from severe infestations. The engorgement weight and egg mass weight of ticks from treated animals were significantly lower throughout the 50-day posttreatment period. Egg hatch was not significantly reduced posttreatment and remained at levels of 80%-90%. A random selection of 9 out of 28 commercial formulations of ivermectin sold in Colombia were analyzed by High Performance Liquid Chromatography (HPLC). All were within the expected labeled concentration (∓15% deviation) of 1% and 3.15% ivermectin except for one. A popular unregistered injectable widely used in both farms and labeled as “natural pyrethrin”, was found to contain 10.5% ivermectin. An adult immersion test was conducted to evaluate the efficacy of topical acaricides to recommended concentrations of five commercial products and/or their combinations. Efficacy was determined by comparing the reproductive index of each treated group to that of the control group. Cypermethrin (150 ppm) was completely ineffective at both farms. Amitraz (208 ppm) exhibited low and intermediate efficacies of 14% and 56%. The combination of amitraz (100 ppm) and cypermethrin (150 ppm) was less efficacious than the amitraz alone. A generic product based on amitraz + citronella (208 ppm + 10 ppm, respectively) was shown to be less efficacious than the name-brand amitraz product. Products containing the organophosphate chlorpyrifos or trichlorfon exhibited intermediate efficacies of approximately 60% at the Tarso farm. We conclude that at these two locations, there is a high degree of resistance to many of the acaricides available in Colombia and confirm suspicions that ivermectin is no longer able to eliminate tick infestations.

The ticks Rhipicephalus (Boophilus) annulatus and R. (B.) microplus, commonly known as cattle and southern cattle tick, respectively, impede the development and sustainability of livestock industries throughout tropical and other world regions. They affect animal productivity and wellbeing directly through their obligate blood feeding habit and indirectly by serving as vectors of the infectious agents causing bovine babesiosis and anaplasmosis. The monumental scientific discovery of certain arthropod species as vectors of infectious agents is associated with the history of research on bovine babesiosis and R. annulatus. Together, R. microplus and R. annulatus are referred to as cattle fever ticks (CFT). Bovine babesiosis became a regulated foreign animal disease in the United States of America (U.S.) through efforts of the Cattle Fever Tick Eradication Program (CFTEP) established in 1906. The U.S. was declared free of CFT in 1943, with the exception of a permanent quarantine zone in south Texas along the border with Mexico. This achievement contributed greatly to the development and productivity of animal agriculture in the U.S. The permanent quarantine zone buffers CFT incursions from Mexico where both ticks and babesiosis are endemic. Until recently, the elimination of CFT outbreaks relied solely on the use of coumaphos, an organophosphate acaricide, in dipping vats or as a spray to treat livestock, or the vacation of pastures. However, ecological, societal, and economical changes are shifting the paradigm of systematically treating livestock to eradicate CFT. Keeping the U.S. CFT-free is a critical animal health issue affecting the economic stability of livestock and wildlife enterprises. Here, we describe vulnerabilities associated with global change forces challenging the CFTEP. The concept of integrated CFT eradication is discussed in reference to global change.
