Antlers are genetically coded to have bilateral symmetry. However, environmental stressors cause asymmetries between antlers. Previous studies have investigated fluctuating asymmetries on harvested white-tailed deer (Odocoileus virginianus Zimmermann, 1780). Cast antlers provide underutilized metrics that are not available prior to shedding. The objectives of this study were to quantify relative fluctuating asymmetry (RFA) between age groups and identify the best age-specific pre- and post-cast antler metrics to confirm an antler pair. We hypothesized lower RFA values for post-cast measurements than pre-cast measurements due to a lessened chance for damage when atop the head. Additionally, younger individuals were hypothesized to have higher RFA values due to greater susceptibility to environmental stressors. Cast antler pairs from 196 white-tailed deer were collected in Nebraska. We measured 14 available antler metrics per cast antler side classified by age group. The most symmetric measurements between antler sides included pedicle seal area, main beam length, and circumference. Antlers of older deer were consistently more symmetric than younger deer. When combining the top metrics and testing against random antler pairs, we found an 81.9–92.3% match rate for 1.5 and ≥2.5-year-olds, respectively. The Sahel drought of the 1970s and 1980s was among the most devastating of the twentieth century. Its history sheds light on the topic of anthropogenic climate change, which partly caused the drought, as well as its impact on human societies in the modern era. Drought had a shaping influence on the trajectory of Burkina Faso's revolution (1983–87), especially when the state intervened to reduce mass vulnerability in the face of environmental and subsistence crisis. Although the enduring image of the Sahel drought has been one of helpless famine victims and the largesse of Western donors, this interpretation belies both the resiliency of local agrarian peoples and the proactive response of the revolutionary state as it fought to address the drought. Moreover, the widely acclaimed international donor generosity concealed US and French efforts to use aid as a political tool to contain the nascent revolutionary movement in Burkina Faso, which was viewed as a threat to French neocolonial interests in the region. The potential for infectious salmon anemia virus (ISAV)—an internationally regulated pathogen of salmon—to transmit vertically from parent to offspring is currently unclear. While the highly virulent ISAV phenotype known as ISAV-HPRΔ has been observed intra-ova, evidence for vertical transmission of the avirulent ISAV phenotype known as ISAV-HPR0 is lacking. In this study, we identified ISAV-HPR0-infected Atlantic salmon broodstock during spawning within a government research recirculating aquaculture facility using qPCR. Eggs and milt from infected brood were used to initiate 16 unique family dam-sire crosses from which 29–60 fertilized eggs per cross were screened for ISAV using qPCR (limit of detection ~100 virus genome copies/egg). A portion of eggs (~300) from one family cross was hatched and further reared in biosecure containment and periodically screened for ISAV by gill clipping over a 2-year period. ISAV was not detected in any of the 781 eggs screened from 16 family crosses generated by infected brood, nor in 870 gill clips periodically sampled from the single-family cohort raised for 2 years in biocontainment. Based on these findings, we conclude that ISAV-HPR0 has a limited likelihood for vertical parent-to-offspring transmission in cultured Atlantic salmon. [ABSTRACT FROM AUTHOR]

Infectious salmon anemia virus (ISAV) is an important pathogen in global Atlantic salmon (Salmo salar L.) aquaculture. The existence of both non-pathogenic (ISAV-HPR0) and pathogenic (HPR-deleted ISAV) forms of the virus impacts hatchery management. In November 2016, fish tested positive for ISAV-HPR0 at the National Cold Water Marine Aquaculture Center in Maine. A cohort exposed to the fish testing positive for ISAV were lethally sampled over a 7-month period (February–August 2017). No positive samples were detected during this time. Additional testing aimed to determine the extent of the ISAV infections in the facility's fish and to investigate the water sources as potential virus entry points. Fish testing was designed to detect 2% pathogen prevalence with 95% confidence (assuming diagnostic sensitivity of 85%). Over a three-year period, ISAV-HPR0 was detected in spawning fish annually and once in smolts. Repeat testing of smolts from the affected tank three weeks later failed to detect ISAV-HPR0. Over a one-year period of weekly or biweekly evaluation of the incoming water sources, ISAV was never detected. These findings suggest that ISAV-HPR0 infections in monitored hatchery populations can evade detection and that episodes of high prevalence of ISAV-HPR0 associated with spawning can be highly transient. In both cases, conventional surveillance based on recurrent testing of healthy populations may provide only a very limited indication of the HPR0 status. Instead, targeting surveillance to periods of physiological stress, such as spawning and smoltification, and adjusting the sample sizes to account for a related surge in prevalence, should enhance the detection capacity in hatchery settings while also reducing testing costs. Commercial culture of channel catfish (Ictalurus punctatus) occurs in earthen ponds that are characterized by diel swings in dissolved oxygen concentration that can fall to severe levels of hypoxia, which can suppress appetite and lead to suboptimal growth. Given the significance of the hypothalamus in regulating these processes in other fishes, an investigation into the hypothalamus transcriptome was conducted to identify specific genes and expression patterns responding to hypoxia. Channel catfish in normoxic water were compared with catfish subjected to 12 h of hypoxia (20% oxygen saturation; 1.8 mg O2/L; 27°C) followed by 12 h of recovery in normoxia to mimic 24 h in a catfish aquaculture pond. Fish were sampled at 0-, 6-, 12-, 18-, and 24-h timepoints, with the 6- and 12-h samplings occurring during hypoxia. A total of 190 genes were differentially expressed during the experiment, with most occurring during hypoxia and returning to baseline values within 6 h of normoxia. Differentially expressed genes were sorted by function into Gene Ontology biological processes and revealed that most were categorized as "response to hypoxia," "sprouting angiogenesis," and "cellular response to xenobiotic stimulus." The patterns of gene expression reported here suggest that transcriptome responses to hypoxia are broad and quickly reversibly with the onset of normoxia. Although no genes commonly reported to modulate appetite were found to be differentially expressed in this experiment, several candidates were identified for future studies investigating the interplay between hypoxia and appetite in channel catfish, including adm, igfbp1a, igfbp7, and stc2b. NEW & NOTEWORTHY: Channel catfish are an economically important species that experience diel episodic periods of hypoxia that can reduce appetite. This is the first study to investigate their transcriptome from the hypothalamus in a simulated 24-h span in a commercial catfish pond, with 12 h of hypoxia and 12 h of normoxia. The research revealed functional groups of genes relating to hypoxia, angiogenesis, and glycolysis as well as individual target genes possibly involved in appetite regulation. Infectious salmon anemia virus (ISAV; Isavirus salaris) causes an economically important disease of Atlantic salmon (Salmo salar L.). ISA outbreaks have resulted in significant losses of farmed salmon globally, often with a sudden onset. However, 2 phenotypically distinct variants of ISAV exist, each with divergent disease outcomes, associated regulations, and control measures. ISAV-HPRΔ, also known as ISAV-HPR deleted, is responsible for ISA outbreaks; ISAV-HPR0, is avirulent and is not known to cause fish mortality. Current detection methodology requires genetic sequencing of ISAV-positive samples to differentiate phenotypes, which may slow responses to disease management. To increase the speed of phenotypic determinations of ISAV, we developed a new, rapid multiplex RT-qPCR method capable of 1) detecting if a sample contains any form of ISAV, 2) discriminating whether positive samples contain HPRΔ or HPR0, and 3) validating RNA extractions with an internal control, all in a single reaction. Following assay development and optimization, we validated this new multiplex on 31 ISAV strains collected from North America and Europe (28 ISAV-HPRΔ, 3 ISAV-HPR0). Finally, we completed an inter-laboratory comparison of this multiplex qPCR with commercial ISAV testing and found that both methods provided equivalent results for ISAV detection.
