Geologic carbon sequestration requires CO2 injection into the storage formation at high‐injecting pressure. Such high pressure could induce choked flow accompanying huge variations in thermodynamic properties of CO2 at a converging‐diverging (CD) fractures in the storage formation. In this study, high‐velocity CO2 transport through CD fractures was investigated to quantify the effect of fracture morphology on occurrence of both choked flow and shockwave that constrain the mass flow rate of fluid. In addition, variations in thermodynamic CO2 properties including Mach Number (Ma), defined by the ratio of fluid velocity to sound speed, and shock properties were investigated. The morphological characteristics of CD fractures were determined by nine properties, such as throat diameter, throat length, inlet diameter, outlet diameter, throat diameters of two‐connected CD fractures, fracture wall curvature, roughness amplitude, and frequency. As a result, the throat diameter crucially affected choked flow occurrence and maximum Ma. When the inlet and outlet diameters varied, the profiles for Ma variation were consistent in the converging and diverging segments, respectively. In addition, regardless of change in the throat length, the position of maximum Ma was nearly constant with showing the position length ratio of 0.13–0.14. However, roughness of fracture wall significantly influenced the Ma variation and occurrence of shock. In particular, backflows segregated from the main CO2 flow were observed near the wall roughness. Plain Language Summary: Choked flow is a fluid‐mechanical process that restricts the mass flow rate of fluid and it can be occurred when fluid velocity overcomes its sound speed. When CO2 is injected into the targeted geologic formation, choked flow can occur due to highly pressurized flow within the fractures located near the CO2 injection well. Once choked flow occurred, the shock, which accompanies substantial changes in the thermodynamic properties of CO2, follows in the fractures. In the study, choked flow is generated within the converging‐diverging (CD) fracture which serves as a pathway of injected CO2, and its cross‐sectional area decreases and increases again after the "throat." We assumed that the CD fractures have various morphologic properties and analyzed the relationship between the fracture morphology and shock properties. The results highlighted that the throat diameter and fracture roughness mainly affected choked flow occurrence and shock properties. Findings here are applicable to other site‐specific studies, such as volcanoes, geysers, geothermal fields, and underground hydrogen storage sites where highly compressed fluid often migrates through subsurface fractures or any types of geologic conduits. Introduction: The binary PirA/PirB toxin expressed by Vibrio parahaemolyticus (PirABVp) is a virulent complex that causes acute hepatopancreatic necrosis disease (AHPND) in shrimps, affecting the global shrimp farming industry. AHPND is currently diagnosed by detecting pirA and pirB genes by PCR; however, several V. parahaemolyticus strains do not produce the two toxins as proteins. Thus, an immunoassay using antibodies may be the most effective tool for detecting toxin molecules. In this study, we report a sandwich ELISA-based immunoassay for the detection of PirABVp. Methods: We utilized a single-chain variable fragment (scFv) antibody library to select scFvs against the PirA or PirB subunits. Phage display panning rounds were conducted to screen and identify scFv antibodies directed against each recombinant toxin subunit. Selected scFvs were converted into IgGs to develop a sandwich immunoassay to detect recombinant and bacterial PirABVp. Results: Antibodies produced as IgG forms showed sub-nanomolar to nanomolar affinities (KD), and a pair of anti-PirA antibody as a capture and anti-PirB antibody as a detector showed a limit of detection of 201.7 ng/mL for recombinant PirABVp. The developed immunoassay detected PirABVp in the protein lysates of AHPND-causing V. parahaemolyticus (VpAHPND) and showed a significant detectability in moribund or dead shrimp infected with a VpAHPND virulent strain compared to that in non-infected shrimp. Discussion: These results indicate that the developed immunoassay is a reliable method for diagnosing AHPND by detecting PirABVp at the protein level and could be further utilized to accurately determine the virulence of extant or newly identified VpAHPND in the global shrimp culture industry. Blood–brain barrier (BBB) models are important tools for studying CNS drug delivery, brain development, and brain disease. In vitro BBB models have been obtained from animals and immortalized cell lines; however, brain microvascular endothelial cells (BMECs) derived from them have several limitations. Furthermore, obtaining mature brain microvascular endothelial-like cells (BME-like cells) from human pluripotent stem cells (hPSCs) with desirable properties for establishing BBB models has been challenging. Here, we developed an efficient method for differentiating hPSCs into BMECs that are amenable to the development and application of human BBB models. The established conditions provided an environment similar to that occurring during BBB differentiation in the presence of the co-differentiating neural cell population by the modulation of TGF-β and SHH signaling. The developed BME-like cells showed well-organized tight junctions, appropriate expression of nutrient transporters, and polarized efflux transporter activity. In addition, BME-like cells responded to astrocytes, acquiring substantial barrier properties as measured by transendothelial electrical resistance. Moreover, the BME-like cells exhibited an immune quiescent property of BBB endothelial cells by decreasing the expression of adhesion molecules. Staphylococcal enterotoxin B (SEB) is a potent bacterial toxin that causes inflammatory stimulation and toxic shock, thus it is necessary to detect SEB in food and environmental samples. Here, we developed a sensitive immunodetection system using monoclonal antibodies (mAbs). Our study is the first to employ a baculovirus expression vector system (BEVS) to produce recombinant wild-type SEB. BEVS facilitated high-quantity and pure SEB production from suspension-cultured insect cells, and the SEB produced was characterized by mass spectrometry analysis. The SEB was stable at 4 °C for at least 2 years, maintaining its purity, and was further utilized for mouse immunization to generate mAbs. An optimal pair of mAbs non-competitive to SEB was selected for sandwich enzyme-linked immunosorbent assay-based immunodetection. The limit of detection of the immunodetection method was 0.38 ng/mL. Moreover, it displayed higher sensitivity in detecting SEB than commercially available immunodetection kits and retained detectability in various matrices and S. aureus culture supernatants. Thus, the results indicate that BEVS is useful for producing pure recombinant SEB with its natural immunogenic property in high yield, and that the developed immunodetection assay is reliable and sensitive for routine identification of SEB in various samples, including foods. Molt strategies have received relatively little attention in current ornithology, and knowledge concerning the evolution, variability and extent of molt is sparse in many bird species. This is especially true for East Asian Locustella species where assumptions on molt patterns are based on incomplete information. We provide evidence indicating a complex postbreeding molt strategy and variable molt extent among the Pallas's Grasshopper Warbler Locustella certhiola, based on data from six ringing sites situated along its flyway from the breeding grounds to the wintering areas. Detailed study revealed for the first time that in most individuals wing feather molt proceeds from the center both toward the body and the wing‐tip, a molt pattern known as divergent molt (which is rare among Palearctic passerines). In the Russian Far East, where both breeding birds and passage migrants occur, a third of the adult birds were molting in late summer. In Central Siberia, at the northwestern limit of its distribution, adult individuals commenced their primary molt partly divergently and partly with unknown sequence. During migration in Mongolia, only descendantly (i.e., from the body toward the wing‐tip) molting birds were observed, while further south in Korea, Hong Kong, and Thailand the proportion of potential eccentric and divergent feather renewal was not identifiable since the renewed feathers were already fully grown as expected. We found an increase in the mean number of molted primaries during the progress of the autumn migration. Moderate body mass levels and low‐fat and muscle scores were observed in molting adult birds, without any remarkable increase in the later season. According to optimality models, we suggest that an extremely short season of high food abundance in tall grass habitats and a largely overland route allow autumn migration with low fuel loads combined with molt migration in at least a part of the population. This study highlights the importance of further studying molt strategy as well as stopover behavior decisions and the trade‐offs among migratory birds that are now facing a panoply of anthropogenic threats along their flyways. An efficient ultraviolet (UV) polariser based on multi-layered carbon nanotubes (CNTs) which is obtained by set of CNT films attached to substrate in series with matching orientations is demonstrated. The degree of polarisation in the UV region increases with increasing number of CNT layers, reaching ~91% in the CNT polariser with 12 layers. A thin film with photoalignment polymer exposed with more than eight layers CNT polariser induces excellent homogenous liquid crystal (LC) alignment. The fringe-field switching (FFS) LC device with the photoalignment layer shows excellent electro-optics comparable to that with conventional metallic wire-grid polariser (WGP). The rapid decline of a few Emberiza bunting species is increasing conservation concerns, especially in Asia. However, temporal changes in communities and populations of buntings, ones of the most common migratory songbirds in Korea, have not been quantitatively assessed. To understand how the status of buntings has changed over the past 100 years, we collated abundance data from museum collections and bird-banding records between 1910 and 2019. We also used presence–absence data for buntings collected by a nationwide census scheme between 1997 to 2012. Our analysis showed that bunting communities reconstructed from museum-specimen and bird-banding data were not significantly different; however, community composition differed over time. The Meadow (E. cioides), Yellow-throated (E. elegans), Black-faced (E. spodocephala), Rustic (E. rustica) and Chestnut Buntings (E. rutila), which are still common or were once common species, significantly affected the temporal changes in bunting community composition. There were no recent changes in the presence of Rustic and Chestnut Buntings since 1997, but they caused medium-term changes in the bunting community composition, suggesting that there was a sharp to moderate decline in their numbers in the past. The probability of the presence of six bunting species decreased annually, with the most prominent decline in two common breeders, the Meadow (-2.99%/year) and Yellow-throated Buntings (-1.82%/year). This finding suggests that breeding buntings in Korea are under high pressure, as are the migratory buntings. Moreover, despite its recent population decline, the Yellow-throated Bunting was still a major contributor to the community, suggesting that bunting diversity has also been deteriorating while bunting populations are shrinking. Long-term monitoring schemes across their distribution ranges, international cooperation for identifying major threats and key areas of conservation, and law enforcement against illegal hunting and habitat loss are strongly required to mitigate the on-going decline of buntings in Korea and Asia. Migratory birds have evolved diverse migration strategies in response to a variety of factors, but information about the detailed migration patterns of Asian songbirds is not yet available. To understand the short-distance migration pattern of declining Yellow-throated Buntings (Emberiza elegans) in East Asia, we analyzed stable isotopes from the outermost tail feathers (rectrices) of individual buntings collected in Korea and Japan. Temporal changes in feather hydrogen (δ2Hf) and oxygen (δ18Of) isotopic values at stopover islands suggested that northern populations start migration earlier than southern populations, especially in autumn. Latitudinal gradient in δ2Hf values of three wintering populations implied that northern breeders wintered farther north than southern breeders. The migration pattern of this bunting, known as Type II chain migration, was also inferred from hydrogen isotope-based geographic assignments of feather growth origins. Our data demonstrate that stable isotope analysis may help to bridge current knowledge gaps in songbird migration despite coarsely mapped isoscapes and as-of-yet undetermined isotope calibration functions in Asia. In geologic media, fluids exist in gas, liquid, and supercritical phases, generating multiphase and multicomponent systems. As fluids migrating through geologic fractures reach the speed of sound, choked flow can be developed in microfractures. To elucidate such choked flow, thermodynamic analysis and numerical simulations were conducted with CO2, H2O, and CO2‐H2O mixtures at various phases ranging from supercritical to gaseous CO2 and liquid H2O. Compressible CO2, with a relatively low speed of sound (~225 m/s at 31.1 °C and 7.38 MPa), demonstrated significant changes in thermodynamic properties with small pressure and temperature variations. In contrast, H2O, having a relatively high speed of sound (1,524 m/s), showed little thermodynamic variation. For CO2‐H2O mixtures, a small addition of CO2 (or H2O) dramatically reduced the speed of sound relative to those for pure H2O or CO2. For an idealized converging‐diverging microfracture with CO2 flow, choked flow and a shock wave were generated as outlet pressure was decreased to less than 6.8 MPa. The H2O flow did not generate choked flow at any outlet pressures. For CO2‐H2O mixtures, choked flow was generated when the CO2 void fraction was greater than 0.7 with an outlet pressure of 6.5 MPa, indicating that presence of H2O inhibited occurrence of choked flow. Choked flow and shock waves can occur in various geologic environments including geologic CO2 sequestration, geothermal energy development, geysers, and volcano eruptions. Graphical abstract Abstract Adipogenesis involved in hypertrophy and hyperplasia of adipocytes is responsible for expanding the mass of adipose tissues in obese individuals. Peroxisome proliferator-activated receptor γ (PPARγ) and CCAAT/enhancer-binding protein α (C/EBPα) are two principal transcription factors induced by delicate signaling pathways, including signal transducer and activator of transcription 5 (STAT5), in adipogenesis. Here, we demonstrated a novel role of ginkgetin, a biflavone from Ginkgo biloba leaves, as a STAT5 inhibitor that blocks the differentiation of preadipocytes into adipocytes. During the differentiation of 3T3-L1 cells, ginkgetin treatment during the first 2 days markedly inhibited the formation of lipid-bearing adipocytes. PPARγ and C/EBPα expression was decreased in 3T3-L1 cells during adipogenesis following ginkgetin treatment, whereas no change was observed in C/EBPβ or C/EBPδ expression. Inhibition of PPARγ and C/EBPα expression by ginkgetin occurred through the prevention of STAT5 activation during the initiation phase of adipogenesis. In addition, ginkgetin-mediated the inhibition of adipogenesis was recapitulated in the differentiation of primary preadipocytes. Lastly, we confirmed the inhibitory effects of ginkgetin on the hypertrophy of white adipose tissues from high-fat diet-fed mice. Abstract: The room‐temperature tensile strength, toughness, and high‐temperature creep strength of 2000, 6000, and 7000 series aluminum alloys can be improved significantly by dispersing up to 1 wt% carbon nanotubes (CNTs) into the alloys without sacrificing tensile ductility, electrical conductivity, or thermal conductivity. CNTs act like forest dislocations, except mobile dislocations cannot annihilate with them. Dislocations cannot climb over 1D CNTs unlike 0D dispersoids/precipitates. Also, unlike 2D grain boundaries, even if some debonding happens along 1D CNT/alloy interface, it will be less damaging because fracture intrinsically favors 2D percolating flaws. Good intragranular dispersion of these 1D strengtheners is critical for comprehensive enhancement of composite properties, which entails change of wetting properties and encapsulation of CNTs inside Al grains via surface diffusion‐driven cold welding. Oxidative stress activates macroautophagy/autophagy and contributes to atherogenesis via lipophagic flux, a form of lipid removal by autophagy. However, it is not known exactly how endogenous antioxidant enzymes are involved in lipophagic flux. Here, we demonstrate that the antioxidant PRDX1 (peroxiredoxin 1) has a crucial role in the maintenance of lipophagic flux in macrophages. PRDX1 is more highly expressed than other antioxidant enzymes in monocytes and macrophages. We determined that Prdx1 deficiency induced excessive oxidative stress and impaired maintenance of autophagic flux in macrophages. Prdx1-deficient macrophages had higher intracellular cholesterol mass and lower cholesterol efflux compared with wild type. This perturbation in cholesterol homeostasis was due to impaired lipophagic cholesterol hydrolysis caused by excessive oxidative stress, resulting in the inhibition of free cholesterol formation and the reduction of NR1H3 (nuclear receptor subfamily 1, group H, member 3) activity. Notably, impairment of both lipophagic flux and cholesterol efflux was restored by the 2-Cys PRDX-mimics ebselen and gliotoxin. Consistent with this observation, apoe −/− mice transplanted with bone marrow from prdx1−/−apoe−/− mice had increased plaque formation compared with apoe−/− BM-transplanted recipients. This study reveals that PRDX1 is crucial to regulating lipophagic flux and maintaining macrophage cholesterol homeostasis against oxidative stress. Although carbon nanotubes (CNTs) are used to reinforce aluminum alloys, the underlying strengthening mechanisms have not yet been clarified. Here, we focus on elucidating several of these mechanisms including load transfer, generation of dislocations from thermal mismatch, and the Orowan looping system to explain the enhanced mechanical properties of CNT-reinforced Al. The sample preparation procedure involves mechanically pulverizing Al in paraffin oil mixed with CNTs dispersed in dichloroethane. The mixture was then sintered and melt-blended to produce solid samples. The formation of covalent bonds (Al 4 C 3 ) between Al and CNT walls was confirmed by X-ray diffraction, Raman spectroscopy, and transmission electron microscope measurements. The yield strength and tensile strength were improved by 60% and 23%, respectively, with the addition of 0.2 wt% CNTs. Abstract: Objective: Blocking agents targeting cell adhesion molecules have been developed to prevent cardiovascular diseases such as atherosclerosis, whereas relatively little attention has been paid to the therapeutic potential of vascular cell adhesion molecule (VCAM)-1 as an inflammatory disease target. Two novel, fully human antibodies, H6 and 7H, against human VCAM-1 (hVCAM-1) were developed and tested to validate the hypothesis that blocking VCAM-1 ameliorates atherosclerosis in apolipoprotein E-deficient (ApoE−/−) mice. Methods and results: Treatment with H6 or 7H effectively inhibited VCAM-1 adhesion to inflammatory cells, and reduced RhoA activation and the production of reactive oxygen species in human umbilical cord vascular endothelial cells. As 7H showed binding affinity to both murine VCAM-1 (mVCAM-1) and hVCAM-1, the therapeutic effects of 7H in ApoE−/− mice were tested. After confirming specific in vivo binding activity of 7H to mVCAM-1, we showed that administering 7H resulted in significantly ameliorated plaque formation compared to administering a control antibody in ApoE−/− mice fed a Western diet for 12 weeks. Also, 7H treatment significantly reduced infiltration of CD45+ cells into plaques and reduced inflammation and improved plaque stability. Conclusion: These results indicate that the anti-VCAM-1 antibody attenuates atherosclerosis in ApoE−/− mice, improves plaque inflammation and stability as well as inhibiting the adhesion of inflammatory cell, and suggest that blocking VCAM-1 with a monoclonal antibody may be an effective means of anti-atherosclerotic therapy. The effect of the timing of spring migration on reproductive success differs between the sexes. As a consequence, various sex-specific tactics relating to the timing of migration have evolved in migratory avian groups. Various hypotheses have been proposed to explain differential migration to breeding or wintering grounds, and inter- and intrasexual size differences are often considered one of the proximate mechanisms. We investigated arrival patterns in the spring by individuals of each sex, sexual size dimorphism and related morphological variables, and the relationship between size variation and arrival date in five bunting species that passed through an East Asian migratory flyway stopover site in 2006-08. Males of all the study species arrived before females, and significant sexual dimorphism was observed. Several morphological characters, including total length, wing-length and tail-length, contributed to the size variation. Although larger males arrived earlier, there was no relationship between arrival date and size in females. Our study confirmed that East Asian buntings display a discriminated protandrous migration pattern at the stopover site as well as at the breeding grounds. This is consistent with the view that larger body size in males is favoured due to its association with early arrival to help ensure access to the best resources and hence enhanced mating success. Abstract: The effects of carbon tetrachloride (CCl4) treatment on acute liver damage in knock out (heat shock proteins—HSP70−/−) mice and wild-type (C57BL/6) mice were examined. Acute liver injury was induced by a single intraperitoneal injection of 0.3ML/kg CCl4 in olive oil. Mice were sacrificed at 12, 24, 48 and 72h after treatment. To assess hepatotoxicity, alanine transaminase, neutrophil infiltration and degree of necrosis were measured. Western blot analysis was employed for heat shock proteins. The result revealed that HSP70−/− mice showed higher alanine transaminase levels and a more severe degree of neutrophilic infiltration and necrosis than those of wild-type mice. Furthermore, HSP70−/− mice recovered more slowly from CCl4 treatment. In HSP70−/− mice, HSP47 was overexpressed. Therefore, HSP70−/− mice could be an adequate model of acute liver toxicity study. Increased oxidative stress (OS) is considered a common etiology in the pathogenesis of cardiovascular disease (CVD). Therefore, the precise regulation of reactive oxygen species (ROS) in cardiovascular cells is essential to maintain normal physiological functions. Numerous regulators of cellular homeostasis are reportedly influenced by ROS. Hydrogen peroxide (H2O2), as an endogenous ROS in aerobic cells, is a toxic substance that can induce OS. However, many studies conducted over the past two decades have provided substantial evidence that H2O2 acts as a diffusible intracellular signaling messenger. Antioxidant enzymes, including superoxide dismutases, catalase, glutathione peroxidases, and peroxiredoxins (Prdxs), maintain the balance of ROS levels against augmentation of ROS production during the pathogenesis of CVD. Especially, Prdxs are regulatory sensors of transduced intracellular signals. The intracellular abundance of Prdxs that specifically react with H2O2 act as regulatory proteins. To study the intraspecific variation of the grey-capped greenfinch Chloris sinica (Passeriformes: Fringillidae), we sequenced complete mitochondrial (mt) genome of the C. sinica ssp. prevalent in Ulleung Island, Republic of Korea. The full length of the genome is 16,812 bp, containing 37 genes (2 rRNAs, 13 proteins, and 22 tRNAs) with a putative control region (D-loop). A total of 98 single nucleotide polymorphisms (SNPs) in the full mt genome were retained for Ulleung Island population and these SNPs were greater than those of inland population compared to the reference China subspecies. The analysis of the SNPs revealed 18 SNPs for ND4 gene, with a dominant haplotype shared by Ulleung population compared to the reference China population. The phylogenetic analysis of the C. sinica subspecies showed that they are monophyletic, however, there is clear phylogenetic separation between China and Korea subspecies with strong support (100% bootstrap). We sequenced the complete mitochondrial (mt) genome of Gavia arctica. The circular mt genome is 17,065 bp long, consisting of 37 genes (13 proteins, 22 transfer RNAs, and two ribosomal RNAs) and a control region. Phylogenetic analysis based on the full mt genome sequences confirmed that the genus Gavia is a monophyletic group, containing the G. stellata, G. arctica, and G. pacifica. We sequenced the complete mitochondrial (mt) genome of Chloris sinica ussuriensis. The circular mt genome is 16,813 bp long and encodes 13 proteins, 22 transfer RNAs, and 2 ribosomal RNAs. Phylogenetic analysis based on full mt genome sequences confirmed that the C. s. ussuriensis is monophyletic group of the Chloris sinica. The complete mitochondrial genome of C. s. ussuriensis can provide a valuable data for resolving geographical distribution of evolutionary subdivision within the C. sinica species in East Asia.
