Surgery, chemotherapy, and radiation therapy are all forms of cancer treatment, as well as more recent methods including interventional radiology and immunotherapy. In this study, we synthesize a novel chitosan (CH) nanocomplex (NC)-based polysaccharide Krestin (PSK) for drug delivery. This technique was used to develop PSK@CH@NC. According to the study, PSK@CH@NC had a particle size of around 500 nm, slight polydispersity as observed under a scanning electron microscope, and a strong positive surface charge of 18 mV. Investigation into the in vitro growth inhibition of the MCF-7 cell line after treatment with CH, PSK, and PSK@CH@NC was followed by morphological changes. Compared to other treatment groups, PSK@CH@NC therapy dramatically reduced the fraction of apoptotic cells, cancer cell survival, and proliferation. Fluorescence analysis was used to examine how PSK@CH@NC affected the distribution of cell cycle phases. This study also shows that a promising foundation for creating cancer nanomedicine can be established by employing new polysaccharides.

Mosquitoes are important vectors, which transmit many communicable diseases throughout the world. Synthetic insecticides, such as organophosphate and pyrethroids, are commonly used for their control in the vector control program. Insecticidal compounds from natural sources, notably from plants and synthesized nanoparticles (NPs) are promising tools for managing such vectors. Hence, the study aimed to analyze the insecticidal potentiality of leaf extract of Cipadessa baccifera and synthesized ZnNPs against three major mosquito vectors. The results recorded from UV-Vis spectroscopy show the peak absorption spectrum at 420 nm. In FTIR, the maximum peak value is 562.85 cm−1 assigned to the N–H group (amide group). The EDAX analysis shows a peak around 63.29, which confirms the binding intensity of selenium. In the scanning electron microscopy analysis, the synthesized ZnNPs sizes were ranging from 49.21 to 65.43 nm. The synthesized ZnNPs produced high mortality against Culex quinquefasciatus LC50 = 0.049653 mg·mL−1; LC90 = 0.9842 mg·mL−1), Anopheles stephensi (LC50 = 0.053421 mg·mL−1 and LC90 = 0.027761 mg·mL−1), and Aedes aegypti LC50 = 0.55214 mg·mL−1 and LC90 = 0.7456 mg·mL−1). These results suggest that the C. baccifera leaf extract-mediated biosynthesis of ZnNPs has the potential to be used as an ideal eco-friendly approach toward the control of mosquito vectors at early stages.

Lung cancer, particularly non-small cell lung cancer (NSCLC), is a leading cause of cancer-related deaths worldwide. This study investigates the molecular mechanisms behind the anti-cancer effects of the tropical desert plant Retama raetam (R. raetam) on the A549 NSCLC cell line. The research examined R. raetam's anti-proliferative effects, cytotoxicity, apoptosis, reactive oxygen species (ROS) generation, mitochondrial membrane potential, and cell morphology in NSCLC A549 and L-132 cells. In addition, the influence of R. raetam on DNA fragmentation, apoptotic signaling, and PI3K/Akt pathways for its anti-cancer mechanism was examined. Our results indicated that R. raetam's effects were dose- and time-dependent to exhibit anti-proliferative effects on A549 cells. R. raetam treatment promoted apoptotic cell death cycle arrest, increased apoptotic cells, depolarized the mitochondrial membrane, and induced morphological alterations in cells and nuclei. It also inhibited A549 cell migration (P < 0.05), colonization, and invasiveness. Moreover, the study demonstrated that R. raetam treatment resulted in the upregulation of Bax expression, downregulation of Bcl-2 expression, and apoptotic fragmented DNA in A549 cells. The top five bioactive compounds derived from R. raetam exhibited molecular interactions that inhibit PIK3CA and AKT1. This inhibition leads to an increased frequency of apoptosis and subsequent death of cancer cells. Additionally, R. raetam extract induced an increase in ROS formation and cytochrome c levels, indicating that its toxic effects on A549 cells involve both ROS-dependent cytotoxicity through the disruption of mitochondrial transmembrane potential ΔΨm and ROS-independent cell cycle arrest through downregulation BCL-2, PARP, E-Cadherin, PI3K, and Akt expressions pathways. In this research, cell-free extracts from magnesite mine-isolated actinobacterial strain (M10A62) were used to produce silver sulfide nanoparticles (Ag2SNPs). Streptomyces minutiscleroticus JX905302, actinobacteria capable of producing Ag2SNPs, was used to synthesize Ag2NPs. The UV–vis range was used to confirm the biosynthesized Ag2NPs; Fourier transform infrared spectroscopy (FT-IR), atomic force microscopy (AFM), transmission electron microscopy (TEM), X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDAX), and dynamic light scattering analysis were employed to characterize them further. Surface resonance plasma (SRP) for Ag2SNPs was obtained at 355 nm using UV–visible spectroscopy; FT-IR detected bimolecular and eventually microbial-reduced Ag2SNPs from S. minutiscleroticus culture extract. Furthermore, AFM and TEM analysis confirms that the synthesized Ag2SNPs were spherical in shape. Dynamic light scattering revealed a negatively charged Ag2NPs surface with a diameter of 10 nm. The XRD spectrum showed the crystalline nature of the obtained particles. EDAX revealed a pure crystalline nature, and a significant silver particle signal confirms the presence of metallic silver and sulfide nanoparticles together with the signals of Cu and C atoms. After 40 and 48 h of treatment at 150–200 µg·ml−1, Ag2SNPs produced the highest mortality in Spodoptera litura, H. armigera, Aedes aegypti, and Culex quinquefasciatus larvae. Hence, the biosynthesized Ag2SNPs may be useful for potential pest control in integrated pest management and vector control program as a safer, cost-effective, selective, and environmentally friendly approaches.

Parkinson’s disease (PD) is a gradual deterioration of dopaminergic neurons, leading to motor impairments. Social isolation (SI), a recognized stressor, has recently gained attention as a potential influencing factor in the progress of neurodegenerative illnesses. We aimed to investigate the intricate relationship between SI and PD progression, both independently and in the presence of manganese chloride (MnCl2), while evaluating the punicalagin (PUN) therapeutic effects, a natural compound established for its cytoprotective, anti-inflammatory, and anti-apoptotic activities. In this five-week experiment, seven groups of male albino rats were organized: G1 (normal control), G2 (SI), G3 (MnCl2), G4 (SI + MnCl2), G5 (SI + PUN), G6 (MnCl2 + PUN), and G7 (SI + PUN + MnCl2). The results revealed significant changes in behavior, biochemistry, and histopathology in rats exposed to SI and/or MnCl2, with the most pronounced effects detected in the SI rats concurrently exposed to MnCl2. These effects were associated with augmented oxidative stress biomarkers and reduced antioxidant activity of the Nrf2/HO-1 pathway. Additionally, inflammatory pathways (HMGB1/RAGE/TLR4/NF-ᴋB/NLRP3/Caspase-1 and JAK-2/STAT-3) were upregulated, while dysregulation of signaling pathways (PI3K/AKT/GSK-3β/CREB), sustained endoplasmic reticulum stress by activation PERK/CHOP/Bcl-2, and impaired autophagy (AMPK/SIRT-1/Beclin-1 axis) were observed. Apoptosis induction and a decrease in monoamine levels were also noted. Remarkably, treatment with PUN effectively alleviated behaviour, histopathological changes, and biochemical alterations induced by SI and/or MnCl2. These findings emphasize the role of SI in PD progress and propose PUN as a potential therapeutic intervention to mitigate PD. PUN’s mechanisms of action involve modulation of pathways such as HMGB1/RAGE/TLR4/NF-ᴋB/NLRP3/Caspase-1, JAK-2/STAT-3, PI3K/AKT/GSK-3β/CREB, AMPK/SIRT-1, Nrf2/HO-1, and PERK/CHOP/Bcl-2.

Mosquito-borne diseases are causing serious damage to public health worldwide, and control of these deadly mosquito vectors is a major thrust area for epidemiologists and public health workers. Therefore, the present research reports an eco-friendly solution with multipotency of silver nanoparticle fabricated from shrimp shell biowaste in controlling mosquitoes and bacterial pathogens. The biofabricated chitosan silver nanoparticles (Cs-AgNPs) were confirmed by UV-visible spectroscopy, transmission electron microscopy (TEM), Fourier transform infrared spectroscopy, energy dispersive X-ray spectroscopy, X-ray powder diffraction and zeta potential analysis. The TEM studies showed that the obtained Cs-AgNPs were mostly spherical in shape. Low doses of chitosan and Cs-AgNPs showed high mosquitocidal properties against both larvae and adult of Anopheles stephensi, Aedes aegypti, and Culex quinquefasciatus. The LC50 (lethal concentration 50%) of Cs-AgNPs was 10.240 ppm (fourth instar larvae) and 9.671 ppm (adult) for An. stephensi; 11.349 ppm (fourth instar) and 12.015 ppm (adult) for Ae. aegypti and 12.426 ppm (fourth instar) and 12.965 ppm (adult) for Cx. quinquefasciatus. The concerning part of antibacterial studies showed that Cs-AgNP had significant inhibition on tested bacterial pathogens. Overall, this study shows that chitosan extracted from the shrimp shell wastes can be used as a potential source for controlling major mosquito vectors.

The exponential growth of obesity rates is a pressing issue, as it is now firmly established as a primary driver behind the development of metabolic disorders. Natural products are crucial in drug discovery, prompting an increasing need for further research on bioactive compounds to understand molecular and pharmacological mechanisms and expand available clinical treatments for various diseases. We set out to investigate the therapeutic potential of Retama raetam (RR) extract, a natural compound, in alleviating testicular degeneration and improving sperm quality and quantity in high-fat diet (HFD)-induced type 2 diabetes mellitus (T2DM) and streptozotocin (STZ)-induced type 2 diabetes mellitus. An eight-week high-fat diet (HFD) was administered to type 2 diabetic rats, and then a modest dose of 35 mg/kg of STZ was injected intraperitoneally. The experiment planned to assess the influence of RR extract on the steroidogenesis pathway. For twelve weeks, the rats were given medications orally. The testicular degeneration caused by HFD/STZ is shown by decreasing the accessory sexual glands while reducing the quality of sperm characters and testosterone levels with significantly increased pad fat and leptin levels. Furthermore, administration of RR extract effectively counteracted HFD/STZ-induced oxidation through enhancing the antioxidant status of the testicular tissue through significantly increased SOD and GSH accompanied by decreasing MDA. HFD/STZ rats showed decreased expression of CYP17, STAR, 3βHSD, and BCL2 genes, along with increased BAX gene expression. However, treatment with RR in HFD/STZ rats led to decreased BAX expression and increased expression of CYP17, STAR, 3βHSD, and BCL2 genes, indicating a restoration of gene expression values similar to the control group. The binding site of Bax showed a strong affinity for several bioactive compounds found in R. raetam. The findings from this study suggest that Retama raetam (RR) has the potential as a therapeutic adjunct for managing complications related to type 2 diabetes mellitus (T2DM). Storage grain infestation is a critical problem, with insect pests and rodents causing economic losses and reductions in the quality of food grains and food products. It has been assessed as affecting 9% of goods in developed countries and more than 20% of goods in developing countries. The chemical insecticides used for storing products protect against insect pests, but introduce hazards associated with handling insecticides and the residues on food, which threaten nontarget organisms, and so represent a serious problem to the environment. Biorational methods are low risk, are obtained from animals, plants, bacteria and some minerals, and are segregated into microbial, plant‐associated protectant and biochemical substances. This method encompasses temperature management, modified atmospheric storage, ionizing irradiation, the mixing of inert materials, pheromone traps, botanicals, insect growth regulators, entomopathogens and host plant resistance. Biorational methods have emerged as alternative or supplemental pest control methods. Biopesticides would help in preventing the use of large quantities of pesticides across the globe, thus providing residue‐free food and a safe environment for living. Further investigation is needed to integrate these elements to enhance the efficacy, minimize the losses and diminish the reliance on pesticides for the preservation of stored commodities. The present study was designed to green synthesize titanium dioxide nanoparticles (G-TiO2 NPs) using Salacia reticulata leaf extract as a reducing and capping agent to assess antidiabetic, anti-inflammatory, and antibacterial effects as well as toxicity evaluation in zebrafish. Besides, zebrafish embryos were employed to study the effect of G-TiO2 NPs on embryonic development. Zebrafish embryos were treated with TiO2 as well as G-TiO2 NPs at four different concentrations, i.e., 25, 50, 100, and 200 µg/ml for 24–96-hour post-fertilization (hpf). The SEM analysis of G-TiO2 NPs confirmed that the size was in the range of 32–46 nm and characterized by EDX, X-ray diffraction (XRD), FTIR, UV–vis spectra. During 24–96-hour post-fertilization (hpf), the results showed that 25–100 µg/ml of TiO2 and G-TiO2 NP instigated developmental acute toxicity in these embryos, causing mortality, hatching delay, and malformation. TiO2 and G-TiO2 NPs exposure induced axis bent, tail bent, spinal cord curvature, yolk-sac, and pericardial edema. Exposure of larvae to the highest concentrations of 200 μg/ml TiO2 and G-TiO2 NPs caused maximum mortality at all time points and reached 70% and 50%, respectively, at 96 hpf. Besides, both TiO2 and G-TiO2 NP revealed antidiabetic and anti-inflammatory effects in vitro. In addition, G-TiO2 NPs exhibited antibacterial effects. Taken together, this study provided a valuable insight into the synthesis of TiO2 NPs using green methods and the synthesized G-TiO2 NPs possess moderate toxicity and potent antidiabetic, anti-inflammatory and antibacterial activities. The biogenic manufacture of nanoparticles utilising endophytic fungus is an eco-friendly, cost-effective, and secure alternative to constructing chemical methods. The prime focus of the study was to fabricate ZnONPs using the biomass filtrate of endophytic Xylaria arbuscula isolated from Blumea axillaris Linn. and to evaluate their biological properties. The characterisation of the biosynthesized ZnO-NPs was done utilising both spectroscopic and microscopic methods. The bioinspired NPs showed a surface plasmon peak at 370nm; SEM and TEM micrographs illustrated the hexagonal organisation; XRD spectra proved the crystalline phase as hexagonal wurtzite; EDX analysis confirmed the presence of zinc and oxygen atoms; and the zeta potential analysis proved the stability of ZnONPs. In addition, they also demonstrated significant concentration-dependent inhibition of antimicrobial, antioxidant, antiinflammatory, and antidiabetic potential in comparison with the reference drugs. In vitro cytotoxicity and wound healing potential of ZnONPs were examined in L929 cell lines, illustrating that they accelerated the wound healing process by roughly 95.37 ± 1.12% after a 24-h exposure to ZnONPs. The photocatalytic activity of the ZnONPs was examined by degrading the methylene blue dye under solar irradiation. In conclusion, our outcomes showed that mycosynthesized ZnONPs possessed potent bioactivity and could be an excellent choice for biomedical applications. [ABSTRACT FROM AUTHOR]

Mosquitoes (Diptera: Culicidae) are considered to be huge threat among millions of peoples, animals, and other living organisms in the world. Most of the vector borne diseases such as malaria, filariasis, dengue hemorrhagic fever, chikungunya, Japanese encephalitis etc., created huge impact on humans in all over the world. Vector diseases in Kingdom of Saudi Arabia are increasing day by day and their control measures taken through the government sectors for eradicating the vectors helps in controlling the diseases but still more approaches to be implemented or assimilated. Most of the synthetic or chemical based insecticides to control mosquitoes developed resistance among their communities even though they showed their potential in controlling the vector in initial days. Botanical insecticides from plant‐based origin such as active compounds, essential oils, green synthesized nanomaterials, and microbial secondary metabolites helps more efficiency in controlling vectors. Mode of action against vectors differs based on its persisting active ingredients, such as larvicidal, pupicidal, adulticidal, oviposition, morphological changes etc. Even though number of research works has been carried out against mosquito species, there is only limited number of studies undergone against mosquito vectors from Saudi Arabia origin. Hence this review will give us the current knowledge on the effectiveness of botanical insecticides against major mosquito vectors from Saudi Arabia.
