This case report concerns a 48-year-old man with a history of ischemic stroke at the age of 41 who reported cardiac hypertrophy, registered in his twenties when explained by increased physical activity. Family history was positive for a mother with permanent atrial fibrillation from her mid-thirties. At the age of 44, he had a first episode of persistent atrial fibrillation, accompanied by left atrial thrombosis while on a direct oral anticoagulant. He presented at our clinic at the age of 45 with another episode of persistent atrial fibrillation and decompensated heart failure. Echocardiography revealed a dilated left atrium, reduced left ventricular ejection fraction, and an asymmetric left ventricular hypertrophy. Cardiac magnetic resonance was positive for a cardiomyopathy with diffuse fibrosis, while slow-flow phenomenon was present on coronary angiography. Genetic testing by whole-exome sequencing revealed three variants in the patient, c.309C > A, p.His103Gln in the ACTC1 gene, c.116T > G, p.Leu39Ter in the PLN gene, and c.5827C > T, p.His1943Tyr in the SCN5A gene, the first two associated with hypertrophic cardiomyopathy and the latter possibly with familial atrial fibrillation. This case illustrates the need for advanced diagnostics in unexplained left ventricular hypertrophy, as hypertrophic cardiomyopathy is often overlooked, leading to potentially debilitating health consequences. The consumption of electrical energy for the community every year has increased including the electricity consumption of PLN UP3 Cengkareng customers. Therefore, PLN UP3 Cengkareng must supply electricity to customers in all categories such as Social Category, Household Category, Business Category, Industry Category and Government Category. With customer needs that continue to increase, it is necessary to forecast future electricity needs, so that PLN UP3 Cengkareng can provide the required electrical power. For this reason, it is necessary to predict the electricity demand. This research was conducted to forecast the electricity demand of UP3 Cengkareng by using the Deep Learning Model Long Short-Term Memory (LSTM). The data set used in this study was taken from the PLN UP3 Cengkareng information system, for 10 years, the period from 2012 to 2021. The data used is divided into 2 categories, namely 70% training data and 30% testing data. The results obtained from this prediction are 96,689, with an average neuron value of 32 and an epoch value of 10.

Este artigo situa-se na interface entre a Lexicografia (PORTO DAPENA, 2002; HARTMANN, 2016), a Dialetologia (CARDOSO, 2010; CHAMBERS; THUDGILL, 1994) e a Linguística Computacional (HABERT, 2004; PÉREZ HERNÁNDEZ; MORENO ORTIZ, 2009; HAUSSER, 2014; KURDI, 2016). Objetiva-se discutir a proposta de construção de um banco de dados em XML (Extensible Markup Language), explorando os resultados obtidos com o PLN (Processamento Automático de Linguagem Natural). O arquivo XML também se fundamenta em parâmetros da Lexicografia Dialetal (EZQUERRA, 1997; NAVARRO CARRASCO, 1993) e está sendo alimentado com dados dialetais oriundos do Projeto Atlas Linguístico do Brasil (ALiB) documentados na região Norte do país. Para tanto, utilizou-se como editor de texto o software jEdit e, para gerenciar o banco de dados, o programa BaseX. A extração das informações linguísticas foi realizada, no BaseX, a partir de uma amostra de dados e com o auxílio de expressões X-Query. Assim, foram executadas as seguintes manipulações de dados: i) localização de uma unidade lexical específica; ii) visualização de qualquer dado da microestrutura filtrada pelas variáveis sexo, idade, escolaridade e localidade; iii) seleção de informações a partir de uma das 14 áreas semânticas em que as questões do questionário semântico-lexical do ALiB foram organizadas. Em síntese, entende-se que a construção do banco de dados em XML confere agilidade em relação à extração de informações e compatibilidade dos dados para executar interfaces com outras aplicações como, por exemplo, a elaboração de um produto lexicográfico a ser publicado em suporte on-line.

Este artículo presenta los métodos y herramientas empleados en el tratamiento de textos teatrales del Siglo de Oro para optimizarlos para el análisis digital. El proceso parte de textos obtenidos en archivos de muy diversos fortmatos y termina produciendo un archivo CSV apto para análisis de lectura distante. Los archivos de origen requieren ser limpiados y preparados de diferentes maneras según sus características propias de forma semimanual. Una vez es posible identificar las unidades textuales, se ordena automáticamente la obra en formato tabular, de forma que represente la estructura dramática. Después se escanden los versos y se añaden los resultados a las columnas correspondientes. Se han diseñado herramientas para el propósito, de manera que las tareas se llevan a cabo automáticamente una vez los textos de origen están preparados.

The Padang branch of the State Electricity Company (PLN) is a BUMN responsible for the electricity aspect. Complaints about various electricity problems are difficult for customers, so it is necessary to have a system that can be used as a place for protests and increase PLN's loyalty to customers. The Android-based Claim System is a new application developed to be used by customers as a forum for complaints about electrical problems to the Padang branch of PLN. The Claim System is made into two parts: a Website-based System for PLN and an Android-based Claim Application for customers. Application development using the OOP concept uses UML (Unified Modeling Language) diagrams with the PHP MySql and Android programming languages. The test results were carried out using black box testing with the relevant results. The data of 35 respondents from the assessment questionnaire on the claims system obtained results with an Excellent rating of 78%, a Good rating of 21%, and a Bad rating of 2%. From the analysis results, the Claim System supported by this Android-based Client application can help customers complain about electrical problems quickly and easily.

The current high energy consumption can cause scarcity as Indonesia's population growth has increased quite significantly. Alternative energy is needed to anticipate energy problems in the future. Solar energy is one of the options given the most significant potential for energy produced in Indonesia. This research aims to see the strategies that PLN can do to increase the use of Rooftop PV in Indonesia based on the results of the analysis that has been done. The approach to this research is a qualitative approach with the SWOT and FGD methods. Also, the data used are primary data from survey results on Rooftop PV users in Indonesia. The SWOT analysis results show that PLN is in quadrant I, which means it has an extreme market position. PT PLN has the potential to increase the use of Rooftop PV in Indonesia. Two strategic points can be carried out by PLN, namely, carrying out vertical integration and diversifying conglomerates. Also, the researcher has proposed several alternative strategies, namely the upstream (manufacturing) development plan, the downstream (consumer) development plan, and the internal development plan.

1) Background: Taxol facilitates nerve regeneration but it has unwanted side-effects via the tumor suppressive function of miR-34a which induces XIST expression and affects paclitaxel administration. We aimed to examine the neuroprotective effect of Paclitaxel-loaded niosomes (PLNs) to enhance fertility by combining with the Belpharis Persica (B.P) extract in male Wistar rat model of spinal cord injury (SCI). 2) Methods: PLNs were prepared using the thin-film hydration method. After laminectomy at thoracic level (T9/T10) SCI was induced using the weight drop method. Abstract Background The human L39X phospholamban (PLN) cardiomyopathic mutant has previously been reported as a null mutation but the detailed molecular pathways that lead to the complete lack of detectable protein remain to be clarified. Previous studies have shown the implication between an impaired cellular degradation homeostasis and cardiomyopathy development. Therefore, uncovering the underlying mechanism responsible for the lack of PLN protein has important implications in understanding the patient pathology, chronic human calcium dysregulation and aid the development of potential therapeutics. Methods A panel of mutant and wild-type reporter tagged PLN modified mRNA (modRNA) constructs were transfected in human embryonic stem cell-derived cardiomyocytes. Lysosomal and proteasomal chemical inhibitors were used together with cell imaging and protein analysis tools in order to dissect degradation pathways associated with expressed PLN constructs. Transcriptional profiling of the cardiomyocytes transfected by wild-type or L39X mutant PLN modRNA was analysed with bulk RNA sequencing. Results Our modRNA assay system revealed that transfected L39X mRNA was stable and actively translated in vitro but with only trace amount of protein detectable. Proteasomal inhibition of cardiomyocytes transfected with L39X mutant PLN modRNA showed a fourfold increase in protein expression levels. Additionally, RNA sequencing analysis of protein degradational pathways showed a significant distinct transcriptomic signature between wild-type and L39X mutant PLN modRNA transfected cardiomyocytes. Conclusion Our results demonstrate that the cardiomyopathic PLN null mutant L39X is rapidly, actively and specifically degraded by proteasomal pathways. Herein, and to the best of our knowledge, we report for the first time the usage of modified mRNAs to screen for and illuminate alternative molecular pathways found in genes associated with inherited cardiomyopathies.

Dilated cardiomyopathy (DCM) is an umbrella term entailing a wide variety of genetic and non-genetic etiologies, leading to left ventricular systolic dysfunction and dilatation, not explained by abnormal loading conditions or coronary artery disease. The clinical presentation can vary from asymptomatic to heart failure symptoms or sudden cardiac death (SCD) even in previously asymptomatic individuals. In the last 2 decades, there has been striking progress in the understanding of the complex genetic basis of DCM, with the discovery of additional genes and genotype–phenotype correlation studies. Rigorous clinical work-up of DCM patients, meticulous family screening, and the implementation of advanced imaging techniques pave the way for a more efficient and earlier diagnosis as well as more precise indications for implantable cardioverter defibrillator implantation and prevention of SCD. In the era of precision medicine, genotype-directed therapies have started to emerge. In this review, we focus on updates of the genetic background of DCM, characteristic phenotypes caused by recently described pathogenic variants, specific indications for prevention of SCD in those individuals and genotype-directed treatments under development. The phospholamban (PLN) R14del mutation is associated with arrhythmogenic right ventricular dysplasia (ARVD/C). ARVD/C is a cardiac disease characterized by arrhythmias and structural abnormalities in the right ventricle. Because PLN is a regulator of calcium release, this mutation can have deleterious effects on tissue integrity and contraction. This mutation is a trinucleotide (AGA) deletion that leads to an arginine deletion at position 14 of the PLN structure. Here we show two lines carrying this mutation with typical iPSC morphology, pluripotency, karyotype, ability to differentiate into the three germ layers in vitro, and readily availability for studying pathological mechanisms or ARVD/C.

Aims Guanylyl cyclase-B (GC-B; natriuretic peptide receptor-B, NPR-B) stimulation by C-type natriuretic peptide (CNP) increases cGMP and causes a lusitropic and negative inotropic response in adult myocardium. These effects are not mimicked by NPR-A (GC-A) stimulation by brain natriuretic peptide (BNP), despite similar cGMP increase. More refined methods are needed to better understand the mechanisms of the differential cGMP signalling and compartmentation. The aim of this work was to measure cGMP near proteins involved in regulating contractility to understand compartmentation of cGMP signalling in adult cardiomyocytes. Methods and results We constructed several fluorescence resonance energy transfer (FRET)-based biosensors for cGMP subcellularly targeted to phospholamban (PLB) and troponin I (TnI). CNP stimulation of adult rat cardiomyocytes increased cGMP near PLB and TnI, whereas BNP stimulation increased cGMP near PLB, but not TnI. The phosphodiesterases PDE2 and PDE3 constrained cGMP in both compartments. Local receptor stimulation aided by scanning ion conductance microscopy (SICM) combined with FRET revealed that CNP stimulation both in the t-tubules and on the cell crest increases cGMP similarly near both TnI and PLB. In ventricular strips, CNP stimulation, but not BNP, induced a lusitropic response, enhanced by inhibition of either PDE2 or PDE3, and a negative inotropic response. In cardiomyocytes from heart failure rats, CNP increased cGMP near PLB and TnI more pronounced than in cells from sham-operated animals. Conclusion These targeted biosensors demonstrate that CNP, but not BNP, increases cGMP near TnI in addition to PLB, explaining how CNP, but not BNP, is able to induce lusitropic and negative inotropic responses. This study aims to measure and compare the efficiency level of several distribution units within the PLN service area Semarang located in Semarang City, Central Java Province. Then, using a soft system methodology (SSM) approach, this study also intends to give some ideas on enhancing the less efficient distribution unit. This research used seven variables as input (medium voltage transmission lines, low voltage transmission lines, one-phase transformer capacity, three-phase transformer capacity, the number of electrical substations, the number of house transmission, and the amount of electricity generated) and one variable as output (the amount of electricity sold). Using input-oriented, 57% of distribution units, four units out of 7 units, were considered efficient units, and the efficiency average was equal to 0.972. The less efficiency happens due to losses in medium voltage transmission lines, low-voltage transmission lines, and transformers. In this case, losses in medium voltage due to the electrical cable being too lengthy or not in compliance with the standard, losses in the low-voltage network due to electrical theft, and losses in the transformer due to low-capacity usage. Then to address this issue, several ideas have been generated, such as (i) straightening the bent of the electrical cable to reduce its length or modifying the network configuration by dividing the electrical cable into several sections (make sure that the cable length on each section compliance with the provisions of PLN); (ii) make arrangements for supervising and controlling schedules with the local security agencies; and (iii) carry out promotional activities with attractive offers to attract new customers or encourage existing customers who use small electrical power to switch to higher electrical power [ABSTRACT FROM AUTHOR]

Objective: The purpose of this study was to investigate the relationship between lymph node harvest and the prognosis in locally advanced rectal cancer (LARC) patients after neoadjuvant chemoradiotherapy (nCRT). Methods: Patients who were diagnosed with clinical LARC and treated with nCRT and radical surgery between June 2008 and July 2017 were included in this study. The relationship between lymph node retrieval and prognosis was analyzed. Other lymph node-related indicators were explored. Results: A total of 837 patients with a median follow-up of 61 (7-139) months were included in the study. The five-year DFS and OS rates of all patients were 74.9% and 82.3%, respectively. Multivariate survival analysis suggested that dissection of ≥ 12 lymph nodes did not improve OS or DFS. 7 was selected as the best cutoff value for the total number of lymph nodes retrieved by Cox multivariate analysis (χ2 = 10.072, HR: 0.503, P=0.002). Dissection of ≥ 5 positive lymph nodes (PLNs) was an independent prognostic factor for poorer DFS (HR: 2.104, P=0.004) and OS (HR: 3.471, p<0.001). A positive lymph node ratio (LNR) of more than 0.29 was also an independent prognostic factor for poorer DFS (HR: 1.951, P=0.002) and OS (HR: 2.434, p<0.001). Conclusion: The recommends that at least 7 harvested lymph nodes may be more appropriate for LARC patients with nCRT. PLN and LNR may be prognostic factors for LARC patients with ypN+ after nCRT. This research aimed to design a novel teaching and learning process in the professional experience training. The Successive Approximation Model (SAM) was applied in the design steps. The attainment of the designed framework derived from numbers of theoretical reviews, as well as the analytical data collected from the expert interviews. The proposed framework included the components of a Personal Learning Network (PLN), the process of PLN, and the professional experience procedure. The components of PLN were: learning resources, learning tools, learning content, and learning activities. In addition, the process of PLN consisted of Creation, Connection, Cognition, and Contribution. The professional experience incorporated nine main activities particularly, enrolling in the course, joining three compulsory seminars, doing two major periods of internship, mentor visiting, submitting assignments, and completing the course. These three concepts were combined into the framework of integrated learning activities, which occurred in social networking tool, Course Networking (CN). It operated as the learning tool utilized the newly created addable badges as a part of gamification features. The assessment results from the eleven experts revealed that the teaching and learning practice using personal learning networks on social networking tools with gamification in the professional experience proved to have high levels of standard evaluation, innovative attributes, design process, teaching and learning procedure, and teaching and learning activities. This study investigated the dependence of contraction from extracellular Ca2+, the presence of a functional sarcoplasmic reticulum (SR), and the effects of β-adrenergic stimulation using isometric cardiac muscle preparations. Moreover, the expression of Ca2+-handling proteins such as SR-Ca2+-ATPase (SERCA), phospholamban (PLN), and Na+/Ca2+ exchanger (NCX) were also evaluated in the ventricular tissue of adult African sharptooth catfish, Clarias gariepinus, a facultative air-breathing fish. In summary, we observed that (1) contractility was strongly regulated by extracellular Ca2+; (2) inhibition of SR Ca2+-release by application of ryanodine reduced steady-state force production; (3) ventricular myocardium exhibited clear post-rest decay, even in the presence of ryanodine, indicating a decrease in SR Ca2+ content and NCX as the main pathway for Ca2+ extrusion; (4) a positive force-frequency relationship was observed above 60 bpm (1.0 Hz); (5) ventricular tissue was responsive to β-adrenergic stimulation, which caused significant increases in twitch force, kept a linear force-frequency relationship from 12 to 96 bpm (0.2 to Hz), and improved the cardiac pumping capacity (CPC); and (6) African catfish myocardium exhibited similar expression patterns of NCX, SERCA, and PLN, corroborating our findings that both mechanisms for Ca2+ transport across the SR and sarcolemma contribute to Ca2+ activator. In conclusion, this fish species displays great physiological plasticity of E-C coupling, able to improve the ability to maintain cardiac performance under physiological conditions to ecological and/or adverse environmental conditions, such as hypoxic air-breathing activity. En Colombia, las redes sociales se han convertido en una poderosa herramienta para expresar opiniones políticas, especialmente durante el período de campaña en las elecciones presidenciales. Este trabajo propone un modelo híbrido para predecir el desenlace de la primera vuelta en las elecciones presidenciales de Colombia en 2018 cuyo objetivo es minimizar el error absoluto y mejorar la calidad de la predicción final. Para ello, las actividades de los usuarios en Twitter y Facebook fueron registradas y analizadas mediante algoritmos de inteligencia artificial, obteniendo como resultado una predicción precisa y coherente con la realidad. Como resultados principales se destaca que el RMSE del modelo híbrido propuesto ronda el 2,47%, superando en promedio el RMSE de las firmas encuestadoras tradicionales más prominentes del país. Adicionalmente también se predijo el valor del abstencionismo electoral con un error diferencial de 1,72% con respecto al valor real, demostrando la confiabilidad de la metodología propuesta.

En el presente trabajo se propone la utilización de las técnicas estilométricas modernas para el estudio y clasificación de geolectos en español, realizando un análisis de Estilometría sobre el corpus COSER. Se utiliza el programa Stylo sobre varias entrevistas transcritas para extraer parámetros fiables de categorización. Al final, se han aplicado los resultados concluyentes sobre todo el corpus, consiguiendo una clasificación no aleatoria que, incluso, es explicable desde algunos trabajos dialectales de referencia.

Abstract Background Rapidly progressive glomerulonephritis has been described in dogs that seroreact to Borrelia burgdorferi, but no studies have compared clinicopathologic differences in Lyme‐seroreactive dogs with protein‐losing nephropathy (PLN) versus dogs with Borrelia‐seronegative PLN. Hypothesis/Objectives Dogs with Borrelia C6 antigen‐seroreactive PLN have distinct clinicopathologic findings when compared to dogs with Borrelia seronegative PLN. Obesity and type 2 diabetes (T2D) are on trend to become a huge burden across all ages. They cause harm to almost every organ, especially the heart. For decades, the incidence of heart failure with impaired diastolic function (or called heart failure with preserved ejection fraction, HFpEF) has increased sharply. More and more studies have uncovered obesity and T2D to be closely associated with HFpEF. The sarcoplasmic/endoplasmic reticulum calcium ATPase2a (SERCA2a) microdomain is a key regulator of calcium reuptake into the sarcoplasmic reticulum (SR) during diastole. 3′,5′-cyclic adenosine monophosphate (cAMP) and its downstream effector cAMP dependent protein kinase (PKA) act locally within the SERCA2a microdomain to regulate the phosphorylation state of the small regulatory protein phospholamban (PLN), which forms a complex with SERCA2a. When phosphorylated, PLN promotes calcium reuptake into the SR and diastolic cardiac relaxation by disinhibiting SERCA2a pump function. In this review, we will discuss previous studies investigating the PLN/SERCA2a microdomain in obesity and T2D in order to gain a greater understanding of the underlying mechanisms behind obesity- and T2D-induced diastolic dysfunction, with the aim to identify the current state of knowledge and future work that is needed to guide further research in the field.

Babelfy is an online tool, developed in the context of Natural Language Processing. When an item with more than one meaning is introduced in Babelfy, it chooses the appropriate meaning considering the context. The objective of this research study is to test the Word Sense Disambiguation skills of Babelfy in Spanish from a linguistic approach. To do so, a descriptive and comparative study between Babelfy and native Spanish speakers was carried out. Twenty-two pairs of sentences with an ambiguous word were designed, the first sentence of the pair had a neutral context and the second one a facilitating context. These sentence-pairs were introduced in Babelfy to check which meaning of the ambiguous word was selected and to explore whether there were differences depending on the type of context. These results were then compared to the answers of sixty-two Spanish native speakers. The data show that the behaviour of speakers when encountering an ambiguous word is not equivalent to the way Babelfy performs Word Sense Disambiguation, especially when the context is neutral, and the word has related meanings. El artículo aborda la cuestión de cómo determinar la polaridad u opinión en textos, particularmente en Twitter. El análisis se hace desde múltiples perspectivas: ética de la información, comunicación, ciencia de la información e informática, enfatizando en dos análisis que se complementan: el procesamiento de lenguaje natural y la organización del conocimiento. Se desarrolla un prototipo que propone anotar las palabras cargadas de opinión como los adjetivos y los adverbios contextualizadas por su núcleo: sustantivos y verbos respectivamente. Estos núcleos sirven también para modelar la terminología del dominio y se propone la creación de un lexicón ad hoc.

Os provérbios são expressões de uso generalizado, utilizados em diferentes situações conversacionais e assumindo diferentes funções no discurso em que se integram. Do ponto de vista sintático, este tipo de expressões apresenta uma grande variedade de estruturas. Tendo isto em conta, o objetivo principal deste trabalho é estabelecer uma classificação formal sintática dos provérbios do português europeu. Para tal, pretendemos desenvolver e aprofundar a tipologia de classificação formal proposta por Rassi et al. (2014). Por conseguinte, será considerada uma subclassificação para as classes que apresentam um elevado número de provérbios e uma eventual reclassificação de alguns dos tipos. A proposta de classificação foi validada pela anotação independente por dois linguistas de uma lista de provérbios muito usuais, medindo-se depois o acordo entre anotadores, que foi muito elevado. Esta classificação, por sua vez, será o ponto de partida para o desenvolvimento de um procedimento de classificação automática deste tipo de estruturas, e contribuir assim para a elaboração de recursos para diferentes aplicações em Processamento da Linguagem Natural (PLN).

With the undeveloped BAPV(Building Applied Photovoltaics) at Office Building and Public Facilities, Caused the high price of solar modules and the lack the economic study on the use of solar modules that housed in the Banda Aceh region. Furthermore, the price of solar modules is expensive, so people think it will cost so much funds to building a BAPV’s system. These problems could be overcomed if the existing technical studies and economic studies of the application of the BAPV’s system. This study aims to assess the application of the BAPV’s system on institutions building in terms of technical and economic value, in this case the building that is used as the study object is the Polytechnic Aceh’s Building. The method that used in the technical studies are theoretical calculations and simulations using helioscope software, while the methods used for economic studies is using the methods of cost-benefit analysis (cost benefit analysis). The method used to find the NPV (Net Present Value), PP (Payback Period), IRR (Internal Rate of Return), and BCR (Benefit Cost Ratio). If the average value of solar radiation reaching 4.79 kWh / m2 / day and the average daily energy requirement is 592 kWh, the energy generated from BAPV-PLN hybrid system on the roof of the object building will reach the amount of 237 MWh/year with the capacity charge controller used is 7490 A and the capacity of the battery used is 64.487 Ah. Panel tilt angle used is 25o and the type of panel used is Monocrystalline manifold. From the economic value will obtained NPV value of Rp. 20.022.106.937, PP during 5,2 years, IRR of 36% and 3,49 of BCR. Based on the evaluation results of the feasibility study, the project of hybrid BAPV-PLN’s system on the roof of the Polytechnic Aceh’s building can be realized, because its already meet the criteria of the feasibility study to make the systems get established in real term.

The main adverse effect of tyrosine kinase inhibitors, such as sunitinib, is cardiac contractile dysfunction; however, the molecular mechanisms of this effect remain largely obscure. MicroRNAs (miRNAs) are key regulatory factors in both cardiovascular diseases and the tyrosine kinase pathway. Therefore, we analyzed the differential expression of miRNAs in the myocardium in mice after exposure to sunitinib using miRNA microarray. A significant downregulation of miR-146a was observed in the myocardium of sunitinib-treated mice, along with a 20% decrease in left ventricle ejection fraction (LVEF). The downregulation of miR-146a was further validated by RT-qPCR. Among the potential targets of miR-146a, we focused on Pln and Ank2, which are closely related to cardiac contractile dysfunction. Results of luciferase reporter assay confirmed that miR-146a directly targeted the 3′ untranslated region of Pln and Ank2. Significant upregulation of PLN and ANK2 at the mRNA and protein levels was observed in the myocardium of sunitinib-treated mice. Cardiac-specific overexpression of miR-146a prevented the deteriorate effect of SNT on calcium transients, thereby alleviating the decreased contractility of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). SiRNA knockdown of PLN or ANK2 prevented sunitinib-induced suppression of contractility in hiPSC-CMs. Therefore, our in vivo and in vitro results showed that sunitinib downregulated miR-146a, which contributes to cardiac contractile dysfunction by regulating the downstream targets PLN and ANK2, and that upregulation of miR-146a alleviated the inhibitory effect of SNT on cardiac contractility. Thus, miR-146a could be a useful protective agent against sunitinib-induced cardiac dysfunction.

Background: Rapidly progressive glomerulonephritis has been described in dogs that seroreact to Borrelia burgdorferi, but no studies have compared clinicopathologic differences in Lyme‐seroreactive dogs with protein‐losing nephropathy (PLN) versus dogs with Borrelia‐seronegative PLN. Hypothesis/Objectives: Dogs with Borrelia C6 antigen‐seroreactive PLN have distinct clinicopathologic findings when compared to dogs with Borrelia seronegative PLN. Animals: Forty dogs with PLN and Borrelia C6 antigen seroreactivity and 78 C6‐seronegative temporally matched dogs with PLN. Methods: Retrospective prevalence case‐control study. Clinical information was retrieved from records of dogs examined at the University of California, Davis, Veterinary Medical Teaching Hospital. Histopathologic findings in renal tissue procured by biopsy or necropsy of dogs with PLN were reviewed. Results: Retrievers and retriever mixes were overrepresented in seroreactive dogs (P < .001). Seroreactive dogs were more likely to have thrombocytopenia (P < .001), azotemia (P =.002), hyperphosphatemia (P < .001), anemia (P < .001), and neutrophilia (P =.003). Hematuria, glucosuria, and pyuria despite negative urine culture were more likely in seroreactive dogs (all P ≤ .002). Histopathologic findings were consistent with immune‐complex glomerulonephritis in 16 of 16 case dogs and 7 of 23 control dogs (P = 006). Prevalence of polyarthritis was not different between groups (P =.17). Conclusions and Clinical Importance: C6 seroreactivity in dogs with PLN is associated with a clinicopathologically distinct syndrome when compared with other types of PLN. Abstract Background Pathogenic variants in human phospholamban coding gene (PLN) are known to cause hereditary dilated cardiomyopathy with heart failure in an autosomal dominant mode. Methods We performed high-depth targeted next-generation sequencing using a cardiomyopathy-panel containing 80 disease-related genes in 650 unrelated patients with non-ischemic cardiomyopathy to identify potential pathogenic PLN variants. To comprehensively evaluate the genetic cause of the proband and his pedigree, whole-exome sequencing and Sanger sequencing were performed. Results A novel homozygous nonsense variant (p.Glu2Ter, c.4G>T) in PLN was identified in a 36-year-old male suffering from dilated cardiomyopathy with severe heart failure. No more cardiomyopathy-causing variant or likely pathogenic copy number variation was identified. This variant was not detected in 800 unrelated healthy controls. Furthermore, the variant is not in the Exome Aggregation Consortium or the Genome Aggregation databases. Western blots showed that this variant significantly reduced the expression of phospholamban. Furthermore, in pedigree analysis, we found that all five heterozygous PLN -p.Glu2Ter carriers (including four elder relatives) had normal heart size and cardiac function, which revealed a novel autosomal recessive inheritance mode. Conclusions Our study identified a novel pathogenic variant of PLN , and revealed a novel pathogenic inheritance mode of PLN causing dilated cardiomyopathy with heart failure. This paper describes an unsupervised system for sentiment analysis is presented in Spanish. The system performs a complete fine grain analysis, where the most important characteristics or aspects of an opinion are identified in order to determine their sentiment associated. The unsupervised approach used allows to extract, identify and sentiment classify, from the analysis of opinions in Spanish in a specific domain, allowing to scale to another language and domain with great ease. For the validation of AspectSA, several experiments were carried out using corpus of opinions in the restaurant domain. Objectives: Thrombosis is a well‐recognized phenomenon in dogs and cats with a significant impact on morbidity and mortality. Despite growing awareness of thrombosis and increased use of antithrombotic therapy, there is little information in the veterinary literature to guide the use of anticoagulant and antiplatelet medications. The goal of Domain 1 was to explore the association between disease and thrombosis in a number of conditions identified as potential risk factors in the current veterinary literature, to provide the basis for prescribing recommendations. Design: A population exposure comparison outcome format was used to represent patient, exposure, comparison, and outcome. Population Exposure Comparison Outcome questions were distributed to worksheet authors who performed comprehensive searches, summarized the evidence, and created guideline recommendations that were reviewed by domain chairs. Revised guidelines then underwent the Delphi survey process to reach consensus on the final guidelines. Diseases evaluated included immune‐mediated hemolytic anemia, protein‐losing nephropathy, pancreatitis, glucocorticoid therapy, hyperadrenocorticism, neoplasia, sepsis, cerebrovascular disease, and cardiac disease. Settings: Academic and referral veterinary medical centers. Results: Of the diseases evaluated, a high risk for thrombosis was defined as dogs with immune‐mediated hemolytic anemia or protein‐losing nephropathy, cats with cardiomyopathy and associated risk factors, or dogs/cats with >1 disease or risk factor for thrombosis. Low or moderate risk for thrombosis was defined as dogs or cats with a single risk factor or disease, or dogs or cats with known risk factor conditions that are likely to resolve in days to weeks following treatment. Conclusions: Documented disease associations with thrombosis provide the basis for recommendations on prescribing provided in subsequent domains. Feedlot cattle often contain Salmonella . The number of bacteria that initiate colonization of different cattle organs and the bacterial migration within these large animals are poorly understood. To investigate these questions, we constructed w ild-type i sogenic t agged s trains (WITS) of Salmonella by inserting 21-base barcodes flanked by Illumina sequencing primers into a neutral genome location. We then delivered several different pools of uniquely barcoded clones orally and into multiple intradermal sites, in individual Holstein steers, and subsequently performed Salmonella -directed s equence t ag-based a nalysis of m icrobial p opulations (STAMP). Using high-throughput sequencing of the barcodes of Salmonella grown from steer lymph nodes, organs and feces, we monitored how individual barcoded clones travel from different entry sites within animals. Data showed that gastrointestinal colonization was established by up to hundreds of Salmonella founder cells, whereas peripheral lymph nodes were usually colonized by very low numbers of founding bacteria, often originating from the nearest draining intradermal delivery site. Transmission of Salmonella from the gastrointestinal tract to the lymphatic system was frequently observed, whereas entry of intradermally delivered bacteria into the gut was rare. Bacteria undergo limited extraintestinal proliferation within or prior to arrival at peripheral lymph nodes. Overall, the application of the STAMP technique facilitated characterization of the migration routes and founder population size of Salmonella within feedlot cattle and their organs and lymph nodes in unprecedented detail. While integrating linguistic knowledge of any kind is becoming an almost implicit practice in natural language understanding systems, the inclusion of cultural or world knowledge in these tools might have been neglected sometimes. However, a NLP system or knowledge base enriched with cultural information is a more robust, better cohesioned instrument for natural language understanding processes. The integration of this type of knowledge in NLP systems may be proven to contribute to solving some phenomena that occur in natural language, such as anaphor, metaphor and metonymy, ambiguity or co-reference, amongst others. The objective of this article is to describe the way FunGramKB (a knowledge base) integrates cultural knowledge in its conceptual modules and, in particular, how the information contained in the Onomasticon module of FunGramKB can contribute to maximising the informativeness and completeness of the whole system, thus resolving ambiguity problems in a determined linguistic phenomenon: anaphora.

Many faculty and staff in higher education have turned to digitally-enhanced professional learning networks (PLNs) as a means for situated learning that can help them grow in their various professional responsibilities. However, there is scant research on what these PLNs mean to the professional lives of higher educators. We report findings of a qualitative study that investigated participants' perspectives on their PLN experiences through analysis of survey data from 151 higher education faculty and professionals. Data analysis suggested that the anytime, anywhere availability of expansive PLNs, and their capacity to respond to educators' diverse interests and needs, fostered new learning experiences. Participants reported that their PLNs supported professional growth, specifically in the areas of teaching and learning. Background Interleukin-18 (IL-18) neutralization protects against lipopolysaccharide (LPS)-induced injuries, including myocardial dysfunction. However, the mechanism is yet to be fully elucidated. The aim of the present study was to determine whether IL-18 gene deletion prevents sepsis-induced cardiac dysfunction and to elucidate the potential mechanisms underlying IL-18-mediated cardiotoxicity by LPS. Methods and results Ten-week-old male wild-type (WT) and IL-18 knockout (IL-18 KO) mice were intraperitoneally administered LPS. Serial echocardiography showed better systolic pump function and less left ventricular (LV) dilatation in LPS-treated IL-18 KO mice compared with those in LPS-treated WT mice. LPS treatment significantly decreased the levels of phospholamban (PLN) and Akt phosphorylation in WT mice compared with those in saline-treated WT mice, while the LPS-induced decrease in the phosphorylation levels was attenuated in IL-18 KO mice compared with that in WT mice. IL-18 gene deletion also attenuated an LPS-induced increase of type 2 protein phosphatase 2A (PP2A) activity, a molecule that dephosphorylates PLN and Akt. There was no difference in type 1 protein phosphatase (PP1) activity. To address whether IL-18 affects PLN and Akt phosphorylation via PP2A activation in cardiomyocytes, rat neonatal cardiac myocytes were cultured and stimulated using 100 ng/ml of recombinant rat IL-18. Exogenous IL-18 decreased the level of PLN and Akt phosphorylation in cardiomyocytes. PP2A activity but not PP1 activity was increased by IL-18 stimulation in cardiomyocytes. Conclusions IL-18 plays a pivotal role in advancing sepsis-induced cardiac dysfunction, and the mechanisms underlying IL-18-mediated cardiotoxicity potentially involve the regulation of PLN and Akt phosphorylation through PP2A activity. It is widely acknowledged that to be able to teach language and literacy with digital technologies, teachers need to engage in relevant professional learning. Existing formal models of professional learning are often criticised for being ineffective. In contrast, informal and self-initiated forms of learning have been recently recognised as meaningful and relevant but little is known about these authentic learning experiences for professional purposes. This article investigates three teachers’ self-initiated professional learning about digital technologies through their Personal Learning Networks. The analysis identified that the participants’ everyday digital literacy practices provided opportunities for a range of professional practices that supported learning about digital technologies: information retrieval and resources aggregation, cooperation, collaboration, reflections and socialising. It also revealed that the teachers considered this form of learning as valuable. The article concludes by discussing how knowledge about self-initiated professional learning might be used to draw out some implications for teachers’ professional development. [ABSTRACT FROM AUTHOR]
