Bacterial infection is one of the main reasons for the clinical failure of oral titanium restorations. In this study, biodegradable polymer/TiO2 nanotube nanoarray capsules were constructed on a titanium substrate surface to locally deliver drugs for long-lasting antibacterial properties. Anodization was applied to prepreparation TiO2 nanotube array film on titanium substrate, and then, the upward opening TiO2 nanotubes were sealed with biodegradable polymer (chitosan and polyethylene glycol) through electrochemical deposition. Scanning electron microscope (SEM) and X-ray photoelectron spectroscopy (XPS) were used to analyze the characterization of this system. The drug release characteristics and the antibacterial activity demonstrated that the polymer coating significantly reduced burst release and enhanced lasting antibacterial properties. The nanoarray capsules still preserved integrity with a little degradation after 2 days. The strategy described herein provides a versatile route for designing targeted drug delivery systems in orthopaedical and other biomedical fields. [ABSTRACT FROM AUTHOR]

Selective laser melting used in manufacturing custom-made titanium implants becomes more popular. In view of the important role played by osteoclasts in peri-implant bone resorption and osseointegration, we modified selective laser melting-manufactured titanium surfaces using sandblasting/alkali-heating and sandblasting/acid-etching, and investigated their effect on osteoclast differentiation as well as their underlying mechanisms. The properties of the surfaces, including elements, roughness, wettability and topography, were analyzed. We evaluated the proliferation and morphology of primary mouse bone marrow-derived monocytes, as well as induced osteoclasts derived from bone marrow-derived monocytes, on samples. Then, osteoclast differentiation was determined by the tartrate-resistant acid phosphatase activity assay, calcitonin receptors immunofluorescence staining and the expression of osteoclast-related genes. The results showed that sandblasting/alkali-heating established nanonet structure with the lowest water contact angle, and both sandblasting/alkali-heating and sandblasting/acid-etching significantly decreased surface roughness and heterogeneity compared with selective laser melting. Surface modifications of selective laser melting-produced titanium altered bone marrow-derived monocyte morphology and suppressed bone marrow-derived monocyte proliferation and osteoclastogenesis in vitro (sandblasting/alkali-heating>sandblasting/acid-etching>selective laser melting). These surface modifications reduced the activation of extracellular signal-regulated kinase and c-Jun N-terminal kinases compared to native-selective laser melting. Sandblasting/alkali-heating additionally blocked tumor necrosis factor receptor-associated factor 6 recruitment. The results suggested that sandblasting/alkali-heating and sandblasting/acid-etching modifications on selective laser melting titanium could inhibit osteoclast differentiation through suppressing extracellular signal-regulated kinase and c-Jun N-terminal kinase phosphorylation in mitogen-activated protein kinase signaling pathway and provide a promising technique which might reduce peri-implant bone resorption for optimizing native-selective laser melting implants. Selective laser melting (SLM) titanium is a suitable material for use in customized implants. However, long-term implant survival requires both successful osseointegration and promising antibacterial characteristics. Native SLM titanium thus requires proper modifications to improve its bioactivity and antibacterial efficacy. Micro-arc oxidation (MAO) was conducted on sandblasted SLM substrate to form a microporous TiO2 coating. Subsequently, hydrothermal treatment was applied to fabricate micro-nano zinc-incorporated coatings with different Zn content (1 mM-Zn and 100 μM-Zn). Surface characterization was performed using scanning electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, a three-dimensional profilometer, and a contact angle measuring device. The osteoblast-like cell line MC3T3-E1, Subclone 14, was used in cell viability assays to evaluate adhesion, proliferation, and ALP activity. An antibacterial assay was conducted using Streptococcus sanguinis and Fusobacterium nucleatum. Zn-incorporated samples exhibited higher adhesion, proliferation, and differentiation than did SLM and MAO samples. 100 μM Zn samples exhibited the highest proliferation, and 1 mM-Zn samples manifested the highest ALP activity. In addition, Zn-incorporated samples exerted inhibitory effects on both Streptococcus sanguinis and Fusobacterium nucleatum. Combining micro-arc oxidation and hydrothermal treatment, we successfully fabricated a novel Zn-incorporated coating on a microporous SLM surface which possesses both outstanding bioactivity and antibacterial efficacy. Abstract: A composite structure, TiO2 nanotube array/CdSe nanoparticle/TiO2 layer, was fabricated by inserting CdSe nanoparticles into anodized TiO2 nanotube arrays via ultrasonic-assisted cyclic voltammetry electrochemical deposition and subsequently wrapped by a TiO2 thin layer via TiCl4 hydrolysis. The films were characterized by X-ray diffraction, field emission scanning electron microscopy, energy dispersive spectroscopy and UV–vis diffuse reflectance spectroscopy. For the film used as the photoanode, linear sweep voltammetry and transient photocurrent were investigated in a three-electrode system under visible light illumination. And the photocatalytic activity was evaluated by the oxidation of glucose in the supporting electrolyte at different potential bias under visible light illumination. The results showed that CdSe nanoparticles are prone to disperse into the nanotube arrays via cyclic voltammetry electrochemical deposition under ultrasonic condition. Increasing cyclic voltammetry cycle increases the loading of CdSe on the TiO2 nanotube arrays. The CdSe-sensitized TiO2 nanotube array films exhibit a wide-absorption response in the visible light region. The TiO2 nanotube array/CdSe/TiO2 film, prepared by ultrasonic-assisted electrochemical deposition with 50cyclic voltammetry cycles and subsequently coating a TiO2 thin layer, exhibits better protection against photocorrosion of CdSe. The TiO2NTA/CdSe/TiO2 film possesses visible light photoelectrocatalytic activity on the degradation of glucose. [Copyright &y& Elsevier]

Selective laser melting (SLM) has promising prospects in manufacturing customized implants, however the rough surface of SLM titanium specimen can facilitate bacterial adherence and biofilm formation, which is a risk to implant success. Therefore, surface modification is required to enhance its antibacterial efficacy. Sandblasting, anodization and electrochemical deposition were applied to construct a novel composite nanostructure of nanophase calcium phosphate embedded to TiO 2 nanotubes on microrough SLM titanium substrates (NTN). NTN samples were compared with TiO 2 nanotubes (NT) samples, mechanical polished (MP) samples and untreated SLM titanium samples. Surface characterization were analyzed using scanning electron microscope, energy dispersive spectroscopy, x-ray photoelectron spectroscopy, x-ray diffraction, a three dimensional profilometer and a contact angle measuring device. Bacteria adhesion assay for bacteria colony counting and bacteria LIVE/DEAD staining was conducted using Streptococcus mutans and Streptococcus sanguinis. Both S. mutans and S. sanguinis adherence on SLM samples were significantly higher than on NTN, NT and MP samples. The antibacterial efficacy of NTN samples was superior compared to NT and had no significant difference with MP samples, despite the fact that NTN samples had much rougher surface than MP samples. This study elucidates an efficient method to enhance antibacterial efficacy on rough SLM surfaces, which contributes to its application in dental and other biomedical implants.

The selective laser melting (SLM) technique is a recent additive manufacturing (AM) technique. Several studies have reported success in the SLM-based production of biocompatible orthopaedic implants and three-dimensional bone defect constructs. In this study, we evaluated the surface properties and biocompatibility of an SLM titanium implant in vitro and compared them with those of a machined (MA) titanium control surface. In addition, we evaluated the osseointegration capability of the SLM implants in vivo and compared it with those of MA and Nobel-speedy (Nobel-S) implants. SLM microtopographical surface analysis revealed porous and high roughness with varied geometry compared with a smooth surface in MA Ti samples but with similar favourable wettability. Osteoblast proliferation and alkaline phosphatase activity were significantly enhanced on the SLM surface. Histological analysis of the bone-implant contact ratio revealed no significant difference among SLM, MA, and Nobel-S implants. Micro-CT assessment indicated that there was no significant difference in bone volume fraction around the implant among SLM implants and other types of surface modification implants. The removal torque value measurement of SLM implants was significantly lower that of than Nobel-S implants P < 0.001 and higher than that of MA implants. The study demonstrates the capability of SLM implants to integrate with living bone. The SLM technique holds promise as a new dental implant manufacturing technique.
