Chronic inflammation and oxidative stress can contribute to the development of cardiovascular diseases. Isoproterenol (ISO), a synthetic catecholamine, is used to study the effects of drugs on cardiotoxicity. Agmatine (AGM) is a type of biogenic amine produced through the decarboxylation of arginine. The purpose of the current study was to evaluate the effects of AGM against ISO-induced cardiotoxicity due to the described roles of AGM in cardiovascular disease. Four groups of thirty-two Wistar Albino rats were divided equally as control, ISO, AGM+ISO, and ISO+AGM. ISO was administered intraperitoneally (i.p.) twice at a dose of 150 mg/kg, at 24-hour intervals. Prior to and after ISO injection, 20 mg/kg of AGM was injected i.p. Hemodynamic measurements and serum and tissue biochemical analyses were evaluated at the end of the experiment. The levels of glutathione (GSH), catalase (CAT), superoxide dismutase (SOD), and malondialdehyde (MDA) in the tissue were measured. Ovarian torsion is a gynecological emergency characterized by ovarian ischemic damage. This study aimed to investigate the possible protective effects of dexpanthenol (DEX)-an antioxidant molecule-against ovarian ischemic damage in rats. The rats were simple randomly grouped into (1) Sham group (n=8); (2) Ischemia (I) group (n=8) (2 hours of I in ovaries); (3) DEX+I group (n=8) (500 mg/kg DEX administration 30 minutes before 2 hours of I.) The removed ovaries were examined histopathologically and biochemically. In the DEX+I group, necrosis-pycnosis severity was significantly reduced compared to the I group; but congestion-hemorrhage severity was similar. In the I group, malondialdehyde (MDA) and total oxidant status (TOS) levels were slightly increased, and glutathione (GSH) levels were significantly decreased compared to the Sham group. In the DEX+I group, MDA and TOS levels were slightly reduced and GSH levels were slightly increased compared to the I group. DEX prevents ischemic damage in rat ovaries via an antioxidant effect. We aimed to investigate the potential benefits of two doses of ivabradine (IVA)-an If sodium channel blocker against isoproterenol (ISO)-induced myocardial damage in rats.The rats were randomly divided into 4 groups: Control group (n=8); Saline was administered, ISO group (n=12); 150 mg/kg ISO was administered for 2 days, ISO+IVA (1 mg/kg) group (n=12); 1 mg/kg IVA was administered for 4 days in addition to ISO. ISO+IVA (10 mg/kg) group (n=12): 10 mg/kg IVA was administered for 4 days in addition to ISO. Thereafter, hemodynamic, histopathological, and biochemical studies were performed. In the ISO+IVA (1 mg/kg) and ISO+IVA (10 mg/kg) groups, ISO-induced myocardial changes including an increase in density of granulation tissue and degenerated cardiomyocyte were equally decreased. HR was mildly reduced, and arterial blood pressures were slightly increased in the IVA-treated groups versus the ISO group. In the hearts of IVA-treated groups, malondialdehyde (MDA) levels were significantly reduced and glutathione (GSH) level and catalase (CAT) activity were mildly increased compared to the ISO group. Elevation of GSH and CAT activity were more pronounced in the ISO+IVA (10 mg/kg) group. Our results indicate that both 1 mg/kg and 10 mg/kg doses of IVA are effective against heart damage induced by ISO via its negative chronotropic, anti-oxidant (dose-dependent), anti-inflammatory and anti-degenerative properties. Atypical antidepressant mirtazapine (MIR) is primarily used to treat major depressive disorder. It has not been clarified whether cardiovascular uncertainties and mechanisms of action emerge as problems during the use of mirtazapine. Cisplatin (CIS) is an effective anti-cancer medication used to treat a variety of human malignancies. There were four groups of 32 Wistar albino male rats in all. Rats were split into 4 groups at random. 1. Control Group, 2. CIS Group, 3. MIR Group, 4. MIR+CIS Group. On the 15th day of the study, ECG, heart rate, and blood pressure were determined. Histopathological and biochemical analyses were carried out on cardiac and vascular tissue samples. Aim:Carbon tetrachloride (CCI4) is a volatile organic chemical agent that can cause damage to many tissues. Caffeic acid phenethyl ester (CAPE), which is structurally similar to flavonoids, is an active component of honeybee propolis. CAPE is known for its antitoxic, antioxidant, and anti-inflammatory effects. In this study, we aimed to investigate the effects of CAPE against testicular damage caused by CCI4.Materials and Methods:Twenty-eight Wistar albino rats were divided into 4 groups (n=7) as, Group 1: control (5% ethanol, 1 mL/day/ip), Group 2: olive oil (0.5 mL/day over/ip), Group 3: CCI4 (0.5 mL/kg over/ip), Group 4: CCI4+CAPE (10 μmol/kg/day/ip). Tissue samples collected at the end of the experiment were detected in 10% formaldehyde and embedded in paraffin. Five-micron-thick sections taken from paraffin blocks were stained with hematoxylin-eosin. To evaluate testicular damage, 100 tubules from each section were randomly examined at 20x magnification under a light microscope and classified as intact, atrophic, and degenerated tubules. Sections were examined by using Leica DFC 280 light microscope and Leica Q Win Image Analysis system (Leica Micros Imaging Solutions Ltd. Cambridge, UK).Results:The testicular sections of the control group and the olive oil group had a normal histological appearance. In the CCI4 group, 55.00±4.22% of the seminiferous tubules were intact, 25.00±2.67% were atrophic and 20.00±1.88% were degenerative. Objective(s): Varenicline is a selective partial agonist for the nicotinic acetylcholine receptor a4b2 subtype, which is widely used to treat smoking addiction. However, there is still no data about its potential toxic effects on tissues. In this study, we aimed to determine the varenicline-induced toxicity on reproductive and renal tissues in rats.Materials and Methods: Rats were randomly divided into two groups: control (n=10) and varenicline (n=24). Then, rats in each group were sub-divided equally as acute and chronic groups. The control rats were orally given distilled water only. Varenicline was administrated orally as follows: 1st–3rd days 9 µg/kg/day, 4th–7th days 9 µg/kg twice daily, and 8th–90th days 18 µg/kg twice daily. The rats of acute and chronic groups were sacrificed on the 45th and 90th days, respectively. Some tissue markers related to oxidative stress were measured, and sperm characteristics were observed.Results: In the acute group, varenicline led to a significant decrease in SOD activities in both kidney and testis tissues. In the chronic group, varenicline significantly increased MDA and MPO production, and reduced CAT and GPx levels in the kidneys and testes. Also, SOD and GSH levels significantly decreased in the testes. Moreover, sperm characteristics were negatively affected; histopathological deformation was observed in the testes and kidneys in all groups.Conclusion: This study showed that varenicline could detrimentally affect the kidneys and testes in both acute and chronic term usage. Further studies will provide more insights into the molecular dynamics of this damage.

Objective(s): The current study was designed to investigate the therapeutic and protective effects of montelukast (ML) against doxorubicin (DOX)-induced acute kidney damage in rats.Materials and Methods: Thirty-five Wistar albino female rats were randomly divided into 5 groups as follows: Group I: Control; Group II: Control+ML; Group III: DOX; Group IV: DOX+ML; Group V: ML+DOX. At the end of the experiment, the kidney tissues of rats were collected. Thiobarbituric acid reactive substance (TBARS), reduced glutathione, superoxide dismutase (SOD), and catalase levels were determined from the kidney tissues. In addition, the kidney tissues were examined histologically.Results: DOX induced a significant increase in the kidney TBARS levels, whereas SOD contents significantly decreased when compared with the control group. On the other hand, ML administration before and after DOX injection caused significant decreases in TBARS production and also increases in SOD levels. Histologically, the most remarkable damage was glomerulosclerosis and tubular changes in the DOX group. Moreover, marked tubular necrosis and swelling in tubular epithelial cells were observed in this group. Contrarily, although glomerulosclerosis was recognized as alleviated also in both DOX+ML and ML+DOX groups, the lesions did not completely ameliorate. However, treatment with ML after DOX injection was more effective than treatment with ML before DOX injection with respect to the protection of tubular structures. Conclusion: It was determined that ML treatment after DOX injection caused therapeutic effects against DOX-induced kidney damage. Thence, ML treatment is of some clinical properties for oxidative stress damage in kidney tissues.

RESUMO Objetivo O objetivo deste estudo foi investigar os efeitos agudos e crônicos da vareniclina no tecido pulmonar em um estudo experimental. Métodos Um total de 34 ratos foi alocado aleatoriamente em grupos de estudo (vareniclina) e controle. Assim, os ratos foram divididos em dois grupos: (i) grupo controle e (ii) grupo vareniclina. A seguir, os ratos de cada grupo foram, por sua vez, subdivididos igualmente em agudos (C1; V1) e crônicos (C2; V2), e todos os ratos dos grupos agudos e crônicos foram sacrificados sob anestesia: no 45.º dia, para o grupo agudo [C1 (n=5) e V1 (n=12)], e no 90.º dia, para o grupo crônico [C2 (n=5) e V2 (n=12)], respectivamente. Em seguida, foram realizadas análises bioquímicas e histopatológicas. Resultados Trinta e quatro ratos completaram o estudo. Destes ratos, 24 estavam no grupo vareniclina e 10 no grupo controle. Na exposição crônica à vareniclina, os níveis de oxidante composto por malondialdeído (MDA) e mieloperoxidase (MPO) aumentaram, e os níveis de superóxido dismutase (SOD), catalase (CAT), glutationa (GSH) e glutationa peroxidase (GPx), nomeados como antioxidantes, diminuiram significativamente quando comparados com o grupo controle. Os níveis de MDA e MPO também foram significativamente mais elevados e os níveis de SOD, CAT, GPx e GSH foram significativamente mais baixos no grupo vareniclina crônico, quando comparado ao grupo vareniclina agudo. Estes achados também foram confirmados por observações histopatológicas. Conclusões Este é o primeiro estudo que avaliou os efeitos pulmonares da vareniclina experimentalmente em um modelo animal. Observamos que o tratamento crônico da vareniclina causa inflamação e lesão pulmonar.

Peripheral nerve blocks are commonly preferred worldwide for the purposes of anesthesia application and postoperative analgesia. In this study, we investigated the effects of phenytoin which has a similar mechanism to local anesthetics in terms of the duration of analgesia and quality. The study was performed on 32 Sprague-Dawley male rats. Rats were randomly grouped into 4 groups. Group S: Sham group (n: 8); 0,2 ml saline perineural unilateral sciatic nerve. Group L: Perineural levobupivacaine (0,2 ml 0,5% levobupivacaine, n: 8); Group Ph: Perineural phenytoin (0,2 ml 62,5 mg / kg, n: 8); Group L + PH: Perineural phenytoin and levobupivacaine (0,2 ml 0,5% levobupivacaine + 62,5 mg / kg phenytoin, n: 8). Hot-plate and tail- flick tests were performed to measure acute thermal pain and histological changes were evaluated. The latency time at 30 minute in Group L+Ph were significantly increased when compared to the other groups during evaluation of the hot plate test. There was a significant difference in terms of latency time at 30 minute in Group L+Ph in the Tail Flick test and the latency time in Group L+Ph was longer when compared to the other groups (p [Med-Science 2018; 7(4.000): 891-7]

As aim of this study, the knowledge of whether vinpocetine (VINPO) is cardioprotective or not following isoprotenol (ISO) induced cardiac ischemia in rats. In myocard infarction, the one of the responsible mechanisms of injury is oxidative damage and inflammmation. The effect of Vinpo which is the potent antioxidant and anti inflammatory agent aimed to reveal as the biochemical, electrophysiological, and histopathologic parameters. In this study, thirty- two Wistar-Albino male rats (in the estimated weight of 250-400gm.) were divided into four groups, each consisting of eight rats. The group 1 named as sham, no any drug used in this group. Group 2 named as iso group, only isoprotenol was adminestered, the group 3 named as vinpo and iso group, here initially vinpocetine then isopretenol were used, and the group 4 named as iso and vinpo, here initially isoprotenol then vinpocetine were used. For the rats in group 2, in first and second day isoprotenol adminestered at a dose of 120mg/kg using an intraperitoneal injection. At third and fourth day no any drug used. And at fifth day the experiment terminated. For the rats in group 3, at the first and second day Vinponcetine adminestered at adose of 20 mg/kg using an intraperitoneal injection, after 30 minute isoprotenol administered at adose of 120mg/kg using an intraperitoneal injection. At third and fourth day no any drug was used. And at fifth day experiment was terminated. Lastly for the rats in group4, first and second day isoprotenol administered at adose of 120mg/kg using an intraperitoneal injection, at third and fourth day vinpocetine adminestered at adose of 20 mg/kg using an intraperitoneal injection, and at fifth day experiment terminated. Prior to termination of experiment the pulse rate and ECG changes were recorded. After completion of experiment protocols blood samples and cardiac tissue samples were recieved. For the knowledge of effects of vinpocetine experimental miyocardial ischemia induced in rats, and the serum myoglobulin, total creatine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanin aminotransferase (ALT), measured. In myocardial tissue as an antioxidative system and an oxidative stres markers; malonylaldehate (MDA), Superoxidedismutase (SOD), Catalase (CAT), Glutathionperoxidase (GPx), GSH, Total oxidant status (TOS),Total antioxidant status(TAS),Oxidative stres index (OSI) studied. We investigated the effects of acute high doses of acetylsalicylic acid (ASA) on liver tissue and the protective and therapeutic effects of melatonin on ASA related damage. Forty rats were randomly divided into five groups of eight: group 1, control; group 2, administered 200 mg/kg ASA; group 3, administered 5 mg/kg melatonin 45 min before ASA; group 4, administered 5 mg/kg melatonin 45 min after ASA; group 5, administered 5 mg/kg melatonin. We measured malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH), myeloperoxidase (MPO) aspartate aminotransferase (AST) and alanine transaminase (ALT) in the liver. ASA treatment significantly increased MDA and MPO production, whereas it significantly decreased levels of SOD, CAT, GPx and GSH in the liver. Melatonin significantly decreased MDA and MPO production, whereas it caused increased levels of antioxidants. AST and ALT levels were higher after ASA treatment, whereas these levels were reduced significantly after melatonin administration. Our histopathological findings, including apoptosis, were consistent with the biochemical results. Melatonin exhibits beneficial effects against high dose ASA induced hepatotoxicity. This study was aimed to investigate possible harmful effects of formaldehyde (FA) exposure on the morphology of fetus liver and kidney development during pregnancy and also to determinate possible protective role of chrysin (CH) against these harmful effects. For this aim, after pregnancy was induced, 58 female rats were divided into 6 groups. Serum physiologic (SF) was injected to the Group I rats intraperitoneally (i.p.). 20 mg/kg CH was given to the Group II via gavage. 0.1 mg/kg FA was applied to the Group III (i.p.), 1 mg/kg FA was injected to Group IV (i.p.) 0.1 mg/kg FA was given to Group V i.p., and 20 mg/kg CH was given to the same group via gavage. 1 mg/kg FA was applied to Group VI i.p., and 20 mg/kg CH was given to the same group via gavage. Fetuses were taken from each pregnant rat with cesarean section on the 20th day of the pregnancy. The morphological analyses of the fetuses, liver and kidney; biochemical and histological analyses of the liver and kidney were performed. The fetal body, liver and kidney weight of the FA groups demonstrated a statistically significant decrease the compared to control group. Also the FA-1 group were observed histopathological changes on the fetus liver and kidneys. FA exposure causes harmful effects on fetus the liver and kidneys. CH reduces the negative effect on morphological variables statistically. Although CH is insufficient to fix the histopathological changes that occur in the liver, damaging effects that occur in the kidney decreased statistically. [Med-Science 2017; 6(1.000): 73-80]

The aim of this study was to investigate the possible protective effects of melatonin and β-D-glucan against AA-induced liver injury in rats. Forty (SpraqueDawley male) rats were randomly divided into 5 experimental groups: sham, acetaminophen only (AA, 900 mg/kg), melatonin (10 mg/kg) + AA (MLT), β-D-glucan (50 mg/kg) + AA (β), and melatonin + β-D-glucan + AA (MLT+β) groups. All of the rats were killed on day 11 of the experiment. Histopathological changes and biochemical parameters including levels of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) and liver tissue malondialdehyde (MDA), activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were determined to assess the liver function. MDA levels were the highest in the AA group whereas activities of SOD, CAT, and GPx in the liver tissue were found as lowest in this group. MDA activities were significantly lower in the MLT+β group than in the AA group. Only GPx activities in the MLT+β group were significantly higher than those in the MLT and β groups. The serum AST and ALT levels were increased significantly following treatment with AA (p < 0.001). Pretreatment with the antioxidant compounds decreased AST levels significantly but again, the levels were still significantly higher than the sham levels (p < 0.001). There were no statistically significant differences in the microscopic damage between the S, MLT, β, and MLT+β groups (p > 0.05). This finding can be attributed to the higher efficacy of the combination of melatonin and β-D-glucan in scavenging free radicals and stimulating the antioxidant enzymes. The effects of montelukast against methotrexate-induced liver damage were investigated. 35 Wistar albino female rats were divided into 5 groups as follows: group I: control; group II: montelukast (ML); group III: methotrexate (Mtx); group IV: montelukast treatment after methotrexate application (Mtx + ML); group V: montelukast treatment before methotrexate application (ML + Mtx). At the end of the experiment, the liver tissues of rats were removed. Malondialdehyde (MDA), myeloperoxidase (MPO), and reduced glutathione levels were determined from liver tissues. In addition, the liver tissues were examined histologically. MDA and MPO levels of Mtx group were significantly increased when compared to control group. In Mtx + ML group, these parameters were decreased as compared to Mtx group. Mtx injection exhibited major histological alterations such as eosinophilic staining and swelling of hepatocytes. The glycogen storage in hepatocytes was observed as decreased by periodic acid schiff staining in Mtx group as compared to controls. ML treatment did not completely ameliorate the lesions and milder degenerative alterations as loss of the glycogen content was still present. It was showed that montelukast treatment after methotrexate application could reduce methotrexate-induced experimental liver damage.

Objective: Sepsis causes the release of free oxygen radicals that disrupt membrane integrity, and damage to Mitochondria due to the production of free oxygen radicals and oxidation leads to exacerbation of sepsis, Cecal ligation and puncture (CLP) in rats mimics the characteristics and course of clinical sepsis (Hubbard et al., 2005). Methods: We evaluated the antioxidant effects of Petroselinum crispum (Pc) in a cecal ligation and puncture (CLP)-induced rat sepsis model, in a rat model of sepsis caused by cecal ligation and puncture (CLP). Wistar albino rats were separated into four groups of eight groups: a sham group with incised and sutured abdomens; a Pc extract (PcE) group, which was given 2 g/kg parsley extract for 14 days by gastric gavage; a CLP group, which was subjected to sepsis caused by CLP; and a PcE + CLP group, which was given parsley extract for 14 days. PcE was given for 14 days, after which sepsis was induced via the CLP procedure. The groups were compared in terms of hemogram, biochemical and histological characteristics. Results: The administration of PcE before CLP-induced sepsis increases neutrophil counts, PLTs and TASs, which decrease with sepsis, and decreases biochemical changes (BUN, AST, ALT, LDH, TOS, and OSI), which increase with sepsis, to protect against sepsis. Compared with that in the CLP group, the severity of intestinal infiltration was significantly lower in the PcE + CLP group; however, the degree of epithelial damage in the PcE + CLP group was similar to that in the CLP group. In the PcE + CLP group, the crypt and villus lengths were greater, and the decrease in Paneth cell degranulation intensity was greater than that in the CLP group. Conclusion: Additionally, the morphology of the cells in the PcE + CLP group was similar to that in the sham group. PcE has potential as a prophylactic agent for sepsis.

Objective: Amikacin (AK) is a wide-spectrum antibiotic routinely used to treat gram-negative and some gram-positive bacterial infections. However, its use is limited due to its potential to cause nephrotoxicity due to an increase in reactive oxygen radicals. The main goal of this study was to investigate the effect of carvacrol (CAR) on AK-induced nephrotoxicity in rats. Methods: Thirty-two Sprague Dawley rats were randomly separated into four groups: the control (0.9% NaCl solution and sunflower oil), AK (400 mg/kg), CAR+AK (80 mg/kg CAR+400 mg/kg AK), and AK+CAR (400 mg/kg AK+80 mg/kg CAR) groups. AK and CAR were administered intramuscularly and orally, respectively for 7 days. Blood and kidney tissue samples were collected at the end of the experiment. The level of catalase, superoxide dismutase, malondialdehyde, and reduced glutathione, which are parameters of oxidative stress, were detected while comparing renal function and histopathological changes. Results: Histopathological findings (necrotic changes, dilatation and inflammatory cell infiltration) were significantly greater in the AK group than in the control group. Objective: To explore the effects of Nigella sativa oil (NSO) on the histopathological and biochemical changes that inhaled formaldehyde (FA) induces on the testicular tissue of rats. Methods: Thirty three adult male rats were separated into five groups as follows: C, the control group; 4FA group which received FA for 4 weeks; 13FA group which was given FA for 13 weeks; 4FA+NSO group which was administered FA plus NSO for 4 weeks; 13FA+NSO group which was treated with FA plus NSO for 13 weeks. FA was administered through inhalation for 8 h 5 days a week at a dose of 5 ppm in a special glass cage, and NSO was administered orally 1 mL/kg once daily. Rats were decapitated at the end of the experiment and testicular tissue specimens were harvested for histopathologic and biochemical assessment. Results: Compared to the C group, reduction was observed in the number of intact tubules and in the mean germinative epithelium thickness of the FA groups. Abstract During restoration of blood flow of the ischemic heart induced by coronary occlusion, free radicals cause lipid peroxidation with myocardial injury. Lipid peroxidation end-products, such as malondialdehyde (MDA), have been used to assess oxygen free radical-mediated injury of the ischemic-reperfused (I/R) myocardium in rats. This experimental study assessed the preventive effect of caffeic acid phenthyl ester (CAPE), antioxidant, on I/R-induced lipid peroxidation in the rat heart. We are also interested in the role of CAPE on glutathione (GSH) levels, an antioxidant whose levels are influenced by oxidative stress. I/R leads to the depletion of GSH which is the major intracellular nonprotein sulphydryl and plays an important role in the maintenance of cellular proteins and lipid in their functional state and acts primarily to protect these important structures against the threat of oxidation. In addition, we also examined morphologic changes in the heart by using light microscopy. The left coronary artery was occluded for 30 min and then reperfused for 120 min more before the experiment was terminated. CAPE (50 µM kg-1) was administered 10 min prior to ischemia and during occlusion by infusion. At the end of the reperfusion period, rats were sacrificed, and the heart was quickly removed for biochemical determination and histopathological analysis. I/R was accompanied by a significant increase in MDA production and decrease in GSH content in the rat heart. Administration of CAPE reduced MDA production and prevented depletion of GSH content. These beneficial changes in these biochemical parameters were also associated with parallel changes in histopathological appearance. These findings imply that I/R plays a causal role in heart injury due to overproduction of oxygen radicals or insufficient antioxidant and CAPE exert cardioprotective effects probably by the radical scavenging and antioxidant activities.
