Abstract Plants with the C4 photosynthesis pathway typically respond to climate change differently from more common C3-type plants, due to their distinct anatomical and biochemical characteristics. These different responses are expected to drive changes in global C4 and C3 vegetation distributions. However, current C4 vegetation distribution models may not predict this response as they do not capture multiple interacting factors and often lack observational constraints. Here, we used global observations of plant photosynthetic pathways, satellite remote sensing, and photosynthetic optimality theory to produce an observation-constrained global map of C4 vegetation. We find that global C4 vegetation coverage decreased from 17.7% to 17.1% of the land surface during 2001 to 2019. This was the net result of a reduction in C4 natural grass cover due to elevated CO2 favoring C3-type photosynthesis, and an increase in C4 crop cover, mainly from corn (maize) expansion. Using an emergent constraint approach, we estimated that C4 vegetation contributed 19.5% of global photosynthetic carbon assimilation, a value within the range of previous estimates (18–23%) but higher than the ensemble mean of dynamic global vegetation models (14 ± 13%; mean ± one standard deviation). Our study sheds insight on the critical and underappreciated role of C4 plants in the contemporary global carbon cycle.

Abstract Winter cover crops represent a land cover change that may sequester carbon in the soil and improve agricultural sustainability. Their adoption may also change the Earth's radiative balance and result in biophysical feedbacks to climate through alterations in albedo and latent heat fluxes. Understanding the mechanisms underlying these alterations to the radiative balance is important for making reliable future climate projections. However, data on cover crop biophysics are limited, requiring models to rely on data from summer plants for parameterization, likely biasing predictions. To address this gap, we measured the albedo and stomatal conductance of two summer crops and three winter crops on farms in the High Plains region of Texas. We also established a winter cover crop field experiment with two cover crops and fallow fields to estimate the change in albedo and latent heat flux that results from a switch to winter cover cropping. We found that albedo was significantly higher in winter‐like conditions than in summer‐like conditions due to an increase in plant albedo and a reduction in leaf area index. The albedo of winter cover crops was higher than the soil albedo, resulting in an increase in top‐of‐atmosphere reflected radiation of 7%–14% when converting from fallow fields to winter cover cropped fields. There was an additional cooling effect through doubling of the estimated latent heat flux caused by the presence of cover crops. The combined changes in albedo and latent heat resulted in a change in the surface energy balance that is associated with an overall cooling effect of winter cover crops on surface atmospheric temperatures. While this effect is likely to be region‐specific, these results strongly indicate that winter cover crops alter the surface albedo and latent heat flux of agricultural fields and provide a direct cooling effect in the High Plains region of Texas.

Abstract Climate change, interacting with and exacerbating anthropogenic modifications to the landscape, is altering ecosystem structure and function, biodiversity, and species distributions. Among the most visible short‐term impacts are the altered ecological roles of foundation species—those species, native or non‐native—that create locally stable environmental conditions and strongly influence ecosystem services. Understanding the future of these species is crucial for projecting impacts on ecosystem services at both local and regional scales. Here we present foundation species by ecoregion study cases across the US South‐Central Region (Louisiana, New Mexico, Oklahoma, and Texas), including C4 grasses, mesquite, and northern bobwhite in the Southern Great Plains, mangroves and nutria in coastal Louisiana wetlands, tiger salamanders and sandhill cranes in wetlands of the Southern Great Plains, and post and blackjack oaks and eastern redcedar in the Cross Timbers ecoregion. These case studies explore the impacts of climate change on foundation species and the consequences for ecosystem services, the outlook for climate adaptation efforts, and the sustainability of restoration in these systems. We underscore risks and vulnerabilities that stakeholders should consider when managing or restoring natural resources and conserving ecosystem services in an increasingly extreme and variable climate. We show that past management, through a lack of understanding or implementation of actions, has exacerbated shifts in invasive species, resulting in significant changes in ecosystem structure and function. These changes, interacting with landscape fragmentation and shifting land use and exacerbated by climate change, can result in critical losses of biodiversity. Unfortunately, lack of public understanding may hinder political support for restoration efforts and climate adaptation strategies crucial for the continued supply of traditional ecosystem services. Furthermore, the resulting invaded systems may provide opportunities for income via new ecosystem services valued by society that may reduce support for restoration to historical baselines, thus further shifting management priorities. These priorities should be informed by an understanding of past and ongoing ecological trends in region‐specific situations, such as those we present, to highlight the immediacy of climate change impacts on the environment and society and provide evidence for the critical nature of informed management decisions.

The fraction of leaf nitrogen allocated to RuBisCO indicates differing nitrogen use strategies of plants and varies considerably. Here the authors show that this variation is largely driven by leaf thickness and phosphorus content with light intensity, atmospheric dryness and soil pH also having considerable influence.

Abstract Empirical studies have shown that plant photosynthetic responses to environmental change can vary over time due to acclimation, but acclimation responses are often not included in Earth System Models. Photosynthetic least cost theory can be used to develop models of photosynthetic acclimation that are simple and testable. The theory is based on the idea that plants will acclimate to minimize the ratio of carbon costs to photosynthetic assimilation rate (Prentice et al., 2014, https://doi.org/10.1111/ele.12211). Formulations of this theory have been developed for C3 plants, but not C4 plants, which account for over 20% of global photosynthesis and are over‐represented among widely grown crops. Here, we use photosynthetic least cost theory to derive a model for C4 photosynthetic acclimation to above‐ground abiotic conditions. We then compare our model's responses to a similar model of C3 photosynthetic acclimation and find that C4 photosynthesis has the highest simulated advantage over C3 photosynthesis in hot, dry, and low CO2 environments. We find that this advantage predicts C4 abundance globally, but that the shallower CO2 response of C4 as compared to C3 photosynthesis will reduce C4 plant competitiveness under future conditions, despite higher temperatures. We also show that an acclimated model predicts similar or faster rates of C4 under all conditions than a model that does not consider acclimation, suggesting that Earth System Models (ESMs) are underestimating future C4 carbon uptake by not including acclimation. Our model is designed for easy incorporation into such ESMs.

Abstract Background Breast cancer is the most common invasive cancer among women worldwide. Next-generation sequencing (NGS) has revolutionized the study of cancer across research labs around the globe; however, genomic testing in clinical settings remains limited. Advances in sequencing reliability, pipeline analysis, accumulation of relevant data, and the reduction of costs are rapidly increasing the feasibility of NGS-based clinical decision making. Methods We report the development of MammaSeq, a breast cancer-specific NGS panel, targeting 79 genes and 1369 mutations, optimized for use in primary and metastatic breast cancer. To validate the panel, 46 solid tumors and 14 plasma circulating tumor DNA (ctDNA) samples were sequenced to a mean depth of 2311× and 1820×, respectively. Variants were called using Ion Torrent Suite 4.0 and annotated with cravat CHASM. CNVKit was used to call copy number variants in the solid tumor cohort. The oncoKB Precision Oncology Database was used to identify clinically actionable variants. Droplet digital PCR was used to validate select ctDNA mutations. Results In cohorts of 46 solid tumors and 14 ctDNA samples from patients with advanced breast cancer, we identified 592 and 43 protein-coding mutations. Mutations per sample in the solid tumor cohort ranged from 1 to 128 (median 3), and the ctDNA cohort ranged from 0 to 26 (median 2.5). Copy number analysis in the solid tumor cohort identified 46 amplifications and 35 deletions. We identified 26 clinically actionable variants (levels 1–3) annotated by OncoKB, distributed across 20 out of 46 cases (40%), in the solid tumor cohort. Allele frequencies of ESR1 and FOXA1 mutations correlated with CA.27.29 levels in patient-matched blood draws. Conclusions In solid tumor biopsies and ctDNA, MammaSeq detects clinically actionable mutations (OncoKB levels 1–3) in 22/46 (48%) solid tumors and in 4/14 (29%) of ctDNA samples. MammaSeq is a targeted panel suitable for clinically actionable mutation detection in breast cancer.

Climate change and land management are altering forest fire frequency and intensity worldwide. In some Northeast U.S. forests, pitch pine (Pinus rigida Miller) is not suffering from presence but rather a lack of wildfire events. In their absence, prescribed fire is being used to diminish fuel loads, open canopies and reduce competition. Pyrogenic carbon (PyC) produced by the fires may also improve soil moisture retention and plant physiological processes. Where the application of prescribed fire is not feasible due to nearby human populations, we reason prescribed fire PyC could be replaced by anthropogenic PyC product to provide similar soil benefits. We tested this hypothesis with pitch pine seedlings at a site absent overstory planted in submerged tree pots with control and PyC-imbued soils. Investigators found anthropogenic and forest PyC fostered similar growth, soil moisture retention and photosynthetic intrinsic water use efficiency, both significantly higher than unamended soils. We conclude anthropogenic subsurface PyC soil amendment provides a conservation management tool for enhancing benefits in ecosystems where prescribed fire is not a viable option in northerly forests in the U.S.

Abstract Photosynthetic temperature acclimation is a commonly observed process that is increasingly being incorporated into Earth System Models (ESMs). While short‐term acclimation has been shown to increase carbon storage in the future, it is uncertain whether acclimation will directly influence simulated future climate through biophysical mechanisms. Here, we used coupled atmosphere‐biosphere simulations using the Community Earth System Model (CESM) to assess how acclimation‐induced changes in photosynthesis influence global climate under present‐day and future (RCP 8.5) conditions. We ran four 30 year simulations that differed only in sea surface temperatures and atmospheric CO2 (present or future) and whether a mechanism for photosynthetic temperature acclimation was included (yes or no). Acclimation increased future photosynthesis and, consequently, the proportion of energy returned to the atmosphere as latent heat, resulting in reduced surface air temperatures in areas and seasons where acclimation caused the biggest increase in photosynthesis. However, this was partially offset by temperature increases elsewhere, resulting in a small, but significant, global cooling of 0.05°C in the future, similar to that expected from acclimation‐induced increases in future land carbon storage found in previous studies. In the present‐day simulations, the photosynthetic response was not as strong and cooling in highly vegetated regions was less than warming elsewhere, leading to a net global increase in temperatures of 0.04°C. Precipitation responses were variable and rates did not change globally in either time period. These results, combined with carbon‐cycle effects, suggest that models without acclimation may be overestimating positive feedbacks between climate and the land surface in the future.
