The number of low-income families in Singapore is increasing. Young children from impoverished backgrounds are at risk of development gaps and challenges. Research has shown that the accumulation of risk factors from adverse childhood experiences can lead to weaker outcomes later in life. The NTUC First Campus (NFC) launched the Child Support Model (CSM) to serve children and families from vulnerable, low-income backgrounds with a range of financial, social and learning support. A 3-year evaluation study using mixed-methods design was conducted to understand the effects of the CSM and the active ingredients in CSM. A cohort of 58 children from families making a monthly income of SGD$3,500 or less participated in the quantitative component where children's language and cognitive outcomes and classroom engagement were measured at three time points in 2019 (Kindergarten 1) and 2020 (Kindergarten 2). Participating children's parents and teachers were interviewed in the qualitative component and reported their perspectives and experiences in CSM.

The "Pre-K Classroom Assessment Scoring System" (CLASS) is commonly used to measure three domains of teacher-child interactional quality in preschool classrooms (Emotional Support, Classroom Organization, and Instructional Support). However, there is considerable debate regarding the validity of its three-domain factor structure and the applicability of CLASS across different countries and educational settings. This study contributes to this debate with an investigation of the factor structure of the CLASS in Singapore, comparing the factor structure for preschool classrooms serving typically developing children (n = 184) and early intervention classrooms serving children who require early intervention support (n = 182). Bifactor measurement models were used to explore the dimensionality of the CLASS in these settings. A best-fitting bifactor model with one general (Responsive Teaching) and two specific factors (Proactive Management and Routines, Cognitive Facilitation) was a good fit for the overall data. Comparison of preschool and early intervention classrooms indicated partial metric invariance, suggesting that while the factor structure was equivalent across the different settings, the loadings on some dimensions differed between preschool and early intervention classrooms. Future research could examine how different aspects of interactions in different settings contribute to children's learning.

As a result of globalization, kindergarten curriculum frameworks in Asia have been strongly influenced by Western theories, pedagogies, and values. In this article, we argue that Singapore's "Nurturing Early Learners" and Hong Kong's "Kindergarten Education Curriculum Guide" present key notions that are inconsistent with cultural values that are deeply rooted in these two societies. To overcome the challenges these inconsistencies trigger for teachers, principals, teacher educators, and parents, we advocate for the design of 'glocal' (global/local, explicitly hybrid) curriculum frameworks, based on principles that are culturally appropriate and socially situated. Drawing on recent research studies, we analyze current curriculum/practice gaps in relation to the notions of "Child-Centeredness," "Quality Interactions," "Creativity and Self-Expression," and "Play." In seeking the global/local balance that is needed in Singapore and Hong Kong, four alternative glocal notions are proposed: "Child-Appropriateness," "Pedagogical Quality," "Arts Engagement," and "Child-Led Activities." We conclude there is an urgent need for generating a solid corpus of local research in both jurisdictions, which should guide subsequent curriculum reforms and teacher preparation models. Our final aim is to contribute to early childhood education policy discussions in Asia, against the background of internationalization.

Research shows that executive functions (EF) and fine motor skills (FMS) contribute to early academic skills, possibly in overlapping ways. We examine whether and how EF and FMS interact in the concurrent prediction of maths, reading and spelling skills at the start of kindergarten. Structural Equation Modelling (SEM) on data from 1248 five-year-olds supports a compensatory account of EF and FMS in contributing towards maths and spelling skills. Controlling for socio-economic status, age, time spent in kindergarten and intelligence, the influence of EF on spelling achievement was larger for children with poorer compared to better FMS, and vice versa; FMS significantly predicted maths achievement only in children with high but not low EF, and vice versa. Identifying EF or FMS difficulties at or before the start of kindergarten may be important. Different approaches to intervention involving EF and FMS may be appropriate for maths versus spelling skills. We suggest for early childhood curricula to enhance opportunities for FMS development, especially for children who enter kindergarten with poor FMS. [Translation from English by Mercè Rius.]

The current study explored the home and school factors that are important for Singaporean pre-schoolers' (N = 1440) early English development from K1 to P1. Language input quantity and quality factors at home (e.g., number of English books,) and at school (e.g., instructional support in class) were used to predict children's English vocabulary and word reading development using linear mixed-effects models. Children's internal factors (e.g., non-verbal reasoning) were controlled. The results revealed that children who had more English input and higher input quality at home demonstrated faster receptive vocabulary growth. Children's word reading skill was more influenced by the English input quality at home. Children who were frequently read to in English demonstrated faster development in word reading competence. Mother's educational level also mattered: higher maternal education was associated with faster growth in reading skill. The length of time children stayed at school and teachers' qualification also significantly and positively associated with children's English learning. These findings indicate the substantial contribution of the home literacy environment to Singaporean children's early English language development, compared to the quality of teacher-child interactions in preschool. The necessity of more parental involvement in early English language learning at home is discussed under the realm of usage-based theory.

The early childhood sector in Singapore has witnessed vast changes in the past two decades. One of the key policy aims is to improve classroom quality. To ensure a rigorous evaluation of the quality of early childhood environments in Singapore, it is important to determine whether commonly used assessments of quality are valid indicators across different national and community contexts. This study investigated the validity of the Early Childhood Environment Rating Scale-Revised (ECERS-R) in Singapore focused on three major aspects: response process, structural, and criterion validity. Data were collected from 108 classrooms of the main providers of kindergarten programs in Singapore. Initial analyses showed that the quality indicators in most of the ECERS-R items were disordered, which indicated problems with the hypothesized response process underlying the standard ECERS-R scoring system in which high-level indicators are only rated if low-level indicators are met (stop-scoring). To deal with the problem of the "stop-scoring" method, we used the data from a full-scoring method (in which every indicator on every item was rated) in subsequent factor analyses to investigate the internal structure of the ECERS-R. Twenty-two of the 34 items were retained and found to load on two factors, one relating to activities/materials and the other related to language/interactions. Both of the identified subscales and the combined scale showed good internal consistency. The factor score on the combined scale mirrored the ECERS-R full score and could discriminate between classrooms in the top and bottom quartiles of quality scores. Findings on the relationship between the identified factors to the teacher-child ratio and to an alternative measure of classroom quality provided further evidence for criterion validity of the ECERS-R. Implications for the future development and use of the ECERS-R are discussed.

Mesenchymal stromal cells (MSCs) are promising therapeutic agents for cartilage regeneration, including the potential of cells to promote chondrogenesis in vivo. However, process development and regulatory approval of MSCs as cell therapy products benefit from facile in vitro approaches that can predict potency for a given production run. Current standard in vitro approaches include a 21 day 3D differentiation assay followed by quantification of cartilage matrix proteins. We propose a novel biophysical marker that is cell population-based and can be measured from in vitro monolayer culture of MSCs. We hypothesized that the self-assembly pattern that emerges from collective-cell behavior would predict chondrogenesis motivated by our observation that certain features in this pattern, namely, topological defects, corresponded to mesenchymal condensations. Indeed, we observed a strong predictive correlation between the degree-of-order of the pattern at day 9 of the monolayer culture and chondrogenic potential later estimated from in vitro 3D chondrogenic differentiation at day 21. These findings provide the rationale and the proof-of-concept for using self-assembly patterns to monitor chondrogenic commitment of cell populations. Such correlations across multiple MSC donors and production batches suggest that self-assembly patterns can be used as a candidate biophysical attribute to predict quality and efficacy for MSCs employed therapeutically for cartilage regeneration. Solving algebraic word problems involves multiple cognitive phases. The authors used a multitask approach to examine the extent to which working memory and executive functioning are associated with generating problem models and producing solutions. They tested 255 11-year-olds on working memory (Counting Recall, Letter Memory, and Keep Track), ability to inhibit inappropriate responses (inhibition: numeric Stroop, Stop Signal), mental flexibility (switching: Number-Letter and Plus-Minus), English literacy, and algebraic problem-solving skills (problem representation, solution generation, and other subcomponents). Working memory explained about a quarter of the variance in both representation and solution formation. Literacy explained an additional 20% of the variance in representation formation. Ability to discern quantitative relationships explained an additional 10%. The findings go beyond a demonstration of an association between working memory and problem-solving accuracy. They show that success in word problems is particularly reliant on ability to decode and assign mathematical operators to quantitative relationships, 2 phases of problem solving that also draw heavily on working memory resources. The current study examined the mediating role of stress in the linkages between attachment to parents and adjustment to attending higher learning institutions in another country. Self-reports from 114 Malaysian late adolescents studying in Singapore supported the primacy of stress for adjustment; more pertinent, stress mediated all the attachment-adjustment linkages that met the conditions for mediation testing, although this mediating role of stress applied mostly to the late adolescent girls and was also dependent on parent gender.

The behavioral variable employment-related motivational distortion (EMD) was defined and measured. In Study 1, a sample of 250 undergraduates completed the California Psychological Inventory (CPI), Form 434, on two separate occasions in which they responded (a) honestly and (b) as if applying for a job they valued. Four different change measures indexed participants response changes between the two conditions on a CPI-based scale measuring counter productivity (CPI-Cp). A composite of these four measures provided an EMD criterion, which enabled the development, in Study 2, of a 56-item CPI-based EMD predictor scale. On the basis of eight samples comprising a total Nof 2,074, internal consistency and test-retest reliability estimates for the CPI-EMD scale ranged between .72 and .86. Correlations with NEO PI-R domain and facet scales provided construct information about EMD. In Study 3, CPI-Cp scale scores adjusted for EMD manifested (a) reliability ranging from .71 to .87 over eight samples and (b) somewhat different correlations than for the unadjusted scale with Big Five personality scales. In addition, some evidence was found for gains in criterion correlations with EMD adjustments.

Introduction: Sex determination in eutherian mammals is controlled by the Y-linked gene Sry, which drives the formation of testes in male embryos. Despite extensive study, the genetic steps linking Sry action and male sex determination remain largely unknown. Here, we focused on Mmd2, a gene that encodes a member of the progestin and adipoQ receptor (PAQR) family. Mmd2 is expressed during the sex-determining period in XY but not XX gonads, suggesting a specific role in testis development. Methods: We used CRISPR to generate mouse strains deficient in Mmd2 and its 2 closely related PAQR family members, Mmd and Paqr8, which are also expressed during testis development. Following characterization of Mmd2 expression in the developing testis, we studied sex determination in embryos from single knockout as well as Mmd2;Mmd and Mmd2;Paqr8 double knockout lines using quantitative RT-PCR and immunofluorescence. Results: Analysis of knockout mice deficient in Sox9 and Nr5a1 revealed that Mmd2 operates downstream of these known sex-determining genes. However, fetal testis development progressed normally in Mmd2-null embryos. To determine if other genes might have compensated for the loss of Mmd2, we analyzed Paqr8 and Mmd-null embryos and confirmed that in both knockout lines, sex determination occurred normally. Finally, we generated Mmd2;Mmd and Mmd2;Paqr8 double-null embryos and again observed normal testis development. Discussion: These results may reflect functional redundancy among PAQR factors, or their dispensability in gonadal development. Our findings highlight the difficulties involved in identifying genes with a functional role in sex determination and gonadal development through expression screening and loss-of-function analyses of individual candidate genes and may help to explain the paucity of genes in which variations have been found to cause human disorders/differences of sex development. Background and Objectives: There is scarce data about the epidemiology, clinical features, investigations, diagnosis, treatment, and outcome in patients attending Singapore emergency departments (EDs) with nontraumatic headache. We sought to describe these characteristics of adult patients presenting to the ED with a primary complaint of headache. Materials and Methods: We performed a cross-sectional study on adult patients with nontraumatic headache over 4 consecutive weeks from 18 March 2019 to 14 April 2019 across four EDs in Singapore. Exclusion criteria were history of head trauma within 48 h of presentation, missing records, interhospital transfers, representation with the same headache as a recent previous visit and headache as an associated symptom. Results: During the study period, 579 patients (representing 1.8% of the total ED census) comprising 55.3% males and with a median age of 36 years presented to the four Singapore EDs with a primary complaint of nontraumatic headache. Paracetamol (41.5%), non-steroidal anti-inflammatory drugs (34.4%) and tramadol (31.5%) were the three commonest analgesics used either singly or in combination. Prochlorperazine (22.9%) and metoclopramide (17.4%) were frequent anti-emetic adjuncts. One-third of patients had computed tomography of the brain performed, which found abnormalities among 20.9% of them. ED diagnoses of primary headache conditions were made in 73.6% of patients. Conclusions: Primary headaches constituted most ED headache diagnoses. ED imaging of selected patients yielded a relatively high pick-up rate for significant intracranial abnormalities. Opioid use for symptomatic relief of headaches in the ED was found to be high, underscoring the need for improvement in headache analgesia relief practices in the ED. [ABSTRACT FROM AUTHOR]

Cultured meat (CM) has emerged as a breakthrough technology, which has the potential to minimise future negative impacts associated with the rapidly growing world population; such as serious environmental, sustainability, global public health, and animal welfare concerns. Recently numerous researchers in academia and companies have been putting significant efforts into scientific and translational development in this field. Since various pillars of CM manufacturing hold substantial translational potential, there has been a steady interest in filing patent applications globally in the past decade. For the first time, we have performed a patentometric analysis for cultured meat by highlighting the current patents landscape, determining prolific organisations and geographical locations active in the area, and identifying the overall outlook for the field. In total, > 190 patents published in the English language were collected from 1997 to 2020 for the analysis. Relevant data were extracted from the patents, including technology focus, country of origin, assignees, priority date, cooperative patent classification (CPC) code, to analyse the current patent landscape. Previous studies on individual differences in mathematical abilities have shown that working memory contributes to early arithmetic performance. In this study, we extended the investigation to algebraic word problem solving. A total of 151 10-year-olds were administered algebraic word problems and measures of working memory, intelligence quotient (IQ), and reading ability. Regression results were consistent with findings from the arithmetic literature showing that a literacy composite measure provided greater contribution than did executive function capacity. However, a series of path analyses showed that the overall contribution of executive function was comparable to that of literacy; the effect of executive function was mediated by that of literacy. Both the phonological loop and the visual spatial sketchpad failed to contribute directly; they contributed only indirectly by way of literacy and performance IQ, respectively.

SRY is the Y‐chromosomal gene that determines male sex development in humans and most other mammals. After three decades of study, we still lack a detailed understanding of which domains of the SRY protein are required to engage the pathway of gene activity leading to testis development. Some insight has been gained from the study of genetic variations underlying differences/disorders of sex determination (DSD), but the lack of a system of experimentally generating SRY mutations and studying their consequences in vivo has limited progress in the field. To address this issue, we generated a mouse model carrying a human SRY transgene able to drive testis determination in XX mice. Using CRISPR‐Cas9 gene editing, we generated novel genetic modifications in each of SRY's three domains (N‐terminal, HMG box, and C‐terminal) and performed a detailed analysis of their molecular and cellular effects on embryonic testis development. Our results provide new functional insights unique to human SRY and present a versatile and powerful system in which to functionally analyze variations of SRY including known and novel pathogenic variants found in DSD.
