Plant electrophysiology carries a strong potential for assessing the health of a plant. Current literature for the classification of plant electrophysiology generally comprises classical methods based on signal features that portray a simplification of the raw data and introduce a high computational cost. The Deep Learning (DL) techniques automatically learn the classification targets from the input data, overcoming the need for precalculated features. However, they are scarcely explored for identifying plant stress on electrophysiological recordings. This study applies DL techniques to the raw electrophysiological data from 16 tomato plants growing in typical production conditions to detect the presence of stress caused by a nitrogen deficiency. The proposed approach predicts the stressed state with an accuracy of around 88%, which could be increased to over 96% using a combination of the obtained prediction confidences. It outperforms the current state-of-the-art with over 8% higher accuracy and a potential for a direct application in production conditions. Moreover, the proposed approach demonstrates the ability to detect the presence of stress at its early stage. Automated monitoring of plant health is becoming a crucial component for optimizing agricultural production. Recently, several studies have shown that plant electrophysiology could be used as a tool to determine plant status related to applied stressors. However, to the best of our knowledge, there have been no studies relating electrical plant response to general stress responses as a proxy for plant health. This study models general stress of plants exposed to either biotic or abiotic stressors, namely drought, nutrient deficiencies or infestation with spider mites, using electrophysiological signals acquired from 36 plants. Moreover, in the signal processing procedure, the proposed workflow reuses information from the previous steps, therefore considerably reducing computation time regarding recent related approaches in the literature. Careful choice of the principal parameters leads to a classification of the general stress in plants with more than 80% accuracy. The main descriptive statistics measured together with the Hjorth complexity provide the most discriminative information for such classification. Herbivorous arthropods, such as spider mites, are one of the major causes of annual crop losses. They are usually hard to spot before a severe infestation takes place. When feeding, these insects cause external perturbation that triggers changes in the underlying physiological process of a plant, which are expressed by a generation of distinct variations of electrical potential. Therefore, plant electrophysiology data portray information of the plant state. Analyses involving machine learning techniques applied to plant electrical response triggered by spider mite infestation have not been previously reported. This study investigates plant electrophysiological signals recorded from 12 commercial tomatoes plants contaminated with spider mites and proposes a workflow based on Gradient Boosted Tree algorithm for an automated differentiation of the plant's normal state from the stressed state caused by infestation. The classification model built using the signal samples recorded during daylight and employing a reduced feature subset performs with an accuracy of 80% in identifying the plant's stressed state. Furthermore, the Hjorth complexity encloses the most relevant information for discrimination of the plant status. The obtained findings open novel access towards automated detection of insect infestation in greenhouse crops and, consequently, more optimal prevention and treatment approaches. Fluorine‐19 (19F) MRI of injected perfluorocarbon emulsions (PFCs) allows for the non‐invasive quantification of inflammation and cell tracking, but suffers from a low signal‐to‐noise ratio and extended scan time. To address this limitation, we tested the hypotheses that a 19F MRI pulse sequence that combines a specific undersampling regime with signal averaging has both increased sensitivity and robustness against motion artifacts compared with a non‐averaged fully sampled pulse sequence, when both datasets are reconstructed with compressed sensing. As a proof of principle, numerical simulations and phantom experiments were performed on selected variable ranges to characterize the point spread function of undersampling patterns, as well as the vulnerability to noise of undersampling and reconstruction parameters with paired numbers of x signal averages and acceleration factor x (NAx‐AFx). The numerical simulations demonstrated that a probability density function that uses 25% of the samples to fully sample the k‐space central area allowed for an optimal balance between limited blurring and artifact incoherence. At all investigated noise levels, the Dice similarity coefficient (DSC) strongly depended on the regularization parameters and acceleration factor. In phantoms, the motion robustness of an NA8‐AF8 undersampling pattern versus NA1‐AF1 was evaluated with simulated and real motion patterns. Differences were assessed with the DSC, which was consistently higher for the NA8‐AF8 compared with the NA1‐AF1 strategy, for both simulated and real cyclic motion patterns (P < 0.001). Both strategies were validated in vivo in mice (n = 2) injected with perfluoropolyether. Here, the images displayed a sharper delineation of the liver with the NA8‐AF8 strategy than with the NA1‐AF1 strategy. In conclusion, we validated the hypotheses that in 19F MRI the combination of undersampling and averaging improves both the sensitivity and the robustness against motion artifacts. The structural complexity of the thalamus, due to its mixed composition of gray and white matter, make it challenging to disjoint and quantify each tissue contribution to the thalamic anatomy. This work promotes the use of partial‐volume‐based over probabilistic‐based tissue segmentation approaches to better capture thalamic gray matter differences between patients at different stages of psychosis (early and chronic) and healthy controls. The study was performed on a cohort of 23 patients with schizophrenia, 41 with early psychosis and 69 age and sex‐matched healthy subjects. Six tissue segmentation approaches were employed to obtain the gray matter concentration/probability images. The statistical tests were applied at three different anatomical scales: whole thalamus, thalamic subregions and voxel‐wise. The results suggest that the partial volume model estimation of gray matter is more sensitive to detect atrophies within the thalamus of patients with psychosis. However all the methods detected gray matter deficit in the pulvinar, particularly in early stages of psychosis. This study demonstrates also that the gray matter decrease varies nonlinearly with age and between nuclei. While a gray matter loss was found in the pulvinar of patients in both stages of psychosis, reduced gray matter in the mediodorsal was only observed in early psychosis subjects. Finally, our analyses point to alterations in a sub‐region comprising the lateral posterior and ventral posterior nuclei. The obtained results reinforce the hypothesis that thalamic gray matter assessment is more reliable when the tissues segmentation method takes into account the partial volume effect. Purpose: The thalamus is an important brain structure and neurosurgical target, but its constituting nuclei are challenging to image non‐invasively. Recently, susceptibility‐weighted imaging (SWI) at ultra‐high field has shown promising capabilities for thalamic nuclei mapping. In this work, several methodological improvements were explored to enhance SWI quality and contrast, and specifically its ability for thalamic imaging. Methods: High‐resolution SWI was performed at 7T in healthy participants, and the following techniques were applied: (a) monitoring and retrospective correction of head motion and B0 perturbations using integrated MR navigators, (b) segmentation and removal of venous vessels on the SWI data using vessel enhancement filtering, and (c) contrast enhancement by tuning the parameters of the SWI phase‐magnitude combination. The resulting improvements were evaluated with quantitative metrics of image quality, and by comparison to anatomo‐histological thalamic atlases. Results: Even with sub‐millimeter motion and natural breathing, motion and field correction produced clear improvements in both magnitude and phase data quality (76% and 41%, respectively). The improvements were stronger in cases of larger motion/field deviations, mitigating the dependence of image quality on subject performance. Optimizing the SWI phase‐magnitude combination yielded substantial improvements in image contrast, particularly in the thalamus, well beyond previously reported SWI results. The atlas comparisons provided compelling evidence of anatomical correspondence between SWI features and several thalamic nuclei, for example, the ventral intermediate nucleus. Vein detection performed favorably inside the thalamus, and vein removal further improved visualization. Introduction: Drug-resistant essential tremor (ET) can benefit from open standard stereotactic procedures, such as deep-brain stimulation or radiofrequency thalamotomy. Non-surgical candidates can be offered either high-focused ultrasound (HIFU) or radiosurgery (RS). All procedures aim to target the same thalamic site, the ventro-intermediate nucleus (e.g., Vim). The mechanisms by which tremor stops after Vim RS or HIFU remain unknown. We used voxel-based morphometry (VBM) on pretherapeutic neuroimaging data and assessed which anatomical site would best correlate with tremor arrest 1 year after Vim RS.Methods: Fifty-two patients (30 male, 22 female; mean age 71.6 years, range 49–82) with right-sided ET benefited from left unilateral Vim RS in Marseille, France. Targeting was performed in a uniform manner, using 130 Gy and a single 4-mm collimator. Neurological (pretherapeutic and 1 year after) and neuroimaging (baseline) assessments were completed. Tremor score on the treated hand (TSTH) at 1 year after Vim RS was included in a statistical parametric mapping analysis of variance (ANOVA) model as a continuous variable with pretherapeutic neuroimaging data. Pretherapeutic gray matter density (GMD) was further correlated with TSTH improvement. No a priori hypothesis was used in the statistical model.Results: The only statistically significant region was right Brodmann area (BA) 18 (visual association area V2, p = 0.05, cluster size Kc = 71). Higher baseline GMD correlated with better TSTH improvement at 1 year after Vim RS (Spearman’s rank correlation coefficient = 0.002).Conclusions: Routine baseline structural neuroimaging predicts TSTH improvement 1 year after Vim RS. The relevant anatomical area is the right visual association cortex (BA 18, V2). The question whether visual areas should be included in the targeting remains open. [ABSTRACT FROM AUTHOR]

A 71-year-old male has been diagnosed with drug-resistant essential tremor (ET), in a familial context, with symptoms that started in childhood, at the age of 11. Tremor was predominant in the right upper limb, in a right-handed patient, provoking dramatic functional impairment. Due to a relative contra-indication to DBS (anticoagulation treatment for pulmonary embolism), an initial Gamma Knife (GK) thalamotomy of the left ventro-intermediate nucleus (Vim) was performed. After progressive and dramatic clinical alleviation, which persisted up to two years, the tremor relapsed. Magnetic resonance imaging (MRI) showed the presence of a small contrastenhancement (CE) surrounding a hypodense T1 necrotic core which, after co-registration with the dosimetry palnning in the Leksell GammaPlan, corresponded to the GK targeting. Additionally, the DWI data made possible the semi-automated segmentation of 7 clusters of thalamic nuclei, with the ventro-lateral ventral (VLV, nomenclature as from Morel et al.) cluster containing the contrast-enhancement area, as corresponding to the GKS target and in a ventral position, having anatomical relevance. A deep-brain stimulation (DBS) procedure was decided after multidisciplinary discussion. Due to the fact that the patient had bilateral tremor, and DBS could offer bilateral implantation without additional risks, it has been decided to treat both sides. For the left previously treated by GK, we aimed at stimulating the vicinity of the preexisting lesion. On this side, intraoperative microrecording showed that the center of the CE visualized lesion on MRI was silent (with no cells) and was surrounded by an area of normally active neurons; however, there was a clear difference in terms of potentials of action between the left (with previous GK) and the right (with no previous GK), raising the question whether new or surviving cells were present on the left. Additional data while comparing left and right tracks showed that neuronal noise, single unit amplitude and frequency were significantly higher on the non-GK treated side, suggesting that the cells on the previously treated left GK side were surviving and not new ones, different than those on the right. The patient had immediate and complete clinical alleviation after DBS. However, three days after the DBS procedure, he presented with dysarthria and left hemisyndrome. A CT scan and angio-CT showed right sylvian hypoperfusion, with right carotid artery thrombus. An MRI performed at 1.5T revealed an ischemia on the territory of the right anterior choroidal artery and progression of the carotid thrombus. The patient has been putted under anticoagulation treatment, with further disappearance of symptoms. Additionally, tremor on the left side remained suppressed after the stroke, with no need for stimulation on that specific side. Eighteen months latter, this important alleviation persisted bilaterally, by unilateral stimulation. Electrophysiological findings suggest functional reorganization at the periphery of the necrotic core visualized on the followup MRI after GK. Consequently, this peripheral area containing neuronal activity is most probably the one responding to DBS, although the former is not identhical on both sides, but with a final identhical clinical efect. Deep-brain stimulation after GK thalamotomy, aiming at a target closed to the previous lesion is possible and can lead to tremor suppression with durable effect. The lack of possibility to directly visualize the Ventro-intermediate thalamic nucleus (Vim) on current routine MRI lead to application of indirect targeting methods in stereotactic neurosurgical procedures, such as Gamma Knife Surgery (GKS) and Deep Brain Stimulation (DBS), for treatment of drug-resistant tremor. Recently, our group proposed an automated robust and reproducible method across healthy subjects and tremor patients for parceling the thalamus in seven main groups of nuclei while exploring the local diffusion information from the spherical harmonics representation of the orientation distribution functions (ODFs) in k-means clustering framework. One among the resulting parcel is the Ventral-Lateral-Ventral group (VLV) enclosing all motor-related nuclei including the Vim. We aim at further automatic subdivision the VLV cluster. To this end, within the VLV, we first build a k-nn graph with edges corresponding to the respective ODFs distances and then, we perform a partition in 3 sub-clusters using the Normalized Cut algorithm. The outcome of the proposed subdivision was compared to the radiological response in the follow-up images of 17 patients treated for tremor with GKS unilaterally. The MRI protocol included pre-operative diffusion-weighted images with 64 (or 72) gradient direction, b=1000s/mm2 and voxel size of 2.2x2.2x2.2mm3, pre-operative and post-operative MPRAGEs with isotropic resolution of ~1mm3, all of them acquired at field of 3T. In general, the sub-partition followed a spatial-distribution pattern and for 12 out of 17 cases we observed that one specific sub-cluster encloses entirely or the major part of the contrast enhancement corresponding to the GKS target that appeared on the follow-up images.The evaluation of the sub-partition outcome was further extended in one additional tremor patient treated with DBS, bilaterally. Similar pre-operative MRI data as for the GKS-treated patients was acquired. We observed that in each hemisphere both initial targeting point and the final position of the electrode are inside the anticipated sub-cluster. This study shows that the diffusion-MRI-based sub-clustering of the VLV thalamic nuclei could potentially allow, in an automated manner, to narrow the area of Vim's localization. Our preliminary results will be further investigated in a larger cohort of patients (treated by either GKS or DBS) and on diffusion images with higher spatial resolution. [ABSTRACT FROM AUTHOR]

Background: Tremor is a main feature of two different movement disorders, Parkinson's disease (PD) and essential tremor (ET), for which previously published data suggested functionally different underlying brain circuitry. Objectives: We analyzed resting-state fMRI data of 16 patients: 8 with tremor-dominant PD, and 8 with ET (mean age 74 years). Tremor was predominant in the right upper limb in all patients. In PD, 7 out of 8 were under Levodopa medication at the time of scanning, with a mean dose of 525 mg (range 0-800). Methods: The functional connectivity of the tremor network has been probed using a seed-to-voxel (further referred to as seed connectivity analysis) and ROI-wise (further referred to as network analysis) approach. Four seeds (dentate nucleus, red nucleus, thalamus, precentral gyrus) in both hemispheres were investigated. Results: We report exclusive negative functional connectivity (FC) during resting-state in ET between the right dentate and left red nucleus towards the left precentral gyrus. Furthermore, the same connectivity is consistently positive in PD (independent of the medication levels). Conclusion: The major differences in FC between two groups suggest an altered tremor network in PD and ET and further point toward the involvement of the putamen and globus pallidus (both internal and external) on one side, as well as the right ventral anterior thalamic nuclei, on the other side. In summary, our findings suggest that rs-fMRI could be used as an additional confirmation test, beside the clinical history and examination and the eventual adding of a DAT scan, in the differential diagnosis between PD and ET. Objective: To assess for the first time structural brain changes, by voxel-based morphometry (VBM), before and after unilateral Gamma Knife thalamotomy (GKT) for drug-resistant tremor. To identify differences between clinical responders and nonresponders to GKT. Methods: Thirty-eight patients (mean age 71.8 years) with severe refractory right essential tremor (ET) were treated with unilateral left GKT. Targeting of ventro-intermediate nucleus (Vim) was performed with Leksell Gamma Knife using a single 4- mm collimator and 130 Gy. Neurological, neuropsychological and neuroimaging (3 Tesla, including 3D T1 weighted) assessment had been done at baseline and 1 year after GKT. Clinical responders were considered those improved in tremor score (Fahn-Tolosa-Marin) with at least 45%. Results: Thirty-one (81.6%) patients were responders (R) and 7 (18.4%) non-responders (NR). With regard to GM changes after GKT, independently of clinical answer, atrophy was present in extensive areas (right globus pallidus, left putamen, left thalamus, right anterior and medio-dorsal thalamus, cerebellar, right premotor and supplementary motor area, left and right visual association cortex, right ventral temporal, left parahippocampal and posterior cingulate gyrus). The interaction between R - NR with time showed brain plasticity in R remote areas, within left temporal pole (BA 38) and cluster including left occipital cortex (BA 19), visual areas V4 and V5, parahippocampal place area (punc<0.005, k>120). Conclusions: Our results show brain plasticity after unilateral left GKT. Responders present changes in areas involved in motion, mainly locomotor monitoring towards the local and distant environment, suggesting the requirement to recruit in the targeting specific visuomotor networks. Objective: The tremor circuitry has commonly been hypothesized to be driven by one or multiple pacemakers within the cerebello-thalamo-cortical pathway, including the cerebellum, contralateral motor thalamus, and primary motor cortex. However, previous studies, using multiple methodologies, have advocated that tremor could be influenced by changes within the right extrastriate cortex, at both the structural and functional level. The purpose of this work was to evaluate the role of the extrastriate cortex in tremor generation and further arrest after left unilateral stereotactic radiosurgery thalamotomy (SRS-T)., Methods: The authors considered 12 healthy controls (HCs, group 1); 15 patients with essential tremor (ET, right-sided, drug-resistant; group 2) before left unilateral SRS-T; and the same 15 patients (group 3) 1 year after the intervention, to account for delayed effects. Blood oxygenation level-dependent functional MRI during resting state was used to characterize the dynamic interactions of the right extrastriate cortex, comparing HC subjects against patients with ET before and 1 year after SRS-T. In particular, the authors applied coactivation pattern analysis to extract recurring whole-brain spatial patterns of brain activity over time., Results: The authors found 3 different sets of coactivating regions within the right extrastriate cortex in HCs and patients with pretherapeutic ET, reminiscent of the "cerebello-visuo-motor," "thalamo-visuo-motor" (including the targeted thalamus), and "basal ganglia and extrastriate" networks. The occurrence of the first pattern was decreased in pretherapeutic ET compared to HCs, whereas the other two patterns showed increased occurrences. This suggests a misbalance between the more prominent cerebellar circuitry and the thalamo-visuo-motor and basal ganglia networks. Multiple regression analysis showed that pretherapeutic standard tremor scores negatively correlated with the increased occurrence of the thalamo-visuo-motor network, suggesting a compensatory pathophysiological trait. Clinical improvement after SRS-T was related to changes in occurrences of the basal ganglia and extrastriate cortex circuitry, which returned to HC values after the intervention, suggesting that the dynamics of the extrastriate cortex had a role in tremor generation and further arrest after the intervention., Conclusions: The data in this study point to a broader implication of the visual system in tremor generation, and not only through visual feedback, given its connections to the dorsal visual stream pathway and the cerebello-thalamo-cortical circuitry, with which its dynamic balance seems to be a crucial feature for reduced tremor. Furthermore, SRS-T seems to bring abnormal pretherapeutic connectivity of the extrastriate cortex to levels comparable to those of HC subjects.
