Abstract: Abstract The branching fraction of D + → K S 0 π 0 e + ν e $$ {D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e $$ is measured for the first time using 7.93 fb −1 of e + e − annihilation data collected at the center-of-mass energy s $$ \sqrt{s} $$ = 3.773 GeV with the BESIII detector operating at the BEPCII collider, and is determined to be B D + → K S 0 π 0 e + ν e = 0.881 ± 0.017 stat . ± 0.016 syst . % $$ \mathcal{B}\left({D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e\right)=\left(0.881\pm {0.017}_{\textrm{stat}.}\pm {0.016}_{\textrm{syst}.}\right)\% $$ . Based on an analysis of the D + → K S 0 π 0 e + ν e $$ {D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e $$ decay dynamics, we observe the S-wave and P-wave components with fractions of f S-wave = (6.13 ± 0.27stat. ± 0.30syst. )% and f K ¯ ∗ 892 0 = 93.88 ± 0.27 stat . ± 0.29 syst . % $$ {f}_{{\overline{K}}^{\ast }{(892)}^0}=\left(93.88\pm {0.27}_{\textrm{stat}.}\pm {0.29}_{\textrm{syst}.}\right)\% $$ , respectively. From these results, we obtain the branching fractions B D + → K S 0 π 0 S − wave e + ν e = 5.41 ± 0.35 stat . ± 0.37 syst . × 10 − 4 $$ \mathcal{B}\left({D}^{+}\to {\left({K}_S^0{\pi}^0\right)}_{S-\textrm{wave}}{e}^{+}{\nu}_e\right)=\left(5.41\pm {0.35}_{\textrm{stat}.}\pm {0.37}_{\textrm{syst}.}\right)\times {10}^{-4} $$ and B D + → K ¯ ∗ 892 0 e + ν e = 4.97 ± 0.11 stat . ± 0.12 syst . % $$ \mathcal{B}\left({D}^{+}\to {\overline{K}}^{\ast }{(892)}^0{e}^{+}{\nu}_e\right)=\left(4.97\pm {0.11}_{\textrm{stat}.}\pm {0.12}_{\textrm{syst}.}\right)\% $$ . In addition, the hadronic form-factor ratios of D + → K ¯ ∗ 892 0 e + ν e $$ {D}^{+}\to {\overline{K}}^{\ast }{(892)}^0{e}^{+}{\nu}_e $$ at q 2 = 0, assuming a single-pole dominance parameterization, are determined to be r V = V 0 A 1 0 = 1.43 ± 0.07 stat . ± 0.03 syst . $$ {r}_V=\frac{V(0)}{A_1(0)}=1.43\pm {0.07}_{\textrm{stat}.}\pm {0.03}_{\textrm{syst}.} $$ and r 2 = A 2 0 A 1 0 = 0.72 ± 0.06 stat . ± 0.02 syst . $$ {r}_2=\frac{A_2(0)}{A_1(0)}=0.72\pm {0.06}_{\textrm{stat}.}\pm {0.02}_{\textrm{syst}.} $$ .

Abstract: Abstract Using e + e − annihilation data sets corresponding to an integrated luminosity of 4.5 fb −1, collected with the BESIII detector at center-of-mass energies between 4.600 and 4.699 GeV, we report the first measurements of the absolute branching fractions B Λ c + → p K L 0 $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0\right) $$ = (1.67 ± 0.06 ± 0.04)%, B Λ c + → p K L 0 π + π − $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0{\pi}^{+}{\pi}^{-}\right) $$ = (1.69 ± 0.10 ± 0.05)%, and B Λ c + → p K L 0 π 0 $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0{\pi}^0\right) $$ = (2.02 ± 0.13 ± 0.05)%, where the first uncertainties are statistical and the second systematic. Combining with the known branching fractions of Λ c + → p K S 0 $$ {\Lambda}_c^{+}\to p{K}_S^0 $$ , Λ c + → p K S 0 π + π − $$ {\Lambda}_c^{+}\to p{K}_S^0{\pi}^{+}{\pi}^{-} $$ , and Λ c + → p K S 0 π 0 $$ {\Lambda}_c^{+}\to p{K}_S^0{\pi}^0 $$ , we present the first measurements of the K S 0 $$ {K}_S^0 $$ - K L 0 $$ {K}_L^0 $$ asymmetries R Λ c + K S , L 0 X = B Λ c + → K S 0 X − B Λ c + → K L 0 X B Λ c + → K S 0 X + B Λ c + → K L 0 X $$ R\left({\Lambda}_c^{+},{K}_{S,L}^0X\right)=\frac{\mathcal{B}\left({\Lambda}_c^{+}\to {K}_S^0X\right)-\mathcal{B}\left({\Lambda}_c^{+}\to {K}_L^0X\right)}{\mathcal{B}\left({\Lambda}_c^{+}\to {K}_S^0X\right)+\mathcal{B}\left({\Lambda}_c^{+}\to {K}_L^0X\right)} $$ in charmed baryon decays: R Λ c + p K S , L 0 = − 0.025 ± 0.031 $$ R\left({\Lambda}_c^{+},p{K}_{S,L}^0\right)=-0.025\pm 0.031 $$ , R Λ c + p K S , L 0 π + π − = − 0.027 ± 0.048 $$ R\left({\Lambda}_c^{+},p{K}_{S,L}^0{\pi}^{+}{\pi}^{-}\right)=-0.027\pm 0.048 $$ and R Λ c + p K S , L 0 π 0 = − 0.015 ± 0.046 $$ R\left({\Lambda}_c^{+},p{K}_{S,L}^0{\pi}^0\right)=-0.015\pm 0.046 $$ . No significant asymmetries with statistical significance are observed.

Abstract: Abstract The processes h c → γP (P = η′, η, π 0) are studied with a sample of (27.12 ± 0.14) × 108 ψ(3686) events collected by the BESIII detector at the BEPCII collider. The decay h c → γη is observed for the first time with the significance of 9.0 σ, and the branching fraction is determined to be (3.77 ± 0.55 ± 0.13 ± 0.26) × 10 −4, while B $$ \mathcal{B} $$ (h c → γη′) is measured to be (1.40 ± 0.11 ± 0.04 ± 0.10) × 10 −3, where the first uncertainties are statistical, the second systematic, and the third from the branching fraction of ψ(3686) → π 0 h c . The combination of these results allows for a precise determination of R h c = B h c → γη B h c → γ η ′ , $$ {R}_{h_c}=\frac{\mathcal{B}\left({h}_c\to {\pi}^0\gamma \eta \right)}{\mathcal{B}\left({h}_c\to {\pi}^0\gamma {\eta}^{\prime}\right)}, $$ which is calculated to be (27.0 ± 4.4 ± 1.0)%. The results are valuable for gaining a deeper understanding of η − η′ mixing, and its manifestation within quantum chromodynamics. No significant signal is found for the decay h c → γπ 0, and an upper limit is placed on its branching fraction of B $$ \mathcal{B} $$ (h c → γπ 0) < 5.0 × 10 −5, at the 90% confidence level.

Abstract: Abstract Using e + e − collision data collected with the BESIII detector at the BEPCII collider at center-of-mass energies between 3.510 and 4.914 GeV, corresponding to an integrated luminosity of 25 fb −1, we measure the Born cross sections for the process e + e − → K − Ξ ¯ + Λ / Σ 0 $$ {e}^{+}{e}^{-}\to {K}^{-}{\overline{\Xi}}^{+}\Lambda /{\Sigma}^0 $$ at thirty-five energy points with a partial-reconstruction strategy. By fitting the dressed cross sections of e + e − → K − Ξ ¯ + Λ / Σ 0 $$ {e}^{+}{e}^{-}\to {K}^{-}{\overline{\Xi}}^{+}\Lambda /{\Sigma}^0 $$ , evidence for ψ 4160 → K − Ξ ¯ + Λ $$ \psi (4160)\to {K}^{-}{\overline{\Xi}}^{+}\Lambda $$ is found for the first time with a significance of 4.4σ, including systematic uncertainties. No evidence for other possible resonances is found. In addition, the products of electronic partial width and branching fraction for all assumed resonances decaying into K − Ξ ¯ + Λ / Σ 0 $$ {K}^{-}{\overline{\Xi}}^{+}\Lambda /{\Sigma}^0 $$ are determined.

Abstract: Abstract Using e + e − collision data collected at the BESIII detector at center-of-mass energies between 4.128 and 4.226 GeV, corresponding to an integrated luminosity of 7.33 fb −1, we determine the absolute branching fractions of fifteen hadronic D s + $$ {D}_s^{+} $$ decays with a double-tag technique. In particular, we make precise measurements of the branching fractions B D s + → K + K − π + = 5.49 ± 0.04 ± 0.07 % $$ \mathcal{B}\left({D}_s^{+}\to {K}^{+}{K}^{-}{\pi}^{+}\right)=\left(5.49\pm 0.04\pm 0.07\right)\% $$ , B D s + → K S 0 K + = 1.50 ± 0.01 ± 0.01 % $$ \mathcal{B}\left({D}_s^{+}\to {K}_S^0{K}^{+}\right)=\left(1.50\pm 0.01\pm 0.01\right)\% $$ and B D s + → K + K − π + π 0 = 5.50 ± 0.05 ± 0.11 % $$ \mathcal{B}\left({D}_s^{+}\to {K}^{+}{K}^{-}{\pi}^{+}{\pi}^0\right)=\left(5.50\pm 0.05\pm 0.11\right)\% $$ , where the first uncertainties are statistical and the second ones are systematic. The CP asymmetries in these decays are also measured and all are found to be compatible with zero.

Abstract: Abstract Using (27.12 ± 0.14) × 108 ψ(3686) events collected by the BESIII detector at the center-of-mass energy of s $$ \sqrt{s} $$ = 3.686 GeV, we search for the first time for two nonleptonic hyperon decays that change strangeness by two units, Ω − → Σ0 π − and Ω − → nK − . No significant signal is observed. The upper limits on their decay branching fractions are determined to be B $$ \mathcal{B} $$ (Ω − → Σ0 π − ) < 5.4 × 10 −4 and B $$ \mathcal{B} $$ (Ω − → nK − ) < 2.4 × 10 −4 at the 90% confidence level.

Abstract: Abstract Using 24.1 fb −1 of e + e − collision data collected with the BESIII detector at the BEPCII collider, the Born cross sections and effective form factors of the e + e − → Σ + Σ ¯ − $$ {e}^{+}{e}^{-}\to {\Sigma}^{+}{\overline{\Sigma}}^{-} $$ reaction are measured. The measurements are performed at center-of-mass energies ranging from 3.510 to 4.951 GeV. No significant evidence for the decay of the charmonium(-like) states, ψ(3770), ψ(4040), ψ(4160), Y(4230), Y(4360), ψ(4415), and Y(4660), into a Σ + Σ ¯ − $$ {\Sigma}^{+}{\overline{\Sigma}}^{-} $$ final state is observed. Consequently, upper limits for the products of the branching fractions and the electronic partial widths at the 90% confidence level are reported for these decays.

Abstract: Abstract Using (27.12 ± 0.14) × 108 ψ(3686) events collected with the BESIII detector at BEPCII, the decay of ψ(3686) → Ω − K + Ξ ¯ 0 $$ {\varOmega}^{-}{K}^{+}{\overline{\Xi}}^0 $$ + c.c. is observed for the first time. The branching fraction of this decay is measured to be B ψ 3686 → Ω − K + Ξ ¯ 0 + c . c . $$ {\mathcal{B}}_{\psi (3686)\to {\varOmega}^{-}{K}^{+}{\overline{\Xi}}^0+\textrm{c}.\textrm{c}.} $$ = (2.78 ± 0.40 ± 0.18) × 10 −6, where the first uncertainty is statistical and the second is systematic. Possible baryon excited states are searched for in this decay, but no evident intermediate state is observed with the current sample size.

9. Intensive Radiosonde Observations of Environmental Conditions on the Development of a Mesoscale Convective System in the Baiu Frontal Zone

Abstract: Abstract Mesoscale convective systems (MCSs) that occur in the Baiu frontal zone (BFZ) can cause devastating flash floods during early summer in Japan; however, the environmental conditions necessary for their development require further investigation. High‐frequency atmospheric soundings, conducted using multiple marine vessels in the East China Sea on 19 June 2022, captured the detailed environmental conditions pertaining to the development of an MCS within the BFZ. The MCS, which developed rapidly without any remarkable preceding synoptic or mesoscale disturbance in the mid‐ or upper troposphere, caused intense precipitation exceeding 80 mm/hr. The MCS persisted for approximately 6 hr, and it intensified when the influx of nearly saturated air near the sea surface toward a weak surface front overlapped with the influx of free‐tropospheric moist air. The influx of nearly saturated air near the sea surface ensured conditional instability within the lower troposphere. The influx of moist air in the free troposphere contributed to the near‐saturation conditions above the boundary layer, a feature inherent to the BFZ, and played an important role in minimizing the reduction in the buoyancy of air parcels. The results of this study indicate that a better forecast of the horizontal distribution of free tropospheric moist air is beneficial for limiting the potential area of genesis of MCS in the BFZ, and a more comprehensive understanding of the vertical variations in moisture transport contributes to an improved forecast skill for MCS in the BFZ.

Abstract: A massless particle beyond the Standard Model is searched for in the two-body decay Σ+→p+invisible using (1.0087±0.0044)×1010 J/ψ events collected at a center-of-mass energy of s=3.097GeV with the BESIII detector at the BEPCII collider. No significant signal is observed, and the upper limit on the branching fraction B(Σ+→p+invisible) is determined to be 3.2×10−5 at the 90% confidence level. This is the first search for a flavor-changing neutral current process with missing energy in hyperon decays which plays an important role in constraining new physics models.

11. Secondary aerosol formation during a special dust transport event: impacts from unusually enhanced ozone and dust backflows over the ocean

Abstract: In the autumn of 2019, a 5 d long-lasting dust event was observed using a synergy of field measurement techniques in Shanghai. This particular dust event stood out from others due to its unique characteristics, including low wind speed, high relative humidity, elevated levels of gaseous precursors, and contrasting wind patterns at different altitudes. During this event, three distinct dust stages were identified. The first stage was a typical dust invasion characterized by high concentrations of particulate matters but relatively short duration. In contrast, the second stage exhibited an unusual enhancement of ozone, attributed to compound causes of a weak synoptic system, transport from the ocean, and subsidence of high-altitude ozone downdrafted by dust. Consequently, gas-phase oxidation served as the major formation pathway of sulfate and nitrate. In the third stage of dust, a noteworthy phenomenon known as dust backflow occurred. The dust plume originated from the Shandong Peninsula and slowly drifted over the Yellow Sea and the East China Sea before eventually returning to Shanghai. Evidence of this backflow was found through the enrichment of marine vessel emissions (V and Ni) and increased solubility of calcium. Under the influence of humid oceanic breezes, the formation of nitrate was dominated by aqueous processing. Additionally, parts of nitrate and sulfate were directly transported via sea salts, evidenced by their co-variation with Na+ and confirmed through thermodynamic modeling. The uptake of NH3 on particles, influenced by the contributions of alkali metal ions and aerosol pH, regulated the formation potential of secondary aerosol. By developing an upstream–receptor relationship method, the quantities of transported and secondarily formed aerosol species were separated. This study highlights that the transport pathway of dust, coupled with environmental conditions, can significantly modify the aerosol properties, especially at the complex land–sea interface.

12. Moisture Sources of the Tohoku Heavy Rainfalls in August 2022 and the Influences of Tropical Storms

Abstract: Abstract In August 2022, the Tohoku region of Japan suffered three major heavy rainfall events, while one was related to an anticyclone, and the others were induced by a “Baiu‐like” stationary front. By using the Lagrangian backward trajectories, we found the moisture sources of the three events varied much due to the rapidly changing atmospheric conditions. In general, moisture from the Sea of Japan contributed the most in all events, while the western Pacific also played a certain role. Surprisingly, moisture from tropical storms and the East China Sea did not contribute much due to the along‐transport precipitation. But, based on forward‐traced trajectories, we confirmed that tropical storms did enhance moisture transport from subtropical regions via their southerly wind. Overall, this study highlights the role of the moisture gains nearby and the outer flows of tropical storms during heavy rainfalls in a northern region.

13. Identifying the spatiotemporal variations in ozone formation regimes across China from 2005 to 2019 based on polynomial simulation and causality analysis

Abstract: Ozone formation regimes are closely related to the ratio of volatile organic compounds (VOCs) to NOx. Different ranges of HCHO/NO2 indicate three formation regimes, including VOC-limited, transitional, and NOx-limited regimes. Due to the unstable interactions between a diversity of precursors, the range of the transitional regime, which plays a key role in identifying ozone formation regimes, remains unclear. To overcome the uncertainties from single models and the lack of reference data, we employed two models, polynomial simulation and convergent cross-mapping (CCM), to identify the ranges of HCHO/NO2 across China based on ground observations and remote sensing datasets. The ranges of the transitional regime estimated by polynomial simulation and CCM were [1.0, 1.9] and [1.0, 1.8]. Since 2013, the ozone formation regime has changed to the transitional and NOx-limited regime all over China, indicating that ozone concentrations across China were mainly controlled by NOx. However, despite the NO2 concentrations, HCHO concentrations continuously exert a positive influence on ozone concentrations under transitional and NOx-limited regimes. Under the circumstance of national NOx reduction policies, the increase in VOCs became the major driver for the soaring ozone pollution across China. For an effective management of ozone pollution across China, the emission reduction in VOCs and NOx should be equally considered.

14. Responses of Arctic black carbon and surface temperature to multi-region emission reductions: a Hemispheric Transport of Air Pollution Phase 2 (HTAP2) ensemble modeling study

Abstract: Black carbon (BC) emissions play an important role in regional climate change in the Arctic. It is necessary to pay attention to the impact of long-range transport from regions outside the Arctic as BC emissions from local sources in the Arctic were relatively small. The task force Hemispheric Transport of Air Pollution Phase 2 (HTAP2) set up a series of simulation scenarios to investigate the response of BC in a given region to different source regions. This study investigated the responses of Arctic BC concentrations and surface temperature to 20 % anthropogenic emission reductions from six regions in 2010 within the framework of HTAP2 based on ensemble modeling results. Emission reductions from East Asia (EAS) had the most (monthly contributions: 0.2–1.5 ng m−3) significant impact on the Arctic near-surface BC concentrations, while the monthly contributions from Europe (EUR), Middle East (MDE), North America (NAM), Russia–Belarus–Ukraine (RBU), and South Asia (SAS) were 0.2–1.0, 0.001–0.01, 0.1–0.3, 0.1–0.7, and 0.0–0.2 ng m−3, respectively. The responses of the vertical profiles of the Arctic BC to the six regions were found to be different due to multiple transport pathways. Emission reductions from NAM, RBU, EUR, and EAS mainly influenced the BC concentrations in the low troposphere of the Arctic, while most of the BC in the upper troposphere of the Arctic derived from SAS. The response of the Arctic BC to emission reductions in six source regions became less significant with the increase in the latitude. The benefit of BC emission reductions in terms of slowing down surface warming in the Arctic was evaluated by using absolute regional temperature change potential (ARTP). Compared to the response of global temperature to BC emission reductions, the response of Arctic temperature was substantially more sensitive, highlighting the need for curbing global BC emissions.

15. Importance of gas-particle partitioning of ammonia in haze formation in the rural agricultural environment

Abstract: Ammonia in the atmosphere is essential for the formation of fine particles that impact air quality and climate. Despite extensive prior research to disentangle the relationship between ammonia and haze pollution, the role of ammonia in haze formation in high ammonia-emitting regions is still not well understood. Aiming to better understand secondary inorganic aerosol (sulfate, nitrate, ammonium – SNA) formation mechanisms under high-ammonia conditions, 1-year hourly measurement of water-soluble inorganic species (gas and particle) was conducted at a rural supersite in Shanghai. Exceedingly high levels of agricultural ammonia, constantly around 30 µg m−3, were observed. We find that gas-particle partitioning of ammonia (ε(NH4+)), as opposed to ammonia concentrations, plays a critical role in SNA formation during the haze period. From an assessment of the effects of various parameters, including temperature (T), aerosol water content (AWC), aerosol pH, and activity coefficient, it seems that AWC plays predominant regulating roles for ε(NH4+). We propose a self-amplifying feedback mechanism associated with ε(NH4+) for the formation of SNA, which is consistent with diurnal variations in ε(NH4+), AWC, and SNA. Our results imply that a reduction in ammonia emissions alone may not reduce SNA effectively, at least at rural agricultural sites in China.

Abstract: The edge turbulence characteristics and the induced radial transport have been investigated in edge localized mode (ELM) suppression by using the $n = $ 4 resonant magnetic perturbation coils on EAST, with ${q_{95}} = $ 3.6 and the electron collisionality $\nu _{\text{e}}^* \approx $ 0.5. During ELM suppression, the edge turbulence is enhanced dramatically, as measured by the reciprocating probe and the poloidal correlation reflectometry. In the near SOL, the low frequency turbulence (250 kHz), which could be beneficial to form the observed turbulent transport. The estimated upstream cross-field particle flux is consistent with the total particle flux deposited on divertor targets, demonstrating that the enhanced radial turbulent particle transport is an important mechanism for particle exhaust in ELM suppression.

Abstract: A newly developed radial directional electron probe (DEP) has been applied to the unidirectional electron current measurement on EAST tokamak. The DEP consists of two radial arrays of channels which have opposite directions and align along the local magnetic field line. Each radial array has 6 holes with a radial interval of 5 mm. Every channel has a hole with 0.5 mm radial width, 3 mm depth and 15° poloidal opening angle. The graphite collector embedded inside the hole is biased to positive potential to repel low energy ions, and high energy ions are blocked by the hole surface because their Larmor radii are larger than the radial width of hole. In consequence, the ion current collected by the DEP collector can be ignored in contrast with the electron current, as demonstrated by the I-V characteristics in a DEP commissioning experiment. The difference of collected current between two opposite channels signifies the unidirectional electron current in the flux tube. In a lower hybrid wave (LHW) modulation experiment, the amplitude and radial structure of unidirectional electron current induced by LHW is measured directly by this radial DEP array, and the LHW filament current covers over 20 mm radial region with a maximum of 20 A/cm2.

18. Detection of a single circulating tumor cell using a genetically engineered antibody-like phage nanofiber probe

Abstract: Detection of circulating tumor cells (CTCs), at the single-cell level, can be of great value for subsequent treatment of tumors but remains a significant challenge. Herein, we report a phage-based detection strategy for the identification of single CTC. The key assay component is the flexible recombinant M13@anti-CEA-scFv probe (M13, CEA, and scFv stand for M13 phage, carcinoembryonic antigen, and single-chain variable region fragment, respectively), which has a high, monoclonal antibody-like binding affinity for tumor cell-expressed CEA antigen, as confirmed by enzyme-linked immunosorbent assay (ELISA). Cell immunofluorescence experiments by confocal microscopy also demonstrate that the M13@anti-CEA-scFv could specifically bind to CEA-positive colon cancer cells such as Caco-2 and HT29, instead of CEA-negative HEK293T cells even when in great excess. Quantitative polymerase chain reaction (qPCR) further proves the ability to pin down single Caco-2 cell in 1 mL of spiked samples by 1,000 copies of M13@anti-CEA-scFv probe, after merely 15 min incubation. Another probe, the rigid magnetic microparticle (MMP), loaded with anti-CEA antibody, is used for the initial screening of the single CTC, which is mixed with numerous lymphocytes. The CEA on the resultant MMPs/CTC complex is subsequently recognized by numerous copies of M13@anti-CEA-scFv to form an MMP/CTC/M13@anti-CEA-scFv sandwich complex. Probing the phage DNA in this complex by qPCR leads to the unambiguous identification of the single Caco-2 cell in 1 mL of spiked blood samples. The phage-based detection strategy holds promise for the single-cell detection of any CTCs since the scFv displayed at the end of phage can be customized to the unique antigen of a specific CTC.

20. Plant functional traits determine latitudinal variations in soil microbial function: evidence from forests in China

Abstract: Plant functional traits have increasingly been studied as determinants of ecosystem properties, especially for soil biogeochemical processes. While the relationships between biological community structures and ecological functions are a central issue in ecological theory, these relationships remain poorly understood at the large scale. We selected nine forests along the North–South Transect of Eastern China (NSTEC) to determine how plant functional traits influence the latitudinal pattern of soil microbial functions and how soil microbial communities and functions are linked at the regional scale. We found that there was considerable latitudinal variation in the profiles of different substrate use along the NSTEC. Specifically, we found that the substrate use by microorganisms was highest in the temperate forest soils (soil microbial substrate use intensities of 10–12), followed by the subtropical forest soils (soil microbial substrate use intensities of 7–10), and was least in the coniferous forest soils (soil microbial substrate use intensities of 4–7). The latitudinal variation in soil microbial function was more closely related to plant functional traits (leaf dry matter content, leaf C concentrations, and leaf N concentrations, P=0.002) than climate (mean annual precipitation, P=0.022). The soil silt, leaf dry matter, and leaf C and N contents were the main controls on the biogeographical patterns of microbial substrate use in these forest soils. The soil microbial community structures and functions were significantly correlated along the NSTEC. Soil carbohydrate and polymer substrate use were mainly related to soil Gram-positive (G+) bacterial and actinomycic phospholipid fatty acids (PLFAs), while the use of amine and miscellaneous substrates were related to soil Gram-negative (G−) bacterial and fungal PLFAs. The enzyme production varied with changes in the soil microbial communities. The soil enzyme activities were positively correlated with the bacterial PLFAs but were not correlated with the fungal PLFAs. The soil organic matter (SOM) decomposition rates were significantly higher in the temperate forests than in the subtropical and tropical forests, emphasizing the rapid degradability of high-energy substrates such as soil microbial biomass carbon, carbohydrates, and amino acids. The SOM decomposition rates were significantly and negatively related to soil dissolved organic carbon concentrations, carboxylic acids, polymers, and miscellaneous substrate use. The relationships between soil PLFAs and microbial substrate use, enzyme activities, and SOM decomposition rate show that as the soil microbial community structure changes, soil biogeochemical processes also change.

21. Characteristics of atmospheric mercury in a suburban area of east China: sources, formation mechanisms, and regional transport

Abstract: Speciated atmospheric mercury including gaseous elemental mercury (GEM), gaseous oxidized mercury (GOM), and particulate-bound mercury (PBM) were measured continuously for a 1-year period at a suburban site, representing a regional transport intersection zone, in east China. Annual mean concentrations of GEM, PBM, and GOM reached 2.77 ng m−3, 60.8 pg m−3, and 82.1 pg m−3, respectively. GEM concentrations were elevated in all the seasons except autumn. High mercury concentrations were related to winds from the south, southwest, and north of the measurement site. Combining analysis results from using various source apportionment methods, it was found that GEM concentration was higher when quasi-local sources dominated over long-range transport. Six source factors belonging to the anthropogenic sources of GEM were identified, including the common sectors previously identified (industrial and biomass burning, coal combustion, iron and steel production, cement production, and incineration), as well as an additional factor of shipping emissions (accounting for 19.5 % of the total), which was found to be important in east China where marine vessel shipping activities are intense. Emissions of GEM from natural surfaces were also found to be as important as those from anthropogenic sources for GEM observed at this site. Concurrences of high GOM concentrations with elevated O3 and temperature, along with the lagged variations in GEM and GOM during daytime demonstrated that the very high GOM concentrations were partially ascribed to intense in situ oxidation of GEM. Strong gas–particle partitioning was also identified when PM2.5 was above a threshold value, in which case GOM decreased with increasing PM2.5.

22. Shrub type dominates the vertical distribution of leaf C : N : P stoichiometry across an extensive altitudinal gradient

Abstract: Understanding leaf stoichiometric patterns is crucial for improving predictions of plant responses to environmental changes. Leaf stoichiometry of terrestrial ecosystems has been widely investigated along latitudinal and longitudinal gradients. However, very little is known about the vertical distribution of leaf C : N : P and the relative effects of environmental parameters, especially for shrubs. Here, we analyzed the shrub leaf C, N and P patterns in 125 mountainous sites over an extensive altitudinal gradient (523–4685 m) on the Tibetan Plateau. Results showed that the shrub leaf C and C : N were 7.3–47.5 % higher than those of other regional and global flora, whereas the leaf N and N : P were 10.2–75.8 % lower. Leaf C increased with rising altitude and decreasing temperature, supporting the physiological acclimation mechanism that high leaf C (e.g., alpine or evergreen shrub) could balance the cell osmotic pressure and resist freezing. The largest leaf N and high leaf P occurred in valley region (altitude 1500 m), likely due to the large nutrient leaching from higher elevations, faster litter decomposition and nutrient resorption ability of deciduous broadleaf shrub. Leaf N : P ratio further indicated increasing N limitation at higher altitudes. Interestingly, drought severity was the only climatic factor positively correlated with leaf N and P, which was more appropriate for evaluating the impact of water status than precipitation. Among the shrub ecosystem and functional types (alpine, subalpine, montane, valley, evergreen, deciduous, broadleaf, and conifer), their leaf element contents and responses to environments were remarkably different. Shrub type was the largest contributor to the total variations in leaf stoichiometry, while climate indirectly affected the leaf C : N : P via its interactive effects on shrub type or soil. Collectively, the large heterogeneity in shrub type was the most important factor explaining the overall leaf C : N : P variations, despite the broad climate gradient on the plateau. Temperature and drought induced shifts in shrub type distribution will influence the nutrient accumulation in mountainous shrubs.

Abstract: Soil microorganisms play an important role in regulating nutrient cycling in terrestrial ecosystems. Most of the studies conducted thus far have been confined to a single forest biome or have focused on one or two controlling factors, and few have dealt with the integrated effects of climate, vegetation, and soil substrate availability on soil microbial communities and functions among different forests. In this study, we used phospholipid-derived fatty acid (PLFA) analysis to investigate soil microbial community structure and extracellular enzymatic activities to evaluate the functional potential of soil microbes of different types of forests in three different climatic zones along the north–south transect in eastern China (NSTEC). Both climate and forest type had significant effects on soil enzyme activities and microbial communities with considerable interactive effects. Except for soil acid phosphatase (AP), the other three enzyme activities were much higher in the warm temperate zone than in the temperate and the subtropical climate zones. The soil total PLFAs and bacteria were much higher in the temperate zone than in the warm temperate and the subtropical zones. The soil β-glucosidase (BG) and N-acetylglucosaminidase (NAG) activities were highest in the coniferous forest. Except for the soil fungi and fungi–bacteria (F/B), the different groups of microbial PLFAs were much higher in the conifer broad-leaved mixed forests than in the coniferous forests and the broad-leaved forests. In general, soil enzyme activities and microbial PLFAs were higher in primary forests than in secondary forests in temperate and warm temperate regions. In the subtropical region, soil enzyme activities were lower in the primary forests than in the secondary forests and microbial PLFAs did not differ significantly between primary and secondary forests. Different compositions of the tree species may cause variations in soil microbial communities and enzyme activities. Our results showed that the main controls on soil microbes and functions vary in different climatic zones and that the effects of soil moisture content, soil temperature, clay content, and the soil N ∕ P ratio were considerable. This information will add value to the modeling of microbial processes and will contribute to carbon cycling in large-scale carbon models.

Abstract: In the context of global warming attributable to the increasing levels of CO2, severe drought may be more frequent in areas that already experience chronic water shortages (semiarid areas). This necessitates research on the interactions between increased levels of CO2 and drought and their effect on plant photosynthesis. It is commonly reported that 13C fractionation occurs as CO2 gas diffuses from the atmosphere to the substomatal cavity. Few researchers have investigated 13C fractionation at the site of carboxylation to cytoplasm before sugars are exported outward from the leaf. This process typically progresses in response to variations in environmental conditions (i.e., CO2 concentrations and water stress), including in their interaction. Therefore, saplings of two typical plant species (Platycladus orientalis and Quercus variabilis) from semiarid areas of northern China were selected and cultivated in growth chambers with orthogonal treatments (four CO2 concentration ([CO2]) × five soil volumetric water content (SWC)). The δ13C of water-soluble compounds extracted from leaves of saplings was determined for an assessment of instantaneous water use efficiency (WUEcp) after cultivation. Instantaneous water use efficiency derived from gas-exchange measurements (WUEge) was integrated to estimate differences in δ13C signal variation before leaf-level translocation of primary assimilates. The WUEge values in P. orientalis and Q. variabilis both decreased with increased soil moisture at 35–80 % of field capacity (FC) and increased with elevated [CO2] by increasing photosynthetic capacity and reducing transpiration. Instantaneous water use efficiency (iWUE) according to environmental changes differed between the two species. The WUEge in P. orientalis was significantly greater than that in Q. variabilis, while an opposite tendency was observed when comparing WUEcp between the two species. Total 13C fractionation at the site of carboxylation to cytoplasm before sugar export (total 13C fractionation) was species-specific, as demonstrated in the interaction of [CO2] and SWC. Rising [CO2] coupled with moistened soil generated increasing disparities in δ13C between water-soluble compounds (δ13CWSC) and estimates based on gas-exchange observations (δ13Cobs) in P. orientalis, ranging between 0.0328 and 0.0472 ‰. Differences between δ13CWSC and δ13Cobs in Q. variabilis increased as [CO2] and SWC increased (0.0384–0.0466 ‰). The 13C fractionation from mesophyll conductance (gm) and post-carboxylation both contributed to the total 13C fractionation that was determined by δ13C of water-soluble compounds and gas-exchange measurements. Total 13C fractionation was linearly dependent on stomatal conductance, indicating that post-carboxylation fractionation could be attributed to environmental variation. The magnitude and environmental dependence of apparent post-carboxylation fractionation is worth our attention when addressing photosynthetic fractionation.

Abstract: Earth surface systems are controlled by a combination of global and local factors, which cannot be understood without accounting for both the local and global components. The system dynamics cannot be recovered from the global or local controls alone. Ground forest inventory is able to accurately estimate forest carbon stocks in sample plots, but these sample plots are too sparse to support the spatial simulation of carbon stocks with required accuracy. Satellite observation is an important source of global information for the simulation of carbon stocks. Satellite remote sensing can supply spatially continuous information about the surface of forest carbon stocks, which is impossible from ground-based investigations, but their description has considerable uncertainty. In this paper, we validated the Kriging method for spatial interpolation of ground sample plots and a satellite-observation-based approach as well as an approach for fusing the ground sample plots with satellite observations. The validation results indicated that the data fusion approach reduced the uncertainty of estimating carbon stocks. The data fusion had the lowest uncertainty by using an existing method for high-accuracy surface modelling to fuse the ground sample plots with the satellite observations (HASM-S). The estimates produced with HASM-S were 26.1 and 28.4 % more accurate than the satellite-based approach and spatial interpolation of the sample plots respectively. Forest carbon stocks of 7.08 Pg were estimated for China during the period from 2004 to 2008, an increase of 2.24 Pg from 1984 to 2008, using the preferred HASM-S method.

26. Temporal and spatial variations of precipitation in the Jinsha River basin during 1961–2010

Abstract: Knowing the variations of precipitation at the basin scale is very important to study the impacts of climate change on water resources and hydrological processes. To achieve the temporal and spatial variations of precipitation on long time scales and some extreme indicators in the Jinsha River basin, some typical precipitation indices were analysed based on daily precipitation data for 1961–2010 for the research area. The results showed that AP had a certain increasing tendency without passing the significance test, while AP in the lower reach of the basin decreased slightly. PFS had no obvious changes, while MP through a year (except rainfall in September and December) had a slight increasing tendency. In addition, AP and PFS showed obvious spatial differences, and the higher rainfall area was located in the lower basin especially in the Hengduan Mountain area. LRD and MRD increased slightly in the upper and middle regions, while they decreased slightly in the lower basin. HRD increased over most of the whole basin, but it had a decreasing tendency in the headwater region and around Dege station but did not pass the significance test. DD and CDD in one year showed similar spatial change patterns and had an obvious decreasing tendency in the upper and middle basin, while they had an obvious increasing tendency in the lower basin. CWD almost decreased over the whole basin, and decreased significantly in a small part of the lower basin. The temporal changes of the typical precipitation indices may confirm the possible increasing tendency for occurrence of drier climate and even drought events in the downstream of Jinsha River basin.

27. Impact of global dimming on reference evapotranspiration in Hai River basin, China

Abstract: Global radiation can impact the surface energy balance and hydrological cycle. This study analysed changes in global radiation in Hai River basin, China during 1960 to 2012. Global radiation decreased significantly from 1960 to 1989 and increased from 1990 to 2012, which has been described as “from dimming to brightening”. Sunshine duration was used as a proxy index where radiation records were unavailable. Results showed that sunshine duration (for 45 stations) show a significant decreasing trend from 1960 to 2012, which was a little different to global radiation (for 6 stations) during the brightening phase. Penman-Monteith model was used for estimating trends of reference evapotranspiration. The consistency of temporal and spatial variations in sunshine duration and reference evapotranspiration was examined during the periods 1960−1989 and 1990−2012. The results showed that the annual reference evapotranspiration trend was consistent with the sunshine duration trend (both were decreasing for 37 of the 45 stations) during 1960 to 1989, followed by an upward trend (for 26 of the 45 stations) from 1990, while the annual sunshine duration indicated a downward trend (for all 45 stations) during the whole period 1960−2012. Overall, global dimming could be the dominant factor for the decreased reference evapotranspiration from 1960 to 1989, while the increased reference evapotranspiration from 1990 to 2012 could be driven by other meteorological variables, especially global warming.

28. Field 13CO2 pulse labeling reveals differential partitioning patterns of photoassimilated carbon in response to livestock exclosure in a Kobresia meadow

Abstract: Livestock exclosure has been widely used as an approach for grassland restoration. However, the effects of exclosures on grasslands are controversial and can depend on many factors, such as the grassland ecosystem types, evolutionary history and so on. In this study, we conduct field experiments to investigate the variations of the ecosystem function in response to livestock exclosure in a Kobresia humilis meadow with 6 years of grazing exclosure on the Qinghai–Tibetan Plateau. We focused on two ecosystem functions: plant community structure and ecosystem carbon cycling. The plant aboveground productivity, plant diversity and the composition of plant functional groups of the meadow were addressed as the indicators of the plant community structure. The 13C isotope pulse labeling technique was applied to evaluate the alterations of ecosystem carbon cycling during a short term. The results showed that the plant community structure was changed after being fenced in for 6 years, with significantly decreased aboveground productivity, species loss and varied composition of the four plant functional groups (grasses, sedges, legumes and forbs). Using the pulse labeling technique, we found a lower cycling rate of 13C in the plant–soil system of the fenced plots compared with the grazed sites during the first 24 h after labeling. A higher proportion of recovered 13C in the plant–soil system migrated into the soil as root exudates immediately after labeling at both fenced and control grazed sites, with a significantly lower proportion in the fenced site, coinciding with the lower proportion of 13C lost from soil respiration. Thirty-two days after labeling, 37% of the recovered 13C remained in the soil of the fenced plots, with significant differences compared to in the grazed plots (47%). In addition, less 13C (5 vs. 7%) was lost by soil respiration in the fenced plots during the chase period of 32 days. Overall, our study suggests that livestock exclosures have negative effects on the plant community structure and partitioning patterns of the photoassimilated carbon in the Kobresia meadow, and the effects on photoassimilated carbon cycling are likely to result from the variations of community structures in the ecosystem.

Abstract: In this experimental study of continuous collapse of reinforced concrete frame structure, the authors design a 2*2 layer of reinforced concrete frame model and simulate the continuous collapse process of frame structure with the pseudo static test method, and hybrid control force and displacement loading method. While the Angle collapsed failure process and plastic hinge form development are observed, the date on resistance, reinforcement and concrete strain is collected and the characteristics of resistance to the continuous collapse of reinforced concrete frame structure is analyzed. By using ABAQUS finite element software to simulate the process of continuous collapse of reinforced concrete frame structures prism failure, resistance curves are spotted and are compared with the experiment result, and the congruency identified between them shows that the simulation can better simulate the process of continuous collapses of the model. On the basis of the corner posts failure simulation, the progressive collapse process is simulated and analyzed with ABAQUS when the reinforced concrete frame structural long side of the column are failure, and accordingly what is discussed is the collapse of the force during the conversion mechanism. The simulation results also show: Firstly, that during the conversion process of column failure, framework has gone through three stages ranging from the elastic stage, the stage of formation of plastic hinge, to the stage of concrete damage and suspension cable; Secondly, the beam in frame failure area has experienced the conversion process by bending to the suspension cable to provide continuous collapse resistance.

30. A possible theory for the interaction between convective activities and vortical flows

Abstract: Theoretical studies usually attribute convections to the developments of instabilities such as the static or symmetric instabilities of the basic flows. However, the following three facts make the validities of these basic theories unconvincing. First, it seems that in most cases the basic flow with balance property cannot exist as the exact solution, so one cannot formulate appropriate problems of stability. Second, neither linear nor nonlinear theories of dynamical instability are able to describe a two-way interaction between convection and its background, because the basic state which must be an exact solution of the nonlinear equations of motion is prescribed in these issues. And third, the dynamical instability needs some extra initial disturbance to trigger it, which is usually another point of uncertainty. The present study suggests that convective activities can be recognized in the perspective of the interaction of convection with vortical flow. It is demonstrated that convective activities can be regarded as the superposition of free modes of convection and the response to the forcing induced by the imbalance of the unstably stratified vortical flow. An imbalanced vortical flow provides not only an initial condition from which unstable free modes of convection can develop but also a forcing on the convection. So, convection is more appropriately to be regarded as a spontaneous phenomenon rather than a disturbance-triggered phenomenon which is indicated by any theory of dynamical instability. Meanwhile, convection, particularly the forced part, has also a reaction on the basic flow by preventing the imbalance of the vortical flow from further increase and maintaining an approximately balanced flow.

Abstract: Responses of the chlorophyll, antioxidant enzymes and lipid peroxidation in maize growing in soil with zinc (Zn) and elemental sulfur (S) were studied. The results showed that sulfur alleviated the high toxicity of Zn and increased the concentration of chlorophyll. The activities of superoxide dismutase (SOD) and peroxidase (POD) increased significantly and catalase (CAT) activity significantly decreased in plants treated with 1000 mg/kg Zn alone. Compared with the control, SOD and POD activity increased from 62.3 to 77.3 U/mg protein and 28 to41 nmol/min/mg protein, respectively. However, CAT activity decreased from 1.96 to 1.48 µmol H2O2/min/mg protein. The concentration of malondialdehyde (MDA) in maize leaves significantly increased as the concentration of Zn increased. Moderate concentrations of S (32 and 160 mg/kg) alleviated the increase of both SOD and POD activity and the decrease of CAT activity that were observed under conditions of Zn stress. The greatest decrease of SOD and POD activity were 17% and 21% and both were observed in the treatment of 160 mg/kg S combined with 1000 mg/kg Zn. In this treatment, the greatest increase of CAT activity (11%) was also observed. The results suggest that moderate supplementation with S may ameliorate the toxicity caused by excess Zn and plays an important role in protecting plants from oxidative stress induced by excess Zn.

Abstract: PCR-SSCP was applied to analyze the polymorphisms of A-FABP gene in 4 lines of Beijing ducks (n = 400). The results showed that six SNPs were found in intron 3. There were no polymorphisms found in exon 3 or exon 4. The discovered SNPs were deposited in GenBank (Acc. No.: EU306611 and EU306610). The frequencies of haplotypes A/B in the Z4, Z2, Cherry Valley, Z4 × Z2 populations were 0.745/0.255, 0.764/0.236, 0.552/0.448, 0.672/0.328, respectively. The linkage disequilibrium was stated. The above described SNPs of A-FABP gene allow the incoming association analysis.

33. Simulation of Thermal Flow Problems via a Hybrid Immersed Boundary-Lattice Boltzmann Method

Abstract: A hybrid immersed boundary-lattice Boltzmann method (IB-LBM) is presented in this work to simulate the thermal flow problems. In current approach, the flow field is resolved by using our recently developed boundary condition-enforced IB-LBM (Wu and Shu, (2009)). The nonslip boundary condition on the solid boundary is enforced in simulation. At the same time, to capture the temperature development, the conventional energy equation is resolved. To model the effect of immersed boundary on temperature field, the heat source term is introduced. Different from previous studies, the heat source term is set as unknown rather than predetermined. Inspired by the idea in (Wu and Shu, (2009)), the unknown is calculated in such a way that the temperature at the boundary interpolated from the corrected temperature field accurately satisfies the thermal boundary condition. In addition, based on the resolved temperature correction, an efficient way to compute the local and average Nusselt numbers is also proposed in this work. As compared with traditional implementation, no approximation for temperature gradients is required. To validate the present method, the numerical simulations of forced convection are carried out. The obtained results show good agreement with data in the literature.

Abstract: This study aimed to explore the roles of monocyte chemotactic protein 1 (MCP-1) and nuclear factor kappa B (NF-κB) in immune response to spinal tuberculosis in a New Zealand white rabbit model. Forty-eight New Zealand white rabbits were collected and divided into four groups: experimental group (n=30, spinal tuberculosis model was established), the sham group (n=15, sham operation was performed) and the blank group (n=3). The qRT-PCR assay and western blotting were applied to detect the mRNA and protein expressions of MCP-1 and NF-κB in peripheral blood. ELISA was used to measure serum levels of MCP-1, NF-κB, IFN-γ, IL-2, IL-4, and IL-10. Flow cytometry was adopted to assess the distributions of CD4+, CD8+ lymphocytes and CD4+ CD25+ Foxp3 lymphocyte subsets. Compared with the sham and blank groups, the mRNA and protein expressions of MCP-1 and NF-κB in the experimental group were significantly increased. The experimental group had lower serum levels of IL-2 and IFN-γ and higher serum level of IL-10 than the sham and blank groups. In comparison to the sham and blank groups, CD4+ T lymphocyte subsets percentage, CD4+/CD8+ ratio and CD4+ CD25+ Foxp3+ Tregs subsets accounting for CD4+ lymphocyte in the experimental group were lower, while percentage of CD8+ T lymphocyte subsets was higher. Our study provided evidence that higher expression of MCP-1 and NF-κB may be associated with decreased immune function of spinal tuberculosis, which can provide a new treatment direction for spinal tuberculosis.

Abstract: PRDM (PRDI-BF1 and RIZ homology domain containing) family members are sequence-specific transcriptional regulators involved in cell identity and fate determination, often dysregulated in cancer. The PRDM15 gene is of particular interest, given its low expression in adult tissues and its overexpression in B-cell lymphomas. Despite its well characterized role in stem cell biology and during early development, the role of PRDM15 in cancer remains obscure. Herein, we demonstrate that while PRDM15 is largely dispensable for mouse adult somatic cell homeostasis in vivo, it plays a critical role in B-cell lymphomagenesis. Mechanistically, PRDM15 regulates a transcriptional program that sustains the activity of the PI3K/AKT/mTOR pathway and glycolysis in B-cell lymphomas. Abrogation of PRDM15 induces a metabolic crisis and selective death of lymphoma cells. Collectively, our data demonstrate that PRDM15 fuels the metabolic requirement of B-cell lymphomas and validate it as an attractive and previously unrecognized target in oncology., The transcriptional regulator PRDM15 is expressed at low levels in normal tissues but overexpressed in B-cell lymphomas. Abstract: Detection of circulating tumor cells (CTCs), at the single-cell level, can be of great value for subsequent treatment of tumors but remains a significant challenge. Herein, we report a phage-based detection strategy for the identification of single CTC. The key assay component is the flexible recombinant M13@anti-CEA-scFv probe (M13, CEA, and scFv stand for M13 phage, carcinoembryonic antigen, and single-chain variable region fragment, respectively), which has a high, monoclonal antibody-like binding affinity for tumor cell-expressed CEA antigen, as confirmed by enzyme-linked immunosorbent assay (ELISA). Cell immunofluorescence experiments by confocal microscopy also demonstrate that the M13@anti-CEA-scFv could specifically bind to CEA-positive colon cancer cells such as Caco-2 and HT29, instead of CEA-negative HEK293T cells even when in great excess. Quantitative polymerase chain reaction (qPCR) further proves the ability to pin down single Caco-2 cell in 1 mL of spiked samples by 1,000 copies of M13@anti-CEA-scFv probe, after merely 15 min incubation. Another probe, the rigid magnetic microparticle (MMP), loaded with anti-CEA antibody, is used for the initial screening of the single CTC, which is mixed with numerous lymphocytes. The CEA on the resultant MMPs/CTC complex is subsequently recognized by numerous copies of M13@anti-CEA-scFv to form an MMP/CTC/M13@anti-CEA-scFv sandwich complex. Probing the phage DNA in this complex by qPCR leads to the unambiguous identification of the single Caco-2 cell in 1 mL of spiked blood samples. The phage-based detection strategy holds promise for the single-cell detection of any CTCs since the scFv displayed at the end of phage can be customized to the unique antigen of a specific CTC. Abstract: Upstream density profiles in the scrape-off layer (SOL) have been examined in low-confinement mode (L-mode) and high-confinement mode (H-mode) plasmas in the EAST superconducting tokamak. A weak density shoulder forms in the near SOL region in upper single-null configurations when the neutral pressure measured at the lower divertor exceeds a threshold value of 2 × 10−2 Pa in L-mode plasmas. When the neutral pressure is below this threshold, the weak density shoulder is absent and the sidebands of the lower hybrid waves associated with SOL parametric instabilities are reduced. Active detachment control with neon–deuterium seeding demonstrate that the weak density shoulder can form before the onset of the outer divertor detachment as long as the neutral pressure is above the threshold. Furthermore, no remarkable expansion of a shoulder is observed during divertor detachment, suggesting that divertor detachment is not a necessary condition for the formation or growth of a density shoulder. Through the increase in neutral pressure in the lower divertor by an order of magnitude, the weak shoulder was observed to expand into the far SOL and reach the leading edge of the limiter. The results in L-mode discharges identified the neutral pressure in the lower divertor as a primary factor for the formation of an SOL density shoulder in the upper single-null discharges. For the type-I ELMy H-mode plasmas, a similar density shoulder was detected during the inter-ELM phase when the neutral pressure in the lower divertor exceeded a threshold value of 4 × 10−2 Pa. On the other hand, the shoulder was absent when the divertor neutral pressure went below this threshold even though the plasma discharge was conducted with a higher core line-averaged density and divertor collisionality. This is consistent with the observations in L-mode plasmas. The neutral particle ionization of the working gas is thus believed to play a key role during the formation of the SOL density shoulder in the EAST tokamak.

Abstract: Mayer-Rokitansky-Küster-Hauser syndrome (MRKHS) is associated with congenital absence of the uterus, cervix, and the upper part of the vagina; it is a sex-limited trait. Disrupted development of the Müllerian ducts (MD)/Wölffian ducts (WD) through multifactorial mechanisms has been proposed to underlie MRKHS. In this study, exome sequencing (ES) was performed on a Chinese discovery cohort (442 affected subjects and 941 female control subjects) and a replication MRKHS cohort (150 affected subjects of mixed ethnicity from North America, South America, and Europe). Phenotypic follow-up of the female reproductive system was performed on an additional cohort of PAX8-associated congenital hypothyroidism (CH) (n = 5, Chinese). By analyzing 19 candidate genes essential for MD/WD development, we identified 12 likely gene-disrupting (LGD) variants in 7 genes: PAX8 (n = 4), BMP4 (n = 2), BMP7 (n = 2), TBX6 (n = 1), HOXA10 (n = 1), EMX2 (n = 1), and WNT9B (n = 1), while LGD variants in these genes were not detected in control samples (p = 1.27E−06). Interestingly, a sex-limited penetrance with paternal inheritance was observed in multiple families. One additional PAX8 LGD variant from the replication cohort and two missense variants from both cohorts were revealed to cause loss-of-function of the protein. From the PAX8-associated CH cohort, we identified one individual presenting a syndromic condition characterized by CH and MRKHS (CH-MRKHS). Our study demonstrates the comprehensive utilization of knowledge from developmental biology toward elucidating genetic perturbations, i.e., rare pathogenic alleles involving the same loci, contributing to human birth defects. © 2020 American Society of Human Genetics

Abstract: The source region of the Yellow River is an important source of water conservation in the Qinghai-Tibet Plateau. It is also an animal husbandry base that has a major impact on China. Its ecological environment changes will have a major impact on the safe and sustainable development of water use in Asia. The alpine grassland is an important part of the ecosystem of the Yellow River source area, and the spatial differentiation characteristics of the plaque have important indication significance for the ecological environment succession. In this paper, the alpine grassland in the source region of the Yellow River is taken as the research object. Based on the aerial image of the unmanned aerial vehicle, the EGI threshold method is used to extract the vegetation patches and the bare plaques to obtain high-precision field monitoring data, and the landscape ecology principle is used to analyze the four types of alpine grassland. The morphological characteristics of plaques in different grassland types are studied, and the spatial indications of spatial sequences are used to study the ecological significance of plaque succession. The results show that the number of plaques in swamp meadows, alpine meadows, degraded meadows and alpine grassland increase significantly, the degree of fragmentation increase, and the area of vegetation patches decrease significantly. The area of bare plaque increase significantly as the main trend; the distribution and dominance of landscape plaques are analyzed; the swamp meadow have the highest spread, the best aggregation, and the largest proportion of vegetation plaque, the highest degree of dominance, alpine grassland Conversely; in terms of the complexity of landscape patch shape; the area-weighted shape index and the area-weighted fractal dimension increase with the order of swamp meadows, alpine meadows, degraded meadows and alpine grasslands, tending to be complex plaque space. The ecological risk intensity index of alpine grassland in the source region of the Yellow River varies greatly, and the ecological risk of alpine grassland is high. The results of this study provide data support for elucidating the mechanism of spatial differentiation of alpine grass plaques, provide scientific assistance for grassland monitoring and management in the source area, and it provides an important basis for further discussion on ecological system protection, animal husbandry economy and sustainable development of alpine grassland in China. At the same time, it provides important theoretical support and ecological indication significance for the understanding of the alpine grassland ecosystem succession in the source area of the Yellow River.

Abstract: The wheels of power and trailer wheelset show different polygonal characteristics since their structures are obviously different. Therefore, the frictional self-excited vibration models of wheelset-track systems are established based on the viewpoint of the frictional self-excited vibration in reducing the wheel polygonal wear. Then, the motion stability of wheelset-track systems is studied by using the complex eigenvalue method. The results show that when the creep force between the wheel and rail is saturated, the unstable vibration frequency of the power wheelset is prone to induce 19-20th-order polygonal wear of the wheel, and the trailer wheelset is prone to induce 20-21th-order polygonal wear of the wheel. Meanwhile, the wheel polygonal wear can be effectively alleviated through changing the gearbox position of the power wheelset. And avoiding disc braking at high speeds can suppress the occurrence of wheel polygonal wear. In addition, the development tendency of wheel polygonal wear can be reduced by increasing the Young’s modulus of the brake pad, but Poisson’s ratio has little effect on the development tendency.

Abstract: C-C chemokine receptor 2 (CCR2) is a key driver of monocyte/macrophage trafficking to sites of inflammation and has long been considered a target for intervention in autoimmune disease. However, systemic administration of CCR2 antagonists is associated with marked increases in CCL2, a CCR2 ligand, in the blood. This heretofore unexplained phenomenon complicates interpretation of in vivo responses to CCR2 antagonism. We report that CCL2 elevation after pharmacological CCR2 blockade is due to interruption in a balance between CCL2 secretion by a variety of cells and its uptake by constitutive internalization and recycling of CCR2. We observed this phenomenon in response to structurally diverse CCR2 antagonists in wild-type mice, and also found substantially higher CCL2 plasma levels in mice lacking the CCR2 gene. Our findings suggest that CCL2 is cleared from blood in a CCR2-dependent but G protein (Gαi, Gαs or Gαq/11)–independent manner. Abstract: Due to the limited bandwidth resource and the interference among networks, it is challengeable to coordinate the bandwidth resource of multiple IEEE 802.15.4-based wireless personal area networks (WPANs) with heterogeneous traffics, especially in a distributed mode. In this paper, to handle this problem, we first propose a renewal carrier sense multiple access (CSMA)-based self-admission access mechanism for coexisting WPANs in order to maximize the frequency resource utilization and satisfy the diverse rate requirements of heterogeneous traffics. Secondly, we propose the time-space-hard core point process (TS-HCPP) to abstract the renewal CSMA-based self-admission access process for the IEEE 802.15.4 network with multi-channels. TS-HCPP considers the correlation of time and space, and appropriately judges the strong interference between coexisting WPANs, which can solve the density underestimation problems of traditional HCPP. Finally, relying on the TS-HCPP, we obtain the optimum combination of access parameters, which meets the minimum service rate requirements for heterogeneous traffics and maximizes the frequency resource utilization. The simulation results show that the density of coexisting WPANs evaluated by the TS-HCPP matches the experimental results, and an improvement in spectral efficiency of coexisting WPANs can be achieved in our proposed self-admission framework.

The role of mitochondria in imatinib-treated GIST is largely unknown. Experimental Design: We quantified the metabolic activity of several human GIST cell lines. We treated human GIST xenografts and genetically engineered KitV558del/+ mice with the mitochondrial oxidative phosphorylation inhibitor VLX600 in combination with imatinib and analyzed tumor volume, weight, histology, molecular signaling, and cell cycle activity. In vitro assays on human GIST cell lines were also performed. Results: Imatinib therapy decreased glucose uptake and downstream glycolytic activity in GIST-T1 and HG129 cells by approximately half and upregulated mitochondrial enzymes and improved mitochondrial respiratory capacity. Mitochondrial inhibition with VLX600 had a direct antitumor effect in vitro while appearing to promote glycolysis through increased AKT signaling and glucose transporter expression. When combined with imatinib, VLX600 prevented imatinib-induced cell cycle escape and reduced p27 expression, leading to increased apoptosis when compared to imatinib alone. In KitV558del/+ mice, VLX600 alone did not induce tumor cell death, but had a profound antitumor effect when combined with imatinib. Conclusions: Our findings show that imatinib alters the metabolic phenotype of GIST, and this may contribute to imatinib resistance. Abstract: In this study, the key factors affecting the formation of humic substances (HS) during green waste composting(GWC) and their potential relationships were investigated by setting up different initial particle sizes (IPS) (2 and 5 mm) and C/N (23, 30, and 37) for composting, evaluating the dynamic of environmental factors and humification parameters during the composting, and performing high-throughput sequencing to detect the bacterial structure and diversity dynamics. The results showed that the 2 mm and C/N=23 treatment could promote the degradation of organic matter, increase the fermentation temperature and germination index (GI), and generate a high HS content and humification degree, indicating that the IPS and C/N ratio were the potential controlling factors for composting maturation. Taxonomic analysis showed that the bacterial community structure was significantly different at various stages of composting, while the bacterial community composition of the same composting stage for the different treatments was similar. In addition, a correlation analysis and redundancy analysis showed that the formation of HS was significantly affected by temperature, GI, polyphenols and polysaccharides. Anaerolineales, Cytophagales and Blastocatellales were the important microbial markers of the maturation process. Abstract: Pancreatic ductal adenocarcinoma is a lethal cancer with fewer than 7% of patients surviving past 5 years. T-cell immunity has been linked to the exceptional outcome of the few long-term survivors, yet the relevant antigens remain unknown. Here we use genetic, immunohistochemical and transcriptional immunoprofiling, computational biophysics, and functional assays to identify T-cell antigens in long-term survivors of pancreatic cancer. Using whole-exome sequencing and in silico neoantigen prediction, we found that tumours with both the highest neoantigen number and the most abundant CD8+ T-cell infiltrates, but neither alone, stratified patients with the longest survival. Investigating the specific neoantigen qualities promoting T-cell activation in long-term survivors, we discovered that these individuals were enriched in neoantigen qualities defined by a fitness model, and neoantigens in the tumour antigen MUC16 (also known as CA125). A neoantigen quality fitness model conferring greater immunogenicity to neoantigens with differential presentation and homology to infectious disease-derived peptides identified long-term survivors in two independent datasets, whereas a neoantigen quantity model ascribing greater immunogenicity to increasing neoantigen number alone did not. We detected intratumoural and lasting circulating T-cell reactivity to both high-quality and MUC16 neoantigens in long-term survivors of pancreatic cancer, including clones with specificity to both high-quality neoantigens and predicted cross-reactive microbial epitopes, consistent with neoantigen molecular mimicry. Notably, we observed selective loss of high-quality and MUC16 neoantigenic clones on metastatic progression, suggesting neoantigen immunoediting. Our results identify neoantigens with unique qualities as T-cell targets in pancreatic ductal adenocarcinoma. More broadly, we identify neoantigen quality as a biomarker for immunogenic tumours that may guide the application of immunotherapies.

Abstract: C-C chemokine receptor 2 (CCR2) is a key driver of monocyte/macrophage trafficking to sites of inflammation and has long been considered a target for intervention in autoimmune disease. However, systemic administration of CCR2 antagonists is associated with marked increases in CCL2, a CCR2 ligand, in the blood. This heretofore unexplained phenomenon complicates interpretation of in vivo responses to CCR2 antagonism. We report that CCL2 elevation after pharmacological CCR2 blockade is due to interruption in a balance between CCL2 secretion by a variety of cells and its uptake by constitutive internalization and recycling of CCR2. We observed this phenomenon in response to structurally diverse CCR2 antagonists in wild-type mice, and also found substantially higher CCL2 plasma levels in mice lacking the CCR2 gene. Our findings suggest that CCL2 is cleared from blood in a CCR2-dependent but G protein (Gαi, Gαs or Gαq/11)--independent manner. This constitutive internalization is rapid: on a given monocyte, the entire cell surface CCR2 population is turned over in <30 minutes. We also found that constitutive receptor internalization/recycling and ligand uptake are not universal across monocyte-expressed chemokine receptors. For example, CXCR4 does not internalize constitutively. In summary, we describe a mechanism that explains the numerous preclinical and clinical reports of increased CCL2 plasma levels following in vivo administration of CCR2 antagonists. These findings suggest that constitutive CCL2 secretion by monocytes and other cell types is counteracted by constant uptake and internalization by CCR2-expressing cells. Abstract: Cementum is a mineralized tissue covering the tooth root that functions in tooth attachment and posteruptive adjustment of tooth position. During formation of the apically located cellular cementum, some cementoblasts become embedded in the cementoid matrix and become cementocytes. As apparently terminally differentiated cells embedded in a mineralized extracellular matrix, cementocytes are part of a select group of specialized cells, also including osteocytes, hypertrophic chondrocytes, and odontoblasts. The differentiation and potential function(s) of cementocytes are virtually unknown, and the question may be posed whether the cementocyte is a dynamic actor in cementum in comparable fashion with the osteocyte in the skeleton, responding to changing tooth functions and endocrine signals and actively directing local cementum metabolism. This review summarizes the literature with regard to cementocytes, comparing them to their closest “cousins,” the osteocytes, where insights gained from osteocyte studies serve to inform the critical examination of cementocytes. The review identifies important unanswered questions about these cells regarding their origins, differentiation, morphology and lacuno-canalicular system, selective markers, and potential functions.

Abstract: The hexagonal single-crystal aluminum nitride nanowires with straight or zigzag morphologies were successfully synthesized by the reaction of aluminum alloy in an ammonia atmosphere at 1100 degree Celsius. The results indicated that the crystal tropism of the nanowires is along [0001] whereas the growth direction of the zigzag nanowires shift between [2111] and [2111].

Abstract: Bimetallic Pd/Cu and Pd/Ag catalytic systems were used for borylation of propargylic alcohol derivatives. The substrate scope includes even terminal alkynes. The reactions proceed stererospecifically with formal S(N)2' pathways to give allenyl boronates.
