The present study aims to assess the Myristica malabarica Bark (MM) extracts in the diabetes-induced cognitive impaired rat model for its nootropic and neuroprotective activity. Memory enhancing activity was evaluated by Y maze and Morris water maze test, respectively. Neuroprotective effects of MM bark extracts were assessed by measuring the acetylcholinesterase, lipid peroxide, superoxide dismutase (SOD), total nitric oxide (NO), catalase (CAT) and glutathione (GSH) levels in the brain of diabetic rats. The Myristica malabarica bark methyl alcohol extract (MMMA) (100 and 200 mg/kg) was observed to affect a significant improvement in spontaneous alteration (P<0.01, P<0.001) and transfer latency (P< 0.01 P<0.001) in retention trials on Y maze and Morris water maze test respectively. However, a significant reduction in acetylcholinesterase activity (P<0.001), lipid peroxide (P<0.001), total NO (P< 0.001) and a substantial increase in SOD, CAT and GSH (P<0.001) levels was observed in animals treated with MMMA (200 mg/kg) related to the diabetic control group. The current results indicate that Myristica malabarica extracts were defending the cognitive decline in diabetes condition and which requires some additional studies to clarify its mode of action. This study intends to explore the neuroprotective and memory-enhancing properties of Myristica malabarica (MM) leaf extracts on diabetic-induced cognitive decline rats. The models of Y maze and Morris water maze were selected for nootropic activity and neuroprotective properties were deliberate by assessing the levels of Ach, malondialdehyde (MDA), superoxide dismutase (SOD), nitric oxide (NO), catalase (CAT) and glutathione hormone (GSH) in diabetic rat brains. Significantly improved memory and learning with 100 mg/kg (P < 0.01) and 200 mg/kg (P < 0.001) of EAMM (L) and MAMM (L) in Y maze and Morris water maze models. Significant effect was observed in all the extracts with two doses when compared with diabetic control on day 75. The levels of AChE (P < 0.001), MDA (P < 0.01), and NO (P < 0.05) were decreased significantly and significantly raised (P < 0.01) levels of SOD, CAT, and GSH with MAMM (100 and 200 mg/kg) compared with disease control. Phytochemical investigation of the methanol extract of the fruit rind of Myristica malabarica led to the isolation of eight known compounds that were identified as malabaricones A-D, promalabaricones B and C, 1-(2,6-dihydroxyphenyl)tetradecan-1-one, and ericanone by comparison with literature spectroscopic data. The structures of malabaricones A-D, promalabaricone B, and 1-(2,6-dihydroxyphenyl)tetradecan-1-one were confirmed by X-ray crystallography. In vitro assay of the isolated phenols indicated that they exhibited moderate anti-proliferative activity against the A2780 human ovarian cancer cell. Compounds (1, 3, 5, 6 and 7) had the most potent activities, whereas the anti-proliferative activities of compounds 2 and 4 were less potent., (© 2022 Wiley-VHCA AG, Zurich, Switzerland.)

Essential oils from the leaves of Myristica malabarica and Gymnacranthera canarica (Myristicaceae) were isolated by hydrodistillation and analyzed by GC/FID and GC/MS. Seventy-six constituents (98.5%) with β-caryophyllene (27.3%), α-humulene (13.8%) and α-copaene (11.5%) as major components were identified from the leaf oil of M. malabarica. Also, seventy-six constituents (98.1%) with β-caryophyllene (23.4%), linalool (13.4%) and α-humulene (11.3%) as major constituents were identified from the leaf oil of G. canarica. Sesquiterpene hydrocarbons constituted 77.3% in M. malabarica leaf oil and 58.1% in G. canarica leaf oil. [ABSTRACT FROM AUTHOR]
