Abstract Racial and ethnic disparities persist in cancer survival rates across the United States, despite overall improvements. This comprehensive analysis examines trends in 5-year relative survival rates from 2002–2006 to 2015–2019 for major cancer types, elucidating differences among racial/ethnic groups to guide equitable healthcare strategies. Data from the SEER Program spanning 2000–2020 were analyzed, focusing on breast, colorectal, prostate, lung, pancreatic cancers, non-Hodgkin lymphoma, acute leukemia, and multiple myeloma. Age-standardized relative survival rates were calculated to assess racial (White, Black, American Indian/Alaska Native, Asian/Pacific Islander) and ethnic (Hispanic, Non-Hispanic) disparities, utilizing period analysis for recent estimates and excluding cases identified solely through autopsy or death certificates. While significant survival improvements were observed for most cancers, notable disparities persisted. Non-Hispanic Blacks exhibited the largest gain in breast cancer survival, with an increase of 5.2% points (from 77.6 to 82.8%); however, the survival rate remained lower than that of Non-Hispanic Whites (92.1%). Colorectal cancer survival declined overall (64.7–64.1%), marked by a 6.2% point drop for Non-Hispanic American Indian/Alaska Natives (66.3–60.1%). Prostate cancer survival declined across all races, with Non-Hispanic American Indian/Alaska Natives showing a decrease of 7.7% points (from 96.9 to 89.2%). Lung cancer, acute leukemia, and multiple myeloma showed notable increases across groups. Substantial racial/ethnic disparities in cancer survival underscore the notable need for tailored strategies ensuring equitable access to advanced treatments, particularly addressing significant trends in colorectal and pancreatic cancers among specific minority groups. Careful interpretation of statistical significance is warranted given the large dataset.

Abstract Metal-organic frameworks (MOFs) with diverse chemistry, structures, and properties have emerged as appealing materials for miniaturized solid-state devices. The incorporation of MOF films in these devices, such as the integrated microelectronics and nanophotonics, requires robust patterning methods. However, existing MOF patterning methods suffer from some combinations of limited material adaptability, compromised patterning resolution and scalability, and degraded properties. Here we report a universal, crosslinking-induced patterning approach for various MOFs, termed as CLIP-MOF. Via resist-free, direct photo- and electron-beam (e-beam) lithography, the ligand crosslinking chemistry leads to drastically reduced solubility of colloidal MOFs, permitting selective removal of unexposed MOF films with developer solvents. This enables scalable, micro-/nanoscale (≈70 nm resolution), and multimaterial patterning of MOFs on large-area, rigid or flexible substrates. Patterned MOF films preserve their crystallinity, porosity, and other properties tailored for targeted applications, such as diffractive gas sensors and electrochromic pixels. The combined features of CLIP-MOF create more possibilities in the system-level integration of MOFs in various electronic, photonic, and biomedical devices.

Abstract Deep mining stress increase and thick hard rock layer endowment drive rock body fissure expansion, leading to sudden fracture of the roof plate accidents. The roof slab of 11,129 working face in Zhangji Mine is a 16 m thick sandstone composite roof slab, whose compressive strength reaches 89.8 MPa. The hardness of the roof slab is high, its bearing capacity is high, and it is difficult to collapse. The hardness of the roof plate is high, its bearing capacity is high, and it is difficult to fall. If the roof plate does not fall in time, it will lead to the energy accumulation of itself, and the sudden fracture and fall will make the working face and the two roadways show the drastic mine pressure, which will lead to the risk of the coal wall appearing the slice of gangs, the roof falling and the pressure frame. Therefore, for the 11,129 working face thick hard sandstone roof plate direct cover and mining technology conditions. It is very necessary and imperative to research the artificial roof release technology of deep hole pre-cracking blasting to improve the adventitious fall of the thick hard roof plate, reduce the overhanging roof area of the mining hollow area, and reduce the incoming pressure strength of the roof plate. This paper adopts the research method combining theory, physical experiment, numerical simulation and field test, focusing on research and analysis of the deflection change of the roof plate after pre-cracking and blasting, the relationship between the roof plate and the role of the bracket, and the effect of artificial blasting to release the roof. The results show that the theoretical part deduces and analyzes the relationship equation of the influence of the rotating and sinking of the roof plate on the pressure of the support, and combined with the law of the roof plate-support action can be divided into three stages: (1) Initial stage of mining, (2) Rotating and sinking stage, and (3) Breakdown and fragmentation stage. The deep stress increase and the rock layer's own characteristics (hardness and thickness) are the main controlling factors to determine the bearing capacity of the stent. As the depth of coal mining increases, the pressure at the minimum roof control distance of the stent gradually increases from 11.5 to 34.6 MPa. The increase of rock hardness relatively reduces the force acting on the stent, and the stent bearing capacity decreases by 75% when the hardness increases from 8 to 32 GPa. The 11,129 engineering geological parameters are brought into the calculation to obtain the pressure in the stent control distance of 13.5–20.5 MPa. The simulation part analyzes the stress relief degree of the roof plate before and after the roof blasting, and the peak stress in front of the coal wall is reduced from 46.7 to 30.9 MPa, which is an obvious effect of pressure relief, and the simulation results are analyzed in comparison with the theoretical results to validate the reliability of the simulation results. Further, the physical similarity experiments were conducted to analyze the deformation behavior of mining instability and crack evolution characteristics of the overburden rock of the quarry after cutting the roof, and the peak stress of the coal wall was in the range of 22.7–27.8 MPa after the blasting decompression, which was mutually verified with the theoretical analysis and the simulation results to ensure the reliability of the calculation results. Combined with the geological conditions of the mine area, the "fan-shaped" blasting hole arrangement is proposed to carry out segmental blasting, and the specific parameters of different locations of the holes are given. The effect of pre-cracking blasting was examined, and it was found that the rock mass at the hole opening had a high degree of fragmentation, and the ring and strike cracks appeared alternately in the middle and lower part of the drilling holes, and the polygonal energy slit played a very effective role, and achieved the purpose of artificially releasing the roof. At the same time, it is improved the load-bearing environment of the mining support, which further improves the safety and efficiency production of the coal mine.

Ethylene-insensitive 3/Ethylene-insensitive3-like proteins (EIN3/EIL) represent a group of transcription factors critical for the ethylene signaling transduction that manipulate downstream ethylene-responsive genes, thereby regulating plant growth, development, and stress responses. However, the identification, evolution, and divergence of the EIL family remain to be studied in Sorghum bicolor. Here, we identified eight SbEILs, which were expanded due to whole-genome-duplication (WGD) events. Characterization of the protein sequences and expression atlas demonstrates that the WGD-duplicated SbEILs could become divergent due to the differential expression patterns, rather than domain and motif architectures. Comparative expression analysis was performed between the RNA-seq data sets of internodes from several sorghum cultivars to understand the potential roles of SbEIL members in internode elongation and maturation. Our results identified SbEIL3 and 7 (the latter as a homolog of OsEIL7/OsEIL1) to be the highly expressed SbEIL genes in sorghum internodes and revealed a potential functional link between SbEIL7 and internode maturation. The co-expression analysis and comparative expression analysis with ethylene-regulated gene sets found that SbEIL7 was co-regulated with a set of ubiquitin-related protein degradation genes, suggesting possible involvement of SbEIL7 in protein degradation and processing during the post-anthesis stages. Altogether, our findings lay a foundation for future functional studies of ethylene signaling-mediated gene regulation and improvement of sorghum internode development.

Red root rot disease is a devastating fungal disease of rubber trees caused by Ganoderma pseudoferreum (Wakef). Biocontrols using beneficial microorganisms are safe and sustainable. We isolated a DHR18 endophytic bacterium from a healthy rubber tree to obtain a new efficient antagonistic bacterium for red root rot disease affecting rubber trees and evaluated the mechanism of action involved using a double culture assay, genome annotation analysis, and the ethyl acetate extraction method. The results revealed that the DHR18 strain inhibits G. pseudoferreum growth and has broad-spectrum antifungal activity by secreting cell wall hydrolases (proteases and chitinases), indole-3-acetic acid, and siderophores. Furthermore, it fixes nitrogen and is involved in biofilm formation and phosphate solubilisation, improving disease resistance and tree growth. The results showed that the antifungal substances secreted by DHR18 are mainly lipopeptides. Simultaneously, DHR18 enhanced the rubber tree resistance to G. pseudoferreum by increasing the activities of defence enzymes superoxide dismutase, phenylalanine ammonia lyase, peroxidase, catalase, and polyphenol oxidase. The results indicate that B. arboris DHR18 has biocontrol potential and could be used as a candidate strain for the control of red root rot disease in rubber trees.

Growth-regulating factor (GRF) is a plant-specific family of transcription factors crucial for meristem development and plant growth. Sorghum (Sorghum bicolor L. Moench) is a cereal species widely used for food, feed and fuel. While sorghum stems are important biomass components, the regulation of stem development and the carbohydrate composition of the stem tissues remain largely unknown. Here, we identified 11 SbGRF-encoding genes and found the SbGRF expansion driven by whole-genome duplication events. By comparative analyses of GRFs between rice and sorghum, we demonstrated the divergence of whole-genome duplication (WGD)-derived OsGRFs and SbGRFs. A comparison of SbGRFs’ expression profiles supports that the WGD-duplicated OsGRFs and SbGRFs experienced distinct evolutionary trajectories, possibly leading to diverged functions. RNA-seq analysis of the internode tissues identified several SbGRFs involved in internode elongation, maturation and cell wall metabolism. We constructed co-expression networks with the RNA-seq data of sorghum internodes. Network analysis discovered that SbGRF1, 5 and 7 could be involved in the down-regulation of the biosynthesis of cell wall components, while SbGRF4, 6, 8 and 9 could be associated with the regulation of cell wall loosening, reassembly and/or starch biosynthesis. In summary, our genome-wide analysis of SbGRFs reveals the distinct evolutionary trajectories of WGD-derived SbGRF pairs. Importantly, expression analyses highlight previously unknown functions of several SbGRFs in internode elongation, maturation and the potential involvement in the metabolism of the cell wall and starch during post-anthesis stages.

Modifications of mRNA, especially methylation of adenosine, have recently drawn much attention. The much rarer modification, 5-hydroxymethylation of cytosine (5hmC), is not well understood and is the subject of this study. Vertebrate Tet proteins are 5-methylcytosine (5mC) hydroxylases and catalyze the transition of 5mC to 5hmC in DNA. These enzymes have recently been shown to have the same function in messenger RNAs in both vertebrates and in Drosophila. The Tet gene is essential in Drosophila as Tet knock-out animals do not reach adulthood. We describe the identification of Tet-target genes in the embryo and larval brain by mapping one, Tet DNA-binding sites throughout the genome and two, the Tet-dependent 5hmrC modifications transcriptome-wide. 5hmrC modifications are distributed along the entire transcript, while Tet DNA-binding sites are preferentially located at the promoter where they overlap with histone H3K4me3 peaks. The identified mRNAs are preferentially involved in neuron and axon development and Tet knock-out led to a reduction of 5hmrC marks on specific mRNAs. Among the Tet-target genes were the robo2 receptor and its slit ligand that function in axon guidance in Drosophila and in vertebrates. Tet knock-out embryos show overlapping phenotypes with robo2 and both Robo2 and Slit protein levels were markedly reduced in Tet KO larval brains. Our results establish a role for Tet-dependent 5hmrC in facilitating the translation of modified mRNAs primarily in cells of the nervous system.

【Objective】Natural rubber is an important industrial raw materials and strategic materials, and played a very important role in national economic development. As a resource constraint industry, it was mainly produced by Brazilian rubber tree (Hevea brasiliensis) and it has the highest yield and the best quality among the mankind known rubber-producing plants. Harvesting latex is the main economic purpose of Brazilian rubber tree, latex yield is a quantitative trait controlled by minorgene, with many influencing factors, and its phenotypic value is affected by heredity and environmental effect. To determine the relationship among the yield and agronomic traits of the wild germplasm resources of Hevea brasiliensis, this study provides a scientific basis for the evaluation and screening of rubber tree germplasm resources and the selection and breeding of new varieties.【Method】The yield, growth and other related agronomic traits of 13 wild germplasm resources of Hevea brasiliensis planted in the experimental field of Chinese Academy of Tropical Agricultural Sciences (CATAS) were studied and analyzed by grey relational analysis.【Result】The results showed that the association order of the eight main agronomic traits related to the dry rubber yield per plant were stem girth, dry rubber content, average number of laticifer cells, average number of laticifer columns, bark thickness, mean stem girth increment, lateral vein latex grade and gum value ratio, and the weights of each trait were 0.1380, 0.1338, 0.1322, 0.1264, 0.1205, 0.1182, 0.1178, 0.1143. In summary, stem girth, dry rubber content, quantity of laticifers bark thickness and other traits are closely related to and have a great impact on dry rubber yield per plant.【Conclusion】Among the main agronomic traits, the stem girth, dry rubber content and quantity of laticifer should be paid more attention on the evaluation of rubber tree germplasm resources, at the same time the potential impact of other traits on yield also need to be laid some stress on.

Graded permittivity materials have gained significant attention due to their exceptional ability to regulate electric fields. Multi-material stereolithography (SLA) 3D printing has opened up new possibilities for creating such materials. However, conventional SLA printers typically generate graded material using fixed printing parameters and multiple feedstocks with limited differences, resulting in a constrained capacity for modulating the electric field distribution. To address this limitation, we have developed a multi-material, parameter-controllable SLA strategy, enabling us to assign varying printing parameters for each building layer and switch between feedstocks with significant differences. Solid insulators with graded permittivity are optimised through electric field distribution simulations and subsequently manufactured using our innovative multi-material SLA approach. A 4-layered graded insulator effectively decreases the maximum electric field strength from 82.5–30.8 kV/mm. Both flashover tests and partial discharge signals confirm that graded insulators outperform homogeneous ones in electrical insulation.

Natural rubber is an important national strategic and industrial raw material. The leaf anthracnose of rubber trees caused by the Colletotrichum species is one of the important factors restricting the yields of natural rubber. In this study, we isolated and identified strain Bacillus velezensis SF334, which exhibited significant antagonistic activity against both C. australisinense and C. siamense, the dominant species of Colletotrichum causing rubber tree leaf anthracnose in the Hainan province of China, from a pool of 223 bacterial strains. The cell suspensions of SF334 had a significant prevention effect for the leaf anthracnose of rubber trees, with an efficacy of 79.67% against C. siamense and 71.8% against C. australisinense. We demonstrated that SF334 can lead to the lysis of C. australisinense and C. siamense mycelia by causing mycelial expansion, resulting in mycelial rupture and subsequent death. B. velezensis SF334 also harbors some plant probiotic traits, such as secreting siderophore, protease, cellulase, pectinase, and the auxin of indole-3-acetic acid (IAA), and it has broad-spectrum antifungal activity against some important plant pathogenic fungi. The genome combined with comparative genomic analyses indicated that SF334 possesses most genes of the central metabolic and gene clusters of secondary metabolites in B. velezensis strains. To our knowledge, this is the first time a Bacillus velezensis strain has been reported as a promising biocontrol agent against the leaf anthracnose of rubber trees caused by C. siamense and C. australisinense. The results suggest that B. velezensis could be a potential candidate agent for the leaf anthracnose of rubber trees.

Bacterial leaf streak (BLS) of rice is a severe disease caused by the bacterial pathogen Xanthomonas oryzae pv. oryzicola (Xoc) that has gradually become the fourth major disease on rice in some rice-growing regions in southern China. Previously, we isolated a Bacillus velezensis strain 504 that exhibited apparent antagonistic activity against the Xoc wild-type strain RS105, and found that B. velezensis 504 was a potential biocontrol agent for BLS. However, the underlying mechanisms of antagonism and biocontrol are not completely understood. Here we mine the genomic data of B. velezensis 504, and the comparative transcriptomic data of Xoc RS105 treated by the cell-free supernatants (CFSs) of B. velezensis 504 to define differentially expressed genes (DEGs). We show that B. velezensis 504 shares over 89% conserved genes with FZB42 and SQR9, two representative model strains of B. velezensis, but 504 is more closely related to FZB42 than SQR9, as well as B. velezensis 504 possesses the secondary metabolite gene clusters encoding the essential anti-Xoc agents difficidin and bacilysin. We conclude that approximately 77% of Xoc RS105 coding sequences are differentially expressed by the CFSs of B. velezensis 504, which significantly downregulates genes involved in signal transduction, oxidative phosphorylation, transmembrane transport, cell motility, cell division, DNA translation, and five physiological metabolisms, as well as depresses an additional set of virulence-associated genes encoding the type III secretion, type II secretion system, type VI secretion system, type IV pilus, lipopolysaccharides and exopolysaccharides. We also show that B. velezensis 504 is a potential biocontrol agent for bacterial blight of rice exhibiting relative control efficiencies over 70% on two susceptible cultivars, and can efficiently antagonize against some important plant pathogenic fungi including Colletotrichum siamense and C. australisinense that are thought to be the two dominant pathogenic species causing leaf anthracnose of rubber tree in Hainan province of China. B. velezensis 504 also harbors some characteristics of plant growth-promoting rhizobacterium such as secreting protease and siderophore, and stimulating plant growth. This study reveals the potential biocontrol mechanisms of B. velezensis against BLS, and also suggests that B. velezensis 504 is a versatile plant probiotic bacterium.

Sorghum (Sorghum bicolor L. Moench), a monocot C4 crop, is an important staple crop for many countries in arid and semi-arid regions worldwide. Because sorghum has outstanding tolerance and adaptability to a variety of abiotic stresses, including drought, salt, and alkaline, and heavy metal stressors, it is valuable research material for better understanding the molecular mechanisms of stress tolerance in crops and for mining new genes for their genetic improvement of abiotic stress tolerance. Here, we compile recent progress achieved using physiological, transcriptome, proteome, and metabolome approaches; discuss the similarities and differences in how sorghum responds to differing stresses; and summarize the candidate genes involved in the process of responding to and regulating abiotic stresses. More importantly, we exemplify the differences between combined stresses and a single stress, emphasizing the necessity to strengthen future studies regarding the molecular responses and mechanisms of combined abiotic stresses, which has greater practical significance for food security. Our review lays a foundation for future functional studies of stress-tolerance-related genes and provides new insights into the molecular breeding of stress-tolerant sorghum genotypes, as well as listing a catalog of candidate genes for improving the stress tolerance for other key monocot crops, such as maize, rice, and sugarcane.

Abstract Background circular RNAs (circRNAs) have been reported to play crucial roles in the biology of different cancers. However, little is known about the function of circSTX6 (hsa_circ_0007905) in pancreatic ductal adenocarcinoma (PDAC). Methods circSTX6, a circRNA containing exons 4, 5, 6 and 7 of the STX6 gene, was identified by RNA sequencing and detected by quantitative reverse transcription PCR (qRT–PCR). The biological function of circSTX6 was assessed in vitro and in vivo. The relationship between circSTX6 and miR-449b-5p was confirmed by biotin-coupled circRNA capture, fluorescence in situ hybridization (FISH) and luciferase reporter assays. The interaction of circSTX6 with Cullin 2 (CUL2) was verified by RNA–protein RNA pull-down, RNA immunoprecipitation (RIP) and western blotting assays. Results circSTX6 was frequently upregulated in PDAC tissues, and circSTX6 overexpression promoted tumor proliferation and metastasis both in vitro and in vivo. Furthermore, circSTX6 expression was associated with tumor differentiation and N stage. Mechanistically, circSTX6 regulated the expression of non-muscle myosin heavy chain 9 (MYH9) by sponging miR-449b-5p. Moreover, circSTX6 was confirmed to participate in the ubiquitin-dependent degradation of hypoxia-inducible factor 1-alpha (HIF1A) by interacting with CUL2 and subsequently accelerating the transcription of MYH9. Conclusions Our findings indicate that circSTX6 facilitates proliferation and metastasis of PDAC cells by regulating the expression of MYH9 through the circSTX6/miR-449b-5p axis and circSTX6/CUL2/HIF1A signaling pathway. Therefore, circSTX6 could serve as a potential therapeutic target for the treatment of PDAC.

【Objective】Powdery mildew of rubber tree is the most serious leaf disease of rubber tree in China so far. The PMA-qPCR detection system for powdery mildew of rubber tree was constructed by combining propidium monoazide (PMA) with real-time fluorescence quantitative PCR (qPCR), which provided technical support for the timely and efficient detection of viable conidia of powdery mildew in rubber tree.【Method】Primers were designed according to the conserved region of ribosomal internal transcribed spacer (ITS) sequence of Erysiphe quercicola and its specificity was verified. The standard curve was drawn with the recombinant plasmid pMD-19T-Eq. A PMA-qPCR detection system for powdery mildew of rubber tree was established to evaluate its repeatability and sensitivity. The system was used to detect the viable count of powdery mildew after propiconazole treatment.【Result】The designed primers had good specificity. A good linear relationship between cycle threshold (Ct) and template concentration was found in the constructed qPCR standard curve for Erysiphe quercicola, the correlation coefficient was 0.9981, and the amplification efficiency was 107.97%. PMA-qPCR had little effect on the amplification of live spore suspension, and the detection sensitivity was 4.77×102 copies/μL. After treatment with propiconazole for 15 min, the proportion of live bacteria decreased from 66.13% to 23.18%.【Conclusion】The PMA-qPCR system could rapidly and quantitatively detect the number of live bacteria of powdery mildew in rubber tree, which had the advantages of sensitivity, accuracy and efficiency, and provided theoretical basis and technical means for the prediction and control of the disease.

In regions with high-stress roadway stress, anchor cables frequently experience damage, leading to risky pull-outs and ejections. This study aimed to determine the dynamics of such incidents, refine protective devices, and validate their efficacy in enhancing safety. Drawing from an ejection accident in the 1632 (3) roadway of Pan San Mine, a combination of laboratory experiments, theoretical analysis, simulations, and field applications was utilized. The kinetic energy and speed of cable ejections were determined from single-axis tension test data. Based on these insights, a spring-based protection device was conceptualized. Subsequent experiments and simulations evaluated the energy absorption and deformation characteristics of these devices with different diameters. The results included the following: A cable, during ejection, moved at 48 m/s. Spring protective devices of 4 mm can absorb more energy than the 5 mm, but the anti-ejection effect is poor respectively. Increasing the device diameter improved its performance, especially in controlling spring deformation rate and preventing cable lock-ups. This devised protection mechanism showed promising results when implemented in the 1511 (1) roadway of Zhangji Mine.

During tunneling in weakly consolidated soft rock strata, lithology, joints, and fractures of rock strata play an important role in controlling overall roof stability of the roadway. A case study based on weakly consolidated soft rock roadway in Bojianghaizi mine was conducted to address disasters caused by sudden changes and collapse as well as difficulties in maintenance of weakly consolidated soft rock roadway. First, physical and mechanical properties of weakly consolidated soft rocks were investigated by X-ray diffraction mineral component analysis and scanning electron microscopy. Second, failure characteristics and instability mechanism of the roadway were analyzed by combining field survey and theoretical analysis. A mechanical model of fracture development on the composite roof of the roadway was built, and the initiation angle and critical stress for fracture development were deduced. The instability criteria of cracks based on cracking angle (θ0) were established. Moreover, the instability mechanism caused by roof falling was disclosed. Fractures were formed upon shear failure at vertex angle of the roadway. The fracture belt extended to the soft bedding plane of the separation stratum along the fracture propagation angle and connected with it, thereby inducing roof falling. On this basis, a high-strength and high-preload “inverted trapezoidal” anchoring mesh-beam string supporting structure + arched roadway cross-section with vertical walls + full-section guniting coupling control technology was proposed, which achieved good site application effects.

RNA-seq has become a state-of-the-art technique for transcriptomic studies. Advances in both RNA-seq techniques and the corresponding analysis tools and pipelines have unprecedently shaped our understanding in almost every aspects of plant sciences. Notably, the integration of huge amount of RNA-seq with other omic data sets in the model plants and major crop species have facilitated plant regulomics, while the RNA-seq analysis has still been primarily used for differential expression analysis in many less-studied plant species. To unleash the analytical power of RNA-seq in plant species, especially less-studied species and biomass crops, we summarize recent achievements of RNA-seq analysis in the major plant species and representative tools in the four types of application: (1) transcriptome assembly, (2) construction of expression atlas, (3) network analysis, and (4) structural alteration. We emphasize the importance of expression atlas, coexpression networks and predictions of gene regulatory relationships in moving plant transcriptomes toward regulomics, an omic view of genome-wide transcription regulation. We highlight what can be achieved in plant research with RNA-seq by introducing a list of representative RNA-seq analysis tools and resources that are developed for certain minor species or suitable for the analysis without species limitation. In summary, we provide an updated digest on RNA-seq tools, resources and the diverse applications for plant research, and our perspective on the power and challenges of short-read RNA-seq analysis from a regulomic point view. A full utilization of these fruitful RNA-seq resources will promote plant omic research to a higher level, especially in those less studied species.

Abstract Fish sex identification is a basic technique of great importance for both fish genetic studies and fisheries. Due to the sexual reversal phenomenon in many fish species, a simple and rapid molecular identification method for fish genetic sex is urgently needed to suit versatile detection scenarios, such as point-of-need applications. In this study, we took Cynoglossus semilaevis as an example, established a recombinase-aided amplification (RAA)-based method for sex identification, and combined the RAA-detection with two result visualization approaches with distinct features, capillary electrophoresis (CE) and lateral flow dipstick (LFD). Specific primers and probe were designed to specifically detect the sex chromosome W of C. semilaevis in order to distinguish the genetic sex between males, pseudo-males and females. To evaluate the performance of our methods, the genetic sex for twenty-eight males, sixty-eight pseudo-males and fifty-four females were examined with the RAA-based method and classical PCR-based genotyping method, demonstrating the consistent results of sex identification between both methods. The RAA-LFD method is operationally simple, rapid (~ 30 min) and holds great potential for point-of-need applications of fish sex identification, including fishery fields. The method presented here could be effective for identifying fish gender with the ZW karyotype.

Aiming at the mining disaster of a thick hard roof, based on the analysis of the mining instability influence of the thick hard roof, this study constructs the mining bearing mechanical model of the thick hard roof by using mechanical theory and obtains the mechanical distribution equation of mining bearing and energy accumulation, the mining instability energy release equation, and the dynamic load response equation of a hydraulic support in the working face, as well as the dynamic load response characteristics of the hydraulic support in the working face, putting forward the technical countermeasures for the strong dynamic pressure control of the thick hard roof in the working face. This research shows that 1) the larger the overburden load and suspension span of the thick hard roof, the more serious the mining bearing state and energy accumulation evolution; the greater the rock thickness and elastic modulus of the thick hard roof, the greater the flexural stiffness of the roof, resulting in the increase of the roof mining limit breaking span, which indirectly aggravates the mining bearing state and self-energy accumulation evolution; 2) the dynamic support resistance of the hydraulic support is composed of the dynamic support resistance caused by the release of elastic energy accumulated by mining of the thick hard roof, the work done by the overlying load, and the static support resistance caused by the direct roof gravity; 3) the dynamic support resistance caused by the work of the overlying load accounts for the highest proportion, followed by the dynamic support resistance caused by the release of mining elastic energy by the thick hard roof; the cause of mining instability and the strong dynamic pressure of the thick hard roof lie in the large span of the mining suspended roof, and the large-scale mining suspension structure of the thick hard roof leads to a high overlying load and large accumulated energy; and 4) the mining instability of the thick hard roof leads to a strong dynamic load response of the hydraulic support; adopting pre-splitting and roof cutting technology to reduce the breaking span of the thick hard roof and reducing the impact dynamic load caused by mining instability of the thick hard roof can effectively eliminate the potential safety hazard of overlimit bearing of the hydraulic support.

The key problem to be solved urgently is how to avoid the occurrence of support break-off and water inrush in the stoping of sandstone straight roof under the action of load transfer in unconsolidated aquifer. For this reason, taking the thin bedrock 1602 (3) working face of Huainan (the middle part of Anhui Province) Panyi Coal Mine as the engineering background, this study establishes the stope mining model by using the discrete element UDEC software and the mathematics mechanical model of the support load, and analyzes the reason of support crushing and decides to re-mining the working face by using the compulsive roof caving method. It is concluded that when the working face of sandstone straight roof is broken, the “voussoir beam” structure cannot be formed and acts on the support in the form of cantilever beam, but only when it falls to the high key stratum can the “voussoir beam” structure be formed and at this point, at this time, the bracket bears the weight of the rock layer in the range from the fractured sandstone layer to the lower critical layer. The working resistance of the support increases with the increase of the thickness and the breaking length of straight sandstone roof. When the breaking length of the roof reaches a certain extreme value, the support crushing accidents will occur. Managing roof with compulsive roof caving method can reduce the intensity of rock pressure in the stope, and the working face can be safely stoped, which provides a certain reference for similar conditions. Fracture of a critical stratum on a coal seam roof may cause strong strata pressure behaviours. In particular, a large-scale sudden fracture of the critical stratum, which is formed by hard thick sandstones on a mining face with a large mining height, can easily cause dynamic disasters of wells. In this study, a simulation analysis of the deformation and failure laws of a hard thick roof in an island mining face with a large mining height in Xinji 2# Mine 210106 after the mining of Coal bed A1 was performed in accordance with the occurrence characteristics of sandstones. Results demonstrate that large-scale fracture suddenly occurs at a gob roof after the working face is advanced by 120 m. A mechanical analysis model of the island working face was constructed. This model comprises 17.9 m of fracture on the 16 m thick sandstone roof. Hydraulic support provides 16854.1 kN of supporting resistance to maintain the stability of the rock beam and thus prevent threats to the supporting devices of the working face. The hard thick roof was weakened every 15 m in the transport gateway and air return in advance through an advanced deep-hole pre-splitting blasting technique. In accordance with the measurement results of underground pressure, the forced caving of thick sandstones relieves the influence of roof loads on the support and thus realizes safe mining.

Pancreatic ductal adenocarcinoma (PDAC) is one of the common malignant tumors with high lethal rate and poor prognosis. Dysregulation of many genes have been reported to be involved in the occurrence and development of PDAC. However, as a highly conserved gene in eukaryotes, the role of Fasciculation and Elongation protein Zeta 2 (FEZ2) in pancreatic cancer progression is not clear. In this study, we identified the oncogenic effect of FEZ2 on PDAC. By mining of The Cancer Genome Atlas (TCGA) database, we found that FEZ2 was upregulated in PDAC tissues and FEZ2 expression was negatively regulated by its methylation. Moreover, high expression and low methylation of FEZ2 correlated with poor prognosis in PDAC patients. Besides, we found that FEZ2 could promote PDAC cells proliferation, migration and 5-FU resistance in vitro. Furthermore, Gene pathway enrichment analysis demonstrated a positive correlation between Wnt signaling activation and FEZ2 expression in PDAC patients. Western blot showed that FEZ2 knockdown significantly suppressed β-catenin expression. Collectively, our finding revealed that FEZ2 functioned as a potential oncogene on PDAC progression and migration, and the expression of FEZ2 had guidance value for the treatment and chemotherapy program of PDAC patients.

Elucidating and understanding the dynamic fracture characteristics of rocks play an essential role in the application of rock engineering and geophysics. In this study, based on a self-developed dynamic damage model, a rock notched semi-circle bend test with the Split Hopkinson Pressure Bar technique is numerically simulated. The study focuses on three aspects including damage evolution, energy evolution, and failure mode of rock under different loading velocities. From the simulated results, the following conclusions can be conducted: 1) the damage range increases gradually with the increase of loading velocity; 2) the crack propagates to the loading point along the symmetry axis of the samples under different loading velocities; 3) the loading velocity has an important influence on the failure mode of straight notch semi-circular marble, whose mechanism can be explained by that the local high strain rate leads to the obvious randomness and uncertainty of crack activation in rock; and 4) the energy evolution of notched semi-circle bend is vitally affected by loading velocity, and the deformation and the failure process of straight notch semicircular marble under dynamic loading can be divided into five stages according to the ratio of internal energy to total energy. The beneficial findings may provide some references in practice design from engineering problems.

PurposeThis study aims to integrate pancreatic cancer TCGA, GEO, and single-cell RNA-sequencing (scRNA-seq) datasets, and explore the potential prognostic markers and underlying mechanisms of the immune microenvironment of pancreatic cancer through bioinformatics methods, in vitro and in vivo assays.MethodsExpression data and clinicopathological data of pancreatic cancer TCGA, GEO (GSE131050), single cell sequencing (PAAD_CRA001160) dataset were downloaded. We used R/Bioconductor edgeR for differential expression analysis. ClusterProfiler was utilized to perform GO enrichment analysis on differentially expressed genes. The online software CIBERSORT was used to reanalyze the mRNA expression data of pancreatic cancer. CellRanger, RunPCA, FindNeighbors, FindClusters, RunTSNE and RunUMAP were used to perform preprocessing, cell clustering and expression profile analysis on single-cell sequencing data sets. We analyzed intracellular pH with or without CA9 inhibitor SLC-0111. Indirect co-culture model of human pancreatic cancer cell lines and healthy individual-derived PBMCs were used to determine the effect of CA9-related Acidic Microenvironment on CD8+ T cells.ResultsThe CIBERSORT analysis of TCGA pancreatic cancer transcriptome sequencing data showed that among the 22 immune microenvironment components, CD8+ T cell infiltration was significantly correlated with the prognosis of pancreatic cancer patients. The differential expression analysis of the TCGA data grouped by the level of CD8+ T cell infiltration indicates that the expression of carbonic anhydrase 9 (CA9) is the most significant, and the survival analysis suggests that CA9 is associated with the overall survival of pancreatic cancer. TCGA data and GEO data set GSE131050 expression correlation analysis suggests that CA9 and CD8 expression are closely related. Pancreatic cancer single-cell sequencing data set PAAD_CRA001160 analysis results show that CA9 is mainly expressed in pancreatic cancer cell clusters, and the expression of the cancer cell subgroup CA9 in the single-cell data set is correlated with CD8+ T cell infiltration.ConclusionPancreatic cancer cells may inhibit the infiltration of CD8+ T cells through CA9. Further exploration of its related mechanisms can be used to explore the immune escape pathway of pancreatic cancer and provides new perspectives immune targeted therapy.

The advanced mining stress on a large working face can easily lead to the failure of the roof and coal mass; coal wall spalling and roof caving occur. The degree of coal wall failure and the depth of the plastic fracture zone are closely related to the advanced mining stress. The mechanical analysis model is established, and the stress distribution in the fracture and plastic zone is analyzed by the elastic-plastic theory, and the depth function expressions of two zones are determined. Comparative analysis of the factors affecting mining strength shows that the mining depth, mining height, and strength of coal are the key factors affecting the stability of the coal wall. The A1 coal of Zhangji mine in Huainan is soft and thick, and the direct roof is easily separated by the organic membrane, which is easy to form arc-shaped sliding instability. The increase in the unsupported roof area causes the direct roof caving and reduces its bearing capacity and stiffness. It is unable to provide enough support for the broken block of the overlying key stratum, which makes the broken block reverse rotation and further aggravate the roof fall; the synchronous instability of the coal wall and direct roof is formed.

In this study, based on the example of the ground vibration damage response of a thick and hard rock layer fracture type mine quake, and by applying the theories of mine pressure, rock mechanics and vibration energy principle, the concept of “vibration damage boundary” of mine quake ground, which uses particle vibration velocity to evaluate the vibration damage, is proposed. In addition, the quantitative prediction method of the ground vibration damage boundary is preliminarily established. The research results reveal that the elastic deformation of the fixed support end of the thick and hard rock layer structure in the stope is the main energy source for the formation of mine quakes, and that the particle vibration velocity caused by the propagation of focal energy to the ground can reasonably reflect the response degree of vibration damage. The proposed prediction method considers the “instantaneous” motion characteristics of mining thick and hard rock layer and the dynamic load effect of “mine quakes.” The method can deepen our understanding of mining ground damage prediction, and increase the reliability of ground damage boundary prediction. Finally, the results are used to predict the ground vibration damage boundary of limestone primary fracture in the longwall 16101 face of the Fuping Coal Mine.

A pressure relief and permeability enhancement method through short-distance floor roadway was proposed to solve the difficult outburst prevention during the gas extraction at the coal roadway strips in deep outburst coal seams with high ground stress and low gas permeability. On the basis of an equivalent model of the surrounding rock in a deep roadway, the analytical solutions of deep roadway excavation to the stress and deformation of pressure relief at overlying short-distance coal roadway strips were obtained using the unified strength theory and nonassociated flow rules. Next, the criteria for determining the reasonable position of floor roadway were established, and a mechanical model of short-distance floor roadway for the pressure relief and permeability enhancement zone at the overlying coal seam was constructed. Finally, the scope of the zonal disintegration at the coal roadway strips in the elastic and elastic–plastic zones of the surrounding rock in the roadway, as well as the expression of gas permeability change, was given. The engineering trial calculation and practice showed that the stress and strain of the surrounding rock in the roadway were evidently influenced by the intermediate principal stress coefficient. Moreover, the vertical stress and vertical displacement of overlying coal seam were gradually reduced with the increase in the intermediate principal stress coefficient and vertical distance of the floor roadway. The minimum reasonable distance arranged for the 213 floor roadway in Qujiang Coal Mine was 6.21 m, and the effective pressure relief should be conducted within 10.6 m from the coal seam floor. When the pressure relief was located at 9.0 m from the coal seam floor, the investigation results were basically consistent with the theoretical analysis results, exerting obvious pressure relief and permeability enhancement effects on the overlying short-distance coal roadway strips.

Abstract Background Sorghum bicolor (L.) is an important bioenergy source. The stems of sweet sorghum function as carbon sinks and accumulate large amounts of sugars and lignocellulosic biomass and considerable amounts of starch, therefore providing a model of carbon allocation and accumulation for other bioenergy crops. While omics data sets for sugar accumulation have been reported in different genotypes, the common features of primary metabolism in sweet genotypes remain unclear. To obtain a cohesive and comparative picture of carbohydrate metabolism between sorghum genotypes, we compared the phenotypes and transcriptome dynamics of sugar-accumulating internodes among three different sweet genotypes (Della, Rio, and SIL-05) and two non-sweet genotypes (BTx406 and R9188). Results Field experiments showed that Della and Rio had similar dynamics and internode patterns of sugar concentration, albeit distinct other phenotypes. Interestingly, cellulose synthases for primary cell wall and key genes in starch synthesis and degradation were coordinately upregulated in sweet genotypes. Sweet sorghums maintained active monolignol biosynthesis compared to the non-sweet genotypes. Comparative RNA-seq results support the role of candidate Tonoplast Sugar Transporter gene (TST), but not the Sugars Will Eventually be Exported Transporter genes (SWEETs) in the different sugar accumulations between sweet and non-sweet genotypes. Conclusions Comparisons of the expression dynamics of carbon metabolic genes across the RNA-seq data sets identify several candidate genes with contrasting expression patterns between sweet and non-sweet sorghum lines, including genes required for cellulose and monolignol synthesis (CesA, PTAL, and CCR), starch metabolism (AGPase, SS, SBE, and G6P-translocator SbGPT2), and sucrose metabolism and transport (TPP and TST2). The common transcriptome features of primary metabolism identified here suggest the metabolic networks contributing to carbon sink strength in sorghum internodes, prioritize the candidate genes for manipulating carbon allocation with bioenergy purposes, and provide a comparative and cohesive picture of the complexity of carbon sink strength in sorghum stem.

Background: Psychological status is a decisive factor for regulating the lung cancer chemotherapy patients’ levels of fatigue and hope. Using the PERMA (Positive Emotion, Engagement, Relationships, Meaning, and Accomplishment) framework. We aimed to explore the influences of the psychological intervention on the patients’ negative emotion, cancer-related fatigue, and level of hope. Method: A total of 100 lung cancer chemotherapy patients admitted in Wuhan No.4 Hospital, China, from Jan 2018 to Aug 2019 were enrolled as research objects divided into the control group and observation group. Positive psychological intervention using the PERMA framework was given to the observation group. The scores of Post-Traumatic Growth Inventory (PTGI), Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS), Cancer Fatigue Scale (CFS), and Herth Hope Index (HHI) were evaluated and compared in the two groups. Protective seam mining is an effective gas pressure relief method in deep mining. Effective theoretical calculation methods in the current studies on the prediction of pressure relief protection effect of interbed coal and rock masses and their distribution laws are lacking. Thus, the evaluation and research with respect to pressure relief effect in protective seam mining relatively lag behind. This situation restricts the engineering feasibility evaluation and decision making in the protective seam mining. Therefore, the influence of upper protective seam mining on the pressure relief protection effect of coal and rock mass between underlying beds was investigated in this study. On the basis of an analysis of concrete engineering projects, a mechanical model was constructed for the pressure relief protection effect of upper protective seam mining on the coal and rock mass between underlying beds. The distribution equation of pressure relief expansion ratio in the underlying protected seam was also derived. The influence laws of main influencing factors on the pressure relief protection effect of the protected seam were revealed as well. In the end, the pressure relief effect was analyzed and verified for the protected seam before and after mining through numerical simulation and similarity simulation test. The pressure relief effect of upper protective seam mining on the coal and rock mass between underlying beds and the distribution characteristics were deeply explored in this study, which could provide a theoretical reference for the decision making in the gas extraction engineering design and pre-evaluation of extraction effect. Results show that the effective pressure relief zone (expansion rate>0.3%) of the protected seam beneath the goaf is located within the range of approximately 40 m from the coal wall to the rear part. It also presents an approximate “Λ-shaped distribution characteristic,” that is, it experiences migration and evolution with the advancement in the working face. Moreover, the peak pressure relief lags behind the coal wall on the working face by nearly 10–20 m. In the numerical simulation, the expansion ratio in the goaf also presents an approximate “Λ-shaped distribution.” Its effective pressure relief zone is the 50 m range from the coal wall to the rear part of the goaf, and the peak value lags behind the coal wall by around 15 m. The theoretical results and numerical simulation results are basically consistent with the physical experiment results. The expansion rates are 1.25%, 1.268%, and 1.32%, respectively. The elastic modulus E of coal seam and interbed spacing H are the main influencing factors of the swelling deformation and are negatively correlated with the expansion ratio. In the actual mining process, E and H of the protected layer can be measured to infer the expansion deformation of the protected layer.

The conditions of the hard roof in my country vary greatly, ranging from a few meters to tens of meters or even hundreds of meters in thickness. The coal reserves under the hard roof account for about one-third of the total reserves. At present, nearly 40% of fully mechanized mining faces that belong to the hard roof working face has the problem of mining in the hard roof working face. This has a serious impact on the load-bearing stability of the fully mechanized support, and it is urgent to solve the problem of strong underground pressure dynamic disaster under the condition of the hard roof. Based on the research background of 11129 working face in Zhangji Coal Mine in Huainan, this paper constructs a mechanical model of the interaction between the cantilever beam of the hard roof of the stope and the support and then the force distribution equation of the bearing capacity of the supports at different positions of the roof during the periodical rotation of the working face is obtained, which is combined with numerical simulation and engineering site to verify. The research results show that the bearing stability of the support is significantly affected by factors such as the buried depth H, the roof elastic modulus E, the roof thickness h, and the roof cantilever length l0, but most of the influencing factors belong to the geological occurrence conditions of the coal seam itself. Presplit blasting of the roof in advance can effectively destroy the integrity of the roof itself and reduce the periodic breaking distance, thereby improving the apparent environment of roof rock pressure and reducing the force on the working face support. According to the specific geological environment of the 11129 working face, the cutting plan of the cut hole is given out, along the groove 0∼200 and 200∼700 m of the concrete presplitting blasting. The stent force of the top-cutting section fluctuates in the range of 3360.8–4347.9 kN in the range of control top distance (5275∼6175 mm). The load-bearing pressure of the stent before top-cutting is about 1.8 times of that after top-cutting. The pressure distribution of the hydraulic support in the numerical simulation stope is approximately “Λ” in the middle and the low on the two sides. The simulated value is slightly smaller than the theoretical calculation value. The reason is that the goaf is backfilled during the simulation process, and the roof has a certain ability to bear the load. Real-time understanding of the “roof-support” mechanical relationship can effectively ensure the safe and efficient mining of the 11129 working face and also provide experience for the subsequent mining of group B coal in the later period.

Due to the change of ground stress environment caused by underground coal mining, the intense lateral mining stress concentration is formed around the stope; so section coal pillar is generally set up to bear the mining pressure, but the different sizes of coal pillars have obvious influence on the bearing capacity of those pillars and the characteristics of mining pressure. Mastering the mechanism characteristics by which coal pillars bearing capacity and mining stress distribution is crucial to identify the reasonable coal pillar size and give full play to the bearing role of section coal pillar, given their importance for the safety and bearing stability of engineering rock mass in underground coal mining. Therefore, the bearing characteristics of section coal pillar and the redistribution of mining stress are achieved with a mechanical model analysis on the basis of the analysis of coal pillar bearing and mining influence characteristics. Moreover, applying the elastic-plastic mechanics theory revealed the mechanical equations of the effective bearing size of coal pillar and redistribution of mining stress in longwall face. Combined with the analysis of a specific engineering example, the research results are as follows. During a roadway excavation, the continuous mining stress transfer occurs “stress redistribution” and the mechanical failure of bearing coal pillar consists of lateral mining and roadway side failures. The bearing coal pillar has two critical dimensions (i.e., the critical dimension We of the self-bearing stability coal pillar and the critical dimension Wp of failure through the coal pillar). The mechanical state of the lateral mining stress redistribution and bearing coal pillar is divided into the three situations: ① when the width of coal pillar W We, three stress concentration areas are present, and the coal pillar at this stage is in a self-bearing stable state. Among all these factors, only the size of coal pillar is completely controllable, so the aspects of safe bearing and reserved size design of coal pillar, after estimating the critical size of coal pillar, the coal pillar size design is carried out according to the mine pressure control needs of mining engineering, and the cohesion, internal friction angle, interlayer friction coefficient, and coal seam mining height are improved by artificial technology, so as to realize the resource safe and efficient mining of all kinds of coal seam mining conditions; in the calculation of wide coal pillar size, the advance mining stress concentration at the end of the self-working face should be taken as the mining load condition, and the reserved size meets the condition of W > We, thereby ensuring the stable bearing of the wide coal pillar despite the advanced mining stress concentration during the self-working face mining; in the calculation of narrow coal pillar size, the lateral mining stress concentration before mining should be taken as the mining load condition and the reserved size meets the condition W

Rock dynamic constitutive model plays an important role in understanding dynamic response and addressing rock dynamic problems. Based on elastoplastic mechanics and damage mechanics, a dynamic constitutive model of rock coupled with elastoplastic damage is established. In this model, unified strength theory is taken as the yield criterion; to reflect the different damage evolution law of rocks under tension and pressure conditions, the effective plastic strain and volumetric plastic strain are used to represent the compressive damage variable and the equivalent plastic strain is used to represent the tensile damage variable; the plastic hardening behavior and strain rate effect of rocks are characterized by piecewise function and dynamic increase factor function, respectively; Fortran language and LS-DYNA User-Defined Interface (Umat) are used to numerically implement the constitutive model; the constitutive model is verified by three classical examples of rock uniaxial and triaxial compression tests, rock uniaxial tensile test, and rock ballistic test. The results show that the constitutive model can describe the dynamic and static mechanical behavior of rock comprehensively.

Sorghum is an important crop widely used for food, feed, and fuel. Transcriptome-wide studies of 3′ untranslated regions (3′UTR) using regular RNA-seq remain scarce in sorghum, while transcriptomes have been characterized extensively using Illumina short-read sequencing platforms for many sorghum varieties under various conditions or developmental contexts. 3′UTR is a critical regulatory component of genes, controlling the translation, transport, and stability of messenger RNAs. In the present study, we profiled the alternative 3′UTRs at the transcriptome level in three genetically related but phenotypically contrasting lines of sorghum: Rio, BTx406, and R9188. A total of 1,197 transcripts with alternative 3′UTRs were detected using RNA-seq data. Their categorization identified 612 high-confidence alternative 3′UTRs. Importantly, the high-confidence alternative 3′UTR genes significantly overlapped with the genesets that are associated with RNA N6-methyladenosine (m6A) modification, suggesting a clear indication between alternative 3′UTR and m6A methylation in sorghum. Moreover, taking advantage of sorghum genetics, we provided evidence of genotype specificity of alternative 3′UTR usage. In summary, our work exemplifies a transcriptome-wide profiling of alternative 3′UTRs using regular RNA-seq data in non-model crops and gains insights into alternative 3′UTRs and their genotype specificity.

The mining spatial structure of isolated island face in extra-thick fully mechanized top-coal caving mining is unique, which leads to a complex mining stress distribution and serious safety hazards. In this study, combined with a specific engineering example, the mining stress distribution characteristics of isolated island face are expounded, and a bearing structural mechanical model of the continuous beam of overlying strata is established using elastic–plastic mechanics theory. The mechanical equations of the mining stress distribution and failure depth of coal–rock mass are then obtained. Comparison of theoretical calculation results with numerical simulation and field measurement results shows basically consistent stress distribution characteristics. The derived mechanical equations can provide an estimation method for the analysis of mining dynamics on isolated island face in extra-thick fully mechanized top-coal caving mining. The following conclusions are acquired. The coal–rock mass should bear not only the lateral mining superposition influence but also the advance mining influence in front of the coal wall, so the isolated island face is in the complex environment of multiple mining stress superposition. In the mining process, the maximum advance mining stress concentration factor is 4.0–6.0 and is located at the upper and lower ends of the isolated island face. The lateral mining failure depth of the coal wall of the isolated island face increases by 2.0–5.0 m under the influence of advance mining. Therefore, compared with the nonisolated island face, the mining pressure appearance is intense. The mining influence in the range of 20–30 m of the upper and lower ends is intense, and the mining stress in this area is characterized by “cone distribution.” This zone is an important hidden danger area with coal–rock mass mining instability on isolated island face, which requires special attention to avoid mining disasters. According to the analysis of the influencing mining factors and laws of isolated island face, it is concluded that the longer the isolated island face size is, the closer the goaf size on both sides of the isolated island face is, the smaller the coal seam buried depth is, the better the mechanical conditions of coal and rock medium are, and the smaller the mining height of coal seam is, the more favorable the safe mining of isolated island face is.

The application of roof-cutting and pressure-relief gob-side entry retention plays a critical role in controlling the stability of the surrounding rock at the entry, easing continuity tension and improving resource recovery ratio. The excavation of the 360,803 airway in Xinji No. 1 Mine is affected by intense mining of the 360,805 working face. Hence, to address the stability problem of surrounding rock in the 360,803 airway, rock mass blast weakening theory was used in this study to analyze the blasting stress of columnar charged rock mass and obtain the radiuses of crushed, fractured, and vibration zones under uncoupled charging conditions. The reasonable array pitch, length, and dip angle of boreholes were determined according to the pressure-relief range of the blasting fracture. The migration laws of roof strata were explored based on a mechanical model of overlying roof strata structure on the working face. Subsequently, the horizon, breaking span, and caving sequence of hard roof strata were obtained to determine the roof-cutting height of this entry. On the basis of the theory of key stratum, the number of sequences at the roof caving limit stratum and hanging roof length in the goaf were calculated, the analytical solution to critical coal pillar width was acquired, the evaluation indexes for the stability of entry-protecting coal pillars were determined, and the engineering requirements for the 25 m entry-protecting coal pillars in the 360,803 airway were met. Moreover, various indexes such as roof separation fracture, displacement of surrounding rock, and loose circle of surrounding rock in the gob-side entry were analyzed. The stability and cementation status of surrounding rock in the 360,803 airway were evaluated, and tunneling safety was ensured.

Kernel hardness is one of the most important single traits of wheat seed. It classifies wheat cultivars, determines milling quality and affects many end-use qualities. Starch granule surfaces, polar lipids, storage protein matrices and Puroindolines potentially form a four-way interaction that controls wheat kernel hardness. As a genetic factor, Puroindoline polymorphism explains over 60% of the variation in kernel hardness. However, genetic factors other than Puroindolines remain to be exploited. Over the past two decades, efforts using population genetics have been increasing, and numerous kernel hardness-associated quantitative trait loci (QTLs) have been identified on almost every chromosome in wheat. Here, we summarize the state of the art for mapping kernel hardness. We emphasize that these steps in progress have benefitted from (1) the standardized methods for measuring kernel hardness, (2) the use of the appropriate germplasm and mapping population, and (3) the improvements in genotyping methods. Recently, abundant genomic resources have become available in wheat and related Triticeae species, including the high-quality reference genomes and advanced genotyping technologies. Finally, we provide perspectives on future research directions that will enhance our understanding of kernel hardness through the identification of multiple QTLs and will address challenges involved in fine-tuning kernel hardness and, consequently, food properties.

Multiseam longwall mining-induced strata deformation and fracture propagation patterns are different from those of single-seam mining. This difference is due to interaction of the caved zones as a result of longwall mining activity at different coal seams, which severely impacts formation of subsidence and permeability of the strata after multiseam mining. Understanding this phenomenon is of great importance in order to predict the multiseam subsidence reliably, evaluate the risk of water inrush and take suitable preventive measures, and determine suitable locations for placing gas drainage boreholes. In this study, scaled physical modelling techniques are utilised to investigate strata deformation, fracture propagation characteristics, and vertical subsidence above multiseam longwall panels. The results show that magnitude of the incremental multiseam subsidence increases significantly after multiseam extraction in comparison with single-seam mining. This increase occurs to different extent depending on the multiseam mining configuration. In addition, interstrata fractures above the abutment areas of the overlapping panels propagate further towards the ground surface in multiseam extractions compared with single-seam extractions. These fractures increase the risk of water inrush in presence of underground/surface water and create highly permeable areas suitable for placing gas drainage boreholes.

The stability of panel floor, which is above confined water, is the key to determine the water inrush from the panel floor. Based on the characteristics of “lower three zones” of the panel floor, the mechanics analysis model of a floor water-resisting rock beam is established. Then, by the principle of virtual work and energy functional variational conditions, the trends of deflection and internal stress are researched in the effective water-resisting rock beam under the combined action of mining stress and water pressure. And how to determine its stability is acquired. According to the geological and mining conditions of A3 coal seam in Panxie mining area of Huainan Mining Group, three factors influencing on the stability of the floor rock beam are analyzed, such as elastic modulus, coefficient of viscosity, and water pressure. It is shown that the elastic modulus plays the most important role on the deformation of the rock beam. So, for improving the mechanical properties of the rock beam, the reinforcing floor technique has been proposed. On the one hand, it is contributed to improve the ability for resisting floor deformation. On the other hand, it can increase the coefficient of rock viscosity in water damage zones and reduce the speeds of loading and deformation in the whole rock beam. Hydrophobic decompression can effectively reduce the stress on the boundary of the rock beam, and the stability is enhanced. The research results have a guiding significance for determining whether there are water inrush risks in the panel above the confined aquifer.

The stress change law of a collapse column and the failure depth of a coal seam floor before and after mining when the fully mechanized coal mining face passes through the collapse column are investigated. Here, we present the constructed program in FISH language, render the damage variable in FLAC3D to establish the numerical model, and complete the numerical calculation. The 10–115 working faces that pass the collapse column at a coal mine in Tuanbai are identified as the research object. The floor failure is numerically simulated to assess the damage. The following results were obtained: the failure depth of the full floor is stabilized at 14.6 m; the maximum failure depth of the floor near the collapse column is 18.2 m; and the stress concentration coefficient is 1.27 times greater than that of normal mining. The calculated depth failure of the floor of the working face without structural defects is 14.6–14.7 m based on the Hoek–Brown criterion. With the collapse column, the failure depth of the floor is 16.8–17.8 m. According to the water injection test, the maximum failure depth of the floor is 18 m. The three derived values agree well with one another.

A thin basement coal seam mining model under different overlying strata conditions was developed using the discrete element software UDEC. This approach is used to discuss the safety mining of the thin basement coal seams. Fracture development in overlying and rock strata movement law in the stope was discussed. The relationship between support and surrounding rocks under different overlying strata conditions was analyzed. Lastly, a field industrial test was conducted based on the research results. A few major conclusions could be drawn. Under load transmission in loose water-bearing strata, causing a large-scaled rock strata movement to advance into the working face is easy when only one bearing stratum exists in the overlying strata. Meanwhile, the support bears strong loads, which can easily be collapsed. When two bearing strata exist in the overlying one, the upper bearing stratum can form a voussoir beam structure. Loads on the support decreased substantially compared with those under single bearing stratum, whilst the probability of pressing frame was reduced accordingly. A weathered zone above the stope was reinforced by ground J-type drilling pregrouting, thereby improving the physical and mechanical properties and increasing the bearing capacity of the rock strata in the grouting range and safety mining of the working face in the lower coal seams. Research results provide important references for the safety mining of thin basement coal seams under similar conditions.

Moso bamboo is an important forest species with a variety of ecological, economic, and cultural values. However, the gene annotation information of moso bamboo is only based on the transcriptome sequencing, lacking the evidence of proteome. The lignification and fiber in moso bamboo leads to a difficulty in the extraction of protein using conventional methods, which seriously hinders research on the proteomics of moso bamboo. The purpose of this study is to establish efficient methods for extracting the total proteins from moso bamboo for following mass spectrometry-based quantitative proteome identification. Here, we have successfully established a set of efficient methods for extracting total proteins of moso bamboo followed by mass spectrometry-based label-free quantitative proteome identification, which further improved the protein annotation of moso bamboo genes. In this study, 10,376 predicted coding genes were confirmed by quantitative proteomics, accounting for 35.8% of all annotated protein-coding genes. Proteome analysis also revealed the protein-coding potential of 1015 predicted long noncoding RNA (lncRNA), accounting for 51.03% of annotated lncRNAs. Thus, mass spectrometry-based proteomics provides a reliable method for gene annotation. Especially, quantitative proteomics revealed the translation patterns of proteins in moso bamboo. In addition, the 3284 transcript isoforms from 2663 genes identified by Pacific BioSciences (PacBio) single-molecule real-time long-read isoform sequencing (Iso-Seq) was confirmed on the protein level by mass spectrometry. Furthermore, domain analysis of mass spectrometry-identified proteins encoded in the same genomic locus revealed variations in domain composition pointing towards a functional diversification of protein isoform. Finally, we found that part transcripts targeted by nonsense-mediated mRNA decay (NMD) could also be translated into proteins. In summary, proteomic analysis in this study improves the proteomics-assisted genome annotation of moso bamboo and is valuable to the large-scale research of functional genomics in moso bamboo. In summary, this study provided a theoretical basis and technical support for directional gene function analysis at the proteomics level in moso bamboo.

Hepatocellular carcinoma (HCC) is a prevalent problem worldwide. Chemotherapy, especially cisplatin (CDDP)-based systemic chemotherapy, is the best option for advanced liver cancer. However, CDDP resistance is becoming common and hindering the clinical application of CDDP. Meanwhile, no consensus has been reached regarding the chemotherapeutic use of vasohibin 2 (VASH2), which promotes the angiogenesis and proliferation of cancer cells. In this work, a tissue microarray was used to observe VASH2 and its possible role in cancer treatment. Results showed that VASH2 was highly expressed in HCC tissues and was significantly correlated with cancer differentiation. To further investigate the efficacy and mechanism of the combination of VASH2 with anti-cancer drugs in liver cancer cells, we stably built VASH2 overexpression and knockdown cell lines. We found that VASH2 can influence the CDDP sensitivity and that the cell overexpression of VASH2 had a higher cell viability and lower apoptosis rate after CDDP exposure. We also observed that VASH2 overexpression downregulated wild-type p53, as well as suppressed the expression of the pro-apoptotic protein BCL2-associated X protein (Bax) and cleaved caspase-3 (CC-3) after treatment by CDDP. Conversely, the knockdown of VASH2 significantly inhibited these effects. In an in vivo chemosensitivity study, nude mice were subcutaneously injected with tumor cells and received CDDP treatment through intraperitoneal administration every 3 days. We found that VASH2 knockdown markedly limited the tumor growth and enhanced the CDDP toxicity and apoptosis of tumor cells. Western blot analysis revealed that tumor cells with downregulated VASH2 had a higher expression of wild-type p53, Bax, and CC-3 than control cells. Overall, our results indicated the novel roles of VASH2 in the chemoresistance of hepatocarcinoma cells to CDDP and suggested that VASH2 may be a promising anticancer target.

Atmospheric pressure room temperature plasma jets (APRTP-Js) that can emit a mixture of different active species have recently found entry in various medical applications. Apoptosis is a key event in APRTP-Js-induced cellular toxicity, but the exact biological mechanisms underlying remain elusive. Here, we explored the role of reactive oxygen species (ROS) and reactive nitrogen species (RNS) in APRTP-Js-induced apoptosis using in vitro model of HepG2 cells. We found that APRTP-Js facilitated the accumulation of ROS and RNS in cells, which resulted in the compromised cellular antioxidant defense system, as evidenced by the inactivation of cellular antioxidants including glutathione (GSH), superoxide dismutase (SOD) and catalase. Nitrotyrosine and protein carbonyl content analysis indicated that APRTP-Js treatment caused nitrative and oxidative injury of cells. Meanwhile, intracellular calcium homeostasis was disturbed along with the alteration in the expressions of GRP78, CHOP and pro-caspase12. These effects accumulated and eventually culminated into the cellular dysfunction and endoplasmic reticulum stress (ER stress)-mediated apoptosis. The apoptosis could be markedly attenuated by N-acetylcysteine (NAC, a free radical scavenger), which confirmed the involvement of oxidative and nitrative stress in the process leading to HepG2 cell apoptosis by APRTP-Js treatment.

Introduction: The risk of osteoporotic vertebral refracture (OVRF) is substantially increased after initial osteoporotic vertebral fracture (OVF). In this study, we comprehensively analyzed risk factors for OVRF in patients undergoing conservative treatment and further evaluated the predictive value of various risk factors. Methods: Basic information, past medical history and imaging parameters were collected from 151 postmenopausal OVFwomen treated conservatively. In univariate analysis, chi-square testwas used for categorical variables and independent sample t-test was used for continuous variables. Binary logistic regression was used to identify independent risk factors for variables with significant differences in univariate analysis. Then receiver operating characteristic curve analysis (ROC Curve) were drawn for the predictors and area under the curve (AUC) were calculated to evaluate the prediction accuracy. Results: Univariate analysis and binary logistic regression analysis showed that age (p < 0.05) and L3/4 paraspinal muscle fat infiltration ratio (FIR) (p < 0.05) were independent risk factors of OVRF. According to the ROC curve, the prediction accuracy of age, FIR and the two parameters together were 0.730, 0.778 and 0.813 respectively. The critical values of age and FIRwere 63.5 years and 37.1%, respectively. Conclusion: OVRF is affected by multiple factors. Age and FIR are independent risk factors of OVRF. The prediction value of integrated two independent risk factors was higher than that of a single risk factor. Postmenopausal women with initial OVF aged over 63.5 years and FIR over 37.1% were more likely to develop OVRF. [ABSTRACT FROM AUTHOR]
