Extracting statistical characteristics from radiographic films is vital for film system classification and contrast sensitivity evaluation and serves as a basis for film noise simulation. A new method for digitising radiographic films in order to extract these characteristics is presented. The method consists of a camera-based setup and image processing procedure to digitise films. Correct optical density values and granularity can be extracted from the digitised images, which are equal to results obtained by standardised measurement procedures. Specific statistical characteristics of film noise are theoretically derived and subsequently verified by the obtained data, including characteristics such as Gaussianity and spatial spectral characteristics of the optical density fluctuations. It is shown that the presented method correctly measures the granularity of film noise and can therefore replace time-consuming microdensitometer measurements traditionally required for film system classifications. Additionally, the inherent unsharpness of film systems was investigated and compared with literature data. This comparison serves as another validation approach of the presented method.

Purpose Microbeam radiation therapy is a developing technique that promises superior tumour control and better normal tissue tolerance using spatially fractionated X-ray beams only tens of micrometres wide. Radiochromic film dosimetry at micrometric scale was performed using a microdensitometer, but this instrument presents limitations in accuracy and precision, therefore the use of a microscope is suggested as alternative. The detailed procedures developed to use the two devices are reported allowing a comparison. Methods Films were irradiated with single microbeams and with arrays of 50 µm wide microbeams spaced by a 400 µm pitch, using a polychromatic beam with mean energy of 100 keV. The film dose measurements were performed using two independent instruments: a microdensitometer (MDM) and an optical microscope (OM). Results The mean values of the absolute dose measured with the two instruments differ by less than 5% but the OM provides reproducibility with a standard deviation of 1.2% compared to up to 7% for the MDM. The resolution of the OM was determined to be ~ 1 to 2 µm in both planar directions able to resolve pencil beams irradiation, while the MDM reaches at the best 20 µm resolution along scanning direction. The uncertainties related to the data acquisition are 2.5–3% for the OM and 9–15% for the MDM. Conclusion The comparison between the two devices validates that the OM provides equivalent results to the MDM with better precision, reproducibility and resolution. In addition, the possibility to study dose distributions in two-dimensions over wider areas definitely sanctions the OM as substitute of the MDM.

We describe the new, fast, high-precision microdensitometer SuperCOSMOS. Some aspects of hardware and software design that enable high-precision astrometry from photographic plates are explained. We show that the positioning repeatability of the measuring machine is less than 0.1 μ μ m standard error in either coordinate, and the absolute positional accuracy is about 0.15 μ m standard error. Furthermore, measurements of the same plate in different orientations show that the sampling errors are small (e.g. ∼ 0.2 μ m, rising to ∼ 1.0 μ m at the plate limit, for stellar images in a IIIaJ emulsion), thus allowing the extraction of relative positional information from Schmidt plates at accuracies less than 1 μ m. We demonstrate that SuperCOSMOS is capable of measuring the positions of bright stars (i.e. those more than ∼ 4 mag above the plate limit) to a precision ∼ 0.5 μ m with survey–grade photographic plates employing fine–grained emulsions.

We present an original method of digitizing and astrometrically reducing "Carte du Ciel" plate material using an inexpensive flatbed scanner, to demonstrate that for this material there is an alternative to more specialized measuring machines that are very few in number and thus not readily available. The sample of plates chosen to develop this method are original "Carte du Ciel" plates of the San Fernando zone, photographic material with a mean epoch 1903.6, and a limiting photographic magnitude ~14.5, covering the declination range of -10 < dec < -2. Digitization has been made using a commercial flatbed scanner, demonstrating the internal precision that can be attained with such a device. A variety of post-scan corrections are shown to be necessary. In particular, the large distortion introduced by the non-uniform action of the scanner is modelled using multiple scans of each plate. We also tackle the specific problems associated with the triple-exposure images on some plates and the grid lines present on all. The final measures are reduced to celestial coordinates using the Tycho-2 Catalogue. The internal precision obtained over a single plate, 3microns ~ 0.18" in each axis, is comparable to what is realized with similar plate material using slower, less affordable, and less widely available conventional measuring machines, such as a PDS microdensitometer. The accuracy attained over large multi-plate areas, employing an overlapping plate technique, is estimated at 0.2"., 16 pages, 19 figures and 3 tables. Accepted for publication in A&A

We have identified a large sample of probable low mass members of the young open cluster IC2391 based on optical (VRIZ) and Infrared (JHK$_s$) photometry. Our sample includes 50 probable members and 82 possible members, both very low mass stars and brown dwarfs. We also provide accurate positions for these stars and brown dwarf candidates derived from red UK Schmidt plates measured using the microdensitometer SuperCOSMOS. Assuming an age of 53 Myr, we estimate that we have reached a mass of 0.025 M$_\odot$, if the identified objects are indeed members of IC2391., Accepted ApJ

We present a new survey of emission line galaxies, performed with the ESO 1 m Schmidt telescope equipped with the 4° objective prism using IIIa-J photographic emulsion. The plates are digitized with the MAMA microdensitometer. A subsequent reduction of the block scans gives redshifts with a mean accuracy of 160 km/s‐1, and spectrophotometric measurements of the intensity and equivalent widths of the principal emission lines. A brief discussion is given of the possible extension of quantitative reduction of slitless spectroscopy to archive plates and future large CCD array frames.

Quantitative techniques in immunohistochemistry are needed, but they are rarely applied because of doubtful reproducibility. We have developed a method for the detection of collagen types I and III in situ. The method applied was a two-step immuno-alkaline phosphatase technique with visualization of the end-product with Fast Red. The staining intensity was measured with a microdensitometer and the results expressed as ratios. The method yielded results that were unaffected by variations in tissue section thickness but which were proportionally related to time and antigen concentrations. Leiomyoma tissue, with a ratio of collagen types I and III of approximately 1.0, was used to establish the appropriate dilutions of the antibodies, thus assuring identical optical densities. By having the leiomyoma tissue sections incubated together with the heart tissue specimens, leiomyoma tissue was also helpful in correcting deviations from the 1.0 ratio. Accurate measurements of collagen type I/III ratios in normal human heart specimens were obtained with the present quantitative immunohistochemical technique.

We have developed microdensitometer-computer correlation techniques to analyze the arrangement of microtubule arms and bridges (i.e., microtubule-associated proteins [MAPs]). A microdensitometer was used to scan immediately adjacent to the wall of longitudinally sectioned microtubules in positive transparency electron micrographs. Signal enhancement procedures were applied to the digitized densitometer output to produce a binary sequence representing the apparent axial spacing of MAP projections. These enhanced records were analyzed in two ways. (a) Autocorrelograms were formed for each record and correlogram peaks from a group of scans were pooled to construct a peak frequency histogram. (b) Cross-correlation was used to optimize the match between each enhanced record and templates predicted by different models of MAP organization. Seven symmetrical superlattices were considered as well as single axial repeats. The analyses were repeated with randomly generated records to establish confidence levels. Using the above methods, we analyzed the intrarow bridges of the Saccinobaculus axostyle and the MAP2 projections associated with brain microtubules synthesized in vitro. We confirmed a strict 16-nm axial repeat for axostyle bridges. For 26 MAP2 records, the only significant match was to a 12-dimer superlattice model (P less than 0.002). However, we also found some axial distances between MAP2 projections which were compatible with the additional spacings predicted by a 6-dimer superlattice. Therefore, we propose that MAP2 projections are arranged in a "saturated 12-dimer, unsaturated 6-dimer" superlattice, which may be characteristic of a wide variety of MAPs.

The Feulgen-DNA contents of human leukocytes, sperm, and oral squames were investigated by scanning and integrating microdensitometry, both with and without correction for residual distribution error and glare. Maximally stained sperm had absorbances which at lambdamax exceeded the measuring range of the Vickers M86 microdensitometer; this potential source of error could be avoided either by using shorter hydrolysis times or by measuring at an off-peak wavelength. Small but statistically significant apparent differences between leukocyte types were found in uncorrected but not fully corrected measurements, and some apparent differences disappeared when only one of the residual instrumental errors was eliminated. In uncorrected measurements, the apparent Feulgen-DNA content of maximally stained polymorphs measured at lambdamax was significantly lower than that of squames, while in all experimental series uncorrected measurements showed apparent diploid:haploid ratios significantly greater than two. In fully corrected measurements no significant differences were found between leukocytes and squames, and in four independent estimations the lowest diploid:haploid ratio found was 1.99 +/- 0.05, and the highest 2.03 +/- 0.05. Discrepancies found in uncorrected measurements could be correlated with morphology of the nuclei concerned. Glare particularly affected measurements of relatively compact nuclei such as those of sperm, polymorphs and lymphocytes, while residual distribution error was especially marked with nuclei having a high perimeter:area ratio (e.g. sperm and polymorphs). Uncorrected instrumental errors, especially residual distribution error and glare, probably account for at least some of the previously reported apparent differences between the Feulgen-DNA contents of different cell types. On the basis of our experimental evidence, and a consideration of the published work of others, it appears that within the rather narrow limits of random experimental error there seems little or no reason to postulate either genuine differences in the amounts of DNA present in the cells studied, or nonstoichiometry of a correctly performed Feulgen reaction.

The temperature dependence of single-crystal elastic constants of synthetic stoichiometric MgAl_2O_4 spinel has been measured by the light-sound scattering technique in the Raman-Nath region. The crystal is set into forced vibration by a single crystal LiNbO_3 transducer coupled to one crystal face. A He-Ne Laser beam is diffracted by the stress-induced birefringence inside the crystal. The diffraction angle is determined from the distance between two spots exposed on a photographic plate by the first order diffracted beams as measured by a microdensitometer. The sound wavelength inside the crystal is then inferred from the laser diffraction angle. Combining the sound wavelength with the measured transducer frequency, the velocity inside the crystal is determined typically to a precision of 0·05 per cent. In this method, the measurement of velocity is not dependent on either the determination of sample length or on phase shifts at sample-transducer interface. Velocities of four pure modes, L//[001], T//[001], L//[110], and T//[110](P//[110] are measured in the temperature range between 293 and 423°K. A linear temperature dependence is fit to the data by a least square method.

Functional differences among various portions of the cerebral cortex are often correlated with differing cortical layering patterns. Convenient, accurate techniques for scoring layering should therefore prove useful in electrophysiological as well as anatomical investigations. We report the application of a computer-controlled scanning microdensitometer as a means of rapid measurement of optical densities in histological sections of monkey visual cortices, areas 17 and 18. The technique readily permits recognition of the previously defined cortical layers and suggests that still finer consistent layering patterns exist; it provides objective "fingerprints" of cortical regions which facilitate comparisons of structure from area to area and from animal to animal. The procedure should serve also to score the positions of autoradiographic grains, degenerating axonal terminals, and other labeled structures, and to allow the comparison of preparations stained by various techniques.

The existence of precursory bulge along an incipient fracture zone in a uniaxially compressed Westerly granite sample has been investigated by two optical methods. The first method is the method of slit diffraction. The cross section of a cylindrical rock sample is monitored by bringing two straightedges next to the rock sample to form two slits (each slit being formed between one straightedge and one side of the rock sample) and by illuminating alternately the two slits with a collimated laser beam. The Fraunhofer diffraction pattern is recorded on film in the direction perpendicular to the straightedge and can be interpreted as the absolute value squared of the one-dimensional spatial Fourier transform of the slit under certain conditions, thereby providing a simple method of magnification of the rock surface geometry. The conditions under which the Fraunhofer diffraction pattern can be interpreted as the absolute value squared of a one-dimensional Fourier transform are related to the radius of the rock sample, width of the slit, position of the recording film plane, and nature of deformation of the rock surface and are presented in this paper. The films are digitized by a microdensitometer. The data are analyzed by digital filtering and interpolation techniques to give a strain resolution of 10^(−5). During a test with the slit diffraction method, strain inhomogeneities in terms of local bulges indicative of incipient failure zones were found to develop at ∼92% of the uniaxial compressive strength, and their propagation is traced at 2.66-s intervals until failure. Local strains in the incipient failure zones are of the order of 10^(−2) before failure takes place. Because of the large amplitude of the strain inhomogeneity prior to failure recorded by the slit diffraction method, we then tried the faster method of recording without magnification by a motion picture camera. In the second test a precursory bulge in the middle of the sample first appeared at ∼3.75 s prior to failure at a load of >99.7% of the uniaxial compressive strength. The bulge developed rapidly in successive frames until eventually a failure plane passed through this sharp bulge. The results from both tests demonstrate the formation of a concentrated weak zone as a result of the interaction and coalescence among the microcracks in the final stage of the test, which then develop into fracture zones. The bulging is the result of accentuated deformation in the weak zone because of its reduced deformation moduli. It is considered that the local bulge and orientation of the fracture zones in the first test were controlled by the stress concentration at the sample-load block interface, whereas those in the second test were controlled mainly by the inhomogeneity in the material properties within the sample. The precursor times of both tests do not fit into the empirical relationship between precursor time and fault dimension as derived from earthquakes and mine rock bursts. The precursor times of these tests are too long by 3 orders of magnitude in comparison with those given by the empirical relationship.

SUMMARY A simple and easily adaptable interface between a Vickers M86 scanning microdensitometer and microinterferometer and a Z80 CP/M based microcomputer (Zenith Z89) is described. The hardware is supplemented by a software package written in Fortran-66, linked with an assembly routine for two-dimensional spot control and measurement. Facilities are provided for background correction, image processing, calibration of the spot movement, correction for glare, diffraction and finite spotsize and computer-aided focusing. Results can be plotted on a matrix printer and on a graphical unit as ruled surface plot, as isophase or isodensity contour diagram or in three-dimensional projection with hidden line removal.
