Abstract Hyperaccumulating plants are able to (hyper)accumulate high concentrations of metal(loid)s in their above-ground tissues without any signs of toxicity. Studies on the root-associated microbiome have been previously conducted in relation to hyperaccumulators, yet much remains unknown about the interactions between hyperaccumulating hosts and their microbiomes, as well as the dynamics within these microbial communities. Here, we assess the impact of the plant host on shaping microbial communities of three naturally occurring populations of Noccaea species in Slovenia: Noccaea praecox and co-occurring N. caerulescens from the non-metalliferous site and N. praecox from the metalliferous site. We investigated the effect of metal enrichment on microbial communities and explored the interactions within microbial groups and their environment. The abundance of bacterial phyla was more homogeneous than fungal classes across all three Noccaea populations and across the three root-associated compartments (roots, rhizosphere, and bulk soil). While most fungal and bacterial Operational Taxonomic Units (OTUs) were found at both sites, the metalliferous site comprised more unique OTUs in the root and rhizosphere compartments than the non-metalliferous site. In contrast to fungi, bacteria exhibited differentially significant abundance between the metalliferous and non-metalliferous sites as well as statistically significant correlations with most of the soil parameters. Results revealed N. caerulescens had the highest number of negative correlations between the bacterial phyla, whereas the population from the metalliferous site had the fewest. This decrease was accompanied by a big perturbation in the bacterial community at the metalliferous site, indicating increased selection between the bacterial taxa and the formation of potentially less stable rhizobiomes. These findings provide fundamentals for future research on the dynamics between hyperaccumulators and their associated microbiome.

Metal contaminated soils are increasing worldwide. Metal-tolerant plants growing on metalliferous soils are fascinating genetic and microbial resources. Seeds can vertically transmit endophytic microorganisms that can assist next generations to cope with environmental stresses, through yet poorly understood mechanisms. The aims of this study were to identify the core seed endophyte microbiome of the pioneer metallophyte Crotalaria pumila throughout three generations, and to better understand the plant colonisation of the seed endophyte Methylobacterium sp. Cp3. Strain Cp3 was detected in C. pumila seeds across three successive generations and showed the most dominant community member. When inoculated in the soil at the time of flowering, strain Cp3 migrated from soil to seeds. Using confocal microscopy, Cp3-mCherry was demonstrated to colonise the root cortex cells and xylem vessels of the stem under metal stress. Moreover, strain Cp3 showed genetic and in planta potential to promote seed germination and seedling development. We revealed, for the first time, that the seed microbiome of a pioneer plant growing in its natural environment, and the colonisation behaviour of an important plant growth promoting systemic seed endophyte. The highly specialized flora of localities affected by former metal ore mining and metallurgy is endangered by succession or intentional afforestation all over Europe. Its last remnants therefore deserve our attention. We examined whether Scots pine encroaching on a heavy-metal grassland (Olkusz Zn-Pb mining area, S Poland) is outcompeting specialized herbaceous species, as has been observed elsewhere. Plant species composition and richness sampled at 124 plots were analysed in relation to pine stand parameters (canopy cover, stand age, stand basal area), abiotic environmental factors (e.g. soil properties) and spatial variables (e.g. plot coordinates). Plots were divided into three shading categories and compared in terms of vegetation and habitat parameters. Scots pine outcompeted several light-demanding species, leading to a decrease of total species richness and cover. Characteristic species of this grassland ( Biscutella laevigata, Silene vulgaris) and some metal-tolerant plants were clearly insensitive to shading. For these early successional species, more important was the availability of microsites with shallow skeletal soil or bare subsoil. Tree stand parameters differently affected grassland vegetation: canopy cover caused primarily a compositional shift in the community, while stand age was the principal agent of decline in species richness. Scots pine increased the soil concentrations of available Ca and Mg, and negatively affected soil development (organic matter and mineral particle accumulation), which might be beneficial to some shade-tolerant grassland species. Maintaining the studied grassland's present species richness and composition would require cutting woody plants less frequently than recommended for dry grasslands of non-metalliferous sites, and disturbing the soil surface. Campo rupestre is a kind of Brazilian rocky outcrop with high biodiversity and many endemic and threatened species. It occurs mainly in the Espinhaço Range in a contact region between Cerrado and/or Caatinga and Atlantic Forest. The Espinhaço Range is recognized as a region with the highest floristic diversity in South America and with many endemic species, most of which are associated with rocky outcrop environments. These, among other peculiarities, recently granted the Espinhaço Range the status of Biosphere Reserve. The relationship between soil and vegetation was studied in campo rupestre areas with quartzite and itabirite rocks. Three habitats in both lithologies were defined by geomorphology as: 1. Slopes with grasslands; 2. Plateaus with grasslands and 3. Valleys with woody savannas. In each lithology, 30 plots (10 × 10 m), 10 in each habitat, were defined. The species and their respective coverage were recorded and soil was sampled to perform chemical and physical analyses. The analyzed soils were similar in being sandy, acidic and with low fertility. Nevertheless, they exhibited differences in chemical and physical properties. Altogether there were 272 species, belonging to 70 families. The canonical correspondence analysis of soil variables and species coverage showed a clear segregation of lithological sites due mainly to the exchangeable content of Ca, Cu, Mg, Mn and S; soil particle size - central tendency and sorting; and the percentage of silt, fine soil and bare rocks. Heavy metal distribution in medicinal plants is gaining importance not only as an alternative medicine, but also for possible concern due to effects of metal toxicity. The present study has been focused on emphasizing the heavy metal status and bioaccumulation factors of V, Mn, Fe, Co, Cu, Zn, Se (essential metals) and Cr, Ni, Cd, As and Pb (potentially toxic metals) in medicinal plants grown under two different environmental conditions e.g., near to Khetri copper mine and those in fertile soils of Haridwar, both in India, using Instrumental Neutron Activation Analysis (relative method) and Atomic Absorption Spectrometry. The copper levels in the medicinal plants from Khetri were found to be 3-4 folds higher (31.6-76.5 mg kg-1) than those from Haridwar samples (7.40-15.3 mg kg-1), which is correlated with very high copper levels (763 mg kg-1) in Khetri soil. Among various heavy metals, Cr (2.60-5.92 mg kg-1), Cd (1.47-2.97 mg kg-1) and Pb (3.97-6.63 mg kg-1) are also higher in concentration in the medicinal plants from Khetri. The essential metals like Mn (36.4-69.3 mg kg-1), Fe (192-601 mg kg-1), Zn (24.9-49.9 mg kg-1) and Se (0.13-0.91 mg kg-1) and potentially toxic metals like Ni (3.09-9.01 mg kg-1) and As (0.41-2.09 mg kg-1) did not show much variations in concentration in the medicinal plants from both Khetri and Haridwar. The medicinal plants from Khetri, e.g., Ocimum sanctum, Cassia fistula, Withania somnifera and Azadirachta Indica were found rich in Ca and Mg contents while Aloe barbadensis showed moderately high Ca and Mg. Higher levels of Ca-Mg were found to correlate with Zn (except Azadirachta Indica). The bioaccumulation factors (BAFS) of the heavy metals were estimated to understand the soil-to-plant transfer pattern of the heavy metals. Significantly lower BAF values of Cu and Cr were found in the medicinal plants from Khetri, indicating majority fraction of these metals are precipitated and were immobilized species unsuitable for plant uptake.
