4. Third Time's a Charm: a Tripartite Approach for Teaching Project Organization to Students

Abstract

Collaborative statisticians are instrumental to maintaining the rigor and reproducibility of multidisciplinary projects on which they collaborate. Clear, complete, well-annotated records of data, its revising, analysis, interpretation, and presentation are essential. Many students do not know how to systematically and consistently organize their digital files and cannot reliably replicate (or find!) work that may have been performed on a collaborative project. This article describes a newly developed and required one semester collaborative statistics course that includes the explicit teaching of project organization in lecture, emphasis of these skills on linked homework assignments, and their closely supervised practice through a mentored collaborative project with multidisciplinary researchers. The tripartite design provided exposure to project organization concepts during the course, encouraged straightforward implementation through homework assignments, and then challenged students with a real-world experience during a collaborative project. Abstract

Background: Ovarian hormones are known to modulate the immune system in the female genital tract (FGT). We sought to define the impact of the menstrual cycle on the mucosal HIV target cell levels, and tissue-resident CD4 T cells. Materials and methods: Here, we characterized the distribution, phenotype, and function of CD4 T cells with special emphasis on HIV target cells (CCR5+ and a4b7+) as well as tissue-resident memory (TRM; CD69+ and CD103+) CD4 T cells in FGT of cycling women. Peripheral blood and Endocervical cells (ECcollected from cytobrush) were collected from 105 healthy women and performed multicolor flow cytometry to characterize the various subsets of CD4 T cells. Cervicovaginal lavage (CVL) were collected for cytokine analysis and plasma were collected for hormonal analysis. All parameters were compared between follicular and luteal phase of menstrual cycle. Results: Our findings revealed no significant difference in the blood CD4 T cell subsets between the follicular and luteal phase. However, in EC, the proportion of several cell types was higher in the follicular phase compared to the luteal phase of menstrual cycle, including CCR5+a4b7-cells (p=0.01), CD69+CD103+ TRM (p=0.02), CCR5+CD69+CD103+ TRM (p=0.001) and FoxP3+ CD4 T cells (p=0.0005). In contrast, a4b7+ CCR5-cells were higher in the luteal phase (p=0.0004) compared to the follicular phase. In addition, we also found that hormonal levels (P4/E2 ratio) and cytokines (IL-5 and IL-6) were correlated with CCR5+ CD4 T cells subsets during the follicular phase of the menstrual cycle. Conclusion: Overall, these findings suggest the difference in the expression of CCR5 and a4b7 in TRM CD4 T cell subsets in endocervix of HIV seronegative women between the follicular and luteal phase. Increase in the CCR5+ expression on TRM subsets could increase susceptibility to HIV infection during follicular phase of the menstrual cycle. [ABSTRACT FROM AUTHOR]

Abstract

Background: The syndemic of HIV, substance use (SU), and mental illness has serious implications for HIV disease progression among women. We described co-utilization of HIV care, SU treatment, and mental health treatment among women with or at risk for HIV. Methods: We included data from women with or at risk for HIV (n = 2559) enrolled in all 10 sites of the Women's Interagency HIV Study (WIHS) from 2013 to 2020. Current SU was defined as self-reported, non-medical use of drugs in the past year, excluding use of only marijuana. Tobacco and alcohol were assessed separately. We described co-utilization of SU treatment, tobacco and alcohol use treatment, HIV care, and mental health care in the past year among women who were eligible for each service. We compared service utilization by those who did/did not utilize SU treatment using Wald Chi-square tests. Results: Among women with current SU (n = 358), 42% reported utilizing SU treatment. Among those with current SU+HIV (n = 224), 84% saw their HIV provider, and 34% saw a mental health provider. Among women with current SU+heavy alcohol use (n = 95), 18% utilized alcohol use treatment; among current SU+tobacco use (n = 276), 8% utilized tobacco use treatment. Women who utilized SU treatment had higher utilization of alcohol use treatment (59% vs. 5%; P <.001) and tobacco use treatment (12% vs. 5%; P =.028). HIV care engagement was high regardless of SU treatment. Conclusions: We found high engagement in SU and HIV care, but low engagement in alcohol and tobacco use treatment. Integrated SU treatment services for women, including tobacco/alcohol treatment and harm reduction, are needed to optimize treatment engagement and HIV care continuum outcomes. Abstract

Rice bean (Vigna umbellata) is a legume that belongs to Vigna genus. Native to Indo-Chinese region, it is considered to be an ‘under-utilized’ or ‘orphan’ crop. Rice bean is known to possess high nutritional potential and antioxidant activity. But the use of rice bean supplementation in routine diet is limited despite its high nutritional profile due to the presence of non-nutritional factors. Thus, various pre-treatments like soaking, germination, oven roasting, sand roasting, boiling and pressure cooking at different time and temperature were carried out to reduce the anti-nutritional content of rice bean and to study its effect on antioxidant activity and phytochemical content. All the pre-treatments were able to significantly reduce the anti-nutrient content in rice bean, but germination showed the maximum reduction. Also germinated rice bean showed the maximum antioxidant potential and maximum content of total phenols, total flavonoids, vitamin C and carotenoids. Rice bean has been underutilized so far, owing to its antinutrient content and low popularity. This experiment attempted to use low cost processing to reduce the content of antinutrients and track the antioxidant content in rice bean. The concluded processing could be adopted for commercial applications for dietary supplementation.

Abstract

Integrase strand-transfer inhibitors (INSTIs) are associated with weight gain in women living with HIV (WLH). Relationships between drug exposure, baseline obesity, and INSTI-associated weight gain remain unclear. Data from 2006 to 2016 were analyzed from virally suppressed WLH enrolled in the Women's Interagency HIV Study, who switched/added an INSTI to antiretroviral therapy: [raltegravir (RAL), dolutegravir (DTG), or elvitegravir (EVG)]. Percent body weight change was calculated from weights obtained a median 6 months pre-INSTI and 14 months post-INSTI initiation. Hair concentrations were measured with validated liquid chromatography-mass spectrometry (MS)/MS assays. Baseline (preswitch) weight status evaluated obese (body mass index, BMI, ≥30 kg/m2) versus nonobese (BMI <30 kg/m2). Mixed models examined the drug hair concentration*baseline obesity status interaction for each INSTI. There were 169 WLH included: 53 (31%) switched to RAL, 72 (43%) to DTG, and 44 (26%) to EVG. Women were median age 47–52 years, predominantly Non-Hispanic Black, median CD4 counts >500 cells/mm3, >75% with undetectable HIV-1 RNA. Over ∼1 year, women experienced median increases in body weight: 1.71% (−1.78, 5.00) with RAL; 2.40% (−2.82, 6.50) with EVG; and 2.48% (−3.60, 7.88) with DTG. Baseline obesity status modified the relationship between hair concentrations and percent weight change for DTG and RAL (p's < 0.05): higher DTG, yet lower RAL concentrations were associated with greater weight gain among nonobese women. Abstract

Specific energy prediction is critically important to enhance field performance of agricultural implements. It enables optimal utilization of tractor power, reduced inefficiencies, and identification of comprehensive inputs for designing energy-efficient implements. In this study, A 3-5-1 artificial neural network (ANN) model was developed to estimate specific energy requirement of a vertical axis rotor type intra-row weeding tool. The depth of operation in soil bed, soil cone index, and forward/implement speed ratio (u/v) were selected as the input variables. Soil bin investigations were conducted using the vertical axis rotor (RVA), interfaced with draft, torque, speed sensors, and data acquisition system to record dynamic forces employed during soil–tool interaction at ranges of different operating parameters. The depth of operation (DO) had the maximum influence on the specific energy requirement of the RVA, followed by the cone index (CI) and the u/v ratio. The developed ANN model was able to predict the specific energy requirements of RVA at high accuracies as indicated by high R2 (0.91), low RMSE (0.0197) and low MAE (0.0479). Findings highlight the potential of the ANN as an efficient technique for modeling soil–tool interactions under specific experimental conditions. Abstract

Abstract Exposure to mechanical stresses developed due to different components of spraying system may cause permanent damage to the microbes of bio-pesticide solutions. To achieve maximum effectiveness of spray application, proper selection of nozzle size and operating pressure is critical. Two different classes of bio-pesticides viz. bacterial-based Bacillus thuringiensis (BBP solution) and fungal-based Beauveria bassiana (FBP solution) were evaluated for spray application by three hollow cone nozzles (1.14, 2.35 and 3.56 mm in diameter) at four operating pressures (145, 245, 345 and 445 kPa). It was observed that viability of bio-pesticide solutions increases with decrease in operating pressure and increase in nozzle size. The nozzle orifice diameter and operating pressure of the spraying system were optimized to get desired application rate, volume median diameter (VMD) and coefficient of variation (CV) of spray for maximum microbial viability of bio-pesticide solutions. The optimized values of application rate (323.94-and 325.58 L ha−1), VMD (225.88 and 231.89 µm), CV (84.49 and 85.99%) and viability (6.63 × 106 and 6.06 × 106 cfu ml−1) were obtained for BBP and FBP solutions, respectively at nozzle orifice size of 3.56 mm and 145 kPa operating pressure. Abstract

Objectives. The seat dimensions of self-propelled combine harvesters are designed without consideration of body dimensions of the user population and vary with make and models of combines. Methods. This research proposes a method to determine the percentage of seat match, upper mismatch and lower mismatch using the anthropometric body dimensions of Indian harvester operators. The seat dimensions of five popular combine harvester makes were measured and compared to the body dimensions of the operators. Results. The selected seats had 100, 44–70, 81–96, 98–100, 63–83, 59–94 and 55–97% mismatch for seat height, seat length, seat pan breadth, seat backrest height, upper backrest breadth, lower backrest breadth and steering wheel clearance, respectively. These data were used to recommend seat dimensions for combine harvesters. Seat height, seat length, seat pan breadth, seat backrest height, upper backrest breadth, lower backrest breadth and steering wheel clearance are recommended as 399, 362, 456, 400, 243, 386 and 190 mm, respectively. Conclusions. The recommended seat dimensions matched the user population anthropometric dimensions 94–100%. This approach will help to assess seat dimensions based on anthropometric data for a comfortable posture to prevent health risks such as musculoskeletal disorders (MSDs) among operators. Abstract

Background To evaluate the effect of cumulative human immunodeficiency virus (HIV)-1 viremia on aging-related multimorbidity among women with HIV (WWH), we analyzed data collected prospectively among women who achieved viral suppression after antiretroviral therapy (ART) initiation (1997–2019). Methods We included WWH with ≥2 plasma HIV-1 viral loads (VL) <200 copies/mL within a 2-year period (baseline) following self-reported ART use. Primary outcome was multimorbidity (≥2 nonacquired immune deficiency syndrome comorbidities [NACM] of 5 total assessed). The trapezoidal rule calculated viremia copy-years (VCY) as area-under-the-VL-curve. Cox proportional hazard models estimated the association of time-updated cumulative VCY with incident multimorbidity and with incidence of each NACM, adjusting for important covariates (eg, age, CD4 count, etc). Results Eight hundred six WWH contributed 6368 women-years, with median 12 (Q1–Q3, 7–23) VL per participant. At baseline, median age was 39 years, 56% were Black, and median CD4 was 534 cells/mm3. Median time-updated cumulative VCY was 5.4 (Q1–Q3, 4.7–6.9) log10 copy-years/mL. Of 211 (26%) WWH who developed multimorbidity, 162 (77%) had incident hypertension, 133 (63%) had dyslipidemia, 60 (28%) had diabetes, 52 (25%) had cardiovascular disease, and 32 (15%) had kidney disease. Compared with WWH who had time-updated cumulative VCY <5 log10, the adjusted hazard ratio of multimorbidity was 1.99 (95% confidence interval [CI], 1.29–3.08) and 3.78 (95% CI, 2.17–6.58) for those with VCY 5–6.9 and ≥7 log10 copy-years/mL, respectively (P <.0001). Higher time-updated cumulative VCY increased the risk of each NACM. Conclusions Among ART-treated WWH, greater cumulative viremia increased the risk of multimorbidity and of developing each NACM, and hence this may be a prognostically useful biomarker for NACM risk assessment in this population. Abstract

Background Substance use (SU) contributes to poor health outcomes, yet limited data exist to inform strategies to optimize SU treatment among persons with human immunodeficiency virus (HIV). We describe SU and SU treatment utilization among women with and without HIV in the Women's Interagency HIV Study (WIHS). Methods We included data from women enrolled in WIHS from 2013 to 2020. Current SU was self-reported, nonmedical use of drugs in the past year, excluding use of only marijuana. SU treatment utilization was self-reported use of a drug treatment program in the past year. Multivariable regression models were used to investigate associations between participant characteristics and SU treatment. Results Among 2559 women (1802 women living with HIV [WWH], 757 women without HIV), 14% reported current SU. Among those with current SU (n = 367), 71% reported crack/cocaine followed by 40% reporting opioids, and 42% reported any treatment in the past year. The most common treatments were methadone (64%), Narcotics Anonymous (29%), inpatient programs (28%), and outpatient programs (16%). Among women using opioids (n = 147), 67% reported methadone use in the past year compared to 5% using buprenorphine/naloxone. Multivariable analysis showed lower odds of treatment utilization among WWH with concurrent alcohol or marijuana use. Visiting a psychiatrist/counselor was associated with higher odds of treatment. Among WWH, SU treatment was not associated with HIV-related clinical outcomes. Conclusions Treatment utilization was high, especially for methadone use. Our results highlight opportunities for accessing SU treatment for WWH, such as the need to prioritize buprenorphine and comprehensive, wraparound services in HIV care settings. Data analyzed for stability parameters and highly significant mean squares were observed forgenotypes, genotype x environment interaction and environment (linear). IBWl-2007-1 was the most stable genotype underirrigated condition of Chhattisgarh plains for kharif planting situations as it had high mean, regression coefficient not deviatedfrom unity and non significant deviation from regression. 47. Advice for isolated statisticians collaborating in academic healthcare centre settings.

Abstract

A substantial number of statisticians work in isolated domain science departments without access to support networks and resources typical of larger statistical units. These isolated statisticians face many challenges including limited professional networks, non‐traditional roles with idiosyncratic expectations and unique career paths. Furthermore, “the curse of success” lies ahead, as success of the department may only lead to greater demand and lagging support for the services needed. We provide suggestions for career development in an isolated environment by urging statisticians to build their network and become engaged with other statisticians to reduce isolation and identify mentors. We discuss the importance of regular communication with their supervisors to define one's role and navigate the change in expectations in the transition from a supporting role to a co‐investigator to a core leader. To help with “the curse of success,” we explain how to develop a business model with a plan for future expansion and professional development. We conclude that isolation should not be perceived as a limitation. Opportunities and resources exist within the American Statistical Association (ASA) to reduce isolation and support career development. However, there is a need for a formal career path along with evaluation criteria and example business models to support isolated statisticians striving to become core leaders. [ABSTRACT FROM AUTHOR]
