BackgroundNucleated red blood cells (nRBC) are precursor cells of the erythropoiesis that are absent from the peripheral blood under physiological conditions. Their presence is associated with adverse outcomes in critically ill patients. This study aimed to evaluate the predictive value of nRBC on mortality in intensive care unit (ICU) patients with COVID-19 acute respiratory distress syndrome (ARDS).Material and methodsThis retrospective, observational cohort study analyzed data on 206 ICU patients diagnosed with COVID-19 ARDS between March 2020 and March 2022. The primary endpoint was ICU mortality, and secondary endpoints included ICU and hospital stay lengths, ventilation hours, and the time courses of disease severity scores and clinical and laboratory parameters.ResultsAmong the included patients, 68.9% tested positive for nRBC at least once during their ICU stay. A maximum nRBC of 105 µl-1 had the highest accuracy in predicting ICU mortality (area under the curve of the receiver operating characteristic [AUCROC] 0.780, p < 0.001, sensitivity 69.0%, specificity 75.5%). Mortality was significantly higher among patients with nRBC >105 µl-1 than ≤105 µl-1 (86.5% vs. 51.3%, p = 0.008). Compared to patients negative for nRBC in their peripheral blood, those positive for nRBC required longer mechanical ventilation (127 [44 - 289] h vs. 517 [255 - 950] h, p < 0.001), ICU stays (12 [8 – 19] vs. 27 [13 – 51] d, p < 0.001), and hospital stays (19 [12 - 29] d vs. 31 [16 - 58] d, p < 0.001). Peak Sepsis-related Organ Failure Assessment (SOFA), Simplified Acute Physiology Score, PaO2/FiO2, interleukin-6, and procalcitonin values were reached before the peak nRBC level. However, the predictive performance of the SOFA (AUCROC 0.842, p < 0.001) was considerably improved when a maximum SOFA score >8 and nRBC >105 µl-1 were combined.DiscussionnRBC predict ICU mortality and indicate disease severity among patients with COVID-19 ARDS, and they should be considered a clinical alarm signal for a worse outcome. nRBC are a late predictor of ICU mortality compared to other established clinical scoring systems and laboratory parameters but improve the prediction accuracy when combined with the SOFA score.

The Visayan Sea is one of the major traditional fishing grounds in the Philippines and is likewise considered as a top contributor to the country’s overall fisheries production. However, fisheries surveys conducted over the past couple of decades have documented changes in the abundance and composition of catches and stocks toward an increasing dominance of invertebrates and low trophic levels fish families. Assessments of the commercially-important pelagic and demersal stocks were used to estimate population parameters that already exceed the sustainable reference limits. These traditional assessments point exclusively to fishing as the likely cause of the observed status and trends, but they fail to incorporate the potential influence of trophic interactions and variable environmental conditions despite a nationally-legislated mandate to apply an ecosystem approach to assessing and managing the country’s fisheries. Thus, an ecosystem model (Ecopath with Ecosim) of the Visayan Sea was constructed to investigate the role and extent of the fishery, trophic interactions, and changing environmental conditions as drivers of stock abundance and ecosystem dynamics. The results suggest that fishing and its corresponding trophic effects are the major drivers of invertebrate and fish biomass and catches in the Visayan Sea over the past two decades, while the environmental trends provide explanations for patterns that cannot be explained by fishing alone. Incorporating sea surface temperature variations and primary productivity anomalies produced the best-fitting models for most of the observed biomass and catch trends. The catches of the small pelagics (i.e., sardines and mackerels) were likewise found to be particularly affected by rainfall, while the biomass and catch of the small demersal fishes also show a sensitivity to chlorophyll-a concentrations. The primary productivity anomalies further showed a significant correlation with the Pacific Decadal Oscillation (PDO), which suggests that bottom-up trophic controls in the system may be affected by long-term regional climatic changes. These findings validate the applicability and necessity of the ecosystem modeling approach in assessing exploited marine ecosystems to complement the analyses from typically single-species population assessments, especially in the face of increasing environmental variability and projected climate change scenarios.

Abstract Background COVID-19 can induce acute respiratory distress syndrome (ARDS). In patients with congenital heart disease, established treatment strategies are often limited due to their unique cardiovascular anatomy and passive pulmonary perfusion. Case presentation We report the first case of an adult with single-ventricle physiology and bidirectional cavopulmonary shunt who suffered from severe COVID-19 ARDS. Treatment strategies were successfully adopted, and pulmonary vascular resistance was reduced, both medically and through prone positioning, leading to a favorable outcome. Conclusion ARDS treatment strategies including ventilatory settings, prone positioning therapy and cannulation techniques for extracorporeal oxygenation must be adopted carefully considering the passive venous return in patients with single-ventricle physiology.

Abstract Introduction Treatment of high-risk pulmonary embolism (PE) in perioperative patients remains challenging. Systemic thrombolysis is associated with a high risk of major bleedings and intracranial haemorrhage. High mortality rates are reported for open pulmonary embolectomy. Therefore, postoperative surgical patients may benefit substantially from catheter-directed ultrasound-accelerated thrombolysis (USAT). Case presentation We report two cases of high-risk perioperative PE. Both patients developed severe haemodynamic instability leading to cardiac arrest. After the implantation of a veno-arterial extracorporeal membrane oxygenation (ECMO), they were both successfully treated with USAT. Adequate improvement of right ventricular function was achieved; thus, ECMO could be successfully weaned after 3 and 4 days, respectively. Both patients showed favourable outcomes and could be discharged to rehabilitation. Conclusion Current guidelines on treatment of PE offer no specific therapies for perioperative patients with high-risk PE. However, systemic thrombolysis is often excluded due to the perioperative setting and the risk of major bleeding. Catheter-directed thrombolysis was shown to utilise less thrombolytic agent while obtaining comparable thrombolytic effects. The risk for major bleeding (including intracranial haemorrhage) is also significantly lowered. Until further trials determining the value of adopted treatment strategies of high-risk PE in perioperative patients are available, USAT should be considered in similar cases.

The concept of intelligent agents is—roughly speaking—based on an architecture and a set of behavioral programs that primarily serve to solve problems autonomously. Increasing the degree of autonomy and improving cognitive performance, which can be assessed using cognitive and behavioral tests, are two important research trends. The degree of autonomy can be increased using higher-level psychological modules with which needs and motives are taken into account. In our approach we integrate these modules in architecture for an embodied, enactive multi-agent system, such that distributed problem solutions can be achieved. Furthermore, after uncovering some weaknesses in the cognitive performance of traditionally designed agents, we focus on two major aspects. On the one hand, the knowledge processing of cognitive agents is based on logical formalisms, which have deficiencies in the representation and processing of incomplete or uncertain knowledge. On the other hand, in order to fully understand the performance of cognitive agents, explanations at the symbolic and subsymbolic levels are required. Both aspects can be addressed by quantum-inspired cognitive agents. To investigate this approach, we consider two tasks in the sphere of Shannon's famous mouse-maze problem: namely classifying target objects and ontology inference. First, the classification of an unknown target object in the mouse-maze, such as cheese, water, and bacon, is based on sensory data that measure characteristics such as odor, color, shape, or nature. For an intelligent agent, we need a classifier with good prediction accuracy and explanatory power on a symbolic level. Boolean logic classifiers do work on a symbolic level but are not adequate for dealing with continuous data. Therefore, we demonstrate and evaluate a quantum-logic-inspired classifier in comparison to Boolean-logic-based classifiers. Second, ontology inference is iteratively achieved by a quantum-inspired agent through maze exploration. This requires the agent to be able to manipulate its own state by performing actions and by collecting sensory data during perception. We suggest an algebraic approach where both kinds of behaviors are uniquely described by quantum operators. The agent's state space is then iteratively constructed by carrying out unitary action operators, while Hermitian perception operators act as observables on quantum eigenstates. As a result, an ontology emerges as the simultaneous solution of the respective eigenvalue equations.

Softness is one of the essential properties of hygiene tissue products. Reliably measuring it is of utmost importance to ensure the balance between customer expectations and cost-effective tissue production. This study presents a method for assessing softness by analyzing acoustic emissions produced while tearing a tissue specimen. The aim was to train neural network models using the corrected results of human panel tests as the ground truth labels and to predict the tissue softness in two- and three-class recognition tasks. We also investigate the possibility of predicting some production parameters related to the softness property. The results proved that tissue softness and production parameters could be reliably estimated only by the tearing noise.

The coast of Peru lies within the tropics under the influence of the cold, nutrient-rich waters of the Humboldt Current and the interannual onslaught of the El Niño phenomenon. The Peruvian upwelling system is exceptionally productive and is comprised of subsystems at different scales along the coast. We aimed to understand the differences between two shallow coastal systems along a latitudinal gradient: Sechura Bay in the north (at the convergence of Humboldt and tropical waters) and Independencia Bay in the central-south (under typical upwelling conditions). We compared their biodiversity, trophic dynamics, community energetics, resource use and underlying abiotic conditions. Our analysis revealed that over the past two decades, Sechura has shown a warming trend, while Independencia has maintained its cold water conditions. Chlorophyll concentrations have risen significantly in both systems, higher values in Sechura suggesting there is an increase in local pressures that could lead to eutrophication. Trophic models of the La Niña 07/08 period revealed that both systems are bottom-up driven with high biomass and production at the lower trophic levels, though top-down controls were also shown, particularly in Sechura. While primary productivity was similar in both systems, differences were found in the structure and size of energy flows. More cycling and higher transfer efficiency were found in Independencia, where phytoplankton-based food chains played the main role in the overall dynamic. In contrast, the detritus food chain appears to be more relevant for energy flow in Sechura. Differences in biota and flow structure relate to the systems’ environmental conditions, i.e., more diverse warm-water species in the north and mostly cold water adapted species (mainly invertebrate filter-feeders and their predators) in the central-south. Catches in both systems were dominated by the diving fisheries and comprised mostly scallops (bottom-cultured), snails and fish in Sechura, and mussels, clams, crabs and fish in Independencia. Overall, system indicators suggest that Sechura is a comparatively less developed system. Independencia shall likely maintain its general highly productive system features, whereas Sechura will continue to be more frequently disturbed by El Niño and ongoing human-driven activities, reducing its overall stability and functionality. In the context of climate change, acknowledging these differences is essential for future adaptive management regimes.

Cognitive agents that act independently and solve problems in their environment on behalf of a user are referred to as autonomous. In order to increase the degree of autonomy, advanced cognitive architectures also contain higher-level psychological modules with which needs and motives of the agent are also taken into account and with which the behavior of the agent can be controlled. Regardless of the level of autonomy, successful behavior is based on interacting with the environment and being able to communicate with other agents or users. The agent can use these skills to learn a truthful knowledge model of the environment and thus predict the consequences of its own actions. For this purpose, the symbolic information received during the interaction and communication must be converted into representational data structures so that they can be stored in the knowledge model, processed logically and retrieved from there. Here, we firstly outline a grammar-based transformation mechanism that unifies the description of physical interaction and linguistic communication and on which the language acquisition is based. Specifically, we use minimalist grammar (MG) for this aim, which is a recent computational implementation of generative linguistics. In order to develop proper cognitive information and communication technologies, we are using utterance meaning transducers (UMT) that are based on semantic parsers and a mental lexicon, comprising syntactic and semantic features of the language under consideration. This lexicon must be acquired by a cognitive agent during interaction with its users. To this aim we outline a reinforcement learning algorithm for the acquisition of syntax and semantics of English utterances. English declarative sentences are presented to the agent by a teacher in form of utterance meaning pairs (UMP) where the meanings are encoded as formulas of predicate logic. Since MG codifies universal linguistic competence through inference rules, thereby separating innate linguistic knowledge from the contingently acquired lexicon, our approach unifies generative grammar and reinforcement learning, hence potentially resolving the still pending Chomsky-Skinner controversy.

Based on the fact that cogwheels are indispensable parts in manufacturing, we present the acoustic resonance testing (ART) of small data on sintered cogwheels for quality control in the context of non-destructive testing (NDT). Considering the lack of extensive studies on cogwheel data by means of ART in combination with machine learning (ML), we utilize time-frequency domain feature analysis and apply ML algorithms to the obtained feature sets in order to detect damaged samples in two ways: one-class and binary classification. In each case, despite small data, our approach delivers robust performance: All damaged test samples reflecting real-world scenarios are recognized in two one-class classifiers (also called detectors), and one intact test sample is misclassified in binary ones. This shows the usefulness of ML and time-frequency domain feature analysis in ART on a sintered cogwheel dataset.

Encouraging people’s pro-environmental behaviors is an objective of Education for Sustainable Development. In the context of small-scale fisheries, unsustainable fishing practices are compromising the integrity of coastal communities and ecosystems. Ecopath with Ecosim (EwE) is an ecosystem modeling software that presents interactions/changes in the food web as a result of fishing. Despite the multiple applications of EwE in fisheries management, it is unknown from a quantitative perspective whether the application of EwE trophic modeling in environmental education processes and management produces effects on norms and ecological beliefs, and if it alters behavioral intentions of the participants receiving ecosystem modeling information. We conducted a behavior change intervention with gillnet fishers in the Gulf of Nicoya, Costa Rica, to compare antecedents of pro-environmental behavior between participants who received an ecosystem-based intervention (lectures containing EwE models; treatment) and those who received lectures that didn’t involve EwE (control). Based on theories of environmental psychology, we used a pre–post survey design, to evaluate changes between control/treatment, and to assess the influence of psychometric constructs and fishing characteristics on the behavioral intentions to support sustainable fishing measures and owning a fishing license (revealed behavior). Personal norms and values were significant at explaining management measures’ support, along with some fishing characteristics (e.g., fishing site). Deliberating about possible future scenarios (via EwE-modeling) helped reduce uncertainties, increasing legitimacy and a perceived behavioral control (PBC) to support measures. Currently, licenses in the Gulf aren’t granted under defined ecological criteria, and although altruistic-biospheric values scored highly before the intervention began, due to mistrust and high illegal-unlicensed fishing, fishers may be underestimating how much others care about the environment. Value-oriented and ecosystem-based interventions may assist to effectively redesign the licensing system and encourage fishers to support sustainable measures. Our research indicates the importance of education interventions that teach about the impacts of fishing in the ecosystem while helping participants to perceive themselves as capable of implementing actions (PBC) and expressing biospheric-altruistic values to restore trust. Redirecting human behaviors to reconnect with ecosystem resilience can be a leverage point for sustainability and for the compliance of small-scale fisheries management measures.

The Gulf of Nicoya is a large tropical estuary located on the Pacific coast of Costa Rica and accounts for the country’s main fish production. It is increasingly impacted not only by its fisheries, but also by urbanization, tourism developments, the input of pollutants from urban centers and agriculture activities in the catchment areas of large rivers entering the gulf. Similar to other coastal ecosystems, the gulf is particularly sensitive to short- and long-term changes in the climate, such as the precipitation rate, Sea Surface Temperature, wind speed and current regimes. While the gulf has been studied for decades, until now no attempt has been made to combine the analysis of the impacts of fishing and environmental changes on the ecosystem. By following a holistic approach that uses fisheries and environmental time series data from collaborative research and simulation models, this study aims at identifying the main drivers of the observed changes in the Gulf of Nicoya ecosystem. While the model simulations indicate that variations in the catch of some target species (e.g., cephalopods and shrimps) are mainly driven by the fisheries’ exploitation rates, several other species (e.g., corvina, snook, small pelagics, and crabs) are also substantially affected by climate variations, particularly during El Niño periods of high Sea Surface Temperature extremes and increased precipitation. During these periods, phytoplankton productivity and zooplankton biomass deceases with bottom–up effects on the entire food chain. Indicators of the environmental state of the system thus have to be considered along with regulations of the fishing pressure to form the basis for the development of appropriate management strategies.

The early administration of fibrinogen has gained wide acceptance for the treatment of major hemorrhage, whereas the substitution of coagulation factor XIII (FXIII) is only supported by a low level of evidence. This study aimed to answer the question of whether a combined therapy of fibrinogen/FXIII substitution performs superiorly to sole fibrinogen administration in the treatment of dilutional coagulopathy. An in-vitro model of massive transfusion was used to compare the effect of combined fibrinogen/FXIII administration to that of sole fibrinogen therapy for the treatment of dilutional coagulopathy. For this purpose, the blood of red blood cell concentrates, fresh frozen plasma, and platelet concentrates were reconstituted in a ratio of 4:4:1, and then diluted with gelatin by 20% and 40%, respectively. Clot formation and stability were analyzed by thrombelastography. Both sole fibrinogen therapy (equivalent to 50 mg/kg) and the combined administration of fibrinogen (equivalent to 50 mg/kg) and FXIII (equivalent to 75 International Units (IU)/kg) increased fibrinogen-dependent mean clot firmness independently of the degree of dilution (20% dilution: 7 (6.3–7.8) mm; 20% dilution fibrinogen: 13.5 (13–17.3) mm; 20% dilution fibrinogen/FXIII: 16.5 (15.3–18.8) mm; 40% dilution: 3 (2–3.8) mm; 40% dilution fibrinogen: 8 (7–11.3) mm; 40% dilution fibrinogen/FXIII: 10 (8.3–11.8) mm; all p < 0.01). However, no differences were identified between the two treatment arms. Compared to fibrinogen therapy, no beneficial effect of the combined administration of fibrinogen and FXIII for the treatment of dilutional coagulopathy was detected in this in-vitro massive transfusion model. The result was independent of the degree of dilution.

Trophic models of the Ecopath with Ecosim (EwE) type and local ecological knowledge (LEK) have widely been applied to fisheries assessment and management. However, there are no specific methodologies describing how LEK from local fishers can be incorporated with the scientific data from the models in the context of ecosystem-based fisheries management. To our knowledge this is the first contribution exploring a systematic integration of LEK with EwE modeled output. An EwE food web model of the Nicoya Gulf ecosystem constructed 20 years ago and recently revisited by the authors and collaborators, was used in workshops to stimulate discussion among local stakeholders regarding changes in the marine ecosystem. For this study, 58 artisanal fishers were recruited to eight workshops. To assess the LEK, we documented the discussions, and the qualitative data were analyzed with quantitative frequency of responses to identify trends. Next, we systematically compared the changes in the fishery over time through an analysis of similar, complementary, and contradictory information across knowledge systems. In general, the analysis across systems reflected changes in species composition of the catches, paralleled by a harvest reduction in high-trophic-level species, as well as economic losses due to a shift in harvesting low-value species and due to an increase in operational costs. Particularly, we identified (1) similar pieces of information that delivered the same message, providing robust evidence of changes in the social–ecological system; (2) information complementary to each other, which together provided a broader picture (descriptors and attributes) of the changes of some fishing resources; and (3) conflicting pieces of information that indicated mismatches between sources of knowledge, which might suggest the cause of management problems. This study demonstrated how integrating knowledge systems can enhance our understanding of the state and changes in ecosystems, helping to improve fisheries management. We also found that an EwE model can be an effective communication tool to be used with fishers and to promote discussion and engagement. Our aspiration is to bring new and replicable tools to the policy interface in Latin-American fisheries, based on both stakeholder participation (including LEK) and the best scientific information available.

Small-scale fisheries (SSF) remain a largely under-assessed and overlooked sector by governments and researchers, despite contributing approximately 50% to global fish landings and providing food and income for millions of people. The multi-species, multi-gear and data-poor nature of SSF makes implementation of traditional single-species management approaches – like catch-quotas or size limits – particularly challenging and insufficient. A more holistic approach is thus required, which demands assessment of ecological impacts. Here we carried out an estimation of selected ecological indicators of the impact of fisheries (mean length, maximum body size, mean trophic level, trophic and spatial guilds, threatened species and landed by-catch) based on the nominal catch of different gears in three representative SSF along the Colombian Pacific using landings data collected in multiple years (2011–2017). Results showed that taxonomic, size-based, functional and conservation features of the nominal catch vary greatly with geographical location and gear type used. Overall, handlines and longlines tend to select larger sizes and higher trophic levels than nets, but they also catch a higher proportion of intrinsically vulnerable species and species of conservation concern. This challenges the idea that more selective gears have overall lower ecological impacts. In contrast, nets target a wider size range – although focusing on small or medium sized fish – and include a higher diversity of trophic and spatial guilds, which could arguably be considered a more “balanced harvest” type of fishing that retains ecosystem structure and functionality. Bottom trawls, though, exhibited a relatively high percentage of landed by-catch, an undesirable feature for any fisheries in terms of sustainability. We propose that the assessment of a suite of ecological indicators, like those implemented here, should be included as part of periodic evaluations of multi-gear and multi-species SSF in tropical coastal areas, as a practical step toward ecosystem-based fisheries management.

Effective management is essential for small-scale fisheries to continue providing food and livelihoods for households, particularly in developing countries where other options are often limited. Studies on the population dynamics and stock assessment on fishery target species are thus imperative to sustain their fisheries and the benefits for the society. In Lake Koka (Ethiopia), very little is known about the vital population parameters and exploitation status of the fishery target species: tilapia Oreochromis niloticus,common carp Cyprinus carpióand catfish Clarias gariepinus.Our study, therefore, aimed at determining the vital population parameters and assessing the status of these target species in Lake Koka using length frequency data collected quarterly from commercial catches from 2007-2012. A total of 20 097 fish specimens (distributed as 7 933 tilapia, 6 025 catfish and 6 139 common carp) were measured for the analysis. Von Bertalanffy growth parameters and their confidence intervals were determined from modal progression analysis using ELEFAN I and applying the jackknife technique. Mortality parameters were determined from length-converted catch curves and empirical models. The exploitation status of these target species were then assessed by computing exploitation rates (E) from mortality parameters as well as from size indicators i.e., assessing the size distribution of fish catches relative to the size at maturity (L m),the size that provides maximum cohort biomass (Lopt) and the abundance of mega-spawners. The mean value of growth parameters L x, Kand the growth performance index 0' were 44.5 cm, 0.41/year and 2.90 for O. niloticus,74.1 cm, 0.28/year and 3.19 for C. carpioand 121.9 cm, 0.16/year and 3.36 for C. gariepinus,respectively. The 95 % confidence intervals of the estimates were also computed. Total mortality (Z) estimates were 1.47, 0.83 and 0.72/year for O. niloticus, C. carpioand C. gariepinus,respectively. Our study suggest that O. niloticusis in a healthy state, while C. gariepinusshow signs of growth overfishing (when both exploitation rate (E)and size indicators were considered). In case of C. carpio,the low exploitation rate encountered would point to underfishing, while the size indicators of the catches would suggest that too small fish are harvested leading to growth overfishing. We concluded that fisheries production in Lake Koka could be enhanced by increasing Etoward optimum level of exploitation (E opt)for the underexploited C. carpioand by increasing the size at first capture (Lc)toward the L opt range for all target species.

El camarón de aguas someras, es uno de los recursos de mayor importancia socioeconómica en la región Caribe. La falta de información biológica pesquera, es una preocupación de los administradores pesqueros. Por tanto, el presente estudio tiene como objetivo conocer la época reproductiva, la estructura de tallas y la talla media de madurez del camarón de aguas someras (Farfantepenaeus notialis) como insumo para la ordenación y manejo pesquero en el Caribe colombiano. El estudio se llevó a cabo desde junio del 2012 hasta mayo del 2013 en el Caribe colombiano. Se recolectaron 5 356 individuos, los cuales fueron identificados, clasificados y conservados para su análisis en el laboratorio, donde se analizaron las muestras tomando datos de talla, peso, sexo y estadio gonadal. Se encontraron diferencias significativas en la razón de sexo durante todos los meses muestreados, con una mayor proporción de hembras. Se presentaron hembras maduras durante todo el año, pero con dos picos reproductivos de máximo desove, uno entre octubre y diciembre y el otro entre abril y junio. La talla media de captura (TMC) durante todo el muestreo para las hembras fue de 148.0mm de longitud total (LT) y para los machos fue de 122.5mm LT. La talla media de madurez (LT50%) para las hembras y machos fue de 129.34mm LT y 97.77mm LT, respectivamente. La TMC siempre fue mayor que la LT50% para ambos sexos. Debido a la gran disminución del esfuerzo pesquero en el Caribe colombiano durante los últimos 10 años, se espera que este recurso se encuentre en una fase de recuperación o ya esté recuperado.

Background: The Aryl Hydrocarbon Receptor Nuclear Translocator (ARNT, HIF-1β) is a member of the basic-Helix-Loop-Helix PER/ARNT/SIM (bHLH/PAS) protein family and a vital transcriptional regulator regarding development and physiological adaptation processes. ARNT is discussed to be linked with cancer, and other diseases. ARNT is known to be translocated into the cell nucleus, where accumulation of the protein takes place. ARNT is a heterodimerisation partner of the xenobiotic ligand activated Aryl Hydrocarbon Receptor (AhR), the Single Minded proteins (SIM), the cardiovascular helix-loop-helix factor 1 and the Hypoxia Inducible Factor proteins (HIF-α). ARNT is obligatory for HIF-1, HIF-2 and HIF-3 binding to DNA. Whereas degradation of the HIF-α subunits is suppressed by hypoxia, ARNT is generally regarded as constitutively expressed in excess within the cell, and stabilisation is commonly thought to be oxygen-independent. However, we provide evidence that the regulation of ARNT is far more complex. The aim of our study was to reevaluate the regulation of ARNT expression. Methods: We examined cell lines of different origin like MCF-7 and T47D (human breast cancer), HeLa (human cervix carcinoma), Hep3B and HepG2 (human hepatoma), Kelly (human neuroblastoma), REPC (human kidney) and Cos7 (primary primate kidney) cells. We used immunoblot analysis, densitometry, RT-PCR and transient transfection. Results and Conclusion: Our results show that ARNT protein levels are influenced by hypoxia and hypoxia mimetics such as cobalt(II)-chloride (CoCl2) and dimethyloxalylglycine (DMOG) in a cell line specific manner. We demonstrate that this effect might be triggered by HIF-1α which plays an important role in the process of stabilizing ARNT in hypoxia.

Eastern Boundary Current systems (EBCSs) are among the most productive fishing areas in the world. High primary and secondary productivity supports a large biomass of small planktivorous pelagic fish, “small pelagics”, which are important drivers of production to the entire system whereby they can influence both higher and lower trophic levels. Environmental variability causes changes in plankton (food) quality and quantity, which can affect population sizes, distribution and domi-nance among small pelagics. This variability combined with impacts from the fishery complicate the development of management strategies. Consequently, much recent work has been in the development of multispecies trophic models to better understand interdependencies and system dynamics. Despite similarities in extent, structure and primary productivity between EBCSs, the Peruvian system greatly differs from the others in the magnitude of fish catches, due mainly to the incredible production of the anchovy Engraulis ringens. This paper reviews literature concerning EBCSs dynamics and the state-of-the-art in the trophic modeling of EBCSs. The objective is to critically analyze the potential of this approach for system understanding and management and to adapt existing steady-state models of the Peruvian system for use in (future) dynamic simulations. A guideline for the construction of trophodynamic models is presented taking into account the important trophic and environmental interactions. In consideration of the importance of small pelagics for the system dynamics, emphasis is placed on developing appropriate model compartmentalization and spatial delineation that facilitates dynamic simulations. Methods of model validation to historical changes are presented to support hypotheses concerning EBCS dynamics and as a critical step to the development of predictive models. Finally, the identification of direct model links to easily obtainable abiotic parameters is emphasized to add practicality to the model as a predictive tool.

The brown sea cucumber fishery is active in the Galapagos Islands since the year 1991 after its collapse in mainland Ecuador. This paper analyzes the Galapagos Sea cucumber fishery over the past decade and the reasons for its management pitfalls and chronic over fishing, and proposes an improved strategy for estimating stock size and harvest potential. Based on the historical distribution of the fishing fleet and past fishery surveys, 15 macrozones were defined; their areas were estimated from the coastline to the 30m isobaths and the numbers of sample replicates per macrozone were calculated for a density estimate precision of ±25%. Overall stock size was calculated by summing over all macrozones and was multiplied by 0.122 to obtain the annual quota. This multiplier was derived by inserting an exploitation rate of E=0.3 and a published natural mortality value of M=0.17 into Cadimas formula, thereby obtaining a more conservative precautionary quota estimate. Pre-fishery stock densities in 2009 were below the legal threshold value and the fishery remained closed. Mean densities were significantly lower in the deeper (>15m) than in the shallower (

Seagrass beds are highly productive and valuable habitats, which fulfill a key role in coastal ecosystems. Spatial distribution, biomass, density, productivity and leaf dynamics of the dominant seagrass species Thalassia testudinum were studied at five locations in the Gandoca-Manzanillo National Wildlife Refuge, Caribbean coast of Costa Rica, from November 2005 until March 2006. Seagrass beds within the study area cover approximately 16 ha. Spatial and temporal differences in ecological seagrass parameters were examined along gradients of riverine inputs and related to a range of environmental parameters (depth, grain size distribution, nutrient concentrations, salinity and temperature). Average canopy cover and above-ground biomass of T. testudinum inversely correlated with depth, but did not vary significantly between study sites when compared along depth ranges. Considerable spatio-temporal variations in shoot densities, areal productivity and leaf sizes seem to be related to riverine inputs and wave energy. It appeared that T. testudinum at exposed sites respond to increased environmental disturbance related to the offset of the rainy season with clonal recruitment, whereby shoot densities increase and average leaf sizes are reduced. Lower shoot densities and greater leaf sizes, in contrast, are characteristic for locations with rather consistent environmental conditions, where seagrasses are sheltered. T. testudinum in the refuge has higher shoot densities but shorter leaves and lower productivity compared to sites in 15 other Caribbean countries. The seagrass beds appear to be in a relatively healthy state, however, observations of temporal increased stocks of filamentous epiphytes and macroalgae indicate temporal environmental stress in the system. La Guajira es un ecosistema de surgencia explotado en la costa del Caribe colombiano. Con el propósito de integrar la información disponible sobre el sistema se construyó un modelo trófico de 27 grupos funcionales usando el programa Ecopath 5.0 Beta. Se obtuvo una base cuantitativa para compararlo con otros sistemas de surgencia. La Guajira presenta una biomasa total de 68 t/km²/año, la producción primaria neta del sistema es de 1 248.51 t/km²/año y las transferencias totales del sistema son de 3 275 t/km²/año, lo que indica un ecosistema con una surgencia moderada comparada con otros sitios. Las mayores transferencias de energía se dan del nivel trófico I-II (68.93 %), aunque existe una importante proporción de flujos al detritus (32 %). Los resultados en los atributos de madurez del sistema sugieren que La Guajira es un ecosistema inmaduro debido a que la relación P/R excede a 1 y está en desarrollo debido a la baja ascendencia (33.7 %) y alta capacidad de desarrollo (66.3 %) como ocurre con otras surgencias donde se presentan valores de ascendencia entre 20 % y 35 %. Aunque los datos básicos de entrada en el modelo fueron buenos, gracias a las investigaciones que se han elaborado entre 1995 y 2000, existen algunos grupos donde no se encuentra aún información adecuada; especialmente en los valores de biomasa del fitoplancton, invertebrados, bagres, peces depredadores pelágicos, así como estimaciones de producción en invertebrados, peces depredadores pelágicos y peces pequeños pelágicos. No existe una cuantificación de las poblaciones de mamíferos y aves en el área de estudio, los cuales constituyen depredadores superiores y hacen parte esencial en sistemas de surgenciaBiomass flow and structure of a tropical upwelling ecosystem in La Guajira, Colombian Caribbean. La Guajira is an exploited tropical upwelling ecosystem in the Colombian Caribbean coast. A trophic model of 27 functional groups was constructed using the ECOPATH 5.0 Beta software to integrate the available information on the ecosystem. The model allowed a comparison with other trophic flow models of upwelling ecosystems. Total system biomass (68 t/km²/year), net system production (1248.5 t/km²/year), and total system throughput (3 275 t/km²/year) make La Guajira moderate when compared with other systems. The largest amount of energy throughput is achieved from trophic level I to II (68.93 %), although an important proportion of the total flow originates from detritus (32 %). The production/respiration ratio exceeds 1, suggesting that La Guajira is an immature ecosystem and is in development, as determined by its low ascendency (33.7 %) and high development capacity (66.3 %), similar to other upwellings that have values of ascendency between 20 % and 35 %. Although the basic input data were good and covered 1995 to 2000, appropriate information is still not available on some trophic groups such as biomass (for phytoplankton, invertebrates, catfishes and pelagic predator fishes), secondary production data (invertebrates, pelagic predator fishes, and small pelagic fishes), and seabird and mammal populations, which are top trophic levels and an essential part of upwelling ecosystems. The population dynamics of an intertidal population of Megapitaria squalida was studied from September 2002 to February 2003 in Bahía Magdalena, Mexico. To obtain information about the artisanal and recreational fishery of M. squalida, local fishermen at different parts of the bay were interviewed. Clam densities were determined at one unexploited and two exploited sites along perpendicular transects from the shore line. Average clam density was 2.01±1.41 ind/m², densities did not differ significantly between the three sampling sites. The length-total mass relation was Mtot = 0.0001*SL3.1644 (r2 = 0.9804, n = 92) and the length-tissue mass relation was Mtis = 0.0002*SL2.7144 (r² = 0.945, n = 92). Von Bertalanffy-growth parameters were determined using marked individuals. Strong interindividual and also seasonal differences in growth were observed. Average annual values were: K = 0.655, L∞ = 83 mm. Since M. squalida can reach a shell length of more than 135 mm in deeper waters, it is necessary to consider intertidal and subtidal populations separately. Average total mortality (Z) was 1.61 y-1. No fishing mortality was calculated because total mortality Z at the unexploited control site was higher than Z at the exploited sites. It appears that M. squalida migrates into deeper water with increasing shell length, as average shell length increased at lower tidal levels and marked specimens showed a clear migration towards deeper waters. La densidad se determinó en un sitio no explotado y dos sitios explotados utilizando transectos perpendiculares desde la línea costera. También se entrevistó a pescadores locales de diferentes partes de la bahía para obtener información acerca de la pesquería artesanal y recreacional de M. Squalida. La densidad promedio estimada fue de 2.01±1.41 ind/m², sin diferencias significativas entre sitios. La relación de masa y longitud total fue de Mtot = 0.0001*SL3.1644 (r² = 0.9804, n = 92) y la relación de masa y longitud de tejido fue de Mtis = 0.0002*SL2.7144 (r² = 0.945, n = 92). Los parámetros de crecimiento Von Bertalanffy se determinaron utilizando marcadores individuales. Hay grandes diferencias en crecimiento tanto interindividuales como estacionales. Los valores anuales promedio fueron: K = 0.655, L∞ = 83 mm. Debido a que la longitud de la concha puede superar los 135 mm en aguas profundas, es necesario considerar las poblaciones intermareal e inframareal de manera separada. La mortalidad promedio total (Z) fue de 1.61 y-1. No se calculó la mortalidad de pesca porque la mortalidad total (Z) del sitio no explotado fue mayor que la de sitios explotados. Aparentemente M. squalida migra hacia aguas más profundas al crecer. No se encontró evidencia de que estas poblaciones estén sobreexplotadas.

The West Indian Topshell Cittarium pica is artisanally collected on rocky shores along the Caribbean Coast of Costa Rica. There are neither data on the state of its exploitation nor exist any regulation of the fishery. From October 2000 to March 2001, the population dynamics of this species were studied at an unexploited and two exploited sites to determine the present impact of the fishery on the resource. Average population density with 14 ind./m² about three times higher at the unexploited than at the exploited sites. Length-frequeney histograms showed a strong shift towards smaller specimens at the exploited sites, which is also reflected in significantly higher rates of total mortality (Z = 4.05 and 4.47) when compared to the unexploited site (Z = 1.47). Von Bertalanffy growth parameters were estimated as k = 0.19-0.28 (yr-1) and L∞= 104 mm. No significant differences were found among sites. From these values a range of the growth performance index Φ was computed ( Φ = 3.31-3.48) which lies at the lower end of the values reported for other tropical marine gastropods. The size at first maturity for both sexes combined was estimated as 29.20 ± 1.14 mm. Exploitation rates >0.6 for both exploited sites and a large fraction of small specimens (El caracol Cittarium pica (West Indian Top Shell) es recolectado en forma artesanal en zonas rocosas de la costa Caribe de Costa Rica. A la fecha no hay datos sobre esta extracción ni existe regulación de su pesquería. La dinámica poblacional de esta especie fue evaluada, desde octubre del 2000 hasta marzo del 2001, en dos sitios en los cuales la especies es recolectada (Playa Negra y Cahuita), y en un sitio protegido de la actividad pesquera (Isla Uvita). La densidad promedio de la población fue 14 ind/m², cerca de tres veces más alta en el sitio protegido que en los dos no protegidos. Los histogramas de frecuencia de tallas mostraron un fuerte sesgo hacia los ejemplares más pequeños en los sitios no protegidos, lo que se refleja también en tasas de mortalidad total significativamente más altas (Z = 4.05 y 4.47) cuando se les compara con el sitio protegido (Z = 1.47). Los parámetros de crecimiento según von Bertalanffy fueron estirnados en k = 0.19 - 0.28 / año y L∞= 104 mm. No se encontró diferencias significativas entre los sitios. A partir de estos valores el índice Φ (performance index Φ) estuvo en un αmbito de 3.31 a 3.48, el cual se encuentra entre los valores bajos informados para otros gastrópodos tropicales. La edad a la primera madurez sexual para ambos sexos combinados fue estimada en 29.20 ± 1.14 mm. Las tasas de explotación fueron mayores a 0.6 para los sitios no protegidos y un alto componente de ejemplares pequeños (menos de 30 mm) en las recolectas, sugieren una sobre explotación de los adultos y sobrepesca en el reclutamiento. Con base en la estimación de la captura máxima sostenible (maximum sustainable yield), se recomienda algunas medidas reguladores de la pesquería como el control de un tamaño mínimo de desernbarque de 40 mm y la veda de la pesquería durante los rneses de reproducción (de julio a noviembre).

The present article gives an overview on the leading concepts and modelling approaches for marine ecosystems’ research including (1) The trophodynamic theory of pelagic ecosystems, (2) Compartment/network models, (3) Mesocosm experiments and (4) Individual based modelling approaches and virtual ecosystems (VE). The main research questions addressed, as well as the potential and limits of each approach, are summarized and discussed and it is shown how the concept of ecosystem has changed over time. Aquatic biomas spectra (derived from the theory of pelagic ecosystems) can give insight into the trophic structure of different systems, and can show how organism sizes are distributed within the system and how different size groups participate in the system’s metabolism and production. Compartment/network models allow for a more detailed description of the trophic structure of ecosystems and of the energy/biomass fluxes through the explicit modelling of P/B-and food consumption rates and biomasses for each system compartment. Moreover, system indices for a characterization and comparison with other systems can be obtained such as average trophic efficiency, energy throughput, and degree of connectivity, degree of maturity, and others. Recent dynamic extensions of trophic network models allow for exploring past and future impacts of fishing and environmental disturbances as well as to explore policies such as marine protected areas. Mesocosm experiments address a multitude of questions related to aquatic processes (i.e. primary production, grazing, predation, energy transfer between trophic levels etc.) and the behaviour of organisms (i.e. growth, migration, response to contaminants etc.) under semi-natural conditions. As processes within mesocosms often differ in rate and magnitude from those occurring in nature, mesocosms should be viewed as large in vitro experiments designed to test selected components of the ecosystem and not as an attempt to enclose a multitude of interacting processes. Models that use individual organisms as units can provide insight into the causes of natural variability within populations (growth, phenotype, behaviour) and into the role of intraspecific variation for interspecific processes, succession, and feedback mechanisms. In biological oceanography, interdisciplinary research is increasingly using "Virtual Ecosystems" to simulate non-linear interactions between the dynamics of fluctuating ocean circulation, the physics of air-sea interaction, turbulence and optics, biogeochemistry, and the physiology and behaviour of plankton, which can be compared with real observations. The different approaches available for the analysis of aquatic ecosystems should be seen as complementary ways for the description and understanding of ecosystems. The modern view of marine ecosystems, as has emerged from ecosystem analysis over the last decades, is that of a composite of loosely coupled subsystems of desynchron dynamics which through their combined action maintain the fundamental structure and function of the wholeEste artículo es una revisión de los conceptos y enfoques predominantes en la modelación e investigación de los ecosistemas marinos, tales como: (1) la Teoría Trofodinámica de ecosistemas pelágicos, (2) modelos de compartimentos/ red (compartment/network models), (3) experimentos de mesocosmos, y (4) modelos basados en enfoques individuales y ecosistemas virtuales. Se resumen y discuten preguntas relevantes para la investigación así como las limitaciones de cada enfoque, y se muestra como el concepto de ecosistema ha cambiado a través del tiempo. El espectro de biomasa acuática (obtenido de la teoría de ecosistemas pelágicos) puede revelar la estructura trófica de los diferentes ecosistemas; puede mostrar como el tamaño de los organismos se distribuyen dentro del ecosistema y como los diferentes grupos, de acuerdo al tamaño, participan en el metabolismo y producción del mismo. Los modelos de compartimentos/redes permiten describir más detalladamente la estructura trófica y el flujo de energía-/ biomasa en los ecosistemas, particularmente, con el modelamiento explícito de P/B y las tasas de consumo de alimento y biomasa de cada compartimento. Además, se pueden obtener índices para la caracterización y comparación entre sistemas, como por ejemplo la eficiencia trófica promedio, el rendimiento energético, los grados de conectividad y de madurez, y otros. Novedosas ampliaciones dinámicas de los modelos tróficos de red, permiten explorar los impactos pasados y futuros de las pesquerías y de las perturbaciones ambientales, así como sondear políticas de manejo como por ejemplo, las áreas marinas protegidas. Los experimentos de mesocosmos tratan con una multitud de preguntas relacionadas con procesos acuáticos (i.e. producción primaria, pastoreo, depredación, paso de energía entre niveles tróficos, etc.) y el comportamiento de los organismos (i.e. crecimiento, migración, reacción a los contaminantes, etc.) bajo condiciones seminaturales. Como los procesos dentro del mesocosmos frecuentemente difieren de los naturales en tasa y magnitud, éstos deberán ser considerados como grandes experimentos in vitro, diseñados para probar selectos componentes del ecosistema y no como intentos de abarcar múltiples procesos interactivos. Los modelos que utilizan organismos individuales como unidades, pueden revelar las causas de la variabilidad natural dentro de las poblaciones (crecimiento, fenotipo, comportamiento) y del papel de la variación intraespecífica de los procesos interespecíficos, de la sucesión y de los mecanismos retroactivos. Los ecosistemas virtuales están siendo utilizados ampliamente en la investigación interdisciplinaria dentro de la oceanografía biológica para simular interacciones no lineares entre las fluctuaciones dinámicas de la circulación oceánica, la física de las interacciones aire- mar, turbulencia y óptica, biogeoquímica, y en la fisiología y comportamiento del plancton. Todos estos aspectos pueden ser comparados con observaciones reales. Los diferentes enfoques disponibles para el análisis de ecosistemas acuáticos deberán ser considerados como medios complementarios para la descripción y comprensión de los ecosistemas. We developed a transdisciplinary modeling approach for the Huave Lagoon System (HLS), Mexico. This lagoon was selected because it has been used sustainably in various complimentary ways by different ethnic groups since pre-Hispanic times. Over the last few years, however, the ecological impact of artisanal fisheries in the region has grown significantly, thus endangering the balance between society and nature. Because the Huaves and the Zapotecs are ancestral fishing cultures with a profound knowledge of ecosystem resources and local property rights, the aim of this study was to identify ecosystem-level management alternatives capable of diminishing fishing impacts to the HLS. We used a consensus-building process and applied the user's traditional ecological knowledge (TEK). Our counterintuitive results show that specific management strategies should be considered for each particular fishing seascape within the HLS while taking into account the differences among ecological structures and fishery dynamics. The insights from this research aid in defining holistic management policies and support spatial allocations of use rights in local fisheries.
