As part of a project aiming at the discovery of environmentally friendly alternatives to copper in organic agriculture, a 96% ethanolic extract from the leaves of Inga sapindoides showed potent inhibitory activity against grapevine downy mildew (Plasmopara viticola) in vitro (MIC100 25 μg/mL). Separation of the n-BuOH soluble fraction by silica gel column chromatography followed by a combination of RP18 and HILIC HPLC resulted in the isolation of a series of bidesmosidic saponins characterized by the presence of a monoterpenoid unit attached to a triterpenoid aglycone, a p-methoxycinnamoyl residue, and rare sugar residues such as N-acetyl-d-glucosamine, d-quinovose, and d-fucose. The isolated compounds inhibited the formation or activity of P. viticola zoospores with MIC100 values of 3 or 6 μg/mL, respectively. I. sapindoides, a tree which is often cultivated for shading coffee plantations in Central America, may represent a sustainable source of fungicidal products to be used in the replacement of copper.

While the interest in finding medical solutions for the worldwide antibiotics crisis is rising, the legal possibility of simplified authorization of herbal veterinary medicinal products is dwindling. An important basis for both the preservation and development of knowledge in veterinary herbal medicine are pharmacological and clinical studies on the performance of herbal remedies, based on historical written sources on the treatment of farm animals with medicinal plants, as well as current ethnoveterinary research. Nevertheless, there is only limited systematic ethnoveterinary research in Europe, with the exceptions of the Mediterranean region, Switzerland and Austria. We conducted a survey on the ethnoveterinary knowledge of farmers in Bavaria, and analyzed two regional historical textbooks.We documented the local veterinary knowledge about livestock in Bavaria based upon local historical textbooks and upon ethnoveterinary interviews to discover opportunities for the future development of European veterinary herbal medicine.In 2018/2019 we conducted 77 semi-structured interviews with 101 farmers from different types of farms. Detailed information about homemade herbal remedies (plant species, plant part, manufacturing process, source of knowledge) and the corresponding use reports (target animal species, category of use, route of administration, dosage, source of knowledge, frequency of use, last time of use and farmers' satisfaction) were collected. To compare our data with the literature, the use reports of two local historical textbooks were analyzed and compared with the data from the interviews.716 homemade remedy reports (HRs) for altogether 884 use reports (URs) were documented in this study. We picked the 363 HRs that consisted of a single plant species with or without other natural products (HSHRs) for a deeper analysis. These HSHRs were prepared from 108 plant species that belonged to 57 botanical families. The most URs were documented for the families of: Asteraceae, Linaceae and Urticaceae. Calendula officinalis L. (Asteraceae), Linum usitatissimum L. (Linaceae) and Urtica dioica L. (Urticaceae) were the most often documented single species. A total of 448 URs were gathered for the 363 HSHRs. The largest number of URs was for treatments of gastrointestinal disorders and metabolic dysfunctions, followed by skin alterations and sores. For nearly half of the URs the source of knowledge was family and friends. For 80 URs the source of knowledge was different from that of the corresponding HSHRs. For 68% of the URs farmers mentioned at least one use during the last 5 years. Half of the plant species that were mentioned in the historical literature were also mentioned in URs by the interviewees.In Bavaria, medicinal plants are actively used by farmers to treat their livestock with a high level of satisfaction. The knowledge is not passed on from generation to generation in a purely static way, but is dynamically developed by the users in almost one fifth of the URs. Ethnoveterinary research combined with data from regional historical textbooks may facilitate pharmacological and clinical studies in veterinary medicine, and the discussion about a simplified registration for traditional herbal veterinary medicinal products.

Summary Globally, more than six million people are infected with Trypanosoma cruzi, the causative protozoan parasite of the vector-borne Chagas disease (CD). We conducted a cross-sectional ethnopharmacological field study in Bolivia among different ethnic groups where CD is hyperendemic. A total of 775 extracts of botanical drugs used in Bolivia in the context of CD and botanical drugs from unrelated indications from the Mediterranean De Materia Medica compiled by Dioscorides two thousand years ago were profiled in a multidimensional assay uncovering different antichagasic natural product classes. Intriguingly, the phylobioactive anthraquinone hotspot matched the antichagasic activity of Senna chloroclada, the taxon with the strongest ethnomedical consensus for treating CD among the Izoceño-Guaraní. Testing common 9,10-anthracenedione derivatives in T. cruzi cellular infection assays demarcates hydroxyanthraquinone as a potential antichagasic lead scaffold. Highly selective mTOR inhibitors have been discovered through the exploration of the heteroaromatic ring engaging the binding affinity region in mTOR kinase. Compound 11 showed predicted BBB permeability in a MDCK-MDR1 permeability in vitro assay, being the first pyrimido-pyrrolo-oxazine with potential application in the treatment of neurological disorders., Here we present the first pyrimido-pyrrolo-oxazine-based mTOR kinase inhibitor (11) predicted to penetrate the blood brain barrier (BBB). Thus, 11 has a potential in treatments of neurological disorders.

In contrast to natural and historical diets of wild and domesticated ruminants, the diversity of plant species is limited in diets of modern dairy cows. Are "production diseases" linked to this? We conducted a trial to test the effects of a multicomponent herbal feed additive (HFA) on health, performance and fertility traits. A dose-finding study (DF) with 62 cows on 11 commercial farms compared a low (50 g) and a high (100 g) dose of HFA (HFA-50, HFA-100) with a placebo (PL). In a subsequent field trial (FT) with 280 cows on 30 commercial farms, HFA-100 was compared to PL. Cows were randomly assigned to HFA and PL groups and received HFA or PL individually daily from 14 days pre- to 300 days post-calving. Data were analysed with mixed effects models. No differences between HFA and PL were found regarding performance, body condition score and overall culling rates. A tendency towards lower milk urea for HFA-100 compared to PL (p = .06) was found in DF. HFA significantly reduced elevated milk acetone observations (≥10 mg/L) in the first 10 lactation weeks (HFA-100: 4%; HFA-50: 4%; PL: 12%) in DF. HFA-50 significantly reduced lameness incidence (HFA-100: 11%; HFA-50: 2%; PL: 14%) in DF. Calving intervals were 15 days shorter in HFA compared to PL in both trials, which could be confirmed by tendency (p = .07) in FT. In both trials, the proportion of test days with elevated somatic cell score (≥3.0) was significantly lower in HFA compared to PL (DF: HFA-100: 40%, HFA-50: 45% and PL: 55%; FT: HFA-100: 38% and PL: 55%) which is also reflected by tendency (p = .08) in lower culling rates due to udder diseases in FT. HFA showed no negative impact on any of the measured parameters. The effects of HFA indicate a potential of phytochemically rich and diverse feed additives for dairy cows' nutrition and physiology.

A blinded placebo-controlled multi-center on-farm trial was conducted in dairy cows with subclinical ketosis to investigate effects of a multicomponent herbal extract. Blood ketone levels were measured weekly in early lactating cows from 16 Swiss herds. Cows were subclassified based on their initial blood-β-hydroxybutyrate levels (≥ 1.0 [KET-low, 84 cows] and > 1.2 mmol/L [KET-high, 39 cows]) and randomly distributed to 3 groups treated orally with herbal extract containing Camellia sinensis, Cichcorium intybus, Gentiana lutea, Glycyrrhiza glabra, Taraxacum officinale, Trigonella foenum-graecum, and Zingiber officinale, sodium propionate, or placebo twice a day for 5 days. Milk yield, milk acetone, blood-β-hydroxybutyrate, glucose, nonesterified fatty acids, gamma-glutamyl transferase, and glutamate dehydrogenase were analyzed over 2 wk. Linear mixed effect models were used for data analysis. No effects were found for nonesterifed fatty acids, gamma-glutamyl transferase, and glucose. Significantly higher glutamate dehydrogenase (29.71 U/L) values were found in herbal extract-treated animals compared to sodium propionate on day 7 (22.33 U/L). By trend, higher blood-β-hydroxybutyrate levels (1.36 mmol/L) were found in the placebo group of KET-high-cows on day 14 compared to the sodium propionate group (0.91 mmol/L). Milk yields of all treatment groups increased. Milking time and treatment showed a significant interaction for milk acetone: sodium propionate led to an immediate decrease, whereas herbal extracts resulted in a milk acetone decrease from day 7 on, reaching significantly lower milk acetone on day 14 (3.17 mg/L) when compared to placebo (4.89 mg/L). In conclusion, herbal extracts and sodium propionate are both likely to improve subclinical ketosis in dairy cows, however, by different modes of action.

Mitochondrial dysfunction plays a major role not only in the pathogenesis of many oxidative stress or age-related diseases such as neurodegenerative as well as mental disorders but also in normal aging. There is evidence that oxidative stress and mitochondrial dysfunction are the most upstream and common events in the pathomechanisms of neurodegeneration. Cyclopia species are endemic South African plants and some have a long tradition of use as herbal tea, known as honeybush tea. Extracts of the tea are gaining more scientific attention due to their phenolic composition. In the present study, we tested not only the in vitro mitochondria-enhancing properties of honeybush extracts under physiological conditions but also their ameliorative properties under oxidative stress situations. Hot water and ethanolic extracts of C. subternata, C. genistoides, and C. longifolia were investigated. Pretreatment of human neuroblastoma SH-SY5Y cells with honeybush extracts, at a concentration range of 0.1-1 ng/ml, had a beneficial effect on bioenergetics as it increased ATP production, respiration, and mitochondrial membrane potential (MMP) after 24 hours under physiological conditions. The aqueous extracts of C. subternata and C. genistoides, in particular, showed a protective effect by rescuing the bioenergetic and mitochondrial deficits under oxidative stress conditions (400 μM H2O2 for 3 hours). These findings indicate that honeybush extracts could constitute candidates for the prevention of oxidative stress with an impact on aging processes and age-related neurodegenerative disorders potentially leading to the development of a condition-specific nutraceutical.

Although botanical drugs are used widely among different ethnic groups in Bolivia, studies challenging the hypothesis that effective antitrypanosomal medicinal agents were identified empirically are lacking. We conducted a cross-sectional ethnopharmacological field study in Bolivia among different ethnic groups in the Chaco, Chiquitanía and Inter-Andean valleys. We compared botanical drugs used in Bolivia in the context of CD with botanical drugs from unrelated indications from the MediterraneanDe Materia Medica (DMM) compiled by Dioscorides two thousand years ago. A total of 775 ethyl acetate plant extracts with and without ethnomedical indications for CD treatment were profiled againstT. cruziepimastigote and procyclicT. bruceiproliferation, parasite release fromT. cruzitrypomastigote infected cells, as well as for host cell cytotoxicityin vitro. Inhibition of parasite release was monitored using a flow cytometry-based celluar assay. At 25 µg/mL, less than 5% of all extracts exhibited selective toxicity forT. cruzi. We found no evidence that ethnomedicine-inspired bioprospecting significantly increased the probability of finding selective antichagasic botanical drugs. The ethnomedical data further indicate a discrepancy between local and scientific concepts about CD among the studied ethnic groups. Intriguingly, the phylobioactive anthraquinone hotspot identified in this study matched the antichagasic activity ofSenna chloroclada, the taxon with the strongest consensus for treating CD among the Izoceño-Guaraní. Selected antitrypanosomal plant extracts fromDMMwere subjected to HPLC-based activity profiling and targeted isolation of active compounds yielding sesquiterpene lactones, naphtoquinones and anthraquinones. Because the anthraquinone emodin selectively and potently inhibitedT. cruziin host cell infection, we performed a preliminary structure-activity relationship analysis for the 9,10-anthracenedione scaffold, exploring the impact of differential hydroxylation. This study shows that the multidimensional phylobioactivity-guided identification of antichagasic natural products enables comparative bioprospecting and is suitable to challenge ethnopharmacological hypotheses.Author summaryChagas disease (CD) is a parasitic disease caused by the protozoanTrypanosoma cruzi. In Bolivia, CD is a major health problem among indigenous communities, which frequently use traditional medicine to treat the chronic symptoms of the disease related to cardiomyopathy. However, the ethnomedical context of the use of such remedies is largely unclear and it remains unknown whether the botanical drugs have any effect on parasitemia. In a field study among different ethnic groups in the Chaco, Chiquitanía and Inter-Andean valleys the authors collected ethnobotanical and ethnopharmacological information. Later, they profiled and compared the CD botanical drug extract library from Bolivia with a botanical drug extract library from the MediterraneanDe Materia Medica with no association to CD. Using phylogenetic and biological information, they identified bioactive hotspots among different taxa and isolated antichagasic natural products. This led to a first structure-activity relationship study of the natural product class called anthraquinones. While there was no overall statistical difference between the libraries, it is noteworthy that the botanical drug derived fromSenna chlorocladawith the highest consensus among the Guaraní communities, also belonged to the anthraquinone cluster, potentially providing a molecular explanation for its use.

Claw diseases and lameness constitute one of the most severe challenges in livestock production, both for animal welfare and economic reasons. From 2011 to 2014 we conducted five ethnoveterinary surveys in 19 German speaking Swiss cantons, and in the Italian speaking canton of Ticino. A total of 208 interviews were carried out with 273 farmers, 1671 use reports (UR) were recorded, comprising detailed information about plant species, plant part used, the manufacturing process for the end-product, dosing, administration, and therapeutic intention. Among them, 89 UR with 22 different plant species were specifically linked to claw diseases of cattle (80), goats (6), sheep (2) and pigs(1). For seven species more than five UR were reported. We determined the concentration of dry plant material in the final product (g herb/100g product; median, minimum-maximum): Malva ssp (22 UR (21 herb, 1 flower); 0.24g/100g; 0.002-1.82), Matricaria chamomilla L. (11 UR (10 flower, 1 herb); 0.46g/100g; 0.04-2.5), Picea abies (L.) H. KARST. (10 UR (resin); 24.7g/100g; 9.9-100), Calendula officinalis L. (7 UR (flower); 0.2g/100g; 0.0003-0.82), Thymus vulgaris L. (6 UR (herb); 0.02g/100g; 0.015-0.43), Senecio ovatus (P. GAERTN., B.MEY. & SCHERB.) WILLD. (6 UR (herb); 0.37g/100g; 0.07-0.57), and Sanicula europaea L. (6 UR (herb); 0.17g/100g; 0.07-0.27). Senecio ovatus could not be recommended due to the content of pyrrolizidine alkaloids. Besides, insufficient pharmacological data are available for Sanicula europaea. For the remaining five species antimicrobial, anti-inflammatory and wound healing properties are well known, and clinical data with extracts of these plants have been published before.

Ethnopharmacological relevance The term “traditional” is well established in European (human) medicine and even appears in recent European regulations on herbal medicinal products connected to a simplified registration. In contrast, a scientific discussion of a Traditional European Veterinary Herbal Medicine is still lacking in spite of a rising interest, in herbal medicine for animals in particular of veterinarians. There is only limited systematic ethnoveterinary research in Europe, with exception of the Mediterranean region, Switzerland and Austria. We conducted a survey on the ethnoveterinary knowledge of farmers in the pre-alpine and alpine regions of the Swiss cantons of Bern and Lucerne. We compared the findings with earlier studies conducted in Switzerland and with recent and past human and veterinary medicinal literature. Aim of the study We wanted to know to what extent (dependent to different definition of the term “traditional”) the ethnoveterinary knowledge of Swiss farmers could be considered as “traditional” in a European veterinary medicinal context. Material and methods Semistructured interviews with 44 dialog partners, mainly smallholder cattle farmers, were conducted in 2014. Detailed information about homemade herbal remedies (plant species, plant part, manufacturing process) and the corresponding use reports (target animal species, category of use, route of administration, dosage, source of knowledge, frequency of use, last time of use and farmers satisfaction) were collected. To compare our data with literature, one German book of veterinary pharmacology published in 1900, one typescript of Swiss lectures in veterinary pharmacology from 1944, four books of veterinary herbal medicine published between 1984 and 2016, and one recent publication comparing current Swiss (human) ethnomedicinal plant knowledge with modern and past literature were analyzed. Results Information on a total of 315 homemade remedies containing one single plant species (homemade single species herbal remedy reports, HSHR) was collected from participating farmers. These HSHR were prepared from 83 plant species belonging to 46 botanical families. Plants of the families Asteraceae, Lamiaceae and Rosaceae were most frequently used. Matricaria recutita L., Calendula officinalis L., Quercus robur L., Thymus vulgaris L. and Symphytum officinale L. were the most frequently documented species. A total of 404 use reports (UR) were gathered for the 315 HSHR. The largest number of UR was for treatments of gastrointestinal disorders and metabolic dysfunctions, followed by skin alterations and sores. For more than half of the UR the source of knowledge was family, ancestors and friends, and for approximately one quarter the information was from courses and other educational events. For nearly 90% of the UR farmers mentioned at least one use during the last 10 years, and in more than 50% of the UR the last use was within the past year. Compared with recent and past literature and depending on different definitions of the term “traditional” as used for human medicine between 43% and 84% of all 83 and between 57% and 100% of the most often mentioned 21 plant species show a “traditional” European veterinary use. Conclusion For the treatment of their animals farmers in the pre-alpine and alpine regions of the Swiss cantons of Bern and Lucerne mostly used plants which have a track record as medicinal herbs in Europe over several centuries. Almost half of the plant species had specific veterinary uses for about 120 years and even more for at least 30 years. The majority of the plant species thus fulfill the criteria of “traditional” according to several definitions and even more up to a certain degree the criteria of “traditional use” as defined in European regulations for human medicinal products. Ethnoveterinary research combined with data from historical sources may serve as a sound foundation for the development and definition of a Traditional European Veterinary Herbal Medicine maybe even with regard to the recent discussion about a simplified registration for Traditional Herbal Veterinary Medicinal Products.

In organic agriculture, the control of several diseases is largely depending on copper fungicides. Yet, copper can accumulate in the soil if the annual input exceeds annual uptake by plants, which can have a negative impact on soil fertility. Its use should thus be avoided or reduced. The aim of the present study was to evaluate the efficacy of a bark extract of Magnolia officinalis Rehder and Wilson to control three pathogens including Plasmopara viticola (Berk. & M.A. Curtis) Berl. & De Toni (causing grapevine downy mildew), Venturia inaequalis (Cooke) G. Winter (causing apple scab), and Phytophthora infestans (Mont.) De Bary (causing potato and tomato late blight) under controlled and field conditions, and to identify compounds responsible for the antifungal activity of the extract. Under controlled conditions, M. officinalis bark extract showed a mean efficacy of 97% (P. viticola) and 93% (V. inaequalis) at 1 mg mL−1, and EC50 between 0.14 and 0.20 mg mL−1. Efficacy against P. infestans was comparatively low (52% at 1 mg mL−1). Magnolol and honokiol were identified as the main active compounds, both with EC50 ≤ 0.08 mg mL−1 against P. viticola and V. inaequalis. Under field conditions, preliminary formulations reached efficacies up to 71% at 1–2 mg plant extract mL−1 against grapevine downy mildew, whereas activity against apple scab could not be confirmed. Magnolia officinalis is a promising candidate for the development of a sustainable plant protection product against grapevine downy mildew due to a combination of good efficacy, high availability of the raw material at affordable prices, reasonable extraction efficiency, and expected low human toxicity due to its longstanding use in traditional Chinese medicine.

The acid-induced rearrangement of three epoxyderivatives of nobilin 1, the most abundant sesquiterpene lactone in Anthemis nobilis flowers, was investigated. From the 1,10-epoxyderivative 2, furanoheliangolide 5 was obtained, while the 4,5-epoxy group of 3 did not react. Conversely, when the 3-hydroxy function of nobilin was acetylated (12), the 4,5-epoxy derivative did cyclize into cadinanes (15 and 16) under Lewis acid catalysis. The reactivity of the 4,5- and 1,10-epoxy derivatives of nobilin (2 and 3) was compared with that of parthenolide, and rationalized on the basis of quantum chemical calculations. All isolated reaction products were fully characterized by spectroscopic and computational methods, and their in vitro anti-protozoal activity was evaluated. The paper could provide new insights into the biosynthesis of this class of natural products.

An earlier prospective, randomised, placebo-controlled clinical trial had suggested that Bryophyllum pinnatum might have potential in the treatment of overactive bladder. Here we investigated the effects of B. pinnatum leaf press juice, fractions enriched in flavonoids and bufadienolides, and a flavonoid aglycon mixture and individual aglycons on detrusor contractility as a major target in overactive bladder treatment. The strength of the detrusor contractions was investigated using porcine muscle strips stimulated with KCl. B. pinnatum leaf press juice increased the contraction force of muscle strips. Treatment with the flavonoid-enriched fraction had almost no effect on contractility, while the bufadienolide-enriched fraction and flavonoid aglycons led to a concentration-dependent lowering of the contraction force. The data indicate that several components of B. pinnatum leaf press juice may contribute to the inhibitory effect on detrusor contractility, which in turn provides support to overactive bladder treatment with B. pinnatum.

Background Ethnoveterinary knowledge in Europe may play an important role as a basis for sustainable treatment options for livestock. Aims of our study were (a) to compare the ethnoveterinary practices of two culturally and sociodemographically different regions of Switzerland, (b) to compare results with earlier ethnoveterinary studies conducted in Switzerland and in adjacent Italian regions and, (c) to evaluate possible reasons for regional differences in European ethnoveterinary medicine. Methods 25 interviews were conducted in 2014 in all Italian speaking regions (ItR) of Switzerland, and 31 interviews were held in five north-western German speaking Cantons (GeC). Semi-structured questionnaires were used to collect detailed information regarding plant species, mode of preparation, dosage, route of administration, category of use, origin of knowledge, frequency of use, and satisfaction with outcomes of the treatments. Results A total of 162 homemade remedies in ItR and 219 in GeC were reported, out of which 125 and 145, respectively, were reported to contain only one plant species (homemade single species herbal remedy report, HSHR). 44 ItR and 43 GeC plant species were reported to treat livestock, of which only a half were used in both regions. For each HSHR, we classified the treatment intention of all use reports (UR), leading to a total of 205 and 219 UR in ItR and GeC respectively. While cattle were the most often treated livestock species in both study regions, in ItR 40% of UR were administered to small ruminants. Main indications in both regions were gastrointestinal diseases and skin afflictions, but in ItR a high number of URs were reported as antiparasitics. URs were mainly handed down from the past generation, but in GeC the source of knowledge for 20% of URs were from courses. Regarding the used plant species, ItR showed a higher concordance with Swiss than Italian studies, but with some differences to all regions. A total of 22 (14 ItR; 8 GeC) plant species in this study have not been reported before in ethnoveterinary studies of Swiss and Italian alpine regions. Conclusions ItR and GeC, show differences and similarities with respect to their own ethnoveterinary practices and earlier Swiss and Italian ethnoveterinary studies. Linguistic, geographical, as well as social and farm-structural conditions influence the regional ethnoveterinary knowledge. However, political borders seem to be more important than language or geographical barriers. A surface extract of the aerial parts of Salvia chamaedryoides afforded 13 diterpenes (1-13), with seven compounds (1, 3, 4, 7-9, 12) described for the first time. The structures of the new compounds were established using 1D and 2D NMR spectroscopic methods, HRESIMS, and ECD data. The potential hypoglycemic effects of the crude extract, fractions, and pure compounds from S. chamaedryoides were investigated by inhibition of α-glucosidase and α-amylase enzymes. The extract and its fractions showed a moderate dose-dependent inhibition; the pure compounds exhibited differential inhibitory activity against these two enzymes. Molecular modeling studies were also performed to suggest the interaction mode of compound 3 in the α-glucosidase enzyme active site. The antimicrobial activity of the purified compounds was investigated against 26 clinical pathogens. No activity was detected for the Gram-negative species tested nor on Candida albicans and C. glabrata, while variable susceptibilities were observed using Gram-positive staphylococcal and enterococcal species.

An in-house library of more than 3000 extracts of plant and fungal origin was screened against some major plant pathogens. As one of the hits, an ethyl acetate extract from inflorescences of Verbesina lanata showed significant inhibitory activity in vitro against grapevine downy mildew (Plasmopara viticola), with a MIC100 value of 35 μg/mL. An emulsifiable concentrate formulation with 50 mg/g of the extract was developed for in vivo evaluation. A suspension of the formulation containing 1 mg/mL of extract lowered leaf surface infection of grapevine seedling by 82% compared to the nontreated control. With the aid of HPLC-based activity profiling, the antifungal activity was correlated with a series of lipophilic compounds. Preparative isolation by a combination of chromatographic techniques afforded 16 eudesmane sesquiterpenes including eight new congeners. Nine compounds were obtained in sufficient quantities to be tested in vitro and were found to inhibit the zoospore activity of P. viticola with MIC100 values ranging from 4 to 50 μg/mL. The two major compounds, 6β-cinnamoyloxy-4β,9β,15-trihydroxyeudesmane (9) and 6β-cinnamoyloxy-1β,15-dihydroxyeudesm-4-en-3-one (13), showed MIC100 values of 5 and 31 μg/mL, respectively.

Sesquiterpene lactones (STLs) are natural products that have potent antitrypanosomal activity in vitro and, in the case of cynaropicrin, also reduce parasitemia in the murine model of trypanosomiasis. To explore their structure-antitrypanosomal activity relationships, a set of 34 natural and semi-synthetic STLs and amino-STLs was tested in vitro against T. b. rhodesiense (which causes East African sleeping sickness) and mammalian cancer cells (rat bone myoblast L6 cells). It was found that the alpha-methylene-gamma-lactone moiety is necessary for both antitrypanosomal effects and cytotoxicity. Antitrypanosomal selectivity is facilitated by 2-(hydroxymethyl)acrylate or 3,4-dihydroxy-2-methylenebutylate side chains, and by the presence of cyclopentenone rings. Semi-synthetic STL amines with morpholino and dimethylamino groups showed improved in vitro activity over the native STLs. The dimethylamino derivative of cynaropicrin was prepared and tested orally in the T. b. rhodesiense acute mouse model, where it showed reduced toxicity over cynaropicrin, but also lost antitrypanosomal activity

International audience; The indolo[2,1-b]quinazoline alkaloid tryptanthrin was previously identified as a potent anti-inflammatory compound with a unique pharmacological profile. It is a potent inhibitor of cyclooxygenase-2, 5-lipooxygenase-catalyzed leukotriene synthesis, and nitric oxide production catalyzed by the inducible nitric oxide synthase. To characterize the pharmacokinetic properties of tryptanthrin, we performed a pilot in vivo study in male Sprague-Dawley rats (2 mg/kg bw i. v.). Moreover, the ability of tryptanthrin to cross the blood-brain barrier was evaluated in three in vitro human and animal blood-brain barrier models. Bioanalytical UPLC-MS/MS methods used were validated according to current international guidelines. A half-life of 40.63 ± 6.66 min and a clearance of 1.00 ± 0.36 L/h/kg were found in the in vivo pharmacokinetic study. In vitro data obtained with the two primary animal blood-brain barrier models showed a good correlation with an immortalized human monoculture blood-brain barrier model (hBMEC cell line), and were indicative of a high blood-brain barrier permeation potential of tryptanthrin. These findings were corroborated by the in silico prediction of blood-brain barrier penetration. P-glycoprotein interaction of tryptanthrin was assessed by calculation of the efflux ratio in bidirectional permeability assays. An efflux ratio below 2 indicated that tryptanthrin is not subjected to active efflux.

Myotonic dystrophy type I (DM1) is a disabling neuromuscular disease with no causal treatment available. This disease is caused by expanded CTG trinucleotide repeats in the 3′ UTR of the dystrophia myotonica protein kinase gene. On the RNA level, expanded (CUG)n repeats form hairpin structures that sequester splicing factors such as muscleblind-like 1 (MBNL1). Lack of available MBNL1 leads to misregulated alternative splicing of many target pre-mRNAs, leading to the multisystemic symptoms in DM1. Many studies aiming to identify small molecules that target the (CUG)n-MBNL1 complex focused on synthetic molecules. In an effort to identify new small molecules that liberate sequestered MBNL1 from (CUG)n RNA, we focused specifically on small molecules of natural origin. Natural products remain an important source for drugs and play a significant role in providing novel leads and pharmacophores for medicinal chemistry. In a new DM1 mechanism-based biochemical assay, we screened a collection of isolated natural compounds and a library of over 2100 extracts from plants and fungal strains. HPLC-based activity profiling in combination with spectroscopic methods were used to identify the active principles in the extracts. The bioactivity of the identified compounds was investigated in a human cell model and in a mouse model of DM1. We identified several alkaloids, including the β-carboline harmine and the isoquinoline berberine, that ameliorated certain aspects of the DM1 pathology in these models. Alkaloids as a compound class may have potential for drug discovery in other RNA-mediated diseases.

Five new diterpenes (1–5) and a megastigmane derivative (6) were isolated from the aerial parts of Euphorbia laurifolia, along with several known compounds. Their structures were elucidated by NMR, MS, and ECD and by chemical methods. A chemical proteomics drug affinity responsive target stability (DARTS) approach to investigate the lathyrane diterpene 1, laurifolioside, on its putative cellular target(s) was performed. Clathrin heavy chain 1, a protein mainly involved in selective uptake of proteins, viruses, and other macromolecules at the plasma membrane of cells, was identified as the major interaction partner of compound 1. The modulation of clathrin activity by 1 was studied through microscopy, molecular docking, and molecular dynamics studies, suggesting a new activity of lathyrane diterpenes in the modulation of trafficking pathways.

Background Berberis orthobotrys Bien ex Aitch. (Berberidaceae) is a plant indigenous of Pakistan that is locally used for the treatment of hypertension. Hypothesis This study evaluated the vasoactive properties of a Berberis orthobotrys root extract and its fractions, and investigated the role of the endothelium and the underlying mechanism. Study design An aqueous methanolic extract of Berberis orthobotrys roots was prepared and submitted to a multi-step liquid-liquid fractionation with solvents of increasing polarity. Vascular reactivity of the different fractions was assessed using porcine coronary artery rings either with or without endothelium, and in the presence or absence of specific pharmacological tools. The ability of Berberis orthobotrys extracts to affect phosphodiesterase (PDE) activity was evaluated using a radioenzymatic method and purified phosphodiesterases. Results The aqueous methanol extract induced similar relaxations in coronary artery rings with and without endothelium, and, amongst the three derived preparations, the butanol fraction (BFBO) was slightly but significantly more effective than the ethyl acetate fraction and the aqueous residue in rings without endothelium. Analysis of the butanol fraction (BFBO) by LC-ELSD-MS indicated the presence of four major isoquinoline alkaloids including berberine. BFBO significantly potentiated the relaxations induced by cyclic GMP- and cyclic AMP-dependent relaxing agonists, and inhibited contractions to KCl, CaCl2, and U46619 in endothelium denuded rings. In contrast, BFBO did not affect relaxations to endothelium-dependent vasodilators. BFBO concentration-dependently inhibited the cyclic GMP-hydrolyzing activity of basal PDE1, calmodulin-activated PDE1 and PDE5, and of cyclic AMP-hydrolyzing activity of PDE3 and PDE4 with IC50 values ranging from 40 to 130 µg/ml. Conclusion The butanol fraction of the aqueous methanol extract of Berberis orthobotrys roots induced pronounced endothelium-independent relaxations and inhibited contractile responses by acting directly at the vascular smooth muscle in the coronary artery. Moreover, BFBO potentiated relaxations induced by both cyclic GMP- and cyclic AMP-dependent vasodilators most likely due to its ability to inhibit several vascular PDEs, and in particular PDE4 and PDE5.
