Abstract The phenomenon of mixed/heterogenous treatment responses to cancer therapies within an individual patient presents a challenging clinical scenario. Furthermore, the molecular basis of mixed intra-patient tumor responses remains unclear. Here, we show that patients with metastatic lung adenocarcinoma harbouring co-mutations of EGFR and TP53, are more likely to have mixed intra-patient tumor responses to EGFR tyrosine kinase inhibition (TKI), compared to those with an EGFR mutation alone. The combined presence of whole genome doubling (WGD) and TP53 co-mutations leads to increased genome instability and genomic copy number aberrations in genes implicated in EGFR TKI resistance. Using mouse models and an in vitro isogenic p53-mutant model system, we provide evidence that WGD provides diverse routes to drug resistance by increasing the probability of acquiring copy-number gains or losses relative to non-WGD cells. These data provide a molecular basis for mixed tumor responses to targeted therapy, within an individual patient, with implications for therapeutic strategies.

Стаття розкриває питання формування електронних архівів кваліфікаційних праць, акцентуючи на міжнародному досвіді. У контексті зростаючої потреби в цифровізації освітніх та наукових матеріалів для забезпечення їх доступності й ефективної обробки автор висвітлює ключові аспекти переходу на електронне архівування. Основна увага фокусується на досвіді цифровізації традиційних архівів для підвищення ефективності обробки та збільшення доступності кваліфікаційних праць. Значний обсяг дослідження приділяється аналізу роботи міжнародних баз даних, таких як ProQuest Dissertations and Theses, OATD, NDLTD, DART-Europe та їхнім методам індексування і зберігання кваліфікаційних праць. Розглядається також роль протоколу OAI-PMH у спрощенні обміну метаданими між різними репозитаріями, що сприяє інтеграції наукових ресурсів у світовий освітній простір. Подальші дослідження мають на меті розробити рекомендації щодо оптимізації формування та інтеграції електронних архівів в Україні для підвищення престижу вітчизняної освіти та науки.

Стаття аналізує потенціал використання технологій оптичного розпізнавання символів (OCR) від Google для ефективної цифровізації архівних документів. Автор описує, як інтеграція Google Cloud Vision API може пришвидшити перетворення фізичних документів у цифровий формат, знижуючи вартість і підвищуючи доступність оцифрування для архівних установ різного масштабу. Також розглядаються переваги збільшення точності та якості оцифрованих даних, що сприяє їх подальшому науковому та освітньому використанню. Аналіз включає огляд потенційних технічних та організаційних проблем, які можуть виникати під час упровадження OCR-технологій, зокрема сумісність систем та вимоги до безпеки даних. Пропонується розробка вебзастосунку для кінцевих користувачів, який зробить передові можливості OCR доступнішими та зручнішими для використання. Метою дослідження є підвищення ефективності збереження інформації та забезпечення швидкого доступу до неї, зважаючи на актуальні потреби управління архівними ресурсами. Стаття також надає огляд останніх досліджень і публікацій, що демонструють ефективність Google OCR порівняно до інших продуктів і підкреслює можливості подальшого розвитку цих технологій для оптимізації процесів оцифрування в архівах.

Висвітлено теоретичні концепції в галузі семіотики, теорії інформації, теорії комунікації та архівознавства, що сприяють розумінню природи й значення інформаційних зв’язків у контексті оптимізації архівної діяльності. Доведено, що застосування спеціальних методик аналізу, класифікації та систематизації архівних документів надає можливість ефективно визначати інформаційні зв’язки між ними, забезпечуючи логічну організацію архівних фондів та полегшуючи їхнє подальше використання. Розкрито значення теоретичного й методичного підґрунтя для вдосконалення процесів систематизації, збереження архівних інформаційних ресурсів з метою забезпечення доступності та зручності використання історичної документації.

У статті надано покроковий опис та особливості оцифрування службових документів, на прикладі створення електронного архіву організаційно-розпорядчих документів закладу вищої освіти, а саме Національного університету «Львівська політехніка». Наведено переваги та проблеми, що можуть виникнути під час проведення цієї процедури. Досліджено та розкрито особливості процесу сканування документів, а також основні атрибути, що дозволяють здійснювати релевантний пошук інформації. Проведено аналіз програмного забезпечення, який дозволяє здійснювати оптичне розпізнавання вмісту документів. Обрано та описано технологічне рішення щодо створення інформаційного ресурсу, який міститиме оцифровані організаційно-розпорядчі документи освітнього закладу — система керування вмістом WordPress, подано переваги його використання та задіяні плагіни.

Abstract Background Recent advances in genomics and proteomics have allowed us to study the nuances of the Warburg effect – a long-standing puzzle in cancer energy metabolism – at an unprecedented level of detail. While modern next-generation sequencing technologies are extremely powerful, the lack of appropriate data analysis tools makes this study difficult. To meet this challenge, we developed a novel application for comparative analysis of gene expression and visualization of RNA-Seq data. Results We analyzed two biological samples (normal human brain tissue and human cancer cell lines) with high-energy, metabolic requirements. We calculated digital topology and the copy number of every expressed transcript. We observed subtle but remarkable qualitative and quantitative differences between the citric acid (TCA) cycle and glycolysis pathways. We found that in the first three steps of the TCA cycle, digital expression of aconitase 2 (ACO2) in the brain exceeded both citrate synthase (CS) and isocitrate dehydrogenase 2 (IDH2), while in cancer cells this trend was quite the opposite. In the glycolysis pathway, all genes showed higher expression levels in cancer cell lines; and most notably, digital gene expression of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and enolase (ENO) were considerably increased when compared to the brain sample. Conclusions The variations we observed should affect the rates and quantities of ATP production. We expect that the developed tool will provide insights into the subtleties related to the causality between the Warburg effect and neoplastic transformation. Even though we focused on well-known and extensively studied metabolic pathways, the data analysis and visualization pipeline that we developed is particularly valuable as it is global and pathway-independent.

Abstract Background One of the major challenges in biology is the correct identification of promoter regions. Computational methods based on motif searching have been the traditional approach taken. Recent studies have shown that DNA structural properties, such as curvature, stacking energy, and stress-induced duplex destabilization (SIDD) are useful in promoter prediction, as well. In this paper, the currently used SIDD energy threshold method is compared to the proposed artificial neural network (ANN) approach for finding promoters based on SIDD profile data. Results When compared to the SIDD threshold prediction method, artificial neural networks showed noticeable improvements for precision, recall, and F-score over a range of values. The maximal F-score for the ANN classifier was 62.3 and 56.8 for the threshold-based classifier. Conclusions Artificial neural networks were used to predict promoters based on SIDD profile data. Results using this technique were an improvement over the previous SIDD threshold approach. Over a wide range of precision-recall values, artificial neural networks were more capable of identifying distinctive characteristics of promoter regions than threshold based methods.

11. A new Ornithoctona (Diptera: Hippoboscidae) species from Baikal State Nature Reserve (Russia)

Louse flies from the family Hippoboscidae parasitise birds and mammals. Both males and females feed on the blood and carry many dangerous diseases. Representatives of the genus Ornithoctona are full-winged, widely specialised parasites of birds. Before this study it was considered, that the genus Ornithoctona includes 12 species. Among them, only two species were recorded in the Palaearctic (from Kazakhstan, Russian Far East, and Kuril Islands). Other ones inhabit Africa, tropical Asia, the Pacific Islands, and the New World. During the annual bird ringing in the Baikal State Nature Reserve (Russia), a new species of the genus Ornithoctona (Diptera: Hippoboscidae) (Ornithoctona zootherae sp. nov.) was collected and described. Ornithoctona zootherae sp. nov. was found on Zoothera dauma, a common species, that breeds in Eastern Siberia and winters in China. Ornithoctona zootherae sp. nov. differs from all known Palearctic and Asian Ornithoctona species in larger body size (length of the head + thorax is 5.5–6.0 mm). Additionally, the new species differs from O. plicata in the wing-setulae, while O. plicata has no wing-setulae. It differs from O. soror and O. australaisiae in the ventral and dorsal view of the abdominal apices. A single key for all World species of the genus Ornithoctona is composed.

Розглянуто інформаційне забезпечення тьютора для сприяння адаптації студентів у закладах вищої освіти (ЗВО). Означено основні засади тьюторства, зокрема акцентовано, що воно є сучасним освітнім трендом, який активно починає впроваджуватися у ЗВО України. Його основна мета спрямована на вдосконалення студента, розвиток його практичних навичок та вмінь, виховання самостійності студента. Акцентовано, що тьютор повинен бути наставником та другом для свого підопічного, допомагати студентові знайти своє призначення, супроводжувати його від початку вступу й до закінчення навчання в університеті. Подано інформаційну модель, функціональну модель, що демонструє, як організована робота тьютора та напрями його роботи, організацію системи тьюторства, яка спрямовується для формування рекомендацій для студентів у навчальній, науковій, творчій та громадській діяльності. Означено, що інформаційне забезпечення тьютора вкрай необхідне для його ефективної та успішної роботи.

Abstract Osimertinib, an epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI), potently and selectively inhibits EGFR-TKI-sensitizing and EGFR T790M resistance mutations. In the Phase III FLAURA study (NCT02296125), first-line osimertinib improved outcomes vs comparator EGFR-TKIs in EGFRm advanced non-small cell lung cancer. This analysis identifies acquired resistance mechanisms to first-line osimertinib. Next-generation sequencing assesses circulating-tumor DNA from paired plasma samples (baseline and disease progression/treatment discontinuation) in patients with baseline EGFRm. No EGFR T790M-mediated acquired resistance are observed; most frequent resistance mechanisms are MET amplification (n = 17; 16%) and EGFR C797S mutations (n = 7; 6%). Future research investigating non-genetic acquired resistance mechanisms is warranted.

Abstract Third-generation EGFR tyrosine kinase inhibitors (EGFR-TKIs), including osimertinib, an irreversible EGFR-TKI, are important treatments for non-small cell lung cancer with EGFR-TKI sensitizing or EGFR T790M resistance mutations. While patients treated with osimertinib show clinical benefit, disease progression and drug resistance are common. Emergence of de novo acquired resistance from a drug tolerant persister (DTP) cell population is one mechanism proposed to explain progression on osimertinib and other targeted cancer therapies. Here we profiled osimertinib DTPs using RNA-seq and ATAC-seq to characterize the features of these cells and performed drug screens to identify therapeutic vulnerabilities. We identified several vulnerabilities in osimertinib DTPs that were common across models, including sensitivity to MEK, AURKB, BRD4, and TEAD inhibition. We linked several of these vulnerabilities to gene regulatory changes, for example, TEAD vulnerability was consistent with evidence of Hippo pathway turning off in osimertinib DTPs. Last, we used genetic approaches using siRNA knockdown or CRISPR knockout to validate AURKB, BRD4, and TEAD as the direct targets responsible for the vulnerabilities observed in the drug screen.

The article provides clinical data and an algorithm for the management of patients with symptomatic uterine fibroids complicated by metrorrhagia and venous thrombotic events. Objective: to improve the treatment of patients with uterine fibroids complicated by abnormal menstrual bleeding, anemia and venous thrombotic events due to the implementation of the developed algorithm for diagnosis and treatment. Materials and methods. The study included 15 patients aged 32–49 years with a diagnosis of uterine fibroids with menorrhagia and iron deficiency anemia, complicated by deep vein thrombosis of the lower extremities. Thromboembolism of small branches of the pulmonary artery was diagnosed in 5 (30%) of these patients. The examination algorithm included general clinical tests (general blood and urine analysis, biochemical blood analysis, coagulogram), evaluation of the D-dimer, soluble fibrin monomer complexes, ultrasound of the pelvic organs, ultrasound duplex scanning of the veins of lower extremities, electrocardiography, echocardiography, multispiral computed tomography of chest. All patients underwent endometrial biopsy to exclude oncological pathology. The proposed treatment included: uterine artery embolization to stop bleeding, correction of hemostatic parameters, and anticoagulant therapy of venous thrombotic events after stopping bleeding. Results. Bleeding was stopped in all 100% of patients after endovascular uterine artery embolization. The duration of anticoagulant therapy depended on the causes of venous thrombosis and was at least 3–6 months; it was extended for more than 6 months if there were concomitant risk factors. Conclusions. Uterine artery embolization allows quickly and reliably stopping bleeding and immediately starting adequate anticoagulant therapy for venous thrombotic events.

Він слугує для них засобом оперативного надання інформації особистого та архівного значення, взаємодії зі споживачами інформації, громадянами, сприяє інноваційній діяльності в умовах інформатизації суспільства. Вебсайт допомагає розвивати теорію та практику архівної справи. Проаналізовано переваги наявності вебсайту та висвітлено етапи створення порталу для архіву Національного університету «Львівська політехніка». Проєкт вебсайту створено на базі CMS WordPress; ця система має зрозумілий інтерфейс, вона легка для опанування навіть тим, хто ніколи не створював вебпортали. Реалізований проєкт, що показано в статті, повністю готовий до використання та впровадження в роботу університетського архіву. Проаналізовано відповідну літературу з метою підкреслити перспективи використання сучасних технологій в архівній справі та створено діаграми, які демонструють роботу порталу зсередини.

Introduction: Osimertinib is a third-generation EGFR tyrosine kinase inhibitor (TKI) that is approved for the use of EGFR -mutant non-small cell lung cancer (NSCLC) patients. In this study, we investigated the acquired resistance mechanisms in NSCLC patients and patient-derived preclinical models. Methods: Formalin-fixed paraffin-embedded tumor samples and plasma samples from 55 NSCLC patients who were treated with osimertinib were collected at baseline and at progressive disease (PD). Next-generation sequencing was performed in tumor and plasma samples using a 600-gene hybrid capture panel designed by AstraZeneca. Osimertinib-resistant cell lines and patient-derived xenografts and cells were generated and whole exome sequencing and RNA sequencing were performed. In vitro experiments were performed to functionally study the acquired mutations identified. Results: A total of 55 patients and a total of 149 samples (57 tumor samples and 92 plasma samples) were analyzed, and among them 36 patients had matched pre- and post-treatment samples. EGFR C797S (14%) mutation was the most frequent EGFR- dependent mechanism identified in all available progression samples, followed by EGFR G824D (6%), V726M (3%), and V843I (3%). Matched pre- and post-treatment sample analysis revealed in-depth acquired mechanisms of resistance. EGFR C797S was still most frequent (11%) among EGFR-dependent mechanism, while among EGFR-independent mechanisms, PIK3CA, ALK , BRAF, EP300, KRAS , and RAF1 mutations were detected. Among Osimertinib-resistant cell lines and patient-derived models, we noted acquired mutations which were potentially targetable such as NRAS p.Q61K, in which resistance could be overcome with combination of osimertinib and trametinib. A patient-derived xenograft established from osimertinib-resistant patient revealed KRAS p.G12D mutation which could be overcome with combination of osimertinib, trametinib, and buparlisib. Conclusion: In this study, we explored the genetic profiles of osimertinib-resistant NSCLC patient samples using targeted deep sequencing. In vitro and in vivo models harboring osimertinib resistance revealed potential novel treatment strategies after osimertinib failure.

Запропоновано інформаційну та функціональну моделі реалізації інформаційного ресурсу для організації діловодних процесів медичного закладу Благодійного фонду «Шпиталь імені Митрополита Андрея Шептицького» у вигляді вебсайту для внутрішнього використання на основі інтранет-технологій. Задіяні моделі, спрямовані на реалізацію інформаційного ресурсу за допомогою інтранет-технологій, допоможуть секретареві медичного закладу вести документообіг без використання вартісних спеціалізованих систем електронного документообігу. Запропонований інформаційний ресурс допоможе здійснювати пошук інформації за певними критеріями. Спільне використання інформаційного ресурсу діловодних процесів та сховищ інформації дозволить систематизувати й поєднувати інформацію, що полегшить її аналіз і складання звітів, а також підвищити точність та оперативність у наданні статистичної, аналітичної і довідкової інформації.

This exploratory, post hoc analysis aimed to model circulating tumor DNA (ctDNA) dynamics and predict disease progression in patients with treatment‐naïve locally advanced/metastatic epidermal growth factor receptor mutation (EGFRm)‐positive non‐small cell lung cancer, from the FLAURA trial (NCT02296125). Patients were randomized 1:1 and received osimertinib 80 mg once daily (q.d.) or comparator EGFR‐TKIs (gefitinib 250 mg q.d. or erlotinib 150 mg q.d.). Plasma was collected at baseline and multiple timepoints until treatment discontinuation. Patients with Response Evaluation Criteria in Solid Tumors (RECIST) imaging data and detectable EGFR mutations (Ex19del/L858R) at baseline and ≥ 3 additional timepoints were evaluable. Joint modeling was conducted to characterize the relationship between longitudinal changes in ctDNA and probability of progression‐free survival (PFS). A Bayesian joint model of ctDNA and PFS was developed solving differential equations with the ctDNA dynamics and the PFS time‐to‐event probability. Of 556 patients, 353 had detectable ctDNA at baseline. Evaluable patients (with available imaging and ≥ 3 additional timepoints, n = 320; ctDNA set) were divided into training (n = 259) and validation (n = 61) sets. In the validation set, the model predicted a median PFS of 17.7 months (95% confidence interval (CI): 11.9–28.3) for osimertinib (n = 23) and 9.1 months (95% CI: 6.3–14.8) for comparator (n = 38), consistent with observed RECIST PFS (16.4 months and 9.7, respectively). The model demonstrates that EGFRm ctDNA dynamics can predict the risk of disease progression in this patient population and could be used to predict RECIST‐defined disease progression. Despite the fact that aortic valve replacement surgery is well developed and accompanied by high efficiency, it is still far from ideal and associated with a large number of complications. In this relation, the methods of surgical treatment based on valve-preserving philosophy surpass traditional prosthetics, since they may decrease risks at early and long-term periods after the operation, which is especially important in elderly and senile patients. The manuscript presents the results of a study on application of the ultrasonic decalcification in 70 patients with calcified aortic valve stenosis. After surgery, up to 13 months, a significant decrease in the systolic pressure gradient on the valve and an increase in the area of the effective valve opening were observed. This shows the effectiveness of the ultrasonic decalcification in the elderly patient population.

Abstract Background Haemoproteus parasites are widespread, and several species cause diseases both in birds and blood-sucking insects. These pathogens are transmitted by dipterans belonging to the Ceratopogonidae and Hippoboscidae, however certain vector species remain unknown for the majority of Haemoproteus spp. Owls are often infected by Haemoproteus parasites, but experimental studies on vectors of these infections are lacking. The aim of this study was to investigate sporogonic development of two widespread Haemoproteus parasites of owls, H. noctuae and H. syrnii in experimentally infected biting midges Culicoides impunctatus and Culicoides nubeculosus. We also followed in vitro sporogonic development of these infections and determined their phylogenetic relationships with Haemoproteus spp., for which vectors have been identified. Methods Wild-caught C. impunctatus and laboratory reared C. nubeculosus were infected experimentally by allowing them to take blood meals on one individual long-eared owl (Asio otus) and one tawny owl (Strix aluco) harbouring mature gametocytes of H. noctuae (lineage hCIRCUM01) and H. syrnii (hCULCIB01), respectively. The engorged insects were maintained in the laboratory at 16–18 °C, and dissected at intervals in order to follow the development of ookinetes, oocysts and sporozoites. We also observed in vitro development of sexual stages of both parasites by exposure of infected blood to air. The parasite lineages were determined by polymerase chain reaction-based methods. Bayesian phylogeny was constructed in order to determine the relationships of owl parasites with other avian Haemoproteus spp., for which vectors have been identified. Results Both H. noctuae and H. syrnii completed sporogony in C. nubeculosus, and H. noctuae completed sporogony in C. impunctatus. Ookinetes, oocysts and sporozoites of these parasites were reported and described. Gametes and ookinetes of both species readily developed in vitro. In accordance with sporogony data, the phylogenetic analysis placed both parasite lineages in a clade of Culicoides spp.-transmitted avian Haemoproteus (Parahaemoproteus) spp. Conclusions Culicoides nubeculosus and C. impunctatus are vectors of H. noctuae and H. syrnii. Phylogenies based on cytochrome b gene indicate parasite-vector relationships, and we recommend using them in predicting possible parasite-vector relationships and planning research on avian Haemoproteus spp. vectors in wildlife.

Pond bat Myotis dasycneme is distributed from Belgium to Eastern Siberia, between 44°–50° N and 60° N. The total population of M. dasycneme is probably declining, but supporting data are incomplete. The conservation status of the species is not yet completely assessed. Surveys of hibernacula and foraging sites have been carried out recently in Northwest Russia. Evidence of a recent increase in numbers of wintering bats have been recorded, a northwards expansion is supposed. However, the local pond bat is still under threat as its most significant population is concentrated in only one hibernaculum, which is not well protected. [ABSTRACT FROM AUTHOR]

Here we studied cell-free plasma DNA (cfDNA) collected from subjects with advanced lung cancer whose tumors had developed resistance to the epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) AZD9291. We first performed next-generation sequencing of cfDNA from seven subjects and detected an acquired EGFR C797S mutation in one; expression of this mutant EGFR construct in a cell line rendered it resistant to AZD9291. We then performed droplet digital PCR on serial cfDNA specimens collected from 15 AZD9291-treated subjects. All were positive for the T790M mutation before treatment, but upon developing AZD9291 resistance three molecular subtypes emerged: six cases acquired the C797S mutation, five cases maintained the T790M mutation but did not acquire the C797S mutation and four cases lost the T790M mutation despite the presence of the underlying EGFR activating mutation. Despite the fact that aortic valve replacement surgery is well developed and accompanied by high efficiency, it is still far from ideal and associated with a large number of complications. In this relation, the methods of surgical treatment based on valve-preserving philosophy surpass traditional prosthetics, since they may decrease risks at early and long-term periods after the operation, which is especially important in elderly and senile patients. The manuscript presents the results of a study on application of the ultrasonic decalcification in 70 patients with calcified aortic valve stenosis. After surgery, up to 13 months, a significant decrease in the systolic pressure gradient on the valve and an increase in the area of the effective valve opening were observed. This shows the effectiveness of the ultrasonic decalcification in the elderly patient population.

University library acting an important role in preparing future qualified specialists in various fields by providing them with the necessary informational resources. This paper is focused on its processes. Modern development of automation of library processes in University quite rapidly passes. Implementation of computer and telecommunication technologies in the work of libraries is no longer fashion, but an urgent requirement to improve productivity and quality of library and information services through the creation, usage and integration of electronic resources and the automation of library processes. Created electronic catalogs and arrays of digital documents together with the means of telecommunication are necessary for the fulfillment of the main task is providing users' access to different types of informational resources of libraries at minimum total cost. Acquisition and organizational processes of university library differ from other types of libraries through the connection with the educational process. Informatization performances a huge role in library functioning. Introduction of new systems, including web-oriented library system, such as Koha, helps to provide quality services to the library users, and in a co-pandemic period provide these services remotely. And the statistics presented in the publication confirm this. To solve the current problems completing the library fund is a constant exchange of data between different structural units of high school: teaching management, scientific research department, department, institutes, departments, etc. Relevance of data in such an exchange is quite low, as in the best-case data is updated twice a year, and human resources that are responsible for status of this information is quite large. The library and the appropriate University departments are interested in obtaining relevant data in an automated mode.
