Simple Summary: Hepatocellular cancer (HCC) is the most common and lethal subtype of liver cancer without effective therapeutics. Understanding and targeting cancer stem cells (CSCs), a stem-cell-like subpopulation, which are emerging as effective ways to decipher tumor biology and develop therapies, may help to revolutionize cancer management. Cancer/testis antigen Maelstrom (MAEL) has been implicated in the regulation of CSC phenotypes, while the role of CSCs remains unclear. We demonstrated that MAEL positively regulates cancer stem-cell-like properties in HCC, and MAEL silencing provokes tumor cells' sensitivity to sorafenib. We further discovered that the MAEL-dependent stemness was operated via PGST2/IL8/AKT/STAT3 signaling. Collectively, our study suggests the MAEL/PGST2 axis as a potential therapeutic target against CSC and sorafenib resistance in HCC. Collectively, our work identifies MAEL as an important stemness regulation gene in HCC. 3. Maelstrom promotes tumor metastasis through regulation of FGFR4 and epithelial-mesenchymal transition in epithelial ovarian cancer.

Background: Increasing evidence has indicated that Maelstrom (MAEL) plays an oncogenic role in various human carcinomas. However, the exact function and mechanisms by which MAEL acts in epithelial ovarian cancer (EOC) remain unclear. Results: This study demonstrated that MAEL was frequently overexpressed in EOC tissues and cell lines. Overexpression of MAEL was positively correlated with the histological grade of tumors, FIGO stage, and pT/pN/pM status (p < 0.05), and it also acted as an independent predictor of poor patient survival (p < 0.001). Ectopic overexpression of MAEL substantially promoted invasiveness/metastasis and induced epithelial-mesenchymal transition (EMT), whereas silencing MAEL by short hairpin RNA effectively inhibited its oncogenic function and attenuated EMT. Further study demonstrated that fibroblast growth factor receptor 4 (FGFR4) was a critical downstream target of MAEL in EOC, and the expression levels of FGFR4 were significantly associated with MAEL. Conclusion: Our findings suggest that overexpression of MAEL plays a crucial oncogenic role in the development and progression of EOC through the upregulation of FGFR4 and subsequent induction of EMT, and also provide new insights on its potential as a therapeutic target for EOC. The spermatogenesis of animal is essential for the reproduction and a very large number of genes participate in this procession. The Maelstrom (Mael) is identified essential for spermatogenesis in both Drosophila and mouse, though the mechanisms appear to differ. It was initially found that Mael gene is necessary for axis specification of oocytes in Drosophila , and recent studies suggested that Mael participates in the piRNA pathway. In this study, we obtained Bombyx mori Mael mutants by using a binary transgenic CRISPR/Cas9 system and analyzed the function of Mael in B. mori , a model lepidopteran insect. The results showed that BmMael is not necessary for piRNA pathway in the ovary of silkworm, whereas it might be essential for transposon elements (TEs) repression in testis. The BmMael mutation resulted in male sterility, and further analysis established that BmMael was essential for spermatogenesis. The spermatogenesis defects occurred in the elongation stage and resulted in nuclei concentration arrest. RNA-seq and qRT-PCR analyses demonstrated that spermatogenesis defects were associated with tight junctions and apoptosis. We also found that BmMael was not involved in the silkworm sex determination pathway. Our data provide insights into the biological function of BmMael in male spermatogenesis and might be useful for developing novel methods to control lepidopteron pests. Terminal regions of the Drosophila embryo are patterned by the localized activation of the Torso-RTK pathway, which promotes the downregulation of Capicua. In the short-germ beetle Tribolium, the function of the terminal system appears to be rather different, as the pathway promotes axis elongation and, in addition, is required for patterning the extra-embryonic serosa at the anterior. Here, we show that Torso signalling also induces gene expression by relieving Capicua-mediated repression in Tribolium. Given that the majority of Torso target genes remain to be identified, we established a differential gene-expression screen. A subset of 50 putative terminal target genes was screened for functions in early embryonic patterning. Of those, 13 genes show early terminal expression domains and also phenotypes were related to terminal patterning. Among others, we found the PIWI-interacting RNA factor Maelstrom to be crucial for early embryonic polarization. Tc-mael is required for proper serosal size regulation and head morphogenesis. Moreover, Tc-mael promotes growth-zone formation and axis elongation. Our results suggest that posterior patterning by Torso may be realized through Maelstrom-dependent activation of posterior Wnt domains. [ABSTRACT FROM AUTHOR]

L'œuvre d'Edgar Allan Poe contient une série d'éléments qui peuvent être décrits comme des espaces clos littéraires : ces espaces textuels ne s'ouvrent pas au mouvement libre, à la communication, à l'échange, mais ils restent fermés sur eux et sont marqués d'un mouvement en spirale. La présente contribution ambitionne de mettre en relief cette caractéristique de l'œuvre littéraire de Poe en se fondant en particulier sur le conte The Purloined Letter et prenant aussi en compte la traduction française du texte par Charles Baudelaire. Celle-ci sert en même temps de base pour une réflexion sur des aspects de traductibilité. L'article part d'une microanalyse du premier paragraphe du récit de Poe pour montrer à quel point ce passage est composé selon une logique de la circularité et de la fermeture. Dans une deuxième étape, l'examen des espaces clos s'étend au niveau du récit global ainsi qu'à d'autres textes de Poe. L'analyse se focalise en particulier sur la structure de la mise en abyme et sur les formes de la spirale, du tourbillon, du maelström comme géométries immanentes, autoréflexives de l'œuvre littéraire de Poe. However, the mechanism whereby Mael is involved in this pathway remains unknown. Recent biochemical and structural studies, along with bioinformatic analyses of Mael-associating RNAs in vivo, have shed light on the functional aspects of Mael in the piRNA pathway. Chromatin of male and female gametes undergoes a number of reprogramming events during the transition from germ cell to embryonic developmental programs. Although the rearrangement of DNA methylation patterns occurring in the zygote has been extensively characterized, little is known about the dynamics of DNA modifications during spermatid maturation. Here, we demonstrate that the dynamics of 5-carboxylcytosine (5caC) correlate with active transcription of LINE-1 retroelements during murine spermiogenesis. We show that the open reading frames of active and evolutionary young LINE-1s are 5caC-enriched in round spermatids and 5caC is eliminated from LINE-1s and spermiogenesis-specific genes during spermatid maturation, being simultaneously retained at promoters and introns of developmental genes. Our results reveal an association of 5caC with activity of LINE-1 retrotransposons suggesting a potential direct role for this DNA modification in fine regulation of their transcription., Blythe et al investigate the patterns of oxidized forms of the 5-methylcytosine DNA modification during spermatogenesis. They find that open reading frames of evolutionarily young and transcriptionally active LINE-1 retrotransposons are enriched in 5-carboxylcytosine (5caC) during spermatid maturation, suggesting a link between 5cac and LINE-1 activity.

14. The Rebound Attack: Cryptanalysis of Reduced Whirlpool and Grøstl.

- Abstract
In this work, we propose the rebound attack, a new tool for the cryptanalysis of hash functions. The idea of the rebound attack is to use the available degrees of freedom in a collision attack to efficiently bypass the low probability parts of a differential trail. The rebound attack consists of an inbound phase with a match-in-the-middle part to exploit the available degrees of freedom, and a subsequent probabilistic outbound phase. Especially on AES based hash functions, the rebound attack leads to new attacks for a surprisingly high number of rounds. We use the rebound attack to construct collisions for 4.5 rounds of the 512-bit hash function Whirlpool with a complexity of 2
120 compression function evaluations and negligible memory requirements. The attack can be extended to a near-collision on 7.5 rounds of the compression function of Whirlpool and 8.5 rounds of the similar hash function Maelstrom. Additionally, we apply the rebound attack to the SHA-3 submission Grøstl, which leads to an attack on 6 rounds of the Grøstl-256 compression function with a complexity of 2120 and memory requirements of about 264 . Pachytene pi RNAs are a class of Piwi-interacting small RNAs abundant in spermatids of the adult mouse testis. They are processed from pi RNA primary transcripts by a poorly understood mechanism and, unlike fetal transposon-derived pi RNAs, lack complementary targets in the spermatid transcriptome. We report that immunopurified complexes of a conserved pi RNA pathway protein Maelstrom ( MAEL) are enriched in MIWI (Piwi partner of pachytene piRNAs), Tudor-domain proteins and processing intermediates of pachytene pi RNA primary transcripts. We provide evidence of functional significance of these complexes in Mael 129 knockout mice that exhibit spermiogenic arrest with acrosome and flagellum malformation. Mael 129-null mutant testes possess low levels of pi RNAs derived from MAEL-associated piRNA precursors and exhibit reduced translation of numerous spermiogenic mRNAs including those encoding acrosome and flagellum proteins. These translation defects in haploid round spermatids are likely indirect, as neither MAEL nor pi RNA precursors associate with polyribosomes, and they may arise from an imbalance between pachytene pi RNAs and MIWI. Further, the roles and relationships between student and lecturer are explored and related to the Māori concept of ako, the idea of reciprocal learning and teaching. This exploration is held by the tenet that the arts process is integral to the teaching environment at Whitecliffe College, with the argument being that trusting in the poietic process supports and facilitates embodied knowledge. The piece further provides a contemplation of students’ experiences of their individual maelstrom (their deep journeys into themselves), the roles of the lecturers as those who help hold those journeys, and the learning gems that can be found along the way.

Mansaram is an Indo-Canadian artist who immigrated to Canada in 1966 with a prior interest in the media ideas of Marshall McLuhan, sparked by reading about him in LIFE Magazine. In Toronto the media guru soon introduced him to Av Isaacs at his Yonge Street gallery, which led to a 1967 Happening there called East-West Intersect, influenced by McLuhan’s ideas. Between 1966 and 1972 Mansaram worked on his Rear View Mirror series of paintings and collages, to one of which McLuhan contributed several items of textual content. Collage with its mosaic structures appealed to McLuhan because he thought it better represented the post-literate “allatonceness” of electronic media and acoustic space, which better integrated the full human sensorium and called for pattern recognition for comprehension. McLuhan had a high regard for artists for their integral awareness and sensitivity to changes in sense perception, enabling them to act as a distant early warning (DEW) line against potentially harmful effects of technology. He viewed their art as anti-environments to the electronic media maelstrom. Mansaram has enjoyed increasing recognition through recent exhibitions, but some of Canada’s premier art galleries have yet to acquire or recognize his art, although the Royal Ontario Museum is planning to do so. In Drosophila, Maelstrom is a conserved component of the perinuclear nuage, a germline-unique structure that appears to serve as a site for Piwi-interacting RNA (piRNA) production to repress deleterious transposons. Maelstrom also functions in the nucleus as a transcriptional regulator to repress the expression of microRNA-7, a process that is essential for the proper differentiation of germline stem cells. In this paper, we report another function of Maelstrom in regulating oocyte determination independently of its transposon silencing and germline stem cell differentiation activities. In Drosophila, the conserved serine 138 residue in Maelstrom is required for its phosphorylation, an event that promotes oocyte determination. Phosphorylation of Maelstrom is required for the repression of the pachytene checkpoint protein Sir2, but not for transposon silencing or for germline stem cell differentiation. We identify Polo as a kinase that mediates the phosphorylation of Maelstrom. Our results suggest that the Polo-mediated phosphorylation of Maelstrom may be a mechanism that controls oocyte determination by inactivating the pachytene checkpoint via the repression of Sir2 in Drosophila ovaries. Abstract: Integrity of the germline genome is essential for the production of viable gametes and successful reproduction. In mammals, the generation of gametes involves extensive epigenetic changes (DNA methylation and histone modification) in conjunction with changes in chromosome structure to ensure flawless progression through meiotic recombination and packaging of the genome into mature gametes. Although epigenetic reprogramming is essential for mammalian reproduction, reprogramming also provides a permissive window for exploitation by transposable elements (TEs), autonomously replicating endogenous elements. Expression and propagation of TEs during the reprogramming period can result in insertional mutagenesis that compromises genome integrity leading to reproductive problems and sporadic inherited diseases in offspring. Recent work has identified the germ cell associated PIWI Interacting RNA (piRNA) pathway in conjunction with the DNA methylation and histone modification machinery in silencing TEs. In this review we will highlight these recent advances in piRNA mediated regulation of TEs in the mouse germline, as well as mention the repercussions of failure to properly regulate TEs. Ecosystems result from the integration of populations of different species in a common environment. They rarely remain steady for long, and fluctuations lie in the very essence of the ecosystems and of every one of the … populations. The truth of Margalef's observation was not apparent to scientists, fishermen, or politicians in the 1860s. The conclusion that ecosystems vary, and also the concepts used (population, environment, steady state, and even ecosystem) were virtually unknown. At that time, it was apparent only that catches varied from year to year despite the best efforts of the fishermen. The reality of this variation was painfully apparent. The amount of fish caught, and hence available as food, was much less in some years, leading at times to hunger. However, in other years the amount of fish caught was so high that prices plummeted and much went to waste, leading to economic difficulties. Catches might increase steadily for several years, encouraging investment in fishing gear and processing equipment, and leading to relocation of work forces, only to subsequently decline. Investments were lost, and people's economic plans and security compromised. The value of resources that are so variable is lessened. As Hugh Miller pointed out in an 1829 description of a Scottish herring fishery in the Inverness Courier: Its value must therefore be estimated in the same way that Directors of an insurance office would calculate upon that of the life of one who had a hereditary tendency to apoplexy. The mouse maelstrom (MAEL) gene has been found to be expressed in male germ cells and to play a role in spermatogenesis. Here, we cloned the human MAEL gene by digital differential display and found that, among human tissues, MAEL is only expressed in the testis, but it is also expressed in various cancer cell lines. The transcription start site of the MAEL gene is 74-bp upstream of the start codon. The region from −216 to +150 is the basal promoter of the MAEL gene, and a CpG island (−295 to +148) is located in this region. Treatment with the demethylating agent 5′-Aza-2-Deoxycytidine significantly upregulated MAEL expression. These results suggest that MAEL is a novel cancer/testis-associated gene and is regulated by DNA methylation. [ABSTRACT FROM AUTHOR]

This paper discusses the current state of thought amongst the Sustainability Indicator (SI) community, what has been achieved and where we are succeeding and failing. Recent years have witnessed the rise of “alternative facts” and “fake news” and this paper discusses how SIs fit into this maelstrom, especially as they are themselves designed to encapsulate complexity into condensed signals and it has long been known that SIs can be selectively used to support polarized sides of a debate. This paper draws from chapters in a new edited volume, the “Routledge Handbook of Sustainability Indicators and Indices”, edited by the authors. The book has 34 chapters written by a total of 59 SI experts from a wide range of backgrounds, and attempts to provide a picture of the past and present, strengths and weaknesses of SI development today. This paper is an “analysis of those analyses”—a mindful reflection on reflection, and an assessment of the malign and benign forces at work in 2018 within the SI arena. Finally, we seek to identify where SIs may be going over the coming, unpredictable years.

Length and complexity of illness and treatment in early breast cancer mean that the acute phase may extend over months. Employing concepts of “trajectory” and “work” from chronic illness, we explored the temporality of the illness experience for women as it unfolded from diagnosis through acute treatment. We performed multiple, qualitative interviews with 14 women at critical points in their first year postdiagnosis. We differentiated between subphases of the acute breast cancer trajectory: becoming a person with breast cancer: a turning point; first steps in treatment: struggling to protect self-concept; being and feeling ill: “cure” as suffering; and conditional “recovering”: an uncertain future. Each subphase represented the distinctive context in which illness management took place with consequences for the work women and those close to them engaged in, to manage the practical, emotional, relational, and existential demands of an illness that intruded every aspect of their lives.

Si bien su estudio ha sido en gran parte ignorado, su análisis es crucial para la comprensión del pensamiento mexicano durante y después del conflicto. Tras un balance historiográfico, este trabajo define el escenario de dichos debates de opinión, identifica su punto más álgido durante el año 1917 y analiza su evolución en tres de sus principales foros: la prensa, el Poder Legislativo y las asambleas estudiantiles. While in the thick of a revolutionary maelstrom, Mexico was the setting for animated debates centered on the direct implications that World War I might have on the nation’s future. Although its study has been largely ignored, analysis of the Great War is crucial for understanding Mexican thought during and after the conflict. In addition to offering a historiographical assessment, this work elaborates the scenes of said debates, identifies their most decisive moment during the year 1917, and analyzes their evolution in three of their principle forums: the press, the Legislative Branch, and student assemblies.

Summary In Drosophila , 23–30 nt long PIWI-interacting RNAs (piRNAs) direct the protein Piwi to silence germline transposon transcription. Most germline piRNAs derive from dual-strand piRNA clusters, heterochromatic transposon graveyards that are transcribed from both genomic strands. These piRNA sources are marked by the heterochromatin protein 1 homolog Rhino (Rhi), which facilitates their promoter-independent transcription, suppresses splicing, and inhibits transcriptional termination. Here, we report that the protein Maelstrom (Mael) represses canonical, promoter-dependent transcription in dual-strand clusters, allowing Rhi to initiate piRNA precursor transcription. Mael also represses promoter-dependent transcription at sites outside clusters. At some loci, Mael repression requires the piRNA pathway, while at others, piRNAs play no role. We propose that by repressing canonical transcription of individual transposon mRNAs, Mael helps Rhi drive non-canonical transcription of piRNA precursors without generating mRNAs encoding transposon proteins. En este sentido, la presente investigación trata de justificar el interés y necesidad del crecimiento de la competencia mediática como forma de expresión autónoma y crítica. Esta temática centra el interés de diferentes organismos e instituciones de carácter nacional e internacional y guarda estrecha relación con la educación mediática que debe llevarse a cabo por los formadores y también por los profesionales de la comunicación The communicative ecosystem develops and takes place in today’s technological society where hypercommunication is prioritized. Within this context, an enormous number of screens broadcast audiovisual contents that shape the recipients’ opinion and behavior. The different types of face-to-face and virtual interaction, as well as the media maelstrom, require training and literacy, not only communicative but also audiovisual, aimed at the attainment of a critical attitude through the development of the skills necessary for face-to-face and virtual communication. In this sense, this research intends to justify the interest and need for growth of media competence as a form of autonomous and critical expression. This topic attracts the interest of different national and international organizations and institutions, and is closely related to the media education that should be carried out by trainers and communication professionals O ecossistema comunicativo cresce e ocorre na sociedade tecnológica de hoje, onde a hipercomunicação é o fator principal. Between 1967 and 1973 Barbara Weinberger and Charles Madge undertook a longitudinal study of art students at Coventry and Birmingham Colleges of Art, incorporating pre-diploma to final year students and their teachers, with a focus on fine art students. This article revisits the study in light of accounts from participants, archival documents and reviews. It also looks at the study's value both as an accurate description of art school socialisation and its effectiveness as a piece of sociology. It then looks at some of the main issues raised by the study and how they have influenced writing since and their value today. The article aims to clarify some of the obscurities of the study and offer a critical context within which it can be read as a now unique historical document.

The authors draw attention to certain salient features of the process, which has combined elements of freedom and bondage, playing out largely on the terrain of employment-systems that have combined for four centuries the free trafficking of slaves and indentured servants with the contested liberty of wage-workers to abandon their employer. One of the most significant episodes in the history of European migration was just playing out: a migratory movement destined to continue until the outbreak of the war of 1914-1918. On the eve of the war of 1914-1918, industrial capital was already beginning to understand that it could not continue to rely on purely local recruitment of its assembly-line workers. In certain cases, the transition from sugar plantations to export-processing zones (EPZs) was remarkably smooth. Unhappily, there has been a maelstrom of problems for dark matter theories over the last few years and many serious difficulties still have no resolution in sight. This article reviews the evidence for dark matter in galaxies. The haloes built up by hierarchical merging in dark matter cosmogonies are cusped and dominated by dark matter at the center. Evidence from the microlensing optical depth towards Baade's Window and from dynamical modelling of the Galactic bar already suggests that the Galactic halo is not cusped. Similarly, evidence from the stability of unbarred disk galaxies, as well as the survival of fast bars in barred galaxies, suggests that the this result holds good more generally. Judged on the data from galactic scales alone, the case for dark matter is weak and non-standard theories of gravity provide a better description. The roles of music publishers and record companies especially [may be] about to alter. In an institutional economic analysis, I will analyze the present situation in this industry, where Information Technology impacts on this system, and how in general the industry is likely to change. The establishment of body axes in multicellular organisms requires accurate control of microtubule polarization. Mutations in Drosophila PIWI-interacting RNA (piRNA) pathway genes often disrupt the axes of the oocyte. This results from the activation of the DNA damage checkpoint factor Checkpoint kinase 2 (Chk2) due to transposon derepression. A piRNA pathway gene, maelstrom (mael), is critical for the establishment of oocyte polarity in the developing egg chamber during Drosophila oogenesis. We show that Mael forms complexes with microtubule-organizing center (MTOC) components, including Centrosomin, Mini spindles, and γTubulin. We also show that Mael colocalizes with αTubulin and γTubulin to centrosomes in dividing cyst cells and follicle cells. MTOC components mislocalize in mael mutant germarium and egg chambers, leading to centrosome migration defects. During oogenesis, the loss of mael affects oocyte determination and induces egg chamber fusion. Finally, we show that the axis specification defects in mael mutants are not suppressed by a mutation in mnk, which encodes a Chk2 homolog. These findings suggest a model in which Mael serves as a platform that nucleates other MTOC components to form a functional MTOC in early oocyte development, which is independent of Chk2 activation and DNA damage signaling. PIWI-interacting RNA (piRNA) is a germline-specific class of small non-coding RNAs that repress transposons in the gonads. Mael, which comprises a high mobility group box and a MAEL domain, is one of the key players in piRNA-mediated transposon silencing. However, the mechanism whereby Mael is involved in this pathway remains unknown. Recent biochemical and structural studies, along with bioinformatic analyses of Mael-associating RNAs in vivo, have shed light on the functional aspects of Mael in the piRNA pathway. We summarize the current understanding of Mael functions in the piRNA pathway, particularly in Drosophila and in mice.
