Colorectal cancer (CRC) ranks third in global incidence and second in mortality. However, a comprehensive predictive model for CRC prognosis, immunotherapy response, and drug sensitivity is still lacking. Various types of programmed cell death (PCD) are crucial for cancer occurrence, progression, and treatment, indicating their potential as valuable predictors. Fourteen PCD genes were collected and subjected to dimensionality reduction using regression methods to identify key hub genes. Predictive models were constructed and validated based on bulk transcriptomes and single-cell transcriptomes. Furthermore, the tumor microenvironment, immunotherapy response, and drug sensitivity profiles among patients with CRC were explored and stratified by risk. A risk score incorporating the PCD genes FABP4, AQP8, and NAT1 was developed and validated across four independent datasets. Patients with CRC who had a high-risk score exhibited a poorer prognosis. Unsupervised clustering algorithms were used to identify two molecular subtypes of CRC with distinct features. The risk score was combined with the clinical features to create a nomogram model with superior predictive performance. Additionally, patients with high-risk scores exhibited decreased immune cell infiltration, higher stromal scores, and reduced responsiveness to immunotherapy and first-line clinical drugs compared with low-risk patients. Furthermore, the top ten non-clinical first-line drugs for treating CRC were selected based on their predicted IC50 values. Our results indicate the efficacy of the model and its potential value in predicting prognosis, response to immunotherapy, and sensitivity to different drugs in patients with CRC. Drought is expected to increase in temperate regions in the coming decades due to global climate change and will strongly affect nitrogen (N) transformation and balance. However, the underlying mechanisms (soil microenvironment vs. biotic community) of rainfall reduction induced changes in soil N transformation rates, and whether the responses of soil N transformation rates to rainfall reduction vary over time remains poorly understood. In this study, we experimentally examined rainfall reduction effects on soil N transformation rates over two (the second and third experimental years) growing‐seasons in a temperate meadow. We applied four treatments: ambient rainfall, 30%, 50%, and 70% reduction in rainfall. Our findings demonstrated that, 70% rainfall reduction significantly decreased the average net N mineralization and nitrification rates by 68.4% and 72.0%, respectively. Soil nitrous oxide emission was profoundly varied between the experimental years, with it was sensitive no only to changes in annual precipitation amount, but also to seasonal distribution, the highest emissions appeared at the early growing‐season. Moreover, plant diversity, microbial biomass, and soil NO3−‐N content mainly regulated soil N transformation rates in response to rainfall reduction in the greater annual precipitation year; but soil N transformation rates were mainly controlled by soil water content in the less annual precipitation year. We suggest that to accurately project future ecosystem functions and improving the prediction of N cycling responses to changing precipitation patterns, stronger focus should be allocated not only to changes in precipitation amounts, but also to inter‐ and intra‐annual differences in precipitation distribution. To understand how the soil N transformation rates respond to different rainfall levels, we used rainout shelters to simulate reduced precipitation amount in a temperate meadow. We examined a suite of soil N transformations responses. Our results showed that soil N transformation rates decreased along the gradient of declining in rainfall amount over the two sampling years, with inter‐ and intra‐annual variation in precipitation distribution also played important roles in determining soil N transformation rates. The high values of each soil N transformation rates were appeared at different periods of the growing‐seasons. We also examined the pathways associated with the responses of soil N transformation rates to rainfall reduction in the two contrasting precipitation years. In summary, we conclude that rainfall reduction led to reductions in soil N transformation rates through a combination of plant, microbial, and soil factors during the greater annual precipitation. Nonetheless, in the less annual precipitation, the key factor determining N transformations was found to be soil water content. These findings offer valuable insights for forecasting interactions within the plant‐microbe‐soil N cycling system under forthcoming precipitation patterns within the context of climate change. Cooking methods can change the composition of foods and have important effects on human health. The Chinese people have developed many distinct and unique cooking methods. However, the daily cooking patterns of Chinese people and the characteristics and evolution of trends in cooking patterns commonly used by Chinese consumers remain unclear. The objective of this study was to identify the major cooking patterns and discuss their effects on human health, as well as to identify the cooking pattern consumer clusters and the evolution of trends in Chinese consumer cooking patterns. From March to June 2021, this study interviewed 4,710 residents in Eastern China regarding the consumption frequency of each cooking method when food is prepared at home or when eating out. Exploratory factor analysis, K-Means cluster analysis, Chi-square test, pairwise comparisons of multiple sample rates, and multivariate linear regression were used to identify the cooking patterns and cooking pattern consumer clusters, to assess differences in consumption preferences between consumer clusters, and to examine the relationship between demographic characteristic variables and different cooking patterns. Results revealed three major cooking patterns, namely traditional Chinese (cooking methods with native Chinese characteristics), bland, and high-temperature cooking patterns, as well as seven cooking pattern consumer clusters and their demographic characteristics in the Eastern Chinese population. With increases in age, education level, and income, consumers tended to choose the healthy "Bland" cooking pattern. Further, there was a higher proportion of people aged 36–65 years in the C3 cluster, which is characterized by the "Bland" cooking pattern. However, participants who were male and younger made fewer healthy choices in their cooking patterns. Specifically, a higher proportion of participants aged 21–35 years were found in the C5 cluster, which is characterized by the unhealthy "High-temperature" cooking pattern. Therefore, culinary health education should focus on individuals who are male and young. Specifically, the shift in cooking patterns among people aged 21–35 years should receive special attention. Questions: Anthropogenic climate change has become a critical environmental problem and is profoundly affecting various ecosystems, especially desert ecosystems. However, very few studies have explored the effects of extreme precipitation on the community structure and ecosystem functions in drylands. Moreover, whether local microtopography will interact with extreme precipitation on plant growth by changing water retention and other processes is still unclear. Location: The Gurbantunggut Desert in Central Asia. Methods: We conducted a two‐year field experiment to assess the effect of extreme precipitation (+65% of ambient growing‐season precipitation amounts) on the composition and productivity of a desert ephemeral plant community growing in sand dunes of the Gurbantunggut Desert in Central Asia. We further determined how much slope position modulates this response. Results: We found that the cover, density, height, species richness, and biomass of ephemeral plants increased with an increase in precipitation, and the strongest direct effect of biomass was a positive effect on density of ephemeral plants. Additionally, no significant differences in the sensitivity of biomass to the increased precipitation were found between slope positions. Conclusions: The results indicate that extreme precipitation primarily increased the above‐ground net primary production by increasing the density, rather than the height and species richness, of the ephemeral community; no interactive effects were observed between sand dune slope positions and the sensitivity of biomass to the extreme precipitation treatment. Background: Ferroptosis is a novel form of regulated cell death that plays a critical role in tumorigenesis. The purpose of this study was to establish a ferroptosis-associated gene (FRG) signature and assess its clinical outcome in gastric cancer (GC). Methods: Differentially expressed FRGs were identified using gene expression profiles from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) database. Univariate and least absolute shrinkage and selection operator (LASSO) Cox regression analyses were performed to construct a prognostic signature. The model was validated using an independent GEO dataset, and a genomic-clinicopathologic nomogram integrating risk scores and clinicopathological features was established. Results: An 8-FRG signature was constructed to calculate the risk score and classify GC patients into two risk groups (high- and low-risk) according to the median value of the risk score. The signature showed a robust predictive capacity in the stratification analysis. A high-risk score was associated with advanced clinicopathological features and an unfavorable prognosis. The predictive accuracy of the signature was confirmed using an independent GSE84437 dataset. Patients in the two groups showed different enrichment of immune cells and immune-related pathways. Finally, we established a genomic-clinicopathologic nomogram (based on risk score, age, and tumor stage) to predict the overall survival (OS) of GC patients. Global change factors, such as variation in precipitation regimes and nitrogen (N) deposition, are likely to occur simultaneously and may have profound impacts on the relative abundance of grasses differing in functional traits, such as C3 and C4 species. We conducted an extreme drought and re-watering experiment to understand differences in the resistance and recovery abilities of C3 and C4 grasses under different N deposition scenarios. A C3 perennial grass (Leymus chinensis) and two C4 grasses (annual species Chloris virgata and perennial species Hemarthria altissima) that co-occur in Northeast China were selected as experimental plants. For both C3 and C4 grasses, N addition caused a strong increase in biomass and resulted in more severe drought stress, leading to a change in the dominant photosynthetic limitation during the drought periods. Although N addition increased antioxidant enzyme activities and protective solute concentrations, the carbon fixing capacity did not fully recover to pre-drought levels by the end of the re-watering period. N addition resulted in lower resilience under the drought conditions and lower resistance at the end of the re-watering. However, N addition led to faster recovery of photosynthesis, especially in the C3 grass, which indicate that the effect of N addition on photosynthesis during drought was asymmetric, especially in the plants with different photosynthetic nitrogen use efficiency (PNUE). These findings demonstrated that nitrogen deposition may significant alter the susceptibility of C3 and C4 grass species to drought stress and re-watering, highlighting the asymmetry between resistance and resilience and to improve our understanding about plant responses to climate change. Backgrounds: Periprocedural myocardial injury (PMI) after elective percutaneous coronary intervention (PCI) significantly influences the prognosis of coronary artery disease (CAD). However, it was unclear whether the occurrence of PMI was associated with a series of controllable factors, such as PCI strategy or severity of CAD.Methods: A total of 544 consecutive stable CAD patients underwent elective PCI were enrolled. The main outcome is PMI, defined as troponin T after PCI was at least one value above the 99th percentile upper reference limit. Major adverse cardiac events (MACE), including all-cause death, repeat myocardial infarction and target vessel revascularization were record in the period of follow-up. Univariate and multivariate analysis was applied to assess predictors for the occurrence of PMI.Results: The incidence of PMI was 38.8% in the study. Compared with non-PMI patients (n = 333), PMI patients (n = 211) had more diseased vessels, higher Gensini and Syntax score. Meanwhile, there were higher incidence of MACE in PMI groups (9.5% vs. 3.2%, P < 0.01). We found that PMI patients underwent higher proportion of multi-vessel PCI simultaneously (32.2% vs. 10.5%, P < 0.01) and had more stents implanted (1.8 ± 0.8 vs. 1.4 ± 0.6, P < 0.01). Importantly, after simultaneously adjusted by other factors (such as age, diabetes, total cholesterol, number of diseased vessels, Gensini score and stent length), the risk of PMI was still increased 84% by multi-vessel PCI independently (OR = 1.654, 95% CI = 1.004-2.720, P < 0.05).Conclusions: The phenomenon of PMI occurred more commonly in stable CAD patients underwent multi-vessel PCI. Variations in the carbon isotope signature of leaf dark-respired CO2 (δ13CR) within a single night is a widely observed phenomenon. However, it is unclear whether there are plant functional type differences with regard to the amplitude of the nighttime variation in δ13CR. These differences, if present, would be important for interpreting the short-term variations in the stable carbon signature of ecosystem respiration and the partitioning of carbon fluxes. To assess the plant functional type differences relating to the magnitude of the nighttime variation in δ13CR and the respiratory apparent fractionation, we measured the δ13CR, the leaf gas exchange, and the δ13C of the respiratory substrates of 22 species present in the agricultural-pastoral zone of the Songnen Plain, northeast China. The species studied were grouped into C3 and C4 plants, trees, grasses, and herbs. A significant nocturnal shift in δ13CR was detected in 20 of the studied species, with the magnitude of the shift ranging from 1‰ to 5.8‰. The magnitude of the nighttime variation in δ13CR was strongly correlated with the daytime cumulative carbon assimilation, which suggests that variation in δ13CR were influenced, to some extent, by changes in the contribution of malate decarboxylation to total respiratory CO2 flux. There were no differences in the magnitude of the nighttime variation in δ13CR between the C3 and C4 plants, as well as among the woody plants, herbs and graminoids. Leaf respired CO2 was enriched in 13C compared to biomass, soluble carbohydrates and lipids; however the magnitude of enrichment differed between 8 pm and 4 am, which were mainly caused by the changes in δ13CR. We also detected the plant functional type differences in respiratory apparent fractionation relative to biomass at 4 am, which suggests that caution should be exercised when using the δ13C of bulk leaf material as a proxy for the δ13C of leaf-respired CO2. Objectives: Coronary artery disease (CAD) severity is associated with patient prognosis. However, few efficient scoring systems have been developed to screen severe CAD in patients with stable angina and suspected CAD before coronary angiography. Here, we present a novel scoring system for CAD severity before elective coronary angiography. Methods: Five hundred fifty-one patients with stable angina who were admitted for coronary angiography were enrolled in this study. Patients were divided into training (n = 347) and validation (n = 204) cohorts. Severe CAD was defined as having a Gensini score of 20 or more. All patients underwent echocardiography (ECG) to detect ejection fraction and aortic valve calcification (AVC). Multivariable analysis was applied to determine independent risk factors and develop the scoring system. Results: In the training cohort, age, male sex, AVC, abnormal ECG, diabetes, hyperlipidemia, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol were identified as independent factors for severe CAD by multivariable analysis, and the Severe Prediction Scoring (SPS) system was developed. C-indices of receiver operating characteristic (ROC) curves for severe CAD were 0.744 and 0.710 in the training and validation groups, respectively. The SPS system also performed well during calibration, as demonstrated by Hosmer-Lemeshow analysis in the validation group. Compared with the Diamond-Forrester score, the SPS system performed better for severe CAD prediction before elective coronary angiography. Conclusions: Severe CAD prediction was achieved by analyzing age, sex, AVC, ECG, diabetes status, and lipid levels. ABSTRACT Differences in light quality penetration within a leaf and absorption by the photosystems alter rates of CO2 assimilation in C3 plants. It is also expected that light quality will have a profound impact on C4 photosynthesis due to disrupted coordination of the C4 and C3 cycles. To test this hypothesis, we measured leaf gas exchange, 13CO2 discrimination (Δ13C), photosynthetic metabolite pools and Rubisco activation state in Zea mays and Miscanthus × giganteus under steady-state red, green, blue and white light. Photosynthetic rates, quantum yield of CO2 assimilation, and maximum phosphoenolpyruvate carboxylase activity were significantly lower under blue light than white, red and green light in both species. However, similar leakiness under all light treatments suggests the C4 and C3 cycles were coordinated to maintain the photosynthetic efficiency. Measurements of photosynthetic metabolite pools also suggest coordination of C4 and C3 cycles across light treatments. The energy limitation under blue light affected both C4 and C3 cycles, as we observed a reduction in C4 pumping of CO2 into bundle-sheath cells and a limitation in the conversion of C3 metabolite phosphoglycerate to triose phosphate. Overall, light quality affects rates of CO2 assimilation, but not the efficiency of CO2 concentrating mechanism. [ABSTRACT FROM AUTHOR]

In order to illustrate rotate-tilt slope zone structures, sedimentary evolution and it's controls on lithological reservoir, a case study in Banqiao Slope of Qikou Sag was presented, based on the seismic data, well-log information, core and core slice, analyzed the evolution characteristics of Banqiao Slope tectonic, sedimentary characteristics of different construction phase and its relationship with lithological reservoir. Banqiao Slope experienced two stages, average settlement and rotate-tilt. Effected by tectonic evolution, palaeogeomorphology was featured by early gentle slope and later steep slope, tectonics of E3s3-E3s1x episode showed whole settlement, with gentle palaeogeomorphology, provenances from Cangxian Lift were deposited in large areas, developed braided-river delta sedimentation.Slope rotate-tilted at the episode of E3s1z- E3d , the main structure of slope reversed, developing dustpan-shaped tectonics, with the characteristics of rotate-tilted slope controlling sandbodies, faults in source rocks and lithologic traps controlling reservoir, advantageous phase accumulation and high productivity. Sand bodies developed by sub-water channel of delta front were overlapped and distributed widely in middle and low slopes at the time of E3S1x, with good reservoir properties.Matched with faults in source rocks,they are the favorable exploration area of lithological reservoirs. According to those, some lithologic traps were found in gentle slope areas, 12 wells have been drilled,and attained high yield flow of oil and gas, drilling success rates can reach 85%. Objective: Given the crucial role of mitochondria in the prognosis and treatment of hepatocellular carcinoma (HCC), we aim to develop two independent mitochondrial scoring systems to separately predict patient prognosis and the likelihood of transarterial chemoembolization non-response (TACE NR). Methods: Mitochondria-related candidate genes were selected and analyzed using univariate Cox and LASSO Cox regression analyses to create a risk prognosis score (RPS). Univariate and LASSO logistic regression analyses were used to establish the risk diagnosis score (RDS). Alternative therapies for patients with TACE NR were explored using TIDE and oncoPredict algorithms. The Seurat package was used to study the involvement of the RDS genes in HCC differentiation. Results: The RPS accurately predicts the 1-5 year survival rates of patients with HCC, where higher RPS values were associated with poorer survival outcomes. The RDS model demonstrated a commendable performance in diagnosing TACE NR, as patients with a higher RDS exhibited a greater likelihood of TACE NR. RDS was associated with the infiltration of various immune cells, and patients with lower RDS tended to have higher response rates to immunotherapy and increased sensitivity to JAK1, rapamycin, and AZD2014. By contrast, patients with higher RDS values and a higher probability of TACE NR had more responsive to paclitaxel, dasatinib, and vincristine, suggesting that these drugs are potential alternative therapies. Single-cell sequencing studies have identified ACSM2A as a key player in HCC differentiation and a potential target for therapeutic intervention. Conclusion: The RPS and RDS are important reference points for predicting outcomes and guiding treatment decisions in patients with HCC. Resources in water-limited ecosystems are highly variable and unpredictable, and the maintenance of functional diversity among coexisting species is a crucial ecological strategy through which plants mitigate environmental stress. The comparison of differential eco-physiological responses among co-occurring plants in harsh environments could help provide deep insights into the coexistence mechanisms of competing species. Two coexisting desert shrubs with different photosynthetic pathways ( Haloxylon ammodendron and Tamarix ramosissima ) were selected in the Gurbantunggut Desert located in northwest China. This study detected variations in the water sources, photosynthetic parameters, stem water status, and non-structural carbohydrates of the two shrubs at three sites with different groundwater table depths during the growing seasons of 2015 and 2016 to identify distinct eco-physiological performances in coexisting plants with different functional types under fluctuating water conditions. The water sources of H. ammodendron shifted from soil water to groundwater, while T. ramosissima extracted water mainly from deep soil layers at both sites. Significant reductions in carbon assimilation and stomatal conductance in H. ammodendron with deeper groundwater table depth were detected during most drought periods, but no significant decreases in transpiration rate were detected with declining groundwater table depth. For T. ramosissima , all of these gas exchange parameters decreased with the progression of summer drought, and their relative reduction rates were larger compared with those of H. ammodendron . The stem water status of H. ammodendron deteriorated, and the relative reduction rates of water potential increased with deeper groundwater, whereas those of T. ramosissima did not differ with greater groundwater depth. These findings indicated that prolonged drought would intensify the impact of declining groundwater depth on the eco-physiology of both shrubs, but the extent to which the shrubs would respond differed. The two shrubs were segregated along the water-carbon balance continuum: the C 3 shrub T. ramosissima maximized its carbon fixation at an enormous cost of water, while greater carbon fixation was achieved with far greater water economy for H. ammodendron . These results demonstrated that the two shrubs prioritized carbon gain and water loss differently when faced with limited water sources. Knowledge of adequate diets can improve an individual's health status. Although previous studies have identified the main resources from which Chinese people acquire dietary knowledge, it is still unclear whether information sources regarding diets (ISRDs) can increase individuals' knowledge of adequate diets (KAD) and which ISRDs are most effective in conveying KAD to the Chinese population. In this study, we interviewed 4,710 residents in Eastern China regarding their ISRDs and KAD. Descriptive statistics, ANOVA, and multivariate linear regression were used to analyze the effectiveness of different ISRDs in transmitting KAD to Chinese individuals and to determine the relationship between ISRDs and KAD. Results showed that the KAD scores of the respondents were low overall in Eastern China. Providing dietary information through expert lectures, books, newspapers, magazines, and social media could significantly improve the average KAD score of Chinese individuals. Respondents with a greater number of ISRDs were more likely to have higher KAD scores. These findings suggest that the number of ISRDs should be increased.
