Maize ( Zea mays ) is a major staple crop in Africa, where its yield and the livelihood of millions are compromised by the parasitic witchweed Striga . Germination of Striga is induced by strigolactones exuded from maize roots into the rhizosphere. In a maize germplasm collection, we identified two strigolactones, zealactol and zealactonoic acid, which stimulate less Striga germination than the major maize strigolactone, zealactone. We then showed that a single cytochrome P450, ZmCYP706C37, catalyzes a series of oxidative steps in the maize-strigolactone biosynthetic pathway. Reduction in activity of this enzyme and two others involved in the pathway, ZmMAX1b and ZmCLAMT1, can change strigolactone composition and reduce Striga germination and infection. These results offer prospects for breeding Striga -resistant maize.

Climate change is affecting how energy and matter flow through ecosystems, thereby altering global carbon and nutrient cycles. Microorganisms play a fundamental role in carbon and nutrient cycling and are thus an integral link between ecosystems and climate. Here, we highlight a major black box hindering our ability to anticipate ecosystem climate responses: viral infections within complex microbial food webs. We show how understanding and predicting ecosystem responses to warming could be challenging—if not impossible—without accounting for the direct and indirect effects of viral infections on different microbes (bacteria, archaea, fungi, protists) that together perform diverse ecosystem functions. Importantly, understanding how rising temperatures associated with climate change influence viruses and virus-host dynamics is crucial to this task, yet is severely understudied. In this perspective, we (i) synthesize existing knowledge about virus-microbe-temperature interactions and (ii) identify important gaps to guide future investigations regarding how climate change might alter microbial food web effects on ecosystem functioning. To provide real-world context, we consider how these processes may operate in peatlands—globally significant carbon sinks that are threatened by climate change. We stress that understanding how warming affects biogeochemical cycles in any ecosystem hinges on disentangling complex interactions and temperature responses within microbial food webs.

ObjectiveThe causality and pathogenic mechanism of microbiome composition remain elusive in many diseases, including autoimmune diseases such as rheumatoid arthritis (RA). This study aimed to elucidate gut microbiome’s role in RA pathology by a comprehensive metagenome-wide association study (MWAS).MethodsWe conducted MWAS of the RA gut microbiome in the Japanese population (ncase=82, ncontrol=42) by using whole-genome shotgun sequencing of high depth (average 13 Gb per sample). Our MWAS consisted of three major bioinformatic analytic pipelines (phylogenetic analysis, functional gene analysis and pathway analysis).ResultsPhylogenetic case–control association tests showed high abundance of multiple species belonging to the genus Prevotella (e.g., Prevotella denticola) in the RA case metagenome. The non-linear machine learning method efficiently deconvoluted the case–control phylogenetic discrepancy. Gene functional assessments showed that the abundance of one redox reaction-related gene (R6FCZ7) was significantly decreased in the RA metagenome compared with controls. A variety of biological pathways including those related to metabolism (e.g., fatty acid biosynthesis and glycosaminoglycan degradation) were enriched in the case–control comparison. A population-specific link between the metagenome and host genome was identified by comparing biological pathway enrichment between the RA metagenome and the RA genome-wide association study results. No apparent discrepancy in alpha or beta diversities of metagenome was found between RA cases and controls.ConclusionOur shotgun sequencing-based MWAS highlights a novel link among the gut microbiome, host genome and pathology of RA, which contributes to our understanding of the microbiome’s role in RA aetiology.

When monitoring phenology at ground level, it would be more important to continue observations in long terms and to detect the timing of various phenological events such as leafing, flowering and autumn senescence. In this study, to develop the methodology for plant phenology monitoring by using digital camera, we examined how multiple image indices, which are derived from multi-temporal visible images, respond to the changes of colors of leaves and flowers for several target species of plants, and tried to detect various phenology events by tracing time series changes of the coordinate in the feature spaces of two indices. As a result, we found out that it was possible to understand the characteristics of the phenological events for different species from each image index. Also, it was identified that the utility of combination with two indices would be effective to detect the timing of phenology events in the feature space of two indices. In the actual phenology monitoring, it would be effective to use a single index for understanding the seasonal characteristics and to use the combination of two indices for detection of the timing of phenology events by tracing the time series changes in the feature space.

MicroRNAs (miRNAs) modulate the post-transcriptional regulation of target genes and are related to biology of complex human traits, but genetic landscape of miRNAs remains largely unknown. Given the strikingly tissue-specific miRNA expression profiles, we here expand a previous method to quantitatively evaluate enrichment of genome-wide association study (GWAS) signals on miRNA–target gene networks (MIGWAS) to further estimate tissue-specific enrichment. Our approach integrates tissue-specific expression profiles of miRNAs (∼1800 miRNAs in 179 cells) with GWAS to test whether polygenic signals enrich in miRNA–target gene networks and whether they fall within specific tissues. We applied MIGWAS to 49 GWASs (nTotal = 3 520 246), and successfully identified biologically relevant tissues. Further, MIGWAS could point miRNAs as candidate biomarkers of the trait. As an illustrative example, we performed differentially expressed miRNA analysis between rheumatoid arthritis (RA) patients and healthy controls (n = 63). We identified novel biomarker miRNAs (e.g. hsa-miR-762) by integrating differentially expressed miRNAs with MIGWAS results for RA, as well as novel associated loci with significant genetic risk (rs56656810 at MIR762 at 16q11; n = 91 482, P = 3.6 × 10−8). Our result highlighted that miRNA–target gene network contributes to human disease genetics in a cell type-specific manner, which could yield an efficient screening of miRNAs as promising biomarkers.

Photosynthetic photon flux density (PPFD: µmol m−2 s−1) is indispensable for plant physiology processes in photosynthesis. However, PPFD is seldom measured, so that PPFD has been estimated by using solar radiation (SR: W m−2) measured in world wide. In method using SR, there are two steps: first to estimate photosynthetically active radiation (PAR: W m−2) by the fraction of PAR to SR (PF) and second: to convert PAR to PPFD using the ratio of quanta to energy (Q / E: µmol J−1). PF and Q/E usually have been used as the constant values, however, recent studies point out that PF and Q / E would not be constants under various sky conditions. In this study, we use the numeric data of sky-conditions factors such cloud cover, sun appearance/hiding and relative sky brightness derived from whole-sky image processing and examine the influences of sky-conditions factors on PF and Q / E of global and diffuse PAR. Furthermore, we discuss our results by comparing with the existing methods.

Observation of sea ice thickness is one of key issues to understand regional effect of global warming. One of approaches to monitor sea ice in large area is microwave remote sensing data analysis. However, ground truth must be necessary to discuss the effectivity of this kind of approach. The conventional method to acquire ground truth of ice thickness is drilling ice layer and directly measuring the thickness by a ruler. However, this method is destructive, time-consuming and limited spatial resolution. Although there are several methods to acquire ice thickness in non-destructive way, ground penetrating radar (GPR) can be effective solution because it can discriminate snow-ice and ice-sea water interface. In this paper, we carried out GPR measurement in Lake Saroma for relatively large area (200 m by 300 m, approximately) aiming to obtain grand truth for remote sensing data. GPR survey was conducted at 5 locations in the area. The direct measurement was also conducted simultaneously in order to calibrate GPR data for thickness estimation and to validate the result. Although GPR Bscan image obtained from 600MHz contains the reflection which may come from a structure under snow, the origin of the reflection is not obvious. Therefore, further analysis and interpretation of the GPR image, such as numerical simulation, additional signal processing and use of 200 MHz antenna, are required to move on thickness estimation.

Observation of sea ice thickness by remote sensing is one of key issues to understand an effect of global warming. However, ground truth must be necessary to discuss this kind of approach. Although there are several methods to acquire ice thickness, Ground Penetrating Radar (GPR) can be good solution because it can discriminate snow-ice and ice-sea water interface thanks to comparative higher spatial resolution than the other methods. In this paper, we carried out GPR measurement in brackish lake and an electromagnetic field analysis in order to interpret the GPR data. The simulation model was assumed considering the actual snow and ice thickness acquired in field measurement. From the simulation results, although it seems difficult to identify the reflection at snow and ice interface due to a thin layer thickness and a low dielectric constant, snow and ice thickness may be estimated by using multiple reflection components.

Solar radiation is a source of energy for ground surfaces. Clouds control energy budget on both local and global scales. We propose the simple ground-based observation method for various sky conditions instead of the existing visual observation. In the paper, we mainly describe how to discriminate clouds area on whole sky images, how to overlay the cloud areas to the satellite-based images and the test experiment. As for the discrimination of sky conditions, we use Sky Index (SI) and Brightness Index (BI) calculated from whole sky imageries. SI shows the extent of the blueness and grayscale and BI indicates the extent of the brightness. Sun, cloud and blue sky areas are divided by SI and BI threshold. As an extension of ground-based observation to satellite-based observation, clouds area discriminated by ground-based observation overlay to satellite images. Through this experiment, the utilities of ground-based cloud observation are discussed. The proposed ground-based cloud observation method and its satellite-based extension should be connected to satellite products with high quality by implementation of sky and clouds observation covering a wide area.

OBJECTIVES: A large-scale survey of doses to patients undergoing the most frequent radiological examinations was carried out in health services in São Paulo (347 radiological exami nations per 1 000 inhabitants), the most populous Brazilian state. METHODS: A postal dosimetric kit with thermoluminescence dosimeters was used to evalu ate the entrance surface dose (ESD) to patients. A stratified sampling technique applied to the national health database furnished important data on the distribution of equipment and the an nual number of examinations. Chest, head (skull and sinus), and spine (cervical, thoracic, and lumbar) examinations were included in the trial. A total of 83 rooms and 868 patients were in cluded, and 1 415 values of ESD were measured. RESULTS: The data show large coefficients of variation in tube charge, giving rise to large variations in ESD values. Also, a series of high ESD values associated with unnecessary lo calizing fluoroscopy were detected. Diagnostic reference levels were determined, based on the 75th percentile (third quartile) of the ESD distributions. For adult patients, the diagnostic ref erence levels achieved are very similar to those obtained in international surveys. However, the situation is different for pediatric patients: the ESD values found in this survey are twice as large as the international recommendations for chest radiographs of children. CONCLUSIONS: Despite the reduced number of ESD values and rooms for the pediatric pa tient group, it is recommended that practices in chest examinations be revised and that specific national reference doses and image quality be established after a broader survey is carried out.OBJETIVOS: Estudio a gran escala de las dosis aplicadas a pacientes sometidos a los exámenes radiológicos más frecuentes realizados en los servicios de salud de São Paulo (347 exámenes radiológicos por 1 000 habitantes), el estado más poblado de Brasil. MÉTODOS: Se empleó un sistema dosimétrico postal con dosímetros termoluminiscentes para evaluar la dosis en la superficie de entrada (DSE) de los pacientes. Se realizó un muestreo estratificado de la base de datos nacional de salud que aportó información importante sobre la distribución del equipamiento y el número de pruebas realizadas anualmente. El estudio abarcó los exámenes de tórax, cabeza (cráneo y senos paranasales) y columna vertebral (cervical, dorsal y lumbar). Se incluyeron 83 instalaciones y 868 pacientes; se obtuvieron 1 415 mediciones de DSE. RESULTADOS: Los resultados mostraron grandes coeficientes de variación en la carga del tubo, lo que originó variaciones considerables en las DSE. Además, se detectaron valores elevados de DSE asociados con fluoroscopias de localización innecesarias. Los niveles de referencia diagnósticos se determinaron a partir del percentil 75 (tres cuartiles) de las distribuciones de las DSE. Para pacientes adultos, los niveles de referencia diagnósticos alcanzados fueron muy similares a los obtenidos en estudios internacionales. No obstante, la situación fue diferente en los pacientes pediátricos: los valores de DSE obtenidos en este estudio fueron dos veces mayores que los recomendados internacionalmente para radiografías de tórax en niños. CONCLUSIONES: A pesar del reducido número de valores de DSE e instalaciones en el grupo de pacientes pediátricos, se recomienda revisar la práctica de estudios radiológicos de tórax y establecer referencias nacionales específicas para las dosis y la calidad de las imágenes, después de realizar un estudio más amplio.

For nearly 40 years, there was no generally accepted staging system for malignant pleural mesothelioma. In 1994, members of the International Mesothelioma Interest Group, in collaboration with the International Association for the Study of Lung Cancer, proposed a TNM staging system based on analyses of outcomes in retrospective surgical series and small clinical trials. Subsequently accepted by the American Joint Commission on Cancer and the Union for International Cancer Control for the sixth editions of their staging manuals, this system has since been the international staging standard. However, it has significant limitations, particularly with respect to clinical staging and to the categories for lymph node staging. Here we provide an overview of the development of the International Association for the Study of Lung Cancer malignant pleural mesothelioma staging database, which was designed to address these limitations through the development of a large international data set. Introduction: The current T component for malignant pleural mesothelioma (MPM) has been predominantly informed by surgical data sets and consensus. The International Association for the Study of Lung Cancer undertook revision of the seventh edition of the staging system for MPM with the goal of developing recommendations for the eighth edition. Methods: Data elements including detailed T descriptors were developed by consensus. Tumor thickness at three pleural levels was also recorded. An electronic data capture system was established to facilitate data submission. Results: A total of 3519 cases were submitted to the database. Of those eligible for T-component analysis, 509 cases had only clinical staging, 836 cases had only surgical staging, and 642 cases had both available. Survival was examined for T categories according to the current seventh edition staging system. There was clear separation between all clinically staged categories except T1a versus T1b (hazard ratio = 0.99, p = 0.95) and T3 versus T4 (hazard ratio = 1.22, p = 0.09), although the numbers of T4 cases were small. Pathological staging failed to demonstrate a survival difference between adjacent categories with the exception of T3 versus T4. Performance improved with collapse of T1a and T1b into a single T1 category; no current descriptors were shifted or eliminated. Tumor thickness and nodular or rindlike morphology were significantly associated with survival. Conclusions: A recommendation to collapse both clinical and pathological T1a and T1b into a T1 classification will be made for the eighth edition staging system. Simple measurement of pleural thickness has prognostic significance and should be examined further with a view to incorporation into future staging. The structures of radial electric field and transport at the magnetic island are investigated using n/m=1/1 external perturbation coils in the Large Helical Device (LHD). The radial profiles of plasma potential, as well as the electron temperature and density, shows flattening inside the magnetic island and the large shear of the radial electric field is observed at the boundary of the magnetic island. When the current of n/m=1/1 external perturbation coils becomes large enough, the finite radial electric field appears inside the magnetic island. The abrupt appearance of plasma flow inside the magnetic island suggests the non-linearity of the viscous force at the boundary of the magnetic island. The thermal diffusivity perpendicular to the magnetic filed inside the magnetic island is estimated with the cold pulse propagation, which is produced by a tracer-encapsulated solid pellet (TESPEL). The time delay and amplitude of the electron temperature of the cold pulse show much lower thermal diffusivity inside the magnetic island (0.16 m2ls; than that outside the magnetic island (2 m2ls;, which is a clear evidence for the significant reduction of heat transport inside the magnetic island.

Stomach contents or foregut content samples of the most abundant species were collected from 26 species (lO families). The fish assemblage occupied the bottom stream. The similarity analysis of spalía! occupation of species grouped four habitat use guilds: A) "Iambaris" (Aslyanax sp. and Deutetodon langei), Characidium spp. (C. lane; and C. plerosliclum) and Rineloricaria kronei used the bottom in deep sites and waters with middle current; B) Pimelodella pappenheimi and Corydoras barbalus used the bottom in siles with JOwer cUI'l"ént; C) Mimagoniates microlepis used the surface of the water column; and D) Phalloceros caudimacullUus used shallow sítes and waters without current. Species with few records were analysed descriptively. Diet similarity suggested seven trophic guilds: Microglanis sp. and PiTMIodella pappenheimi: omnivorouslca:mivorous gui14; Corydoras barbatus: omnivorous/insectivorous guild; Characidium [ane;: aquatic insectivorous guild, Ilillinly aquatic insects; Mimagoniates microlepis: terrestriál insectivorous guild, mainly te.rrestrial insects; Deuterodon [angei and ASlyanax sp.: omnivorous/herbivorous guild; Rineloricaria kronei, Kronichth.yssubteres, Schizolecis guntheri. We describe a very rare case of anomalous origin of the anterior choroidal artery. In our case the anterior choroidal artery arises from the internal carotid artery proximal to the posterior communicating artery.

The fundamental problem of how tunneling in thermal medium is completed is addressed, and a new time scale of order 1/friction for its termination, which is usually much shorter than the Hubble time, is pointed out. Enhanced non-linear resonance is responsible for this short time scale. Time evolution of the decay process of unstable particles is investigated in field theory models. We first formulate how to renormalize the non-decay amplitude beyond perturbation theory and then discuss short-time behavior of very long-lived particles. Two different formalisms, one that does and one that does not, assume existence of the asymptotic field of unstable particles are considered. The non-decay amplitude is then calculated by introducing a finite time resolution of measurement, which makes it possible to discuss both renormalizable and non-renormalizable decay interaction including the nucleon decay. In ordinary circumstances the onset of the exponential decay law starts at times as early as at roughly the resolution time, but with an enhanced amplitude which may be measurable. It is confirmed that the short-time formula $1 - \Gamma t$ of the exponential decay law may be used to set limits on the nucleon decay rate in underground experiments. 1. The effects of the volatile anaesthetics, halothane, isoflurane and enflurane, on rat hippocampal CA1 and CA3 neurones in in vitro preparations were studied by intracellular recording methods. 2. The three anaesthetics, at concentrations similar to those used clinically (0.2-1.2 mM), initially increased and then subsequently decreased the spontaneous firing of CA1 neurones without affecting the resting membrane properties or the EPSPs evoked by focal stimuli. 3. The anaesthetics at these concentrations depressed both the fast after-hyperpolarization of the soma spike and the post-tetanic hyperpolarization induced by repetitive stimulation. They also decreased the IPSPs evoked by focal stimuli. 4. The threshold for spike generation was gradually elevated by as much as 4-6 mV during application of the anaesthetics at these concentrations. The subthreshold potential oscillations (which are likely to be associated with periodic alterations in non-inactivating Ca2+ and Na+ currents) were enhanced in the low concentrations (0.2-0.5 mM), but were depressed in the high concentrations (0.8-1.2 mM). 5. The results suggest that the transient increase in the firing frequency was caused by a depression of both the spike after-hyperpolarization and the post-tetanic hyperpolarization, and that the reduction of spontaneous firing was mainly due to an elevated threshold for spike generation. 6. The three anaesthetics altered the pattern of spontaneous spike-firing in CA3 neurones from solitary spiking to burst firing without affecting the resting membrane properties. 7. The effects of the anaesthetics on the active membrane properties and the postsynaptic potentials in CA3 neurones were similar to the effects in CA1 neurones. 8. In the majority of CA3 neurones, soma spikes elicited by depolarizing current pulses were followed by a Ca2+-dependent after-depolarization, which was in turn followed by a prolonged after-hyperpolarization (post-burst hyperpolarization). The anaesthetics facilitated the after-depolarizing potential, while they depressed the post-burst hyperpolarization. Combination of the two effects would give rise to the highly stereotyped burst (about 1 Hz in frequency) in the presence of the volatile anaesthetics.
