Abstract The KM3NeT/ARCA neutrino detector is currently under construction at 3500 m depth offshore Capo Passero, Sicily, in the Mediterranean Sea. The main science objectives are the detection of high-energy cosmic neutrinos and the discovery of their sources. Simulations were conducted for the full KM3NeT/ARCA detector, instrumenting a volume of 1 km $$^3$$ 3 , to estimate the sensitivity and discovery potential to point-like neutrino sources. This paper covers the reconstruction of track- and shower-like signatures, as well as the criteria employed for neutrino event selection. With an angular resolution below 0.1 $$^\circ $$ ∘ for tracks and under 2 $$^\circ $$ ∘ for showers, the sensitivity to point-like neutrino sources surpasses existing observed limits across the entire sky.

Climate warming and land-use change are reshuffling the distribution of wild organisms on a global scale. Some species may expand their ranges and colonize new regions, which may greatly affect ecological interactions among pre-existing species and colonizers. In the last decades, such processes have originated a unique condition of sympatry among three Eurasian small Falco species (common kestrel F. tinnunculus, lesser kestrel F. naumanni, red-footed falcon F. vespertinus) in the intensively cultivated farmland habitats of the Po Plain (Northern Italy). This provides an excellent opportunity to investigate patterns of spatial niche overlap during the initial phases of the establishment of sympatry. To investigate spatial niche overlap of the three falcon species, we relied on Environmental Niche Models (ENMs) based on widespread breeding occurrence data obtained through field surveys and citizen science programs (during the 2018–2020 period). ENMs were based on bioclimatic and land-use variables in an ensemble modelling framework. We estimated species-specific relative contributions of each climatic and land-use variable and its response curves effect. Eventually, we generated spatial correlation maps of the potential species’ distributions to derive spatially-explicit predictions of potential co-occurrence areas among the three species. Overall, eco-climatic determinants of the distribution of lesser kestrel and red-footed falcon were similar, resulting in a strong association with intensive arable lands and dry continental climate. Consistently, we found a high spatial correlation between the suitability maps of the two species, with highly suitable areas located in the Central-Eastern area of the Po Plain, corresponding to the core range of both species. Conversely, the common kestrel emerged as a habitat generalist and was widely distributed throughout the Po Plain. Our findings suggest that the recent sympatry between lesser kestrels and red-footed falcons in the Po Plain may promote ecological interactions and intra-guild competition.

Summary: Background: Integrase inhibitor (INSTI) with boosted darunavir (DRV/r), a regimen with a high-resistance barrier, avoiding NRTI toxicities, might be a switching option in children living with HIV (CLWHIV). Methods: SMILE is a randomised non-inferiority trial evaluating safety and antiviral efficacy of once-daily INSTI + DRV/r vs. continuing on current standard-of-care (SOC) triple ART (2NRTI + boosted PI/NNRTI) in virologically-suppressed CLWHIV aged 6–18 years. The primary outcome is the proportion with confirmed HIV-RNA ≥50 copies/mL by week 48, estimated by Kaplan–Meier method. Non-inferiority margin was 10%. Registration number for SMILE are: ISRCTN11193709, NCT #: NCT02383108. Findings: Between 10th June 2016 and 30th August 2019, 318 participants were enrolled from Africa 53%, Europe 24%, Thailand 15% and Latin America 8%, 158 INSTI + DRV/r [153 Dolutegravir (DTG); 5 Elvitegravir (EVG)], 160 SOC. Median (range) age was 14.7 years (7.6–18.0); CD4 count 782 cells/mm3 (227–1647); 61% female. Median follow-up was 64.3 weeks with no loss to follow-up. By 48 weeks, 8 INSTI + DRV/r vs. 12 SOC had confirmed HIV-RNA ≥50 copies/mL; difference (INSTI + DRV/r-SOC) −2.5% (95% CI: −7.6, 2.5%), showing non-inferiority. No major PI or INSTI resistance mutations were observed. There were no differences in safety between arms. By week 48, difference (INSTI + DRV/r-SOC) in mean CD4 count change from baseline was −48.3 cells/mm3 (95% CI: −93.4, −3.2; p = 0.036). Difference (INSTI + DRV/r-SOC) in mean HDL change from baseline was −4.1 mg/dL (95% CI: −6.7, −1.4; p = 0.003). Abstract The next generation of water Cherenkov neutrino telescopes in the Mediterranean Sea are under construction offshore France (KM3NeT/ORCA) and Sicily (KM3NeT/ARCA). The KM3NeT/ORCA detector features an energy detection threshold which allows to collect atmospheric neutrinos to study flavour oscillation. This paper reports the KM3NeT/ORCA sensitivity to this phenomenon. The event reconstruction, selection and classification are described. The sensitivity to determine the neutrino mass ordering was evaluated and found to be 4.4 $$\sigma $$ σ if the true ordering is normal and 2.3 $$\sigma $$ σ if inverted, after 3 years of data taking. The precision to measure $$\varDelta m^2_{32}$$ Δ m 32 2 and $$\theta _{23}$$ θ 23 were also estimated and found to be $$85 . 10^{-6}~{\mathrm{eV}^{2}}$$ 85 . 10 - 6 eV 2 and $$(^{+1.9}_{-3.1})^{\circ }$$ ( - 3.1 + 1.9 ) ∘ for normal neutrino mass ordering and, $$75 . 10^{-6}~{\mathrm{eV}^{2}}$$ 75 . 10 - 6 eV 2 and $$(^{+2.0}_{-7.0})^{\circ }$$ ( - 7.0 + 2.0 ) ∘ for inverted ordering. Finally, a unitarity test of the leptonic mixing matrix by measuring the rate of tau neutrinos is described. Three years of data taking were found to be sufficient to exclude event rate variations larger than 20% at $$3\sigma $$ 3 σ level.

Abstract Imaging using scintillators is a widespread and cost-effective approach in radiography. While different types of scintillator and sensor configurations exist, it can be stated that the detection efficiency and resolution of a scintillator-based system strongly depend on the scintillator material and its thickness. Recently developed event-driven detectors are capable of registering spots of light emitted by the scintillator after a particle interaction, allowing to reconstruct the Center-of-Mass of the interaction within the scintillator. This results in a more precise location of the event and therefore provides a pathway to overcome the scintillator thickness limitation and increase the effective spatial resolution of the system. Utilizing this principle, we present a detector capable of Time-of-Flight imaging with an adjustable field-of-view, ad-hoc binning and re-binning of data based on the requirements of the experiment including the possibility of particle discrimination via the analysis of the event shape in space and time. It is considered that this novel concept might replace regular cameras in neutron imaging detectors as it provides superior detection capabilities with the most recent results providing an increase by a factor 3 in image resolution and an increase by up to a factor of 7.5 in signal-to-noise for thermal neutron imaging.

Abstract The KM3NeT research infrastructure is under construction in the Mediterranean Sea. It consists of two water Cherenkov neutrino detectors, ARCA and ORCA, aimed at neutrino astrophysics and oscillation research, respectively. Instrumenting a large volume of sea water with $$\sim {6200}$$ ∼ 6200 optical modules comprising a total of $$\sim {200{,}000}$$ ∼ 200 , 000 photomultiplier tubes, KM3NeT will achieve sensitivity to $$\sim {10} \ \mathrm{MeV}$$ ∼ 10 MeV neutrinos from Galactic and near-Galactic core-collapse supernovae through the observation of coincident hits in photomultipliers above the background. In this paper, the sensitivity of KM3NeT to a supernova explosion is estimated from detailed analyses of background data from the first KM3NeT detection units and simulations of the neutrino signal. The KM3NeT observational horizon (for a $$5\,\sigma $$ 5 σ discovery) covers essentially the Milky-Way and for the most optimistic model, extends to the Small Magellanic Cloud ( $$\sim {60} \ \mathrm{kpc}$$ ∼ 60 kpc ). Detailed studies of the time profile of the neutrino signal allow assessment of the KM3NeT capability to determine the arrival time of the neutrino burst with a few milliseconds precision for sources up to 5–8 kpc away, and detecting the peculiar signature of the standing accretion shock instability if the core-collapse supernova explosion happens closer than 3–5 kpc, depending on the progenitor mass. KM3NeT’s capability to measure the neutrino flux spectral parameters is also presented.

The emergent excitation dynamics of an ultracold gas of Rydberg atoms exhibits features analogous to epidemic spreading on networks. Wintermantel et al. propose a controllable experimental system for studying network dynamics at the interface of mathematical models and real-world complex systems.

Abstract Background Endoscopic management of low risk upper tract urothelial carcinoma (UTUC) may be considered in select clinical scenarios, which allows sparing the morbidity of radical nephroureterectomy while achieving acceptable oncological outcomes and preservation of kidney function. Herein, we present a case with UTUC in a solitary kidney managed with 532 nm laser vaporization through a percutaneous approach. Case presentation The patient in this video (Additional file 1) is an 85-year-old woman who presented with a bulky tumor in the collecting system of a congenital solitary left kidney, which was a biopsy proven low grade urothelial carcinoma. Prior to the procedure, a lower pole percutaneous nephrostomy tube was successfully placed under sedation by Interventional Radiology. The procedure was done in a prone split leg position. The mass, which was predominantly localized to the renal pelvis was efficiently vaporized with the 532 nm laser in a systematic manner with continuous irrigation of normal saline through the cystoscope. The patient was discharged home on postoperative day 2 with the nephroureterostomy catheter open to drainage. This catheter was subsequently clamped and removed two weeks later without complications. Follow up uretroscopy showed excellent treatment response and the patient remains well without complications. Conclusion This case report details the potential utility of 532 nm laser vaporization of UTUC, however, ongoing studies are required to demonstrate peri-operative safety and durable oncologic efficacy.

Abstract KM3NeT is a research infrastructure located in the Mediterranean Sea, that will consist of two deep-sea Cherenkov neutrino detectors. With one detector (ARCA), the KM3NeT Collaboration aims at identifying and studying TeV–PeV astrophysical neutrino sources. With the other detector (ORCA), the neutrino mass ordering will be determined by studying GeV-scale atmospheric neutrino oscillations. The first KM3NeT detection units were deployed at the Italian and French sites between 2015 and 2017. In this paper, a description of the detector is presented, together with a summary of the procedures used to calibrate the detector in-situ. Finally, the measurement of the atmospheric muon flux between 2232–3386 m seawater depth is obtained.

Summary: The NHGRI Genomic Data Science Analysis, Visualization, and Informatics Lab-space (AnVIL; https://anvilproject.org) was developed to address a widespread community need for a unified computing environment for genomics data storage, management, and analysis. In this perspective, we present AnVIL, describe its ecosystem and interoperability with other platforms, and highlight how this platform and associated initiatives contribute to improved genomic data sharing efforts. The AnVIL is a federated cloud platform designed to manage and store genomics and related data, enable population-scale analysis, and facilitate collaboration through the sharing of data, code, and analysis results. By inverting the traditional model of data sharing, the AnVIL eliminates the need for data movement while also adding security measures for active threat detection and monitoring and provides scalable, shared computing resources for any researcher. We describe the core data management and analysis components of the AnVIL, which currently consists of Terra, Gen3, Galaxy, RStudio/Bioconductor, Dockstore, and Jupyter, and describe several flagship genomics datasets available within the AnVIL. We continue to extend and innovate the AnVIL ecosystem by implementing new capabilities, including mechanisms for interoperability and responsible data sharing, while streamlining access management. The AnVIL opens many new opportunities for analysis, collaboration, and data sharing that are needed to drive research and to make discoveries through the joint analysis of hundreds of thousands to millions of genomes along with associated clinical and molecular data types.

Immunization polices in Italy has recently reached important milestones, including the approval of the National Immunization Prevention Plan and of a new law mandatory immunization; this stimulating a lively debate at the scientific, political and societal-level, reflected on the media. We applied a model previously published to quantitatively and qualitatively assess media coverage on vaccines and immunization-related topics on the most read Italian newspaper, “Corriere della Sera”, over an 11-year study period (2007-2017). We retrieved relevant key words and articles, reported on included articles’ topic, position, approach to immunization and on other selected indicators’ summary statistics, temporal trends and correspondence with key epidemiological and policy events. Over the study period the quote “vaccin*” was mentioned on average 325 times per year; with an increase of 150% after the approval of the new law on mandatory immunization in 2017. In the same year, on average, two first-page articles per week were published on the topic. We report a clear association between key events (i.e the H1N1 influenza pandemic, the “Fluad case”, the approval of the new law on mandatory immunization) and their media coverage. Overall, 84% of articles had a positive attitude towards immunization, this share decreasing to 79% when only considering articles published after the approval of the law on mandatory vaccination. Media play a crucial role in channelling health-related information and significantly influence health behaviours. We urge public institutions, health authorities and the scientific community not to underestimate the opportunity to monitor media coverage on key healthcare topics and to convey evidence-based health education messages through the media.

Abstract Recurrent deletions of the CDKN2A/ARF/CDKN2B genes encoded at chromosome 9p21 have been described in both pediatric and adult acute lymphoblastic leukemia (ALL), but their prognostic value remains controversial, with limited data on adult T-ALL. Here, we investigated the presence of homozygous and heterozygous deletions of the CDKN2A/ARF and CDKN2B genes in 64 adult T-ALL patients enrolled in two consecutive trials from the Spanish PETHEMA group. Alterations in CDKN2A/ARF/CDKN2B were detected in 35/64 patients (55%). Most of them consisted of 9p21 losses involving homozygous deletions of the CDKNA/ARF gene (26/64), as confirmed by single nucleotide polymorphism (SNP) arrays and interphase fluorescence in situ hybridization (iFISH). Deletions involving the CDKN2A/ARF/CDKN2B locus correlated with a higher frequency of cortical T cell phenotype and a better clearance of minimal residual disease (MRD) after induction therapy. Moreover, the combination of an altered copy-number-value (CNV) involving the CDKN2A/ARF/CDKN2B gene locus and undetectable MRD (≤ 0.01%) values allowed the identification of a subset of T-ALL with better overall survival in the absence of hematopoietic stem cell transplantation.

Abstract Studying atmospheric neutrino oscillations in the few-GeV range with a multi-megaton detector promises to determine the neutrino mass hierarchy. This is the main science goal pursued by the future KM3NeT/ORCA water Cherenkov detector in the Mediterranean Sea. In this paper, the processes that limit the obtainable resolution in both energy and direction in charged-current neutrino events in the ORCA detector are investigated. These processes include the composition of the hadronic fragmentation products, the subsequent particle propagation and the photon-sampling fraction of the detector. GEANT simulations of neutrino interactions in seawater produced by GENIE are used to study the effects in the 1-20 GeV range. It is found that fluctuations in the hadronic cascade in conjunction with the variation of the inelasticity y are most detrimental to the resolutions. The effect of limited photon sampling in the detector is of significantly less importance. These results will therefore also be applicable to similar detectors/media, such as those in ice.

Blood serum proteins are significant indicators of animal health. Nevertheless, several factors should be considered to appropriately interpret their concentrations in blood. Therefore, the objectives of this study were (1) to assess the effect of herd productivity, breed, age and stage of lactation on serum proteins and (2) to investigate association between serum proteins and somatic cell count (SCC) in dairy cattle. Milk and blood samples were collected from 1508 cows of six different breeds (Holstein Friesian, Brown Swiss, Jersey, Simmental, Rendena and Alpine Grey) that were housed in 41 multi-breed herds. Milk samples were analyzed for composition and SCC, while blood samples were analyzed for serum proteins (i.e. total protein, albumin, globulin and albumin-to-globulin ratio (A : G)). Herds were classified as low or high production, according to the cow’s average daily milk energy yield adjusted for breed, days in milk (DIM) and parity. Data were analyzed using a linear mixed model that included the fixed effects of DIM, parity, SCS, breed, herd productivity and the random effect of the Herd-test date within productivity level. Cows in high producing herds (characterized also by greater use of concentrates in the diet) had greater serum albumin concentrations. Breed differences were reported for all traits, highlighting a possible genetic mechanism. The specialized breed Jersey and the two dual-purpose local breeds (Alpine Grey and Rendena) had the lowest globulin concentration and greatest A : G. Changes in serum proteins were observed through lactation. Total protein reached the highest concentration during the 4th month of lactation. Blood albumin increased with DIM following a quadratic pattern, while globulin decreased linearly. As a consequence, A : G increased linearly during lactation. Older cows had greater total protein and globulin concentrations, while albumin concentration seemed to be not particularly affected by age. A linear relationship between serum proteins and SCS was observed. High milk SCS was associated with greater total protein and globulin concentrations in blood. The rise in globulin concentration, together with a decrease in albumin concentrations, resulted in a decline in A : G as SCS of milk increased. In conclusion, such non-genetic factors must be considered to appropriately interpret serum proteins as potential animal welfare indicator and their evaluation represents an important first-step for future analysis based on the integration of metabolomics, genetic and genomic information for improving the robustness of dairy cows.

Thirty-five multiparous Holstein dairy cows were selected from a high-producing dairy farm in northeastern Italy: 16 in second lactation (L2), 10 in third lactation (L3) and 9 in fourth lactation (L4). Blood sampling was carried out 7 ± 5 days before calving (Pre/C) and 7 ± 5 days after calving (Post/C). Serum triiodothyronine (T3), thyroxine (T4) and a thyroid-stimulating hormone (TSH) were assessed. Two-way analysis of variance (ANOVA) showed statistically significant effects of class (L2, L3, L4) and of period (Pre/C, Post/C) on the parameters studied (T3, T4, TSH). In particular, Bonferroni's multiple comparison test showed lower values in post-calving than in the pre-calving in L2 and L3 for TSH; lower values in post-calving than in the pre-calving in L2 for T3; and lower values in post-calving than in the pre-calving in L2, L3 and L4 for T4. Our results improve the knowledge of endocrine and metabolic changes occurring in dairy cows during transition periods and may be useful to supply a new strategy for the improvement of dairy cow farm management and reproductive performance.

A study using 20 Holstein Friesian cows was conducted to investigate the influence of calf gender on metabolism during the transition period in dairy cattle. Blood samples were collected at three time points: 2–4 days prepartum (time 1), and 1 week and 2 weeks postpartum (time 2 and time 3 respectively). Serum samples obtained were analysed for total proteins, albumin, urea, glucose, non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHB). Depending on the sex of the offspring, cows were divided into two groups: Group 1 consisted of cows with heifer offspring (n = 12) and Group 2 consisted of cows with bull offspring (n = 8). A two-way repeated measures ANOVA and t tests for unpaired data were used to analyse the pattern of studied parameters and differences between the two groups. The results indicate differences in metabolic parameters between the two groups. These results highlight the importance of considering fetal sex as a factor that influences maternal metabolism during the early transition period in dairy cows.

The Metafor project has developed a common information model (CIM) using the ISO19100 series formalism to describe numerical experiments carried out by the Earth system modelling community, the models they use, and the simulations that result. Here we describe the mechanism by which the CIM was developed, and its key properties. We introduce the conceptual and application versions and the controlled vocabularies developed in the context of supporting the fifth Coupled Model Intercomparison Project (CMIP5). We describe how the CIM has been used in experiments to describe model coupling properties and describe the near term expected evolution of the CIM.

The aim of this study was to evaluate the variations of Acute Phase Proteins (APPs) and other blood constituents during the onset of the sub-acute ruminal acidosis (SARA) pathological status. A total of 108 cows from 12 dairy herds were randomly selected and divided into three Groups of 36 animals each. All animals were subjected to a rumenocentesis. Group A was composed by subjects with a rumen pH>5.8, Group B was composed by subjects with a rumen pH ≤5.5≤5.8 and Group C was composed by subjects with a rumen pH

The circadian rhythm of body core and surface temperature in 10 Comisana sheep kept under a natural photoperiod (06:30/19:00) was evaluated. Every 3 h for 24 consecutive hours rectal temperature (RT) and foot temperature were recorded. Particularly, foot temperature was recorded by infrared thermography, an ideal technique for evaluating the temperature not only at one point but also at the eight points as follows: in the right front of the interdigital area (FA1), in the left front of the interdigital area (FA2), in the right rear of the interdigital area (RA1), in the left rear of the interdigital area (RA2), in the right front of the interdigital line (FL1), in the left front of the interdigital line (FL2), in the right rear of the interdigital line (RL1) and in the left rear of the interdigital line (RL2). Two-way repeated measures ANOVA using SPSS, followed by Scheffé's test, showed a significant effect of the time of day and side of temperature collection (P < 0.05) on temperature values. The single cosinor procedure showed a daily rhythmicity of RT and foot temperature in all tested sides. The circadian oscillations of RT reflected the familiar circadian patterns of endogenous sources and the results of foot temperatures supported the idea that the daily rhythm was, at least in part, influenced by variation in the blood flow to the extremities. The infrared thermography providing more information on the development of disturbances in the peripheral circulation may be used with an advantage in occupational health examinations and in special clinical work.

Avaliou-se a atividade oxidativa sérica da ceruloplasmina como indicadora dos teores sérico e hepático de cobre e compararam-se dois métodos de determinação da ceruloplasmina, pelo parafenileno e pela ortodianisidina. Foram colhidas 56 amostras de soro e de fígado (biópsias hepáticas) de novilhas Nelore para determinação da atividade sérica da ceruloplasmina e da concentração de cobre. As correlações entre a concentração sérica de cobre e a atividade sérica da ceruloplasmina determinadas pelos métodos do parafemileno e da ortodianisidina foram 0,75 e 0,62, respectivamente. As correlações entre a concentração hepática de cobre e a atividade sérica da ceruloplasmina pelos métodos citados foram 0,15 e 0,12. Não foi observada correlação entre os valores séricos e hepáticos de cobre. A correlação entre determinação da atividade sérica de ceruloplasmina, que utilizou o parafenileno como substrato, e o cobre sérico foi maior do que a correlação entre esse mineral e o método que usou a ortodianisidina.The ceruloplasmin oxidase activity as an indicator of serum and liver copper levels was evaluated and two different methods of ceruloplasmin determination (paraphenylen and ortodianisidine) were compared. Fifty six serum and liver samples from Nelore heifers were used to evaluate copper concentration and ceruloplasmin serum activity. The correlation coefficients between cooper serum values and ceruloplasmin serum activity using paraphenylen and ortodianisidine were 0.75 and 0.62, respectively. The correlation between ceruloplasmin serum activity and copper liver concentrations were 0.15 and 0.12, respectively, for paraphenylen and ortodianisidine. No relationship between serum and liver copper levels was observed. Cooper serum determination by paraphenylen method showed higher correlation with liver copper level in comparison to copper serum determination by ortodianisidine method.

Meat processing by drying, after salting or fermentation and before a long ripening period, is an ancient and widespread process, used to preserve meat from spoilage. Among the wide variety of dry meat products made in Europe, the most famous ones are made from pork (Italian and Spanish dry-cured hams, fermented sausages), but other interesting products are obtained from meat of different species: i.e., the “bresaola”, from bovine, horse, goat, etc. (Paleari et al., 2002). Deer meat and meat products are interesting in this picture, since they include the high degree of consumers’ appreciation for dried products and considering the growing interest for food obtained by natural husbandry and technology...

The left and right hams of forty heavy pig carcasses were thermographed to evaluate whether surface temperature differences were related to meat and ham quality. Thermal imaging analysis showed no differences in average surface temperature among classes of pH or of L* colour co-ordinate nor among classes of ham defects such as veining and red skin. However, hams with a lower fat cover displayed a significantly warmer average temperature surface. Infrared thermography seems to be a practical and non-invasive method to detect hams with a fat cover below the minimum requested to be submitted to the dry-curing process.

The microbiological and physicochemical properties of a typical fermented meat product called Pitina, prepared using ground sheep meat mixed with a variable proportion of pork lard, were studied in order to firstly assess them and to evaluate the effect of two levels of lard pork content (10% vs. 30%, weight basis) and of two ripening conditions adopted by producers (traditional vs. modified atmosphere) on the quality of final products. Both, the level of lard and the ripening condition influenced the physicochemical values and then the chemical characteristics of the ripened product. Instead, the microbiological profile seemed not to be strongly affected by the studied conditions and all batches examined showed high count of enterobacteria, coliforms and gram negative bacteria at the end of the sixtyday maturation period.

The effects of lambs separation and manual milking, which are stressors usually associated with the productive cycle of dairy sheep farm, were investigated by comparing short-term (24 h) adaptive responses in strongly versus weakly lateralized dairy ewes. Functional lateralization was scored with a series of behavioural tests (direction of free turning, direction of obstacle turning, preferential leg use in front of an obstacle) followed by a daily period of hormonal, biochemical and clinical parameters assayed on two groups of 6 strongly (SLS) and two groups of 6 weakly (WLS) lateralized ewes divided for lamb presence or separation. Differences were observed for several parameters as a function of the degree of lateralization score and stressors application. The results suggest that behavioural lateralization parameters should be taken into consideration as a crucial variable to understand stress responses and their modulation in relation to animal welfare.

In order to verify the effect of the introduction of concentrates without GMO risk and at low aflatoxin risk in the diet of grazing milk ewes on the quanti-qualitative production of meat of their milk-fed light lambs, two trials were carried out - in Sicily, on 32 Comisana lambs, slaughtered at 49±4 days (trial 1); and in Sardinia, on 28 Sarda lambs, slaughtered at 31±4 days(trial 2) - comparing the following grazing dams’ feeding regimes: High stocking rate + Organic (barley – tickbean or pea) Concentrate (HO); High stocking rate + Conventional (maize-soybean) Concentrate (HC); Low stocking rate + Organic Concentrate (LO); Low stocking rate + Conventional Concentrate (LC). Lamb performances, carcass quality, meat colour and lipid content were not modified by dam’s feeding regimen. However, significant differences were observed in the fatty acid (FA) composition of the intramuscular fat of the older suckling lambs of trial 1. The main variation concerned n-3 polyunsaturated FAs and conjugated linoleic acids.

The characteristics of ripened cheeses depend on a large number of factors, of which animal feeding plays an important role. Several researches showed influences of factors linked to forage, such as quality or method of conservation (Verdier-Metz et al., 1998).

The whole seed of sweet lupin var. Multitalia with a protein content higher than 35% and a low content in alkaloids constitutes a good protein source alternative to soybean meal, today deriving for more than 60% from OGM (Genetically Modified Organisms) cultivation. Moreover, the poor national production and the cost of feeds for animals forces the livestock compartment to utilize alternative biomasses the availability of which highly depends on from the environment and the season.

Today, after more than 70 years of continued progress on silicon technology, about 85% of cumulative installed photovolatic (PV) modules are based on crystalline silicon (c-Si). PV devices based on silicon are the most common solar cells currently being produced, and it is mainly due to silicon technology that the PV has grown by 40% per year over the last decade. An additional step in the silicon solar cell development is ongoing, and it is related to a further efficiency improvement through defect control, device optimization, surface modification, and nanotechnology approaches. This paper attempts to briefly review the most important advances and current technologies used to produce crystalline silicon solar devices and in the meantime the most challenging and promising strategies acting to increase the efficiency to cost/ratio of silicon solar cells. Eventually, the impact and the potentiality of using a nanotechnology approach in a silicon-based solar cell are also described.

Testosterone has rapid nongenomic vasodilator effects which could be involved in protective cardiovascular actions. Several authors suggested specific mechanisms to explain this effect, but this matter was not clarified yet. We studied the actions of testosterone and cholesterol on endothelium-denuded rat aorta and their effects on the L-type Ca2+ current (ICa,L) and potassium current (IK). Testosterone (1–100 μM) totally relaxed, in a rapid and concentration-dependent way, the aortic rings contracted by KCl or by (−)-Bay K8644 (BAY). Cholesterol also fully relaxed the contractions induced by KCl. None of the potassium channel antagonists tested (glibenclamide, tetraethylammonium and 4-aminopyridine) modified significantly the relaxant effect of testosterone. The antagonist of classic testosterone receptors, flutamide, did not modify the vasorelaxant effect of testosterone. Furthermore, testosterone and cholesterol inhibited either basal and BAY-stimulated ICa,L in A7r5 cells and they have no effects on IK. In summary, our results demonstrate that cholesterol and testosterone relax rat aorta by inhibiting LTCC. This effect of testosterone is not mediated by the classic hormone receptor or by potassium channel activation. These results suggest that the vasodilator mechanism of cholesterol and testosterone is the same.

The present paper will discuss the work in progress at the Department of Physics of the University of Pisa in collaboration with the IFAC laboratory of CNR in Florence to develop pressure sensors with outstanding sensitivity in the acoustic and ultrasonic ranges. These devices are based on optically-pumped fiber lasers, where the mirrors are Bragg gratings written into the fiber core.

We present here the design, synthesis, and analysis of a series of receptors for peptide ligands inspired by the hydrogen-bonding pattern of protein β-sheets. The receptors themselves can be regarded as strands 1 and 3 of a three-stranded β-sheet, with cross-linking between the chains through the 4-position of adjacent phenylalanine residues. We also report on the conformational equilibria of these receptors in solution as well as on their tendency to dimerize. 1H NMR titration experiments are used to quantify the dimerization constants, as well as the association constant values of the 1:1 complexes formed between the receptors and a series of diamides and dipeptides. The receptors show moderate levels of selectivity in the molecular recognition of the hydrogen-bonding pattern present in the diamide series, selecting the α-amino acid-related hydrogen-bonding functionality. Only one of the two cyclic receptors shows modest signs of enantioselectivity and moderate diastereoselectivity in the recognition of the enantiomers and diastereoisomers of the Ala-Ala dipeptide (ΔΔG01 (DD-DL) = −1.08 kcal/mol and ΔΔG01 (DD-LD) = −0.89 kcal/mol). Surprisingly, the linear synthetic precursors show higher levels of stereoselectivity than their cyclic counterparts.

The adsorption kinetics of human serum albumin (HSA) on bare and poly-L-arginine (PARG)-modified silica substrates were investigated using reflectometry and atomic force microscopy (AFM). Measurements were carried out at various pHs, flow rates and albumin concentrations in the 10 and 150 mM NaCl solutions. The mass transfer rate constants and the maximum protein coverages were determined for the bare silica at pH 4.0 and theoretically interpreted in terms of the hybrid random sequential adsorption model. These results were used as reference data for the analysis of adsorption kinetics at larger pHs. It was shown that the adsorption on bare silica rapidly decreased with pH and became negligible at pH 7.4. The albumin adsorption on PARG-functionalized silica showed an opposite trend, i.e., it was negligible at pH 4 and attained maximum values at pH 7.4 and 150 mM NaCl, the conditions corresponding to the blood serum environment. These results were interpreted as the evidence of a significant role of electrostatic interactions in the albumin adsorption on the bare and PARG-modified silica. It was also argued that our results can serve as useful reference data enabling a proper interpretation of protein adsorption on substrates functionalized by polyelectrolytes.

Deposition kinetics of polymer particles characterized by a prolate spheroid shape on gold sensors modified by the adsorption of poly(allylamine) was investigated using a quartz crystal microbalance and atomic force microscopy. Reference measurements were also performed for polymer particles of a spherical shape and the same diameter as the spheroid shorter axis. Primarily, the frequency and dissipation shifts for various overtones were measured as a function of time. These kinetic data were transformed into the dependence of the complex impedance, scaled up by the inertia impedance, upon the particle size to the hydrodynamic boundary layer ratio. The results obtained for low particle coverage were interpolated, which enabled the derivation of Sauerbrey-like equations, yielding the real particle coverage using the experimental frequency or dissipation (bandwidth) shifts. Experiments carried out for a long deposition time confirmed that, for spheroids, the imaginary and real impedance components were equal to each other for all overtones and for a large range of particle coverage. This result was explained in terms of a hydrodynamic, lubrication-like contact of particles with the sensor, enabling their sliding motion. In contrast, the experimental data obtained for spheres, where the impedance ratio was a complicated function of overtones and particle coverage, showed that the contact was rather stiff, preventing their motion over the sensor. It was concluded that results obtained in this work can be exploited as useful reference systems for a quantitative interpretation of bioparticle, especially bacteria, deposition kinetics on macroion-modified surfaces.

We show by extensive experimental characterization combined with molecular simulations that pH has a major impact on the assembly mechanism and properties of poly(L-lysine) (PLL) and poly(L-glutamic acid) (PGA) complexes. A combination of dynamic light scattering (DLS) and laser Doppler velocimetry (LDV) is used to assess the complexation, charge state, and other physical characteristics of the complexes, isothermal titration calorimetry (ITC) is used to examine the complexation thermodynamics, and circular dichroism (CD) is used to extract the polypeptides' secondary structure. For enhanced analysis and interpretation of the data, analytical ultracentrifugation (AUC) is used to define the precise molecular weights and solution association of the peptides. Molecular dynamics simulations reveal the associated intra- and intermolecular binding changes in terms of intrinsic vs. extrinsic charge compensation, the role of hydrogen bonding, and secondary structure changes, aiding in the interpretation of the experimental data. We combine the data to reveal the pH dependency of PLL/PGA complexation and the associated molecular level mechanisms. This work shows that not only pH provides a means to control complex formation but also that the associated changes in the secondary structure and binding conformation can be systematically used to control materials assembly. This gives access to rational design of peptide materials via pH control.

Deposition kinetics of positively charged polymer microparticles, characterized by prolate spheroid shape, at silica and gold sensors was investigated using the quartz microbalance (QCM) technique. Reference measurements were also performed for positively charged polymer particles of spherical shape and the same mass as the spheroids. Primarily, the frequency and bandwidth shifts for various overtones were measured as a function of time. It is shown that the ratio of these signals is close to unity for all overtones. These results were converted to the dependence of the frequency shift on the particle coverage, directly determined by atomic force microscopy and theoretically interpreted in terms of the hydrodynamic model. A quantitative agreement with experiments was attained considering particle slip relative to the ambient oscillating flow. In contrast, the theoretical results pertinent to the rigid contact model proved inadequate. The particle deposition kinetics derived from the QCM method was compared with theoretical modeling performed according to the random sequential adsorption approach. This allowed to assess the feasibility of the QCM technique to furnish proper deposition kinetics for anisotropic particles. It is argued that the hydrodynamic slip effect should be considered in the interpretation of QCM kinetic results acquired for bioparticles, especially viruses.

Mechanisms and kinetic of particle deposition at solid surfaces leading to the formation of self-assembled layers of controlled structure and density were reviewed. In the first part theoretical aspects were briefly discussed, comprising limiting analytical solutions for the linear transport under flow and diffusion. Methods of the deposition kinetics analysis for non-linear regimes affected by surface blocking were also considered. Characteristic monolayer formation times under diffusion and flow for the nanoparticle size range were calculated. In the second part illustrative experimental results obtained for micro- and nanoparticles were discussed. Deposition at planar substrates was analyzed with emphasis focused on the stability of layers and the release kinetics of silver particles. Applicability of the quartz microbalance measurements (QCM) for quantitative studies of nanoparticle deposition kinetic was also discussed. Except for noble metal and polymer particles, representative results for virus deposition at abiotic surfaces were analyzed. Final part of the review was devoted to nanoparticle corona formation at polymer carrier particles investigated by combination of the concentration depletion, AFM, SEM and the in situ electrokinetic method. It is argued that the results obtained for colloid particles can be used as reliable reference systems for interpretation of protein and other bioparticle deposition, confirming the thesis that simple is universal., (Copyright © 2022 Elsevier B.V. Physicochemical properties of poly-L-arginine (P-Arg) molecules in NaCl solutions were determined by molecular dynamics (MD) modeling and various experimental techniques. Primarily, the molecule conformations, the monomer length and the chain diameter were theoretically calculated. These results were used to interpret experimental data, which comprised the molecule secondary structure, the diffusion coefficient, the hydrodynamic diameter and the electrophoretic mobility determined at various ionic strengths and pHs. Using these data, the electrokinetic charge and the effective ionization degree of P-Arg molecules were determined. In addition, the dynamic viscosity measurements for dilute P-Arg solutions enabledto determine the molecule intrinsic viscosity, which was equal to 500 and 90 for ionic strength of 10 -5 and 0.15 M, respectively. This confirmed that P-Arg molecules assumed extended conformations and approached the slender body limit at the low range of ionic strength. The experimental data were also used to determine the molecule length and the chain diameter, which agreed with theoretical predictions. Exploiting these results, a robust method for determining the molar mass of P-Arg samples, the hydrodynamic diameter, the radius of gyration and the sedimentation coefficient was proposed.

Microsecond-long all-atom molecular dynamics (MD) simulations, circular dichroism, laser Doppler velocimetry, and dynamic light-scattering techniques have been used to investigate pH-induced changes in the secondary structure, charge, and conformation of poly l-lysine (PLL) and poly l-glutamic acid (PGA). The employed combination of the experimental methods reveals for both PLL and PGA a narrow pH range at which they are charged enough to form stable colloidal suspensions, maintaining their α-helix content above 60%; an elevated charge state of the peptides required for colloidal stability promotes the peptide solvation as a random coil. To obtain a more microscopic view on the conformations and to verify the modeling performance, peptide secondary structure and conformations rising in MD simulations are also examined using three different force fields, i.e., OPLS-AA, CHARMM27, and AMBER99SB*-ILDNP. Ramachandran plots reveal that in the examined setup the α-helix content is systematically overestimated in CHARMM27, while OPLS-AA overestimates the β-sheet fraction at lower ionization degrees. At high ionization degrees, the OPLS-AA force-field-predicted secondary structure fractions match the experimentally measured distribution most closely. However, the pH-induced changes in PLL and PGA secondary structure are reasonably captured only by the AMBER99SB*-ILDNP force field, with the exception of the fully charged PGA in which the α-helix content is overestimated. The comparison to simulations results shows that the examined force fields involve significant deviations in their predictions for charged homopolypeptides. The detailed mapping of secondary structure dependency on pH for the polypeptides, especially finding the stable colloidal α-helical regime for both examined peptides, has significant potential for practical applications of the charged homopolypeptides. The findings raise attention especially to the pH fine tuning as an underappreciated control factor in surface modification and self-assembly.

Deposition kinetics of fibrinogen/polystyrene particle complexes on mica and the silicon/silica substrates was studied using the direct optical and atomic force microscopy. Initially, basic physicochemical characteristics of fibrinogen and the microparticles were acquired using the dynamic light scattering and the electrophoretic mobility methods, whereas the zeta potential of the substrates was determined using the streaming potential measurements. Subsequently an efficient method for the preparation of fibrinogen/polymer microparticle complexes characterized by controlled coverage and molecule orientation was developed. It was demonstrated that for a lower suspension concentration the complexes are stable for pH range 3-9 and for a large concentration for pH below 4.5 and above 5.5. This enabled to carry out thorough pH cycling experiments where their isoelectric point was determined to appear at pH 5. Kinetic measurements showed that the deposition rate of the complexes vanished at pH above 5, whereas the kinetics of the positively charged amidine particles, used as control, remained at maximum for pH up to 9. These results were theoretically interpreted using the hybrid random sequential adsorption model. It was confirmed that the deposition kinetics of the complexes can be adequately analyzed in terms of the mean-field approach, analogously to the ordinary colloid particle behavior. This is in contrast to the fibrinogen molecule behavior, which efficiently adsorb on negatively charged substrates for the entire range pHs up to 9.7. Various physicochemical parameters of poly-l-lysine (PLL) solutions comprising the diffusion coefficient, the electrophoretic mobility, the density, and the intrinsic viscosity were determined for the pH range 3.0-9.2. This allowed us to calculate derivative parameters characterizing the PLL molecule such as: zeta potential, the number of electrokinetic charges, ionization degree, contour length, and cross section area. These data were exploited in theoretical calculations of PLL adsorption kinetics on solid substrates under diffusion transport. A hybrid approach was used comprising a blocking function derived from the random sequential adsorption (RSA) model. In experiments, the PLL adsorption on mica was studied using the streaming potential measurements and interpreted in terms of a general electrokinetic model. This confirmed a side-on adsorption mechanism of the macroion molecules at the examined pH range. Additionally, using this method, the stability of PLL monolayers was determined performing in situ desorption kinetic experiments. In this way, the equilibrium adsorption constant and the energy minimum depth were determined. It was confirmed that the monolayer stability decreases with pH following the decrease in the number of electrokinetic charges per molecule. This confirmed the electrostatic interaction driven adsorption mechanism of PLL. It is also predicted that at pH 5.7-7.4 the monolayers were stable under diffusion-controlled desorption over the time exceeding 100 h. In addition to their significance for basic science, the results obtained in this work can be exploited for developing procedures for preparing stable PLL monolayers of well controlled coverage and electrokinetic properties.

The kinetics of positively charged gold nanoparticle self-assembly on oxidized silicon substrates (wafers) under diffusion-controlled transport was studied using scanning electron microscopy (SEM) and atomic force microscopy (AFM). The latter technique allowed the roughness parameters of the monolayer (root mean square) to be determined as a function of the particle coverage. These results were adequately interpreted in terms of a theoretical model developed for surfaces covered by features of spherical shape considering the tip convolution effect. The stability and the electrokinetic characteristics (zeta potential) of the monolayers were also acquired using streaming potential measurements. It was shown that the inversion of the negative zeta potential of the bare substrate (overcharging) occurs at the particle coverage equal to 0.15, and for larger coverages positive zeta potential values were asymptotically attained. Additionally, the desorption kinetics of the particles was investigated by the streaming potential method, which confirmed the stability of the monolayers for a broad range of pHs. It was argued that these results enable to develop an efficient method for the preparation of gold sensors exhibiting a well-controlled surface roughness and electrostatic charge comprising both negative and positive values.

Quantitative studies on self-assembled hematite/silica nanoparticle (NP) bilayers on mica were performed by applying scanning electron microscopy (SEM), atomic force microscopy (AFM), and streaming potential measurements. The coverage of the supporting hematite layers was adjusted by changing the bulk concentration of the suspension and the deposition time. The coverage was determined by direct enumeration of deposited particles from AFM images and SEM micrographs. Afterward, silica nanoparticle monolayers were assembled under diffusion-controlled transport. A unique functional relationship was derived connecting the silica coverage with the hematite precursor layer coverage. The formation of the hematite monolayer and the hematite/silica bilayer was also monitored in situ by streaming potential measurements. It was confirmed that the zeta potential of the bilayers was independent of the supporting layer coverage, exceeding 0.15. These measurements were theoretically interpreted in terms of the general electrokinetic model that allowed for deriving a formula for calculating nanoparticle coverage in the bilayers. Additionally, from desorption experiments, the interactions among hematite/silica particles in the bilayers were determined using DLVO theory. These results facilitate the development of a robust method of preparing nanoparticle bilayers with controlled properties, with potential applications in catalytic processes.

Proteins at interfaces are a key for many applications in the biomedical field, in biotechnologies, in biocatalysis, in food industry, etc. The development of surface layers that allow to control and manipulate proteins is thus highly desired. In previous works, we have shown that mixed polymer brushes combining the protein-repellent properties of poly(ethylene oxide) (PEO) and the stimuli-responsive adsorption behavior of poly(acrylic acid) (PAA) could be synthesized and used to achieve switchable protein adsorption. With the present work, we bring more insight into the rational design of such smart thin films by unravelling the role of PEO on the adsorption/desorption of proteins. The PEO content of the mixed PEO/PAA brushes was regulated, on the one hand, by using PEO with different molar masses and, on the other hand, by varying the ratio of PEO and PAA in the solutions used to synthesize the brushes. The influence of ionic strength on the protein adsorption behavior was also further examined. The behavior of three proteins-human serum albumin, lysozyme, and human fibrinogen, which have very different size, shape, and isoelectric point-was investigated. X-ray photoelectron spectroscopy, quartz crystal microbalance, atomic force microscopy, and streaming potential measurements were used to characterize the mixed polymer brushes and, in particular, to estimate the fraction of each polymer within the brushes. Protein adsorption and desorption conditions were selected based on previous studies. While brushes with a lower PEO content allowed the higher protein adsorption to occur, fully reversible adsorption could only be achieved when the PEO surface density was at least 25 PEO units per nm 2 . Taken together, the results increase the ability to finely tune protein adsorption, especially with temporal control. This opens up possibilities of applications in biosensor design, separation technologies, nanotransport, etc.
