1. Competencies based innovative learning solutions for co-development of climate services in West Africa

Abstract: In developing countries, and particularly in West Africa, the role of Climate Services (CS) for sustainable development is growing thanks to wide spreading collaboration among European institutions, including National Meteorological and Hydrological Services (NMHS) research centers, universities, and homologue local institutions. Operationally, the implementation of CSs in developing countries is mainly pivoted on NMHS, which, according to the World Meteorological Organization (WMO), are dramatically affected by unmet learning demand. The global scale of learning needs for co-development of CSs calls for innovative solutions and a range of flexible modalities to reach learners in a variety of ways, and for sharing resources and successful strategies within the global education and training community. In order to harmonize expected learning outcomes, WMO defined a competency framework (CF) for CSs to be used in the implementation of training initiatives and knowledge sharing tools. This paper presents the strategic and methodological approach adopted in the implementation of the TOPaCS, a new knowledge-based distance learning initiative, aiming to provide a flexible learning environment within the CSs CF of WMO ensuring coherence with other WMO education initiatives (Global Campus, other RTCs, etc.). The methodological approach adopted is based on the competency-based approach to training, where competencies are composed by elements of knowledge and skill. TOPaCS integrates the WMO CF for CSs into a taxonomy co-designed with stakeholders at different levels, and allows the definition of learning paths, which are a further interactive opportunity for co-development of CSs within the TOPaCS learning ecosystem. Indeed, the approach aims also to guide further instructional strategies and assessments and becomes a starting point to build a common language enabling a better cooperation and exchange between the different CSs training initiatives.

2. Learning and teaching about seasonal climate forecasts: a Mediterranean educational experience toward operational climate services

Abstract: During the World Climate Conference-3, Capacity Development has been acknowledged as a transversal component underpinning all the other Pillars of the Global Framework for Climate Services. Within the Mediterranean basin, the interest of climate services based on seasonal climate forecasts is rising because they provide an opportunity for developing a proactive approach towards water management. In 2014, the Regional Training Center (RTC) in Italy, in agreement with World Meteorological Organization (WMO) and member countries of Region I and VI, identified seasonal climate forecasts as a strategic subject of capacity development for the Mediterranean Region. Following design-based research methods, this paper presents the evolution of the training approaches adopted, from classroom lessons to a blend of practical and theoretical classroom and distance learning. This evolution, as well as the rising satisfaction of trainees' expectations encouraged WMO and the RTC to widen the spectrum of beneficiaries and to make the resulting course materials available for other regions and RTCs as a course package. The course package provides essential guidelines to facilitate adoption and adaptation of the course by different institutions and instructors, including those in other WMO Regions, based on regional or institutional learning needs and standards, while also serving the needs of individual learners.

Abstract: Cosmological constraints from key probes of the Euclid imaging survey rely critically on the accurate determination of the true redshift distributions, $n(z)$, of tomographic redshift bins. We determine whether the mean redshift, $$, of ten Euclid tomographic redshift bins can be calibrated to the Euclid target uncertainties of $\sigma()$ is measured in each tomographic redshift bin to an accuracy of order 0.01 or better. By measuring the clustering redshifts on subsets of the full Flagship area, we construct scaling relations that allow us to extrapolate the method performance to larger sky areas than are currently available in the mock. For the full expected Euclid, BOSS, and DESI overlap region of approximately 6000 deg$^{2}$, the uncertainties attainable by clustering redshifts exceeds the Euclid requirement by at least a factor of three for both $n(z)$ models considered, although systematic biases limit the accuracy. Clustering redshifts are an extremely effective method for redshift calibration for Euclid if the sources of systematic biases can be determined and removed, or calibrated-out with sufficiently realistic simulations. Abstract: The Euclid space telescope will survey a large dataset of cosmic voids traced by dense samples of galaxies. In this work we estimate its expected performance when exploiting angular photometric void clustering, galaxy weak lensing and their cross-correlation. To this aim, we implement a Fisher matrix approach tailored for voids from the Euclid photometric dataset and present the first forecasts on cosmological parameters that include the void-lensing correlation. We examine two different probe settings, pessimistic and optimistic, both for void clustering and galaxy lensing. We carry out forecast analyses in four model cosmologies, accounting for a varying total neutrino mass, $M_\nu$, and a dynamical dark energy (DE) equation of state, $w(z)$, described by the CPL parametrisation. We find that void clustering constraints on $h$ and $\Omega_b$ are competitive with galaxy lensing alone, while errors on $n_s$ decrease thanks to the orthogonality of the two probes in the 2D-projected parameter space. We also note that, as a whole, the inclusion of the void-lensing cross-correlation signal improves parameter constraints by $10-15\%$, and enhances the joint void clustering and galaxy lensing Figure of Merit (FoM) by $10\%$ and $25\%$, in the pessimistic and optimistic scenarios, respectively. Finally, when further combining with the spectroscopic galaxy clustering, assumed as an independent probe, we find that, in the most competitive case, the FoM increases by a factor of 4 with respect to the combination of weak lensing and spectroscopic galaxy clustering taken as independent probes. The forecasts presented in this work show that photometric void-clustering and its cross-correlation with galaxy lensing deserve to be exploited in the data analysis of the Euclid galaxy survey and promise to improve its constraining power, especially on $h$, $\Omega_b$, the neutrino mass, and the DE evolution., Comment: 22 pages, 9 figures - published in A&A

Loureiro et al., We present a tomographic weak lensing analysis of the Kilo Degree Survey Data Release 4 (KiDS-1000), using a new pseudo angular power spectrum estimator (pseudo-Cℓ) under development for the ESA Euclid mission. Over 21 million galaxies with shape information are divided into five tomographic redshift bins, ranging from 0.1 to 1.2 in photometric redshift. 6. MR Micro-Neurography and a Segmentation Protocol Applied to Diabetic Neuropathy

Abstract: The aim of this study was to assess with MRI morphometric ultrastructural changes in nerves affected by diabetic peripheral neuropathy (DPN). We used an MR micro-neurography imaging protocol and a semiautomated technique of tissue segmentation to visualize and measure the volume of internal nerve components, such as the epineurium and nerve fascicles. The tibial nerves of 16 patients affected by DPN and of 15 healthy volunteers were imaged. Nerves volume (NV), fascicles volume (FV), fascicles to nerve ratio (FNR), and nerves cross-sectional areas (CSA) were obtained. In patients with DPN the NV was increased and the FNR was decreased, as a result of an increase of the epineurium (FNR in diabetic neuropathy 0,665; in controls 0,699, p=0,040). CSA was increased in subjects with DPN (12,84 mm2 versus 10,22 mm2, p=0,003). The FV was increased in patients with moderate to severe DPN. We have demonstrated structural changes occurring in nerves affected by DPN, which otherwise are assessable only with an invasive biopsy. MR micro-neurography appears to be suitable for the study of microscopic changes in tibial nerves of diabetic patients.

Abstract: Euclid Consortium: M. S. Cagliari et al., [Context] The ESA Euclid mission will produce photometric galaxy samples over 15 000 square degrees of the sky that will be rich for clustering and weak lensing statistics. The accuracy of the cosmological constraints derived from these measurements will depend on the knowledge of the underlying redshift distributions based on photometric redshift calibrations., [Aims] A new approach is proposed to use the stacked spectra from Euclid slitless spectroscopy to augment broad-band photometric information to constrain the redshift distribution with spectral energy distribution fitting. The high spectral resolution available in the stacked spectra complements the photometry and helps to break the colour-redshift degeneracy and constrain the redshift distribution of galaxy samples., [Methods] We modelled the stacked spectra as a linear mixture of spectral templates. The mixture may be inverted to infer the underlying redshift distribution using constrained regression algorithms. We demonstrate the method on simulated Vera C. Rubin Observatory and Euclid mock survey data sets based on the Euclid Flagship mock galaxy catalogue. We assess the accuracy of the reconstruction by considering the inference of the baryon acoustic scale from angular two-point correlation function measurements., [Results] We selected mock photometric galaxy samples at redshift z > 1 using the self-organising map algorithm. Considering the idealised case without dust attenuation, we find that the redshift distributions of these samples can be recovered with 0.5% accuracy on the baryon acoustic scale. The estimates are not significantly degraded by the spectroscopic measurement noise due to the large sample size. However, the error degrades to 2% when the dust attenuation model is left free. Abstract: Euclid Consortium: S. Nesseris et al., [Context] The standard cosmological model is based on the fundamental assumptions of a spatially homogeneous and isotropic universe on large scales. An observational detection of a violation of these assumptions at any redshift would immediately indicate the presence of new physics., [Aims] We quantify the ability of the Euclid mission, together with contemporary surveys, to improve the current sensitivity of null tests of the canonical cosmological constant Λ and the cold dark matter (ΛCDM) model in the redshift range 0, [Methods] We considered both currently available data and simulated Euclid and external data products based on a ΛCDM fiducial model, an evolving dark energy model assuming the Chevallier-Polarski-Linder parameterization or an inhomogeneous Lemaître-Tolman-Bondi model with a cosmological constant Λ, and carried out two separate but complementary analyses: a machine learning reconstruction of the null tests based on genetic algorithms, and a theory-agnostic parametric approach based on Taylor expansion and binning of the data, in order to avoid assumptions about any particular model., [Results] We find that in combination with external probes, Euclid can improve current constraints on null tests of the ΛCDM by approximately a factor of three when using the machine learning approach and by a further factor of two in the case of the parametric approach. However, we also find that in certain cases, the parametric approach may be biased against or missing some features of models far from ΛCDM., [Conclusions] Our analysis highlights the importance of synergies between Euclid and other surveys. Abstract: Flood estimation and flood management have traditionally been the domain of hydrologists, water resources engineers and statisticians, and disciplinary approaches abound. Dominant views have been shaped; one example is the catchment perspective: floods are formed and influenced by the interaction of local, catchment-specific characteristics, such as meteorology, topography and geology. These traditional views have been beneficial, but they have a narrow framing. In this paper we contrast traditional views with broader perspectives that are emerging from an improved understanding of the climatic context of floods. We come to the following conclusions: (1) extending the traditional system boundaries (local catchment, recent decades, hydrological/hydraulic processes) opens up exciting possibilities for better understanding and improved tools for flood risk assessment and management. (2) Statistical approaches in flood estimation need to be complemented by the search for the causal mechanisms and dominant processes in the atmosphere, catchment and river system that leave their fingerprints on flood characteristics. (3) Natural climate variability leads to time-varying flood characteristics, and this variation may be partially quantifiable and predictable, with the perspective of dynamic, climate-informed flood risk management. (4) Efforts are needed to fully account for factors that contribute to changes in all three risk components (hazard, exposure, vulnerability) and to better understand the interactions between society and floods. (5) Given the global scale and societal importance, we call for the organization of an international multidisciplinary collaboration and data-sharing initiative to further understand the links between climate and flooding and to advance flood research.

Abstract: Euclid Consortium: N. Hamaus et al., Euclid is poised to survey galaxies across a cosmological volume of unprecedented size, providing observations of more than a billion objects distributed over a third of the full sky. Approximately 20 million of these galaxies will have their spectroscopy available, allowing us to map the three-dimensional large-scale structure of the Universe in great detail. This paper investigates prospects for the detection of cosmic voids therein and the unique benefit they provide for cosmological studies. In particular, we study the imprints of dynamic (redshift-space) and geometric (Alcock–Paczynski) distortions of average void shapes and their constraining power on the growth of structure and cosmological distance ratios. To this end, we made use of the Flagship mock catalog, a state-of-the-art simulation of the data expected to be observed with Euclid. We arranged the data into four adjacent redshift bins, each of which contains about 11 000 voids and we estimated the stacked void-galaxy cross-correlation function in every bin. Fitting a linear-theory model to the data, we obtained constraints on f/b and DMH, where f is the linear growth rate of density fluctuations, b the galaxy bias, DM the comoving angular diameter distance, and H the Hubble rate. In addition, we marginalized over two nuisance parameters included in our model to account for unknown systematic effects in the analysis. With this approach, Euclid will be able to reach a relative precision of about 4% on measurements of f/b and 0.5% on DMH in each redshift bin. Better modeling or calibration of the nuisance parameters may further increase this precision to 1% and 0.4%, respectively. Our results show that the exploitation of cosmic voids in Euclid will provide competitive constraints on cosmology even as a stand-alone probe. Abstract: An accurate covariance matrix is essential for obtaining reliable cosmological results when using a Gaussian likelihood. In this paper we study the covariance of pseudo-$C_\ell$ estimates of tomographic cosmic shear power spectra. Using two existing publicly available codes in combination, we calculate the full covariance matrix, including mode-coupling contributions arising from both partial sky coverage and non-linear structure growth. For three different sky masks, we compare the theoretical covariance matrix to that estimated from publicly available N-body weak lensing simulations, finding good agreement. We find that as a more extreme sky cut is applied, a corresponding increase in both Gaussian off-diagonal covariance and non-Gaussian super-sample covariance is observed in both theory and simulations, in accordance with expectations. Studying the different contributions to the covariance in detail, we find that the Gaussian covariance dominates along the main diagonal and the closest off-diagonals, but further away from the main diagonal the super-sample covariance is dominant. Forming mock constraints in parameters describing matter clustering and dark energy, we find that neglecting non-Gaussian contributions to the covariance can lead to underestimating the true size of confidence regions by up to 70 per cent. The dominant non-Gaussian covariance component is the super-sample covariance, but neglecting the smaller connected non-Gaussian covariance can still lead to the underestimation of uncertainties by 10--20 per cent. A real cosmological analysis will require marginalisation over many nuisance parameters, which will decrease the relative importance of all cosmological contributions to the covariance, so these values should be taken as upper limits on the importance of each component., 15 pages, 8 figures; matches version accepted by A&A; code available at https://github.com/robinupham/shear_pcl_cov

12. Jetstream and rainfall distribution in the Mediterranean region

Abstract: This is a study on the impact of the jetstream in the Euro-Atlantic region on the rainfall distribution in the Mediterranean region; the study, based on data analysis, is restricted to the Mediterranean rainy season, which lasts from September to May. During this season, most of the weather systems originate over the Atlantic, and are carried towards the Mediterranean region by the westerly flow. In the upper troposphere of the Euro-Atlantic region this flow is characterized by two jets: the Atlantic jet, which crosses the ocean with a northeasterly tilt, and the African jet, which flows above the coast of North Africa. This study shows that the cross-jet circulation of the Atlantic jet favors storm activity in its exit region, while the cross-jet circulation of the African jet suppresses this kind of activity in its entrance region, with the 1st jet-stormtrack covariance mode explaining nearly 50% of the variability. It follows that the rainfall distribution downstream to these cross-jet circulations is strongly influenced by their relative positions. Specifically, in fall, rainfall is abundant in the western Mediterranean basin (WM), when the Atlantic jet is relatively strong but its northeasterly tilt is small, and the African jet is in its easternmost position. In winter, rainfall is abundant in the eastern Mediterranean basin (EM); this is when the Atlantic jet reaches the Scandinavian peninsula and the African jet is in its westernmost position. In spring, when the two jets weaken, the Atlantic jet retreats over the ocean, but the African jet stays in its winter position, rainfall is abundant in the Alpine region and in the Balkans. In addition, the covariance between precipitation and the jetstream has been evaluated. In fall, the latitudinal displacement of the Atlantic jet and the longitudinal displacement of the African jet modulate rainfall anomalies in the WM, with 38% explained covariance. In winter, the latitudinal displacement of the Atlantic jet produces rainfall anomalies in the western and central Mediterranean, with 45% explained covariance. In spring, the latitudinal displacement of the African jet produces rainfall anomalies, with 38% explained covariance.

For both cases of reds in Bordeaux and Tuscany the growing season temperatures and heat units were significantly higher for the good, compared with the poor ranked vintages. For Tuscany specific weather types (WT) are associated with each group. Abstract: Non-classical congenital adrenal hyperplasia (NCAH) is a morbid condition sustained by the reduced function of one of the enzymes involved in the adrenal steroid biosynthesis pathway, mainly the 21-hydroxylase. Different degrees of enzyme activity impairment determine different clinical pictures, with childhood or post-pubertal onset. The aim of this study was to evaluate the relationship between genotype, phenotype, and adrenal hormonal levels in a group of 66 patients affected by NCAH attending outpatient pediatric or endocrinological Clinics. Our findings show that age at pubarche/menarche was significantly younger, height SD score) and Δ bone age-chronological age were significantly higher in patients with a more severe enzyme activity impairment, while cutaneous androgenization and menstrual irregularities in post-pubertal girls were not related to the grading of genotype.

Abstract: Research into land-atmosphere coupling within the African Monsoon Multidisciplinary Analysis has highlighted the atmospheric impact of soil moisture on space scales of 5 km upwards and time scales of several days. Observational and modelling studies have shown how antecedent rainfall patterns affect new storms in the Sahel. The land feedback operates through various mechanisms, including a direct link to afternoon storm initiation from surface-induced mesoscale circulations, and indirectly via a large-scale moisture transport in the nocturnal monsoon. The results suggest potential for significant improvements in weather forecasting through assimilation of satellite data. Intriguing questions remain about the importance of vegetation memory on seasonal-interannual scales.

25. Changes in means and extreme events of temperature and precipitation in the Asia-Pacific Network region, 19552007

Abstract: In this study, spatial and temporal patterns of changes in extreme events of temperature and precipitation at 143 weather stations in ten Asia-Pacific Network (APN) countries and their associations with changes in climate means are examined for the 1955-2007 period. Averaged over the APN region, annual frequency of cool nights (days) has decreased by 6.4 days/decade (3.3 days/decade), whereas the frequency of warm nights (days) has increased by 5.4 days/decade (3.9 days/decade). The change rates in the annual frequency of warm nights (days) over the last 20 years (1988-2007) have exceeded those over the full 1955-2007 period by a factor of 1.8 (3.4). Seasonally, the frequencies of summer warm nights and days are changing more rapidly per unit change in mean temperatures than the corresponding frequencies for cool nights and days. However, normalization of the extreme and mean series shows that the rate of changes in extreme temperature events are generally less than that of mean temperatures, except for winter cold nights which are changing as rapidly as the winter mean minimum temperature. These results indicate that there have been seasonally and diurnally asymmetric changes in extreme temperature events relative to recent increases in temperature means in the APN region. There are no systematic, regional trends over the study period in total precipitation, or in the frequency and duration of extreme precipitation events. Statistically significant trends in extreme precipitation events are observed at fewer than 30% of all weather stations, with no spatially coherent pattern of change, whereas statistically significant changes in extreme temperature events have occurred at more than 70% of all weather stations, forming strongly coherent spatial patterns.

26. Catechin, epicatechin, quercetin, rutin and resveratrol in red grape: Content, in vitro antioxidant activity and interactions

Abstract: The extracts obtained from skin and seeds of 10 native Tuscan and international Vitis vinifera varieties were evaluated for their antioxidant activity, total phenolic and anthocyanin content and subjected to HPLC–UV analysis to quantify the content of five phenolic constituents of biological interest: catechin and epicatechin in seeds and quercetin, rutin and resveratrol in skin extracts. The antioxidant activity of the extracts and pure compounds was assessed by means of two different in vitro tests: scavenging of the stable DPPH radical and of authentic peroxynitrite (ONOO−). All the extracts showed significant antiradical capacity: Merlot skin was most active towards both radicals. All the five phenols investigated possessed strong antiradical activity. Quercetin, catechin and epicatechin showed maximum activity (respectively, IC 50 ( DPPH ) 5.5, 6.7 and 6.8 μM, IC 50 ( ONOO - ) 48.8, 55.7 and 56.7 μM). Potential antiradical interactive effects among the five compounds were also investigated and results indicated possible synergy between quercetin, rutin and resveratrol towards ONOO−. The effect was additive for catechin and epicatechin.

Abstract: The Mediterranean climate is affected by several tropical and subtropical systems as illustrated by some evidence presented in this chapter. These factors range from the El Nin~ o Southern Oscillation (ENSO) and tropical hurricanes to the South Asian Monsoon and Saharan dust. This leads to complex features in the Mediterranean climate variability. In the following sections, we review some tropical and subtropical teleconnections to the Mediterranean climate in the following order: El Nin~ o Southern Oscillation is elaborated in Section 2.2, the South Asian Monsoon is discussed in Section 2.3, Section 2.4 is dedicated to African monsoon, tropical cyclones are discussed in Section 2.5 and finally Red Sea Trough intrusions into the Eastern Mediterranean and the Saharan dust are discussed respectively in the last two Sections 2.6 and 2.7.

28. Influence of local SSTa and surface processes on the Sahel Rainfall

Abstract: An intense and rapid cooling of the equatorial Atlantic waters of the Guinea gulf precedes the West Africa monsoon intensifying the sea-land temperature contrast. This temperature gradient exerts, from April to July a significant influence on the West African monsoon, since it intensifies the southwesterly flow which pushes the ITCZ inland. Using rainfall data from GPCP dataset and SST data from NOAA Optimum Interpolation SST dataset in the period 1979-2004, we correlate the rainfall in the Sahel, cumulated in July-September (JAS), with the SSTAs in the gulf of Guinea, averaged within a 3 month sliding window. We find that the correlation between the monsoonal rainfall and the previous fall SSTAs is very high. The rainfall in the year 0 and the October-December (OND) SSTA in the year -1 correlation is R=0.73. The monsoonal dynamics in the Sahel is sensitive to the interannual fluctuations in the meridional gradient of the MSE, moist static energy in the PBL. Wet conditions are related to large gradients and dry conditions are related to weak gradients. In this scenario, the rainfall in the months preceding the rainy season plays an important role, because the boundary-layer MSE content is regulated by the soil wetness via re-evaporation. Applying the same method to the rainfall in the Guinea Coast region, we find a sizable correlation between the JAS Sahel rainfall in the year 0 and the OND Guinea Coast rainfall in the year -1 (R=0.39). A multi-linear regression with cross validation is used to equate the JAS rainfall in the Sahel with the OND SSTA in the Gulf of Guinea and the OND rainfall in the Guinea region. It results a good correlation between the observed and the forecasted rainfall (R=0.65) and the hit rate is 0.55 for dry events and 0.89 for wet events.

Abstract: Objective The aim of the study was to propose a set of tests to clarify the diagnosis of repeated implantation failure in patients undergoing in vitro fertilization (IVF). Study design Fifty-nine patients with at least two unsuccessful IVF attempts were included in the study. Blood samples were evaluated for the presence of underlying thyroid abnormalities, antiphospholipid antibodies (aPL), increased levels of natural killer cells (NK), inherited thrombophilia and mouse embryo assay factor (MEA-f). The same tests were performed on 20 normal fertile control patients. Results Seventy-six percent of IVF patients showed at least one abnormal result. This incidence was higher with respect to that found among control patients (45%). The prevalence of thyroid abnormalities, aPL and increased NK level was higher in IVF patients whereas no differences were observed in terms of prevalence of inherited thrombophilias and MEA-f. Conclusions A better understanding of reproductive failure mechanisms should allow an effective diagnostic flow chart and a focused therapeutic option for patients experiencing repeated IVF failure. With this objective in mind, our data provide two important results: thyroid abnormalities, aPL and increased NK levels are more prevalent in women experiencing IVF failure. No evidence was found for an association between inherited thrombophilia and MEA-f and failure to achieve pregnancy after IVF.

30. Hepatitis C virus-related chronic liver disease with autoantibodies to liver-kidney microsomes (LKM). Clinical characterization from idiopathic LKM-positive disorders

Abstract: This study was carried out on 33 patients who were sero-positive for liver-kidney microsomal antibodies (LKM) in order to examine clinical features and the presence of underlying hepatitis C virus infection. Twenty-four sera were positive for antibodies against HCV (anti-HCV) as detected by enzyme immunoassay and confirmed by recombinant immunoblot assay. These patients had chronic liver disease and the majority of those treated with interferon responded favourably. Three of the nine anti-HCV-negative patients had idiopathic chronic hepatitis and two responded favourably to steroids. Two patients were diagnosed as having toxic hepatitis and the other four had various extrahepatic disorders without evidence of liver involvement. The immunoblotting analysis showed reactivity with a 50 kDa microsomal protein which presumably corresponded to cytochrome P-450 db1 both in anti-HCV-positive and -negative sera. In addition a few anti-HCV-positive sera also reacted with a 35 kDa microsomal antigen. Autoimmune markers different from LKM were absent in both groups. The high prevalence of antibodies to the hepatitis C virus among LKM-positive sera confirms that this infection plays a role in forms of chronic hepatitis that had previously been labelled autoimmune. In patients with LKM the presence of anti-HCV may help to forecast a therapeutic response to interferon, while its absence may forecast response to steroid therapy.

32. Strategies and clinical outcome of 250 cycles of Preimplantation Genetic Diagnosis for single gene disorders.

Abstract: BACKGROUND: We report on our experience with preimplantation genetic diagnosis (PGD) for single gene disorders (SGDs), from 1999 to 2004, describing strategies and overall clinical outcome of 250 cycles in 174 couples for 23 different genetic conditions. METHODS: PGD cycles included 15 for autosomal dominant, 148 for autosomal recessive and 19 for X-linked SGDs. In addition, 68 cycles of PGD for SGDs were performed in combination with HLA matching. The strategy in each case used an initial multiplex PCR, followed by minisequencing to identify the mutation(s) combined with multiplex PCR for closely linked informative markers to increase accuracy. Linkage analysis, using intragenic and/or extragenic polymorphic microsatellite markers, was performed in cases where the disease-causing mutation(s) was unknown or undetectable. RESULTS: In 250 PGD cycles, a total of 1961 cleavage stage embryos were biopsied. PCR was successful in 3409 out of 3149 (92.4%) biopsied blastomeres and a diagnosis was possible in 1849 (94.3%) embryos. Four hundred and twenty-seven embryos were transferred in 211 cycles, resulting in 71 pregnancies (33.6% per embryo transfer), including 15 biochemical pregnancies, six spontaneous miscarriages, two ectopic pregnancies, which were terminated, and nine pregnancies which are still ongoing. The remaining pregnancies were confirmed to be unaffected and went to term without complications, resulting in the birth of 35 healthy babies. CONCLUSIONS: Minisequencing for mutation detection combined with multiplex fluorescence PCR for linkage analysis is an efficient, accurate and widely applicable strategy for PGD of SGDs. Abstract: The expression of intrahepatic delta antigen (HDAg) was studied in relation to the morphologic features of HDV hepatitis and the outcome of liver disease. The study was performed in 101 patients followed up for an average of 12 years; one or more liver biopsies were available from each patient, giving a total of 167 specimens. The histologic features were assessed using numerical scores. A significant positive relation was observed between the number of HDAg-positive cells and the extent of portal inflammation (Spearman's rank coefficient 0.75). The highest degree of inflammation and intrahepatic expression of HDAg was found before the elimination of the virus, while the outcome of HDV disease was unrelated to the severity of the initial morphologic lesion. These results suggest that the individual immune response may play an important role in the pathogenesis of HDV hepatitis.

Abstract: Objective Aim of the study was to verify in a prospective study the sensitivity and specificity of a recently elaborated diagnostic score for the prediction of the presence of mutation of genes associated with periodic fever [1]. Patients and methods Detailed clinical information of 100 Italian patients with a clinical history of periodic fever was collected since June 2007. For each patient the Diagnostic score (www.printo.it/periodicfever) was calculated. According to previous experiences a cut-off > 1.32 was chosen to define those patients at high risk to carry relevant mutations. All patients were screened for mutations of MVK, TNFRSF1A and MEFV genes. Results Ten patients displayed relevant (homozygous or compound heterozygous) mutations for MVK and MEFV genes. No structural mutations of TNFRSF1A gene were found. 10 patients dysplayed low-penetrance mutations of the TNFRSF1A gene (R92Q) or a single mutation of the MEFV gene. 80 patients were negative to all the three genes. The Diagnostic score revealed high sensitivity (90%) and specificity (65%) in discriminating positive and negative patients. The regression tree analysis [1] was able to provide the correct identification of the affected gene in 7 out of the 9 positive identified by the diagnostic score. Conclusion This study confirm the validity of the Diagnostic score as a useful tool for the identification of children at higher risk to carry relevant mutations of genes associated with periodic fever.

35. THE MASS ACCRETION RATE OF GALAXY CLUSTERS: A MEASURABLE QUANTITY.

Abstract: We explore the possibility of measuring the mass accretion rate (MAR) of galaxy clusters from their mass profiles beyond the virial radius R200. We derive the accretion rate from the mass of a spherical shell whose inner radius is 2R200, whose thickness changes with redshift, and whose infall velocity is assumed to be equal to the mean infall velocity of the spherical shells of dark matter halos extracted from N-body simulations. This approximation is rather crude in hierarchical clustering scenarios where both smooth accretion and aggregation of smaller dark matter halos contribute to the mass accretion of clusters. Nevertheless, in the redshift range z = [0, 2], our prescription returns an average MAR within 20%–40% of the average rate derived from the merger trees of dark matter halos extracted from N-body simulations. The MAR of galaxy clusters has been the topic of numerous detailed numerical and theoretical investigations, but so far it has remained inaccessible to measurements in the real universe. Since the measurement of the mass profile of clusters beyond their virial radius can be performed with the caustic technique applied to dense redshift surveys of the cluster outer regions, our result suggests that measuring the mean MAR of a sample of galaxy clusters is actually feasible. Abstract: Next generation telescopes, like Euclid, Rubin/LSST, and Roman, will open new windows on the Universe, allowing us to infer physical properties for tens of millions of galaxies. Machine learning methods are increasingly becoming the most efficient tools to handle this enormous amount of data, because they are often faster and more accurate than traditional methods. We investigate how well redshifts, stellar masses, and star-formation rates (SFR) can be measured with deep learning algorithms for observed galaxies within data mimicking the Euclid and Rubin/LSST surveys. We find that Deep Learning Neural Networks and Convolutional Neutral Networks (CNN), which are dependent on the parameter space of the training sample, perform well in measuring the properties of these galaxies and have a better accuracy than methods based on spectral energy distribution fitting. CNNs allow the processing of multi-band magnitudes together with $H_{\scriptscriptstyle\rm E}$-band images. We find that the estimates of stellar masses improve with the use of an image, but those of redshift and SFR do not. Our best results are deriving i) the redshift within a normalised error of less than 0.15 for 99.9$\%$ of the galaxies with S/N>3 in the $H_{\scriptscriptstyle\rm E}$-band; ii) the stellar mass within a factor of two ($\sim0.3 \rm dex$) for 99.5$\%$ of the considered galaxies; iii) the SFR within a factor of two ($\sim0.3 \rm dex$) for $\sim$70$\%$ of the sample. Abstract: The Complete Calibration of the Color-Redshift Relation survey (C3R2) is a spectroscopic programme designed to empirically calibrate the galaxy color-redshift relation to the Euclid depth (I_E=24.5), a key ingredient for the success of Stage IV dark energy projects based on weak lensing cosmology. A spectroscopic calibration sample as representative as possible of the galaxies in the Euclid weak lensing sample is being collected, selecting galaxies from a self-organizing map (SOM) representation of the galaxy color space. Here, we present the results of a near-infrared H- and K-bands spectroscopic campaign carried out using the LUCI instruments at the LBT. The incidence of chronic wounds worldwide, and the associated economic burden, urge for simple and cheap point-of-care testing (PoCT) devices for fast on-site diagnosis to enable appropriate early treatment. The enzyme myeloperoxidase (MPO), whose activity in infected wounds is about ten times higher than in non-infected wounds, appears to be a suitable biomarker for wound infection diagnosis. Herein, we develop a single-component foldable paper-based device for the detection of MPO in wound fluids. The analyte detection is achieved in two steps: (i) selective immunocapture of MPO, and (ii) reaction of a specific dye with the captured MPO, yielding a purple color with increasing intensity as a function of the MPO activity in infected wounds in the range of 20-85 U/mL. Ex vivo experiments with wound fluids validated the analytic efficiency of the paper-based device, and the results strongly correlate with a spectrophotometric assay.

Abstract: We present a method for fast evaluation of the covariance matrix for a two-point galaxy correlation function (2PCF) measured with the Landy-Szalay estimator. The standard way of evaluating the covariance matrix consists in running the estimator on a large number of mock catalogs, and evaluating their sample covariance. With large random catalog sizes (data-to-random objects ratio M>>1) the computational cost of the standard method is dominated by that of counting the data-random and random-random pairs, while the uncertainty of the estimate is dominated by that of data-data pairs. We present a method called Linear Construction (LC), where the covariance is estimated for small random catalogs of size M = 1 and M = 2, and the covariance for arbitrary M is constructed as a linear combination of these. We validate the method with PINOCCHIO simulations in range r = 20-200 Mpc/h, and show that the covariance estimate is unbiased. With M = 50 and with 2 Mpc/h bins, the theoretical speed-up of the method is a factor of 14. We discuss the impact on the precision matrix and parameter estimation, and derive a formula for the covariance of covariance., 17 pages, 11 figures

Abstract: Animal manure (AM), such as swine, cattle, sheep, horse, as well as other organic waste materials from recycling agri-food or other processes may be used as nutrient source for horticultural annual and perennial crops, increasing nutrient cycling and reducing costs related to acquisition of industrial fertilizers. Additionally, over the years it is expected to modify chemical, physical, and biological soil attributes such as increasing the nutrient content in the soil, which can affect crop productivity, change the food composition, fruit and vegetable storage aptitudes, and impact on the environment. The present review addresses the effect of AM applications on the soil-plant interface, emphasizing the following aspects: (a) changes in chemical, physical and biological attributes in soils with a history of AM applications, (b) effect of application of AM on annual plant productivity and (c) AM as fertilizer: productivity and food composition of horticultural crops. Successive applications of AM in soils tend to increase the chemical and physical attributes, and, increased production of grain crops. Effect of AM fertilizations on quality and nutritional value of fruits is still uncertain; it depends on several factors, including: 1) characteristics of organic matter, 2) pedoclimatic conditions, 3) time of application and 4) plant species.

Abstract: We perform for the first time N-body simulations of Interacting Dark Energy assuming non-Gaussian initial conditions, with the aim of investigating possible degeneracies of these two theoretically independent phenomena in different observational probes. We focus on the large-scale matter distribution, as well as on the statistical and structural properties of collapsed halos and cosmic voids. On very large scales, we show that it is possible to choose the Interaction and non-Gaussian parameters such that their effects on the halo power spectrum cancel, and the power spectrum is indistinguishable from a $\mathrm{\Lambda CDM}$ model. On small scales, measurements of the non-linear matter power spectrum, halo-matter bias, halo and subhalo mass function and cosmic void number function validate the degeneracy determined on large scales. However, the internal structural properties of halos and cosmic voids, namely halo concentration-mass relation and void density profile, are very different from those measured in the $\mathrm{\Lambda CDM}$ model, thereby breaking the degeneracy. In practice, the values of $\mathrm{f_{NL}}$ required to cancel the effect of interaction are already ruled by observations. Abstract: (Abridged) Present cosmological constraints and the absence of a direct detection and identification of any dark matter particle candidate leave room to the possibility that the dark sector of the Universe be actually more complex than it is normally assumed. In particular, more than one new fundamental particle could be responsible for the observed dark matter density in the Universe, and possible new interactions between dark energy and dark matter might characterize the dark sector. In the present work, we investigate the possibility that two dark matter particles exist in nature, with identical physical properties except for the sign of their coupling constant to dark energy. Extending previous works on similar scenarios, we study the evolution of the background cosmology as well as the growth of linear density perturbations for a wide range of parameters of such model. Interestingly, our results show how the simple assumption that dark matter particles carry a "charge" with respect to their interaction with the dark energy field allows for new long-range scalar forces of gravitational strength in the dark sector without conflicting with present observations both at the background and linear levels. Abstract: Objectives: The objectives of the present study were to analyze the morphology and the superficial roughness of enamel and composite resin restorations after three different bleaching regimens. Materials and methods: Enamel specimens were obtained from buccal surfaces of 21 human incisors. Two specimens were obtained for each tooth. Half part of each tooth was maintained in distilled water and served as control; the other part was treated with one of three professional bleaching agents: 38% hydrogen peroxide (Opalescence Boost; Ultradent Products, Inc.), 35% hydrogen peroxide (Whiteness HP Maxx; FGM Produtos Odontológicos Ltda), light-activated 25% hydrogen peroxide (Zoom 2; Discus Dental Inc.). The same bleaching protocols were applied on standardized composite resin (CeramX mono; Dentsply De Trey) surfaces. Composite specimens were divided in four subgroups (n = 7), according to surface treatment (distilled water was used for control group). Two enamel and composite specimens for each group were submitted to a qualitative scanning electron microscopy (SEM) analysis. Remaining specimens were measured for roughness using a 3D optical profilometer (Talysurf CLI 1000; Taylor Hobson). Data were statistically analyzed. Results: A preliminary profilometric analysis of the enamel and composite surfaces after bleaching protocols showed no significant differences in surface roughness parameters (p > 0.05) compared with those at baseline. Results of the correlated SEM analysis showed no relevant alteration on the enamel surfaces. However, some significant changes of composite surface morphology were SEM observed at high resolution (×1000 and ×3000) after conditioning treatments, suggesting a predominance of depressions after each of the three bleaching procedures. Conclusions: Although SEM analysis seems to suggest that professional bleaching procedures may adversely affect the microscopic surface texture of composites, the profilometric results showed that the tested bleaching regimens do not alter enamel and composite surface roughness.

Abstract: Lesbian, gay, bisexual, and transgender individuals ( hereafter people with minoritized sexual orientation and/or gender identities) have limited legal rights and access to resources because of their marginalized status in society. These limitations are associated with notable health disparities and increase experiences of minority stress. For those in a romantic relationship, being able to communicate and cope with one's partner-dyadic coping-can help buffer stress' deleterious effects on well-being. Given the promise of understanding how dyadic coping can mitigate experiences of sexual minority stress, the Dyadic Coping Inventory-Sexual Minority Stress (DCI-SMS) was recently created and validated with those living in the United States to assess how partners cope with sexual minority stress. Answering a global call to expand psychological science beyond a U.S. centric perspective, the purpose of this study was to validate the DCI-SMS in German and Italian using samples from Austria, Germany, Switzerland, and Italy, respectively. Confirmatory factor analysis results, along with tests of convergent and discriminant validity, and measurement invariance, suggest that the DCI-SMS is a valid measure of stress communication and dyadic coping behaviors for those in a same-gender relationship in the countries sampled. Important future directions include examining its efficacy in other countries, such as those with more adverse sociopolitical climates for people with minoritized sexual orientation and/or gender identities in a same-gender relationship. 48. The chronic alcohol consumption influences the gonadal axis in men: Results from a meta-analysis.

Abstract: Background: Low testosterone concentrations affect 2-13% of adult males, with a direct association between reduction in testosterone (T) concentrations and cardiovascular events. Lifestyle habits have been linked to visceral fat accumulation and endocrine disorders like secondary hypogonadism. Alcohol intake has also been a topic of debate, with studies showing a detrimental effect on sperm production and underlying mechanisms. This meta-analysis aims to comprehensively evaluate the effect of alcohol consumption on T serum concentrations in adult men., Methods: The literature search included only controlled clinical trials comparing men who drink alcohol to men who do not, or who assumed placebo or nonalcoholic beverages. The primary outcome was the comparison of total testosterone serum concentrations between the study and control groups. The publications were examined for publication bias using Egger's test., Results: Twenty-one studies were included in the analysis for a total of 30 trials that examined the effects of alcohol consumption on testosterone level in 10,199 subjects. The meta-analysis showed that alcohol consumption overall is related to significant reduction in circulating concentrations of total testosterone (mean difference [MD] = -4.02; 95% CI -6.30, -1.73), free T (MD = -0.17; 95% CI -0.23, -0.12), sex hormone binding globulin (SHBG) (MD = -1.94; 95% CI -3.37, -0.48), an increase in estradiol (E2) (MD = 7.65; 95% CI 1.06, 14.23) and neutral effect on luteinizing hormone (LH) (MD = -0.15; 95% CI -0.36, 0.06), independently by age, body mass index (BMI), E2, and LH serum concentrations and alcohol intake. However, these results are evident only in healthy men exposed to chronic alcohol consumption and not in those with a recognized diagnosis of alcohol use disorder or after acute alcohol intake., Conclusion: This study suggests how chronic alcohol consumption may inhibit the gonadal axis in healthy men, although the exact pathophysiological mechanisms connecting alcohol exposure and steroidogenesis are still not completely clarified., (© 2023 American Society of Andrology and European Academy of Andrology.)

49. Case report: First documented case of cerebral angiostrongyliasis caused by Angiostrongylus costaricensis in a free-ranging opossum.

Abstract: Angiostrongylus costaricensis is a metastrongyloid nematode that primarily infects the mesenteric arteries of wild rodents. This parasite is endemic in several regions of the American continent, and in humans, causes a disease known as abdominal angiostrongyliasis. Despite the important health implications of this nematode, there are limited studies investigating the involvement of wild animals in its life cycle. In this study, we present the clinical manifestations, pathologic findings, and molecular diagnosis, to the best of our current knowledge, of the first documented onset of cerebral angiostrongyliasis because of A. costaricensis infection in a juvenile free-ranging opossum ( Didelphis marsupialis ). Histopathological findings stress the presence of eosinophilic meningoencephalitis with nematodes present within the lesions, and PCR was positive for cox 1 and ITS1 reactions. The obtained sequences for a 279 bp fragment of ITS1 were 100% identical to A. costaricensis from Costa Rica. This case highlights the substantial difficulties in diagnosing neuroangiostrongyliasis, yet underscores the importance of considering A. costaricensis as a potential culprit behind neurological conditions in wild marsupials. 50. Surgery for tracheal and laryngotracheal stenosis: a historical case series.

Abstract: Objectives: Benign (laryngo-)tracheal stenosis is a relatively rare pathology and its surgical treatment is performed only at few specialized centres. This study aims to investigate outcomes after (laryngo-)tracheal resection-anastomosis, to explore potential risk factors for postoperative complications and to assess whether, over a 33-year period, there were major changes in surgical indications, techniques or outcomes., Methods: Retrospective, single-centre review of all consecutive patients who underwent tracheal or laryngo-tracheal resection/anastomosis for benign pathologies from 1990 to 2023., Results: Overall, 211 patients underwent tracheal (149 patients, 70.6%) and laryngo-tracheal (62 patients, 29.4%) resection-anastomosis. Of these, 195 patients (93.8%) were affected by iatrogenic stenosis, while 13 (6.2%) suffered from idiopathic stenosis. The median length of stenosis was 25 mm (interquartile range 1-3, 20-30). The overall morbidity rate was 27.5%, while major morbidity occurred in 10.5% of cases. One patient (0.5%) died in the postoperative period. Glottic oedema (17 patients, 8.1%), granulations (12 patients, 5.7%) and restenosis (10 patients, 4.7%) were the main complications. The only independent risk factor for postoperative complications was the length of the resected airway (P = 0.019). In the latest half of the study period, an older median age was observed, and no patient with idiopathic tracheal stenosis underwent surgery. Postoperative outcomes were comparable between surgical eras., Conclusions: Surgical treatment of (laryngo-)tracheal stenosis is challenging and should be performed by specialized centres. In our experience, morbidity and mortality rates were satisfactory, and in most cases, patients could breathe without tracheostomy. The length of the stenosis was the most significant risk factor for postoperative complications., (© The Author(s) 2024.
