Resistant starch (RS) consumption can have beneficial effects on metabolic health, but the response, in terms of effects on the gut microbiota and host physiology, varies between individuals. Factors predicting the response to RS are not yet established and would be useful for developing precision nutrition approaches that maximize the benefits of dietary fiber intake. We sought to identify predictors of gut microbiota response to RS supplementation. We enrolled 76 healthy adults into a 7-week crossover study with 59 individuals completing the study. Participants consumed RS type 2 (RS2), RS type 4 (RS4), and digestible starch, for 10 d each with 5-d washout periods in between. We collected fecal and saliva samples and food records during each treatment period. We performed 16S rRNA gene sequencing and measured fecal short-chain fatty acids (SCFAs), salivary amylase (AMY1) gene copy number, and salivary amylase activity (SAA). Dietary fiber intake was predictive of the relative abundance of several amplicon sequence variants (ASVs) at the end of both RS treatments. AMY1-related metrics were not predictive of response to RS. SAA was only predictive of the relative abundance of one ASV after digestible starch supplementation. Interestingly, SCFA concentrations increased the most during digestible starch supplementation. Treatment order (the order of consumption of RS2 and RS4), alpha diversity, and a subset of ASVs were predictive of SCFA changes after RS supplementation. Based on our findings, dietary fiber intake and gut microbiome composition would be informative if assessed prior to recommending RS supplementation because these data can be used to predict changes in specific ASVs and fecal SCFA concentrations. These findings lay a foundation to support the premise that using a precision nutrition approach to optimize the benefits of dietary fibers such as RS could be an effective strategy to compensate for the low consumption of dietary fiber nationwide.

This essay employs Dr Martin Luther King, Jr’s sermon, “The Three Dimensions of a Complete Life”, as an acute lens through which to assess and impart new meanings to African–American activists’ strivings to reach an ideal state of humanness and communal holism as they fulfilled their personal, political, and spiritual missions in the food realm during the 1960s Civil Rights era and the contemporary food justice movement. Narrative analyses of these Black activists’ personal testimonies convey that their discrete journeys to completeness—what Dr King called the ideal state of humanity in its fullness—were not only facilitated by a divine calling but were also conditioned by the enactment of their Christian faith, particularly in reconciling the affective tolls engendered by their participation in lunch-counter sit-ins and by their quests to help alleviate food insecurity among impoverish populations in the American South. Indeed, when these individuals consciously endeavored to master the three dimensions of a complete life—recognize their agency, honor the interconnectedness of humanity, and seek God’s guidance in doing both—were they able to embody their best selves and demand the realization of a truly democratic nation.

Acoustic communication serves a crucial role in the social interactions of vocal animals. Duetting—the coordinated singing among pairs of animals—has evolved independently multiple times across diverse taxonomic groups including insects, frogs, birds, and mammals. A crucial first step for understanding how information is encoded and transferred in duets is through quantifying the acoustic repertoire, which can reveal differences and similarities on multiple levels of analysis and provides the groundwork necessary for further studies of the vocal communication patterns of the focal species. Investigating acoustic tradeoffs, such as the tradeoff between the rate of syllable repetition and note bandwidth, can also provide important insights into the evolution of duets, as these tradeoffs may represent the physical and mechanical limits on signal design. In addition, identifying which sex initiates the duet can provide insights into the function of the duets. We have three main goals in the current study: (1) provide a descriptive, fine-scale analysis of Gursky’s spectral tarsier (Tarsius spectrumgurskyae) duets; (2) use unsupervised approaches to investigate sex-specific note repertoires; and (3) test for evidence of acoustic tradeoffs in the rate of note repetition and bandwidth of tarsier duet contributions. We found that both sexes were equally likely to initiate the duets and that pairs differed substantially in the duration of their duets. Our unsupervised clustering analyses indicate that both sexes have highly graded note repertoires. We also found evidence for acoustic tradeoffs in both male and female duet contributions, but the relationship in females was much more pronounced. The prevalence of this tradeoff across diverse taxonomic groups including birds, bats, and primates indicates the constraints that limit the production of rapidly repeating broadband notes may be one of the few ‘universals’ in vocal communication. Future carefully designed playback studies that investigate the behavioral response, and therefore potential information transmitted in duets to conspecifics, will be highly informative.

BackgroundThe mosquito Aedes aegypti is a medically important, globally distributed vector of the viruses that cause dengue, yellow fever, chikungunya, and Zika. Although reproduction and mate choice are key components of vector population dynamics and control, our understanding of the mechanisms of sexual selection in mosquitoes remains poor. In "good genes" models of sexual selection, females use male cues as an indicator of both mate and offspring genetic quality. Recent studies in Ae. aegypti provide evidence that male wingbeats may signal aspects of offspring quality and performance during mate selection in a process known as harmonic convergence. However, the extent to which harmonic convergence may signal overall inherent quality of mates and their offspring remains unknown.Methodology/principal findingsTo examine this, we measured the relationship between acoustic signaling and a broad panel of parent and offspring fitness traits in two generations of field-derived Ae. aegypti originating from dengue-endemic field sites in Thailand. Our data show that in this population of mosquitoes, harmonic convergence does not signal male fertility, female fecundity, or male flight performance traits, which despite displaying robust variability in both parents and their offspring were only weakly heritable.Conclusions/significanceTogether, our findings suggest that vector reproductive control programs should treat harmonic convergence as an indicator of some, but not all aspects of inherent quality, and that sexual selection likely affects Ae. aegypti in a trait-, population-, and environment-dependent manner.

Alternative, biologically-based approaches for pest management are sorely needed and one approach is to use genetically engineered insects. Herein we describe a series of integrated field, laboratory and modeling studies with the diamondback moth, Plutella xylostella, a serious global pest of crucifers. A “self-limiting” strain of Plutella xylostella (OX4319L), genetically engineered to allow the production of male-only cohorts of moths for field releases, was developed as a novel approach to protect crucifer crops. Wild-type females that mate with these self-limiting males will not produce viable female progeny. Our previous greenhouse studies demonstrated that releases of OX4319L males lead to suppression of the target pest population and dilution of insecticide-resistance genes. We report results of the first open-field release of a non-irradiated, genetically engineered self-limiting strain of an agricultural pest insect. In a series of mark-release-recapture field studies with co-releases of adult OX4319L males and wild-type counterparts, the dispersal, persistence and field survival of each strain were measured in a 2.83 ha cabbage field. Abstract A reconnaissance‐level regional resource accounting approach was applied to quantify benefits associated with an advanced quantitative precipitation information (AQPI) system being developed for the San Francisco Bay area. The AQPI system would provide incrementally higher resolution monitoring of rainfall events and longer lead‐time forecasts compared to current practice. AQPI system costs were estimated to have initial costs of $66 M and $3.3 M annual operation and maintenance; these translate to a present value cost (at 10 years and 6%) of $90 M. Benefits were associated with avoided flood damages, increasing water supplies, and enhancement of ecological, recreational, and transportation services. Total incremental benefits are estimated to be $61 M/year. Taken by category about 48% of the benefits are for flood damage mitigation ($29 M/year), with water supply (32%, $19.5 M/year), ecosystem (3.3%, $2 M/year), recreation (7.6%, $4.6 M/year), and transportation (9.5%, $5.8 M/year). These translate to a present value benefit of $449 M which computes to a base‐level B/C ratio of 5 to 1. Sensitivity analysis indicates a range of B/C up to 10 and down to 2. Given that most of the benefits are dependent on appropriate actions by hazards and water management agencies and citizens, then the AQPI project development involves outreach and training to maximize responses.

Background Despite increased access to treatment and reduced incidence, vertical transmission of HIV continues to pose a risk to maternal and child health in sub-Saharan Africa. Performance-based financing (PBF) directed at healthcare providers has shown potential to improve quantity and quality of maternal and child health services. However, the ways in which these PBF initiatives lead to improved service delivery are still under investigation. Methods Therefore, we implemented a longitudinal-controlled proof-of-concept PBF intervention at health facilities and with community-based associations focused on preventing vertical transmission of HIV (PVT) in rural Mozambique. We hypothesized that PBF would increase worker motivation and other aspects of the workplace environment in order to achieve service delivery goals. In this paper, we present two objectives from the PBF intervention with public health facilities (n = 6): first, we describe the implementation of the PBF intervention and second, we assess the impact of the PBF on health worker motivation, key factors in the workplace environment, health worker satisfaction, and thoughts of leaving. Implementation (objective 1) was evaluated through quantitative service delivery data and multiple forms of qualitative data (eg, quarterly meetings, participant observation (n = 120), exit interviews (n = 11)). The impact of PBF on intermediary constructs (objective 2) was evaluated using these qualitative data and quantitative surveys of health workers (n = 83) at intervention baseline, midline, and endline. Results We found that implementation was challenged by administrative barriers, delayed disbursement of incentives, and poor timing of evaluation relative to incentive disbursement (objective 1). Although we did not find an impact on the motivation constructs measured, PBF increased collegial support and worker empowerment, and, in a time of transitioning implementing partners, decreased against desire to leave (objective 2). Conclusion Areas for future research include incentivizing meaningful quality- and process-based performance indicators and evaluating how PBF affects the pathway to service delivery, including interactions between motivation and workplace environment factors.

Compound flooding, resulting from a combination of riverine and coastal processes, is a complex but important hazard to resolve along urbanized shorelines in the vicinity of river mouths. However, inland flooding models rarely consider oceanographic conditions, and vice versa for coastal flood models. Here, we describe the development of an operational, integrated coastal-watershed flooding model to address this issue of compound flooding in a highly urbanized estuarine environment, San Francisco Bay (CA, USA), where the surrounding communities are susceptible to flooding along the bay shoreline and inland rivers and creeks that drain to the bay. The integrated tributary-coastal forecast model (Hydro-Coastal Storm Modeling System, or Hydro-CoSMoS) was developed to provide water managers and other users with flood forecast information beyond what is currently available. Results presented here are focused on the interaction of the Napa River watershed and the San Pablo Bay at the northern end of San Francisco Bay. This paper describes the modeling setup, the scenario used in a tabletop exercise (TTE), and the assessment of the various flood forecast information products. Hydro-CoSMoS successfully demonstrated the capability to provide watershed and coastal flood information at scales and locations where no such information is currently available and was also successful in showing how tributary flows could be used to inform the coastal storm model during a flooding scenario. The TTE provided valuable feedback on how to guide continued model development and to inform what model outputs and formats are most useful to end-users.

BACKGROUND:Clinical reasoning is an important topic in healthcare training, assessment, and research. Virtual patients (VPs) are a safe environment to teach, assess and perform research on clinical reasoning and diagnostic accuracy. Our aim was to explore the details of the clinical reasoning process and diagnostic accuracy of undergraduate medical students when working with VPs using a concept mapping tool. METHODS:Over seven months we provided access to 67 German and 30 English VPs combined with a concept mapping tool to visualize and measure the clinical reasoning process of identifying problems, differential diagnoses, recommended tests and treatment options, and composing a summary statement about a VP. A final diagnosis had to be submitted by the learners in order to conclude the VP scenario. Learners were allowed multiple attempts or could request the correct diagnosis from the system. RESULTS:We analyzed 1,393 completed concept maps from 317 learners. We found significant differences between maps with a correct final diagnosis on one or multiple attempts and maps in which learners gave up and requested the solution from the system. These maps had lower scores, fewer summary statements, and fewer problems, differential diagnoses, tests, and treatments. CONCLUSIONS:The different use patterns and scores between learners who had the correct final diagnosis on one or multiple attempts and those who gave up, indicate that diagnostic accuracy in the form of a correct final diagnosis on the first attempt has to be reconsidered as a sole indicator for clinical reasoning competency. For the training, assessment, and research of clinical reasoning we suggest focusing more on the details of the process to reach a correct diagnosis, rather than whether it was made in the first attempt.

Soil moisture retention (SMR) capacity plays a key role in estimating the direct runoff when a multi-pulse storm event occurs. It is very important to know how much SMR will be recovered during the intervals of no rain of a multi-pulse storm. This study developed a new approach for derivation of the SMR recovery curve (R-curve) at sub-daily time-scales using the Curve Number (CN) method. The methodology was applied using complex storm events in the Napa River basin, California. The R-curve is classified into three sections depending on the recovery rate of SMR during the inter-storm interval of no rain (INR), and this study defines the characteristics. The first section of the R-curve (INR 0–21 h with 0.97 mm/h) is described as gradually recovering SMR, since water is being infiltrated and the upper soil layer is not fully saturated. The second section (INR 21–36 h with 2.11 mm/h) is defined as steeply recovering S due to downward drainage (sub-surface/inter flows) and evaporation without infiltration. The third section (INR 36–68 h with 0.34 mm/h) is described as gradually decreasing recovery dependent on evaporation since percolation and drainage have almost stopped.
