Numerous methods now exist for plant breeders and others to estimate genetic relationships among different germplasm. Two of the most commonly used methods have been restriction fragment length polymorphism (RFLP) and pedigrees. The objective of this study was to compare a relatively new method, DNA amplification fingerprinting (DAF), which is a short, arbitrary-primer, polymerase chain reaction (PCR)-amplification method, to RFLP and pedigree methods for genetic distance estimation among soybean [Glycine max (L.) Merr.] genotypes. The genotypes consisted of agronomically important soybean lines Archer, BSR101, Forrest, Hartwig, 9171, 9341, 9392, 9593, S8330 and the plant introduction, PI437.654. The 10 genotypes were evaluated by means of 53 RFLP clones, a subset of 12 RFLP clones, seven DAF primers, and available pedigrees. The DAF data set and the subset of 12 RFLP clones each consisted of 18 markers. For each method, the data were used to calculate the 45 pairwise genetic-distance estimates among the varieties using the average linkage algorithm in SAS. The genetic-distance estimates were correlated from 0.63 to 0.89 among the four methods. Dendrograms based on each method were therefore considered similarly reliable. For varietal Identification, the DAF and RFLP markers distinguished the varieties with similar efficiency. We conclude that both DAF and RFLP marker technologies and pedigrees are useful tools for estimating genetic distances in soybean., The ability to define genetically crop cultivars or breeding lines is useful for maintaining genetic purity, identifying proprietary germplasm, and estimating genetic relationships. Elementary school teachers (N=222) in Hong Kong completed Conners Teacher's Rating Scale (CTRS) for total of 874 students. Two months later, teachers decided whether students had behavioral or emotional problems requiring referral, considering 55 students as requiring referral. Purpose: To compare the refractive outcome of eyes that underwent phacoemulsification combined with endoscopic cyclophotocoagulation (phaco-ECP) with the eyes that underwent phacoemulsification alone. Methods: A retrospective case series review of 103 consecutive eyes that underwent phaco-ECP and 62 eyes that underwent phacoemulsification between January 2006 and August 2012 was conducted. Post-operative refractive outcomes were obtained by subjective refraction or autorefraction. Demographic data, best corrected visual acuity, intraocular pressure, number of glaucoma agents and mean deviation of the Humphrey visual field test (MD) were also analysed. Results: The total number of eyes included in the study was 62 eyes for the phaco-ECP group and 62 eyes for the control group who underwent phacoemulsification alone. More than 90% of eyes ( n = 56) in phaco-ECP group achieved post-operative refraction within ±1 dioptre (D) of the target refraction. In the control group, 100% of eyes achieved post-operative refraction within ±1D of the target refraction. When comparing the difference between the target and the actual refractive outcome between the phaco-ECP and the control group, there was no statistically significant difference. Conclusions: Refractive outcome after phaco-ECP is comparable to phacoemulsification alone. This study suggests that the intraocular lens power can be selected for cataract surgery alone and that ECP does not change the effective lens position significantly; therefore, no modification of biometry formulae is required. The optical spin Hall effect is a transport phenomenon of exciton polaritons in semiconductor microcavities, caused by the polaritonic spin-orbit interaction, which leads to the formation of spin textures. The control of the optical spin Hall effect via light injection in a double microcavity is demonstrated. Angular rotations of the polarization pattern up to 22° are observed and compared to a simple theoretical model. The device geometry is responsible for the existence of two polariton branches which allows a robust independent control of the polariton spin and hence the polarization state of the emitted light field, a solution technologically relevant for future spin-optronic devices. Background: Photodynamic therapy (PDT) is a promising adjuvant therapy in cancer treatment. However, cancers resistant to PDT, mediated through the efflux of photosensitisers by means of P-glycoprotein or ATP-binding cassette transporter proteins, have been reported. The DNA repair has also been suggested to be responsible for PDT resistance, but little is known about the repair pathways and mechanisms involved. Therefore, this study aimed to investigate the possible function of six major DNA repair mechanisms in glioma cells resistant to Photofrin-mediated PDT (Ph-PDT).Methods: The U87 glioma cells relatively resistant to Ph-PDT were obtained by recovering the viable cells 3 h after PDT treatment. The mRNA and protein expression levels of DNA repair genes were evaluated by quantitative real-time reverse transcription-polymerase chain reaction and western blotting, respectively. Small-interfering RNA and chromatin-immunoprecipitation assays were used to further examine the relationship between AlkB, an alkylation repair homologue 2 (Escherichia coli) (ALKBH2) and Ph-PDT responsiveness, and transcription factors involved in ALKBH2 transcription.Results: The ALKBH2 of DNA damage reversal was significantly increased at both mRNA and protein levels from 30 min to 48 h post-treatment with Ph-PDT. Conversely, down-regulating ALKBH2 expression enhances Ph-PDT efficiency. Furthermore, our data clearly show for the first time that tumour protein (TP53) is directly involved by binding to the promoter of ALKBH2 in mediating Ph-PDT resistance.Conclusion: C The DNA damage reversal mechanisms may have important functions in Ph-PDT resistance through the activation of ALKBH2 by TP53. γ-Tocotrienol (γT3) is known to selectively kill prostate cancer (PCa) cells and to sensitize the cells to docetaxel (DTX)-induced apoptosis. In the present study, the pharmacokinetics of γT3 and the in vivo cytotoxic response of androgen-independent prostate cancer (AIPCa) tumor following γT3 treatment were investigated. Here, we investigated these antitumor effects for PCa tumors in vivo. The pharmacokinetic and tissue distribution of γT3 after exogenous γT3 supplementation were examined. Meanwhile, the response of the tumor to γT3 alone or in combination with DTX were studied by real-time in vivo bioluminescent imaging and by examination of biomarkers associated with cell proliferation and apoptosis. After intraperitoneal injection, γT3 rapidly disappeared from the serum and was selectively deposited in the AIPCa tumor cells. Administration of γT3 alone for 2 weeks resulted in a significant shrinkage of the AIPCa tumors. Meanwhile, further inhibition of the AIPCa tumor growth was achieved by combined treatment of γT3 and DTX (p < 0.002). The in vivo cytotoxic antitumor effects induced by γT3 seem to be associated with a decrease in expression of cell proliferation markers (proliferating cell nuclear antigen, Ki-67 and Id1) and an increase in the rate of cancer cell apoptosis [cleaved caspase 3 and poly(ADP-ribose) polymerase]. Additionally, the combined agents may be more effective at suppressing the invasiveness of AIPCa. Overall, our results indicate that γT3, either alone or in combination with DTX, may provide a treatment strategy that can improve therapeutic efficacy against AIPCa while reducing the toxicity often seen in patients treated with DTX. Objective. To introduce the progress in genetic studies in attention-deficit hyperactivity disorder with particular emphasis on findings in Han Chinese. Methods. The literature review included peer-reviewed articles on Medline, the China National Knowledge Infrastructure database, and references since 1980 were hand-picked. Results. Several candidate genes have a substantial role in the aetiology of attention-deficit hyperactivity disorder. Possible confounding influences include: population admixture, environment and its interactions with genetic factors, diagnostic criteria, ethnicity, and genotype. Conclusions. Diversity of the relationships of genes with attention-deficit hyperactivity disorder exists across different ethnic groups. Hong Kong is an Asian city with nearly 7 million people. Because of the lack of natural resources, the local water supply is not adequate to meet the demand. Fresh water supply in Hong Kong relies on the cross border impart of water from the East River (Dongjiang) in China. To conserve fresh water, seawater has been used for toilet flushing since the 1950s. In this paper, the current status of seawater for toilet flushing in Hong Kong is reviewed, and the future strategy toward sustainable toilet flushing water supply, including other alternatives, is discussed. Hepatocellular carcinoma is a major health problem worldwide. Different treatment strategies have been developed to cope with this problem. Herbal medicine is now widely studied in both Eastern and Western countries. In this study, we used both in vitro and in vivo model to illustrate the anti-tumor effect of a product, CKBM, consisting of herbal medicine and specially processed Saccharomyces cerevisiae. Dose-dependent anti-proliferation effect was observed on in vitro growth of human hepatoma HepG2 cells after 48 hours incubation with CKBM. At the 50% inhibitory concentration(IC50) no significant toxic effect was observed on normal human fibroblasts Hs68 and human liver WRL-68 cells. The results of morphological changes, detection of DNA fragmentation, flow cytometric analysis and Western blot analysis indicated that this anti-tumor effect of CKBM was mediated via the process of apoptosis. In addition, HepG2 cells- bearing nude mice model was used for in vivo anti-tumor study. Our results showed that 14-day treatment with 0.8 ml daily dosage of CKBM could inhibit 54.1% of tumor growth. The plasma activities of enzymes specific for heart and liver, namely creatine kinase, lactate dehydrogenase, aspartate transaminase and alanine transaminase, remained at normal levels, indicated that CKBM did not produce toxicity to the host. Unlabelled: To determine the optimal method of suprapubic aspiration (SPA), the success rates of real-time ultrasound-guided SPA were compared with those of conventional SPA, and factors associated with success were studied. Thirty infants were randomly allocated to group A (for real-time ultrasound-guided SPA) and 30 infants to group B (for blind SPA with a prehydration protocol). The results showed that the overall success rates for all attempts were similar (26/30 or 87% in group A vs 24/30 or 80% in group B, p > 0.05). The first attempts in both groups were equally successful (both 18/30 or 60%). In comparison with failed attempts, successful ultrasound SPA attempts were associated with a greater bladder depth (mean +/- SD: 28 +/- 11 vs 21 +/- 5 mm, p < 0.01), length (32 +/- 12 vs 23 +/- 9 mm, p < 0.05) and volume (17 +/- 13 vs 8 +/- 6 ml, p < 0.01), but similar width (33 +/- 9 vs 29 +/- 5 mm, p > 0.05). In blind SPA, successful attempts were associated with the presence of bladder dullness on percussion (odds ratio 29).Conclusion: This study confirms that ultrasound-guided SPA has a high success rate. Blind SPA could also be equally successful with appropriate preparation. Ultrasound-guided SPA is recommended when the bladder depth exceeds 3 cm, or the bladder length exceeds 3.7 cm. If an ultrasound machine is not available, blind SPA may be an alternative, with attention being paid to prehydration and the demonstration of bladder dullness by percussion. [ABSTRACT FROM AUTHOR]

Many studies have demonstrated the short-term efficacy of stimulant drugs in decreasing the symptoms of Attention Deficit Hyperactivity Disorder (ADHD). However, long-term studies have not shown a similar effect. In this study, we report an audit of the medium-term outcome of 50 children with ADHD referred to a mental health service and treated with stimulants for at least 6 months. A poor outcome group was identified consisting of 30% of the children. As a group, they received a significantly higher dose of stimulant. More studies, with prospective design, are required to examine the medium-term outcome of children with ADHD on medication. [ABSTRACT FROM AUTHOR]

Three groups of hyperactive children (clinic observed, reported and situational) and a normal control group were compared on measures of behavioural, developmental and cognitive functioning. The clinic observed and reported groups did not differ significantly on any of the dependent variables. Situationally hyperactive children were less active, and had fewer behavioural and social difficulties than children with pervasive hyperactivity. Normal control subjects were distinguished by their better academic and social performance, and the absence of behaviour problems, language delay and hearing concerns. The data support the distinction between pervasive and situational hyperactivity, but not the separation of pervasive hyperactivity into clinic observable and reported hyperactivity. In a two-stage community study of 3069 Chinese schoolboys in Hong Kong, those entering stage 2 were classified by scores on Rutter's teacher and parent questionnaires into: (1) a mixed hyperactive/conduct-disturbed (HA-CD) group; (2) a pure hyperactive (HA) group; (3) a pure conduct-disturbed (CD) group; and (4) a normal control group. The four groups of children were compared on a series of psychosocial, cognitive and neurodevelopmental measures. This was followed by a regression analysis to examine the specificity of the differential patterns of associations between HA and CD. There was a mix of negative and positive findings defying a simple, definitive conclusion. However, the positive findings that did emerge supported a growing body of recent literature which favoured a separation of HA from CD and their cross-cultural validity: the former was associated with neurodevelopmental impairments, the latter with family disharmony. - Sixty-one boys, 6-12 years old, met both the diagnostic criteria of ADHD of DSM-III-R and hyperkinetic syndrome of ICD-9. They could be subdivided into two groups: 43 showed hyperactive symptoms in the clinic situation and 18 did not. The former group were younger and showed significantly poorer performance in measures of general intelligence, attention, cognitive styles, motor clumsiness and minor neurological deficits. Controlling for age and intelligence attenuated the differences, but significant differences remained in activity level, attention, and minor neurological deficits. The characteristics of 75 children referred to psychiatrists by medical practitioners in Hong Kong were compared with 56 children who were referred by their families to a community clinic. Their demographic characteristics, and the nature and severity of their illness, were quite similar. Medical practitioners tended to refer children with childhood psychosis and psychophysiological disorder. The reasons leading to different types of referral are discussed. A community child mental health clinic may be a useful model for providing psychiatric care to children in Hong Kong. [ABSTRACT FROM AUTHOR]
