1. Environmental representativeness and the role of emitter and recipient areas in the future trajectory of a protected area under climate change

Abstract

We propose a protocol to estimate the effects of climate change on species inhabiting a reserve by assessing the location of areas with similar environmental conditions to a focal protected area, both now and in the future. Following this protocol it is possible to estimate: (i) the level of change that will occur in the current climatic conditions of a reserve; (ii) the present location of the areas with similar conditions to those this reserve will have in the future (emitter areas); and (iii) the location of the areas that in the future will have similar environmental conditions to those existing in the studied protected area (recipient areas). This knowledge can be used to anticipate and adapt the protected area against future changes. In this study, we used an Iberian reserve representative of the Mediterranean conditions, the Cabañeros National Park, as an example to calculate the extension, fragmentation and location of the areas with climatic conditions similar to those of the reserve. We also determined the connectivity between these areas and their degree of anthropic alteration.

Abstract

The lack of information about the distribution of threatened species inhibits the development of strategies for their conservation. This is a particularly important problem when considering invertebrates. Here we evaluate the effects of species’ traits and data characteristics on the accuracy of species distribution models (SDM) of 20 threatened Iberian invertebrates. We found that the accuracy of the predictions was mostly affected by the characteristics of the data. Species whose distributions were most accurately modelled were those with a greater sample size or smaller relative occurrence area (ROA). Species in habitats that were difficult to detect using GIS data, such as riparian species, tended to be more difficult to predict.

Abstract

We analyzed the altitudinal change in dung beetle species richness and the relative proportion of higher taxa, as well as the turnover in the type of distribution and range size of species in two mountain chains located at the two extremes of Europe (Western Rhodopes Mountains and the Iberian Central System). Both mountain ranges showed a clear substitution among higher taxa (Aphodiinae-Geotrupinae vs. Scarabaeidae) and species richness variation with the altitude was similar. We suggest that East European dung beetle assemblages are conditioned by a horizontal colonization process in which mountains had been reached in relatively recent geological time by elements coming from different latitudes. In spite of these convergences, Rhodopes dung beetle assemblages are characterized by a significantly lower proportion of narrowly distributed species and a lower relevance of Aphodiinae species in lowland places. Although these divergences can be partially attributed to the dissimilar sampling effort accomplished in both regions, we suggest that the low number on narrowly distributed species could be due to the different role of these two mountain zones as refuges during glaciar-interglaciar Pleistocene cycles.

Abstract

Patterns of habitat preference and distribution and abundance of wintering bird fauna in central Spain. Analysis and prediction of the effect of ecological factors This paper analyses the effect of geographic, topographic, land use and habitat structure variables on the composition and structure of wintering bird communities in Central Spain (Iberian peninsula). Parameters describing the avifauna varied in a predictable way considering a small group of coarse-grained variables defining the geographical and altitudinal location of the censuses, and the basic characteristics of the structure and typology of habitats: 49-76% of variance accounted for total bird density and for abundance of four ecological groups, 37-63% accounted for species richness and diversity, and 65% explained the relative abundance of species with conservation problems according to the European scale (SPEC figures). Regarding the most widespread species in the study area, significant models were obtained by means of tree regression analysis for 50 species, with an average reduction of deviance of 39%. Altitude was the most important variable affecting bird community parameters and abundance of each bird species, showing a consistent and marked negative effect. Structural complexity of the vegetation and geographical location followed as the variables of importance explaining variability. The habitats with the lowest bird density, richness and diversity of birds were mountain grasslands/shrublands, young pine re-forestations, and Pyrenean oak forests at 1,200-1,600 m a.s.l. The habitats with the highest values on these parameters were riparian forests, agricultural mosaics, and holmoak “dehesa” parklands, mainly located at the southern and western part of the region and at intermediate altitudes. The total density of birds increased from east to west, was higher in intermediate altitudes than in the extremes of the altitudinal range, and increased with habitat structural complexity (i.e., vertical development and degree of vegetation cover), agricultural use of the land, and the presence of water (e.g., streams, rivers, flooded areas). Density of strictly wintering species in the study region decreased latitudinally from southeast to northwest, being higher at intermediate altitudes in localities with presence of water and woodlands dominated by coniferous trees. Abundance of facultative or obligate frugivorous species was very low. Richness of species was higher toward the western part of the study area, increased with habitat structural complexity, and was lower at higher altitudes. On the other hand, the less diverse bird assemblages were those that inhabit agricultural landscapes and/or areas located at higher altitudes. Habitats and areas with a higher relative abundance of species with conservation problems at the European scale (SPEC scores) were located at intermediate altitudes in the southwest and southeast of the study region. The relative abundance of this group of species was also associated to the presence of water, habitat structural complexity and agricultural use. The relationship between the European conservation status of species (SPEC scores) and the patterns of distribution, abundance, habitat preferences and ecological width of 72 species was also analysed. Species with more conservation problems on the European scale have a marked preference for structurally simpler habitats (e.g., agricultural and grasslands habitats) and have a broader altitudinal and among-habitats distribution. Results from the 44 census localities were extrapolated to the remaining region using a geographical information system in order to build predictive maps for density, species richness, species diversity and weighed European conservation status. This work shows that valuable knowledge can be obtained from fragmentary and dispersed data, in order to describe general patterns of distribution, abundance and habitat preferences of birds. This methodological approach could be a valid in environmentally heterogeneous, large regions, with few qualified bird observers and researchers.

Abstract

Introduction: Inborn errors of immunity (IEI) are an expanding group of rare diseases whose field has been boosted by next-generation sequencing (NGS), revealing several new entities, accelerating routine diagnoses, expanding the number of atypical presentations and generating uncertainties regarding the pathogenic relevance of several novel variants. Methods: Research laboratories that diagnose and provide support for IEI require accurate, reproducible and sustainable phenotypic, cellular and molecular functional assays to explore the pathogenic consequences of human leukocyte gene variants and contribute to their assessment. We have implemented a set of advanced flow cytometry-based assays to better dissect human B-cell biology in a translational research laboratory. We illustrate the utility of these techniques for the in-depth characterization of a novel (c.1685G>A, p.R562Q) de novo gene variant predicted as probably pathogenic but with no previous insights into the protein and cellular effects, located in the tyrosine kinase domain of the Bruton’s tyrosine kinase (BTK) gene, in an apparently healthy 14-year-old male patient referred to our clinic for an incidental finding of low immunoglobulin (Ig) M levels with no history of recurrent infections. Results and discussion: A phenotypic analysis of bone marrow (BM) revealed a slightly high percentage of pre-B-I subset in BM, with no blockage at this stage, as typically observed in classical X-linked agammaglobulinemia (XLA) patients. The phenotypic analysis in peripheral blood also revealed reduced absolute numbers of B cells, all pre-germinal center maturation stages, together with reduced but detectable numbers of different memory and plasma cell isotypes. The R562Q variant allows Btk expression and normal activation of anti-IgMinduced phosphorylation of Y551 but diminished autophosphorylation at Y223 after anti IgM and CXCL12 stimulation. Lastly, we explored the potential impact of the variant protein for downstream Btk signaling in B cells. Within the canonical nuclear factor kappa B (NF-kB) activation pathway, normal IkBa degradation occurs after CD40L stimulation in patient and control cells. In contrast, disturbed IkBa degradation and reduced calcium ion (Ca2+) influx occurs on anti-IgM stimulation in the patient’s B cells, suggesting an enzymatic impairment of the mutated tyrosine kinase domain. Abstract

Edoxaban is used for venous thromboembolism (VTE) treatment. Real-life data are lacking about its use in long-term therapy. We aimed to assess the efficacy and the safety of edoxaban for long-term VTE treatment in a real-life setting. Patients with VTE included in the Registro Informatizado Enfermedad TromboEmbólica (RIETE) registry, receiving edoxaban 60 or 30 mg daily were prospectively followed up to validate the benefit of using different dosages. The main outcome was the composite of VTE recurrences or major bleeding in patients with or without criteria for dose reduction. Multivariable analysis to identify predictors for the composite outcome was performed. From October 2015 to November 2019, 562 patients received edoxaban for long-term therapy. Most (94%) of the 416 patients not meeting criteria for dose reduction received 60 mg daily, and 92 patients meeting criteria (63%) received 30 mg daily. During treatment, two patients developed recurrent VTE, six had major bleeding and nine died (2 from fatal bleeding). Among patients not meeting criteria for dose reduction, those receiving 30 mg daily had a higher rate of the composite event (hazard ratio (HR) 8.37; 95% confidence interval (CI) 1.12–42.4) and a significant higher mortality rate (HR 31.1; 95% CI 4.63–262) than those receiving 60 mg. Among patients meeting criteria for dose reduction, those receiving 60 mg daily had no events, and a nonsignificantly higher mortality rate (HR 5.04; 95% CI 0.54–133) than those receiving 30 mg daily. In conclusion, edoxaban seems to be effective and safe for long-term VTE treatment in real life. Criteria for dose reduction should be reformulated., No funding was received for this work

Abstract

The association between elevated liver enzymes or FIB-4 (fibrosis index 4) and outcome in patients with venous thromboembolism (VTE) has not been evaluated. Data from patients in RIETE (Registro Informatizado Enfermedad TromboEmbólica) were used to assess the association between elevated liver enzymes or FIB-4 levels and the rates of major bleeding or death in apparent liver disease-free patients with acute VTE under anticoagulation therapy. A total of 6206 patients with acute VTE and without liver disease were included. Of them, 92 patients had major bleeding and 168 died under anticoagulation therapy. On multivariable analysis, patients with elevated liver enzymes were at increased mortality risk (HR: 1.58; 95% CI: 1.10–2.28), while those with FIB-4 levels > 2.67 points were at increased risk for major bleeding (HR: 1.69; 95% CI: 1.04–2.74). Evaluation of liver enzymes and FIB-4 index at baseline in liver disease-free patients with VTE may provide additional information on the risk for major bleeding or death during anticoagulation. Abstract

In the current era of patient empowerment and precision medicine, access to timely information is critical to decision-making. Unfortunately, we currently lack patient-specific, real-time data about clinical presentation, risk of thrombotic or hemorrhagic events, key risk factors, and adverse outcomes in patients with venous thromboembolism (VTE). Accordingly, the Registro Informatizado Enfermedad TromboEmbólica (RIETE) investigators developed a tool to provide an open-source, real-time graphic representation of VTE-related data derived from over 90 000 patients with confirmed VTE. This information is intended to facilitate discussion in the informed decision-making process. The current article describes the aims, rationale, methods, and ongoing and future efforts of the real-time VTE infographics developed by the RIETE registry collaborators.

Abstract

The EUCAST EDef 9.3.2 procedure recommends visual readings of azole and amphotericin B MICs against Aspergillus spp. Visual determination of MICs may be challenging. In this work, we aim to obtain and compare visual and spectrophotometric MIC readings of azoles and amphotericin B against Aspergillus fumigatus sensu lato isolates. A total of 847 A. fumigatus sensu lato isolates (A. fumigatus sensu stricto [n = 828] and cryptic species [n = 19]) were tested against amphotericin B, itraconazole, voriconazole, posaconazole, and isavuconazole using the EUCAST EDef 9.3.2 procedure. Isolates were classified as susceptible or resistant/non-wild type according to the 2020 updated breakpoints. The area of technical uncertainty for the azoles was defined in the updated breakpoints. Visual and spectrophotometric (fungal growth reduction of >95% compared to the control, read at 540 nm) MICs were compared. Essential (±1 2-fold dilution) and categorical agreements were calculated. Overall, high essential (97.1%) and categorical (99.6%) agreements were found. We obtained 100% categorical agreements for amphotericin B, itraconazole, and posaconazole, and consequently, no errors were found. Categorical agreements were 98.7 and 99.3% for voriconazole and isavuconazole, respectively. Most of the misclassifications for voriconazole and isavuconazole were found to be associated with MIC results falling either in the area of technical uncertainty or within one 2-fold dilution above the breakpoint. The resistance rate was slightly lower when the MICs were obtained by spectrophotometric readings. However, all relevant cyp51A mutants were correctly classified as resistant. Spectrophotometric determination of azole and amphotericin B MICs against A. fumigatus sensu lato isolates may be a convenient alternative to visual endpoint readings.

Abstract

R-(+)-limonene epoxidation has been focused from different synthesis methods. Thanks to the optically active epoxides obtained from this reaction are widely employed at industrial level, like: agrochemicals, polymers, cosmetics, pharmaceuticals. A key point found in this topic has been the asymmetric induction to improve the yield to the diastereomers of 1,2-limonene oxide. Many catalysts have been developed for the diastereoselective epoxidation, but some oxidation methodology have not been so useful. The enzymes and the Jacobsen's catalyst presented the highest selectivity towards the endocyclic epoxides diastereomers, while other catalysts like metals supported on mesoporous materials were directed to one or more oxidation products, reducing their potential industrial scaling. Also, it was evidenced that controlling the reaction parameters it allows the segregation homogeneous catalyst to a phase different from the reaction products, thus increasing its useful reutilization in several reaction cycles. This minireview confronts the different systems used for the diastereoselective epoxidation of R-(+)-limonene. Challenges, issues and trends of said chemical transformation are also discussed. Abstract

In this paper, we shall apply symmetry analysis to second order functional differential equations with constant coefficients. The determining equations of the admitted Lie group are constructed in a manner different from that of the existing literature for delay differential equations. We define the standard Lie bracket and make a complete classification of the second order linear functional differential equations with constant coefficients, to solvable Lie algebras. We also classify some second order nonlinear functional differential equations with constant coefficients, to solvable Lie algebras. [ABSTRACT FROM AUTHOR]
