For the purpose of mastering the influence of the strip-shaped air-out slits’ width in the fully mechanized working face under wallattached swirling ventilation condition, By using fluent software, numerical simulation of airflow field and dust diffusion in fully mechanized working face under different strip-shaped air-out slits’ width conditions was carried out. The results show that the wall-attached swirling ventilation can effectively control the dust diffusion in the fully mechanized working face, improve the dust collecting effect of the bag precipitator fan, and reduce the particle mass concentration at the driver and the rear of the tunnel; Increasing the air volume of the strip-shaped airout slits can improve the control effect of the swirling air curtain on the dust, appropriate air volume can make the swirling air curtain control the dusty airflow and push it to the end of the heading surface to control, block and propel the dust; When the width of the strip-shaped air-out slits is 0.06 m, the dust collecting effect of the exhaust air cylinder on the dust and the effect of the wall-attached swirling on the plugging of the dust are best.

Spiral channel pressure nozzles are commonly used pressure nozzles in practical workplaces. In this paper, two kinds of spiral channel type pressure nozzles, namely, spiral hole type and spiral non-porous type, the atomization characteristics and dust reduction efficiency under different spray pressures are discussed and compared. Based on the experimental method, based on the self-designed spray dust-reducing roadway experimental platform, the macro-atomization characteristics of the two nozzles, namely the flow rate, the atomization angle, the range, and the droplet size, were measured. The following conclusions were drawn: (1) The flow rates of both nozzles increase with increasing spray pressure, and the flow coefficient of the spiral non-porous nozzle is small. (2) The change of the atomization angle of the two nozzles first increases and then decreases with the increase of the spray pressure, and the atomization angle of the spiral non-porous nozzle is larger. At the same time, the range of the two nozzles gradually increases as the spray pressure increases, and the range of the spiral perforated nozzle is always larger than that of the spiral non-porous nozzle. (3) When the spray pressure is gradually increased, the droplet size of the two nozzles selected in the experiment is gradually reduced, and the droplet size of the spiral perforated nozzle is always larger than that of the spiral non-porous nozzle before 5 MPa, and then gradually Become smaller. The main reason why the droplet size decreases with the increase of the spray pressure is that the increase of the spray pressure leads to an increase in the spray speed of the water droplets, so that the water droplets are completely split when they are ejected from the nozzle, resulting in a smaller droplet size. In summary, when the spray pressure required in the actual working environment is low, the use of a spiral non-porous nozzle is more conducive to dust reduction.

During the spin-torque ferromagnetic resonance (ST-FMR) measurement, the magnetization precession driven by the microwave field yields the radio frequency (rf) oscillating magnetoresistance and its time-averaged change (photoresistance). Here, we find that the strength of photoresistance can be directly determined by using dc bias current Idc modulating the symmetric component VS of the ST-FMR voltage spectrum. By measuring the angular dependence of photoresistance, we can quantify the in-plane and out-of-plane precession angles of ST-FMR, the actual rf current distribution in the magnetic and non-magnetic sublayers, and the magnitude of spin-torque and various magnetoresistance coefficients. These experimentally obtained values and analysis methods can more accurately quantify the spin-torque efficiency of both in-plane and out-of-plane spin polarizations by self-consistent calculation of the precession angle without harsh assumptions. And, we further confirm this universal method in three spintronic systems: the prototypical Pt/Py bilayer with anisotropic magnetoresistance (AMR), Py/Cu/Co20Tb80 spin valve trilayer with AMR and giant magnetoresistance, and [Co/Ni]3/Co/Pt multilayer with AMR and anisotropic interface magnetoresistance. This method eliminates potential deviation in calculating spin-torque efficiency by previously reported line shape analyzation and linewidth modulation methods of the ST-FMR technique and significantly extends its application range in characterizing spintronic materials and nanodevices. Abstract Background Mesenchymal stem cells (MSCs) have been considered to be the attractive vehicles for delivering therapeutic agents toward various tumor diseases. This study was to explore the distribution pattern, kinetic delivery of adenovirus, and therapeutic efficacy of the MSC loading of E1A mutant conditionally replicative adenovirus Adv-Stat3(-) which selectively replicated and expressed high levels of anti-sense Stat3 complementary DNA in breast cancer and melanoma cells. Methods We assessed the release ability of conditionally replicative adenovirus (CRAd) from MSC using crystal violet staining, TCID50 assay, and quantitative PCR. In vitro killing competence of MSCs carrying Adv-Stat3(-) toward breast cancer and melanoma was performed using co-culture system of transwell plates. We examined tumor tropism of MSC by Prussian blue staining and immunofluorescence. In vivo killing competence of MSCs carrying Adv-Stat3(-) toward breast tumor was analyzed by comparison of tumor volumes and survival periods. Results Adv-Stat3(-) amplified in MSCs and were released 4 days after infection. MSCs carrying Adv-Stat3(-) caused viral amplification, depletion of Stat3 and its downstream proteins, and led to significant apoptosis in breast cancer and melanoma cell lines. In vivo experiments confirmed the preferential localization of MSCs in the tumor periphery 24 hours after tail vein injection, and this localization was mainly detected in the tumor parenchyma after 72 hours. Intravenous injection of MSCs carrying Adv-Stat3(-) suppressed the Stat3 pathway, down-regulated Ki67 expression, and recruited CD11b-positive cells in the local tumor, inhibiting tumor growth and increasing the survival of tumor-bearing mice. Conclusions These results indicate that MSCs migrate to the tumor site in a time-dependent manner and could be an effective platform for the targeted delivery of CRAd and the amplification of tumor killing effects.

Abstract Background P21(WAF1/Cip1) binds to cyclin-dependent kinase complexes and inhibits their activities. It was originally described as an inhibitor of cancer cell proliferation. However, many recent studies have shown that p21 promotes tumor progression when accumulated in the cell cytoplasm. So far, little is known about the correlation between cytoplasmic p21 and drug resistance. This study was aimed to investigate the role of p21 in the cisplatin resistance of ovarian cancer. Methods RT-PCR, western blot and immunofluorescence were used to detect p21 expression and location in cisplatin-resistant ovarian cancer cell line C13* and its parental line OV2008. Regulation of cytoplasmic p21 was performed through transfection of p21 siRNA, Akt2 shRNA and Akt2 constitutively active vector in the two cell lines; their effects on cisplatin-induced apoptosis were evaluated by flow cytometry. Tumor tissue sections of clinical samples were analyzed by immunohistochemistry. Results p21 predominantly localizes to the cytoplasm in C13* compared to OV2008. Persistent exposure to low dose cisplatin in OV2008 leads to p21 translocation from nuclear to cytoplasm, while it had not impact on p21 localization in C13*. Knockdown of cytoplasmic p21 by p21 siRNA transfection in C13* notably increased cisplatin-induced apoptosis through activation of caspase 3. Inhibition of p21 translocation into the cytoplasm by transfection of Akt2 shRNA into C13* cells significantly increased cisplatin-induced apoptosis, while induction of p21 translocation into the cytoplasm by transfection of constitutively active Akt2 in OV2008 enhanced the resistance to cisplatin. Immunohistochemical analysis of clinical ovarian tumor tissues demonstrated that cytoplasmic p21 was negatively correlated with the response to cisplatin based treatment. Conclusions Cytoplasmic p21 is a novel biomarker of cisplatin resistance and it may represent a potential therapeutic target for ovarian tumors that are refractory to conventional treatment.

Pulses, the dried seeds of leguminous plants, form an important part of the diets of many cultures, including Caribbean cuisine, and are a rich source of protein, carbohydrates, and antioxidants while being low in fats. This study examined the effect of a traditional home-cooking method on the nutritional characteristics of pulses commonly consumed in the Caribbean: red kidney beans and cranberry beans (Phaseolus vulgaris L.), cowpeas (Vigna unguiculata L.), and pigeon peas (Cajanus cajan L.). Protein quality, determined via three in vitro protein digestibility methods, starch, and phenolic content were determined in pre- and post-cooked samples using established methods. Pulses contained 20–26% protein, and cooking improved protein digestibility on average by 14.0 ± 2.5% (p < 0.05). However, notable differences in digestibility were observed: it was higher in static assays (pH-Drop and pH-Stat) than in the two-step digestibility assay. Average protein digestibility-corrected amino acid score (IVPDCAAS) among cooked pulses was 0.81 ± 0.14, with the highest in cranberry bean (0.82) and cowpea (0.88). Cooking modified pulse starch profiles by increasing total digestible starch. However, resistant starch and slowly digestible starch fractions accounted for approximately 20–25% of total cooked starch content. While total phenolic content (TPC) and antioxidant activity were reduced with cooking, they were within expected ranges for cooked pulse flours; however, they were higher in bean (P. vulgaris) varieties than cowpea and pigeon pea. These findings support the promotion of increased pulse consumption in Caribbean diets. Home cooking is a simple method to enhance pulse protein quality through enhancing digestibility; however, in vitro protein digestibility assays may require further standardization. Maintaining intracellular redox balance is essential for the survival, antibody secretion, and mucosal immune homeostasis of immunoglobulin A (IgA) antibody‐secreting cells (ASCs). However, the relationship between mitochondrial metabolic enzymes and the redox balance in ASCs has yet to be comprehensively studied. Our study unveils the pivotal role of mitochondrial enzyme PCK2 in regulating ASCs' redox balance and intestinal homeostasis. We discover that PCK2 loss, whether globally or in B cells, exacerbates dextran sodium sulphate (DSS)‐induced colitis due to increased IgA ASC cell death and diminished antibody production. Mechanistically, the absence of PCK2 diverts glutamine into the TCA cycle, leading to heightened TCA flux and excessive mitochondrial reactive oxygen species (mtROS) production. In addition, PCK2 loss reduces glutamine availability for glutathione (GSH) synthesis, resulting in a decrease of total glutathione level. The elevated mtROS and reduced GSH expose ASCs to overwhelming oxidative stress, culminating in cell apoptosis. Crucially, we found that the mitochondria‐targeted antioxidant Mitoquinone (Mito‐Q) can mitigate the detrimental effects of PCK2 deficiency in IgA ASCs, thereby alleviating colitis in mice. Cancerous genetic mutations result in a complex and comprehensive post-translational modification (PTM) dynamics, in which protein succinylation is well known for its ability to reprogram cell metabolism and is involved in the malignant evolution. Little is known about the regulatory interactions between succinylation and other PTMs in the PTM network. Here, we developed a conjoint analysis and systematic clustering method to explore the intermodification communications between succinylome and phosphorylome from eight lung cancer patients. We found that the intermodification coorperation in both parallel and series. Besides directly participating in metabolism pathways, some phosphosites out of mitochondria were identified as an upstream regulatory modification directing succinylome dynamics in cancer metabolism reprogramming. Phosphorylated activation of histone deacetylase (HDAC) in lung cancer resulted in the removal of acetylation and favored the occurrence of succinylation modification of mitochondrial proteins. These results suggest a tandem regulation between succinylation and phosphorylation in the PTM network and provide HDAC-related targets for intervening mitochondrial succinylation and cancer metabolism reprogramming.
