16. A survey in rural China of parent-absence through migrant working: the impact on their children's self-concept and loneliness

Abstract

Abstract Background Following the rapid increase of migrant workers in China, the number of "absent migrant parents" children is also rising fast. The "absent migrant parents" children might have an insecure relationship with their parents, have a different view of them, and be prone to have the feeling of loneliness. The purpose of the study was to compare the self-concept and loneliness between the "absent migrant parents" children and comparison children, to examine the relationship between self-concept and loneliness among the two groups, and to study the predictors of self-concept among the two groups. Methods Participants were 230 "absent migrant parents" children and 250 comparison children in the rural area of a county, China. The self-concept and loneliness of children were assessed using Piers-Harris Self-Concept Scale and Childhood Loneliness Scale. Results The "absent migrant parents" children were more likely to dislike their parents or be uncertain whether they like their parents, and they reported less time spent in physical and leisure time activities, higher loneliness and lower self-concept in comparison with the comparison children. Loneliness was significantly negatively correlated with all the dimensions of self-concept among the two groups. Regression analysis showed that self-concept was positively related to the relationship with parents and guardians and time spent in physical and leisure activities among the "absent migrant parents" children. The same factors (except the relationship with guardians) were found for self-concept among the comparison children. Conclusions The "absent migrant parents" children were more inclined to have lower self-concept and higher loneliness. The lower self-concept seemed to contribute to the higher loneliness of the "absent migrant parents" children. The lower self-concept of the "absent migrant parents" children was mainly related with their relationship with parents and guardians. The acceptance and support from their parents could not be fully replaced by that from their guardians.

30. Effect of Ce Addition on Microstructure, Thermal Conductivity, and Mechanical Properties of As-Cast and As-Extruded Mg–3Sn Alloys.

Abstract

In the present research, the impacts of Ce additions at various concentrations (0, 1.0, 3.4, and 4.0 wt.%) on the evolution of the microstructure, mechanical properties, and thermal conductivity of as-cast and as-extruded Mg-3Sn alloys were investigated. The findings demonstrate that adding Ce caused the creation of a new ternary MgSnCe phase in the magnesium matrix. Some new Mg17Ce2 phases are generated in the microstructure when Ce levels reach 4%. The thermal conductivity of the Mg-3Sn alloy is significantly improved due to Ce addition, and the Mg-3Sn-3.4Ce alloy exhibits the highest thermal conductivity, up to 133.8 W/(m·K) at 298 K. After extrusion, both the thermal conductivity and mechanical properties are further improved. The thermal conductivity perpendicular to the extrusion direction of Mg-3Sn-3.4Ce alloy could achieve 136.28 W/(m·K), and the tensile and yield strengths reach 264.3 MPa and 227.2 MPa, with an elongation of 7.9%. Adding Ce decreases the dissolved Sn atoms and breaks the eutectic α-Mg and Mg2Sn network organization, leading to a considerable increase in the thermal conductivity of as-cast Mg-3Sn alloys. Abstract

Current antiangiogenic therapy is limited by its cytostatic property, scarce drug delivery to the tumor, and side toxicity. To address these limitations, we unveiled the role of ZEB1, a tumor endothelium- enriched zinc-finger transcription factor, during tumor progression. We discovered that the patients who had lung adenocarcinomas with high ZEB1 expression in tumor endothelium had increased prevalence of metastases and markedly reduced overall survival after the diagnosis of lung cancer. Endothelial ZEB1 deletion in tumor-bearing mice diminished tumor angiogenesis while eliciting persistent tumor vascular normalization by epigenetically repressing TGF-[beta] signaling. This consequently led to improved blood and oxygen perfusion, enhanced chemotherapy delivery and immune effector cell infiltration, and reduced tumor growth and metastasis. Moreover, targeting vascular ZEB1 remarkably potentiated the anticancer activity of nontoxic low-dose cisplatin. Treatment with low-dose anti-programmed cell death protein 1 (anti-PD-1) antibody elicited tumor regression and markedly extended survival in ZEB1-deleted mice, conferring long-term protective anticancer immunity. Collectively, we demonstrated that inactivation of endothelial ZEB1 may offer alternative opportunities for cancer therapy with minimal side effects. 41. Blocking GPR15 Counteracts Integrin-dependent T Cell Gut Homing in Vivo.

Abstract

Background and Aims The G protein coupled receptor GPR15 is expressed on and functionally important for T cells homing to the large intestine. However, the precise mechanisms by which GPR15 controls gut homing have been unclear. Thus, we aimed to elucidate these mechanisms as well as to explore the potential of targeting GPR15 for interfering with T cell recruitment to the colon in inflammatory bowel disease [IBD]. Methods We used dynamic adhesion and transmigration assays, as well as a humanised in vivo model of intestinal cell trafficking, to study GPR15-dependent effects on gut homing. Moreover, we analysed GPR15 and integrin expression in patients with and without IBD, cross-sectionally and longitudinally. Results GPR15 controlled T cell adhesion to MAdCAM-1 and VCAM-1 upstream of α4β7 and α4β1 integrin, respectively. Consistently, high co-expression of these integrins with GPR15 was found on T cells from patients with IBD, and GPR15 also promoted T cell recruitment to the colon in humanised mice. Anti-GPR15 antibodies effectively blocked T cell gut homing in vitro and in vivo. In vitro data, as well as observations in a cohort of patients treated with vedolizumab, suggest that this might be more effective than inhibiting α4β7. Conclusions GPR15 seems to have a broad, but organ-selective, impact on T cell trafficking and is therefore a promising target for future therapy of IBD. Further studies are needed.
