This study presents an event-based storyline framework to assess the influence of future climatic and socioeconomic conditions on coastal flood impacts to critical infrastructure. The framework combines well-established quantitative methods of sea level rise, coastal inundation, and critical infrastructure (CI) physical damage assessments into an integrated modelling approach. We apply our approach to re-imagine three historic events: storm Xaver, storm Xynthia , and a storm surge event along the coast of Emilia Romagna (Italy). Our results indicate that northern Germany would benefit mostly from coordinated adaptation action to reduce the flood impact, whereas the southwestern coast of France would find the highest damage reduction through asset-level 'autonomous' adaptation action. Our approach helps to improve the scientific understanding of how coastal flood risk are assessed and best managed, and forces a distillation of the science into an accessible narrative to support policymakers and asset owners to make progress towards more climate-resilient coastal communities. We provide a time series of Agulhas leakage anomalies over the last 20-years from satellite altimetry. Until now, measuring the interannual variability of Indo-Atlantic exchange has been the major barrier in the investigation of the dynamics and large scale impact of Agulhas leakage. We compute the difference of transport between the Agulhas Current and Agulhas Return Current, which allows us to deduce Agulhas leakage. The main difficulty is to separate the Agulhas Return Current from the southern limb of the subtropical "supergyre" south of Africa. For this purpose, an algorithm that uses absolute dynamic topography data is developed. The algorithm is applied to a state-of-the-art ocean model. The comparison with a Lagrangian method to measure the leakage allows us to validate the new method. An important result is that it is possible to measure Agulhas leakage in this model using the velocity field along a section that crosses both the Agulhas Current and the Agulhas Return Current. In the model a good correlation is found between measuring leakage using the full depth velocities and using only the surface geostrophic velocities. This allows us to extend the method to along-track absolute dynamic topography from satellites. It is shown that the accuracy of the mean dynamic topography does not allow us to determine the mean leakage but that leakage anomalies can be accurately computed.

A 250 year simulation of a strongly eddying global version of the Parallel Ocean Program (POP) model reveals a new mode of intrinsic multidecadal variability, the Southern Ocean Mode (SOM), with a period of 40-50 year. The peak-to-peak difference in the global ocean heat content within a multidecadal cycle is up to 60 ZJ. This change results from surface heat flux variations in the South Atlantic and propagation of temperature anomalies along the Antarctic Circumpolar Current and into the Weddell gyre around 30°E. The temperature anomalies propagate as deep as 5000 m along the isopycnals between 50°S and 30°S and induce multidecadal changes in the Atlantic Meridional Overturning Circulation. A positive feedback loop between the generation of eddies through baroclinic instability and the dynamics of the mean circulation is essential for the existence of the SOM. The dominant physics appears similar to that responsible for variability found in a three-layer quasi-geostrophic eddy-resolving model. This combined with the fact that the SOM is not found in a noneddying version of the same global POP model further suggests that eddy processes are crucial for its existence and/or excitation.

21. Representing Greenland ice sheet freshwater fluxes in climate models

- Abstract
Here we present a long-term (1850–2200) best estimate of Greenland ice sheet (GrIS) freshwater runoff that improves spatial detail of runoff locations and temporal resolution. Ice discharge is taken from observations since 2000 and assumed constant in time. Surface meltwater runoff is retrieved from regional climate model output for the recent past and parameterized for the future based on significant correlations between runoff and midtropospheric (500 hPa) summer temperature changes over the GrIS. The simplicity of this approach enables assimilation of meltwater runoff into coupled climate models, which is demonstrated here in a case study with the medium-resolution (1?) Community Earth System Model. The model results suggest that the decrease in Atlantic Meridional Overturning Circulation (AMOC) is dominated by warming of the surface ocean and enhanced GrIS freshwater forcing leads to a slightly enhanced (?1.2 sverdrup in the 21st century) weakening of the AMOC.

International audience; Serotonin and its various receptors are involved in numerous brain functions and neuropsychiatric disorders. The 5-HT1A family is the best characterized subtype of the fourteen currently known 5-HT receptors. The 5-HT1A receptor is closely involved in the pathogenesis of anxiety, depression, epilepsy and eating disorders and therefore is an important target for drug therapy. The development in the 1980s of molecules specifically targeting this receptor was followed by the rapid development of corresponding PET neuroimaging. Because this receptor represents a crucial target in neuroscience, a large number of radioligands have been developed by academic and industry centers for visualization and quantification, first in living animals and ultimately in humans. After a brief account of some of the structural and functional characteristics of brain 5-HT1A receptors, this review focuses on the main lines of evolution opened up by preclinical and clinical 5-HT1A PET radiopharmaceuticals, illustrating the potential value of PET for clinical research and drug development.

- Abstract
International audience; Serotonin (5-HT) neurotransmission plays a key modulatory role in the brain. This system is critical for pathophysiological processes and many drug treatments for brain disorders interact with its 14 subtypes of receptors. Positron emission tomography (PET) is a unique tool for the study of the living brain in translational studies from animal models to patients in neurology or psychiatry. This short review is intended to cover the current status of PET radioligands used for imaging human brain 5-HT receptors. Here, we describe the available PET radioligands for the 5-HT1A, 5-HT1B, 5-HT2A, 5-HT4 and 5-HT6 receptors. Finally, we highlight the future challenges for a functional PET imaging of serotonin receptors, including the research towards specific PET radiotracers for yet unexplored serotonin receptors, the need of radiotracers for endogenous serotonin level measurement and the contribution of agonist radiotracers for functional imaging of 5-HT neurotransmission.

Objective: To investigate the anatomic substrates underlying the beneficial effect of prism adaptation in five patients with persistent left neglect following right stroke. Methods: In a functional imaging PET study, we used a covariation analysis to examine linear changes of regional cerebral blood flow over sessions as a function of left neglect improvement. Results: The network of significant brain regions associated with improvement of left neglect performance produced by prism adaptation involved the right cerebellum, the left thalamus, the left temporo-occipital cortex, the left medial temporal cortex, and the right posterior parietal cortex. Conclusion: Our results suggest that the realignment of visuomotor coordinates is processed by the cerebellum and that low level sensorimotor adaptation actively modulates cerebral areas, albeit now relying on intact cerebellocerebral connections. Hence, our data support the hypothesis that the beneficial effect of prism adaptation on the clinical presentation of left neglect derives from modulation of cortical regions implicated in spatial cognition.

East of Madagascar, wind and surface buoyancy fluxes reinforce each other, leading to frontogenesis, outcrop and an eastward along-front flow: the South Indian Ocean Countercurrent (SICC). In the east the Leeuwin Current (LC) is a unique eastern boundary current which flows poleward along Australia. It is often described as a regional coastal current forced by an off-shore meridional density gradient or a sea surface slope, yet little is known of the forcing and dynamics that control these open ocean meridional gadients. To complete this understanding, we make use of both an ocean general circulation model and a conceptual two-layer model. The SICC impinges on west Australia and adds to a sea level slope and a southward geostrophic coastal jet: the Leeuwin Current. The SICC and the LC are thus dynamically connected. An observed transport maximum of the LC around 22° S is directly related to this impingement of the SICC. The circulation of the Indonesian Throughflow (ITF) through the Indian Ocean appears to be partly trapped in the upper layer north of the outcrop line and is redirected along this outcrop line to join the eastward flow of the SICC. Shutdown of the ITF in both models strongly decreases the Leeuwin Current transport and breaks the connection between the LC and SICC. In this case, most of the SICC was found to reconnect to the internal gyre circulation in the Indian Ocean. The Indonesian Throughflow, South Indian Ocean Countercurrent and the Leeuwin Current are thus dynamically coupled. [ABSTRACT FROM AUTHOR]

We provide a time series of Agulhas leakage anomalies over the last 20-years from satellite altimetry. Until now, measuring the interannual variability of Indo- Atlantic exchange has been the major barrier in the investigation of the dynamics and large scale impact of Agulhas leakage. We compute the difference of transport between the Agulhas Current and Agulhas Return Current, which allows us to deduce Agulhas leakage. The main difficulty is to separate the Agulhas Return Current from the southern limb of the subtropical "supergyre" south of Africa. For this purpose, an algorithm that uses absolute dynamic topography data is developed. The algorithm is applied to a state-of-the-art ocean model. The comparison with a Lagrangian method to measure the leakage allows us to validate the new method. An important result is that it is possible to measure Agulhas leakage in this model using the velocity field along a section that crosses both the Agulhas Current and the Agulhas Return Current. In the model a good correlation is found between measuring leakage using the full depth velocities and using only the surface geostrophic velocities. This allows us to extend the method to along-track absolute dynamic topography from satellites. It is shown that the accuracy of the mean dynamic topography does not allow us to determine the mean leakage but that leakage anomalies can be accurately computed. [ABSTRACT FROM AUTHOR]

Using ocean models of different complexity we show that opening the Indonesian Passage between the Pacific and the Indian oceans increases the input of Indian Ocean water into the South Atlantic via the Agulhas leakage. In a strongly eddying global ocean model this response results from an increased Agulhas Current transport and a constant proportion of Agulhas retroflection south of Africa. The leakage increases through an increased frequency of ring shedding events. In an idealized two-layer and flat-bottom eddy resolving model, the proportion of the Agulhas Current transport that retroflects is (for a wide range of wind stress forcing) not affected by an opening of the Indonesian Passage. Using a comparison with a linear model and previous work on the retroflection problem, the result is explained as a balance between two mechanisms: decrease retroflection due to large-scale momentum balance and increase due to local barotropic/baroclinic instabilities. Background and objectives: Brain tumours responsible for longstanding partial epilepsy are characterised by a high prevalence of dysembryoplastic neuroepithelial tumour (DNT), whose natural evolution is much more benign than that of gliomas. The preoperative diagnosis of DNT, which is not yet feasible on the basis of available clinical and imaging data, would help optimise the therapeutic strategy for this type of tumour. This study tested whether [
11 C]-methionine positron emission tomography (MET-PET) could help to distinguish DNTs from other epileptogenic brain tumours. Methods: Prospective study of 27 patients with partial epilepsy of at least six months duration related to a non-rapidly progressing brain tumour on magnetic resonance imaging (MRI). A structured visual analysis, which distinguished between normal, moderately abnormal, or markedly abnormal tumour methionine uptake, as well as various regions of interest and semiquantitative measurements were conducted. Results: Pathological results showed 11 DNTs (41%), 5 gangliogliomas (18%), and 11 gliomas (41%). MET-PET visual findings significantly differed between the various tumour types (p<0.0002), regardless of gadolinium enhancement on MRI, and were confirmed by semiquantitative analysis (p<0.001 for all calculated ratios). All gliomas and gangliogliomas were associated with moderately or markedly increased tumour methionine uptake, whereas 7/11 DNTs had a normal methionine uptake, including all six located in the mesiotemporal structures. No DNT presented with a marked MET-PET abnormality. Conclusion: Normal MET-PET findings in patient with an epileptogenic and non-rapidly progressing brain tumour are suggestive of DNT, whereas a markedly increased tumour methionine uptake makes this diagnosis unlikely. The aim of this study was to demonstrate the ability of a recently developed β[sup +]-range sensitive intracerebral probe (β-Microprobe) to measure the binding kinetics of [[sup 18]F]MPPF, a well-documented 5-HT[sub 1A] serotoninergic receptor ligand, in the dorsal raphe nucleus (DRN) of the anaesthetised rat. This midbrain nucleus presents a high concentration of 5-HT[sub 1A] receptors known to be implicated in the effects of antidepressants. The difficulty confronting this study lay in the fact that the dimensions of the DRN are smaller than the detection volume of the β-Microprobe. In the first part of the study, we studied the feasibility of this measurement from a theoretical point of view by autoradiography and a Monte Carlo simulation. We determined the optimal β-Microprobe location close to the DRN and verified that this configuration allowed accurate determination of [[sup 18]F]MPPF specific binding in the nucleus. In the second part of our study, we measured the in vivo time-concentration curves of [[sup 18]F]MPPF binding in the DRN in comparison with the cerebellum. The specificity of [[sup 18]F] MPPF binding in the DRN was confirmed by its displacement after non-labelled 5-HT[sup 1A] antagonist injection (MPPF or WAY-100635). Moreover, we verified the feasibility of using β-Microprobe monitoring and simultaneous validation by microdialysis to study the effect of an increase in extracellular serotonin, induced by fenfluramine injection, on [[sup 18]F]MPPF binding in the DRN. Our theoretical simulations, confirmed by our experimental results, demonstrate the ability of this new device to monitor in vivo the binding of [[sup 18]F]MPPF in the DRN of anaesthetised rodents. The in vivo behavior of 4-(2′-methoxyphenyl)-1-[2′-[N-(2″-pyridinyl)-p-[
18 F]fluorobenzamido]ethyl]-piperazine (p-[18 F]MPPF), a new serotonin 5-HT1A antagonist, was studied in awake, freely moving rats. Biodistribution studies showed that the carbon-fluorine bond was stable in vivo, that this compound was able to cross the blood-brain barrier, and that a general diffusion equilibrium could account for the availability of the tracer. The great quantity of highly polar metabolites found in plasma did not contribute to the small amounts of metabolites found in hippocampus, frontal cortex, and cerebellum. Exvivo p-[18 F]MPPF and in vitro 8-hydroxy-2-(di-n-[3 H]propylamino)tetralin autoradiography were compared both qualitatively and quantitatively. Qualitative evaluation proved that the same brain regions were labeled and that the p-[18 F]MPPF labeling is (a) in total agreement with the known distribution of 5-HT1A receptors in rats and (b) characterized by very low nonspecific binding. Quantitative comparison demonstrated that the in vivo labeling pattern obtained with p-[18 F]MPPF cannot be explained by differences in regional blood flow, capillary density, or permeability. The 5-HT1A specificity of p-[18 F]MPPF and binding reversibility were confirmed in vivo with displacement experiments.
