Abstract Bats (order: Chiroptera) are known to host a diverse range of viruses, some of which present a human public health risk. Thorough viral surveillance is therefore essential to predict and potentially mitigate zoonotic spillover. Astroviruses (family: Astroviridae) are an understudied group of viruses with a growing amount of indirect evidence for zoonotic transfer. Astroviruses have been detected in bats with significant prevalence and diversity, suggesting that bats may act as important astrovirus hosts. Most astrovirus surveillance in wild bat hosts has, to date, been restricted to single-gene PCR detection and concomitant Sanger sequencing; additionally, many bat species and many geographic regions have not yet been surveyed for astroviruses at all. Here, we use metagenomic Next Generation Sequencing (mNGS) to detect astroviruses in three species of Madagascar fruit bats, Eidolon dupreanum, Pteropus rufus, and Rousettus madagascariensis. We detect numerous partial sequences from all three species and one near-full length astrovirus sequence from Rousettus madagascariensis, which we use to characterize the evolutionary history of astroviruses both within bats and the broader mammalian clade, Mamastrovirus. Taken together, applications of mNGS implicate bats as important astrovirus hosts and demonstrate novel patterns of bat astrovirus evolutionary history, particularly in the Southwest Indian Ocean region.

Abstract Autocatalytic chemical networks play a predominant role in a large number of natural systems such as in metabolic pathways and in ecological networks. Despite recent efforts, the precise impact of thermodynamic constraints on these networks remains elusive. In this work, we present a theoretical framework that allows specific bounds on the thermodynamic affinity and on the concentrations of autocatalysts in mass-action autocatalytic networks. These bounds can be obtained solely from the stoichiometry of the underlying chemical reaction network, and are independent from the numerical values of kinetic parameters. This property holds in the specific regime where all the fluxes of the network are tightly coupled and maximal. Our method is applicable to large networks, and can be used to complement constraints-based modeling methods of metabolic networks, which typically do not provide predictions about thermodynamic properties or concentration ranges of metabolites.

Abstract Background Cocaine craving is a central symptom of cocaine use disorders (CUD). Virtual reality cue-exposure therapy for craving (VRCET) allows more immersive, realistic, and controllable exposure than traditional non-VR cue-exposure therapy (CET), whose efficacy is limited in treating substance use disorders. The purpose of this study is to evaluate the efficacy and acceptability of VRCET, as a stand-alone and add-on intervention (i.e., combined with cognitive therapy), compared to a picture-based CET (PCET), in reducing self-reported cocaine craving in inpatients hospitalized for CUD. Methods Fifty-four inpatients hospitalized for CUD will be randomized in one of two intensive 3-week treatment arms: 10 meetings/2-week treatment of VRCET plus 5 meetings/1-week treatment of memory-focused cognitive therapy (MFCT; experimental arm), or 15 meetings/3-week treatment of PCET (active control arm). The Craving Experience Questionnaire (CEQ – F & S) will be used to assess the primary outcome, i.e., the post-treatment decrease of self-reported cocaine craving frequency (within the past 2 weeks) and intensity scores (in VR exposure to cocaine cues). Secondary endpoints include urinary, physiological, and self-reported cocaine use-related measures. Assessments are scheduled at pretreatment, after 2 weeks of treatment (i.e., VRCET vs. PCET), post-treatment (3 weeks, i.e., VRCET + MFCT vs. PCET), and at 1-month follow-up. Acceptability will be evaluated via (i) the Spatial Presence for Immersive Environments – Cybersickness along VRCET and (ii) the Client Satisfaction Questionnaires after 2 weeks of treatment and post-treatment. Discussion This study will be the first to evaluate the acceptability and efficacy of VRCET for CUD, as a psychotherapeutic add-on, to reduce both cocaine craving frequency and intensity. Additionally, this study will provide evidence about the specific interest of VRCET, compared to a non-VR-based CET, as a cue reactivity and exposure paradigm for treating substance use disorders. En el marco de la Guerra Fría, se examinan las implicancias del Torneo Internacional de Ajedrez de La Serena (TIALAS), Chile (1972) que contó con la presencia del campeón soviético, Vladimir Savon, en el marco del apoyo de la URSS al gobierno de Salvador Allende. El torneo se realizó en el Norte Chico de Chile, lugar caracterizado por la elaboración del pisco (destilado de uva), producto típico que se elabora en ese territorio desde hace más de cuatro siglos y reconocido como la primera Denominación de Origen de América, delimitado como tal en 1931. El TIALAS tenía el potencial de visibilizar, promover y poner en valor el pisco, en el marco de una estrategia de gastropolítica y ludodiplomacia, en el sentido de fortalecer su valor simbólico mediante el acercamiento del producto a figuras de prestigio: así como durante muchos años el marketing de los vinos giraba en torno a figuras de la corona y la nobleza, en este caso la idea era fortalecer el producto a partir de la asociación con los genios del deporte-ciencia, símbolos de la inteligencia. La idea de realizar el torneo surgió del seno de la industria del pisco, y fue apoyado por el gobierno que hizo las gestiones para asegurar la presencia de Savón y financió los costos a través del apoyo de empresas estatales. Fue un proyecto innovador y precursor de iniciativas similares en otros lugares del mundo, como el torneo internacional de Villarrobledo en España, que también convocó figuras mundiales de ajedrez para patrimonializar el queso manchego con éxito. El TIALAS apuntaba a instalar un acontecimiento permanente, lo cual se frustró por las tensiones políticas derivadas de la Guerra Fría y los constantes conflictos entre el gobierno y la industria del pisco. Se concluye que la ludodiplomacia y la gastropolítica representan herramientas interesantes en asociación con productos típicos patrimoniales, pero requieren del consenso entre los actores políticos y económicos para resultar viables.

13. Glial expression of Drosophila UBE3A causes spontaneous seizures that can be modulated by 5-HT signaling

Misexpression of the E3 ubiquitin ligase gene UBE3A is thought to contribute to a range of neurological disorders. In the context of Dup15q syndrome, additional genomic copies of UBE3A give rise to the autism, muscle hypotonia and spontaneous seizures characteristics of the disorder. In a Drosophila model of Dup 15q syndrome, it was recently shown that glial-driven expression of the UBE3A ortholog dube3a led to a “bang-sensitive” phenotype, where mechanical shock triggers convulsions, suggesting glial dube3a expression contributes to hyperexcitability in flies. Here we directly compare the consequences of glial- and neuronal-driven dube3a expression on motor coordination and seizure susceptibility in Drosophila.To quantify seizure-related behavioral events, we developed and trained a hidden Markov model that identified these events based on automated video tracking of fly locomotion. Both glial and neuronal driven dube3a expression led to clear motor phenotypes. However, only glial-driven dube3a expression displayed spontaneous seizure-associated immobilization events, that were clearly observed at high-temperature (38 °C). Using a tethered fly preparation amenable to electrophysiological monitoring of seizure activity, we found glial-driven dube3a flies display aberrant spontaneous spike discharges which are bilaterally synchronized. Neither neuronal-dube3a overexpressing flies, nor control flies displayed these firing patterns. We previously performed a drug screen for FDA approved compounds that can suppress bang-sensitivity in glial-driven dube3a expressing flies and identified certain 5-HT modulators as strong seizure suppressors. Here we found glial-driven dube3a flies fed the serotonin reuptake inhibitor vortioxetine and the 5-HT2A antagonist ketanserin displayed reduced immobilization and spike bursting, consistent with the previous study. Together these findings highlight the potential for glial pathophysiology to drive Dup15q syndrome-related seizure activity.

14. Long-term contributions of VOC sources and their link to ozone pollution in Bronx, New York City

Changes in energy and environmental policies along with changes in the energy markets of New York State over the past two decades, have spurred interest in evaluating their impacts on emissions from various energy generation sectors. This study focused on quantifying these effects on VOC (volatile organic compounds) emissions and their subsequent impacts on air quality within the New York City (NYC) metropolitan area. NYC is an EPA nonattainment region for ozone (O3) and likely is a VOC limited region. NYC has a complex coastal topography and meteorology with low-level jets and sea/bay/land breeze circulation associated with heat waves, leading to summertime O3 exceedances and formation of secondary organic aerosol (SOA). To date, no comprehensive source apportionment studies have been done to understand the contributions of local and long-range sources of VOCs in this area. This study applied an improved Positive Matrix Factorization (PMF) methodology designed to incorporate atmospheric dispersion and photochemical reaction losses of VOCs to provide improved apportionment results. Hourly measurements of VOCs were obtained from a Photochemical Assessment Monitoring Station located at an urban site in the Bronx from 2000 to 2021. The study further explores the role of VOC sources in O3 and SOA formation and leverages advanced machine learning tools, XGBoost and SHAP algorithms, to identify synergistic interactions between sources and provided VOC source impacts on ambient O3 concentrations. Isoprene demonstrated a substantial influence in the source contribution of the biogenic factor, emphasizing its role in O3 formation. Notable contributions from anthropogenic emissions, such as fuel evaporation and various industrial processes, along with significant traffic-related sources that likely contribute to SOA formation, underscore the combined impact of natural and human-made sources on O3 pollution. Findings of this study can assist regulatory agencies in developing appropriate policy and management initiatives to control O3 pollution in NYC.

15. Benthic responses to organic enrichment under a mussel (Mytilus edulis) farm

Bivalve culture is know to affect some interactions between the water column and the benthic environment with the organic enrichment. An in situ mesocosms experiment was used to determine the influence of mussel biodeposition on the benthic ecosystem in St. Peters Bay, Prince Edward Island, eastern Canada. Eight mussel densities (0 to 2266 mussels m-2) were placed above mesocosms to reproduce the organic enrichment from a mussel farm over 12 weeks. The experimental procedure created an accumulation of organic matter on the seafloor with increasing mussel densities. Some benthic parameters had the predicted response to organic enrichment, with the presence of anoxic sediment in sediment profile images, increased water and organic matter content, and reduced infaunal abundance and species richness. Other parameters, including redox potential, total free sulfides, and the response of the biotic index AMBI, had inconsistent responses to mussel biodeposition-related organic enrichment. Given that other studies done in other locations have observed varying results, results suggest that measurement of various parameters is important to interpret the influence of deposition from mussel farms. The conditions of the site and the study duration are also parameters to consider when evaluating the results from mesocosm studies. Results from this study also suggest that St. Peters Bay is likely impacted by biodeposition from the mussels farmed there.

The MoniDiam project is part of the French national collaboration CLaRyS (Contrôle en Ligne de l’hAdronthérapie par RaYonnements Secondaires) for on-line monitoring of hadron therapy. It relies on the imaging of nuclear reaction products that is related to the ion range. The goal here is to provide large area beam detectors with a high detection efficiency for carbon or proton beams giving time and position measurement at 100 MHz count rates (beam tagging hodoscope). High radiation hardness and intrinsic electronic properties make diamonds reliable and very fast detectors with a good signal to noise ratio. Commercial Chemical Vapor Deposited (CVD) poly-crystalline, heteroepitaxial and monocrystalline diamonds were studied. Their applicability as a particle detector was investigated using α and β radioactive sources, 95 MeV/u carbon ion beams at GANIL and 8.5 keV X-ray photon bunches from ESRF. This facility offers the unique capability of providing a focused (~1 μm) beam in bunches of 100 ps duration, with an almost uniform energy deposition in the irradiated detector volume, therefore mimicking the interaction of single ions. A signal rise time resolution ranging from 20 to 90 ps rms and an energy resolution of 7 to 9% were measured using diamonds with aluminum disk shaped surface metallization. This enabled us to conclude that polycrystalline CVD diamond detectors are good candidates for our beam tagging hodoscope development. Recently, double-side stripped metallized diamonds were tested using the XBIC (X Rays Beam Induced Current) set-up of the ID21 beamline at ESRF which permits us to evaluate the capability of diamond to be used as position sensitive detector. The final detector will consist in a mosaic arrangement of double-side stripped diamond sensors read out by a dedicated fast-integrated electronics of several hundreds of channels.

IMPORTANCE:. Ventilator-associated pneumonia (VAP) frequently occurs in patients with cardiac arrest. Diagnosis of VAP after cardiac arrest remains challenging, while the use of current biomarkers such as C-reactive protein (CRP) or procalcitonin (PCT) is debated. OBJECTIVES:. To evaluate biomarkers’ impact in helping VAP diagnosis after cardiac arrest. DESIGN, SETTING, AND PARTICIPANTS:. This is a prospective ancillary study of the randomized, multicenter, double-blind placebo-controlled ANtibiotherapy during Therapeutic HypothermiA to pRevenT Infectious Complications (ANTHARTIC) trial evaluating the impact of antibiotic prophylaxis to prevent VAP in out-of-hospital patients with cardiac arrest secondary to shockable rhythm and treated with therapeutic hypothermia. An adjudication committee blindly evaluated VAP according to predefined clinical, radiologic, and microbiological criteria. All patients with available biomarker(s), sample(s), and consent approval were included. MAIN OUTCOMES AND MEASURES:. The main endpoint was to evaluate the ability of biomarkers to correctly diagnose and predict VAP within 48 hours after sampling. The secondary endpoint was to study the combination of two biomarkers in discriminating VAP. Blood samples were collected at baseline on day 3. Routine and exploratory panel of inflammatory biomarkers measurements were blindly performed. Analyses were adjusted on the randomization group. RESULTS:. Among 161 patients of the ANTHARTIC trial with available biological sample(s), patients with VAP (n = 33) had higher body mass index and Acute Physiology and Chronic Health Evaluation II score, more unwitnessed cardiac arrest, more catecholamines, and experienced more prolonged therapeutic hypothermia duration than patients without VAP (n = 121). In univariate analyses, biomarkers significantly associated with VAP and showing an area under the curve (AUC) greater than 0.70 were CRP (AUC = 0.76), interleukin (IL) 17A and 17C (IL17C) (0.74), macrophage colony-stimulating factor 1 (0.73), PCT (0.72), and vascular endothelial growth factor A (VEGF-A) (0.71). Multivariate analysis combining novel biomarkers revealed several pairs with p value of less than 0.001 and odds ratio greater than 1: VEGF-A + IL12 subunit beta (IL12B), Fms-related tyrosine kinase 3 ligands (Flt3L) + C–C chemokine 20 (CCL20), Flt3L + IL17A, Flt3L + IL6, STAM-binding protein (STAMBP) + CCL20, STAMBP + IL6, CCL20 + 4EBP1, CCL20 + caspase-8 (CASP8), IL6 + 4EBP1, and IL6 + CASP8. Best AUCs were observed for CRP + IL6 (0.79), CRP + CCL20 (0.78), CRP + IL17A, and CRP + IL17C. CONCLUSIONS AND RELEVANCE:. Our exploratory study shows that specific biomarkers, especially CRP combined with IL6, could help to better diagnose or predict early VAP occurrence in cardiac arrest patients.

20. Characterization of detonation waves by simultaneous OH and NO planar laser-induced fluorescence

This study presents a thorough evaluation of the simultaneous planar laser-induced fluorescence (PLIF) on hydroxyl (OH) radicals and nitric oxide (NO), for characterizing hydrogen detonation fronts. The study combines experimental and numerical results to evaluate the benefits and drawbacks of different visualization strategies, namely OH-PLIF alone, NO-PLIF alone, OH-PLIF + shock det. (comparable to simultaneous OH-PLIF and schlieren), and combined OH- and NO-PLIF. Main findings are: (i) For the first time, simultaneous single-shot OH- and NO-PLIF visualizations are demonstrated on detonation waves for three mixtures with varying stability levels: Mixture (a) 2H2-O2-3.76Ar, Mixture (b) 2H2-O2-3.76N2, and Mixture (c) 3H2-CH4-3.5O2-3.76Ar; (ii) the simultaneous visualization evidences the self-similarity of both techniques, with a clear interlock of OH-PLIF imaging into NO-PLIF imaging. This means that the structure of the OH-reaction zone, typically visualized from OH-PLIF, can be accurately described from NO-PLIF imaging alone, from which induction zones can also be visualized. In addition, both can clearly depict the wrinkling of the front and the presence of unburned pockets; (iii) the effectiveness of the four visualization strategies at characterizing the detonation wave of a H2-O2-3.76Ar mixture is determined using ZND and spectroscopic simulations. Interestingly, the induction zone (Δi) is more accurately predicted by NO-PLIF (3% error) than with the combined OH + shock det. (10% error). This study highlights the potential of NO-PLIF diagnostic in characterizing detonation waves with low levels of confinement (i.e., d/λ>>>1) both qualitatively (e.g., reaction zone structure) and quantitatively (e.g., Δi measurements) using a single diagnostic. Additionally, the simultaneous OH- and NO-PLIF diagnostic may offer significant advantages in characterizing the detonation waves in engine-relevant conditions (i.e., in complex geometries with unburned pockets or local quenching events), as combining both diagnostics enables to discriminate unburned pockets and local quenching events.

Background: COVID-19 is primarily known as a respiratory illness; however, many patients present to hospital without respiratory symptoms. The association between non-respiratory presentations of COVID-19 and outcomes remains unclear. We investigated risk factors and clinical outcomes in patients with no respiratory symptoms (NRS) and respiratory symptoms (RS) at hospital admission. Methods: This study describes clinical features, physiological parameters, and outcomes of hospitalised COVID-19 patients, stratified by the presence or absence of respiratory symptoms at hospital admission. RS patients had one or more of: cough, shortness of breath, sore throat, runny nose or wheezing; while NRS patients did not. Results: Of 178,640 patients in the study, 86.4 % presented with RS, while 13.6 % had NRS. NRS patients were older (median age: NRS: 74 vs RS: 65) and less likely to be admitted to the ICU (NRS: 36.7 % vs RS: 37.5 %). NRS patients had a higher crude in-hospital case-fatality ratio (NRS 41.1 % vs. RS 32.0 %), but a lower risk of death after adjusting for confounders (HR 0.88 [0.83–0.93]). Conclusion: Approximately one in seven COVID-19 patients presented at hospital admission without respiratory symptoms. These patients were older, had lower ICU admission rates, and had a lower risk of in-hospital mortality after adjusting for confounders.

IntroductionBovine respiratory disease (BRD) is one of the most important animal health problems in the beef industry. While bacterial culture and antimicrobial susceptibility testing have been used for diagnostic testing, the common practice of examining one isolate per species does not fully reflect the bacterial population in the sample. In contrast, a recent study with metagenomic sequencing of nasal swabs from feedlot cattle is promising in terms of bacterial pathogen identification and detection of antimicrobial resistance genes (ARGs). However, the sensitivity of metagenomic sequencing was impeded by the high proportion of host biomass in the nasal swab samples.MethodsThis pilot study employed a non-selective bacterial enrichment step before nucleic acid extraction to increase the relative proportion of bacterial DNA for sequencing.ResultsNon-selective bacterial enrichment increased the proportion of bacteria relative to host sequence data, allowing increased detection of BRD pathogens compared with unenriched samples. This process also allowed for enhanced detection of ARGs with species-level resolution, including detection of ARGs for bacterial species of interest that were not targeted for culture and susceptibility testing. The long-read sequencing approach enabled ARG detection on individual bacterial reads without the need for assembly. Metagenomics following non-selective bacterial enrichment resulted in substantial agreement for four of six comparisons with culture for respiratory bacteria and substantial or better correlation with qPCR. Comparison between isolate susceptibility results and detection of ARGs was best for macrolide ARGs in Mannheimia haemolytica reads but was also substantial for sulfonamide ARGs within M. haemolytica and Pasteurella multocida reads and tetracycline ARGs in Histophilus somni reads.DiscussionBy increasing the proportion of bacterial DNA relative to host DNA through non-selective enrichment, we demonstrated a corresponding increase in the proportion of sequencing data identifying BRD-associated pathogens and ARGs in deep nasopharyngeal swabs from feedlot cattle using long-read metagenomic sequencing. This method shows promise as a detection strategy for BRD pathogens and ARGs and strikes a balance between processing time, input costs, and generation of on-target data. This approach could serve as a valuable tool to inform antimicrobial management for BRD and support antimicrobial stewardship.

Self-driving vehicles have attracted significant attention in the automotive industry that is heavily investing to reach the level of reliability needed from these safety critical systems. Security of in-vehicle communications is mandatory to achieve this goal. Most of the existing research to detect anomalies for in-vehicle communication does not take into account the low processing power of the in-vehicle Network and ECUs (Electronic Control Units). Also, these approaches do not consider system level isolation challenges such as side-channel vulnerabilities, that may arise due to adoption of new technologies in the automotive domain. This paper introduces and discusses the design of a framework to detect anomalies in in-vehicle communications, including side channel attacks. The proposed framework supports real time monitoring of data exchanges among the components of in-vehicle communication network and ensures the isolation of the components in in-vehicle network by deploying them in Trusted Execution Environments (TEEs). The framework is designed based on the AUTOSAR open standard for automotive software architecture and framework. The paper also discusses the implementation and evaluation of the proposed framework.

Brain development and function are highly reliant on adequate establishment and maintenance of vascular networks. Early impairments in vascular health can impact brain maturation and energy metabolism, which may lead to neurodevelopmental anomalies. Our recent work not only provides novel insights into the development of cerebrovascular networks but also emphasizes the importance of their well-being for proper brain maturation. In particular, we have demonstrated that endothelial dysfunction in autism spectrum disorders (ASD) mouse models is causally related to altered behavior and brain metabolism. In the prenatal human brain, vascular cells change metabolic states in the second trimester. Such findings highlight the need to identify new cellular and molecular players in neurodevelopmental disorders, raising awareness about the importance of a healthy vasculature for brain development. It is thus essential to shift the mostly neuronal point of view in research on ASD and other neurodevelopmental disorders to also include vascular and metabolic features.

Introduction Lung transplantation (LTx) aims at improving survival and quality of life for patients with end-stage lung diseases. Venoarterial extracorporeal membrane oxygenation (VA-ECMO) is used as intraoperative support for LTx, despite no precise guidelines for its initiation. We aim to evaluate two strategies of VA-ECMO initiation in the perioperative period in patients with obstructive or restrictive lung disease requiring bilateral LTx. In the control ‘on-demand’ arm, high haemodynamic and respiratory needs will dictate VA-ECMO initiation; in the experimental ‘systematic’ arm, VA-ECMO will be pre-emptively initiated. We hypothesise a ‘systematic’ strategy will increase the number of ventilatory-free days at day 28.Methods and analysis We designed a multicentre randomised controlled trial in parallel groups. Adult patients with obstructive or restrictive lung disease requiring bilateral LTx, without a formal indication for pre-emptive VA-ECMO before LTx, will be included. Patients with preoperative pulmonary hypertension with haemodynamic collapse, ECMO as a bridge to transplantation, severe hypoxaemia or hypercarbia will be secondarily excluded. In the systematic group, VA-ECMO will be systematically implanted before the first pulmonary artery cross-clamp. In the on-demand group, VA-ECMO will be implanted intraoperatively if haemodynamic or respiratory indices meet preplanned criteria. Non-inclusion, secondary exclusion and VA-ECMO initiation criteria were validated by a Delphi process among investigators. Postoperative weaning of ECMO and mechanical ventilation will be managed according to best practice guidelines. The number of ventilator-free days at 28 days (primary endpoint) will be compared between the two groups in the intention-to-treat population. Secondary endpoints encompass organ failure occurrence, day 28, day 90 and year 1 vital status, and adverse events.Ethics and dissemination The sponsor is the Assistance Publique–Hôpitaux de Paris. The ECMOToP protocol version 2.1 was approved by Comité de Protection des Personnes Ile de France VIII. Results will be published in international peer-reviewed medical journals.Trial registration number NCT05664204.
