The antibiotic azatyrosine [DL-3-(5-hydroxy-2-pyridyl)alanine] suppressed meiotic maturation in oocytes induced by progesterone or the combination of [Val12]p21Ha-ras microinjection and insulin-like growth factor I. The suppression was dose-dependent in the range of 20-250 microM azatyrosine. In addition, azatyrosine blocked the tyrosine phosphorylation of Xp42, a member of the mitogen-activated protein kinase family, after progesterone or [Val12]p21Ha-ras/insulin-like growth factor I stimulation. Activation of maturation-promoting factor, as shown by a decrease in the tyrosine phosphorylation of the Xenopus homolog of p34cdc2, was also suppressed by azatyrosine. Azatyrosine had no effect in vivo or in vitro on the growth factor-induced autophosphorylation of the oocyte insulin-like growth factor I receptor. Azatyrosine has been shown by others [Shindo-Okada, N., Makabe, O., Nagahara, H. & Nishimura, S. (1989) Mol. Carcinog. 2, 159-167] to inhibit the growth of ras-transformed cells without affecting that of nontransformed cells. In oocytes, the antibiotic exerts an inhibitory action on both a ras-dependent and a ras-independent pathway. Lack of an effect of azatyrosine on germinal vesicle breakdown induced by the microinjection of an extract from mature oocytes, however, suggests that azatryosine is acting upstream of maturation-promoting factor activation.

The muscanmc cholinergic antagonist 3-quinudidinyl benzilate (QNB) binds avidly but reversibly to the muscarinic cholinergic receptor of mammalian brain and peripheral tissues. [3H]-QNB binding provides a simple, sensitive and specific assay for the muscarinic cholinergic receptor binding. Inhibition of [3H]-QNB binding to homogenates of brain and guinea pig ileum by muscarinic drugs correlates with their pharmacologic potencies, while nicotinic agents and noncholinergic drugs have negligible affinity. The regional distribution of [3H]-QNB binding throughout rat and monkey brain parallels to a major extent other cholinergic markers, suggesting that the majority of cholinergic synapses in the brain are muscarinic. [3H]-QNB accumulation in various brain regions after intravenous injection provides a means of labelling the muscarinic receptor in vivo. By labelling the receptor in vivo, autoradiographic studies under the light microscope have been performed to visualize the muscarinic receptor.

1. Leucine-enkephalin and some analogues were assayed for activity in vitro on the mouse vas deferens and for binding to opiate receptors from rat brain. 2. The experimental data were analysed in terms of the stringency for glycine, a D-amino acid or an L-amino acid at each position in the peptide. 3. The observed configurational specificity was compared with the stringency that would be predicted to occur if enkephalin adopted certain hydrogen-bonded conformations at the receptor. 4. A small subset of the conformations examined was found to be compatible with the experimental data.

Permeabilization of human platelets with saponin (15-25 micrograms/ml) allows the determination of the ADP-ribosylation of a 41-kDa protein by pertussis toxin. The ADP-ribosylated protein is present in the particulate fraction. ADP-ribosylation of the 41-kDa protein increases for 20 min; it is not affected by indomethacin, prostacyclin, and 1,2-diacylglycerols but is inhibited by 1 mM Ca2+ and phorbol esters. Treatment of platelets with trypsin, thrombin, or collagen before saponin addition precludes subsequent pertussis toxin-induced ADP-ribosylation of the 41-kDa protein. The effect of trypsin or thrombin is blocked by soybean trypsin inhibitor and leupeptin. Trypsin proteolytically cleaves the ADP-ribosylated 41-kDa protein to an ADP-ribosylated fragment slightly smaller than 20 kDa. The results suggest that a modification of a guanine nucleotide-binding regulatory protein is associated with the actions of trypsin, thrombin, and collagen on platelet activation.
