This review article summarises the results of research on forest liming, fertilisation, nutrition flows and cycles in selected European and other countries. The presented studies mostly deal with assessing the effect of liming and fertilisation applied during planting or shortly after planting. The sporadic studies on fertilisation in older stands are also presented. The application of crushed limestone, dolomite or other alkaline rocks or their mixtures is usually used to improve the soil conditions on a large area through the adjustment of soil acidity and to supply any deficient elements, especially calcium and magnesium. These amendments are typically used on naturally nutrient-poor soils or as a curative technique to neutralise the soil chemistry affected by anthropogenic acidification. Artificial fertilisers are usually applied on small spots to individual trees on the surface of the soil shortly after planting or into a planting hole during planting. The purpose is to give some initial support to young trees to better overcome the post-planting shock and to accelerate the height growth. Less frequently, artificial fertilisers are used on large areas of forest stands for the purpose of increasing stem growth. The methods and the extent of forest fertilisation substantially vary in individual countries and different time periods.

This review uses a case study based on an executed afforestation project to comprehensively describe the administrative procedure regarding afforestation of agricultural land in the Czech Republic, Central Europe. We consider the official proceedings for obtaining a permit to land-use change (i.e. changing the nature of land use and the mode of land use in the real estate cadastre) as a very complex and protracted procedure, which can substantially reduce the motivation of people to establish new forests. Therefore, we propose a simplification of the administration process of land-use change. The main idea is to make the procedure as simple as possible in those areas where afforestation is desirable, especially from the environmental point of view. It assumes the possibility of quick, precise and, if possible, automatic distinction whether the afforestation is suitable or not. The situation in the Czech Republic is compared with the surrounding Central-European countries: Poland and Slovakia. In these countries, the official procedure connected with afforestation seems to be rather less complicated.

Seeds of forest species can show germination and survival problems if they are exposed to adverse climatic conditions in the course of germination. Brassinosteroids could help seeds to overcome such environmental stress. We tested the effects of exogenous application of a chosen brassinosteroid compound 2α,3α,17β-trihydroxy-5α-androstan-6-one on the germination capacity and germination energy of European black pine (Pinus nigra J. F. Arnold) seeds. Before germination, the seeds were soaked for 24 hours either in demineralised water (control treatment) or in brassinosteroid solutions of four concentrations (high, higher-medium, lower-medium and low-concentration treatments). In the course of germination, the control and all four concentration treatments were subjected to two alternative temperature regimes: the optimal (unstressed) regime (20/30°C) and the stress regime, during which the seeds were exposed to temporary temperature stress (peaking at 42°C). In the optimal temperature regime, the highest germination energy was recorded in the higher-medium-concentration treatment and a significantly increased germination energy when compared to the control was also observed in a high-concentration treatment. The brassinosteroid, when applied in high, higher-medium and lower-medium concentrations, significantly increased the germination capacity compared to the control. The highest germination capacity was recorded in the high-concentration treatment. The temperature stress substantially reduced the germination of P. nigra. In the stress regime, the seeds of the higher-medium-concentration treatment decidedly showed the highest germination energy and capacity and those of the control treatment the lowest. The seed germination energy in the control was significantly lower than that recorded in the high and higher-medium-concentration treatments. The seed germination capacity in the control was significantly lower than those found in all concentration treatments, except for the seeds in the low-concentration treatment. The brassinosteroid application promoted the germination of black pine and partly alleviated the impact of the temporary heat stress. However, this application did not compensate for the heat stress effects completely. The potential for the use of the brassinosteroid may exist chiefly for seedings of Pinus nigra in forest nurseries where the nursery staff try to maintain optimal climatic conditions, and deviations from these conditions are usually only temporary.

Precision agriculture (PA) relies on a large amount of precise data about a given area and allows that data to be used in accordance with agronomic practices. It offers farmers greater control over existing processes, from crop placement to soil conditions to chemical use. On the other hand, applying precision livestock solutions to these expanding systems is a way to bring animals closer to producers and minimize waste and costs. In Europe, precision agriculture has emerged as a new way to help farmers increase the quantity and quality of agricultural production while using fewer inputs. The spatial and temporal variability in the application of PA and the implications of site-specific application are documented in this article. The objective of this review article is to provide an overview of the application of precision agriculture in Central European countries (Poland, Czech Republic, Austria, Slovakia, Slovenia, Germany, and Hungary) identified through a systematic literature review (SLR). The analyses revealed that with the rapid development of automation in agriculture, the demand for skilled workers will continue to increase and technology will continue to open up new application areas as well as the development of new AgriTech startups in the future. Currently, there are farmers in Germany and the Czech Republic who are leaders in the use of these technologies. However, the application of precision agriculture requires further research to provide more accurate information and is an area that is constantly evolving.

Only 1.82% of the Czech forests are covered by the introduced (exotic, non-native) tree species, they represent only a very minor part of the forest area in the Czech Republic. Despite this fact under certain circumstances and locally, they can play an important role in the forest restoration of declined forests. The main non-native tree species used in the Czech Republic are: black locust (Robinia pseudoacacia L.), Douglas-fir (Pseudotsuga menziesii [Mirbel] Franco), grand fir (Abies grandis [Douglas ex D. Don] Lindl.), black walnut (Juglans nigra L.), northern red oak (Quercus rubra L.), and blue spruce (Picea pungens Engelm.). Other tree species are cultivated on very small areas, despite their much larger potential. The aim of the presented review paper is to summarize information on the introduced tree species, available especially from national sources, and give comprehensive information concerning the potential and risk of their use in the conditions of the Czech Republic. The authors mention also other tree species eligible for silviculture under current as well as future climatic circumstances. The current area and silvicultural potential in the climate changing conditions are analysed and summarized.

A procedure for quantifying the U 5f electronic covalency and degree of localization in U intermetallic compounds is presented. To this end, bulk sensitive hard and soft x-ray photoelectron spectroscopy were utilized in combination with density-functional theory (DFT) plus dynamical mean-field theory (DMFT) calculations. The energy dependence of the photoionization cross sections allows the disentanglement of the U 5f contribution to the valence band from the various other atomic subshells so the computational parameters in the DFT+DMFT can be reliably determined. Applying this method to UGa_{2} and UB_{2} as model compounds from opposite ends of the (de)localization range, we have achieved excellent simulations of the valence band and core-level spectra. The width in the distribution of atomic U 5f configurations contributing to the ground state, as obtained from the calculations, quantifies the correlated nature and degree of localization of the U 5f. The findings permit answering the longstanding question why different spectroscopic techniques give seemingly different numbers for the U 5f valence in intermetallic U compounds.

AbstractPeptides, as potential therapeutics continue to gain importance in the search for active substances for the treatment of numerous human diseases, some of which are, to this day, incurable. As potential therapeutic drugs, peptides have many favorable chemical and pharmacological properties, starting with their great diversity, through their high affinity for binding to all sort of natural receptors, and ending with the various pathways of their breakdown, which produces nothing but amino acids that are nontoxic to the body. Despite these and other advantages, however, they also have their pitfalls. One of these disadvantages is the very low stability of natural peptides. They have a short half-life and tend to be cleared from the organism very quickly. Their instability in the gastrointestinal tract, makes it impossible to administer peptidic drugs orally. To achieve the best pharmacologic effect, it is desirable to look for ways of modifying peptides that enable the use of these substances as pharmaceuticals. There are many ways to modify peptides. Herein we summarize the approaches that are currently in use, including lipidization, PEGylation, glycosylation and others, focusing on lipidization. We describe how individual types of lipidization are achieved and describe their advantages and drawbacks. Peptide modifications are performed with the goal of reaching a longer half-life, reducing immunogenicity and improving bioavailability. In the case of neuropeptides, lipidization aids their activity in the central nervous system after the peripheral administration. At the end of our review, we summarize all lipidized peptide-based drugs that are currently on the market.

The aim of the first experiment was to verify the effectiveness and profitability of providing supplemental feed to piglets. The milk feed mixture of two producers (MFM-1 vs MFM-2) were evaluated (served ad libitum). All piglets were suckled and were fed with the pre-starter feed mixture from the 5th day until the weaning stage. The control group without milk supplements has shown a higher piglet mortality and worse body condition of sows. The usage of milk supplements led to the elimination of the sow body condition loss during the suckling period and reduced feed mixture consumption in sows (P

Ambelline, an alkaloid from the Amaryllidaceae family with a crinane-type skeleton, has not yet demonstrated any outstanding biological activity. However, its analogues prepared by derivatization of the C-11 hydroxyl group show different interesting effects. Continuing our earlier work, twelve novel aromatic esters were developed (10, 14, 16, 17, 22–25, 30–33) and studied, together with previously synthesized derivatives (2–9, 11–13, 15, 18–21, 26–29) in terms of their cytotoxic activity. The cytotoxic potential was determined on a panel of nine human cancer cell lines and one noncancerous cell line to characterize their biological activity spectrum. To describe and foresee the structure–activity relationship for further research, substances synthesized and described in our previous work were also included in this cytotoxicity study. The most significant activity was associated with analogues having methyl (10), methoxy (14–17), or ethoxy (18) substitution on the phenyl condensed to ambelline. However, the 4-chloro-3-nitrobenzoyl derivative (32) showed the most promising IC50 values, ranging from 0.6 ± 0.1 µM to 9.9 ± 0.2 µM. In vitro cytotoxicity studies indicated the most potent antiproliferative activity of 32 in a dose-dependent and time-dependent manner. Besides, 32 was found to be effective in decreasing viability and triggering apoptosis of MOLT-4 T-lymphoblastic leukemia cells.

The fate of high-energy degrees of freedom, such as spin-orbit interactions, in the coherent state of Kondo lattice materials remains unclear. Here, the authors use resonant inelastic x-ray scattering in CePd3 to show how Kondo-quasiparticle excitations are renormalized and develop a pronounced momentum dependence, while maintaining a largely unchanged spin-orbit gap.

Abstract Mitigating the effects of global change on biodiversity requires its understanding in the past. The main proxy of plant diversity, fossil pollen record, has a complex relationship to surrounding vegetation and unknown spatial scale. We explored both using modern pollen spectra in species-rich and species-poor regions in temperate Central Europe. We also considered the biasing effects of the trees by using sites in forests and open habitats in each region. Pollen samples were collected from moss polsters at 60 sites and plant species were recorded along two 1 km-transects at each site. We found a significant positive correlation between pollen and plant richness (alpha diversity) in both complete datasets and for both subsets from open habitats. Pollen richness in forest datasets is not significantly related to floristic data due to canopy interception of pollen rather than to pollen productivity. Variances (beta diversity) of the six pollen and floristic datasets are strongly correlated. The source area of pollen richness is determined by the number of species appearing with increasing distance, which aggregates information on diversity of individual patches within the landscape mosaic and on their compositional similarity. Our results validate pollen as a reconstruction tool for plant diversity in the past.

Abstract Background/Objective Anorexigenic palmitoylated prolactin-releasing peptide (palm11-PrRP) is able to act centrally after peripheral administration in rat and mouse models of obesity, type 2 diabetes mellitus and/or neurodegeneration. Functional leptin and intact leptin signaling pathways are necessary for the body weight reducing and glucose tolerance improving effect of palm11-PrRP. We have previously shown that palm11-PrRP31 had glucose-lowering properties but not anti-obesity effect in Koletsky rats with leptin signaling disturbances, so improvements in glucose metabolism appear to be completely independent of leptin signaling. The purpose of this study was to describe relationship between metabolic and neurodegenerative pathologies and explore if palm11-PrRP31 could ameliorate them in obese fa/fa rat model with leptin signaling disruption. Subject/Methods The fa/fa rats and their age-matched lean controls at the age 32 weeks were used for this study. The rats were infused for 2 months with saline or palm11-PrRP31 (n = 7–8 per group) at a dose of 5 mg/kg per day using Alzet osmotic pumps. During the dosing period food intake and body weight were monitored. At the end of experiment the oral glucose tolerance test was performed; plasma and tissue samples were collected and arterial blood pressure was measured. Then, markers of leptin and insulin signaling, Tau phosphorylation, neuroinflammation, and synaptogenesis were measured by western blotting and immunohistochemistry. Results Fa/fa rats developed obesity, mild glucose intolerance, and peripheral insulin resistance but not hypertension while palm11-PrRP31 treatment neither lowered body weight nor attenuated glucose tolerance but ameliorated leptin and insulin signaling and synaptogenesis in hippocampus. Conclusion We demonstrated that palm11-PrRP31 had neuroprotective features without anti-obesity and glucose lowering effects in fa/fa rats. This data suggest that this analog has the potential to exert neuroprotective effect despite of leptin signaling disturbances in this rat model.

The study of ontogenetic aspects of water and electrolyte metabolism performed in the Institute of Physiology (Czechoslovak Academy of Sciences) led to the research on the increased susceptibility of immature rats to salt-dependent forms of hypertension since 1966. Hemodynamic studies in developing rats paved the way to the evaluation of hemodynamic mechanisms during the development of genetic hypertension in SHR. A particular attention was focused on altered renal function and kidney damage in both salt and genetic hypertension with a special respect to renin-angiotensin system. Renal damage associated with hypertension progression was in the center of interest of several research groups in Prague. The alterations in ion transport, cell calcium handling and membrane structure as well as their relationship to abnormal lipid metabolism were studied in a close cooperation with laboratories in Munich, Glasgow, Montreal and Paris. The role of NO and oxidative stress in various forms of hypertension was a subject of a joint research with our Slovak colleagues focused mainly on NO-deficient hypertension elicited by chronic L-NAME administration. Finally, we adopted a method enabling us to evaluate the balance of vasoconstrictor and vasodilator mechanisms in BP maintenance. Using this method we demonstrated sympathetic hyperactivity and relative NO deficiency in rats with either salt-dependent or genetic hypertension. At the end of the first decennium of this century we were ready to modify our traditional approach towards modern trends in the research of experimental hypertension. [ABSTRACT FROM AUTHOR]

Alzheimer’s disease (AD) is a progressive brain disorder characterized by extracellular amyloid-β (Aβ) plaques, intracellular neurofibrillary tangles formed by hyperphosphorylated Tau protein and neuroinflammation. Previous research has shown that obesity and type 2 diabetes mellitus, underlined by insulin resistance (IR), are risk factors for neurodegenerative disorders. In this study, obesity-induced peripheral and central IR and inflammation were studied in relation to AD-like pathology in the brains and periphery of APP/PS1 mice, a model of Aβ pathology, fed a high-fat diet (HFD). APP/PS1 mice and their wild-type controls fed either a standard diet or HFD were characterized at the ages of 3, 6 and 10 months by metabolic parameters related to obesity via mass spectroscopy, nuclear magnetic resonance, immunoblotting and immunohistochemistry to quantify how obesity affected AD pathology. The HFD induced substantial peripheral IR leading to central IR. APP/PS1-fed HFD mice had more pronounced IR, glucose intolerance and liver steatosis than their WT controls. The HFD worsened Aβ pathology in the hippocampi of APP/PS1 mice and significantly supported both peripheral and central inflammation. This study reveals a deleterious effect of obesity-related mild peripheral inflammation and prediabetes on the development of Aβ and Tau pathology and neuroinflammation in APP/PS1 mice.

Some species of two tribes (Anthidiini and Osmiini) of the bee family Megachilidae utilize empty gastropod shells as nesting cavities. While snail-nesting Osmiini have been more frequently studied and the nesting biology of several species is well-known, much less is known about the habits of snail-nesting Anthidiini. We collected nests of four species of the genus Rhodanthidium (R. septemdentatum, R. sticticum, R. siculum and R. infuscatum) in the Czech Republic, Slovakia, Catalonia (Spain) and Sicily (Italy). We dissected these nests in the laboratory and documented their structure, pollen sources and nest associates. The four species usually choose large snail shells. All four species close their nests with a plug made of resin, sand and fragments of snail shells. However, nests of the four species can be distinguished based on the presence (R. septemdentatum, R. sticticum) or absence (R. siculum, R. infuscatum) of mineral and plant debris in the vestibular space, and the presence (R. septemdentatum, R. infuscatum) or absence (R. sticticum, R. siculum) of a resin partition between the vestibular space and the brood cell. Rhodanthidium septemdentatum, R. sticticum and R. siculum usually build a single brood cell per nest, but all R. infuscatum nests studied contained two or more cells. For three of the species (R. siculum, R. septemdentatum and R. sticticum) we confirmed overwintering in the adult stage. Contrary to R. siculum, R. septemdentatum and R. sticticum do not hide their nest shells and usually use shells under the stones or hidden in crevices within stone walls. Nest associates were very infrequent. We only found two R. sticticum nests parasitized by the chrysidid wasp Chrysura refulgens and seven nests infested with pollen mites Chaetodactylus cf. anthidii. Our pollen analyses confirm that Rhodanthidium are polylectic but show a preference for Fabaceae by R. sticticum.

Abstract Background Obesity leads to low-grade inflammation in the adipose tissue and liver and neuroinflammation in the brain. Obesity-induced insulin resistance (IR) and neuroinflammation seem to intensify neurodegeneration including Alzheimer’s disease. In this study, the impact of high-fat (HF) diet-induced obesity on potential neuroinflammation and peripheral IR was tested separately in males and females of THY-Tau22 mice, a model of tau pathology expressing mutated human tau protein. Methods Three-, 7-, and 11-month-old THY-Tau22 and wild-type males and females were tested for mobility, anxiety-like behavior, and short-term spatial memory in open-field and Y-maze tests. Plasma insulin, free fatty acid, cholesterol, and leptin were evaluated with commercial assays. Liver was stained with hematoxylin and eosin for histology. Brain sections were 3′,3′-diaminobenzidine (DAB) and/or fluorescently detected for ionized calcium-binding adapter molecule 1 (Iba1), glial fibrillary acidic protein (GFAP), and tau phosphorylated at T231 (pTau (T231)), and analyzed. Insulin signaling cascade, pTau, extracellular signal-regulated kinase 1/2 (ERK1/2), and protein phosphatase 2A (PP2A) were quantified by western blotting of hippocampi of 11-month-old mice. Data are mean ± SEM and were subjected to Mann-Whitney t test within age and sex and mixed-effects analysis and Bonferroni’s post hoc test for age comparison. Results Increased age most potently decreased mobility and increased anxiety in all mice. THY-Tau22 males showed impaired short-term spatial memory. HF diet increased body, fat, and liver weights and peripheral IR. HF diet-fed THY-Tau22 males showed massive Iba1+ microgliosis and GFAP+ astrocytosis in the hippocampus and amygdala. Activated astrocytes colocalized with pTau (T231) in THY-Tau22, although no significant difference in hippocampal tau phosphorylation was observed between 11-month-old HF and standard diet-fed THY-Tau22 mice. Eleven-month-old THY-Tau22 females, but not males, on both diets showed decreased synaptic and postsynaptic plasticity. Conclusions Significant sex differences in neurodegenerative signs were found in THY-Tau22. Impaired short-term spatial memory was observed in 11-month-old THY-tau22 males but not females, which corresponded to increased neuroinflammation colocalized with pTau(T231) in the hippocampi and amygdalae of THY-Tau22 males. A robust decrease in synaptic and postsynaptic plasticity was observed in 11-month-old females but not males. HF diet caused peripheral but not central IR in mice of both sexes.

In this study, a new series of aliphatic, cyclic, and heterocyclic derivatives of haemanthamine was designed and synthesized to enhance its inhibitory effect on the proliferation and viability of cancer cells. A library of haemanthamine derivatives was subjected to 10 μM single-dose cytotoxicity screening against a panel of human cell lines of various histotypes. Initial cytotoxicity evaluation of the parent haemanthamine (1) and a series of twenty-nine (2–30) semisynthetic analogues showed that for some of the newly formed derivatives, a certain cytotoxic effect was observed, in one case even higher than that of the parent compound. Specifically, 11-O-(4-chloro-3-nitrobenzoyl)haemanthamine (21) showed an enhanced antiproliferative effect, where the mean growth percent (GP) value was 5% compared to haemanthamine, leading to a decrease in the GP to 25%. Among ten cell lines tested, derivative 21, bearing a substituted aromatic ester bond via C-11 of haemanthamine, had excellent activity for inhibiting the growth of HeLa (IC50 = 0.2 ± 0.1 μM), A549 (IC50 = 1.7 ± 0.1 μM) and HT-29 (IC50 = 2.2 ± 0.1 μM) cells. When evaluating response kinetics, we found that 21 and haemanthamine dose- and time-dependently suppressed the proliferation of A549 cells. In contrast to haemanthamine (1), Trypan blue and lactate dehydrogenase (LDH) release assay revealed that 21 was capable of reducing the survival of A549 cells.

Zucker fatty (fa/fa) rats represent a well-established and widely used model of genetic obesity. Because previous metabolomic studies have only been published for young fa/fa rats up to 20 weeks of age, which can be considered early maturity in male fa/fa rats, the aim of our work was to extend the metabolomic characterization to significantly older animals. Therefore, the urinary profiles of obese fa/fa rats and their lean controls were monitored using untargeted NMR metabolomics between 12 and 40 weeks of age. At the end of the experiment, the rats were also characterized by NMR and LC-MS serum analysis, which was supplemented by a targeted LC-MS analysis of serum bile acids and neurotransmitters. The urine analysis showed that most of the characteristic differences detected in young obese fa/fa rats persisted throughout the experiment, primarily through a decrease in microbial co-metabolite levels, the upregulation of the citrate cycle, and changes in nicotinamide metabolism compared with the age-related controls. The serum of 40-week-old obese rats showed a reduction in several bile acid conjugates and an increase in serotonin. Our study demonstrated that the fa/fa model of genetic obesity is stable up to 40 weeks of age and is therefore suitable for long-term experiments.

Tuberculosis remains a serious killer among infectious diseases due to its incidence, mortality, and occurrence of resistant mycobacterial strains. The challenge to discover new antimycobacterial agents forced us to prepare a series of N-(1-hydroxy-1,3-dihydrobenzo[c][1,2]oxaborol-6-yl)(hetero)aryl-2-carboxamides 1–19 via the acylation of 6-aminobenzo[c][1,2]oxaborol-1(3H)-ol with various activated (hetero)arylcarboxylic acids. These novel compounds have been tested in vitro against a panel of clinically important fungi and bacteria, including mycobacteria. Some of the compounds inhibited the growth of mycobacteria in the range of micromolar concentrations and retained this activity also against multidrug-resistant clinical isolates. Half the maximal inhibitory concentrations against the HepG2 cell line indicated an acceptable toxicological profile. No growth inhibition of other bacteria and fungi demonstrated selectivity of the compounds against mycobacteria. The structure–activity relationships have been derived and supported with a molecular docking study, which confirmed a selectivity toward the potential target leucyl-tRNA synthetase without an impact on the human enzyme. The presented compounds can become important materials in antimycobacterial research.

Understanding the spin dynamics in antiferromagnetic (AFM) thin films is fundamental for designing novel devices based on AFM magnon transport. Here, we study the magnon dynamics in thin films of AFM S=5/2 α-Fe_{2}O_{3} by combining resonant inelastic x-ray scattering, Anderson impurity model plus dynamical mean-field theory, and Heisenberg spin model. Below 100 meV, we observe the thickness-independent (down to 15 nm) acoustic single-magnon mode. At higher energies (100–500 meV), an unexpected sequence of equally spaced, optical modes is resolved and ascribed to ΔS_{z}=1, 2, 3, 4, and 5 magnetic excitations corresponding to multiple, noninteracting magnons. Our study unveils the energy, character, and momentum dependence of single and multimagnons in α-Fe_{2}O_{3} thin films, with impact on AFM magnon transport and its related phenomena. From a broader perspective, we generalize the use of L-edge resonant inelastic x-ray scattering as a multispin-excitation probe up to ΔS_{z}=2S. Our analysis identifies the spin-orbital mixing in the valence shell as the key element for accessing excitations beyond ΔS_{z}=1, and up to, e.g., ΔS_{z}=5. At the same time, we elucidate the novel origin of the spin excitations beyond the ΔS_{z}=2, emphasizing the key role played by the crystal lattice as a reservoir of angular momentum that complements the quanta carried by the absorbed and emitted photons.

Koletsky rats, the genetically obese strain of spontaneously hypertensive rats (SHROB), are the well-accepted animal model of human metabolic syndrome. They are characterized by early onset obesity, spontaneous hypertension, hyperinsulinemia, hyperlipidemia, proteinuria and shortened life-span. One of the factors in the pathogenesis of metabolic syndrome is oxidative stress. The aim of the present study was to compare two parameters related to oxidative stress: the levels of the main intracellular antioxidant, reduced glutathione as well as the indirect indicator of lipid peroxidation damage, thiobarbituric acid-reactive substances (TBARS) in heart, renal cortex and medulla and liver in male lean spontaneously hypertensive rats (SHR) and obese Koletsky rats. We did not find any significant differences in these markers in heart and kidneys. However, we found significantly lower glutathione level in Koletsky rat liver compared with SHR (5.03±0.23 vs. 5.83±0.14 µmol/g tissue, respectively). On the contrary, we observed significantly higher TBARS levels in Koletsky rat liver compared with SHR (28.56±2.15 vs. 21.83±1.60 nmol/mg protein, respectively). We conclude that the liver is the most sensitive tissue to oxidative damage with the significantly decreased concentration of glutathione and the significantly increased concentration of TBARS in obese Koletsky rats in comparison with lean control SHR. Ojcow birch (Betula oycoviensis Besser) is a rare Central European tree taxon, micro-populations of which are found in only several localities. With a view to maintaining the B. oycoviensis gene pool, this study tested the species’ potential for micropropagation, grafting, and propagation by cuttings. Plant material for vegetative propagation was collected from ten genotypes in the Czech Republic. In vitro culture was established from axillary buds surfaces sterilized with 0.1% HgCl2 and cultivated on woody plant (WP) medium supplemented with 1 mg l-1 6-benzylaminopurine (BAP). Two genotypes of the species were successfully multiplied by in vitro propagation using WP medium supplemented with 0-2 mg l-1 BAP. The BAP concentration of 1 mg l-1 proved to be optimal, yielding 2.5 new shoots per explant in genotype 516 and 3.5 shoots per explant in genotype 545. The shoots were rooted on half-strength Murashige and Skoog (MS) medium supplemented with various concentrations of α-naphthylacetic acid (NAA) and indole-3-butyric acid (IBA). The highest rooting percentages (72.5% and 77.5% for genotypes 516 and 545, respectively) were achieved on the medium with the combination of both auxins at concentrations of 0.3 mg l-1. The rooted plants were transferred ex vitro in substrate composed of sand, peat, and perlite (1:1:1) and acclimated in the greenhouse. After 4 weeks, more than 90% of plants survived. Grafting was carried out in spring using Betula pendula as rootstock. The efficiency of this technique ranged from 0% to 50% across genotypes, and 4 out of 10 genotypes were successfully propagated by grafting. The cuttings were treated with commercial root stimulators Stimulax I and Stimulator AS-1, planted in a mixture of peat and sand (1:1) in the greenhouse, and watered regularly. This technique resulted in 0% rooting, however, and no cutting survived until the end of the vegetation period. The results of this study show that protocols for in vitro propagation and grafting can be employed for effective mass propagation of B. oycoviensis, although these processes show genotype-dependent responses.

Wildfire occurrence is influenced by climate, vegetation and human activities. A key challenge for understanding the risk of fires is quantifying the mediating effect of vegetation on fire regimes. Here, we explore the relative importance of Holocene land cover, land use, dominant functional forest type, and climate dynamics on biomass burning in temperate and boreo-nemoral regions of central and eastern Europe over the past 12 kyr. We used an extensive data set of Holocene pollen and sedimentary charcoal records, in combination with climate simulations and statistical modelling. Biomass burning was highest during the early Holocene and lowest during the mid-Holocene in all three ecoregions (Atlantic, continental and boreo-nemoral) but was more spatially variable over the past 3–4 kyr. Although climate explained a significant variance in biomass burning during the early Holocene, tree cover was consistently the highest predictor of past biomass burning over the past 8 kyr. In temperate forests, biomass burning was high at ∼45 % tree cover and decreased to a minimum at between 60 % and 70 % tree cover. In needleleaf-dominated forests, biomass burning was highest at ∼ 60 %–65 % tree cover and steeply declined at >65 % tree cover. Biomass burning also increased when arable lands and grasslands reached ∼ 15 %–20 %, although this relationship was variable depending on land use practice via ignition sources, fuel type and quantities. Higher tree cover reduced the amount of solar radiation reaching the forest floor and could provide moister, more wind-protected microclimates underneath canopies, thereby decreasing fuel flammability. Tree cover at which biomass burning increased appears to be driven by warmer and drier summer conditions during the early Holocene and by increasing human influence on land cover during the late Holocene. We suggest that long-term fire hazard may be effectively reduced through land cover management, given that land cover has controlled fire regimes under the dynamic climates of the Holocene.

Using non-destructive techniques in investigating tree stem rots is a modern approach in arboriculture and urban forestry. We used PiCUS® 3 Sonic tomograph (SoT) and TreeTronic® electrical resistance tomograph (ERT) to inspect the health status of urban and park trees. The process of setting up the device and measuring is time demanding as it requires numerous delicate operations. The aim of the study was to evaluate the time needed for measurement and to propose an optimal workflow. The results of work sampling suggest that scanning of one average-difficulty tree by SoT and ERT resistance tomography takes an average approximately 52 min (when one operator measures one scan), and approx. 37 min (when two operators measure a queue of trees). Working in two-person-team is moderately more efficient. Typically, the overall costs of one scan are approximately EUR 25-30 (~ CZK 650-780), depending on many variables.

Prolactin-releasing peptide (PrRP) is an anorexigenic neuropeptide that has potential for the treatment of obesity and its complications. Recently, we designed a palmitoylated PrRP31 analog (palm11-PrRP31) that is more stable than the natural peptide and able to act centrally after peripheral administration. This analog acted as an anti-obesity and glucose-lowering agent, attenuating lipogenesis in rats and mice with high-fat (HF) diet-induced obesity. In Wistar Kyoto (WKY) rats fed a HF diet for 52 weeks, we explored glucose intolerance, but also prediabetes, liver steatosis and insulin resistance-related changes, as well as neuroinflammation in the brain. A potential beneficial effect of 6 weeks of treatment with palm11-PrRP31 and liraglutide as comparator was investigated. Liver lipid profiles, as well as urinary and plasma metabolomic profiles, were measured by lipidomics and metabolomics, respectively. Old obese WKY rats showed robust glucose intolerance that was attenuated by palm11-PrRP31, but not by liraglutide treatment. On the contrary, liraglutide had a beneficial effect on insulin resistance parameters. Despite obesity and prediabetes, WKY rats did not develop steatosis owing to HF diet feeding, even though liver lipogenesis was enhanced. Plasma triglycerides and cholesterol were not increased by HFD feeding, which points to unincreased lipid transport from the liver. The liver lipid profile was significantly altered by a HF diet that remained unaffected by palm11-PrRP31 or liraglutide treatment. The HF-diet-fed WKY rats revealed astrogliosis in the brain cortex and hippocampus, which was attenuated by treatment. In conclusion, this study suggested multiple beneficial anti-obesity-related effects of palm11-PrRP31 and liraglutide in both the periphery and brain.

Obesity and type 2 diabetes mellitus (T2DM) are preconditions for the development of metabolic syndrome, which is reaching pandemic levels worldwide, but there are still only a few anti-obesity drugs available. One of the promising tools for the treatment of obesity and related metabolic complications is anorexigenic peptides, such as prolactin-releasing peptide (PrRP). PrRP is a centrally acting neuropeptide involved in food intake and body weight (BW) regulation. In its natural form, it has limitations for peripheral administration; thus, we designed analogs of PrRP lipidized at the N-terminal region that showed high binding affinities, increased stability and central anorexigenic effects after peripheral administration. In this review, we summarize the preclinical results of our chronic studies on the pharmacological role of the two most potent palmitoylated PrRP31 analogs in various mouse and rat models of obesity, glucose intolerance, and insulin resistance. We used mice and rats with diet-induced obesity fed a high-fat diet, which is considered to simulate the most common form of human obesity, or rodent models with leptin deficiency or disrupted leptin signaling in which long-term food intake regulation by leptin is distorted. The rodent models described in this review are models of metabolic syndrome with different severities, such as obesity or morbid obesity, prediabetes or diabetes and hypertension. We found that the effects of palmitoylated PrRP31 on food intake and BW but not on glucose intolerance require intact leptin signaling. Thus, palmitoylated PrRP31 analogs have potential as therapeutics for obesity and related metabolic complications.

Clivia miniata (Amaryllidaceae) is an herbaceous evergreen flowering plant that is endemic to South Africa and Swaziland and belongs to one of the top-10 traded medicinal plants in informal medicine markets in South Africa. The species has been reported as the most important component of a traditional healer’s pallet of healing plants. Eighteen known Amaryllidaceae alkaloids (AAs) of various structural types, and one undescribed alkaloid of homolycorine-type, named clivimine B (3), were isolated from Clivia miniata. The chemical structures of the isolated alkaloids were elucidated by a combination of MS, HRMS, 1D and 2D NMR techniques and by comparison with literature data. Compounds isolated in a sufficient quantity, and not tested previously, were evaluated for their in vitro acetylcholinesterase (AChE; E.C. 3.1.1.7) and butyrylcholinesterase (BuChE; E.C. 3.1.1.8) inhibition activities.

Tuberculosis (TB) is a widespread infectious disease caused by Mycobacterium tuberculosis. The increasing incidence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains has created a need for new antiTB agents with new chemical scaffolds to combat the disease. Thus, the key question is: how to search for new antiTB and where to look for them? One of the possibilities is to search among natural products (NPs). In order to search for new antiTB drugs, the detailed phytochemical study of the whole Dicranostigma franchetianum plant was performed isolating wide spectrum of isoquinoline alkaloids (IAs). The chemical structures of the isolated alkaloids were determined by a combination of MS, HRMS, 1D, and 2D NMR techniques, and by comparison with literature data. Alkaloids were screened against Mycobacterium tuberculosis H37Ra and four other mycobacterial strains (M. aurum, M. avium, M. kansasii, and M. smegmatis). Alkaloids 3 and 5 showed moderate antimycobacterial activity against all tested strains (MICs 15.625–31.25 µg/mL). Furthermore, ten semisynthetic berberine (16a–16k) derivatives were developed and tested for antimycobacterial activity. In general, the derivatization of berberine was connected with a significant increase in antimycobacterial activity against all tested strains (MICs 0.39–7.81 μg/mL). Two derivatives (16e, 16k) were identified as compounds with micromolar MICs against M. tuberculosis H37Ra (MIC 2.96 and 2.78 µM). All compounds were also evaluated for their in vitro hepatotoxicity on a hepatocellular carcinoma cell line (HepG2), exerting lower cytotoxicity profile than their MIC values, thereby potentially reaching an effective concentration without revealing toxic side effects.

Obesity, diabetes, insulin resistance, sedentary lifestyle, and Western diet are the key factors underlying non-alcoholic fatty liver disease (NAFLD), one of the most common liver diseases in developed countries. In many cases, NAFLD further progresses to non-alcoholic steatohepatitis (NASH), fibrosis, cirrhosis, and to hepatocellular carcinoma. The hepatic lipotoxicity and non-liver factors, such as adipose tissue inflammation and gastrointestinal imbalances were linked to evolution of NAFLD. Nowadays, the degree of adipose tissue inflammation was shown to directly correlate with the severity of NAFLD. Consumption of higher caloric intake is increasingly emerging as a fuel of metabolic inflammation not only in obesity-related disorders but also NAFLD. However, multiple causes of NAFLD are the reason why the mechanisms of NAFLD progression to NASH are still not well understood. In this review, we explore the role of food intake regulating peptides in NAFLD and NASH mouse models. Leptin, an anorexigenic peptide, is involved in hepatic metabolism, and has an effect on NAFLD experimental models. Glucagon-like peptide-1 (GLP-1), another anorexigenic peptide, and GLP-1 receptor agonists (GLP-1R), represent potential therapeutic agents to prevent NAFLD progression to NASH. On the other hand, the deletion of ghrelin, an orexigenic peptide, prevents age-associated hepatic steatosis in mice. Because of the increasing incidence of NAFLD and NASH worldwide, the selection of appropriate animal models is important to clarify aspects of pathogenesis and progression in this field.

The Jizera Mountains (Jizerské hory) are a part of the Black Triangle, which, in the past, was one of Europe's most polluted regions. Situated on the Czech–Polish border, these mountains were heavily affected by extreme SO2 and NOx loads emitted from coal power plants in the piedmont. During the 1970s and 1980s, the upper plateau of the Jizera Mountains experienced substantial forest decline due to air pollution. Dying stands were felled on more than 12,000 ha. Modernization of the energy industry after 1989 has led to a significant reduction in air pollution in the Black Triangle. Therefore, replanting the clear-cut areas in the Jizera Mountains became possible during the 1990s, and a new generation of forests has covered the upper plateau of the mountains. However, these even-aged, mainly spruce stands urgently need to be diversified in terms of age, structure, and species composition. This is not an easy task due to extreme microclimate, acidified soils, and damage to plants by rodents and deer. In 2007, a project aimed at the diversification of local ecosystems was initiated. The project is based on a system of diversification centers and corridors containing a species admixture (broadleaf trees and silver fir), which is protected from game, to form a web that enriches the age and spatial structure of forests on the upper plateau and complements their species composition. Initially, these centers and corridors were placed in more sheltered and accessible places and planted with the standard planting stock in combination with large-sized transplants (

Birches are generally known for their high genetic and morphological variability, which has resulted in the description of many species. Ojców birch was described in 1809 by Willibald Suibert Joseph Gottlieb Besser in Poland. Since then, several studies assessing its taxonomy were conducted. Today, various authors present Ojców birch at different taxonomic ranks. In Czechia, the Ojców birch is classified a critically endangered taxon and confirmed at one locality consisting of several tens of individuals. However, before a strategy for its conservation can be applied, we consider it necessary to assess the taxonomic position of the endangered Czech population and to evaluate its relationship to the original Polish population. This study aimed to evaluate the morphometric and genetic variability between populations of B. ×oycoviensis in Poland and the Czechia and their relationship to regional populations of B. pendula, one of the putative parental species of the Ojców birch. Altogether, 106 individuals were sampled, including the holotype of B. szaferi, the second putative parental species of B. ×oycoviensis, received from the herbarium of W. Szafer, which is deposited at the Institute of Botany in Kraków. Morphological analyses identified differences in leaves between B. ×oycoviensis and B. pendula. However, no significant differences were found in genome size between selected taxa/working units except for B. pendula sampled in Czechia. The identified difference of the Czech population of B. pendula is probably caused by population variability. Genetic variability between all the taxa under comparison, regardless of their origin, was also very low; only the benchmark taxa (B. nana and B. humilis) clearly differed from all samples analyzed. The results indicate minute morphological and negligible genetic variability between the Czech and Polish populations of B. ×oycoviensis. In light of our results, the classification of B. ×oycoviensis as B. pendula var. oycoviensis seems more accurate than all hitherto presented alternatives (e.g. B. ×oycoviensis as a separate species).

We present a comprehensive study of CaCu_{3}Ru_{4}O_{12} using bulk sensitive hard and soft x-ray spectroscopy combined with local-density approximation+dynamical mean-field theory (DMFT) calculations. Correlation effects on both the Cu and Ru ions can be observed. From the Cu 2p core-level spectra, we deduce the presence of magnetic Cu^{2+} ions hybridized with a reservoir of itinerant electrons. The strong photon energy dependence of the valence band allows us to disentangle the Ru, Cu, and O contributions and, thus, to optimize the DMFT calculations. The calculated spin and charge susceptibilities show that the transition metal oxide CaCu_{3}Ru_{4}O_{12} must be classified as a Kondo system and that the Kondo temperature is in the range of 500–1000 K.

Infrared thermography (IRT) is a noninvasive and safe method of displaying the temperature map of objects that can be used to detect hoof diseases and lameness to reduce significant financial costs and physically stress animals. A qualitative bibliometric method based on the analysis of publications by the authors themselves using sophisticated tools of scientific databases was applied in this work. This review presents the fundamentals of IRT as well as recent developments in IRT detection in dairy science, including preprocessing, segmentation, and classification of objects in IRT images. In addition, recent studies dealing with the detection of hoof diseases and lameness using IRT are reviewed. As a result of this study, select previous studies are confronted in terms of technical aspects of IRT measurements such as emissivity, distance, temperature range, and reflected air temperature. Subsequently, recommendations for future IRT measurements are discussed.

The aim of this work was to suggest a reliable method for distinguishing between diploid and tetraploid taxa of the genus Betula Linnaeus, based on leaf measurements. In total, 97 individuals from 6 locations within the Bohemian Forest region (Czech Republic) were selected. Four leaves from each individual were evaluated. On each leaf, twenty parameters were measured. Each sample was analysed by flow cytometry to determine its actual ploidy. Measured parameters were analysed by principal component analysis and tested for differences between diploid and tetraploid taxa. For actual ploidy prediction, a classification function was designed. The reliability of the classification function was verified on samples from three different regions of the Czech Republic and compared with functions as suggested by other authors. The classification function designed in this work (based on 3 parameters - blade width in the upper 1/4 of blade length, first vein angle and number of leaf teeth between 3rd and 4th vein) correctly determined actual ploidy in 89% of all tested samples.

The technology for intensive broiler breeding using deep litter method provides convenient conditions for production of odour substances inside the barn. As a consequence, there are relatively high odour emissions in the breeding area and its surrounding, which has significant impacts on the life quality of both people and animals. The amount of produced emissions increases in time because it is closely related to the amount of poultry droppings accumulated in litter inside the barn. This paper deals with changes in odour intensity depending on time measured since the beginning of broiler fattening. Odour intensity was measured by methods of dynamic olfactometry. The estimated values gradually increased from 45 ouE·m-3 to the highest value of 259 ouE·m-3, which was determined in the final fattening phase (broilers’ age 33 days). The calculated odour substances emission values were in range from 0.02 ouE·s-1·bird-1 at the beginning of fattening up to 0.10 ouE·s-1·bird-1 in the final fattening phase. Odour emissions increased five times during the fattening period.

The European Pollen Database (EPD) is a community effort to archive and make available pollen sequences from across the European continent. Pollen sequences provide records that may be used to infer past vegetation and vegetation change. We present here maps based on 828 sites from the EPD giving an overview of changes in postglacial pollen assemblages in Europe over the past 15,000 years. The maps show the distribution and abundance of 54 different pollen taxa at 500 year intervals, supported by new age-depth models and associated chronological uncertainty analysis. Results show the individualistic patterns of spread of different pollen taxa, and provide a standardized dataset for further analysis, defining a spatial context for the study of past plant and vegetation changes and other aspects of environmental history in Europe.

The search for novel antimycobacterial drugs is a matter of urgency, since tuberculosis is still one of the top ten causes of death from a single infectious agent, killing more than 1.4 million people worldwide each year. Nine Amaryllidaceae alkaloids (AAs) of various structural types have been screened for their antimycobacterial activity. Unfortunately, all were considered inactive, and thus a pilot series of aromatic esters of galanthamine, 3-O-methylpancracine, vittatine and maritidine were synthesized to increase biological activity. The semisynthetic derivatives of AAs were screened for their in vitro antimycobacterial activity against Mycobacterium tuberculosis H37Ra and two other mycobacterial strains (M. aurum, M. smegmatis) using a modified Microplate Alamar Blue Assay. The most active compounds were also studied for their in vitro hepatotoxicity on the hepatocellular carcinoma cell line HepG2. In general, the derivatization of the original AAs was associated with a significant increase in antimycobacterial activity. Several pilot derivatives were identified as compounds with micromolar MICs against M. tuberculosis H37Ra. Two derivatives of galanthamine, 1i and 1r, were selected for further structure optimalization to increase the selectivity index.

Motivated by recent core-level x-ray photoemission spectroscopy, x-ray absorption spectroscopy (XAS), and resonant inelastic x-ray scattering (RIXS) experiments for the newly discovered superconducting infinite-layer nickelate, we investigate the core-level spectra of the parent compounds NdNiO_{2} and LaNiO_{2} using the combination of local density approximation and dynamical mean-field theory (LDA+DMFT). Adjusting a charge-transfer energy to match the experimental spectra, we determine the optimal model parameters and discuss the nature of the NdNiO_{2} ground state. We find that self-doping from the Nd 5d states in the vicinity of the Fermi energy prohibits opening of a Mott-Hubbard gap in NdNiO_{2}. The present Ni L_{3} XAS and RIXS calculation for LaNiO_{2} cannot explain the difference from NdNiO_{2} spectra.

Alzheimer’s disease (AD) is a multifactorial neurodegenerative condition of the central nervous system (CNS) that is currently treated by cholinesterase inhibitors and the N-methyl-d-aspartate receptor antagonist, memantine. Emerging evidence strongly supports the relevance of targeting butyrylcholinesterase (BuChE) in the more advanced stages of AD. Within this study, we have generated a pilot series of compounds (1–20) structurally inspired from belladine-type Amaryllidaceae alkaloids, namely carltonine A and B, and evaluated their acetylcholinesterase (AChE) and BuChE inhibition properties. Some of the compounds exhibited intriguing inhibition activity for human BuChE (hBuChE), with a preference for BuChE over AChE. Seven compounds were found to possess a hBuChE inhibition profile, with IC50 values below 1 µM. The most potent one, compound 6, showed nanomolar range activity with an IC50 value of 72 nM and an excellent selectivity pattern over AChE, reaching a selectivity index of almost 1400. Compound 6 was further studied by enzyme kinetics, along with in-silico techniques, to reveal the mode of inhibition. The prediction of CNS availability estimates that all the compounds in this survey can pass through the blood-brain barrier (BBB), as disclosed by the BBB score.

The anorexigenic neuropeptide prolactin-releasing peptide (PrRP) is involved in the regulation of food intake and energy expenditure. Lipidization of PrRP stabilizes the peptide, facilitates central effect after peripheral administration and increases its affinity for its receptor, GPR10, and for the neuropeptide FF (NPFF) receptor NPFF-R2. The two most potent palmitoylated analogs with anorectic effects in mice, palm11-PrRP31 and palm-PrRP31, were studied in vitro to determine their agonist/antagonist properties and mechanism of action on GPR10, NPFF-R2 and other potential off-target receptors related to energy homeostasis. Palmitoylation of both PrRP31 analogs increased the binding properties of PrRP31 to anorexigenic receptors GPR10 and NPFF-R2 and resulted in a high affinity for another NPFF receptor, NPFF-R1. Moreover, in CHO-K1 cells expressing GPR10, NPFF-R2 or NPFF-R1, palm11-PrRP and palm-PrRP significantly increased the phosphorylation of extracellular signal-regulated kinase (ERK), protein kinase B (Akt) and cAMP-responsive element-binding protein (CREB). Palm11-PrRP31, unlike palm-PrRP31, did not activate either c-Jun N-terminal kinase (JNK), p38, c-Jun, c-Fos or CREB pathways in cells expressing NPFF-1R. Palm-PrRP31 also has higher binding affinities for off-target receptors, namely, the ghrelin, opioid (KOR, MOR, DOR and OPR-L1) and neuropeptide Y (Y1, Y2 and Y5) receptors. Palm11-PrRP31 exhibited fewer off-target activities; therefore, it has a higher potential to be used as an anti-obesity drug with anorectic effects.
