Patients undergoing major orthopedic surgeries, including total hip replacement, total knee replacement, and hip fracture surgery, represent a group at particularly high risk of venous thromboembolism. Therefore, prophylaxis for thromboembolic events has been of great concern to surgeons. Edoxaban is a novel, orally available, and highly specific and direct factor Xa inhibitor. This new agent was approved for the prevention of venous thromboembolism in patients undergoing major orthopedic surgery, including total hip replacement, total knee replacement, and hip fracture surgery, by the Japanese Ministry of Health, Labor, and Welfare in 2011. Preclinical and Phase I clinical trials demonstrated several promising properties. Its rapid absorption and short life-time in blood are known. Edoxaban inhibits factor Xa activity directly and selectively. It also has a strong antithrombotic effect without any influence of food intake. Coagulation monitoring is not required. Edoxaban has predictable linear pharmacokinetic and pharmacodynamic profiles. Phase II and III clinical trials have been completed to examine its efficacy and safety in patients undergoing major orthopedic surgery. In these clinical trials, oral administration of edoxaban showed efficacy superior to that of oral placebo or subcutaneously administered dalteparin or enoxaparin. © 2015 The Author(s). The BioMart Community Portal (www.biomart.org) is a community-driven effort to provide a unified interface to biomedical databases that are distributed worldwide. The portal provides access to numerous database projects supported by 30 scientific organizations. It includes over 800 different biological datasets spanning genomics, proteomics, model organisms, cancer data, ontology information and more. All resources available through the portal are independently administered and funded by their host organizations. The BioMart data federation technology provides a unified interface to all the available data. The latest version of the portal comes with many new databases that have been created by our ever-growing community. It also comes with better support and extensibility for data analysis and visualization tools. A new addition to our toolbox, the enrichment analysis tool is now accessible through graphical and web service interface. The BioMart community portal averages over one million requests per day. Building on this level of service and the wealth of information that has become available, the BioMart Community Portal has introduced a new, more scalable and cheaper alternative to the large data stores maintained by specialized organizations.

Abstract

Naturally occurring regulatory T (Treg) cells, which specifically express the transcription factor forkhead box P3 (Foxp3), are engaged in the maintenance of immunological self-tolerance and homeostasis. By transcriptional start site cluster analysis, we assessed here how genome-wide patterns of DNA methylation or Foxp3 binding sites were associated with Treg-specific gene expression. We found that Treg-specific DNA hypomethylated regions were closely associated with Treg up-regulated transcriptional start site clusters, whereas Foxp3 binding regions had no significant correlation with either up- or down-regulated clusters in nonactivated Treg cells. However, in activated Treg cells, Foxp3 binding regions showed a strong correlation with down-regulated clusters. In accordance with these findings, the above two features of activation-dependent gene regulation in Treg cells tend to occur at different locations in the genome. The results collectively indicate that Treg-specific DNA hypomethylation is instrumental in gene up-regulation in steady state Treg cells, whereas Foxp3 down-regulates the expression of its target genes in activated Treg cells. Thus, the two events seem to play distinct but complementary roles in Treg-specific gene expression.

Abstract

Obesity confers an increased risk of developing specific cancer forms. Although the mechanisms are unclear, increased fat cell secretion of specific proteins (adipokines) may promote/facilitate development of malignant tumors in obesity via cross-talk between adipose tissue(s) and the tissues prone to develop cancer among obese. We searched for novel adipokines that were overexpressed in adipose tissue of obese subjects as well as in tumor cells derived from cancers commonly associated with obesity. For this purpose expression data from human adipose tissue of obese and non-obese as well as from a large panel of human cancer cell lines and corresponding primary cells and tissues were explored. We found expression of ceruloplasmin to be the most enriched in obesity-associated cancer cells. This gene was also significantly up-regulated in adipose tissue of obese subjects. Ceruloplasmin is the body's main copper carrier and is involved in angiogenesis. We demonstrate that ceruloplasmin is a novel adipokine, which is produced and secreted at increased rates in obesity. In the obese state, adipose tissue contributed markedly (up to 22%) to the total circulating protein level. In summary, we have through bioinformatic screening identified ceruloplasmin as a novel adipokine with increased expression in adipose tissue of obese subjects as well as in cells from obesity-associated cancers. Whether there is a causal relationship between adipose overexpression of ceruloplasmin and cancer development in obesity cannot be answered by these cross-sectional comparisons.

40. Raman scattering from vibrational modes in Si46 clathrates

Abstract

Room-temperature Raman-scattering spectra are reported for the type-II superconductors MxBaySi46 (M =Na,K) that were recently shown to exhibit Tc's ∼3.5 K. These spectra are compared to those of Na8Si46 and K7Si46 clathrates that exhibit normal metallic behavior down to 2 K. In the Si46 system, fifteen of the eighteen Si related first-order Raman frequencies predicted by group theory have been detected, and the frequencies are found to be sensitive to the particular dopants. The Raman linewidths observed for the MxBaySi46 system are comparable to those observed for Na8Si46 and K7Si46. The data, taken collectively, suggest that the line broadening in the metallic Si clathrates is due to important contributions from both the electron-phonon interaction as well as from a random filling of the Si cages and charge-transfer disorder.

Abstract

Background: BioHackathon 2010 was the third in a series of meetings hosted by the Database Center for Life Sciences (DBCLS) in Tokyo, Japan. The overall goal of the BioHackathon series is to improve the quality and accessibility of life science research data on the Web by bringing together representatives from public databases, analytical tool providers, and cyber-infrastructure researchers to jointly tackle important challenges in the area of in silico biological research.Results: The theme of BioHackathon 2010 was the 'Semantic Web', and all attendees gathered with the shared goal of producing Semantic Web data from their respective resources, and/or consuming or interacting those data using their tools and interfaces. We discussed on topics including guidelines for designing semantic data and interoperability of resources. We consequently developed tools and clients for analysis and visualization.Conclusion: We provide a meeting report from BioHackathon 2010, in which we describe the discussions, decisions, and breakthroughs made as we moved towards compliance with Semantic Web technologies - from source provider, through middleware, to the end consumer.

Clinical application of alumina ceramic to femoral head of prostheses has been performed with possible advantage of lower friction of ceramic head / ultra-high molecular weight polyethylene (UHMWPE) or ceramic head /ceramic cup articulations, when compared to that of metal head / UHMWPE cup. Although favorable clinical results of alumina ceramic head / UHMWPE cup combination has been known, breakage of alumina ceramic head has been reported. In this study, we investigated the series of total hip arthroplasties (THAs) with alumina ceramic head, and reported breakage of alumina ceramic head combined with UHMWPE cup and critical clinical problems after tiny modification of the taper system between alumina ceramics head and metal neck. Abstract

The low-temperature isobaric heat capacity (CP) of synthetic stishovite was measured between 2 and 311 K by the thermal relaxation method. The measured CPP is considerably smaller than that of Holm et al. (1967) over the whole temperature range studied. The standard entropy, Due to image rights restrictions, multiple line equation(s) cannot be graphically displayed., of stishovite obtained is 24.0 J/(mol⋅K), which is 3.8 J/(mol⋅K) lower than that of Holm et al. (1967). Using the measured CP, the Debye temperature and thermal Grüneisen parameter at 298.15 K were calculated to be 1109 K and 1.68, respectively. The equilibrium coesite-stishovite transition boundary was calculated using the Due to image rights restrictions, multiple line equation(s) cannot be graphically displayed. of stishovite from published thermodynamic data. The calculated boundary has a slope of 3.2 ± 0.1 MPa/K at 1200-1600 K, which is larger than the slope determined by the high-pressure in situ X-ray diffraction study by Zhang et al. (1996). Abstract

The heat capacity of PbMO3 ( M=Ti, Zr and Hf) at constant pressure was measured using a differential scanning calorimeter (DSC) from room temperature up to 870 K. Large anomalies were found in the heat capacity curves, corresponding to the ferroelectricparaelectric phase transition in PbTiO3 (PT), the antiferroelectric-paraelectric phase transitions in PbZrO3 (PZ) and PbHfO3 (PH). The transition entropies were estimated as 7.3 J K−1 mol−1 (PT), 9.9 J K−1 mol−1 (PZ) and 9.3 J K−1 mol−1 (PH). Abstract

The phase transitions of α,α-trehalose dihydrate ( T h) were investigated by either differential thermal analysis (DTA) with an in-house apparatus of variable-pressure type equipped with an open sample holder or commercially available TG (thermal gravimetry)-DTA apparatus for comparison under the same experimental conditions as to the heating rate (2°C min−1), the type of pan (open), and the particle size of T h (63 μm). The former DTA measurements were carried out under five different total pressures, 101, 75, 61, 48 and 35 kPa, which provided quite helpful information necessary for confirmative assignments of the endothermic peaks due to either melting or dehydration of T h. The usage of largely different amount of T h, 126 and 14 mg for the DTA and TG-DTA measurements respectively, led to their different DTA traces, showing that there were largely different extents of the influence by the measured sample surface exposed to the surrounding atmosphere on its dehydration behavior. In addition the high thermal sensitivity achieved with such mass of T h gave rise to an interesting discovery of an unidentified thermal event at 92°C prior to either melting or dehydration of T h.
