1. AMP‐dependent kinase stimulates the expression of αKlotho

Abstract

Renal αKlotho along with fibroblast growth factor 23 regulates phosphate and vitamin D metabolism. Its cleavage yields soluble Klotho controlling intracellular processes. αKlotho has anti‐inflammatory and antioxidant effects and is nephro‐ and cardioprotective. AMP‐dependent kinase (AMPK) is a nephro‐ and cardioprotective energy sensor. Given that both αKlotho and AMPK have beneficial effects in similar organs, we studied whether AMPK regulates αKlotho gene expression in Madin–Darby canine kidney, normal rat kidney 52E, and human kidney 2 cells. Using quantitative real‐time PCR and western blotting, we measured αKlotho expression upon pharmacological manipulation or siRNA‐mediated knockdown of AMPKα. AMPK activator 5‐aminoimidazole‐4‐carboxamide ribonucleoside (AICAR) enhanced αKlotho expression, an effect reduced in the presence of AMPK inhibitor compound C or siRNA targeting AMPK catalytic α‐subunits (α1 and α2). Similarly, AMPK activators metformin and phenformin upregulated αKlotho transcripts. Taken together, our results suggest that AMPK is a powerful inducer of αKlotho and could thereby contribute to the development of future therapeutic interventions.

Abstract

Objective: Endothelin receptor B (ETB) together with ETA mediates cellular effects of endothelin 1 (ET-1), an autocrine and endocrine peptide produced by the endothelium and other cells. It regulates vascular tone and controls kidney function. Metabolic syndrome is due to high caloric intake and is characterized by insulin resistance, dyslipidemia, and white adipose tissue (WAT) accumulation. ETA/ETB antagonism has been demonstrated to favorably influence insulin resistance. Our study explored the role of ETB in metabolic syndrome. Methods: Wild type (etb+/+) and rescued ETB-deficient (etb−/−) mice were fed a high-fat diet, and energy, glucose, and insulin metabolism were analyzed, and hormones and lipids measured in serum and tissues. Cell culture experiments were performed in HepG2 cells. Results: Compared to etb+/+ mice, etb−/− mice exhibited better glucose tolerance and insulin sensitivity, less WAT accumulation, lower serum triglycerides, and higher energy expenditure. Protection from metabolic syndrome was paralleled by higher hepatic production of fibroblast growth factor 21 (FGF21) and higher serum levels of free thyroxine (fT4), stimulators of energy expenditure. Conclusions: ETB deficiency confers protection from metabolic syndrome by counteracting glucose intolerance, dyslipidemia, and WAT accumulation due to enhanced energy expenditure, effects at least in part dependent on enhanced production of thyroid hormone/FGF21. ETB antagonism may therefore be a novel therapeutic approach in metabolic syndrome.

Abstract

BackgroundNumerous mobile health apps are marketed globally, and these have specific features including physical activity tracking, motivational feedback, and recipe provision. It is important to understand which features individuals prefer and whether these preferences differ between consumer groups. ObjectiveIn this study, we aimed to identify consumers’ most preferred features and rewards for a mobile app that targets healthy eating and physical activity and to reduce the number of individual mobile health app features to a smaller number of key categories as perceived by consumers. In addition, we investigated the impact of differences in consumers’ BMI and self-efficacy on their intention to use and willingness to pay for such an app. Finally, we identified the characteristics of different target groups of consumers and their responses toward app features via cluster analysis. MethodsA total of 212 participants from France, Italy, the United Kingdom, and Germany were recruited via the web to answer questions about app features, motivation, self-efficacy, demographics, and geographic factors. It is important to note that our study included an evenly distributed sample of people in the age range of 23 to 50 years (23-35 and 35-50 years). The app features in question were generated from a 14-day cocreation session by a group of consumers from the United Kingdom and the Republic of Ireland. Results“Home work out suggestions,” “exercise tips,” and “progress charts” were the most preferred app features, whereas “gift vouchers” and “shopping discounts” were the most preferred rewards. “Connections with other communication apps” was the least preferred feature, and “charitable giving” was the least preferred reward. Importantly, consumers’ positive attitude toward the “social support and connectedness and mindfulness” app feature predicted willingness to pay for such an app (β=.229; P=.004). Differences in consumers’ health status, motivational factors, and basic demographics moderated these results and consumers’ intention to use and willingness to pay for such an app. Notably, younger and more motivated consumers with more experience and knowledge about health apps indicated more positive attitudes and intentions to use and willingness to pay for this type of app. ConclusionsThis study indicated that consumers tend to prefer app features that are activity based and demonstrate progress. It also suggested a potential role for monetary rewards in promoting healthy lifestyle behaviors. Moreover, the results highlighted the role of consumers’ health status, motivational factors, and socioeconomic status in predicting their app use. These results provide up-to-date, practical, and pragmatic information for the future design and operation of mobile health apps.

Abstract

Merkel cell carcinoma (MCC) is a very rare, but highly aggressive skin cancer which occurs mainly in elderly patients. MCC cells show an expression pattern of three cell lineages: epithelial, neuroendocrine, and B-cell progenitor. This trilinear expression pattern suggests stemness activity in MCC. The etiopathogenesis of MCC is either linked to the Merkel cell polyomavirus (MCPyV) or in a smaller proportion (20%) to high levels of UV-induced somatic mutations. Both viral presence and accumulation of mutations have been shown to be associated with accelerated DNA methylation Age (DNAmAge) compared to chronological age. The MCC DNAmAge was significantly lower compared to the chronological age, which was irrespective of the viral presence or mutational burden. Although these features indicate some aspects of stemness in MCC cells, gene-expression-based pluripotency testing did not provide evidence for pluripotency of MCC cells.

Abstract

Temporary goals modulate attention to threat. We examined whether attentional bias to angry faces differs depending on whether a temporary background goal is neutral, or threat related, whilst also measuring social anxiety. Participants performed a dot probe task combined with a separate task that induced a temporary goal. Depending on the phase in this goal task, the goal made angry faces or neutral stimuli (i.e., houses) relevant. The dot probe task measured attention to combinations of angry faces, neutral but goal-relevant stimuli (i.e., houses), and neutral control stimuli. Attention was allocated to angry faces when an angry goal was active. This was more pronounced for people scoring high on social phobia. The neutral goal attenuated attention to angry faces and effects of social phobia were no longer apparent. These findings suggest that individual differences in social anxiety interact with current and temporary goals to affect attentional processes.

Abstract

It is estimated that more than half of all online transactions are abandoned before completion. This paper investigates the psychological factors that influence online shopping behavior, with a view to improving transactional success rates. Through a review of the literature, we identify a range of factors which predict abandonment of online shopping, highlighting affective and motivational dimensions in addition to processing style and characteristics of the consumer, device, and product. We conclude that online purchasing and payment systems that boost consumers’ motivation to buy and prevent or attenuate negative affective states will demonstrate the greatest rates of transactional success. However, with rapid advancement in technology, continued research is needed to fully understand the potential impact on future online purchasing behavior.

Abstract

The current paper investigates the value and application of a range of physiological and neuroscientific techniques in applied marketing research and consumer science, highlighting new insights from research in social psychology and neuroscience. We review measures of sweat secretion, heart rate, facial muscle activity, eye movements, and electrical brain activity, using techniques including skin conductance, pupillometry, eyetracking, and magnetic brain imaging. For each measure, after a brief explanation of the underlying technique, we illustrate concepts and mechanisms that the measure allows researchers in marketing and consumer science to investigate, with a focus on consumer attitudes and behavior. By providing reviews on recent research that applied these methods in consumer science and relevant related fields, we also highlight methodological and theoretical strengths and limitations, with an emphasis on ecological validity. We argue that the inclusion of physiological and neuroscientific techniques can advance consumer research by providing insights into the often unconscious mechanisms underlying consumer behavior. Therefore, such technologies can help researchers and marketing practitioners understand the mechanisms of consumer behavior and improve predictions of consumer behavior.

9. Detailing radio frequency heating induced by coronary stents: a 7.0 Tesla magnetic resonance study.

Abstract

The sensitivity gain of ultrahigh field Magnetic Resonance (UHF-MR) holds the promise to enhance spatial and temporal resolution. Such improvements could be beneficial for cardiovascular MR. However, intracoronary stents used for treatment of coronary artery disease are currently considered to be contra-indications for UHF-MR. The antenna effect induced by a stent together with RF wavelength shortening could increase local radiofrequency (RF) power deposition at 7.0 T and bears the potential to induce local heating, which might cause tissue damage. Realizing these constraints, this work examines RF heating effects of stents using electro-magnetic field (EMF) simulations and phantoms with properties that mimic myocardium. For this purpose, RF power deposition that exceeds the clinical limits was induced by a dedicated birdcage coil. Fiber optic probes and MR thermometry were applied for temperature monitoring using agarose phantoms containing copper tubes or coronary stents. The results demonstrate an agreement between RF heating induced temperature changes derived from EMF simulations versus MR thermometry. The birdcage coil tailored for RF heating was capable of irradiating power exceeding the specific-absorption rate (SAR) limits defined by the IEC guidelines by a factor of three. This setup afforded RF induced temperature changes up to +27 K in a reference phantom. The maximum extra temperature increase, induced by a copper tube or a coronary stent was less than 3 K. The coronary stents examined showed an RF heating behavior similar to a copper tube. Our results suggest that, if IEC guidelines for local/global SAR are followed, the extra RF heating induced in myocardial tissue by stents may not be significant versus the baseline heating induced by the energy deposited by a tailored cardiac transmit RF coil at 7.0 T, and may be smaller if not insignificant than the extra RF heating observed under the circumstances used in this study.

Abstract

Water scarcity is a worldwide concern with various implications, such as widespread pollution with heavy metals and organic pollutants, resulting in global threats to the environment and human health, and calling for innovative remediation strategies. We show here that the capability of protein-based carbon aerogels for removing contaminants from water can be greatly improved by transforming proteins into amyloids before their hydrothermal carbonization. Amyloid fibrils were prepared from whey protein isolate, a by-product of the dairy industry, and upon addition to different carbohydrates, the ensued mixtures were used for carbon aerogels production. Furthermore, the generality of the approach for producing carbon aerogels was demonstrated using vegetable amyloids from soy protein. The carbon aerogels are able to adsorb Au(III), Pt(II), Fe(III), and Ag(I) with excellent removal efficiencies. Moreover, the amyloid whey lactose carbon aerogel has an adsorption capacity of 650.08 mg/g for Au(III), significantly higher than that of a carbon aerogel derived from monomer whey lactose. The reusability of the carbon aerogel is also proven over three continuous adsorption/regeneration cycles, with a stable performance in terms of removal efficiency indicating good long-term use. 11. Blind to threat: the presence of temporary goals prevents attention to imminent threat already at early stages of attention allocation

Abstract

Human attention is biased by several motivational factors. For instance, threat attracts attention. People also preferentially pay attention to goal-relevant stimuli even when they are not emotional. How is attention allocated when individuals encounter multiple stimuli with different motivational salience that compete for limited attentional resources? Recent evidence suggests that neutral but goal-relevant stimuli are prioritized over threat when they are simultaneously presented. However, the role of strategic attentional processes in this bias is unknown. To address this issue, we conducted two studies that presented goal-relevant and fear-conditioned threatening stimuli in a dot probe paradigm at early (30 ms) and later stages (250 ms) of attentional processing. Across both studies, attention was allocated to goal-relevant stimuli over threatening stimuli at both presentation times. However, attentional bias to goal-relevant stimuli was larger at later stages of attentional processing. Further, attention to goals over threat was reduced in people high in trait anxiety in the main study. These findings corroborate the conclusion that temporary goals determine attention allocation in a relatively automatic manner, but the influence of goals is stronger at later stages of attention allocation.

12. Wanting to be happy but not knowing how: Poor attentional control and emotion-regulation abilities mediate the association between valuing happiness and depression

Abstract

Recent studies suggest that valuing happiness is associated with negative psychological health outcomes, including increased depression, in US samples. We aimed to replicate these associations in two studies at a UK university (Nstudy one = 151, and Nstudy two = 299). Importantly, we also investigated the role of emotional attentional control and habitual emotion regulation in the relationship between valuing happiness and depression. In both studies, we found that valuing happiness was related to increased depression, confirming the link between valuing happiness and depression in a Western country outside of the USA. Moreover, our findings indicated that the relationship between valuing happiness and depression was strongest in British, rather than non-British participants or participants of dual nationality. Further, our findings revealed that valuing happiness and depression were indirectly associated via the ability to control attention in emotional situations, perceived ability to savor positive experiences, and the extent to which positive emotions feel intrusive. Specifically, increased valuing happiness was associated with lower emotion attention control and lower savoring of positive experiences, which in turn was related to depressive symptoms. These results show that the impaired ability to respond adaptively to emotional situations and to enjoy positive events may underlie the paradoxical relationship between valuing happiness and low well-being.

Abstract

Prior evidence has shown that a person's affective context influences attention to emotional stimuli. The present study investigated whether a crossmodal affective context that is induced by remembering an emotional sound modulates attention to visual emotional stimuli. One group of participants had to remember a positive, negative, or neutral sound during each trial of a dot probe paradigm. A second group of participants also had to encode the valence of the sound. The results revealed that attention was preferentially deployed to stimuli that were emotionally congruent to the affective context. However, this effect was only evident when participants had to encode the valence of the affective context. These findings suggest that a crossmodal affective context modulates the deployment of attention to emotional stimuli provided that the affective connotation of the context is task-relevant.

Prior studies have shown that attention is allocated to events relevant to the current goal of a person. Until now, research has focused on the implementation of a single goal leaving open the question of how attention is allocated when multiple goals are activated. We examined whether the allocation of spatial attention is affected by the prioritizing of one goal over another. The results of two dot probe studies showed that attention is oriented to stimuli relevant to a goal with high value when simultaneously presented with stimuli relevant to a goal with low value (Experiment 1) and to stimuli relevant to a goal with high expectancy of success that were simultaneously presented with stimuli relevant to a goal with low expectancy of success (Experiment 2). These findings demonstrate that the allocation of spatial attention is dependent on the motivational strength of goal pursuit.

Abstract

It is often assumed that attention is automatically allocated to stimuli relevant to one's actual goals. However, the existing evidence for this idea is limited in several ways. We investigated whether words relevant to a person's current goal influence the orienting of attention even when an intention to attend to the goal-relevant stimuli is not present. In two experiments, participants performed a modified spatial cueing paradigm combined with a second task that induced a goal. The results of the experiments showed that the induced goal led to the orientation of attention to goal-relevant words in the spatial cueing task. This effect was not found for words semantically related to the goal-relevant words. The results provide evidence for motivational accounts of attention, which state that the automatic allocation of attention is guided by the current goals of a person.

17. Keeping pain in mind: A motivational account of attention to pain

Abstract

Attention is a key concept in many theories of pain perception. A clinically popular idea is that pain is\ud more intense in persons who are hypervigilant for or bias their attention to pain information. So far,\ud evidence for such bias in pain patients as compared to healthy persons is inconclusive. Furthermore,\ud studies investigating the effects of distracting attention away from pain have shown contradictory\ud results. In this review, we present a motivational perspective on attentional processing of pain that\ud accounts for these inconclusive research findings. We argue that pain always has to be considered within\ud a context of goal pursuit. From this perspective, two largely unexplored theoretical assumptions are\ud introduced. First, when pain occurs during the pursuit of a certain goal, it may unintentionally capture\ud attention although it is not relevant for the goal. Whether such unintentional attentional capture\ud happens is not only dependent upon the characteristics of the pain but also on the characteristics of the\ud focal goal. Second, attention to pain and pain-related information might be driven by a focal goal related\ud to pain. Attentional processing of pain information will be particularly enhanced when the focal goal is\ud related to pain management (e.g., attempting to gain control). Future research should systematically\ud investigate the role of motivation and goal pursuit in the attentional processing of pain-related\ud information. This motivational perspective offers a powerful framework to explain inter- and intraindividual\ud differences in the deployment of attention to pain-related information.

Abstract

Streptococcus agalactiaeis a major pathogen in humans and animals. Virulence factors are often associated with mobile genetic elements, and their expression can be modulated by host factors.S. agalactiaeharbors the genes for C5a peptidase (scpB) and Lmb on a composite transposon structure which is absent in many bovine isolates. To investigate whether these genes participate in the adaptation to human hosts, we determined the influence of human and bovine serum on the promoter activity ofscpBandlmbby using fluorescence-activated cell sorter analysis. Culture in the presence of 1 to 50% human serum resulted in a dose-dependent induction of reporter gene activity forscpBbut notlmb. Reporter gene activity was, however, unchanged following growth in fetal calf serum. Interestingly, a bovine strain did not display any induction ofscpBby either bovine or human serum. Reverse transcription-PCR analysis was used to confirm differential induction ofscpBinS. agalactiaeand showed a similar induction of theStreptococcus pyogenesC5a peptidase genescpAby human but not bovine serum. The specific induction of the streptococcal C5a peptidase by human serum corresponds to the absence ofscpBin many bovineS. agalactiaeisolates and underlines the importance of this virulence factor for human infections.

Abstract

As the knowledge base and importance of mitochondrial physiology to evolution, health and diseaseexpands, the necessity for harmonizing the terminologyconcerning mitochondrial respiratory states and rates has become increasingly apparent. Thechemiosmotic theoryestablishes the mechanism of energy transformationandcoupling in oxidative phosphorylation. Theunifying concept of the protonmotive force providestheframeworkfordeveloping a consistent theoretical foundation ofmitochondrial physiology and bioenergetics.We followthe latest SI guidelines and those of the International Union of Pure and Applied Chemistry(IUPAC)onterminology inphysical chemistry, extended by considerationsofopen systems and thermodynamicsof irreversible processes.Theconcept-driven constructive terminology incorporates the meaning of each quantity and alignsconcepts and symbols withthe nomenclature of classicalbioenergetics.
