Abstract Background Glioma is a common tumor that occurs in the brain and spinal cord. Hypoxia is a crucial feature of the tumor microenvironment. Tumor-associated macrophages/microglia play a crucial role in the advancement of glioma. This study aims to illuminate the detailed mechanisms by which hypoxia regulates microglia and, consequently, influences the progression of glioma. Methods The glioma cell viability and proliferation were analyzed by cell counting kit-8 assay and 5-ethynyl-2’-deoxyuridine assay. Wound healing assay and transwell assay were implemented to detect glioma cell migration and invasion, respectively. Enzyme-linked immunosorbent assay was conducted to detect protein levels in cell culture medium. The protein levels in glioma cells and tumor tissues were evaluated using western blot analysis. The histological morphology of tumor tissue was determined by hematoxylin-eosin staining. The protein expression in tumor tissues was determined using immunohistochemistry. Human glioma xenograft in nude mice was employed to test the influence of hypoxic microglia-derived interleukin-1beta (IL-1β) and heparanase (HPSE) on glioma growth in vivo. Results Hypoxic HMC3 cells promoted proliferation, migration, and invasion abilities of U251 and U87 cells by secreting IL-1β, which was upregulated by hypoxia-induced activation of hypoxia inducible factor-1alpha (HIF-1α). Besides, IL-1β from HMC3 cells promoted glioma progression and caused activation of nuclear factor-κB (NF-κB) and upregulation of HPSE in vivo. We also confirmed that IL-1β facilitated HPSE expression in U251 and U87 cells by activating NF-κB. Hypoxic HMC3 cells-secreted IL-1β facilitated the proliferation, migration, and invasion of U251 and U87 cells via NF-κB-mediated upregulation of HPSE expression. Finally, we revealed that silencing HPSE curbed the proliferation and metastasis of glioma in mice. Conclusion Hypoxia-induced activation of HIF-1α/IL-1β axis in microglia promoted glioma progression via NF-κB-mediated upregulation of HPSE expression.

Imine-linked covalent organic frameworks (COFs) provide distinctive prospects for promoting photocatalytic H2O2 production, yet the intricate involvement of imine-derived linkages chemistry under light irradiation remains incompletely elucidated. Here, imine-linked COF-LZU1 demonstrates a visible light-driven H2O2 evolution rate of 387 ​μmol ​g−1 ​h−1, particularly, negligible dependence on the sacrificial agents. By virtue of electron paramagnetic spectroscopy and solid-state nuclear magnetic resonance, we experimentally tracked the structure evolution and identified its autooxidation to Wurster's salt mimics under the photocatalytic conditioning. This finding coincides with the radical anion ·O2− governed reaction pathway and is further rationalized with additional theoretical calculations. This study provides insights into both photophysical/photochemical aspects over the imine-linkage structure.

Abstract Olfr78, which is a novel receptor for short‐chain fatty acid (SCFA) acetate and propionate, plays essential roles in some cellular processes. The present study aimed to investigate the role of olfr78 in regulating energy metabolism and delineate the underlying mechanisms using olfr78−/− mice. Deletion of olfr78 did not influence the adiposity of mice fed either normal chow or a high‐fat diet (HFD). However, olfr78−/− mice exhibited glucometabolic dysfunction, as evidenced by increased fasting blood glucose levels, decreased serum insulin levels, and impaired oral glucose tolerance under HFD feeding. When compared to wild‐type (WT) mice, olfr78 deficiency enhanced HFD‐induced gluconeogenesis and increased the mRNA expression of key gluconeogenic genes in the liver and kidney. Quantitative real‐time PCR results revealed that olfr78 expression was higher in the colon compared with other tissues in WT mice. Analysis of the gut microbiota in the feces of olfr78−/− mice using 16S rRNA gene sequencing revealed altered relative abundances of some representative SCFAs producers along with decreased levels of SCFAs, including propionic, isobutyric, 2‐methylbutyric, valeric, isovaleric, and 4‐methylvaleric acids. Importantly, mice lacking olfr78 had low circulating levels of glucagon‐like peptide 1 (GLP‐1). In the STC‐1 enteroendocrine cell line, propionate and acetate induced the secretion of the gut hormone GLP‐1, and this effect was partially abolished by olfr78 siRNA‐mediated knockdown. Together, these results suggest a previously undescribed, specific regulatory role for gut olfr78 in glucose homeostasis and highlight olfr78 as a potential target for diabetes treatment.

Previous studies have reported the therapeutic effects of oleuropein (OP) consumption on the early stage of diabetic nephropathy and diabetic cardiomyopathy. However, the efficacy of OP on the long-course of these diabetes complications has not been investigated. Therefore, in this study, to investigate the relieving effects of OP intake on these diseases, and to explore the underlying mechanisms, db/db mice (17-week-old) were orally administrated with OP (200 mg/kg bodyweight) for 15 weeks. We found that OP reduced expansion of the glomerular mesangial matrix, renal inflammation, renal fibrosis, and renal apoptosis. Meanwhile, OP treatment exerted cardiac anti-fibrotic, anti-inflammatory, and anti-apoptosis effects. Notably, transcriptomic and bioinformatic analyses indicated 290 and 267 differentially expressed genes in the kidney and heart replying to OP treatment, respectively. For long-course diabetic nephropathy, OP supplementation significantly upregulated the cyclic guanosine monophosphate-dependent protein kinase (cGMP–PKG) signaling pathway. For long-course diabetic cardiomyopathy, p53 and cellular senescence signaling pathways were significantly downregulated in response to OP supplementation. Furthermore, OP treatment could significantly upregulate the transcriptional expression of the ATPase Na+/K+ transporting subunit alpha 3, which was enriched in the cGMP–PKG signaling pathway. In contrast, OP treatment could significantly downregulate the transcriptional expressions of cyclin-dependent kinase 1, G two S phase expressed protein 1, and cyclin B2, which were enriched in p53 and cellular senescence signal pathways; these genes were confirmed by qPCR validation. Overall, our findings demonstrate that OP ameliorated long-course diabetic nephropathy and cardiomyopathy in db/db mice and highlight the potential benefits of OP as a functional dietary supplement in diabetes complications treatment.

Co-catalysts decorations provide unique opportunity in promoting the photocatalytic water splitting performance of graphite carbon nitride (g-C3N4) system, while mechanistic understanding of this complex catalytic network remains elusive. Here, taking the single-atom-based photocatalysts (M1-g-C3N4) as an unprecedented simplified model system, we theoretically tracked the photocatalytic kinetics for a comprehensive understanding of the photocatalytic process and afforded the descriptor αS1-T1/αT1-S0 (ratio of the extent of S1-T1 and T1-S0 state mixing) and ΔGH∗ (hydrogen adsorpti on free energy) for rational screening of photocatalysts. The targeted Fe1-g-C3N4 yields an excellent H2 evolution rate (ca. 3.2 ⋅mmol·gcat−1·h−1 under full arc), two order of magnitude improvement relative to pristine g-C3N4 counterpart and also outperforms other representative 3d-transition-metal-based photocatalysts. This work presents a comprehensive understanding of the essential role of isolated atomic sites in the photocatalytic course and sheds light on the design of photocatalysts from both photophysical and photochemical aspects.

Serious carrier recombination and limited active sites are the main restrictions on the efficiency of CO2 photoconversion over g-C3N4-based photocatalysts. Herein, a unique tubular C3N4-carbon heterostructure was constructed via thermal polycondensation process, where the inherent open porous C3N4 microstructure was rationally exploited as scaffolds to co-assemble with pyridinic N moieties into composites. Beyond the critical role of serving as a carbon source, the EDTA-2Na additive provides a fairly straightforward pathway for the production of surface cyano groups, which presumably results from the dehydration process of amide bonds. The interfacial cyano groups and work function difference induce strong built-in electric field effects, boosting spatial carrier transfer kinetics. Meanwhile, the incorporated reactive N species endow high affinity for CO2 and thereby remarkable enhancement of uptake capacity. The developed hybrid system modulates the CO2 reduction pathway with favoured 8e− selectivity (56%) toward CH4 products, with an evolution rate up to 2.54 μmol·g-1 h−1, compared with trace of the bare C3N4 counterpart. This work constitutes a prospective target for surface and interface design and paves the way for developing high-performance photocatalysts for multi-electron CO2 reduction.

Operational stability and mechanical robustness remain as engineering bottlenecks in perovskite solar cells technology. Here, Dong et al. introduce an interpenetrating perovskite at the electron-transporting-layer interface that enables a 1000-hour stable operation and high endurance against bending fatigue over 2500 cycles.

Cedryl acetate (CA), also called acetyl cedrene, is approved by the FDA as a flavoring or adjuvant to be added to foods. In this study, we aimed to investigate the preventive benefits of CA on obesity and obesity-related metabolic syndrome caused by a high-fat diet (HFD). Three groups of C57BL/6J mice (ten-week-old) were fed Chow, an HFD, or an HFD with CA supplementation (100 mg/kg) for 19 weeks. We observed that CA supplementation significantly reduced weight gain induced by an HFD, decreased the weight of the visceral fat pads, and prevented adipocyte hypertrophy in mice. Moreover, mice in the CA group showed significant improvements in hepatic lipid accumulation, glucose intolerance, insulin resistance, and gluconeogenesis compared with the mice in the HFD group. Since 16S rRNA analysis revealed that the gut microbiota in the CA and HFD groups were of similar compositions at the phylum and family levels, CA may have limited effects on gut microbiota in HFD-fed mice. The beneficial effects on the metabolic parameters of CA were reflected by CA’s regulation of metabolism-related gene expression in the liver (including Pepck, G6Pase, and Fbp1) and the epididymal white adipose tissues (including PPARγ, C/EBPα, FABP4, FAS, Cytc, PGC-1α, PRDM16, Cidea, and COX4) of the mice. In summary, a potent preventive effect of CA on HFD-induced obesity and related metabolic syndrome was highlighted by our results, and CA could be a promising dietary component for obesity intervention.

The application of STING agonists and the blockade of the SIRPα–CD47 signaling axis are emerging immunotherapeutic strategies. The role of PD-L1 expression on tumor-infiltrating immune cells in promoting tumor immune escape is not fully understood. Here, the authors show that PD-L1 expression by dendritic cells plays a crucial role in hindering anti-tumor T cell responses and is essential for the therapeutic efficacy of PD-L1 blockade.

Interleukin-2 (IL-2) based cancer therapy is limited by severe toxicity and strong Treg amplification at the therapeutic dosage. Here, the authors develop a recombinant IL-2 immunocytokine which is comprised of a tumor-targeting antibody fused to a super mutant IL-2 and show in mouse models that this next-generation IL2 has reduced toxicity and enhanced antitumor activity.

Despite the presence of tumor-infiltrating lymphocytes, many patients do not respond to PD-L1/PD-1 blockade therapy. Here they show that PD-L1 antibody armed with interferon-α (IFNα) improves tumor targeting and antigen presentation while countering innate or T-cell-drive PD-L1 upregulation, and overcomes resistance to checkpoint blockade therapy.

Genetic modification (GM) offers an alternative strategy to conventional animal breeding. The present study was carried out to investigate the potential health effects of the consumption of growth hormone-transgenic triploid carp (GH-ttc) through a 90-day subchronic rodent feeding study. Wistar rats (n = 10/sex/group) were given formulated diets containing GH-ttc or non-GM carp at an incorporated rate of 2.5%, 5%, or 10% (w/w) for 90 days. An additional control group of rats (n = 10/sex/group) was fed a basic rodent diet. During the 90-day study, clinical observation, ophthalmic examination, body weight, and food intake were evaluated. At the end of the study, rats were killed, and the hematology, serum chemistry, urine test, necropsy, and histopathology were assessed. Compared with the non-GM carp and the basic control groups, no biologically significant differences were observed on clinical signs of toxicity, body weights, food intake, hematology, serum chemistry, urinalysis, organ weight, and histopathology on selected organs for the GH-ttc group. The results of this 90-day subchronic feeding study indicated that, at the dose level used in this study, consumption of GH-ttc showed no subchronic toxicity to Wistar rats.

Tisochrysis lutea is a haptophyte microalga important in aquaculture. The 105,837-bp chloroplast genome encodes 113 proteins, 27 different transfer-RNAs and ribosomal-RNAs. Phylogenetic analysis on currently available chloroplast sequences showed T. lutea was most closely related to Emiliania huxleyi. Unlike E. huxleyi chloroplast DNA, T. lutea contains directly-repeated ribosomal repeats following multiple genome rearrangements during evolution. The non-identical 4.9-kb ribosomal direct-repeats are distinguished by 42 nucleotide polymorphisms and three single- or double-nucleotide INDELs. This is unlike land plant and green algal chloroplast genomes in which efficient copy correction by homologous recombination ensures identical long inverted repeats containing the rRNA operons. Imperfect rDNA repeats are also found in the chloroplast genomes of cryptophytes, rhodophytes and other haptophytes. Reduced copy correction rates in these algal lineages raise questions on the mechanism(s) by which genetic uniformity of multi-copy chloroplast genomes is maintained and has implications for developing chloroplast transformation in these species.

Lyme spirochetes have coevolved with ticks to optimize transmission to hosts using tick salivary molecules (TSMs) to counteract host defenses. TSMs modulate various molecular events at the tick–host interface. Lymphotoxin-beta receptor (LTβR) is a vital immune receptor and plays protective roles in host immunity against microbial infections. We found that Ltbr knockout mice were more susceptible to Lyme disease spirochetes, suggesting the involvement of LTβR signaling in tick-borne Borrelia infection. Further investigation showed that a 15-kDa TSM protein from Ixodes persulcatus (I. persulcatus salivary protein; IpSAP) functioned as an immunosuppressant to facilitate the transmission and infection of Lyme disease spirochetes. IpSAP directly interacts with LTβR to block its activation, thus inhibiting the downstream signaling and consequently suppressing immunity. IpSAP immunization provided mice with significant protection against I. persulcatus–mediated Borrelia garinii infection. Notably, the immunization showed considerable cross-protection against other Borrelia infections mediated by other ixodid ticks. One of the IpSAP homologs from other ixodid ticks showed similar effects on Lyme spirochete transmission. Together, our findings suggest that LTβR signaling plays an important role in blocking the transmission and pathogenesis of tick-borne Lyme disease spirochetes, and that IpSAP and its homologs are promising candidates for broad-spectrum vaccine development.
