The interaction between CdTe quantum dots (QDs) with human serum albumin (HSA) and human cell culture was studied by optical spectroscopy technique. Performed research explored the interaction between the CdTe QDs and HSA, and fluorescence imaging efficiency of the QD-HSA bioconjugates in comparison with colloidal QDs. The secondary structure of the HSA is similar to the native form, which suggest a biocompatibility of prepared bionanocomplex. The CdTe QD-HSA bionanoconjugate shows chemical stability in phosphate-buffered saline (PBS) under ambient conditions, furthermore, it is stable in cytoplasm and suitable for cell labeling, tracking, and other bioimaging applications. The CdTe QDs located in an osteosarcoma cancer cells show a high luminescence intensity. The light emission of the CdTe QDs connected with albumin is less than the pure QDs, but it is still satisfactory, and additionally it is stable and have long photoluminescence lifetime. It suggest, that the CdTe NC-HSA bionanocomplex can be used as a fluorescent probe for cell labeling, tracking, and other bioimaging applications. The method of the high voltage electric discharge (HVED) treatment in the kerosene (the dispersion, the activation and the synthesis) is proposed to obtain the new anti-friction alloys based on TiC – AlC – and FeC with inclusions of the Cn0 (n=6 or 7) phases which are characterized by the high heat resistance, the strength and the durability to be used in the aircraft and in the rocket technology. The method is distinguished by the hardening particles, which are not mechanically added to the powder mixture, but instead they are synthesized during the HVED treatment of the “kerosene – Fe (Al) – Ti powder mixture” the disperse system with its simultaneous grinding what enable us to create the composite material with the increased mechanical and performance characteristics. The obtained alloys contain the refractory components (the fullerenes, the carbides), the MAX-phases of the Ti-Al-C system as shown by the micro-Raman data.

Chronic intestinal pseudo-obstruction results in clinical manifestations that resemble intestinal obstruction but in the absence of any physical obstructive process. Fabry disease is an X-linked lysosomal storage disease characterized by the dysfunction of multiple systems, including significant gastrointestinal involvement. We report the occurrence of chronic intestinal pseudo-obstruction in two unrelated patients with Fabry disease and the possible explanation of a direct relation of these two disorders. In Fabry disease, gastrointestinal symptoms occur in approximately 70% of male patients, but the frequency ranges from 19% to 69% in different series. In some patients, colonic dysmotility due glycolipid deposition in autonomic plexus and ganglia can lead to the pseudo-obstruction syndrome, simulating intestinal necrosis. That is why up to this date colostomy has been performed in some cases, even for children with FD without cardiac, renal or cerebrovascular compromise. Early treatment with enzyme replacement therapy in asymptomatic or mildly symptomatic patients may be justified in order to prevent disease progression. Several studies have demonstrated that enzyme replacement therapy alleviates GI manifestations. Because of the non-specific nature of the gastrointestinal symptoms, diagnosis of Fabry disease is often delayed for several years. Gastrointestinal involvement is often misdiagnosed or under-reported. It is therefore very important to consider Fabry disease in the differential diagnosis of chronic intestinal pseudo-obstruction.

Background: Screening for Fabry disease (FD) in high risk populations yields a significant number of individuals with novel, ultra rare genetic variants in the GLA gene, largely without classic manifestations of FD. These variants often have significant residual α-galactosidase A activity. The establishment of the pathogenic character of previously unknown or rare variants is challenging but necessary to guide therapeutic decisions. Objectives: To present 2 cases of non-classical presentations of FD with renal involvement as well as to discuss the importance of high risk population screenings for FD. Results: Our patients with non-classical variants were diagnosed through FD screenings in dialysis units. However, organ damage was not limited to kidneys, since LVH, vertebrobasilar dolichoectasia and cornea verticillata were also present. Lyso-Gb3 concentrations in plasma were in the pathologic range, compatible with late onset FD. Structural studies and in silico analysis of p.(Cys174Gly) and p.(Arg363His), employing different tools, suggest that enzyme destabilization and possibly aggregation could play a role in organ damage. Conclusions: Screening programs for FD in high risk populations are important as FD is a treatable multisystemic disease which is frequently overlooked in patients who present without classical manifestations.

Being a methyl ester of partricin, the mepartricin complex is the active substance of a drug called Ipertrofan (Tricandil), which was proven to be useful in treatment of benign prostatic hyperplasia and chronic nonbacterial prostatitis/chronic pelvic pain syndrome. Nevertheless, no direct structural evidence on the stereochemistry of its components has been presented to date. In this contribution, we have conducted detailed, NMR-driven stereochemical studies on mepartricins A and B, aided by molecular dynamics simulations. The absolute configuration of all the stereogenic centers of mepartricin A and B was defined as 3 R , 7 R , 9 R , 11 S , 13 S , 15 R , 17 S , 18 R , 19 S , 21 R , 36 S , 37 R , and 38 S , and proposed as 41 R . The geometry of the heptaenic chromophore of both compounds has been established as 22 E , 24 E , 26 E , 28 Z , 30 Z , 32 E , and 34 E . Our studies on mepartricin ultimately proved that partricins A and B are structurally identical to the previously described main components of the aureofacin complex: gedamycin and vacidin, respectively. The knowledge of the stereochemistry of this drug is a fundamental matter not only in terms of studies on its molecular mode of action, but also for potential derivatization, aiming at improvement of its pharmacological properties.

During spermiogenesis of an alga Chara vulgaris, which in many aspects resembles that of animals, histones are replaced by protamine-type proteins. Our earlier immunocytochemical studies showed that this replacement started during the short stage V of spermiogenesis, when electronograms revealed an extensive system of cisternae and vesicles of endoplasmic reticulum (ER). The present studies revealed at stage V intensive incorporation of labeled (3)H-arginine and (3)H-lysine quickly translocating into a nucleus visualized with pulse-chase autoradiography of semithin sections. The immunogold technique with the use of the antibodies to protamine-type proteins isolated from Chara tomentosa show that both ER cisternae and vesicles are labeled with gold grains, which are absent from the spermatids not treated with the antibodies; thus, the ER is probably the site of the protamine-type protein synthesis. These proteins then are translocated to a nucleus through ER channels connected with the nuclear envelope, as suggested by gold labeling of an inner membrane of the nuclear envelope adjacent to condensed chromatin. The above results correspond with those of other authors showing that in animals, protamines bind with lamin B receptors localized in the inner membrane of the nuclear envelope. A hypothesis has been put forward that during Chara spermiogenesis the inner membrane of the nuclear envelope invaginates into a nucleus together with protamine-type proteins, which become separated from the membrane and penetrate into chromatin.

The intra-S-phase checkpoint response to hydroxyurea (HU)-mediated arrest of DNA replication was analysed in root meristems of two legumes, Pisum sativum and Vicia faba. The obtained results suggest that a molecular signal which invokes mechanisms allowing the cells to override the S-M dependency control system may be generated by caffeine (CF) and a number of alternative, yet related chemical agents, benzyl-6-aminopurine (BAP), 2-aminopurine (2-AP), and 6-dimethylaminopurine (DMAP). A variety of aberrant mitotic divisions included chromosomal breaks and gaps, lost and lagging chromatids and chromosomes, acentric fragments, chromosome bridges and micronuclei. Furthermore, similar effects induced by sodium vanadate, an inhibitor of protein phosphatases, extend the number of inhibitors capable of inducing premature chromosome condensation (PCC) in root meristem cells, as well as the range of possible regulatory pathways leading to the transition from S-phase arrest towards abnormal mitosis. Until preprophase, FITC-conjugated monoclonal antibodies (alpha-Y(a)b-FITC) that specifically recognize phosphorylated form of threonine indicate no evident cell cycle-dependent changes in an overall phosphorylation status of root meristem cells in the control plants. Irrespective of the stage of interphase, alpha-Y(p)ab-FITC was localized basically in the cytoplasm, whereas nuclear staining was considerably weaker, with a significant fluorescence confined merely to nucleolar regions. The intensity of alpha-Y(p)ab-FITC staining in HU/CF-treated seedlings was found higher than that in the control plants (with the exception of G2 cells), suggesting a general increase in the level of protein phosphorylation, a physiological response mediated probably by an enhanced activity of the cdc-like protein kinase(s).

Previous experiments have shown that extracts obtained from maturing male sex organs of Chara tomentosa, containing a low molecular weight peptide (termed antheridial chromatin condensation factor--ACCF) are capable to induce structural and functional effects in root meristems and fern gametophytes. Our present data point to a number of resemblances between the phenotypic characters of antheridial filaments (a.f.) and the properties of ACCF-treated human lymphocytes and HeLa cells; these comprise primarily a number of morphological changes at the nuclear/chromosomal levels. Mitotic chromosomes become shortened and the relative duration of prophase is reduced, while duration of telophase is prolonged. Nucleolar profiles in ACCF-treated HeLa cells become reduced. Significant decrease in mitotic activity was found in human and yeast cells (Schizosaccharomyces pombe). All the above similarities between the "innate" processes within a.f. and those induced by ACCF provide positive evidence for the presence of a highly specific factor that contributes to nuclear re-patterning of cells undergoing morphogenetic transformations before the onset of spermiogenesis.

Previous results have demonstrated that extracts derived from maturing male sex organs of Chara tomentosa are capable of inducing profound structural and functional effects upon M-phase cells in the primary root meristems of Melandrium noctiflorum and Allium cepa. Evident changes produced by a putative factor engaged in morphogenesis of antheridial filaments are manifested by: (1) significant shortening of chromosomes, (2) decreased mitotic indices, and (3) altered proportions estimated for the prophase and telophase transit times. The present image analysis of late G2 phase nuclei in antheridial filaments of C. tomentosa supports the concepts that progressive changes of their functional activities correspond closely to the increasing proportion of condensed chromatin. Cytophotometric measurements of Feulgen-stained cell nuclei in root meristems after a prolonged incubation in antheridial extracts revealed that cells which previously divided asynchronously became preferentially arrested in G1 (M. noctiflorum) and G2 (A. cepa). The stages at which the cells arrest are supposed to counterpart restriction checkpoints that prevent the initiation of DNA synthesis and mitosis. This assumption has been confirmed by autoradiographic studies using 3H-thymidine. In terms of the "Principal Control Points" (PCP) hypothesis, the obtained results suggest that two PCPs regulate G1-->S and G2-->M transition in a nuclear structure-dependent and a species-specific manner. Although in antheridial extract-treated roots of both M. noctiflorum and A. cepa there are only slight changes in the levels of chromatin condensation, the relative proportions of G1- and G2-arrested cells and their nuclear density profiles differ, as compared with the control and carbohydrate-starved plants.
