The Ixodidae family comprises ticks that are hematophagous ectoparasites and are considered vectors of several hemoparasites from the Anaplasmataceae family and the genus Hepatozoon, Babesia, and Rickettsia. These ectoparasites parasitize domestic and wild animals belonging to several vertebrate groups. Ticks are highly adapted to different biomes and thus possess a wide geographical distribution. In the Brazilian state of Bahia, localized in the Northeast region, there are large rainforest fragments. Studies have rarely been carried out on ticks, and their hemoparasites, that parasitize wild animals in this region. Thus, this study aimed to identify the tick species parasitizing wild animals rescued in rainforest fragments of Bahia and investigate the presence of hemoparasites in tick tissues. During a 2-year period, 238 ticks were collected from 41 wild mammalians, reptiles, and amphibians. These ectoparasites were taxonomically classified according to their morphological characteristics. The ticks identified belonged to five different species from the Ixodidae family: Amblyomma varium, Amblyomma rotundatum, Amblyomma nodosum, Ixodes loricatus, and Rhipicephalus sanguineus. For the first time, an A. rotundatum parasitizing the Mesoclemmys tuberculata turtle was described. PCR assays using DNA extracted from salivary glands or midgut of the ticks were performed to detect specific DNA fragments of hemoparasites from the genus Rickettsia, Ehrlichia, Babesia, Hepatozoon, and from the Anaplasmataceae family. The results showed positive detection of the Rickettsia genus (7.9%), Anaplasmataceae family (15.8%), and Hepatozoon genus (15.8%). Specific DNA from the Ehrlichia and Babesia genera were not detected in these samples. Specific DNA from members of the Anaplasmataceae family was detected in A. varium for the first time. The present work showed that amphibians, reptiles, and mammals from Bahia's Atlantic Forest areparasitized by different tick species, and that these ectoparasites present pathogens in their tissues that impact both humans and animals due to their zoonotic potential.

Rio Grande do Sul is the southernmost state of Brazil, bordering Uruguay. Clinical cases of spotted fever group (SFG) rickettsiosis were recently reported in Rio Grande do Sul. None of these cases was lethal, and all were confirmed by seroconversion to R. rickettsii antigens. Because serological cross-reactions are well known to occur between different SFG agents, the SFG agent responsible for the clinical cases remains unknown in Rio Grande do Sul, where no rickettsial agent is known to infect ticks. During 2013-2014, ticks and blood sera samples were collected from domestic dogs and wild small mammals, and from the vegetation in a SFG-endemic area of Rio Grande do Sul. Dogs were infested by Amblyomma ovale adult ticks, whereas small mammals were infested by immature stages of A. ovale, Ixodes loricatus, and adults of I. loricatus. Ticks collected on vegetation were adults of A. ovale, and immature stages of A. ovale, Amblyomma dubitatum, and Amblyomma longirostre. Three Rickettsia species were detected: Rickettsia bellii in I. loricatus, Rickettsia amblyommii in A. longirostre, and a Rickettsia parkeri-like agent ( Rickettsia sp. strain Atlantic rainforest) in A. ovale. Seroreactivity to SFG antigens were detected in 19.7 % (27/137) canine and 37.5 % (15/40) small mammal sera, with highest titers to R. parkeri. Results indicate that the R. parkeri-like agent, strain Atlantic rainforest, is circulating between A. ovale ticks, dogs and small mammals in the study area, suggesting that this SFG pathogen could be one of the etiological agents of SFG clinical cases in Rio Grande do Sul. Larvae and nymphs of Ixodes loricatus Neumann, 1899 ticks (confirmed by morphological characters and by comparison of 16S mitochondrial rDNA sequences) were collected from Sigmodontinae Wagner, rodents in central and northern Argentina and Uruguay. A total of 100 larvae and 38 nymphs of I. loricatus were collected on the genera Akodon Meyen (n = 36 individuals), Calomys Waterhouse (n = 2), Oligoryzomys Bang (n = 1 2), Oxymycterus Waterhouse (n = 9), and Scapleromys Waterhouse (n = 13). 72 larvae and 18 nymphs were collected on Akodon. Adults of I. loricatus were found in central Argentina and Uruguay on Didelphimorphia of the genera Didelphis Linnaeus and LutreolinaThomas. Ixodes loricatus has been considered a species with strict total specificity to Didelphimorphia. Our results show that this statement may not be justified. Sigmodontinae rodents are sympatric and share habitats with the phylogenetically distant Didelphimorphia; infestation with I. loricatus seems to be its consequence. We tentatively consider I. loricatus moderately specific to Didelphimorphia.

In this study we analyzed seasonal trends of immature stages of Ixodes loricatus parasitizing Akodon azarae (Sigmodontinae) in a locality of the Paraná Delta, Buenos Aires, Argentina. The data encompassed two consecutive years and the analysis included the control of potential confounders using Generalized Linear Mixed Models (GLMM) with a negative binomial response. Larvae had within-year differences in abundance, but the peak months differed drastically between both sampling years, indicating that there was no consistent seasonal pattern. Regarding nymphs, a seasonal pattern was first apparent, but it vanished completely when potential confounders were considered, strongly suggesting that the association with season was spurious and caused by the confounders. Our results showed that the occurrence of immature I. loricatus in its hosts does not follow a distinct seasonal pattern, and highlight the usefulness of controlling for confounding and statistically assessing consistency between years when establishing tick seasonality. An Ixodes loricatus engorged female, infected with Rickettsia bellii, was collected from an opossum ( Didelphis aurita) in Mogi das Cruzes, São Paulo State, Brazil. Two consecutive laboratory tick generations (F1 and F2) reared from this single engorged female were evaluated for Rickettsia infection by polymerase chain reaction (PCR) targeting specific Rickettsia genes. Immature ticks fed on naïve Wistar rats ( Rattus norvegicus) and adult ticks fed on opossum ( D. aurita), both free of ticks and rickettsial infection. PCR performed on individual ticks from the F1 (20 larvae, 10 nymphs, and 10 adults) and the F2 (30 larvae, 30 nymphs, and 15 adults) yielded expected bands compatible with Rickettsia. All the PCR products that were sequenced, targeting gltA gene, resulted in sequences identical to each other and 99.7% (349/350) similar to the corresponding sequence of R. bellii in GenBank. The R. bellii infection on ticks from the second laboratory generation (F2) was confirmed by other PCR protocols and successful isolation of R. bellii in cell culture. We report for the first time a Rickettsia species infecting I. loricatus, and the first report of R. bellii in the tick genus Ixodes. We conclude that there was an efficient transovarial transmission and transstadial survival of this Rickettsia species in the tick I. loricatus. Our results suggest that R. bellii might be maintained in nature solely by transovarial transmission and transstadial survival in ticks (no amplifier vertebrate host is needed), since there has been no direct or indirect evidence of infection of vertebrate hosts by R. bellii.
