However, a recent study suggested that pulmonary hypertension, the pivotal symptom of porcine CARPA, is not mediated by C in cases of polystyrene nanoparticle (PS-NP)-induced reactions.Goals: To characterize PS-NPs and reexamine the contribution of CARPA to their pulmonary reactivity in pigs.Study design: C activation by 200, 500, and 750 nm (diameter) PS-NPs and their opsonization were measured in human and pig sera, respectively, and correlated with hemodynamic effects of the same NPs in pigs in vivo.Methods: Physicochemical characterization of PS-NPs included size, ζ-potential, cryo-transmission electron microscopy, and hydrophobicity analyses. C activation in human serum was measured by ELISA and opsonization of PS-NPs in pig serum by Western blot and flow cytometry. Pulmonary vasoactivity of PS-NPs was quantified in the porcine CARPA model.Results: PS-NPs are monodisperse, highly hydrophobic spheres with strong negative surface charge. In human serum, they caused size-dependent, significant rises in C3a, Bb, and sC5b-9, but not C4d. Exposure to pig serum led within minutes to deposition of C5b-9 and opsonic iC3b on the NPs, and opsonic iC3b fragments (C3dg, C3d) also appeared in serum. PS-NPs caused major hemodynamic changes in pigs, primarily pulmonary hypertension, on the same time scale (minutes) as iC3b fragmentation and opsonization proceeded. There was significant correlation between C activation by different PS-NPs in human serum and pulmonary hypertension in pigs.Conclusion: PS-NPs have extreme surface properties with no relevance to clinically used nanomedicines. They can activate C via the alternative pathway, entailing instantaneous opsonization of NPs in pig serum. Therefore, rather than being solely C-independent reactivity, the mechanism of PS-NP-induced hypersensitivity in pigs may involve C activation. These data are consistent with the “double-hit” concept of nanoparticle-induced hypersensitivity reactions involving both CARPA and C-independent pseudoallergy. Bilateral TAP blocks with 120 mg (1.8 mg/kg) of ropivacaine provided sufficient intra- and postoperative analgesia for insertion of a continuous ambulatory peritoneal dialysis catheter. However, the plasma concentration of ropivacaine reached a maximum of 2.5 µg/mL at 15 minutes after the TAP block, a concentration that was potentially neurotoxic. Although apparent signs of local anesthetic systemic toxicity (LAST) such as convulsion or changes in an electrocardiogram were not observed, the patient became drowsy after the TAP block, which might be one of the mild symptoms of LAST. A TAP block by itself can thus be an anesthetic option for patients undergoing minor abdominal surgery. However, cardiac and renal dysfunction might influence the pharmacokinetics of a local anesthetic used, and attention should be paid to the possibility of LAST even with a low dose of a local anesthetic for patients with cardiac and renal failure. International audience; Muon neutrino and antineutrino disappearance probabilities are identical in the standard three-flavor neutrino oscillation framework, but CPT violation and non-standard interactions can violate this symmetry. In this work we report the measurements of $\sin^{2} \theta_{23}$ and $\Delta m_{32}^2$ independently for neutrinos and antineutrinos. The aforementioned symmetry violation would manifest as an inconsistency in the neutrino and antineutrino oscillation parameters. The analysis discussed here uses a total of 1.97$\times$10$^{21}$ and 1.63$\times$10$^{21}$ protons on target taken with a neutrino and antineutrino beam respectively, and benefits from improved flux and cross-section models, new near detector samples and more than double the data reducing the overall uncertainty of the result. No significant deviation is observed, consistent with the standard neutrino oscillation picture.

Cosmic-ray muons that enter the Super-Kamiokande detector cause hadronic showers due to spallation in water, producing neutrons and radioactive isotopes. Those are a major background source for studies of MeV-scale neutrinos and searches for rare events. Since 2020, gadolinium was introduced in the ultra-pure water in the Super-Kamiokande detector to improve the detection efficiency of neutrons. In this study, the cosmogenic neutron yield was measured using data acquired during the period after the gadolinium loading. The yield was found to be $(2.76 \pm 0.02\,\mathrm{(stat.) \pm 0.19\,\mathrm{(syst.)}}) \times 10^{-4}\,\mu^{-1} \mathrm{g^{-1} cm^{2}}$ at 259 GeV of average muon energy at the Super-Kamiokande detector., Comment: 10 pages, 10 figures, 3 tables

We report the first search result for the flux of astrophysical electron antineutrinos for energies O(10) MeV in the gadolinium-loaded Super-Kamiokande (SK) detector. In June 2020, gadolinium was introduced to the ultra-pure water of the SK detector in order to detect neutrons more efficiently. In this new experimental phase, SK-Gd, we can search for electron antineutrinos via inverse beta decay with efficient background rejection and higher signal efficiency thanks to the high efficiency of the neutron tagging technique. In this paper, we report the result for the initial stage of SK-Gd with a $22.5\times552$ $\rm kton\cdot day$ exposure at 0.01% Gd mass concentration. No significant excess over the expected background in the observed events is found for the neutrino energies below 31.3 MeV. Thus, the flux upper limits are placed at the 90% confidence level. The limits and sensitivities are already comparable with the previous SK result with pure-water ($22.5 \times 2970 \rm kton\cdot day$) owing to the enhanced neutron tagging.

International audience; This paper reports the first measurement of muon neutrino charged-current interactions without pions in the final state using multiple detectors with correlated energy spectra at T2K. The data was collected on hydrocarbon targets using the off-axis T2K near detector (ND280) and the on-axis T2K near detector (INGRID) with neutrino energy spectra peaked at 0.6 GeV and 1.1 GeV respectively. The correlated neutrino flux presents an opportunity to reduce the impact of the flux uncertainty and to study the energy dependence of neutrino interactions. The extracted double-differential cross sections are compared to several Monte Carlo neutrino-nucleus interaction event generators showing the agreement between both detectors individually and with the correlated result.

- Abstract
We searched for proton decay via p→μ+K0 in 0.37 Mton·years of data collected between 1996 and 2018 from the Super-Kamiokande water Cherenkov experiment. The selection criteria were defined separately for KS0 and KL0 channels. No significant event excess has been observed. As a result of this analysis, which extends the previous search by an additional 0.2 Mton·years of exposure and uses an improved event reconstruction, we set a lower limit of 3.6×1033 years on the proton lifetime.

In 2020, the Super-Kamiokande (SK) experiment moved to a new stage (SK-Gd) in which gadolinium (Gd) sulfate octahydrate was added to the water in the detector, enhancing the efficiency to detect thermal neutrons and consequently improving the sensitivity to low energy electron anti-neutrinos from inverse beta decay (IBD) interactions. SK-Gd has the potential to provide early alerts of incipient core-collapse supernovae through detection of electron anti-neutrinos from thermal and nuclear processes responsible for the cooling of massive stars before the gravitational collapse of their cores. These pre-supernova neutrinos emitted during the silicon burning phase can exceed the energy threshold for IBD reactions. We present the sensitivity of SK-Gd to pre-supernova stars and the techniques used for the development of a pre-supernova alarm based on the detection of these neutrinos in SK, as well as prospects for future SK-Gd phases with higher concentrations of Gd. For the current SK-Gd phase, high-confidence alerts for Betelgeuse could be issued up to nine hours in advance of the core-collapse itself., Comment: 20 pages

Due to a very low production rate of electron anti-neutrinos ($\bar{\nu}_e$) via nuclear fusion in the Sun, a flux of solar $\bar{\nu}_e$ is unexpected. An appearance of $\bar{\nu}_e$ in solar neutrino flux opens a new window for the new physics beyond the standard model. In particular, a spin-flavor precession process is expected to convert an electron neutrino into an electron anti-neutrino (${\nu_e\to\bar{\nu}_e}$) when neutrino has a finite magnetic moment. In this work, we have searched for solar $\bar{\nu}_e$ in the Super-Kamiokande experiment, using neutron tagging to identify their inverse beta decay signature. We identified 78 $\bar{\nu}_e$ candidates for neutrino energies of 9.3 to 17.3 MeV in 2970.1 live days with a fiducial volume of 22.5 kiloton water (183.0 kton$\cdot$year exposure). The energy spectrum has been consistent with background predictions and we thus derived a 90% confidence level upper limit of ${4.7\times10^{-4}}$ on the $\nu_e\to\bar{\nu}_e$ conversion probability in the Sun. In this study, therefore, we tried to shed the light on the contributions of metronomic S-1 dosing to the enhanced accumulation and/or spatial distribution of PEGylated liposome within tumor tissue. Tumor priming with metronomic S-1 treatment induced a potent apoptotic response against both angiogenic endothelial cells and tumor cells adjacent to tumor blood vessels, resulting in enhanced tumor blood flow via transient normalization of tumor vasculature, along with alleviation of intratumor pressure. Such a change in the tumor microenvironment imparted by S-1 treatment allows efficient delivery of PEGylated liposome to tumor tissue and permits their deep penetration/distribution into the tumor mass. Such a priming effect of S-1 dosing can be exploited as a promising strategy to enhance the therapeutic efficacy of nanocarrier-based cancer therapeutics suffering from inadequate/heterogeneous delivery to tumor tissues. The patients received teriparatide 20 µg daily by subcutaneous injection, for a maximum of 24 months. Safety and effectiveness analyses were based on data from 1,847 patients who were predominantly female (92.6%) with a mean age of 75.4 years. A total of 157 adverse drug reactions (ADRs) were reported in 140 (7.58%) patients; the most common ADRs were hyperuricemia, nausea, and dizziness. Only six (0.32%) patients reported serious ADRs, the most common being nausea (two patients; 0.1%). Persistence with teriparatide treatment was 60.8% and 39.1% at 18 and 24 months, respectively. There were significant increases in biomarkers for bone formation (procollagen type I N-terminal propeptide and bone-specific alkaline phosphatase) and bone resorption (collagen type I cross-linked C telopeptide and tartrate-resistant acid phosphatase 5b) throughout the study. These were accompanied by significant increases in bone mineral density and low incidences of new vertebral and nonvertebral fractures. Patient-reported measurements for health-related quality of life revealed significant improvements from baseline in back pain and overall health-related quality of life (Short Form-8™ health survey). The results of this 24-month postmarketing surveillance study imply that teriparatide has a favorable safety profile and is effective in the treatment of patients with osteoporosis at high risk of fracture in Japan. Between 2005 and 2014, we examined seven patients with hemophilia A (ten joints: six hips and four knees) receiving total hip or knee arthroplasty (THA or TKA) for hemophilic arthropathy. One male patient (31 years old) showed an intra-articular hematoma formation after THA (case 1). In another male patient (46 years old) receiving TKA, the postoperative trough factor VIII level became lower significantly than reference levels (80%–100% for the 5–10 postoperative days) recommended by the guidelines from the Japanese Society on Thrombosis and Hemostasis, despite sufficient coagulant based on the guidelines being administered (case 2). In the latter patient, deep infection and hematoma formation were observed postoperatively. In this article, we provide a detailed clinical report regarding these two complication cases at the early postoperative periods, and the management of bleeding control for hemophilia patients is discussed.Keywords: hemophilia A, arthroplasty, clotting factor VIII levels, hematoma, infection

We searched for proton decay via $p\to\mu^+K^0$ in 0.37\,Mton$\cdot$years of data collected between 1996 and 2018 from the Super-Kamiokande water Cherenkov experiment. The selection criteria were defined separately for $K^0_S$ and $K^0_L$ channels. No significant event excess has been observed. As a result of this analysis, which extends the previous search by an additional 0.2\,Mton$\cdot$years of exposure and uses an improved event reconstruction, we set a lower limit of $3.6\times10^{33}$ years on the proton lifetime., Comment: 13 pages, 11 figures

However, detailed profiles of PLCβ 3-expressing dorsal root ganglion (DRG) neurons and involvement of PLCβ 3 in inflammatory and postoperative pain have not been fully investigated.Purpose: We evaluated neurochemical char0acteristics of PLCβ 3-expressing DRG neurons in mice and then we examined the effects of selective knockdown of PLCβ 3 expression in DRGs on inflammatory and postoperative pain.Methods: Male C57BL/6-strain mice were used. For the inflammatory model, each mouse received subcutaneous injection of complete Freund’s adjuvant (CFA) in the left hindpaw. For the postoperative pain model, a plantar incision was made in the left hindpaw. PLCβ 3 antisense oligodeoxynucleotide or PLCβ 3 mismatch oligodeoxynucleotide was intrathecally administered once a day for three consecutive days in each model. The time courses of thermal hyperalgesia and mechanical hyperalgesia were investigated. Changes in PLCβ 3 protein levels in DRGs were evaluated by Western blotting.Results: Immunohistochemical analysis showed that high proportion of the PLCβ 3-positive profiles were biotinylated isolectin B4-positive or transient receptor potential vanilloid subfamily 1-positive. PLCβ 3 protein level in DRGs during CFA-induced inflammation was comparable to that at baseline. Intrathecal administration of PLCβ 3 antisense oligodeoxynucleotide, which significantly suppressed PLCβ 3 expression in DRGs, did not affect pain thresholds in normal conditions but inhibited CFA-induced thermal and mechanical hyperalgesia both at the early and late phases compared to that in mismatch oligodeoxynucleotide-treated mice. Intrathecal administration of PLCβ 3 antisense oligodeoxynucleotide also inhibited surgical incision-induced thermal and mechanical hyperalgesia.Conclusion: Our results uncover a unique role of PLCβ 3 in the development and maintenance of inflammatory pain induced by CFA application and in those of surgical incision-induced pain, although PLCβ 3 does not play a major role in thermal nociception or mechanical nociception in normal conditions.Keywords: phospholipase Cβ 3, inflammatory pain, postoperative pain, thermal hyperalgesia, mechanical hyperalgesia
