Abstract Osteoporosis affects millions of people worldwide. As such, this study assessed the macrophage-dependent in vitro anti-osteoporosis, phytochemical profile and hepatotoxicity effects in zebrafish larvae of the stem bark extracts of P. africana. Mouse bone marrow macrophages (BMM) cells were plated in 96-well plates and treated with P. africana methanolic bark extracts at concentrations of 0, 6.25, 12.5, 25, and 50 µg/ml for 24 h. The osteoclast tartrate-resistant acid phosphatase (TRAP) activity and cell viability were measured. Lipopolysaccharides (LPS) induced Nitrite (NO) and interleukin-6 (IL-6) production inhibitory effects of P. africana bark extracts (Methanolic, 150 µg/ml) and β-sitosterol (100 µM) were conducted using RAW 264.7 cells. Additionally, inhibition of IL-1β secretion and TRAP activity were determined for chlorogenic acid, catechin, naringenin and β-sitosterol. For toxicity study, zebrafish larvae were exposed to different concentrations of 25, 50, 100, and 200 µg/ml P. africana methanolic, ethanolic and water bark extracts. Rice bran (RB) is a promising food ingredient that can improve biological function. In this study, we investigated the effects of RB, both unfermented (RB30) and fermented (RBF30), with five different microorganisms on the neurobehavioral activity in zebrafish larvae. Analytical methods such as LC–UV and LC–MS were performed for the analysis of RB30 and RBF30 extracts. Interestingly, niacin content, which is known to improve brain functions such as cognition and emotion, was found to be higher in RBF30 than in RB30. Furthermore, niacin content was highly increased in the RBF30-exposed fish, compared to those in the control fish. Therefore, we profiled behavioral patterns and various neurochemistry in zebrafish larvae following supplementation with RBF30 as well as performed calcium imaging on Tg (huC:GAL4-VP16); (UAS:GCaMP7a) zebrafish larvae to determine the correlation of neural activity. RBF30 revealed greater stimulation of locomotor activity without negatively affecting decision-making behavior in zebrafish larvae, as compared to RB30 or niacin. Its behavioral activation is mainly linked with the elevations of neural activity and several neurochemicals such as serotonergic and dopaminergic systems that are implicated in the control of anxiety and stress. Taken together, these results suggest that RBF30 could be a food material that improves the behavioral health by modulating neural activity and brain neurochemistry.

Non-Down syndrome pediatric acute megakaryoblastic leukemia (AMKL) may be classified according to the presence of recurrent genetic abnormalities with prognostic relevance. In this case study, we report on a girl with AMKL, 32 months old at the time of diagnosis, in whom we confirmed the presence of the cryptic, poor prognosis NUP98::KDM5A fusion. The patient achieved complete remission (CR) with the first course of chemotherapy, underwent haploidentical family donor hematopoietic stem cell transplantation (HSCT) without event, but relapsed 5 months after HSCT. Through this case report, we emphasize the good initial response to chemotherapy, and the early relapse despite undergoing HSCT in first CR. We review the limited literature on NUP98::KDM5A (+) pediatric AML, and underscore the need for further study to improve the outcome of patients with this rare AML subtype.

Solvatochromism plays a pivotal role in various scientific and technological fields including those that explore molecular interactions, sensing technologies, and organic electronics. Notably, despite their ease of manipulation, direct visualization, and potential for single particle‐based sensing, micro‐sized solid particles have been the focus of a surprisingly low number of solvatochromism investigations. In this study, polydiacetylene (PDA) particles are synthesized and their solvatochromism is investigated at the single particle level using optical laser tweezers‐based methods. The findings reveal that unpolymerized monomers within PDA particles at the water/n‐decane interface undergo dissolution in the n‐decane phase to form internal voids in the particles. This phenomenon leads to structural deformation of the PDA which triggers a solvatochromic response. Studies that integrate this phenomenon with established particle‐based methodologies should provide deeper insights into diverse chromism behaviors and potential applications of solvatochromic materials.

Abstract Osteoporosis is a pathological condition characterized by an accelerated bone resorption rate, resulting in decreased bone density and increased susceptibility to fractures, particularly among the elderly population. While conventional treatments for osteoporosis have shown efficacy, they are associated with certain limitations, including limited drug bioavailability, non-specific administration, and the occurrence of adverse effects. In recent years, nanoparticle-based drug delivery systems have emerged as a promising approach for managing osteoporosis. Nanoparticles possess unique physicochemical properties, such as a small size, large surface area-to-volume ratio, and tunable surface characteristics, which enable them to overcome the limitations of conventional therapies. These nanoparticles offer several advantages, including enhanced drug stability, controlled release kinetics, targeted bone tissue delivery, and improved drug bioavailability. This comprehensive review aims to provide insights into the recent advancements in nanoparticle-based therapy for osteoporosis. It elucidates the various types of nanoparticles employed in this context, including silica, polymeric, solid lipid, and metallic nanoparticles, along with their specific processing techniques and inherent properties that render them suitable as potential drug carriers for osteoporosis treatment. Furthermore, this review discusses the challenges and future suggestions associated with the development and translation of nanoparticle drug delivery systems for clinical use. These challenges encompass issues such as scalability, safety assessment, and regulatory considerations. However, despite these challenges, the utilization of nanoparticle-based drug delivery systems holds immense promise in revolutionizing the field of osteoporosis management by enabling more effective and targeted therapies, ultimately leading to improved patient outcomes.

This study conducted a survey to identify the best ergonomic operation method, in-vehicle location, and the effects of their combination on electronic gearshifts. A total of 15 different design alternatives were derived through combinations of three operation methods (lever slide, button push, and dial rotation) and five in-vehicle locations (left wheel spoke, right wheel spoke, upper center fascia, lower center fascia, and center console). A total of 40 respondents with diverse ages and driving experiences evaluated the 15 different design alternatives across nine ergonomic evaluation measures (accuracy, efficiency, rapidity, learnability, intuitiveness, safety, preference, memorability, and satisfaction). The study results indicated that: (1) the lever slide and button push were superior to dial rotation for the operation method; (2) the lower center facia and center console were superior for the in-vehicle location, and (3) implementing the lever slide method in the center console location was found to lead to the best combination of the operation method and in-vehicle location, while implementing the button push method in the right wheel spoke or upper center fascia location also showed relative superiority. The study findings are expected to contribute to the ergonomic design of electronic gearshifts that can enhance the driver’s gear-shifting experience, thereby improving driving performance and safety.

In this paper, the origin of the gate oscillations with a stray capacitance in the load inductor is analyzed with a device/circuit mix-mode simulation. It is found that the gate ringing occurs when the superjunction device reaches its pinch-off potential (the n-pillar and the p-pillar are fully depleted by the lateral depletion process). The progress of the depletion profiles of the superjunction leads to a rapid change of the drain-to-source capacitance and the dV/dt. Finally, the dV/dt causes a sudden change of the current flow rate across the stray capacitance of the load inductor and the device while triggering the parasitic inductances. Based on these results, a comparative study was carried out with an ideal inductive load switching and, finally, the dampers for relieving the gate ringing were investigated.

Diaphragmatic eventration is a rare anomaly. When patients with this condition undergo general anesthesia, anesthetic management should be performed with particular care owing to the risk of diaphragmatic rupture. Such a rupture can be perioperatively diagnosed using multiple tools including lung ultrasonography. This case report describes the anesthetic management of a male infant with osteochondroma in the distal ulna, presenting with diaphragmatic eventration on the right side.
