Rice (Oryza sativa L.) is a commonly consumed crop nourishing more than half of the human population. Different environmental stresses greatly affect cash crops. Among these, drought is a serious abiotic stress effecting rice productivity. Therefore, the current study was designed to know the role of OsTZF8 gene in conferring tolerance to drought stress. For this purpose, OsTZF8 gene was over-expressed in Oryza sativa subspecies indica variety Swat-1. Through Agrobacterium mediated transformation, OsTZF8 gene (pBIG-Ubi-OsTZF8) was successfully transferred to rice calli. From these rice calli transgenic plants were regenerated and established in greenhouse. For the evaluation of OsTZF8 gene role in drought stress tolerance, two weeks old rice seedling were subjected to drought stress. OsTZF8-OX transgenic indica line A and B displayed 69% and 64% survival rates respectively compared to 33% of control. These results confirmed that due to overexpression of OsTZF8 gene, transgenic line A and B displayed considerable level of drought stress tolerance. Additionally, the chlorophyll content of top four leaves of OsTZF8-OX transgenic indica line A and B were observed to be high compared to control. In conclusion our study revealed that over-expression of OsTZF8 gene has significant role in conferring tolerance to drought stress in rice. [ABSTRACT FROM AUTHOR]

3. Phytochemical and antimicrobial study of Alstonia scholaris leaf extracts against multidrug resistant bacterial and fungal strains.

- Abstract
In the present study, Alstonia scholaris leaves were explored for phytochemical constituents, antibacterial and antifungal potentials. Phytochemical screening of the extracts established the presence of glycosides, alkaloids, saponins, terpenoids, anthraquinones, reducing sugars and steroids which later on confirmed through fourier transform infrared spectroscopic analysis. The extract was applied against eight multidrug resistant bacterial and five fungal strains using standard protocols. Methanol and ethyl acetate extracts of the leaves showed highest diameter of inhibition zone (DIZ) of 28mm and 26mm respectively against Enterobacter. Ethanolic extract exhibited prominent DIZ of 26.33mm and 23.67mm against Enterobacter and Pseudomonas respectively. The n-Hexane extract showed DIZ of 23.67mm against Enterobacter. Aqueous extract showed 19.33mm DIZ against methicillin resistant Staphylococcus aureus. Similarly, the n-hexane extract showed highest DIZ of 20.33mm against Aspergillus fumigatus and this activity was highly effective than the control. Ethyl acetate extract showed 18.67mm DIZ against Aspergillus niger whereas methanolic extract showed marked inhibition against Rhizopus and Acremonium with a DIZ of 20mm and 17.03mm respectively. The current study on A. scholaris unveils about the presence of valuable phytochemicals in it having significant antimicrobial properties and further suggests to investigate for the minimum inhibitory concentration (MIC) value of the extracts in prospective research. In the present study various solvent extracts from the leaves of Fernandoa adenophylla were screened for their phytochemical and antimicrobial potentials. Preliminary phytochemical screening of leaves extracts showed the presence of alkaloids, glycosides, flavonoids, terpenoids, tannins, steroids, reducing sugars and anthracenes. Fourier transform infrared spectroscopic studies showed the presence of different functional groups for compounds like alcohol, aldehydes and ketones, saturated and unsaturated hydrocarbons, amides, amines, carboxylic acids, esters, and ethers. All the extracts showed significant antibacterial activities when tested against eight multidrug resistant bacterial strains isolated from urinary tract infection patients. It was concluded that the leaves extracts of F. adenophylla have several vital phytochemical constituents and significant antibacterial activities against multidrug resistant bacterial strains causing urinary tract infections. Adiantum capillus veneris is a medicinally essential plant used for the treatment of diverse infectious diseases. The study of phytochemical and antimicrobial activities of the plant extracts against multidrug-resistant (MDR) bacteria and medically important fungi is of immense significance. Extracts from the leaves, stems, and roots of Adiantum capillus veneris were extracted with water, methanol, ethanol, ethyl acetate, and hexane and screened for their antimicrobial activity against ten MDR bacterial strains and five fungal strains isolated from clinical and water samples. Ash, moisture, and extractive values were determined according to standard protocols. FTIR (Fourier transform infrared Spectroscopy) studies were performed on different phytochemicals isolated from the extracts of Adiantum capillus Veneris. Phytochemical analysis showed the presence of flavonoids, alkaloids, tannins, saponins, cardiac glycosides, terpenoids, steroids, and reducing sugars. Water, methanol, and ethanol extracts of leaves, stems, and roots showed significant antibacterial and antifungal activities against most of the MDR bacterial and fungal strains. This study concluded that extracts of Adiantum capillus veneris have valuable phytochemicals and significant activities against most of the MDR bacterial strains and medically important fungal strains. Various solvent extracts from the whole plant of Thymus linearis were screened for their phytochemical and antimicrobial potentials. Preliminary phytochemical screening of plant extracts showed the existence of terpenoids, flavonoids, tannins, alkaloids, glycosides and reducing sugars. Fourier transform infrared spectroscopy studies were carried out on various phytochemicals extracted from the exhibited T. linearis which resulted in the presence of different compounds like amides, aldehydes, carboxylic acid, ethers, alcohol and ketones. All the extracts of T. linearis showed significant antibacterial and antifungal activities when tested against nine bacterial and four fungal strains. It was concluded from this study that extracts of T. linearis have an array of important phytochemicals and significant activities against some of the multidrug resistant bacterial and medically important fungal strains.
