1. Identification of key enzalutamide-resistance-related genes in castration-resistant prostate cancer and verification of RAD51 functions

Abstract

Patients with castration-resistant prostate cancer (CRPC) often develop drug resistance after treatment with enzalutamide. The goal of our study was to identify the key genes related to enzalutamide resistance in CRPC and to provide new gene targets for future research on improving the efficacy of enzalutamide. Differential expression genes (DEGs) associated with enzalutamide were obtained from the GSE151083 and GSE150807 datasets. We used R software, the DAVID database, protein–protein interaction networks, the Cytoscape program, and Gene Set Cancer Analysis for data analysis. The effect of RAD51 knockdown on prostate cancer (PCa) cell lines was demonstrated using Cell Counting Kit-8, clone formation, and transwell migration experiments. Six hub genes with prognostic values were screened (RAD51, BLM, DTL, RFC2, APOE, and EXO1), which were significantly associated with immune cell infiltration in PCa. High RAD51, BLM, EXO1, and RFC2 expression was associated with androgen receptor signaling pathway activation. Except for APOE, high expression of hub genes showed a significant negative correlation with the IC50 of Navitoclax and NPK76-II-72-1. RAD51 knockdown inhibited the proliferation and migration of PC3 and DU145 cell lines and promoted apoptosis. Additionally, 22Rv1 cell proliferation was more significantly inhibited with RAD51 knockdown than without RAD51 knockdown under enzalutamide treatment. Overall, six key genes associated with enzalutamide resistance were screened (RAD51, BLM, DTL, RFC2, APOE, and EXO1), which are potential therapeutic targets for enzalutamide-resistant PCa in the future.

Abstract

The rapid development of modern urban agglomerations has reshaped the territory of China’s economic geography and reconstructed the spatial forms of Chinese cities and regions. Since the reform and opening-up, China’s urban agglomerations have experienced three stages, i.e. spontaneous cultivation, rapid growth, and the super leading role of planning. The giant development area composed of super urban agglomerations is becoming an emerging geographic unit that improves the urban development quality, promotes regional integration processes and participates in global industrial competition and innovative resource allocations in China. While encountering challenges such as insufficient innovation kinetic energy and implementation difficulties of cross-regional coordination mechanisms, it has also ushered in new opportunities brought about by the shift of the global urban network system focus, the accelerated formation of high-speed rail urban belts, the rise of innovative geographic unit communities, etc. As entering the new era, centering on the high-quality development requirements of urbanization and urban agglomerations, China should establish a new mechanism for more effective coordinated regional development, build an urban pattern of coordinated development of large, medium and small cities and towns with urban agglomerations as the main body, comprehensively enhance the competitiveness of urban agglomerations and regions, and take a steady and innovation-driven road to modernization.

37. The construction of new 'student' identity, focusing on Chinese pre-sessional English learners

Abstract

Identity and language attracts much interest in the relevant research communities; however the majority of existing studies focus on immigrants who have little English-learning experience before they arrive in their new countries. Their lack of experience of English learning, means they do not have many experiences of contacting with English-speakers, and then do not have suffered the evaluations, whatever acceptances or denials from English speakers. But for some Chinese students, after learning English at least for a decade, their English competence have been regarded as not qualified to enroll the Masters programme. Required to attend a pre-sessional programme which means language or identity denials from native speakers, are likely to position Chinese pre-sessional students in a community with fiercer power relations and identity struggles. The investigation of Chinese pre-sessional students' identity construction, including their views on the Chinese Academic Community and the Local British Academic Community, could draw a fuller picture of how language learners perform, negotiate and construct identities within power relations. In order to carry out this investigation, discourse analysis was used in ananalysis of data collected from an 8-week pre-sessional programme. Semi-structured interview, classroom observation and stimulated recall interview were conducted, based on participants' English-learning experience and their opinions in their processes of identity performance, negotiation and construction. The results suggest that participants' new 'student' identity construction cannot replace entirely the identities constructed in the Chinese Academic Community. In addition to the teachers' encouragement of challenging teachers' power positions in the Local British Academic Community which could be seen as teachers' give-up of own powers in the community, their not understanding or speaking Chinese would result in students' non-cooperation or problems in constructing new English 'student' identities. This would imply that educators need to focus on students' identity struggles from the perspectives of power relations.
