In this study, we investigated the bacterial diversity and plant growth‐promoting (PGP) properties of bulk soil bacteria of Vachellia tortilis subsp. raddiana collected from the Taidalt and Amazloug nature preserves area in southern Morocco. Using repetitive extragenic palindromic polymerase chain reaction fingerprinting and 16S rDNA sequencing, 93 strains were identified. They belonged mainly to the genera Bacillus and Peribacillus. In vitro tests for PGP traits showed high levels of activity for strains belonging to the genera Bacillus and Pseudomonas. It was relevant that several Bacillus strains produced auxin (maximum amount 262.61 µg mL−1 for Bacillus sp. LMR1097), solubilized phosphate (maximum amount 22.10 µg mL−1), or produced siderophores, while three strains were able to fix atmospheric nitrogen (LMR1145, LMR1013, and LMR1015). Inoculation of V. tortilis subsp. raddiana plants with selected bacterial strains (LMR881 and LMR1097) increased shoot and root growth parameters. The strain Pantoea sp. LMR881 had the highest mean values for shoot and root length (24.60–14.80 cm) and shoot dry weight (1.31 g), whereas Bacillus sp. LMR1097 showed the lowest mean shoot dry weight. Overall, these results highlight the potential of using selected native bacterial inoculants for improving growth of candidate tree plants to be used in restoration programs of arid degraded areas. Legumes are renowned for their distinctive biological characteristic of forming symbiotic associations with soil bacteria, mostly belonging to the Rhizobiaceae familiy, leading to the establishment of symbiotic root nodules. Within these nodules, rhizobia play a pivotal role in converting atmospheric nitrogen into a plant-assimilable form. However, it has been discerned that root nodules of legumes are not exclusively inhabited by rhizobia; non-rhizobial endophytic bacteria also reside within them, yet their functions remain incompletely elucidated. This comprehensive review synthesizes available data, revealing that Bacillus and Pseudomonas are the most prevalent genera of nodule endophytic bacteria, succeeded by Paenibacillus, Enterobacter, Pantoea, Agrobacterium, and Microbacterium. To date, the bibliographic data available show that Glycine max followed by Vigna radiata, Phaseolus vulgaris and Lens culinaris are the main hosts for nodule endophytic bacteria. Clustering analysis consistently supports the prevalence of Bacillus and Pseudomonas as the most abundant nodule endophytic bacteria, alongside Paenibacillus, Agrobacterium, and Enterobacter. Although non-rhizobial populations within nodules do not induce nodule formation, their presence is associated with various plant growth-promoting properties (PGPs). These properties are known to mediate important mechanisms such as phytostimulation, biofertilization, biocontrol, and stress tolerance, emphasizing the multifaceted roles of nodule endophytes. Importantly, interactions between non-rhizobia and rhizobia within nodules may exert influence on their leguminous host plants. This is particularly shown by co-inoculation of legumes with both types of bacteria, in which synergistic effects on plant growth, yield, and nodulation are often measured. Objective: To investigate the relationship between triterpenoid saponin content and antioxidant, antimicrobial, and a-glucosidase inhibitory activities of 70% ethanolic, butanolic, aqueous, supernate and precipitate extracts of Juglans regia leaves. Methods: Triterpenoid saponins of different Juglans regia leaf extracts were measured by the vanillin method. Antioxidant activity was evaluated against DPPH and ABTS free radicals. We also assessed a-glucosidase inhibitory and antimicrobial activities of the leaf extracts. Pearson's correlation coefficient was evaluated to determine the correlation between the saponin content and biological activities. Results: The butanolic extract was most effective against DPPH with an IC50 of 6.63 µg/mL, while the aqueous extract showed the highest scavenging activity against ABTS free radical with an IC50 of 42.27 µg/mL. Pearson's correlation analysis indicated a strong negative correlation (r = -0.956) between DPPH radical scavenging activity (IC50) and the saponin content in the samples examined. In addition, the aqueous extract showed the best a-glucosidase inhibitory activity compared with other extracts. All the extracts had fair antibacterial activity against Bacillus subtilis, Escherichia coli, and Klebsiella pneumoniae except for the aqueous extract. Conclusions: Juglans regia extracts show potent antioxidant, antimicrobial, and a-glucosidase inhibitory activities. There is a correlation between saponin levels in Juglans regia leaf extracts and the studied activities. However, additional research is required to establish these relationships by identifying the specific saponin molecules responsible for these activities and elucidating their mechanisms of action. The increasing demand for plant-based medicines makes plant research attractive to find natural alternatives that can replace synthetic products or at least reduce their use. Saponins are a group of natural phytochemical compounds in most herbs, vegetables, and legumes. There are two different types of saponin compounds, which can be classified based on the nature of the aglycone bound to the oligosaccharide fragments, whether it is a steroid or triterpenoid. This paper is devoted to saponins with triterpenoid aglycone and aims to study the relationship between their content and several biological activities in different Juglans regia kernel extracts, including antioxidant, antimicrobial and antibacterial activities. Antioxidant activity was assessed against two free radicals: DPPH and ABTS. Oleanolic acid was used as a standard to quantify triterpenoid saponins, measured by the vanillin method, whereas Acarbose and Tetracycline were used as positive controls to evaluate the α-glucosidase inhibitory and antimicrobial activities, respectively, of walnut kernel extracts. Findings show that hydroalcoholic extract (EtOH 70%) has a considerable content of phenolic compounds, with 59.51 ± 0.26 mg GAE/g in total polyphenol contents, while the content of total flavonoids and condensed tannins were 3.14 ± 0.27 mg QE/g and 85.65 ± 0.37 mg CE/g of crude extract, respectively. In terms of antioxidant activity, the butanolic extract proved to be the most effective against DPPH with IC50 = 7.74 ± 1.49 µg/mL, while the precipitated extract showed the highest scavenging activity against the ABTS free radical with IC50 = 33.14 ± 2.96 µg/mL. In addition, butanolic and hydroalcoholic extracts of Juglans regia kernels showed an α-glucosidase inhibitory activity even more efficiently than synthetic drugs such as Acarbose, with IC50 values of 11.66 µg/mL and 16.17 µg/mL, respectively, versus 18.01 µg/mL in Acarbose. The evaluation of the antimicrobial activity against Bacillus subtilis, Escherichia coli and Klebsiella pneumonia strains revealed that some extracts could inhibit these bacteria. This study successfully assessed the bioactivities of the extracts, suggesting that the extracts of medicinal plants can demonstrate stronger bioactivity than pure synthetic compounds. In the present study, we analyzed the genetic diversity, phylogenetic relationships, stress tolerance, phytobeneficial traits, and symbiotic characteristics of rhizobial strains isolated from root nodules of Vachellia tortilis subsp. raddiana grown in soils collected in the extreme Southwest of the Anti-Atlas Mountains in Morocco. Subsequent to Rep-PCR fingerprinting, 16S rDNA gene sequencing of 15 representative strains showed that all of them belong to the genus Ensifer. Phylogenetic analysis and concatenation of the housekeeping genes gyrB , rpoB , recA , and dnaK revealed that the entire collection (except strain LMR678) shared 99.08 % to 99.92% similarity with Ensifer sp. USDA 257 and 96.92% to 98.79% with Sinorhizobium BJ1. Phylogenetic analysis of nodC and nodA sequences showed that all strains but one (LMR678) formed a phylogenetic group with the type strain " E. aridi" LMR001T (similarity over 98%). Moreover, it was relevant that most strains belong to the symbiovar vachelliae. In vitro tests revealed that five strains produced IAA, four solubilized inorganic phosphate, and one produced siderophores. All strains showed tolerance to NaCl concentrations ranging from 2 to 12% and grew at up to 10% of PEG6000. A greenhouse plant inoculation test conducted during five months demonstrated that most rhizobial strains were infective and efficient. Strains LMR688, LMR692, and LMR687 exhibited high relative symbiotic efficiency values (respectively 231.6 %, 171.96 %, and 140.84 %). These strains could be considered as the most suitable candidates for inoculation of V. t. subsp. raddiana , to be used as a pioneer plant for restoring arid soils threatened with desertification. Nodular endophytes of drought-tolerant legumes are understudied. For this reason, we have isolated and studied non-symbiotic endophytic bacteria from nodules of Vachellia tortilis subsp. raddiana, a leguminous tree adapted to the harsh arid climate of Southern Morocco. Rep-PCR analysis followed by 16S rDNA sequencing revealed two main genera, Pseudomonas and Bacillus. Isolates responded variably to salt and water stresses, and mostly produced exopolysaccharides. Differences concerned also plant growth-promoting activities: phosphate, potassium, and zinc solubilization; biological nitrogen fixation; auxin, siderophore, ammonia, and HCN production; and ACC deaminase activity. Some strains exhibited antagonistic activities against phytopathogenic fungi (Fusarium oxysporum and Botrytis cinerea) and showed at least two enzymatic activities (cellulase, protease, chitinase). Four selected strains inoculated to vachellia plants under controlled conditions have shown significant positive impacts on plant growth parameters. These strains are promising bio-inoculants for vachellia plants to be used in reforestation programs in arid areas increasingly threatened by desertification., (© 2022.
