The current status and distributional uncertainties of some Bombus (bumble bee) species across North America has driven the need for monitoring. The central Appalachian Mountain region has historic and recent records of the endangered Bombus affinis (Rusty Patched Bumble Bee), but the species' distribution within the region is unknown. To address the lack of knowledge on distribution of bumble bee species within the region, specifically Rusty Patched Bumble Bee, I randomly generated a network of 70 roadside transects and conducted 764 ten-minute non-lethal surveys. These surveys recorded 11 out of 14 species, including Rusty Patched Bumble Bee, thereby validating that 10-minute surveys using nonlethal techniques at randomly generated roadside locations can document the bumble bee community of the region. Declines in bumble bee species range and abundances are documented across multiple continents and have prompted the need for research to aid species recovery and conservation. The rusty patched bumble bee (Bombus affinis) is the first federally listed bumble bee species in North America. We conducted a range-wide population genetics study of B. affinis from across all extant conservation units to inform conservation efforts. To understand the species' vulnerability and help establish recovery targets, we examined population structure, patterns of genetic diversity, and population differentiation. Additionally, we conducted a site-level analysis of colony abundance to inform prioritizing areas for conservation, translocation, and other recovery actions. We find substantial evidence of population structuring along an east-to-west gradient. Putative populations show evidence of isolation by distance, high inbreeding coefficients, and a range-wide male diploidy rate of ~15%. Our results suggest the Appalachians represent a genetically distinct cluster with high levels of private alleles and substantial differentiation from the rest of the extant range. Site-level analyses suggest low colony abundance estimates for B. affinis compared to similar datasets of stable, co-occurring species. These results lend genetic support to trends from observational studies, suggesting that B. affinis has undergone a recent decline and exhibit substantial spatial structure. The low colony abundances observed here suggest caution in overinterpreting the stability of populations even where B. affinis is reliably detected interannually. These results help delineate informed management units, provide context for the potential risks of translocation programs, and help set clear recovery targets for this and other threatened bumble bee species. Mercury (Hg) is a globally ubiquitous pollutant that has received much attention due to its toxicity to humans and wildlife. The development of non-destructive sampling techniques is a critical step for sustainable monitoring of Hg accumulation. We evaluated the efficacy of non-destructive sampling techniques and assessed spatial, temporal, and demographic factors that influence Hg bioaccumulation in turtles. We collected muscle, blood, nail, and eggs from snapping turtles (Chelydra serpentina) inhabiting an Hg contaminated river. As predicted, all Hg tissue concentrations strongly and positively correlated with each other. Additionally, we validated our mathematical models against two additional Hg contaminated locations and found that tissue relationships developed from the validation sites did not significantly differ from those generated from the original sampling site. The models provided herein will be useful for a wide array of systems where biomonitoring of Hg in turtles needs to be accomplished in a conservation-minded fashion. [Copyright &y& Elsevier]

Amphibians with biphasic life histories occupy aquatic and terrestrial habitats at different times in their lives, leading to a double jeopardy of contaminant risk in both habitats. The present study examines individual and interactive effects of mercury exposure to terrestrial adults and aquatic larvae on fitness-related traits of American toads, Bufo americanus. Eggs from reference mothers or contaminated mothers were allowed to hatch and larvae were fed diets of either no added Hg or 2.5 or 10 µg/g total Hg (dry wt). Both dietary and maternal Hg had adverse effects on developing larvae, but there was no interaction between these factors. Dietary Hg had a marginal effect of decreased survival with increasing Hg in the diet. Animals from Hg-exposed mothers weighed 14% less than those from reference mothers, and size at metamorphosis was directly correlated with hopping performance. Animals from Hg-exposed mothers also took longer to complete metamorphosis and had 2.5 times the prevalence of spinal malformations compared with those from reference mothers. Results of the present study demonstrate that amphibians do indeed face a double jeopardy of contaminant exposure stemming from terrestrial and aquatic environments, because both exposure pathways adversely affected developing offspring. The present study also demonstrates that all possible routes of exposure over an organism's life history must be examined to provide a comprehensive picture of the ecological consequences of habitat contamination.
