Soybean mosaic virus (SMV) is a prevalent pathogen that causes significant yield reduction in soybean production worldwide. SMV belongs to potyvirus and causes typical symptoms such as mild mosaic, mosaic and necrosis. SMV is seed-borne and also transmitted by aphid. Eleven SMV strains, G1 to G7, G5H, G6H, G7H, and G7a were reported in soybean varieties in Korea. A reverse transcription loop-mediated isothermal amplification (RT-LAMP) method allowed one-step detection of gene amplification by simple procedure and needed only a simple incubator for isothermal template. This RT-LAMP method allowed direct detection of RNA from virus-infected plants without thermal cycling and gel electrophoresis. In this study, we designed RT-LAMP primers named SML-F3/B3/FIP/BIP from coat protein gene sequence of SMV. After the reaction of RT-LAMP, products were identified by electrophoresis and with the detective fluorescent dye, SYBR Green I under daylight and UV light. Optimal reaction condition was at 58°C for 60 min and the primers of RT-LAMP showed the specificity for nine SMV strains tested in this study.

Several tomato production regions in Korea were surveyed for tomato yellow leaf curl disease (TYLCD). Tomato leaf samples showing TYLCD-like symptoms were collected from Tongyeong (To), Geoje (Gi), and Gimhae (Gh) cities of the southern part of Korea. Tomato yellow leaf curl virus (TYLCV) was detected and the full-length genomes of the isolates were sequenced. The TYLCV isolates found in Korea shared high sequence identity (> 99%) with TYLCV-IL [JR:Omu:Ng] (AB110217). Phylogenetic relationship analysis revealed that they formed two groups (with little genetic variability), and the To, Gj, and Gh isolates belonged to the TYLCV-IL group. An infectious clone of TYLCV-To (JQ013089) was constructed and agroinoculated into Nicotiana benthamiana, Nicotiana tabacum var. Xanthi, Petunia hybrida, Capsicum annuum, and Lycopersicon esculentum cv. Hausumomotaro. Agroinfection with a dimeric infectious clone of TYLCV-To induced severe leaf curling and stunting symptoms in these plants, excluding C. annuum. Tomato plants then developed typical yellow leaf curl symptoms.

A sequential planting method was developed to screen rice plants with durable resistance against rice blast in a short time, and applied for several years in Korean rice breeding program. In this study, we showed the advantages of a sequential planting method compared to other pathogenicity tests. The correlation analysis among three pathogenicity tests and other factors demonstrated that durable resistance depended on the average of diseased leaf area and the number of compatible pathogens. Significant correlations were found in the nursery test but not in the field test result. In addition, we traced changes in the pathogen population during sequential planting stages through re-isolation of the pathogen. The portion of compatible pathogens was increased during sequential planting. Through this study, we provide an effective sequential planting method and direction of durable resistance in a breeding program.

For application of sodium hypochlorite as a seed disinfectant to the control of bakanae disease caused by Gibberella fujikuroi in rice, we investigated the effects of sodium hypochlorite for antifungal activity, eliminating fungus from seeds and reducing disease occurrence in vitro and greenhouse. The viability of the pathogen was significantly reduced at 80 ml/l concentration of sodium hypochlorite, and the pathogens did not grow at over 100 ml/l concentration of sodium hypochlorite. The effect of eliminating fungus was 90% at treatment of 0.3% sodium hypochlorite solution to infected rice seeds for eight hours. When the rice seeds were soaked into 0.5% and 0.3% sodium hypochlorite solutions for twelve hours, the disease incidences of rice seedling were remarkably reduced to 4.3% and 4.7%, respectively, compared to 97.3% of non-treatment control. The rates of seedling stand were 29.1% and 26.9% higher with the sodium hypochlorite treatment than that of non-treatment control. When prochloraz and sodium hypochlorite was treated to naturally severely infested rice seeds with bakanae disease, the disinfection effect was higher than that of prochloraz alone treatment. When the seeds were soaked in sodium hypochlorite before or after prochloraz, the rate of seed contamination was low as 4.0% or 6.3%, respectively, compared to prochloraz alone as 13.7%. The disease incidence was low as 3.7% or 8.3%, respectively, compared to prochloraz alone as 14.3%. The disinfection effect of treatment with prochloraz after sodium hypochlorite was higher than that of treatment with prochloraz before sodium hypochlorite.

A gene detection strategy using two-component Ac/Ds construct, with the mobile Ds transposon, has been developed to better understand gene functions in crops. Currently, 115,000 Ds insertion lines have been generated through the Ac/Ds gene trap system in Korea using japonica rice Dongjin as donor. Four hundred and thirty-seven mutants from 12,162 Ds-tagged lines were catalogued, including physiological and agronomic traits. Different traits were identified with distinct characteristics in terms of tillers, panicles, leaves, flowers, seed, chlorophyll content, and height. Culm and panicle length, number of panicles, and days to flowering of the Dongjin Ds population revealed high standard deviations compared with the donor cultivar. An evaluation of the Ds distribution on the chromosome revealed that 74.5% of the Ds were reinserted into gene-rich regions, making this Ac/Ds-mediated gene trap system useful in helping to gain an understanding of the function of genes and thus improve the gene-tagging system in rice. Data presented here are associated with the article: âIn-depth proteomic analysis of soybean (Glycine max) seeds during controlled deterioration treatment (CDT) reveals a shift in seed metabolismâ (Min et al., 2017) [1]. Seed deterioration is one of the major problems, affecting the seed quality, viability, and vigor in a negative manner. Here, we display the gel-based and gel-free proteomic data, associated with the CDT in soybean seeds. The present data was obtained from 2-DE, shotgun proteomic analysis (label-free quantitative proteomic analysis) using Q-Exactive, and gene ontology analysis associated with CDT in soybean seeds (Min et al., 2017) [1].

For QTL analysis of agronomic traits based on cultivation of low and high altitude locations, BC1 F5 181 lines were developed from a cross of Tongil type Gayabyeo and japonica Chhomrong originated from Nepal. Plant materials were grown in both of low altitude area of Milyang, Korea and high altitude area of Khumaltar, Nepal. In QTLs analysis, a total of 42 QTLs were detected in days to flowering, culm length, panicle length, number of panicles/hill, panicle exertion, and spikelet ripening ratio. Although many of the QTLs were coincided between the two locations of Korea and Nepal, several QTLs were revealed as location specific in high altitude area of Khumaltar. Especially, the regions harboring marker RM489-RM14281 on chromosome 3, RM5642-RM19049 on chromosome 5, and RM2854-RM6696 on chromosome 12 where QTLs were clustered and coincided between the two locations are considered as positive target regions for environmental independent traits. Furthermore, high rate of location specific QTLs such as panicle exertion and spikelet ripening ratio could be considered carefully for understanding the mechanism of cold tolerance based on cultivation of different altitude location. The groundnut or cultivated peanut (Arachis hypogaea L.) in Korea consists of 36 domestic varieties which have been developed and registered as cultivars for the public during last 25 years. To screen and identify of Korean peanut varieties and genetic resources, we present a simple and reliable method. A methodology based on simple sequence repeat (SSR) markers developed and widely used for prominent gene identification and variety discrimination. For identification of those 36 Korean peanut varieties, 238 unique peanut SSR markers were selected from some previously reported results, synthesized and used for polymerase chain reaction (PCR). Data were taken through acryl amide gel electrophoresis and changed into proper formats for application of data mining analysis using Biomine (all-in-one functional genomics data mining program). Consequently, twelve SSR primers were investigated and revealed the differences between those 36 varieties. These primer pairs amplified 27 alleles with an average of 2.3 allele per primer pair. In addition, those results showed genetic relationship by classification method within 36 varieties. Rice is a staple food for more than 50% of the world's population. Embryo comprises only 2 to 3% of the weight distribution of the entire pericarp but has higher concentration of vitamins, proteins, and essential fatty acids than the other parts of grains. Moreover, α-tocoperol, γ-oryzanol, phytic acid and γ-aminobutric acid that have nutraceutical value are abundant. Increasing the volume of embryo assures the fortification of nutritional value of rice grain. We developed new black waxy giant embryo rice, Milyang 263 by crossing Josaengheugchal, a black waxy rice variety, and get, a giant embryo mutant generated by tissue culture. The nutrient contents and physical properties of Milyang 263 were compared with several giant embryo mutants and normal embryo rice varieties. Changes in the nutrient properties after germination were also observed. Results indicated that this new black waxy giant embryo rice, Milyang 263, offers a promising source for improving nutritional quality of rice especially anthocyanin, essential minerals, and GABA. Recently, peoples are greatly concerned with global temperature change because global warming can be a potential serious effect on agriculture production such as yield reduction and poor grain quality. On the other hand, it can bring some beneficial effects through twice cultivation of rice in temperate region. In order to overcome this situation, we developed extremely early-maturing rice 'Milyang255' which heading date was similar with 'Jinbuolbyeo'. Based on agronomic characteristics of 'Milyang255', it has very short stature as 65 cm of culm length and slightly lower spikelets number per panicle compared with that of 'Jinbuolbyeo'. However, the grain appearance, palatability and other items were better than those of 'Jinbuolbyeo' in panel test of cooked rice. The milled rice yield of 'Milyang255' is 3.94 MT/ha at the early transplanting. Especially, its grain filling rate was higher and faster than Jinbuolbyeo and the head rice ratio in milled rice was also higher. Thus, 'Milyang255' can efficiently reduce the growth duration of rice cultivation and also be useful material for research on twice cultivation of rice in Korea. [ABSTRACT FROM AUTHOR]

'Geumyoung' is a new early maturing rice cultivar developed from the cross between Sambaegbyeo and Iksan423/ Sangju22 at Sangju Substation, NICS, RDA, Korea in 2009. It needs about 116 days from seedling to harvest in mid-mountainous areas. This cultivar has about 72 cm long culm and 19 cm long panicle, which are similar to Odaebyeo. It has higher panicle number per plant, medium to small grains and sometimes has a few awned spikelets. It shows tolerance to cold and resistance to blast but is susceptible to bacterial blight, virus diseases, and insects and pests. Milled rice of 'Geumyoeng' is translucent with non-glutinous endosperm and medium short grain. It has about 18.0% amylose and 6.6% protein content with good palatability of cooked rice. The milled rice yield of this cultivar was 5.53 MT/ha under standard fertilizer level of ordinary transplanting cultivation. 'Geumyong' would be adaptable for ordinary transplanting cultivation in the northern plains, and northern and southern mid-mountainous areas of South Korea. [ABSTRACT FROM AUTHOR]
