The immersion corrosion experiment of laser-based direct energy deposited H13 steel in ADC12 aluminum alloy melt was conducted to evaluate the demands for corrosion resistance after mold repair. The effect of the anisotropic microstructure of the deposited H13 steel on corrosion behavior was elucidated by characterizing the microstructure before and after corrosion. The results show that the microstructural heterogeneity affects the initiation, development and stabilization process of corrosion. Corrosion tends to initiate near the heat-affected zone due to the heterogeneous distribution of grain size, carbides, and dislocation density. Planes with weak microstructural heterogeneity are less likely to develop cracks in the intermetallic compounds (IMCs) layer during the corrosion development stage, thus exhibiting a slower corrosion rate. The IMCs layer of plane with weak microstructural heterogeneity stabilizes more quickly during the corrosion stabilization stage. The plane perpendicular to the build direction exhibits the highest corrosion resistance due to fewer corrosion initiation locations and the weakest microstructural heterogeneity. Furthermore, the composite structure of carbide core + Si phase shell, generated during corrosion, contributes to improving localized corrosion resistance. This study provides insights into understanding the influence of the heterogeneous microstructure from additive manufacturing on the reaction-diffusion process and offers guidance on direction selection when preparing components for applications in contact with aluminum melts using additive manufacturing techniques.

The cellular automaton-lattice Boltzmann method-immersed moving boundary coupled model is established to study the microstructure evolution during the solidification process of 2009Al/SiCp. After several model benchmarks, the natural convection of liquid phase and its effect on particle settlement under different pulling velocities were studied firstly. The results showed that at higher pulling velocity, the particle decelerates downward before contacting with dendrite front due to intensified natural convection. At lower pulling velocity, the particle decelerates gradually until a change in direction and upward movement. As the particle approach the tip of the dendrite, the particle experiences a brief upward movement before contacting the dendritic front. Then the convection induced by particles has an impact on the distribution of solutes in the liquid phase. It has been observed that at lower pulling velocity, there is a significant difference in solute distribution at the solidification front. The primary reason for this phenomenon is the higher solute concentration near the solidification front at lower pulling velocity, rather than the higher liquid phase flowing rate. Finally, our model investigated the microstructure evolution process during the directional solidification process of 2009Al/SiCp at different pulling velocities. The results indicate a good agreement between the simulation results and experiment observations.

The aim of the present study was to construct a molecular spectral database of magnesium oxide crystal arc plasma, thereby allowing for the net emission coefficient of arc to be calculated at different temperatures, pressures and Mg ratios. First, Rydberg-Klein-Rees inversion method was adopted to reconstruct the potential-energy curves of diatomic molecules. Subsequently, the Schrödinger equation was solved to determine the molecular wave function. This solution combined with the electrical transition moment function facilitates the calculation of radiative transition probabilities between different energy levels and the development of this database. Second, calculation models of the bound-free and free-free radiative absorption cross-section were established and use to calculate the continuous radiation of atoms. Finally, the radiative capability of different particles was calculated separately, followed by calculation of the net emission coefficient of plasma through addition The experimental results reveal that the net emission coefficient of air plasma under atmospheric pressure is consistent with existing research in both its magnitude and trend. Given that Mg can be ionized at lower temperatures, the magnesium oxide crystal arc plasma exhibited a pronounced radiative capacity at these temperatures. Additionally, a larger optical transmission distance corresponded to a reduced net emission coefficient of the plasma.

Previous studies have shown that subcutaneous adipose tissue is an important energy supply organ for chicks before and after birth, except yolk. So far, the significance of large deposits of subcutaneous adipose tissue in chicks is unclear. Therefore, this study takes the information interaction between adipocytes and macrophages as the starting point to explore whether adipocytes and macrophages could participate in adipose tissue fibrosis, angiogenesis, adaptive thermogenesis and other related functions in a specific metabolic environment. Under cold stress, the expression levels of genes related to lipidolysis, lipid transport and fatty acid oxidation in adipose tissue of chicks were significantly increased, but the expression levels of genes related to mitochondrial uncoupling were not significantly changed. Through Masson staining of adipose tissue of chicks under cold stress, it was found that the level of vascularization in adipose tissue of chicks was significantly increased. We found that the interaction between adipocyte and macrophage could participate in the angiogenesis related process of adipocytes in chicks through the HIF1A–VEGFA pathway. The analysis of lipid metabolism in subcutaneous adipose tissue of chicks from the perspective of cell heterogeneity will expand the understanding of lipid metabolism in chicks and provide a theoretical basis for chick rearing.

In this paper, coal-based solid waste, including fly ash, desulfurization gypsum, furnace bottom slag, and coal washing sludge in the ratio of 3:2:3:2 with a total of 200 tons/0.07 hm2, were used as the specialized main material for restoring the saline alkali land of Emao River in Huairen of China. The remediation effect and safety of solid waste artificial soil were evaluated by testing the soil samples before and after the remediation. The results showed that the pH value of the soil after remediation decreased from 9.98 to 7.60, which was close to the neutral value and suitable for crop growth. The total amount of water-soluble salts decreased from 8.30 g kg−1 to 4.80 g kg−1 with a decrease of 42.2 %. The organic matter increased from 6.5 g kg−1 to 39.1 g kg−1 with a 5-fold increase. Compared to the original soil, the heavy metal content in the restored soil did not increased, but instead decreased, indicating that the restoration technology was feasible and meets environmental requirements. Corn planting experiment results showed that corn's emergence rate in the original saline alkali soil was extremely low (about 1 %), while in the restored soil reached over 99 %. The average yield of corn in restoration field was 16.56 % higher than the average local yield level. The residual content of heavy metals and organic toxic substances in corn and potatoes grown on restored soil were analyzed, and the results showed that the detected heavy metal content was far lower than the standard values, and the residual organic toxic substances were basically not detected, indicating that the agricultural fruits grown on restored soil were safe, harmless, and edible. This approach could achieve large-scale consumption of coal-based solid waste, increase arable land, and reduce the cost of restoring saline alkali land.

Abstract Background Accurately predicting the response to neoadjuvant chemotherapy (NAC) in breast cancer patients is crucial for guiding treatment strategies and enhancing clinical outcomes. Current studies have primarily focused on a limited set of biomarkers. More importantly, the results of many studies are in conflict. To address this, we conducted a comprehensive evaluation of the predictive value of a diverse range of clinically available molecular biomarkers in breast cancer, including HER2, ER, PR, TOPO II, EGFR, Ki67, CK5/6, AR, and p53. Additionally, we assessed changes in these biomarkers after NAC administration. Methods Our study involved 189 patients with invasive breast cancer who underwent NAC at our institute. We examined biomarker profiles in core-needle biopsies taken before NAC and in surgical specimens obtained after NAC. We examined the association between these biomarkers and NAC outcomes, focusing on two main aspects: the rate of pathological complete response (pCR) and the reduction in tumor size. We used Chi-square and Mann-Whitney U tests to compare biomarker status changes between pCR and non-pCR patients. Linear regression analysis was employed to evaluate the relationship between biomarker status and tumor shrinkage rate. Additionally, we compared the expression status of these biomarkers before and after NAC using Chi-square and Wilcoxon signed-rank tests. Results and conclusions Our results demonstrated significant differences in the expression levels of HER2, ER, PR, TOPO II, EGFR, and Ki67 between pCR and non-pCR patients, underscoring their potential as predictive markers for NAC outcomes. Importantly, our results have shed light on the contentious issue surrounding TOPO II in NAC outcome prediction. We have provided evidence that establishes a significantly positive association between TOPO II expression level and the pCR rate. Notably, tumor size was identified as a relevant predictive factor for achieving pCR. Regarding biomarker profiles, only Ki67 levels and TOPO II status exhibited changes following NAC, resolving previous controversies. While the ER and PR status remained unchanged, their expression values exhibited a slight but significant decrease post-NAC. Our results provide clarity and insights into the value and potential of using these biomarkers to predict NAC responses and prognosis in breast cancer patients.

With the advantages of eco-friendliness, low cost, and low density, Mg2(Si,Sn) solid solutions are promising candidates for thermoelectric applications. In this work, Sb-doped Mg2Si0.4Sn0.6 bulks were prepared with a combined method of solid-state reaction and high pressure synthesis, followed by spark plasma sintering. Our investigations show that Sb doping optimizes the carrier concentration, while Si/Sn alloying effectively suppresses the lattice thermal conductivity and induces a convergence of the two lowest-lying conduction bands. Additionally, numerous coherent Sn-rich nanoprecipitates are formed within micron-sized grains. All these factors contribute synergistically to improving the thermoelectric properties of Mg2Si0.4Sn0.6. The optimal Mg2(Si0.4Sn0.6)0.985Sb0.015 exhibits a power factor higher than 4 000 μW·m−1·K−2 and a lattice thermal conductivity less than 0.8 W·m−1·K−1 at temperatures higher than 600 K, leading to the highest ZT of 1.61 at 823 K. Current work demonstrates an effective approach to enhancing the thermoelectric performance of n-type Mg2X solid solutions through doping, alloying, and microstructure modification.

ABSTRACTIn present paper, a universal method for multi-objective parameter optimisation of additive manufacturing processes was proposed and successfully applied to laser-based directed energy deposition (DED) experiments with mixtures of H13 and M2 steel powders that were deposited on 4Cr5Mo2SiV1 hot work die steel. The DED experiments were designed and completed based on the response surface method with 13 groups of laser parameters. The microstructure of the deposited alloy steel was observed and its mechanical properties were tested. The deposited steel alloy achieved an ultimate tensile strength (UTS) of 1821 ± 30 MPa with a reasonable elongation of approximately 4.5%, and the bond strength specimens achieved a bond toughness of ∼10.66% with a moderate UTS (1329 ± 28 MPa). A multi-objective optimisation method was proposed based on response surfaces which were established according to microstructural characteristics and mechanical properties data. It provided a basis for achieving high strength or high toughness DED-fabricated steel alloys.

The synergistic effect of externally solidified crystals (ESCs) and Fe-rich intermetallic on the fracture behavior of high pressure die casting Al–8Si-0.4 Mg–2Zn alloy was investigated using in-situ tensile test. The results revealed that the localized stress concentration during tensile test were significantly depended on the characteristic of ESCs and Fe-rich intermetallic. The crack preferentially initiated in the region where fine Fe-rich phase was surrounded by the clustered ESCs, this mainly attributed to the inconsistency of stress distribution in the clustered ESCs during tensile loading. Meanwhile, the primary Fe-rich phase with high aspect ratio is also beneficial to the crack initiation. The cracks were propagated both in the grain boundary and internal region of ESCs. The final fracture occurred in both the interfacial region between ESCs and other intermetallic and the internal region of ESCs.

The three-dimensional (3D) self-supporting Pt nanoflower catalyst was prepared by using Pt organic compounds as precursors in a low-temperature molten salt system. The obtained Pt nanoflower nanoparticle can reach 400–500 nm with a face-centered cubic structure, which has the structural characteristics of a loose framework and more exposed active sites. The surfactant-free Pt nanoflower was directly used as methanol electrooxidation reaction (MOR) electrocatalysts without any pretreatment and activated carbon support. The results of electrochemical catalytic oxidation of methanol showed that the Pt nanoflower catalyst exhibit more enhanced electrocatalytic oxidation performance toward methanol compared with the commercial platinum black catalyst. The green and effective method can be developed and expected to enable mass production of fuel cell catalysts.

Transition metal selenides have the leading position in the field of energy storage and conversion due to their high theoretical capacity, good electrical conductivity, and cycling stability. Nickel is widely used for the construction of positive electrodes in devices due to its good conductivity, variable valence state, and ideal redox activity. NiSe materials have high internal resistance and are prone to volume change during charging and discharging, thus affecting the practical application of this electrode material, and the reported NiSe materials have not achieved a more desirable capacity value. Therefore, in this study, N, P-NiSe nanoelectrode materials were prepared using nickel foam as the nickel source and hexachlorocyclotriphonitrile as the nitrogen and phosphorus dopant using an efficient, energy-saving, and simple microwave method. It was also characterised by XRD and XPS to confirm the successful preparation of N, P-NiSe materials. In addition, the material yielded a high capacitance value (3184 F g−1) and good cycling stability (72% of the initial capacitance value was retained after 4000 cycles) in electrochemical tests. To demonstrate its excellent suitability for practical applications, an asymmetric supercapacitor was assembled using N, P-NiSe as the anode and activated carbon as the cathode. At an operating voltage of 1.6 V, the device achieved an energy density of 289.06 Wh kg−1 and a power density of 799.26 W kg−1 and retained 80% of its initial capacity after 20,000 cycles.

This paper investigates the effect of Nb and B microalloying on the wear and corrosion resistance of 20MnCr5 steel after high-temperature carburization at 950 °C. The abrasion and corrosion performance of the steel was compared using electrochemical experiments and dry sliding wear tests. The results show that Nb microalloying reduces the size of residual austenite, thereby improving the wear resistance of 20MnCr5 steel. Furthermore, the combination of Nb and B microalloying reduces the amount of residual austenite, leading to further improvements in wear resistance. Both microalloying methods reduce the wear scar depth and debris accumulation. The corrosion potentials of 20MnCr5, 20MnCr5Nb, and 20MnCr5NbB are 0.645 V, 0.627 V, and 0.675 V, respectively. The addition of Nb to 20MnCr5 steel results in higher corrosion resistance after carburization due to the refinement of needle-like martensite. However, 20MnCr5NbB steel exhibits low corrosion resistance due to the coarsening of needle-like martensite caused by B microalloying. In summary, this study provides new insights into the effects of microalloying elements on the properties of steel. The findings contribute to a deeper understanding of how to design and manufacture gears and other components with superior wear and corrosion resistance.

Abstract Background The chlorophyll content (CC) is a key factor affecting maize photosynthetic efficiency and the final yield. However, its genetic basis remains unclear. The development of statistical methods has enabled researchers to design and apply various GWAS models, including MLM, MLMM, SUPER, FarmCPU, BLINK and 3VmrMLM. Comparative analysis of their results can lead to more effective mining of key genes. Results The heritability of CC was 0.86. Six statistical models (MLM, BLINK, MLMM, FarmCPU, SUPER, and 3VmrMLM) and 1.25 million SNPs were used for the GWAS. A total of 140 quantitative trait nucleotides (QTNs) were detected, with 3VmrMLM and MLM detecting the most (118) and fewest (3) QTNs, respectively. The QTNs were associated with 481 genes and explained 0.29-10.28% of the phenotypic variation. Additionally, 10 co-located QTNs were detected by at least two different models or methods, three co-located QTNs were identified in at least two different environments, and six co-located QTNs were detected by different models or methods in different environments. Moreover, 69 candidate genes within or near these stable QTNs were screened based on the B73 (RefGen_v2) genome. GRMZM2G110408 (ZmCCS3) was identified by multiple models and in multiple environments. The functional characterization of this gene indicated the encoded protein likely contributes to chlorophyll biosynthesis. In addition, the CC differed significantly between the haplotypes of the significant QTN in this gene, and CC was higher for haplotype 1. Conclusion This study’s results broaden our understanding of the genetic basis of CC, mining key genes related to CC and may be relevant for the ideotype-based breeding of new maize varieties with high photosynthetic efficiency.

The authors show that in recent decades compound climate extremes (i.e., hot, dry, and windy events) have increased and have reduced winter wheat yields in the U.S. Great Plains. The area most affected is the same area as that in 1930s Dust Bowl.

The high vacuum die casting (HVDC) AlSiMgMnCu alloy sheets were successfully welded by stationary shoulder friction stir welding (SSFSW) technique, and a post-weld heat treatment (PWHT) was also performed to enhance the mechanical properties of the joints. The microstructure and maximum texture intensity obviously change in different zones before and after the heat treatment. In the as-SSFSW joint, the microstructure in nugget zone (NZ) and thermal mechanically zone (TMAZ) possessed stronger texture of brass components than the heat affect zone (HAZ). After the T6 PWHT, the joints of the alloy showed continuous recrystallization. The texture type is still brass texture in NZ and TMAZ, while the texture intensity in the NZ and TMAZ significant increased. Some grains in NZ and TMAZ rotate 20° towards the . The nano-precipitation β'', Q′ and θ′ phases formed in the Al matrix after the T6 PWHT. The yield strength, ultimate strength and elongation of PWHT joints are 252 MPa, 297 MPa and 4.1%, respectively. The joint welding coefficient of PWHT joint is 96.1%, which was ascribed to dislocation and precipitation strengthening.

Warm temperatures due to increases of greenhouse gas emissions have changed temperature distribution patterns especially for their extremes, which negatively affect crop yields. However, the assessment of these negative impacts remains unclear when surface precipitation patterns are shifted. Using a statistical model along with 23,944 county-year maize-yield data during 1981–2020 in the US Corn Belt, we found that the occurrence of timely precipitation reduced the sensitivity of maize yields to extreme heat by an average of 20% during the growing season with variations across phenological periods. Spatially across the US corn belt, maize in the northern region exhibited more significant benefits from timely precipitation compared to the southern region, despite the pronounced negative effects of extreme heat on yields in cooler regions. This study underscores the necessity of incorporating timely precipitation as a pivotal factor in estimating heat effects under evolving climates, offering valuable insights into complex climate-related challenges.

A method to calculate element concentrations in Al alloy castings, based on the serial block-face scanning electron microscopy (SBFSEM) measurement at nano-scale, was proposed. For the high vacuum die casting (HVDC) Al-10Si-0.4MgMn-0.6Cu alloy, it reveals that the solute fractions of the Si, Mg and Cu in the Al matrix reaches about 0.39 wt%, 0.15 wt%, and 0.10 wt%, respectively, under the as-cast condition. After it was heat treated via T5 process, larger nano-scale precipitation phases with low density and non-uniform distribution were formed in the α-Al matrix. After T6-treated, the narrow neck regions of the seaweed-like α-Fe phase particles were fragmented as well, but could not dissolve into the α-Al matrix. The element concentrations of the Si, Mg and Cu in the α-Al matrix increases to 0.75 wt%, 0.23 wt%, and 0.43 wt%, respectively. Hence, finer nano-scale precipitation phases with high density and uniform distribution are formed in the Al matrix. Consequently, the ultimate tensile strength, yield strength and elongation of the T6-treated alloy simultaneously increase of about 15%, 52% and 63%, respectively. It presents that the solute solution during high pressure die casting solidification has decisive effect on the mechanical properties before and after heat treatment.

Abstract Pulsed amplitude modulation (PAM) chlorophyll a fluorescence provides information about photosynthetic energy transduction. When reliably measured, chlorophyll a fluorescence provides detailed information about critical in vivo photosynthetic processes. Such information has recently provided novel and critical insights into how the yield potential of crops can be improved and it is being used to understand remotely sensed fluorescence, which is termed solar‐induced fluorescence and will be solely measured by a satellite scheduled to be launched this year. While PAM chlorophyll a fluorometers measure fluorescence intensity per se, herein we articulate the axiomatic criteria by which instrumentally detected intensities can be assumed to assess fluorescence yield, a phenomenon quite different than fluorescence intensity and one that provides critical insight about how solar energy is variably partitioned into the biosphere. An integrated mathematical, phenomenological, and practical discussion of many useful chlorophyll a fluorescence parameters is presented. We draw attention to, and provide examples of, potential uncertainties that can result from incorrect methodological practices and potentially problematic instrumental design features. Fundamentals of fluorescence measurements are discussed, including the major assumptions underlying the signals and the methodological caveats about taking measurements during both dark‐ and light‐adapted conditions. Key fluorescence parameters are discussed in the context of recent applications under environmental stress. Nuanced information that can be gleaned from intra‐comparisons of fluorescence‐derived parameters and intercomparisons of fluorescence‐derived parameters with those based on other techniques is elucidated.

Abstract Background Occult metastases in axillary lymph nodes have been reported to be associated with poor prognosis in patients with breast cancer. However, studies on the prognostic value of occult metastases have shown controversial results. This meta-analysis aimed to evaluate the prognostic significance of occult lymph node metastases in breast cancer. Methods Studies published until May, 2020, which retrospectively examined negative lymph nodes by stepsectioning and/or immunohistochemistry, were retrieved from MEDLINE, EMBASE, CNKI, and Cochrane Library databases. The pooled Relative Risk (RR) with 95% confidence interval (95% CI) for overall survival (OS) and disease-free survival (DFS) were calculated to examine the associations between occult metastases and prognosis. Results Patients with occult metastases in axillary lymph nodes had poorer five-year DFS (RR = 0.930; 95% CI = 0.907–0.954) and OS (RR = 0.972; 95% CI = 0.954–0.990). Furthermore, the DFS (RR = 0.887; 95% CI = 0.810–0.972) and OS (RR = 0.896; 95% CI = 0.856–0.939) of patients with occult metastases were significantly lower after a ten-year follow-up. Conclusions Occult metastases in the axillary lymph nodes are associated with poorer DFS andOS of patients with breast cancer. Occult metastases might serve as a predictive factor of survival outcomes in patients with breast cancer.

With high fecundity and short production cycle, poultry is one of the important sources of meat. During the embryonic and post-hatch period, the higher death rate caused huge economic losses in poultry production. Our previous study showed that chick subcutaneous adipose tissue is an important energy supply tissue besides yolk. Therefore, the metabolic mechanism of subcutaneous adipose tissue in chicks could provide a new perspective of brooding. The objectives of the current study were to evaluate the differences between chick subcutaneous adipose tissue and abdominal adipose tissue before and after hatching and reveal the cross-talk of different cells within the chick subcutaneous adipose tissue. The results of RNA-seq and weighted gene co-expression network analysis (WGCNA) of chick subcutaneous and abdominal adipose tissues showed that the function of chick subcutaneous tissue was related to immunoreaction, and macrophage could be the major immune infiltration cell type in chicken subcutaneous adipose tissue, which were also verified by qPCR, HE stain, and IHC. The results of free fatty acids (FFAs)-induced the cross-talk between macrophages and adipocytes showed that FFAs-Ccl2 (chicken CCL26) axis could have an important role in lipid transportation in adipose tissue. The results of Oil Red O and Nile red stain demonstrated that macrophages have the ability to absorb FFAs quickly. Interestingly, according to the genomic organization of CCL family with representative vertebrate species, we found that chicken CCL26 could be the major chemokine in chicken adipocyte as the status of CCL2 in mammal adipocyte. In conclusion, we demonstrate that FFA-induced Ccl2 (chicken CCL26) secretion is crucial in determining fat depot-selective adipose tissue macrophage (ATM) infiltration, which could be an important medium of lipid transportation in chicken subcutaneous adipose tissue. These findings may have multiple important implications for understanding macrophage biology with chick subcutaneous adipose tissue and provide theoretical basis for lipid metabolism in poultry brooding.

BackgroundMitoxantrone hydrochloride injection for tracing (MHI), a new strategy to identify lymph nodes, has not been tested for axillary node staging in breast cancer. This multicenter, self-controlled, non-inferiority trial aimed to evaluate MHI’s efficacy and safety in sentinel lymph node biopsy (SLNB).MethodsThe trial was conducted across seven hospitals from December 2019 to December 2020. Patients with early-stage breast cancer received MHI and technetium-99m (99mTc) during the surgery. Sentinel node detection rates were compared between MHI and 99mTc to evaluate non-inferiority and concordance. Non-inferiority was valid if the lower limit of the 95% CI of sentinel node relative detection rate difference was ≥−5%.ResultsSLN relative detection rate of MHI was 97.31% (362/372). Of the SLNs, 79.69% (871/1093) were co-detected by both tracers. Of the patients, 4.13% (16/387) had adverse events and recovered during the follow-up.ConclusionsMHI is a lymphatic tracer with comparable efficacy to radionuclides and can be used alone or in combination with radioactive substances for SLNB.Clinical Trial Registrationhttp://www.chinadrugtrials.org.cn, CTR20192435.

The microstructural characteristics of the high pressure die casting (HPDC) AlSiMgMnCu alloys in the skin layer, segregation band and the central zone were investigated in three dimensions by serial block-face scanning electron microscopy (SBFSEM). The results present that the volume fractions of α-Al15(Fe, Mn)3Si (α-Fe phase), β-Mg2Si (β phase) and Q-Al5Cu2Mg8Si6 (Q phase) in the skin layer and the central zone are higher than those in the segregation band zone. The volume fractions and sizes of the θ-Al2Cu phase (θ phase) in the skin layer and the central zone are lower than those in the segregation band zone. In the skin layer, the preferential growth orientation of the α-Fe phase with plate-like morphology is parallel to the heat transfer direction, while the other phases exhibit a complex seaweed-like morphology. In the central zone and the segregation band zone, the shearing stress and turbulent flow during the filling process lead to the morphology of α-Fe and β-Mg2Si relatively regular. However, the morphology of Q-Al5Cu2Mg8Si6 and θ-Al2Cu phases are irregular owing to their lower solidification temperatures. The distribution and morphology of β-Mg2Si, Q-Al5Cu2Mg8Si6 and θ-Al2Cu in the segregation band zone mainly determined the distribution and morphology of the α-Fe phase.

Abstract Background Breast cancer (BC) remains a prevalent and common form of cancer with high heterogeneity. Making efforts to explore novel molecular biomarkers and serve as potential disease indicators, which is essential to effectively enhance the prognosis and individualized treatment of BC. FBXO proteins act as the core component of E3 ubiquitin ligase, which play essential regulators roles in multiple cellular processes. Recently, research has indicated that FBXOs also play significant roles in cancer development. However, the molecular functions of these family members in BC have not been fully elucidated. Methods In this research, we investigated the expression data, survival relevance and mutation situation of 10 FBXO members (FBXO1, 2, 5, 6, 16, 17, 22, 28, 31 and 45) in patients with BC from the Oncomine, GEPIA, HPA, Kaplan–Meier Plotter, UALCAN and cBioPortal databases. The high transcriptional levels of FBXO1 in different subtypes of BC were verified by immunohistochemical staining and the specific mutations of FBXO1 were obtained from COSMIC database. Top 10 genes with the highest correlation to FBXO1 were identified through cBioPortal and COXPRESdb tools. Additionally, functional enrichment analysis, PPI network and survival relevance of FBXO1 and co-expressed genes in BC were obtained from DAVID, STRING, UCSC Xena, GEPIA, bc-GenExMiner and Kaplan–Meier Plotter databases. FBXO1 siRNAs were transfected into MCF-7 and MDA-MB-231 cell lines. Expression of FBXO1 in BC cell lines was detected by western-blot and RT-qPCR. Cell proliferation was detected by using CCK-8 kit and colony formation assay. Cell migration was detected by wound‐healing and transwell migration assay. Results We found that FBXO2, FBXO6, FBXO16 and FBXO17 were potential favorable prognostic factors for BC. FBXO1, FBXO5, FBXO22, FBXO28, FBXO31 and FBXO45 may be the independent poor prognostic factors for BC. All of them were correlated to clinicopathological staging. Moreover, knockdown of FBXO1 in MCF7 and MDA-MB-231 cell lines resulted in decreased cell proliferation and migration in vitro. We identified that FBXO1 was an excellent molecular biomarker and therapeutic target for different molecular typing of BC. Conclusion This study implies that FBXO1, FBXO2, FBXO5, FBXO6, FBXO16, FBXO17, FBXO22, FBXO28, FBXO31 and FBXO45 genes are potential clinical targets and prognostic biomarkers for patients with different molecular typing of BC. In addition, the overexpression of FBXO1 is always found in breast cancer and predicts disadvantageous prognosis, implicating it could as an appealing therapeutic target for breast cancer patients.

As important livestock in Qinghai-Tibet Plateau, yak provides meat and other necessities for Tibetans living. Plateau yak has resistance to diseases and stress, yet is nearly unknown in the structure and expression mechanism of yak immunoglobulin loci. Based on the published immunoglobulin genes of bovids (cattle, sheep and goat), the genomic organization of the yak immunoglobulin heavy chain (IgH) and immunoglobulin light chain (IgL) were described. The assemblage diversity of IgH, Igλ and Igκ in yak was similar to that in bovids, and contributes little to the antibody lineage compared with that in humans and mice. Somatic hypermutation (SHM) had a greater effect on immunoglobulin diversity in yak than in goat and sheep, and in addition to the complementarity-determining region (CDR), some loci in the framework region (FR) also showed high frequency mutations. CDR3 diversity showed that immunological lineages in yak were overwhelmingly generated through linkage diversity in IgH rearrangements. The emergence of new high-throughput sequencing technologies and the yak whole genome (2019) publication have greatly improved our understanding of the immune response in yaks. We had a more comprehensive analysis of yak immunoglobulin expression diversity by PE300, which avoided the disadvantage of missing low-frequency recombination in traditional Sanger sequencing. In summary, we described the schematic structure of the genomic organization of yak IgH loci and IgL loci. The analysis of immunoglobulin expression diversity showed that yak made up for the deficiency of V(D)J recombinant diversity by junctional diversity and CDR3 diversity. In addition, yak, like cattle, also had the same ultra-long IgH CDR3 (CDR3H), which provided more contribution to the diverse expression of yak immunoglobulin. These findings might provide a theoretical basis for disease resistance breeding and vaccine development in yak.

Since excess abdominal fat is one of the main problems in the broiler industry for the development of modern broiler and layer industry, the importance of subcutaneous adipose tissue has been neglected. However, chick subcutaneous adipose tissue appeared earlier than abdominal adipose tissue and more than abdominal adipose tissue. Despite a wealth of data, detailed information is lacking about the development and function of chick subcutaneous adipose tissue during the embryonic and posthatch period. Therefore, the objective of the current study was to determine the developmental changes of adipocyte differentiation, lipid synthesis, lipolysis, fatty acid β-oxidation, and lipid contents from E12 to D9.5. The results showed that subcutaneous adipose tissue was another important energy supply tissue during the posthatch period. In this stage, the mitochondrial copy number and fatty acid β-oxidation level significantly increased. It revealed that chick subcutaneous adipose tissue not only has the function of energy supply by lipidolysis but also performs the same function as brown adipose tissue to some extent, despite that the brown adipose tissue does not exist in birds. In addition, this finding improved the theory of energy supply in the embryonic and posthatch period and might provide theoretical basis on physiological characteristics of lipid metabolism in chicks.

The transportation is a crucial phase in beef cattle industry, and the annual losses caused by beef cattle transport stress are substantial. Several studies have described the effect of long distance transportation stress on animal health, such as disorder in nervous, endocrine, immune, and metabolic system. However, molecular mechanisms underlying short distance transportation stress is still poorly understood. Present study aims to investigate the effect of short distance transportation by measuring the hematological indices and transcriptomic analysis. In this study, a total 10 Qinchuan cattle were used to compare the molecular characteristics of blood before and after transportation. We have found that a stress-related marker “white blood cell count (WBC)” increased significantly after transportation. The decrease in triglyceride (TG), cholestenone (CHO), high-density lipoprotein (HDL), and low-density lipoprotein (LDL) showed that energy expenditure was increased after transportation, but not enough to activate fatty decomposition. Intriguingly, the decrease of malondialdehyde (MDA) showed that cattle were more resilience to oxidative stress. The RNA-seq showed that 1,092 differentially expressed genes (DEGs) were found (329 up-regulated and 763 down-regulated) between group before and group after. The GO and KEGG enrichment showed that the metabolic pathway and B cell function related pathways were enriched. Furthermore, median absolute deviation (MAD) top 5,000 genes were used to construct a co-expression network by weighted correlation network analysis (WGCNA), and 11 independent modules were identified. Combing with protein-protein interaction (PPI) analysis, the verification of quantitative real-time PCR (qPCR) and the correlation of B cell function, structural maintenance of chromosomes 3 (SMC3), jun proto-oncogene (JUN), and C-X-C motif chemokine ligand 10 (CXCL10) were suggested as potential molecular markers in identification of short distance transportation. Collectively, the blood RNA-seq analysis and WGCNA indicated that the disorder of B cell differentiation, proliferation, survival, and apoptosis were the potential molecular mechanism in short distance transportation stress. In conclusion, our results provide the novel insight about potential biomarkers for short distance transportation stress, which may serve as for diagnosing and preventing this condition in beef industry.

Background: Cancer patients had been profoundly affected by the outbreak of COVID-19 especially after quarantine restrictions in China. We aimed to explore the treatment changes and delays of early breast cancer (EBC) during the first quarter of 2020. Methods: We did this retrospective, multicentre, cohort study at 97 cancer centres in China. EBC patients who received treatment regardless of preoperative therapy, surgery or postoperative therapy during first quarter of 2020 were included. Findings: 8397 patients were eligible with a median age of 50 (IQR 43–56). 0·2% (15/8397) of EBC patients were confirmed as COVID-19 infection. Only 5·2% of breast cancer diagnosis occurred after quarantine in Hubei compared with 15·3% in other provinces (OR= 0·30, 95%CI 0·24–0·38). postoperative endocrine therapy were least affected compared with different regions after quarantine (OR=0·37 [95%CI 0·19–0·73]). The proportion of surgery decreased from 16·4% in December last year to 2·6% in February in Hubei. Compared with intervals from diagnosis to treatment before quarantine restrictions, the average time increased with significance from 3·5 to 7·7 days in Hubei and 5·7 to 7·7 days in other provinces (p< 0·001). There were also 18·5 and 7·2 days delay in Hubei and other provinces respectively when calculating interval from surgery to postoperative therapy. Interpretation: EBC from high risk regions had a comparative rate of COVID-19 infection. After implementation of COVID-19 quarantine restrictions, fewer diagnosis and surgery with significant delays were seen when compared with treatment before. Transportation is a crucial phase in the beef cattle industry, and the annual losses caused by beef cattle transport stress are substantial. Because of its huge economic losses, such as lower growth rate and even death, long-distance transportation stress has attracted more attention from beef production practitioners because of its huge economic losses. Compared with the long-distance transportation stress, the short-distance transportation stress was ignored for the reason of no obvious symptoms in cattle. Our previous study showed that the disorder of B cell function could be a potential health risk after short-distance transportation. However, the transcriptome details of the changes in the cattle blood after short-distance transportation and the molecular mechanisms for the regulation of the developmental process are not clearly known. In this study, a total of 10 Qinchuan cattle were used to compare the molecular characteristics of blood before and after short-distance transportation. The miRNA-seq showed that 114 differentially expressed miRNAs (DEMs) were found (40 upregulated and 74 downregulated) between two groups before and after transportation. Furthermore, more than 90% of the miRNAs with counts of more than 10 were used to construct a co-expression network by weighted correlation network analysis (WGCNA), and four independent modules were identified. According to their relationship with 30 hub genes, the turquoise module was the key module in this study. The regulator network of hub genes and miRNAs in the turquoise module was constructed by miRNAs targeting genes predicting, and the miRNAs had targeting sites within hub genes that could be identified as hub-miRNAs. Further, it showed that CD40 and ITPKB had the same targeting miRNAs (miR-339a/b), and the newly discovered hub miRNAs filled the gaps in our previous study about the relationship between hub genes in short-distance transportation stress and provided the potential utility for predicting and treatment of short-distance transportation stress in beef cattle.

The high-volume fraction SiC/Al composite is the new type of electronic packaging material, which plays an important role in the field of high-power integrated circuits. In this study, SiC/Al composites with high-volume fraction of SiC particles were prepared by vacuum pressure infiltration. The influence of SiC particle size and NH4HCO3 on the pores in the preform was explored, aiming to accurately adjust the volume fraction of SiC to meet the thermal performance requirements in different fields. In addition, the preform was infiltrated with different Al alloys, and the relationship between the porosity and thermal conductivity of SiC/Al was studied. For the SiC preform, the volume fraction of SiC can be adjusted regularly when 12 μm and 100 μm SiC particles are mixed in different proportions, and the volume fraction reaches the maximum when the proportion of coarse particles is about 77%. NH4HCO3 is conducive to the connectivity of pores in the preform, and about 40 vol.% of NH4HCO3 can effectively increase the porosity of the preform. Thermal conductivity is sensitive to the porosity of composites, especially in the range of 2.5–4.5%. A simple application of the Hasselman–Johnson model and a new thermal conductivity model, λd, established in this article, offer a good agreement with the experimental results.

The purpose of this study was to explore functional variants in the prosaposin (PSAP) three prime untranslated region (3’ UTR) and clarify the relationship between the variants and morphological traits. Through Sanger sequencing, 13 variations were identified in bovine PSAP in four Chinese cattle breeds, with six of them being loci in 3’ UTR. In particular, Nanyang (NY) cattle had a special genotype and haplotype distribution compared to the other three breeds. NY cattle with ACATG and GCGTG haplotypes had higher morphological traits than GTACA and GTACG haplotypes. The results of dual-luciferase reporter assay showed that ACATG and GCGTG haplotypes affected the morphological traits of NY cattle by altering the secondary structure of PSAP 3’ UTR rather than the miR-184 target sites. The findings of this study could be an evidence of a complex and varying mechanism between variants and animal morphological traits and could be used to complement candidate genes for molecular breeding.

Zinc finger BED-type containing 6 (ZBED6), a highly conservative transcription factor of placental mammals, has conservative interaction of insulin-like growth factor 2 (IGF2) based on the 16 bp binding sites of ZBED6 on the IGF2 sequence. IGF2 is related to embryo growth and cell proliferation. At the same time, its functions in muscle and adipose in mammals have been widely mentioned in recent studies. To further investigate the mechanism of ZBED6 on IGF2, we detected the expression of IGF2 and related genes in ZBED6 single allele knockout (ZBED6-SKO) pig tissues and analyzed the transcriptome of ZBED6-SKO pig liver. Through RNA-seq, we captured nine up-regulated genes and eight down-regulated genes which related to lipid metabolism. The results showed that the mRNA of IGF2 had an upward trend after the partial knockout of ZBED6 in liver and had no significant difference in protein expression of IGF2. In summary, ZBED6-SKO could affect the secretion of IGF2 in pig liver and its own lipid metabolism. Our research has provided basic information for revealing the regulatory mechanism of the interaction between ZBED6 and IGF2 in mammals.

The GH growth axis plays an important role in the growth and development of animals and runs through the whole life of animals. Many studies have shown that molecular mutations in key genes of the GH axis will affect the growth and development of animals. The purpose of this study was to explore the distribution characteristics of InDels of GHR, GHRH, and GHRHR in seven Chinese sheep populations, and to further explore the relationship between InDels and sheep growth traits. GHR showed high variation in Chinese sheep, and GHR-53 showed the highest minimum allele frequency (MAF). There was only one InDel mutation site in both GHRH and GHRHR. The genotype frequencies of Hu sheep (HS), Tong sheep (TS), and Lanzhou fat-tail sheep (LFTS) were quite different from other breeds. The association between GHR, GHRH, and GHRHR InDels and body size traits in seven varieties were analyzed. The results showed that there was no significant relationship between GHRH and body size traits in the seven sheep populations. There was a positive association between GHR-21 and hip height of LFSH (p < 0.05). GHR-43 reduced body height and chest depth of Small tail han sheep (STHS) and hip width of TS. GHR-44 significantly affected the body weight of HS, the body height of STHS and the head depth of TS. GHR-53 significantly reduced cannon girth of HS, chest of STHS and forehead width of TS. GHRHR-2 significantly reduced the body weight of LFHS. To sum up, this study revealed the effects of GHR, GHRH, and GHRHR InDels on sheep phenotypic traits, which indicated their potential application prospects in the genetic improvement of mutton sheep.

Ti-based alloys are finding ever-increasing applications in biomaterials due to their excellent mechanical, physical and biological performance. Nowdays, low modulus β-type Ti-based alloys are still being developed. Meanwhile, porous Ti-based alloys are being developed as an alternative orthopedic implant material, as they can provide good biological fixation through bone tissue ingrowth into the porous network. This paper focuses on recent developments of biomedical Ti-based alloys. It can be divided into four main sections. The first section focuses on the fundamental requirements titanium biomaterial should fulfill and its market and application prospects. This section is followed by discussing basic phases, alloying elements and mechanical properties of low modulus β-type Ti-based alloys. Thermal treatment, grain size, texture and properties in Ti-based alloys and their limitations are dicussed in the third section. Finally, the fourth section reviews the influence of microstructural configurations on mechanical properties of porous Ti-based alloys and all known methods for fabricating porous Ti-based alloys. This section also reviews prospects and challenges of porous Ti-based alloys, emphasizing their current status, future opportunities and obstacles for expanded applications. Overall, efforts have been made to reveal the latest scenario of bulk and porous Ti-based materials for biomedical applications.

Al−Si−Mg based alloys can provide high strength and ductility to satisfy the increasing demands of thin wall castings for automotive applications. This study has investigated the effects of T6 heat-treatment on the microstructures, the local mechanical properties of alloy phases and the fracture behavior of high vacuum die-cast AlSiMgMn alloys using in-situ scanning electron microscopy (SEM) in combination with nano-indentation testing. The microstructures of the alloys at as-cast and T6 treated conditions were compared and analyzed. It is found that the T6 heat treatment plays different roles in affecting the hardness and the Young’s modulus of alloy phases. This study also found that the T6 heat treatment would influence the failure modes of the alloys. The mechanisms of crack propagation in the as-cast and T6 treated alloys were also analyzed and discussed.

Intestinal ischemia-reperfusion (I/R) injury is a serious clinical pathophysiological process that may result in acute local intestine and remote liver injury. Protocatechuic acid (PCA), which has been widely studied as a polyphenolic compound, induces expression of antioxidative genes that combat oxidative stress and cell apoptosis. In this study, we investigated the effect of PCA pretreatment for protecting intestinal I/R-induced local intestine and remote liver injury in mice. Intestinal I/R was established by superior mesenteric artery occlusion for 45 min followed by reperfusion for 90 min. After the reperfusion period, PCA pretreatment markedly alleviated intestine and liver injury induced by intestinal I/R as indicated by histological alterations, decreases in serological damage parameters and nuclear factor-kappa B and phospho-foxo3a protein expression levels, and increases in glutathione, glutathione peroxidase, manganese superoxide dismutase protein expression, and Bcl-xL protein expression in the intestine and liver. These parameters were accompanied by PCA-induced adaptor protein p66shc suppression. These results suggest that PCA has a significant protective effect in the intestine and liver following injury induced by intestinal I/R. The protective effect of PCA may be attributed to the suppression of p66shc and the regulation of p66shc-related antioxidative and antiapoptotic factors.

Unprecedented precipitation deficits in the 2022-2023 growing season across the primary wheat-producing region in the United States caused delays in winter wheat emergence and poor crop growth. Using an integrated approach, we quantitatively unraveled a 37% reduction in wheat production as being attributable to both per-harvested acre yield loss and severe crop abandonment, reminiscent of the Dust Bowl years in the 1930s. We used random forest machine learning and game theory analytics to show that the main driver of yield loss was spring drought, whereas fall drought dominated abandonment rates. Furthermore, results revealed, across the US winter wheat belt, the La Niña phase of the El Niño Southern Oscillation (ENSO), increased abandonment rates compared to the El Niño phase. These findings underscore the necessity of simultaneously addressing crop abandonment and yield decline to stabilize wheat production amid extreme climatic conditions and provide a holistic understanding of global-scale ENSO dynamics on wheat production. The bushfires that occurred in Australia in late 2019 and early 2020 were unprecedented in terms of their scale, intensity, and impacts. Using nitrogen dioxide (NO2) and carbon monoxide (CO) data measured by the Tropospheric Monitoring Instrument (TROPOMI), together with fire counts and fire radiative power (FRP) from MODIS, we analyzed the temporal and spatial variation of NO2 and CO column densities over three selected areas covering savanna and temperate forest vegetation. The ΔNO2/ΔCO emission ratio and emission factor were also estimated. The ΔNO2/ΔCO emission ratio was found to be 1.5 ± 1.2 for temperate forest fire and ranged from 2 ± 1.3 to 2.8 ± 1.8 for savanna fire. For savanna and temperate forest fires, satellited-derived NOx emission factors are 1.29 g kg-1 and 1.2 g kg-1 separately, while CO emission factors are 62.34 and 112.5 g kg-1. This study demonstrates that the large-scale emission ratio from the TROPOMI satellite for different biomass burnings can help identify the relative contribution of smoldering and flaming activities in a large region and their impacts on the regional atmospheric composition and air quality.
