2. Art as a distributed ecosystem : mapping the limits of systems-based art

Abstract

My practice-based fine art research is shaped and informed by my transdisciplinary background and specifically my experience of ecology, geography and data visualisation. As a result I draw on the concepts of the 'ecosystem' and the 'information system' to consider how an artwork can be understood in ecological and informational terms, especially in relation to my own engagement with 'Systems Art', a mode of practice which shares a systems thinking foundation with these concepts. This research identifies three periods of systems thinking influencing culture, and this understanding has helped structure the thesis. Firstly, the holistic ecology movements of the mid-1960s which generated expanded expectations of the term 'system', secondly, the shift towards the 'informational' inherent in the critiques of linguistic and institutional systems of the 1980s, finally, our current 'post-systems' condition in which the pervading and infinitely complex nature of socio-environmental systems and their simulation is acknowledged. This three-fold journey is marked by incremental modifications to the definition of a system as the idea takes on greater extension through Latour's Actor-Network Theory and Deleuze and Guattari's Assemblage Theory. Later, the incomprehensibility of complex systems is conjured through references to Timothy Morton's concept of the Hyperobject and Object-Oriented Ontology's notion of 'withdrawal'. The practical component of my research is a body of artwork, which is a product of my engagement with systems thinking, the natural environment, complex information, image processing and painting. Within the material production of two and three-dimensional artworks, the perspective I have gained is that of a researcher exploring the analogical potential of an extensively 'distributed ecosystem'. If this perspective is persuasive, then artists interested in systems should be encouraged to not only map more expansive and complex boundaries for their work, but also articulate the unknowability of these complex systems as a knowable dimension of the distributed meaning of 'Systems Art'.

6. Characterization of mismatch‐repair/microsatellite instability‐discordant endometrial cancers.

Abstract

Background: Mismatch‐repair (MMR)/microsatellite instability (MSI) status has therapeutic implications in endometrial cancer (EC). The authors evaluated the concordance of testing and factors contributing to MMR expression heterogeneity. Methods: Six hundred sixty‐six ECs were characterized using immunohistochemistry (IHC), MSI testing, and mut‐L homolog 1 (MLH1) methylation. Select samples underwent whole‐transcriptome analysis and next‐generation sequencing. MMR expression of metastatic/recurrent sites was evaluated. Results: MSI testing identified 27.3% of cases as MSI‐high (n = 182), MMR IHC identified 25.1% cases as MMR‐deficient (n = 167), and 3.8% of cases (n = 25) demonstrated discordant results. A review of IHC staining explained discordant results in 18 cases, revealing subclonal loss of MLH1/Pms 1 homolog 2 (PMS2) (n = 10) and heterogeneous MMR IHC (mut‐S homolog 6 [MSH6], n = 7; MLH1/PMS2, n = 1). MSH6‐associated Lynch syndrome was diagnosed in three of six cases with heterogeneous expression. Subclonal or heterogeneous cases had a 38.9% recurrence rate (compared with 16.7% in complete MMR‐deficient cases and 9% in MMR‐proficient cases) and had abnormal MMR IHC results in all metastatic recurrent sites (n = 7). Tumors with subclonal MLH1/PMS2 demonstrated 74 differentially expressed genes (determined using digital spatial transcriptomics) when stratified by MLH1 expression, including many associated with epithelial–mesenchymal transition. Conclusions: Subclonal/heterogeneous MMR IHC cases showed epigenetic loss in 66.7%, germline mutations in 16.7%, and somatic mutations in 16.7%. MMR IHC reported as intact/deficient missed 21% of cases of Lynch syndrome. EC with subclonal/heterogeneous MMR expression demonstrated a high recurrence rate, and metastatic/recurrent sites were MMR‐deficient. Transcriptional analysis indicated an increased risk for migration/metastasis, suggesting that clonal MMR deficiency may be a driver for tumor aggressiveness. Reporting MMR IHC only as intact/deficient, without reporting subclonal and heterogeneous staining, misses opportunities for biomarker‐directed therapy. Plain Language Summary: Endometrial cancer is the most common gynecologic cancer, and 20%–40% of tumors have a defect in DNA proofreading known as mismatch‐repair (MMR) deficiency.These results can be used to guide therapy.Tests for this defect can yield differing results, revealing heterogeneous (mixed) proofreading capabilities.Tumors with discordant testing results and mixed MMR findings can have germline or somatic defects in MMR genes.Cells with deficient DNA proofreading in tumors with mixed MMR findings have DNA expression profiles linked to more aggressive characteristics and cancer spread.These MMR‐deficient cells may drive tumor behavior and the risk of spreading cancer. Intratumoral heterogeneity, including subclonal and heterogeneous mismatch‐repair (MMR) expression, can yield discordant results from diagnostic assays, such as immunohistochemistry and polymerase chain reaction‐based microsatellite instability testing. Within endometrial cancers, subclonal or heterogeneous MMR expression may be attributed to germline or somatic mutations in MMR proteins, and the authors demonstrate that these tumors are characterized by clonal selection favoring the propagation of MMR deficiency at sites of metastasis or recurrence. Abstract

Contemporary systems painting directly engages with the material of contemporary culture, not necessarily the technological substrates of computation, social media, the Internet, and artificial intelligence, but the concept of the algorithm and the circulation and patterning of information at the limit of human apprehension. Systems painting emerged as part of the wider category of systems art in the 1960s—a heterogenous collection of artists who were focused on the exploration of social, ecological, and technological systems, and the processes that underpin them. These systemic fields increasingly define and shape our lifeworld in the 21st century, producing an excess of algorithmically generated information. It is, therefore, appropriate to consider the role system painting plays in addressing the conceptual, aesthetic, and affective aspects of information derived from computational, algorithmic, and rule-based processes. This paper discusses the practice of the contemporary systems painter James Hugonin and his series of paintings Fluctuations in Elliptical Form (2015–2021). Karl Popper's theory of three worlds is introduced, and the concepts of 'concrete' and 'abstract' objects are described and applied to Hugonin's painting as a way of understanding the role externalised rules and internal intuitive decisions play in the construction of these complex and visually mesmerising paintings. Abstract

This document is a reply to a previous article discussing the significance of tumors with "unusual staining" from mismatch repair (MMR) immunohistochemistry (IHC) in endometrial cancer. The authors agree that ongoing conversation and multiple testing strategies are needed to improve outcomes for patients. They also emphasize the importance of quality assurance in performing and interpreting MMR IHC, including uniform fixation and grossing protocols. The authors acknowledge the potential benefits of primary assessment of MMR IHC staining on biopsy specimens but note that in their cohort, preoperative biopsies were performed outside their institution and MMR IHC was not performed on these specimens. They also discuss the interpretation of MSI testing in endometrial cancer and the use of the Promega panel for clarifying cases that are MSI-low on the National Cancer Institute panel. Further research is needed to define optimal testing for MMR-deficient/MSI endometrial cancer and predict response to immunotherapy. [Extracted from the article]

Abstract

At both the individual and societal levels, we are entangled within environmental, social, and technological systems that shape our material and emotional states. Contemporary art needs to integrate and challenge the information circulating within these interacting systems to address our increasingly complex lifeworld. This systemic understanding emerged in the 1960s as part of a broader growth in relational thinking within the natural and social sciences, which extended the conceptual boundaries of the artwork. The ecosystem, a model originally developed within ecology, is an example of a systems model as it describes the flow of matter, energy, and information through the physical world. This model has evolved into a powerful analogical tool to describe contemporary culture's entanglement with nature and technology. The ecosystem model is invoked here to describe how information flows through the artwork. The paper suggests that art is a vital form of communication as it can channel noise or unknown information. This channelling is demonstrated with the artwork, The Creation Myth (1998), by Jason Rhoades. This work anticipated the convergence of natural and technological systems, and it demonstrates the ability of the arts to channel unknown messages or noise, thereby disrupting the dominant signals of contemporary culture. Abstract

Dominant mutations in the Ret receptor tyrosine kinase lead to the familial cancer syndrome multiple endocrine neoplasia type 2 (MEN2). Mammalian tissue culture studies suggest that [Ret.sup.MEN2] mutations significantly alter Ret-signaling properties, but the precise mechanisms by which [Ret.sup.MEN2] promotes tumorigenesis remain poorly understood. To determine the signal transduction pathways required for [Re.sup.MEN2] activity, we analyzed analogous mutations in the Drosophila Ret ortholog dRet. Overexpressed d[Re.sup.MEN2] isoforms targeted to the developing retina led to aberrant cell proliferation, inappropriate cell fate specification, and excessive Ras pathway activation. Genetic analysis indicated that d[Ret.sup.MEN2] acts through the Ras-ERK, Src, and Jun kinase pathways. A genetic screen for mutations that dominantly suppress or enhance d[Ret.sup.MEN2] phenotypes identified new genes that are required for the phenotypic outcomes of d[Ret.sup.MEN2] activity. Finally, we identified human orthologs for many of these genes and examined their status in human tumors. Two of these loci showed loss of heterozygosity (LOH) within both sporadic and MEN2-associated pheochromocytomas, suggesting that they may contribute to Ret-dependent oncogenesis.
