Abstract “Tissue engineering,” is a concept that involves the use of scaffolds, cells, and growth factors/signaling molecules in the field of regenerative medicine. The present study was carried out to evaluate the attachment and depth of penetration of the Gingival Mesenchymal Stem Cells (GMSCs) onto the collagen scaffold preconditioned with Fetal Bovine Serum (FBS), Injectable Platelet Rich Fibrin (i-PRF) and a combination of FBS with i-PRF using histological analysis and environmental scanning electron microscopy (ESEM) respectively. In the present study, commercially available collagen membranes were used as scaffolds and divided into 3 groups where Group I was preconditioned with FBS, Group II was preconditioned with i-PRF, and Group III with a combination of FBS and i-PRF. Scaffolds were then seeded with GMSCs and incubated in a CO2 incubator at 37 0C for 7 days. Both histological and ESEM analysis showed proliferation, attachment, and depth of penetration along with the combination of cell morphological changes in all the three groups. In group I cells showed mild depth of penetration along with a round morphological appearance. In group II the cells showed moderate depth of penetration and rounded morphology. In group III maximum depth of penetration was seen along with the round and spheroidal morphology of cells. The combination of the growth media showed the best effect on cell attachment, depth of penetration, and cell morphology. This is the first report demonstrating the effect of the combination of collagen and i-PRF supports the proliferation of GMSCs. Since FBS alone and i-PRF alone exhibited similar cell proliferation it is recommended that i-PRF could be used in clinical scenarios making it xenofree.

Investments in Industry 4.0 are making a significant impact on various sectors by integrating advanced digital technologies. The chances of financial losses for organisations' investments due to new technologies are high, and there is a low possibility of recovering the investments. Reduced spending on long-term costs by organisations due to low investments and consumer demand for durable goods has resulted in sluggish global trade in advanced and emerging markets. Challenges have arisen for companies in securing loans for investments due to past economic slowdowns, resulting in increased corporate borrowing expenses. The literature acknowledges Supply Chain Finance (SCF) as a vital approach to optimising efficiency. Industry leaders are turning to SCF as high costs persist during the transformation process for manufacturers. SCF can unlock substantial working capital by improving cash flow, which can be used to back essential business initiatives. This evolving environment presents opportunities and requires a deeper investigation of possibilities and challenges in advancing SCF to support Industry 4.0 efforts. Given this, the Special Issue released in the International Journal of Production Research pushes forward this crucial research focus. Nine articles accepted in this Special Issue used different methodologies, theories, and contexts to address research needs and offer valuable insights on supply chain finance and industry 4.0 investment challenges. Background With the increase in popularity of cannabis and its use and the lack of large‐scale data on cannabis use and venous thromboembolism and pulmonary embolism (PE), we used a nationally representative cohort of young adults (aged 18–44 years) to compare the odds of admissions and in‐hospital mortality of PE with and without cannabis use disorder (CUD). Methods and Results Identified patients with PE using the National Inpatient Sample (2018) were compared for baseline, comorbidities, and outcomes. Multivariable regression analysis, adjusted for covariates, was used to compare the odds of PE in young patients with CUD (CUD+) versus those without (CUD−) and those with prior venous thromboembolism. Propensity score–matched analysis (1:6) was also performed to assess in‐hospital outcomes. Digital Twins (DTs) hold significant promise in addressing the challenges faced by food supply chains (FSCs). This paper aims to provide critical insights into the potential for Digital Twins to meet the key challenges of the FSC and establish a comprehensive conceptual framework for their implementation. Following a systematic literature review (SLR), the study identified 81 peer-reviewed, high-quality papers published over the last decade (2012–2023). The typology-driven thematic analysis emphasises the emergent nature of DTs within FSCs, highlighting their key characteristics including monitoring, real-time simulation, and scenario analysis. The identified characteristics, applications, implementation drivers and barriers of Digital Twin form the basis for a novel conceptual framework for implementing DTs in FSCs. Leveraging insights from Innovation Adoption Theory and the Technology-Organization-Environment (TOE) framework, the study outlines a structured five step implementation process divided into three stages. Notably, technology assessment and performance evaluation emerge as two innovative steps necessary for the successful implementation of DTs specifically, not previously considered by the theory. The study identifies promising avenues for future research. Background: Traditionally, mechanical thrombectomy performed for pulmonary embolism (PE) necessitates the utilization of iodinated contrast. Intravascular ultrasound (IVUS) has been used as a diagnostic and therapeutic modality in the management of acute high and intermediate-risk PE. Recently, with the shortage of contrast supplies and the considerable incidence of contrast-induced acute kidney injury (CI-AKI), other safer and more feasible IVUS methods have become desirable. The purpose of this systematic review was to evaluate the importance of IVUS in patients with PE undergoing thrombectomy. Methods: Medline/PubMed, Embase, Scopus, and Google Scholar were searched for review studies, case reports, and case series. Clinical characteristics, outcomes and the usage of IVUS-guided mechanical thrombectomy during the treatment of acute high and intermediate-risk PE were examined in a descriptive analysis. Results: In this systematic review, we included one prospective study, two case series, and two case reports from July 2019 to May 2023. A total of 39 patients were evaluated; most were female (53.8%). The main presenting symptoms were dyspnea and chest pain (79.5%); three patients (7.9%) presented with syncope, one with shock and one with cardiac arrest. Biomarkers (troponin and BNP) were elevated in 94.6% of patients. Most patients (87.2%) had intermediate-risk PE, and 12.8% had high-risk PE. All patients presented with right-heart strain (RV/LV ratio ≥ 0.9, n = 39). Most patients (56.4%) had bilateral PE. Mechanical thrombectomy was performed using IVUS without contrast utilization in 39.4% of the patients. After the initial learning curve, contrast usage decreased gradually over time. There was a significant decrease in the composite mean arterial pressure immediately following IVUS-guided thrombectomy from 35.1 ± 7.2 to 25.2 ± 8.3 mmHg (p < 0.001). Post-procedure, there was no reported (0%) CI-AKI, no all-cause mortality, no major bleeding, or other adverse events. There was a significant improvement in symptoms and RV function at the mean follow-up. Conclusions: New evidence suggests that IVUS-guided mechanical thrombectomy is safe, with visualization of the thrombus for optimal intervention, and reduces contrast exposure.

Activation of the CXCL12/CXCR4/ACKR3 axis is known to aid myocardial repair through ischemia-triggered hypoxia-inducible factor-1α (HIF-1α). To enhance the upregulation of HIF-1α, we administered roxadustat, a novel prolyl hydroxylase inhibitor (PHI) clinically approved by the European Medicines Agency 2021 for the treatment of renal anemia, with the purpose of improving LV function and attenuating ischemic cardiomyopathy. Methods: We evaluated roxadustat’s impact on HIF-1 stimulation, cardiac remodeling, and function after MI. Therefore, we analyzed nuclear HIF-1 expression, the mRNA and protein expression of key HIF-1 target genes (RT-PCR, Western blot), inflammatory cell infiltration (immunohistochemistry), and apoptosis (TUNEL staining) 7 days after MI. Additionally, we performed echocardiography in male and female C57BL/6 mice 28 days post-MI. Results: We found a substantial increase in nuclear HIF-1, associated with an upregulation of HIF-1α target genes like CXCL12/CXCR4/ACKR3 at the mRNA and protein levels. Roxadustat increased the proportion of myocardial reparative M2 CD206+ cells, suggesting beneficial alterations in immune cell migration and a trend towards reduced apoptosis. Echocardiography showed that roxadustat treatment significantly preserved ejection fraction and attenuated subsequent ventricular dilatation, thereby reducing adverse remodeling. Conclusions: Our findings suggest that roxadustat is a promising clinically approved treatment option to preserve myocardial function by attenuating adverse remodeling.

Technology has revolutionized many walks of life from business to governance, learning to workplaces. This research aimed to examine students' perceptions towards the digital library services at Visvesvaraya National Institute of Technology. A survey of 176 engineering post-grads was used to accomplish this. Random sampling technique was used. Primary data collected through structured questionnaires. The research showed that respondents are aware of and use digital collections. The report examined students' views on internet libraries and digital collections. The survey also examined students' perceptions of digital library pros and cons. The digital library system's applicability was also examined, with a focus on the prototype model's course material presentation. The study showed that digital libraries can improve student learning. Studying student perspectives helps determine the efficacy and relevance of digital library services in academic settings. This research clarifies the existing state of digital library usage and lays the groundwork for improving digital library infrastructure and services to meet the changing needs of college students. Research on Blockchain implementation in the Pharmaceutical Supply Chains (PSC) is lacking despite its strong potential to overcome conventional supply chain challenges. Thus, this study aims to provide critical insight into the nexus between Blockchain and PSC and further build a conceptual framework for implementation within the pharmaceutical industry. Following a systematic literature review and text mining approach, 65 interdisciplinary articles published between 2010 and 2021 were studied to capture the decade long developments. Descriptive and thematic analysis showcases nascent developments of Blockchain in PSC. The drivers and barriers to adoption, implementation stages, and applications identified through the thematic analysis guide in setting the agenda for future research, primarily focussing on the use of Blockchain for drug counterfeiting, recall issues, along with other sector-specific challenges such as patient privacy, regulations and clinical trials. Research on Blockchain for PSC has been slow compared to other sectors, but has accelerated since the Covid-19 pandemic. Identified influential factors, implementation process and apparent applications are expected to influence researchers and practitioners in developing a roadmap for adopting Blockchain in the pharmaceutical industry. The rehabilitation process after the onset of a stroke primarily deals with assisting in regaining mobility, communication skills, swallowing function, and activities of daily living (ADLs). This entirely depends on the specific regions of the brain that have been affected by the stroke. Patients can learn how to utilize adaptive equipment, regain movement, and reduce muscle spasticity through certain repetitive exercises and therapeutic interventions. These exercises can be performed by wearing soft robotic gloves on the impaired extremity. For post-stroke rehabilitation, we have designed and characterized an interactive hand orthosis with tendon-driven finger actuation mechanisms actuated by servo motors, which consists of a fabric glove and force-sensitive resistors (FSRs) at the tip. The robotic device moves the user’s hand when operated by mobile phone to replicate normal gripping behavior. In this paper, the characterization of finger movements in response to step input commands from a mobile app was carried out for each finger at the proximal interphalangeal (PIP), distal interphalangeal (DIP), and metacarpophalangeal (MCP) joints. In general, servo motor-based hand orthoses are energy-efficient; however, they generate noise during actuation. Here, we quantified the noise generated by servo motor actuation for each finger as well as when a group of fingers is simultaneously activated. To test ADL ability, we evaluated the device’s effectiveness in holding different objects from the Action Research Arm Test (ARAT) kit. Our device, novel hand orthosis actuated by servo motors (NOHAS), was tested on ten healthy human subjects and showed an average of 90% success rate in grasping tasks. Our orthotic hand shows promise for aiding post-stroke subjects recover because of its simplicity of use, lightweight construction, and carefully designed components.

A hexanucleotide repeat expansion of C9orf72 is translated to dipeptide repeat proteins in amyotrophic lateral sclerosis and frontotemporal dementia patients. Here the authors generate a C. elegans model of C9orf72-mediated ALS/FTD and show that translation initiation factor eIF2D regulates the dipeptide repeat protein expression.

Initial double blind period of 16 weeks was followed by open-label phase till week 24. Efficacy assessment was performed at weeks 1, 4, 8, 12, 16, 20 and 24 based on best corrected visual acuity. Change in central macular thickness was assessed by optical coherence tomography from baseline to week 24. Immunogenicity assessment was done in both arms at baseline, week 16 and week 24. Safety evaluation included clinical and ophthalmic examination, adverse events, vital signs, laboratory parameters and immunogenicity in both treatment arms.Results: In the biosimilar test arm, 104 (98.11%) and 105 (99.06%) patients lost fewer than 15 letters in visual acuity at week 16 and week 24, respectively, compared with 53 (100%) at both follow-ups in reference arm. In the test arm, 27 (25.47%) and 34 (32.08%) patients gained at least 15 letters in visual acuity till week 16 and week 24 respectively, compared with 17 (32.08%) and 23 (43.30%) in the reference arm. In the test arm, mean change in central macular thickness at 24 weeks was − 89.93 μm against − 64.42 μm in the reference arm. Difference was statistically not significant for any endpoint at 16 and 24 weeks for the primary and secondary endpoints.Conclusion: The evaluation of efficacy, safety and immunogenicity was concluded to show no meaningful clinical difference for biosimilar ranibizumab with the reference product.Keywords: ranibizumab, biosimilar, AMD, intravitreal, visual acuity

The food grain production in India has progressively risen in the past few decades, whereas the storage capacity has remained limited. The policymakers in India are attempting to close this capacity gap while addressing sustainability objectives. However, the quantification and integration of multiple social sustainability factors have remained a challenge. To improve the overall sustainability, the study attempts to develop a mathematical model considering procurement, transportation, inventory, and location-related issues. Several supply chain network factors are integrated and assessed while focussing on the social sustainability dimension. Three cases of India's largest food grain-producing and consuming states are analysed with the help of two Pareto-based algorithms. Multiple relationships between variations in supply, demand, and the capacity of silos with three defined objectives are evaluated. It is observed that, the demand significantly influences the economic and environmental objectives compared with the supply and silo capacity. The capacity of silos has a more significant impact on social objectives than economic and environmental objectives. Results reveal the importance of establishing a sufficient number of modernised silos, which reduces environmental impact and improves social factors such as farmers' economic condition and welfare, balanced economic development, number of jobs created, and public health level. Background: The aim of transferring critical patient to hospital was providing highest specialized care and maintain the optimal health of patient. Critical ill patient was a person's baseline state of health has significantly changed and their life is at risk because of an illness and injury. Critically ill patient are those who at high risk for actual and potential life-threatening health problems. Critical illness is life threatening multisystem process that can result in significant morbidity and mortality, in most of patient critical illness is physiological deterioration. Objectives: To assess practices on shifting of critical patient among ambulance health care team members in experimental group before and after administration information booklet & to assess effectiveness of information booklet on practices on shifting of critical patient among ambulance health care team members in experimental group. Methods: Quantitative research approach and quasi experimental pre-test post-test control group design was used for the study. 120 ambulance health care team members were selected by purposive sampling technique. 60 samples were in experimental group and 60 samples were in control group. Data collected through structured observation checklist. Results: Majority (56.7%) participants were from 21-30 age group in experimental group and (41.7%) were from 31-40 age group in control group. Maximum (66.7%) and 51.7% were male in both groups. Majority of ambulance health care team members had 1-5 years of experience in both groups. Ambulance health care team members were (25%) of nurses, doctors Multipurpose Purpose Workers & drivers respectively. Pre-test findings were, majority (60%) in experimental and (51.7%) in control group had good practices on shifting of critical patients, whereas in post-test majority (93%) in experimental and (55%) in control group had good practices. The p value was <0.0001. So, result shows that, there was significant effect of information booklet on practices regarding shifting of critical patient among ambulance health care team members from selected hospitals at p<0.05 level of significance. Hence, Information booklet was effective in improving the practices level regarding shifting of critical patient. There is no association between practices and demographic variable among ambulance health care team members. Conclusion: The study concluded that there was improvement in practices after administration of information booklet regarding shifting of critical patient in experimental group. Embelin, isolated from Embelia ribes berries is a major constituent isolated by the Soxhlet process and possesses antimicrobial activity. In this report, we develop and evaluate the topical antimicrobial embe- lin niosomal cream formulation. Niosomal formulation of embelin was formulated using a thin-film hydra- tion method using Tween™ 60, chloroform and phosphate buffer with a rotary evaporator. This niosomal formulation was incorporated into the cream base, where both oil and water phase ingredients were prepared separately at 70 °C by constant stirring to form w/o (water-in-oil) cream formulation. This niosomal cream was evaluated for particle size, entrapment efficacy, spreadability, centrifugation, viscosity, dye test, homogeneity, in vitro antifungal and antibacterial activity against C. albicans and S. aureus, respectively. The zone of inhibition was calculated and further compared with the market formulations. The study proposed that niosomal formulations delivered sustained and prolonged delivery of drug with increase in bioavailability. [ABSTRACT FROM AUTHOR]

Context: Control of pain during dental treatment is an essential aspect of pediatric dentistry. Aims: This study was conducted to evaluate and compare the anesthetic efficacy of 4% articaine buccal infiltration with 2% lignocaine inferior alveolar nerve block (IANB) for primary mandibular molar extractions. Settings and Design: The study was a prospective, split-mouth, randomized controlled trial. Methods: Bilateral symmetrical carious primary mandibular molar (n = 92) extractions in 46 healthy children aged 5–10 years were included in this randomized controlled trial. Extraction was performed on one side using 4% of articaine buccal infiltration and on the contralateral side using 2% lignocaine IANB in two subsequent appointments. Pain and behavior were assessed at baseline, during injection and extraction using Wong–Baker Faces Pain Rating Scale, Modified Behavior Pain Scale (MBPS), and Frankl Behavior Rating Scale. Statistical Analysis Used: Values thus obtained were statistically analyzed by one-way analysis of variance test and compared using independent samples test. Results: According to MBPS, the mean value of pain experienced in the form of cry during injection was reported to be more for 2% lignocaine IANB (1.76) as compared to 4% articaine buccal infiltration (1.30), which was statistically significant (P = 0.024). Comparison of behavior depicted showed no statistically significant difference between the groups. Conclusion: Buccal infiltration with 4% articaine can be utilized as an effective alternative to 2% lignocaine IANB for primary mandibular molar extractions.
